Verfahren zur Gewinnung eines Cyelushormons.
Im Harn und in andern Körperflüssigkeiten finden sich zwei nach verschiedenen Methoden gewinnbare Hormone, die beide den östrischen Cyclus an kastrierten Tieren hervorzurufen imstande sind. Das eine Hormon hat die Analysenformel CigHOg und schmilzt bei 268-269 .
Es wurde von G. F. Marrian dargestellt und im Biochemical Journal Bd. 23, S. 1090,1929 beschrieben. Weitere Arbeiten von Marrian über diesen Gegenstand finden sich im Biochemical Journal Bd. 24, S. 435 und 1021. Das andere hat die Formel C18H22O2 und den Schmelzpunkt 240-243 .
Es wurde nun gefunden, dass das Hormon der Formel CigHOg in das Hormon der Formel C18H22O2 übergeführt werden kann, wenn es mit wasserabspaltenden Mitteln behandelt wird. Durch diese Überführung wird die Wirksamkeit des Hormons beträchtlich erhöht.
Die Wasserabspaltung kann bewirkt werden durch Erhitzen mit Kaliumbisulfat, Phosphorpentoxyd usw. oder katalytisch durch Überleiten der Dämpfe über Tonerde, Bleicherde und andere hochporöse Stoffe.
An Stelle des reinen Hormons C18H24O3 kann man als Ausgangsmaterial auch hormonhaltige Öle oder Präparate verwenden.
Beispiel 1 : 0'1 g Cyclushormon der Formel C18H24O3 wird mit Kaliumbisulfat im Hochvakuum auf 1200 erhitzt und langsam in eine Vorlage überdestilliert. Man erhält 0'085 9 des Hormons der Formel C18H22O2 vom Schmelzpunkt 240-243 .
Beispiel 2 : 59 eines cyclushormonhaltigen Öles, das eine Wirksamkeit von 30.000 M. E. pro Gramm hat, werden mit einer hochporösen Bleicherde vermischt und erhitzt. Durch Abdestillieren im Hochvakuum bei etwa 120 geht ein dunkelbraunes Öl über, von welchem man 4 g erhält und das eine Wirksamkeit von 50-60. 000 M. E. pro Gramm hat. Man kann auch das Ausgangsöl mit der hochporöses Bleicherde vermischen und nun durch langsames Anheizen im Hochvakuum gleichzeitig die Wasserabspaltung und das Abdestillieren des Reaktionsproduktes vornehmen.
Beispiel 3 : 12 mg Cyclushormon der Formel C18H24O3 werden mit 100 my geschmolzenem Kaliumbisulfat innig verrieben, in eine Hochvakuumretorte gefüllt und bei 0#02 mm Hg zunächst 2 Stunden auf 110 , dann weitere 5 Stunden auf 180-200 erhitzt. Nach der Temperaturerhöhung auf 1800 sublimiert das entstandene Hormon in den Retortenansatz, aus dem es mit Alkohol herausgelöst wird. Die konzentrierte alkoholische Lösung liefert nach dem Anspritzen mit Wasser 9 mg Cyclushormon der Formel C18H22O2 vom Schmelzpunkt 250-251 (korr. ) und der physiologischen Wirksamkeit von 8 Millionen M. E. pro Gramm bei Injektion in 01lösung und einmaliger Dosis.

Process for obtaining a cyelus hormone.
In the urine and in other body fluids there are two hormones which can be obtained by different methods and which are both capable of producing the estrous cycle in castrated animals. One hormone has the analytical formula CigHOg and melts at 268-269.
It was presented by G. F. Marrian and described in Biochemical Journal Vol. 23, pp. 1090, 1929. Further work by Marrian on this subject can be found in Biochemical Journal Vol. 24, pp. 435 and 1021. The other has the formula C18H22O2 and the melting point 240-243.
It has now been found that the hormone of the formula CigHOg can be converted into the hormone of the formula C18H22O2 if it is treated with water-releasing agents. This transfer increases the effectiveness of the hormone considerably.
The elimination of water can be effected by heating with potassium bisulphate, phosphorus pentoxide etc. or catalytically by passing the vapors over clay, bleaching earth and other highly porous substances.
Instead of the pure hormone C18H24O3, you can also use hormone-containing oils or preparations as the starting material.
Example 1: 0.1 g of cyclus hormone of the formula C18H24O3 is heated to 1200 with potassium bisulfate in a high vacuum and slowly distilled over into a receiver. 0.085 9 of the hormone of the formula C18H22O2 with a melting point of 240-243 is obtained.
Example 2: 59 of an oil containing cyclus hormones, which has an effectiveness of 30,000 M.U. per gram, are mixed with a highly porous fuller's earth and heated. By distilling off in a high vacuum at about 120, a dark brown oil passes over, of which 4 g are obtained and which has an effectiveness of 50-60. Has 000 MU per gram. The starting oil can also be mixed with the highly porous bleaching earth and the water can be split off and the reaction product is distilled off at the same time by slowly heating up in a high vacuum.
Example 3: 12 mg of cyclus hormone of the formula C18H24O3 are thoroughly triturated with 100 .mu.m of molten potassium bisulfate, filled into a high vacuum retort and heated at 0-02 mm Hg for 2 hours to 110, then for a further 5 hours to 180-200. After the temperature has been increased to 1800, the hormone produced sublimes into the retort, from which it is extracted with alcohol. When injected with water, the concentrated alcoholic solution provides 9 mg of cyclus hormone of the formula C18H22O2 with a melting point of 250-251 (corr.) And the physiological effectiveness of 8 million MU per gram when injected in oil solution and a single dose.
