Verfahren zur Herstellung von neuen Steroidguanylhydrazonen
Gegenstand der österr. Patentschrift Nr. 261816 ist die Herstellung von Kondensationsprodukten aus Steroiden mit mehr als einer Carbonylfunktion und Aminoguanidin bzw. seinen Salzen ; diese Kondensationsprodukte sind herzwirksam.
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Steroidguanylhydrazonen der allgemeinen Formel

Die bei der Kondensation entstehenden neuen Verbindungen, die noch freie Carbonylfunktionen enthalten können, sind herzwirksam.
Die Erzeugnisse des erfindungsgemässen Verfahrens können als solche oder in Form ihrer Salze mit nichttoxischen anorganischen oder organischen Säuren angewendet werden. Geeignete Säuren sind z. B.
Salzsäure, Phosphorsäure, Essigsäure, Propionsäure, Maleinsäure, Fumarsäure, Bernsteinsäure, Weinsäure, Citronensäure, Salicylsäure, Naphthalin-1,5-disulfonsäure usw.
Die als Ausgangsmaterialien verwendbaren substituierten S-Alkylisothiosemicarbazone von Steroiden mit mehr als einer Carbonylfunktion erhält man durch Umsetzung von Steroiden mit mehr als einer Carbonylfunktion mit substituierten S-Alkylisothiosemicarbaziden bzw. substituierten Thiosemicarbaziden und Alkylhalogeniden.
Geeignete Steroide für das erfindungsgemässe Verfahren sind z. B.

2 g Dihydrochlorid des Pregn-4-en-3, 20-bis- (S-methylisothiosemicarbazons) löst man in 25 ml Äthanol, versetzt mit 1 g ss-Pyrrolidinoäthylamin und erhitzt 12 h unter Rückfluss. Dann dampft man i. V. zur Trockne ein, verreibt den Rückstand mit Wasser und trennt das Unlösliche ab. Dieses wird in wenig Äthanol gesetzt und die Lösung in Äther eingerührt. Die ausfallenden Flocken werden abgesaugt, mit Aceton ausgekocht und aus Methanol/Äthanol umkristallisiert. 6 g Tetrahydrochlorid des Pregn-4- en - 3, 20-bis- [4- (ss-pyirolidinoäthyl)-guanylhydrazons], Fp. 273 bis 2760 C Zers.
Beispiel 2 : 2, 8 g Progesteron werden in 100 ml Methanol gelöst und mit einer Lösung von 3 g N-Methyl-N'-amino-S-methyl-isothioharnstoff-hydrochlorid in Methanol versetzt. Man stellt durch Zugabe von methanolischer Salzsäure einen pH-Wert von 2 ein und rührt 24 h bei Raumtemperatur. Dann rührt man in Äther ein, saugt den entstehenden Niederschlag ab, kocht ihn mit Aceton aus und fällt aus Alkohol/Äther um.
