Verfahren zur Gewinnung der wirksamen Bestandteile der Keimdrüsen.
Gemäss vorliegender Erfindung können die wirksamen Bestandteile der Keimdrüsen (Eierstöcke einschliesslich Corpus luteum, Follikel und Plazenta, Hoden) in der Weise gewonnen werden, dass man die betreffenden Organe, zweckmässig nach vorheriger Zerkleinerung und Trocknung, in der üblichen Weise extrahiert, den Extrakt verseift, den unverseifbaren Teil, in dem die wirksamen Stoffe (Hormone) enthalten sind, abtrennt und reinigt.
Die Verseifung des Extrakts geschieht, nach Feststellung der Verseifungszahl, zweckmässig mit alkoholischer Kali-oder Natronlauge. Aus dem Gemisch kann der unverseifbare Teil durch Behandlung mit geeigneten Lösungsmitteln, z. B. mit Äther, abgetrennt werden. Der Äther wird dann abdestilliert.
Zur Reinigung, insbesondere zur Abscheidung von Cholesterin, löst man den Rückstand in Äther, filtriert vom Ungelösten ab, nimmt den Rückstand mit heissem Alkohol auf und bringt, beispielsweise durch Konzentration und starke Abkühlung der Lösung, das Cholesterin zur Ausscheidung. Das Reinigungverfahren kann gegebenenfalls mehrmals wiederholt werden.
Beispiel.
100 Teile Rohazetonextrakt aus getrockneter Plazenta werden mit 16 Teilen Natriumhydroxyd oder 22-4 Teilen Kaliumhydroxyd, gelöst in Alkohol, versetzt und zur Verseilung unter Rühren 20 Minuten gekocht. Der Alkohol wird abdestilliert und die erhaltene Seife in 2000 Teilen Wasser gelöst. Diese Lösung wird dreimal mit je 200 Teilen Äther ausgeschüttelt und die ätherische Lösung abgetrennt. Der Äther wird abdestilliert und das erhaltene Produkt in kochendem Methylalkohol gelöst. Man lässt erkalten und filtriert vom ausgefallenen Cholesterin ab. Der Rückstand nach dem Abdestillieren enthält das Hormon in fast quantitativer Ausbeute.

According to the present invention, the active components of the gonads (ovaries including the corpus luteum, follicles and placenta, testes) can be obtained in such a way that the organs concerned are extracted in the usual way, suitably after previous comminution and drying, and the extract is saponified, the unsaponifiable part, which contains the active substances (hormones), separates and cleans.
The saponification of the extract takes place, after the saponification number has been determined, suitably with alcoholic potash or sodium hydroxide solution. The unsaponifiable part can be removed from the mixture by treatment with suitable solvents, e.g. B. with ether, are separated. The ether is then distilled off.
For purification, in particular to remove cholesterol, the residue is dissolved in ether, the undissolved material is filtered off, the residue is taken up with hot alcohol and the cholesterol is eliminated, for example by concentrating and cooling the solution strongly. The cleaning procedure can be repeated several times if necessary.
Example.
100 parts of crude acetone extract from dried placenta are mixed with 16 parts of sodium hydroxide or 22-4 parts of potassium hydroxide, dissolved in alcohol, and boiled for 20 minutes with stirring for stranding. The alcohol is distilled off and the soap obtained is dissolved in 2000 parts of water. This solution is extracted three times with 200 parts of ether each time and the ethereal solution is separated off. The ether is distilled off and the product obtained is dissolved in boiling methyl alcohol. It is allowed to cool and the precipitated cholesterol is filtered off. The residue after distilling off contains the hormone in almost quantitative yield.
