Verfahren zur Darstellung von 6-Monoalkyl-bzw.-aralkylamino-2-methylhepten- (2).
Zur Beseitigung von spastischen Zuständen wurden in der Therapie bisher hauptsächlich Papaverin bzw. dessen Abkömmlinge verwendet. Diese ziemlich kompliziert gebauten Verbindungen gehören in chemischer Beziehung der Isochinolinreihe an.

Gemäss dem Verfahren werden diese neuen Verbindungen durch Kondensation von 2-Methyl- hepten- (2)-on- (6) mit primären Alkyl-bzw. Aralkylaminen und anschliessende Reduktion des Kon- densationsproduktes erhalten. Diese beiden Reaktionen können auch in einem Arbeitsgang durchgeführt werden.
Es ist bekannt, dass das Sauerstoffatom einer Carbonylgruppe gegen eine substituierte Aminogruppe durch Umsetzung von Aminen mit Carbonylverbindungen und gleichzeitiger Reduktion der entstehenden Kondensationsprodukte ersetzbar ist (C. 1928, II, 2461). Es ist ferner bekannt, dass das aus 2-Methylhepten- (2)-on- (6) und Hydroxylamin erhaltene Oxim bei der Reduktion 6-Amino-2-methylhepten- (2) liefert (A. 309 [1899] 25). Diese Reaktion führt aber im vorliegenden Falle zu bisher unbekannten Verbindungen. Es wurde gefunden, dass diese neue Gruppe ungesättigter Amine neue und unerwartete pharmakologische Wirkungen aufweist.
Die nach dem Verfahren darstellbare sekundären Basen sind meist farblose, leicht bewegliche Öle von charakterstischem Geruch, die in guter Ausbeute und in hervorragende Reinheitsgrad erhalten werden. Mit organischen Säuren, wie z. B. Weinsäure, Citronensäure. Camphersäure, Barbitursäure oder deren Derivaten, erhält man aus den Basen prächtig kristallisierende Verbindungen.

Beispiel 2 : 50'4 g 2-Methylhepten- (2) -on- (6), in 150 cm3 Äther gelöst, werden mit einer Lösung von 15 g Methylamin in 250 cm3 Äther versetzt. Unter geringer Erwärmung findet Kondensation statt.
Die ätherische Lösung wird mit Pottasche getrocknet, eingeengt und der Rückstand unter vermindertem Druck destilliert. Das entstandene 6-[N-Dehydromethylamino ]-2-methylhepten- (2) siedet bei 65-670 unter 8 mm Druck ; verdünnte Säuren zerlegen es in die Komponenten.

2-3 Stunden wird filtriert und das Filtrat unter vermindertem Druck eingeengt ; nichtbasisehe Anteile werden mit Äther oder Benzol ausgeschüttelt. Auf Zusatz von Alkali scheidet sieh das 6-Methylamin- 2-methylhepten- (2) ab. Ausbeute 65-70% der Theorie.
Beispiel 4 : 50'4 2-Methylhepten- (2)-on- (6) und 62 g 30%ige Methylaminlösung werden unter
Zusatz von wenig Alkohol in 250 em Äther gelöst und im Verlaufe von 2 Stunden mit 46 g Natrium unter Zugabe von insgesamt 100 cm3 Wasser unter Riihren am Rückflusskühler reduziert. Die Auf-

Beispiel 5: 31#5 g 2-Methylhepten-(2)-on-(6) und 8#5 g Methylamin, in 150 cm3 50%igem Alkohol gelöst, werden mit einer Lösung von 70 g Methylaminehlorhydrat in 200 cm"Wasser vermischt und kathodisch reduziert. Bei Anwendung einer Bleikathode bedarf es einer Stromdiehte von 3 Amp./100 ran und einer Spannung von 7 bis 8 Volt. Als Anodenflüssigkeit dient gesättigte Sodalösung. Ausbeute an 6-Methylamino-2-methylhepten-(2) 30-40% der Theorie.
Beispiel 6 : 37#8 g 2-Metylhepten-(2)-on-(6) in 250 cm3 Äthr werden in Gegenwart von 12 g Methylamin und 100 cm3 1%iger kolloidaler Platinlösung der katalytischen Reduktion unterworfen. Nach der ziemlich langsam verlaufenden Aufnahme der 1 Mol. entsprechenden Menge Wasserstoff wird die Hydrierung abgebrochen und das Reaktionsprodukt in bekannter Weise aufgearbeitet. Ausbeute an 6-Methylamino-2-methylhepten-(2) 60% der Theorie.

Nach beendeter Reduktion wird filtriert und in bekannter Weise aufgearbeitet. Das 6-Äthylamino-2-methylhepten-(2) ist ein farbloses Öl von angenehm, schwach basischem Geruch vom Kp6 = 66-68 . Ausbeute fast quantitativ.

According to the process, these new compounds are produced by condensation of 2-methyl-hepten- (2) -one- (6) with primary alkyl or. Aralkylamines and subsequent reduction of the condensation product obtained. These two reactions can also be carried out in one operation.
It is known that the oxygen atom of a carbonyl group can be replaced by a substituted amino group by reaction of amines with carbonyl compounds and simultaneous reduction of the condensation products formed (C. 1928, II, 2461). It is also known that the oxime obtained from 2-methylhepten- (2) -one- (6) and hydroxylamine gives 6-amino-2-methylhepten- (2) on reduction (A. 309 [1899] 25). In the present case, however, this reaction leads to previously unknown compounds. This new group of unsaturated amines has been found to have new and unexpected pharmacological effects.
The secondary bases which can be prepared by the process are mostly colorless, easily mobile oils with a characteristic odor, which are obtained in good yield and with an excellent degree of purity. With organic acids, such as. B. tartaric acid, citric acid. Camphoric acid, barbituric acid or their derivatives are obtained from the bases that crystallize perfectly.
