Abstract

La presente divulgación se refiere a un procedimiento para preparar clones infecciosos de ADNc basados en el genoma de un virus atenuado del síndrome respiratorio y reproductivo porcino (PRRSV) y usos de los mismos en la preparación de vacunas eficaces y con seguridad mejorada contra PRRSV. La presente comprende además un clon infeccioso de ADNc obtenible mediante dicho procedimiento. La presente proporciona además un PRRSV atenuado modificado y composiciones inmunogénicas y vacunas que comprenden ese PRRSV atenuado. Se proporciona además el PRRSV atenuado para uso en la profilaxis y/o el tratamiento de infecciones por PRRSV, y el PRRSV atenuado para uso como vacuna o medicamento en la profilaxis y/o el tratamiento de infecciones por PRRSV. Reivindicación 1: Un procedimiento para generar un clon infeccioso de ADNc basado en el genoma de una cepa de PRRSV atenuada, caracterizado porque comprende: a) identificar zonas polimórficas de la secuencia genómica de una cepa atenuada de PRRSV, b) determinar la secuencia más frecuente dentro de las zonas polimórficas identificadas en la etapa a), y c) construir un clon infeccioso de ADNc que comprende la secuencia más frecuente en al menos una de las zonas polimórficas identificadas en la etapa a).
