Abstract
Se producen agentes de suspension genética que comprenden ARN orientado en el sentido opuesto del marco de lectura, bajo la forma de un rizo de ARN antisentido, en células de organismos transgénicos, por ejemplo, plantas, mediante la transcripcion de una construccion de ADN recombinante que comprende, en el orden de 5' a 3', un elemento promotor unido operativamente a más de un ADN orientado en el sentido opuesto del marco de lectura y uno o más elementos de ADN complementarios. Reivindicacion 1:Una construccion de ADN recombinante para suprimir una pluralidad de genes blanco de zeína de maíz caracterizada porque comprende, en le orden de 5' a 3', un elemento promotor unido operativamente a una pluralidad de ADN orientados en el sentido opuesto del marco de lectura de una pluralidad de genes blanco de zeína de maíz, donde los ADN antisentido se designan, en el ordén de 5' a 3', elemento 1, elemento 2, y una pluralidad de elementos de ADN orientados en el sentido del marco de lectura de una pluralidad de elementos de ADN orientados en el sentido del marco de lectura de una pluralidad de genes blanco de zeína de maíz, designados en el orden de 5' a 3', elemento 3, elemento 4, donde el ADN antisentido 2 tiene al menos una porcion de su secuencia en su extremo 3' que no es complementaria consigo misma o con el elemento de ADN orientado en el sentido del marco de lectura3, y donde el ARN orientado en el sentido del marco de lectura transcripto por elementos de ADN orientados en el sentido del marco de lectura es complementario con el extremo 5' del ARN antisentido transcripto por los ADN antisentido. Reivindicacion 9: Un método para generar semillas de maíz que tiene al menos una característica seleccionada entre lisina mejorada, aceite mejorado y triptofano mejorado, caracterizado porque comprende los pasos de: a) transformar una célula vegetal con la construccion de ADN recombinante de la reivindicacion 1 o la reivindicacion 4 para suprimir más de un gen blanco de zeína de maíz; y b) regenerar la célula vegetal en una planta transgénica fértil, donde la planta contiene la construccion en su genoma; y c) cosechar semillas de la planta, donde las semillas tienen al menos una característica seleccionada entre lisina mejorada, aceite mejorado y triptofano mejorado respecto de las semillas de la misma variedad no transformadas con la construccion. Reivindicacion 11: Una planta transgénica caracterizada porque comprende en su genoma una construccion de ADN de la reivindicacion 1 o la reivindicacion 4 para suprimir más de un gen blanco de zeína de maíz. Reivindicacion 12: Una semilla caracterizada porque ha sido cosechada de una planta de reivindicacion 11. Reivindicacion 14: Un producto procesado de la semilla de la reivindicacion 12, caracterizado porque el producto es un alimento, una racion o una composicion de proteínas parcialmente purificadas.
