Abstract

Reivindicación 1: Un proceso de purificación rápido, de alto rendimiento, de polisacáridos capsulares de Neisseria meningitidis serogrupo X, en donde dicho proceso comprende las etapas de: (a) tratar un caldo de fermentación de Neisseria meningitidis serogrupo X mediante agregado de soluciones de sales simples predeterminadas y polvo de sal seca de concentración específica; (b) agregar un volumen específico de solvente orgánico a la solución tratada con sal de la etapa (a); (c) mezclar la solución de la etapa (b) a temperatura ambiente durante una duración específica; (d) llevar a cabo la centrifugación de la solución de la etapa (c) a rpm especificas durante un periodo predeterminado; (e) recolectar el sobrenadante a partir de la solución centrifugada de la etapa (d); (f) someter dicho sobrenadante de la etapa (e) a concentración y diafiltración utilizando una membrana de corte por peso molecular para obtener polisacárido bacteriano parcialmente purificado; (g) precipitar dicho polisacárido bacteriano parcialmente purificado de la etapa (f) con al menos un detergente catiónico bajo condiciones especificas seguido por centrifugación a rpm específicas durante un predeterminado período para obtener un primer pellet; (h) disolver dicho primer pellet en un agente de precipitación inorgánico; (i) agregar a dicho primer pellet disuelto de la etapa (h) al menos un solvente orgánico seguido por incubación en condiciones predeterminadas; (j) someter dicha solución de la etapa (i) a centrifugación en condiciones específicas seguido por recolección de un segundo pellet; (k) disolver dicho segundo pellet en MQW y tratar con reactivos químicos específicos, y solvente orgánico específico, seguido por incubación en condiciones especificas; (l) someter dicha solución incubada de la etapa (k) a centrifugación en condiciones de centrifugación específicas seguido por recolección del sobrenadante; (m) someter dicho sobrenadante de la etapa (l) a filtración por carbón para obtener un filtrado por carbón hasta que se alcance una densidad óptica deseada; (n) filtrar dicho filtrado por carbón de la etapa (m) utilizando un arreglo de filtración PES seguido por concentración y diafiltración en condiciones específicas; (o) filtrar dicho sobrenadante diafiltrado de la etapa (n) utilizando un arreglo de filtración PES específico para obtener polisacárido purificado; (p) almacenar dicho polisacárido purificado de la etapa (o) en condiciones especificas; en donde dicho proceso es económico, escalable y brinda polisacárido purificado de Neisseria meningitidis serogrupo X en un rango de 147 - 323 mg/L de caldo de fermentación.Claim 1: A rapid, high-performance purification process of capsular polysaccharides of Neisseria meningitidis serogroup X, wherein said process comprises the steps of: (a) treating a fermentation broth of Neisseria meningitidis serogroup X by adding salt solutions Simple predetermined and dry salt powder of specific concentration; (b) adding a specific volume of organic solvent to the salt-treated solution of step (a); (c) mixing the solution from step (b) at room temperature for a specific duration; (d) carrying out centrifugation of the solution from step (c) at specified rpm for a predetermined period; (e) collecting the supernatant from the centrifuged solution of step (d); (f) subjecting said supernatant from step (e) to concentration and diafiltration using a molecular weight cutoff membrane to obtain partially purified bacterial polysaccharide; (g) precipitating said partially purified bacterial polysaccharide from step (f) with at least one cationic detergent under specific conditions followed by centrifugation at specific rpm for a predetermined period to obtain a first pellet; (h) dissolving said first pellet in an inorganic precipitation agent; (i) adding to said first dissolved pellet of step (h) at least one organic solvent followed by incubation under predetermined conditions; (j) subjecting said solution from step (i) to centrifugation under specific conditions followed by collecting a second pellet; (k) dissolving said second pellet in MQW and treating with specific chemical reagents, and specific organic solvent, followed by incubation under specific conditions; (l) subjecting said incubated solution from step (k) to centrifugation under specific centrifugation conditions followed by collecting the supernatant; (m) subjecting said supernatant from step (I) to charcoal filtration to obtain a charcoal filtration until a desired optical density is reached; (n) filtering said charcoal filtrate from step (m) using a PES filtration arrangement followed by concentration and diafiltration under specific conditions; (o) filtering said diafiltered supernatant from step (n) using a specific PES filtration arrangement to obtain purified polysaccharide; (p) storing said purified polysaccharide from step (o) under specific conditions; wherein said process is economical, scalable and provides purified polysaccharide from Neisseria meningitidis serogroup X in a range of 147 - 323 mg / L of fermentation broth.
