Verfahren zur Herstellung des neuen Coffeino-8- (ss-Phenyl-isopropyl) -amin
Es ist bereits eine Reihe in der 8-Stellung des Coffeins mit Basen substituierter Derivate aufgebaut worden (E. Fischer, Liebig Ann.
Chem. 215,253 [1882] ; L. Cramer, Ber. dtsch. chem. Ges. 27,3098 [1894] ; A. Einhorn und E. Baumeister, Ber. dtsch. chem. Ges. 31,1138 [1898] und J. Gomberg, Am. chem. J. 23,51 [1901) ; F-F. Blicke u. H. C. Godt Jr., Journ.
Amer. chem. Soc. 76,2835 [1954)]. Eine Bedeutung haben diese Verbindungen nicht erlangen können, da die Substanzen gegenüber dem unsubstituierten Coffein in keiner Richtung irgendwelche Vorteile zeigten, im Gegenteil, es wurde entweder die Wirkung des Coffein herabgesetzt, deren Giftigkeit gesteigert oder schliesslich die Wirkung auch ganz ausgelöscht.
Es wurde nun überraschend gefunden, dass das neue, erfindungsgemässe, noch nicht beschriebene Coffeino- (8) - (ss- Pheny1-isopropyl) -amin der nachfolgenden Konstitution :

welches als Racemat und in seinen Isomeren erhalten werden kann, auf das Zentralnervensystem eine überlegene stimulierende Wirkung gegenüber Coffein besitzt, dass es weiters gegen- über Barbitursäurederivaten antagonistisch wirkt, ohne dass die zentralstimulierende Wirkung bis zur Weckwirkung gesteigert wird. Es ist an sich bekannt, dass Coffein die Schlaf- und Beruhigungswirkung, welche durch Barbiturate im Tierversuch erzeugt wird, nicht in belebenden Sinne zu beeinflussen vermag.
Die verstärkte, zentralstimulierende Wirkung des Coffeino- (8) -isopropyl-phenylamins, welches auch in Tierversuchen keine kreislaufwirksamen Eigenschaften entfaltet, ist insofern überraschend,

Die zentralerregende Wirkung der neuen erfindungsgemässen Verbindung ist, gemessen an der Motilitätssteigerung an Mäusen und Ratten und an deren Versuchsbedingungen, etwa 2-3mal geringer als diejenige von 2-Phenyl-3-methylmorpholin und entschieden stärker als bei Coffein.
Bekanntlich haben solche zentralstimulierende Substanzen die grösste Bedeutung, welche einerseits auch in hohen Dosen keine Kreislaufwirkung entfalten und anderseits mild stimulierend wirken, so dass Ermüdungserscheinungen durch einen gewissen Belebungseintritt beseitigt werden.
Es ist neben der neuen, nach dem erfindunggemässen Verfahren dargestellten Substanz noch keine Substanz beschrieben worden, welche diese Vorteile besitzt.
Die neue Verbindung kann nach an sich bekannten Methoden und Verfahren der Aminoalkylierung hergestellt werden. Vorteilhaft ist die Umsetzung von 8-Halogencoffein mit ss- Phenylisopropylamin, wobei die Umsetzung in

Lösungsmitteln, wie Toluol, Tetralin, Xylolen oder Alkoholen mehrere Stunden gekocht werden.
Alkalibromid und Alkalijodid beschleunigen die Reaktion.
Die neue, erfindungsgemässe Substanz lässt sich mit Mineralsäure, wie Salzsäure, Schwefelsäure oder organischen Säuren, wie Oxalsäure, Bernsteinsäure, Ascorbinsäure, Nikotinsäure in Salze überführen. Sowohl die freie Base wie ihre Salze sollen als Heilmittel bzw. Zwischen-

stufe zur Herstellung von Heilmitteln Verwendung finden.
Beispiel 1 : d, 1-N- [Coffeino- (8)]- (ss-Phenyl- isopropyl)-amin. 44 g 8-Chlorcoffein werden mit 44 ml d, l- (ss-Phenyl)-isopropyl-amin durchtränkt, durchgerührt und innerhalb 15-20 Minuten langsam unter Rühren auf 1400 C erhitzt. Das 8-Chlorcoffein färbt sich anfangs schwach violett und geht langsam in Lösung. Die Temperatur beginnt nun auf 170 C zu steigen. Man erhitzt weiterhin 20-30 Minuten auf 160-170 C, wobei das Reaktionsgut dickflüssig wird und eine gelbe Farbe annimmt, Nun lässt man die Temperatur auf 160-150 C fallen und hält auf dieser Temperatur noch 30 bis 40 Minuten. Hierauf lässt man 2 Stunden abkühlen, wobei eine dickflüssige, gelbe Reaktionsmasse resultiert. Diese wird mit 70 ml 96%igem Äthanol aufgekocht und heiss filtriert.
Man lässt 5 Stunden stehen und entfernt einen eventuellen Niederschlag durch Filtration. Daraufhin verdünnt man mit 20 ml Wasser, wobei eine schwache Trübung eintritt, die wieder verschwindet. Nach 6-12 Stunden ist der Kolbeninhalt in Form farbloser Nadeln auskristallisiert. Man setzt nochmals 10-30 ml Wasser zu und lässt weiter auskristallisieren. Daraufhin saugt man die Kristalle ab. Ausbeute zirka 35 g ; durch Verdünnen der Mutterlauge mit Wasser erhält man weitere Mengen, so dass die Ausbeute zirka 43 g beträgt. F. : 172-174 C ; löslich in Benzol, Alkohol, unlöslich in Wasser und Äther, in Mineralsäure unter Bildung mineralsaurer Salze löslich, ebenso in organischen Carbonsäurelösungen. Zum gleichen Produkt kommt man durch 48 Stunden langes Kochen von 8-Bromcoffein und Phenylisopropylamin in Toluol.
Beispiel 2 : d-N- [Coffeino- (8)]- (ss-Phenyl-iso- propyl)-amin. 44 g 8-Chlorcoffein (oder eine entsprechende Menge von 8-Bromcoffein) werden mit 44 ml d-(ss-Phenyl)-isopropyl-amin, wie in Beispiel 1 erhitzt. Es resultiert eine dickflüssige, gelbe Reaktionsmasse, die mit 50 ml 96%igem Alkohol aufgenommen wird. Es wird heiss filtriert, sodann 15-20 ml Wasser zugesetzt.
Es kristallisiert alles innerhalb 3 Stunden aus. Ausbeute zirka 46 g ; F. : 103-105'C, wird bei 120 C wieder fest und schmilzt bei 167-169 bei 1200 C wieder fest und schmilzt bei 167 bis 169 0 C.
Umkristallisation lässt sich aus Benzol oder zweckmässiger Alkohol und Wasser durchführen.
40 g des Rohproduktes werden in 50 ml 96%igem Äthanol gelöst. Dann gibt man 10 ml Wasser zu und lässt 24 Stunden stehen. Die neue Base fällt in kugelförmigen Kristallen aus,

Umkristallisation beträgt die Ausbeute 35 g.

These compounds were not able to acquire any significance, as the substances showed no advantages in any way over the unsubstituted caffeine, on the contrary, either the effect of the caffeine was reduced, its toxicity increased or the effect was finally extinguished completely.

The central stimulating effect of the new compound according to the invention, measured by the increase in motility in mice and rats and by their test conditions, is about 2-3 times less than that of 2-phenyl-3-methylmorpholine and decidedly stronger than that of caffeine.
It is known that such centrally stimulating substances are of the greatest importance, which on the one hand do not develop any circulatory effects even in high doses and on the other hand have a mild stimulating effect, so that symptoms of fatigue are eliminated by a certain activation.
Apart from the new substance produced by the method according to the invention, no substance has yet been described which has these advantages.

Example 1: d, 1-N- [caffeineo- (8)] - (ss-phenylisopropyl) amine. 44 g of 8-chlorocaffeine are impregnated with 44 ml of d, l- (ss-phenyl) isopropylamine, stirred and slowly heated to 1400 ° C. over the course of 15-20 minutes while stirring. The 8-chlorocaffeine initially turns a pale purple color and slowly dissolves. The temperature now begins to rise to 170 C. The mixture is heated to 160-170 ° C. for 20-30 minutes, during which the reaction mixture becomes viscous and takes on a yellow color. The temperature is now allowed to drop to 160-150 ° C. and is maintained at this temperature for another 30 to 40 minutes. It is then allowed to cool for 2 hours, a viscous, yellow reaction mass resulting. This is boiled with 70 ml of 96% ethanol and filtered hot.

F. 172-174 C; soluble in benzene, alcohol, insoluble in water and ether, soluble in mineral acid with formation of mineral acid salts, also in organic carboxylic acid solutions. The same product is obtained by boiling 8-bromocaffeine and phenylisopropylamine in toluene for 48 hours.
Example 2: d-N- [caffeineo- (8)] - (ss-phenyl-isopropyl) -amine. 44 g of 8-chlorocaffeine (or a corresponding amount of 8-bromocaffeine) are heated with 44 ml of d- (ss-phenyl) -isopropyl-amine as in Example 1. A thick, yellow reaction mass results, which is taken up with 50 ml of 96% alcohol. It is filtered hot, then 15-20 ml of water are added.
Everything crystallizes out within 3 hours. Yield about 46 g; F.: 103-105'C, becomes solid again at 120 C and melts again at 167-169 at 1200 C and melts at 167 to 169 0 C.
Recrystallization can be carried out from benzene or an appropriate alcohol and water.
