Abstract

La presente se refiere a un método para identificar si un compuesto inhibe la entrada de un virus a una célula que comprende: (a) obtener ácido nucleico que codifica una proteína de envoltura viral de un paciente infectado por el virus; (b) co-transfectar dentro de una primera célula (i) el ácido nucleico de la etapa (a), y (ii) un vector de expresión viral que carece del ácido nucleico que codifica una proteína de envoltura, y que comprende un ácido nucleico indicador que produce una senal detectable, de modo que la primera célula produce partículas virales que comprenden la proteína de envoltura codificada por el ácido nucleico obtenido del paciente; (c) poner en contacto las partículas virales producidas en la etapa (b) con una segunda célula en presencia del compuesto, donde la segunda célula expresa un receptor de superficie celular al que se une el virus; (d) medir la magnitud de la senal producida por la segunda célula con el objeto de determinar la infectividad de las partículas virales; y (e) comparar la magnitud de la senal medida en la etapa (d) con la magnitud de la senal producida en ausencia del compuesto, donde una reducción en la magnitud de la senal medida en presencia del compuesto indica que el compuesto inhibe la entrada del virus a la segunda célula. Reivindicación 1: Un método para detectar en un paciente infectado por un virus, el desarrollo de un anticuerpo capaz de bloquear la infección, que comprende: (a) poner en contacto la célula huésped con una preparación de anticuerpo del paciente, donde la célula huésped comprende un ácido nucleico que codifica una proteína viral del paciente y un ácido nucleico que produce una senal detectable; (b) medir la magnitud de la senal detectable producida por la célula huésped; y (c) comparar la magnitud de la senal medida en la etapa (b) con la magnitud de la senal producida en ausencia de la preparación de anticuerpo, donde una reducción en la magnitud de la senal medida en presencia de la preparación de anticuerpo indica que el paciente ha desarrollado una respuesta de anticuerpos a la proteína viral capaz de bloquear la infección.
