Verfahren zur Herstellung von Bis- (pyridyl-4-thio) -alkanen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Bis- (pyridyl-4-thio)-alkanen der allgemeinen Formel :

worin R einen geradkettigen oder verzweigten, gesättigten oder ungesättigten Alkylenrest mit 1-16 Kohlenstoffatomen, dessen Kette gegebenenfalls weiterhin durch Sauerstoff oder Schwefel unterbrochen sein kann, und R'Wasserstoff oder die Methylgruppe bedeuten.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass Pyridyl-pyridinium-halogenide der allgemeinen Formel :

in der R'Wasserstoff oder die Methylgruppe und Hal ein Halogenatom bedeuten, in Gegenwart eines Lösungsmittels und in Anwesenheit einer anorganischen oder organischen Base mit Schwefelwasserstoff und anschliessend mit einem Alkylendihalogenid der Formel :
Hal-R-Hal, in der Rund Hal die vorstehend angegebene Bedeutung haben, umgesetzt werden. Sowohl die Umsetzung mit dem Schwefelwasserstoff als auch die anschliessende Umsetzung mit dem Alkylendihalogenid erfolgt vorzugsweise bei Temperaturen unter 150 C und insbesondere bei Temperaturen zwischen 60 und 1000 C.
Es war bekannt, dass sich Pyridylalkylthioäther nur unter Druck und in mässiger Ausbeute herstellen lassen, es war ferner bekannt, dass sich Alkylen-bis-thioäther, beispielsweise Alkylen-bis-thioäther des p-Nitrothiophenol nur bis zu einer Kettenlänge von maximal 3 Kohlenstoffatomen herstellen lassen. Es ist daher durchaus überraschend, dass sich die Verbindungen der obigen allgemeinen Formel ohne Anwendung von Druck unter milden Reaktionsbedingungen in ausgezeichneter Ausbeute herstellen lassen, wobei der Alkylenrest R bis zu 16 Kohlenstoffatome enthalten kann.
Bei der Durchführung des erfindungsgemässen Verfahrens kann das Pyridyl-pyridinium-halogenid beispielsweise zunächst in einem organischen Lösungsmittel in Gegenwart einer tertiären Base gelöst bzw. aufgeschlämmt werden, worauf das Reaktionsgemisch unter Erwärmen mit Schwefelwasserstoff gesättigt wird. Die tertiäre Base, vorzugsweise Pyridin, kann auch selbst als Lösungsmittel verwendet werden. Anschliessend wird das Reaktionsgemisch bei Temperaturen unter 150 C, vorzugsweise bei Temperaturen zwischen 60 und 100 C, direkt mit dem entsprechenden Alkylendihylogenid einige Stunden erwärmt, nach dem Erkalten alkalisch gemacht und das gebildete Di- (pyridyl-4-thio)-alkan entweder

abgenutscht oder aus dem Reaktionsgemisch mit einem geeigneten Lösungsmittel, beispielsweise Chloroform, extrahiert.
Die Ausbeuten betragen hiebei bis zu 75% der Theorie.
Eine besonders günstige Ausführungsform des Verfahrens besteht darin, dass nach dem Sättigen mit Schwefelwasserstoff die tertiäre Base, die vorzugsweise auch als Lösungsmittel verwendet wird, im Vakuum abdestilliert wird, und die Umsetzung mit dem Alkylendihalogenid in wässeriger, wässerig-alkoholischer oder alkoholischer Lösung in Gegenwart von Alkalihydroxyden oder Alkalicarbonaten im oben angegebenen Temperaturbereich erfolgt.
Auf diese Weise wird die verwendete tertiäre Base quantitativ zurückgewonnen und die Aufarbeitung des Reaktionsgemisches erleichtert, da hiebei kristalline Produkte entstehen, die leicht abgenutscht werden können, während bei Verwendung organischer Basen als säurebindende Mittel, insbesondere bei Verwendung von Pyridin, bisweilen verschmierte Reaktionsprodukte entstehen, die durch Extraktion der Base mittels eines organischen Lösungsmittels aus dem Reaktionsgemisch isoliert werden müssen.
Man kann auch völlig auf die Verwendung organischer Lösungsmittel und organischer Basen verzichten und die Reaktion so durchführen, dass man das 4-Pyridyl-pyridiniumsalz in Wasser unter Zusatz einer anorganischen Base, beispielsweise Natriumbicarbonat, mit H2S in der beschriebenen Weise behandelt und anschliessend die Lösung in Gegenwart der gleichen oder einer andern anorganischen Base, beispielsweise NaOH, mit dem Dihalogenid unter Rühren bei den angegebenen Temperaturen zur Umsetzung bringt. Man hat so den Vorteil, ausschliesslich anorganische Reagentien verwenden zu können, jedoch sind die erzielten Ausbeuten geringer.
Die auf diese Weise erhaltenen Bis- (pyridyl-4-thio)-alkane sind Zwischenprodukte zur Gewinnung von Heilmitteln, die zur Bekämpfung von durch Pilze und Bakterien hervorgerufenen Krankheiten dienen.
Im folgenden soll die Durchführung des erfindungsgemässen Verfahrens an Hand einiger Beispiele erläutert werden.
Beispiel 1 : Di- (pyridyl-4-thio)-methan.
29 g 4-Pyridyl-pyridinium-chlorid-hydrochlorid und 29 cm3 Pyridin werden bei 60-80 C unter Rühren mit Schwefelwasserstoff gesättigt. Anschliessend gibt man 11 g Methylenbromid zu und erwärmt 2 Stunden auf 60-80 C. Der gebildete Niederschlag wird in wenig Wasser suspendiert, mit Natronlauge alkalisch gemacht und nach Kühlung abgesaugt. Nach Umkristallisation aus wässerigem Methanol erhält man 7, 1 g (48% der Theorie) des Dithioäthers vom F. = 148-149 C.

Beispiel 3 : l, 10-Di- (pyridyl-4-thio)-n-decan.
17, 5 g 4-Pyridyl-pyridiniumchlorid-hydrochlorid und 20 cm3 Pyridin werden bei 60-80 C mit Schwefelwasserstoff gesättigt und anschliessend im Vakuum von überschüssigem Pyridin befreit.
Der Rückstand wird mit der berechneten Menge 24%iger Natronlauge versetzt, durch Ausäthern vom restlichen Pyridin befreit und nach Zugabe von 50 cm3 Äthanol sowie 8 g 1, 10-Dichlordecan 3 Stunden am Rückfluss erhitzt. Beim Abkühlen fällt der Dithioäther aus und wird nach dem Absaugen aus Aceton umkristalliesiert. Man erhält so 10 g (73% der Theorie) des Produktes vom F. = 880 C.

Beispiel 4 : l, 10-Di- (pyridyl-4-thio)-n-decan).
20 g 4-Pyridyl-pyridinium-chlorid-hydrochlorid und 18 g NaHCOg in 50 ml Wasser werden bei 800 C mit H2S gesättigt. Nach Zugabe von 6 g NaOH und 11, 2 g Dibromdecan wird die Mischung unter Rühren li Stunden auf dem Wasserbad erhitzt. Das beim Erkalten ausgefallene Produkt liefert nach Absaugen und Umkristallisieren aus Aceton 5 g (35% der Theorie) des Produktes von Schmp. 88 C.

Nach Zugabe von 9 g 1, 5-Dibrom-3-methyl-pentan zur alkalischen Lösung bildet sich bei 4stündigem Erhitzen auf 90 C ein Öl, das nach Extraktion mit Chloroform und Vertreibung des Lösungsmittels im Vakuum bei 0, 15 mm Hg und 225-2300 C übergeht. Eine aus 6, 0 g 4-Pyridylpyridinium-chlorid-hydrochlorid nach vorstehender Vorschrift bereitete alkalische Lösung von 4-Mercaptopyridin wird mit 1, 0 g l, 4-Dichlorbutin- (2) versetzt und 8 Stunden unter Rühren bei 500 C gehalten. Nach Kühlung lässt sich das gebildete Produkt absaugen und nach dem Trocknen aus Essigester umkristallisieren. Die Ausbeute beträgt 1, 0 g (30% der Theorie) ; der Schmelzpunkt liegt bei 128-130 C.
