Description
本発明は、PD−1抗体およびPD−L1抗体ならびにその使用に関する。
コレセプターシグナル伝達は、免疫応答を調和しそして密接に調節するための重要な機序である。αβT細胞の通常の活性化スキームは、HLAクラスIまたはIIによって提示されるペプチド抗原によってもたらされるポジティブなシグナルに依拠する。コレセプターシグナルは、この活性化を増加させるかまたは妨げるかのいずれかである。
本発明は、CNCM寄託番号I−4122の下で入手できるハイブリドーマから得ることのできるPD−1抗体(PD1.3)に関する。
a)PD1.3またはその誘導体;および
b)癌または慢性感染の処置のためのワクチン
を含む、癌または慢性感染の処置のためのキットに関する。
定義
本発明による「抗体」または「免疫グロブリン」は、同じ意味を有し、そして本発明において同等に使用される。本明細書において使用する「抗体」という用語は、免疫グロブリン分子および免疫グロブリン分子の免疫学的に活性な部分、すなわち、抗原と免疫特異的に結合する抗原結合部位を含む分子を指す。従って、抗体という用語は、完全な抗体分子だけでなく、抗体フラグメントまたは誘導体も包含する。抗体フラグメントは、Fv、Fab、F(ab’)2、Fab’、dsFv、scFv、sc(Fv)2およびダイアボディを含むがそれらに限定されない。
本発明は、CNCM寄託番号I−4122の下で入手できるハイブリドーマから得ることのできる単離されたPD−1抗体(PD1.3)に関する。
本発明の抗体を、任意の化学的、生物学的、遺伝学的または酵素的技術など、これらに限定されない当技術分野において公知の任意の技術を単独でまたは組み合わせてのいずれかによって産生し得る。
(i)抗体の発現を可能とするに適した条件下でCNCM I−4122、CNCM I−4080またはCNCM I−4081として寄託したハイブリドーマを培養すること;および
(ii)発現した抗体を回収することを含む。
本発明者らは、PD1.3が、PD−1へのPD−L1およびPD−L2の結合を阻害し、よってこれを癌患者および慢性感染中に観察されるPD−1によって媒介される免疫抑制機序を克服するために使用し得ることを実証した。
a)PD1.3またはその誘導体;および

を含む、癌または慢性感染の処置のためのキットに関する。
要約
プログラム死1分子(PD−1)は、末梢性トレランスおよび持続ウイルス感染の調節、ならびに免疫系からの腫瘍の回避機序に関与する。PD−L1およびPD−L2という2つのリガンドが記載されており、これらは、組織分布、発現の調節およびPD−1へのその結合に関与する残基において異なっている。本発明者らは、組換えタンパク質およびmAbを使用して、表面プラズモン共鳴および細胞表面結合によって、PD−1とそのリガンドとの相互作用の分子機序をさらに調査した。本発明者らは、PD−L1およびPD−L2の両方が、PD−1への結合に関して交差競合することを実証できた。興味深いことにそして同じように、1つの選択したPD−1 mAbが、PD−L1およびPD−L2の両方の結合に干渉し得る。PD−L1およびPD−L2は、同等な親和性で、しかし会合および解離の特徴のレベルにおいて示される顕著な差異をもってPD−1に結合した。従って、PD−L2ではなくPD−L1は、遅延した相互作用を示し、これはコンフォメーション遷移の現象を連想させる。これらの機序を、PD−1からのPD−L1の解離を遅延させ得るPD−L1mAbのお陰でさらに確認した。この機序は、PD−1相互作用に限定されなかった。なぜならPD−L1は、その第2リガンドのCD80と類似した様式で挙動するからである。
構築物
ヒトPD−1およびCTLA−4 cDNAを、RT−PCRによって、(CD3+CD28)活性化T細胞から表1に示すプライマーを使用して生成し、そしてその後、DNA4ベクター(Yang WC et al., Int. Immnol. 2000)にサブクローニングした。ヒトPD−1およびCTLA−4の細胞外領域(aa1〜152およびaa)を、このプラスミドから表1に示すプライマーを使用して増幅し、そしてCos Fcリンクベクター(SmithKline Beecham Pharmaceuticals, King of Prussia, PA)を使用してヒトIgG1配列のFcフラグメントと共にインフレームにクローニングした。ヒトPD−L1およびPD−L2 cDNAを、RT−PCRによって、脳および肺のヒト全RNA(Clontech Laboratories, Inc)からそれぞれ表1に示すプライマーを使用して生成した。PD−1と同じクローニングプロトコールを使用して、Ig融合タンパク質として完全長PD−Lおよびその細胞外部分を生成した。
PD1、PD−L1、PD−L2およびCTLA−4の細胞外ドメインを、Cos Fcリンクベクターを使用して、ヒトIgG1配列のFcフラグメントとライゲーションすることによって、キメラcDNAを構築した。
ヒトPD1、PD−L1およびPD−L2に対するMAbを同じように産生した。雌BALB/cマウスを、フロイントアジュバントと共に10μgのヒトIg融合タンパク質を用いての腹腔内注入によって免疫化した。免疫化は2週間間隔で3回繰り返し、4回目の免疫化は、10μgのIg融合タンパク質を用いての尾への静脈内注入によってなした。3日後、脾臓細胞を、PEG1500(Roche)を用いてX63Ag8骨髄腫細胞と融合し、そしてHAT選択(Sigma)およびハイブリドーマクローニング因子(OrigenのHCF)を用いてクローニングした。ハイブリドーマ上清を、それぞれヒトPD1、PD−L1またはPD−L2をトランスフェクションしたCOS細胞系の細胞表面を染色することによって、トランスフェクションしていないCOS細胞との反応性の欠如についてスクリーニングした。クローンPD1.3(マウス、IgG2b)およびPD1.6(マウス、IgG1)、PDL1.1、PDL1.2およびPDL1.3(マウス、IgG1)およびPD−L2(マウス、IgG1)を、腹水液の産生によって産生し、プロテインAアフィニティカラムを用いて精製し、そしてFACSおよびBiacore分析ならびに機能的研究のための試薬として選択した。一過性トランスフェクトCOS細胞を、製造業者のプロトコール(ROCHE)に従ってFuGENE6トランスフェクション試薬を用いて得た。CD80mAb 2D10.4は以前に報告されている。
Cos−7細胞系を、2mM L−グルタミン(Invitrogen)を含むDMEM10%FBS中で培養した。一過性にトランスフェクションしたCOS細胞の染色は、基本的な手順に従った。簡潔に言えば、細胞を、最適な希釈度のmAbと共にインキュベーションし、2%FBSおよび0.02%アジ化ナトリウムを含む冷PBS中で洗浄し、そしてFITCとコンジュゲーションしたヤギ抗マウス(GAM)(Beckman Coulter)と共にインキュベーションした。洗浄後、細胞を、FACS CANTOフローサイトメーター(Becton Dickinson)で分析した。
表面プラズモン共鳴測定を、Biacore1000アップグレード装置(Biacore GE Healthcare)で25℃において実施した。全てのBiacore実験においてHBS−EP緩衝液(Biacore GE Healthcare)をランニング緩衝液として用い、そしてセンサーグラムをBiaevaluation4.1ソフトウェアを用いて分析した。
iDCを、改変を加えた以前に確立したプロトコール(Charbonnier et al, Eur J Immunol. 1999;29(8):2567-78)に従って単球から調製した。PBMCを健康な個体志願者から入手し、そしてlymphoprep(商標)Axis−Shield(ABCys)勾配遠心分離による分画によって単離した。単球は、製造業者のプロトコール(Miltenyi Biotec)に従って単球単離キットIIヒトを用いてのネガティブセレクションによってPBMCから得た。単球を、6ウェルプレート中の、10%FBSを含むRPMI 1640(Invitrogen)培地中で、2.5×106細胞/ウェル(Falcon, BD Biosciences)で培養し、そして20ng/mlの組換えヒトインターロイキン4(IL−4)および100ng/mlの組換えヒトGM−CSF(ABCys French society)を用いて5日間分化させた。iDCは、一貫してCD14およびCD83陰性であり、CD1aは92%超、CD11bは96%超、HLADRは80%超、CD80細胞は20%超であった。
CD4+T細胞を、製造業者のプロトコール(Miltenyi Biotec)に従ってCD4+T細胞単離キットIIヒトを用いてのネガティブセレクションによりPBMCから単離した。CD4+T細胞は、通例的に95%超の純度であった。CD4+T細胞(2×105/ウェル)を、種々の濃度のPD−1、PD−L1およびPD−L2に対するmAbを加えて、3枚の96ウェル平底プレート(Falcon; BD Biosciences)中で、10%FBSを補充した200μlのRPMI 1640(Invitrogen)中の2×104iDC/ウェルと共に共培養した。アイソタイプの一致したmAb(B9.3、マウスIgG1およびB9.4、マウスIgG2b)を、陰性対照として使用した。培養を5日間インキュベーションした。
その後、上清を回収し、培養上清中のIFN−γおよびIL−10の産生をELISA検出によってOptEIA(商標)ヒトIFN−γおよびIL10セットを使用して製造業者のプロトコール(BD Biosciences)に従って検出した。検出限界は、4pg/mlであった。
PD−1、PD−L1、PD−L2mAbおよびPD−1 Ig、PD−L1 IgおよびPD−L2 Ig融合タンパク質の特徴付け
本研究を実施するために、本発明者らは、PD−1およびそのリガンドに対して作られたmAbを作製し、そしてそれらをFab、ならびにPD−1、PDL−1およびPDL−2の細胞外ドメインおよびヒトIgG1のFc部分に対応する可溶性融合タンパク質として使用した。本発明者らは、mAbのFabフラグメントおよび融合タンパク質の両方の結合特徴を、トランスフェクションした細胞上で、SPRによって、BiacoreT100およびフローサイトメトリーを使用して調査した(データは示さず)。
本発明者らは次に、PD−L1およびPD−L2が一緒にPD−1に結合し得るかどうか、またはPD−1占有に対して交差競合するかどうかを調査した。
追加の1セットの実験において、本発明者らは、PD−1mAbが、逆向きに、PD−L1およびPD−L2の両方の結合を妨げ得るかどうかを調査した。図2パネルBおよびCに示したように、抗PD1.6ではなく抗PD1.3が、PD−1へのPD−L1(図2Bおよび2Cの上のパネル)およびPD−L2(図2Bおよび2Cの下のパネル)の両方の結合を完全に阻害した。相互的に、PD1.3mAbは用量依存的に、PD−L1チップへのPD−1Fcの結合を遮断した(データは示さず)。FACS分析によって、PD1.3mAbは、PD−L1およびPD−L2発現細胞へのPD−1Fcの結合を阻害することができる(図2C)。
本発明者らは次に、PD−1に対して作られたmAbの機能的能力を調査した。本発明者らは、最適以下の活性化に対する同種異型T細胞の活性化、すなわち未成熟な単球由来樹状細胞の活性化を誘導するその能力を試験した。非阻害性のPD1.6ではなく、PD−1リガンドとの相互作用を阻害するmAbであるPD1.3が、IFNγおよびIL10の産生の増加(図2パネルD)およびT細胞増殖(データは示さず)によって示されるように、CD4 T細胞の活性化を増強することができた。
本発明者らは次に、PD−1へのPD−L1およびPD−L2の結合の機序を調査した。本発明者らは、この相互作用の異なる工程を特定するために、注意深い動態研究を実施した。動的結合アッセイを実施して、PD−1と、PD−L1およびPD−L2融合タンパク質との間の平衡解離定数を決定した。最初に結合データを1:1のラングミュアモデルを使用して分析した。PD−L2/PD−1については、適合は非常に良好であり、そして9.97nMのKDが得られた。両方の組換え分析物は二価であったが、ラングミュアモデルは非常に良好にデータに適合し、このことは各分子の2つの結合部位がおそらく同時に相互作用することを示す。従って、KD値は結合価を示す可能性が最も高い。
本発明者らはまた、PD−1へのPD−L1およびPD−L2の相互作用を妨げる、PD−L1およびPD−L2mabの能力を試験した。抗PD−L2および抗PDL1.3mAbは、それぞれPD−1/PD−L2およびPD−L1相互作用を妨げた(データは示さず)。しかしながら、抗PDL1.1および抗PDL1.2は、PD−L1/PD−1相互作用を阻害しなかった(データは示さず)。PD−1へのPD−L1の結合は、PD−L1のコンフォメーション修飾を誘導し得るので、本発明者らは、PD−1/PD−L1の結合に干渉しない2つのmAbは、リガンド−レセプター相互作用に必須であり得る他のPD−L1領域に影響を及ぼし得ると判断した。従って、本発明者らはまた、PD−1への結合を遮断しない抗PD−L1抗体がこの現象に影響を及ぼし得るかどうかを調査した。そうするために、PD−L1 Igタンパク質を、飽和濃度のPDL1.1およびPDL1.2抗体Fabと共にインキュベーションし、そしてPD−1チップ上に注入した(図4A)。本発明者らは、両方の非遮断性抗PD−L1抗体とのプレインキュベーションが、PD−1チップからのPD−L1 Igタンパク質の解離を明瞭に修飾することを観察した。PD−L1がPDL1.1またはPDL1.2Fabのいずれかに結合している場合にはPD−L1の解離はより遅い。PD−L1へのPDL1.1またはPDL1.2Fabの結合は、PD−L1 PD−1の相互作用の安定性を増加させるようであり、これはおそらくPD−L1のコンフォメーション修飾に影響を及ぼすことによってである。これらのデータは、PD−1へのPD−L1の結合の適合のために本発明者らが選択することを好んだ「コンフォメーション修飾モデル」と一致しているようである。しかしながら、コンフォメーション遷移現象を、Biacoreを使用して観察することができるという事実は、PD−L1状態の修飾が、急速な現象ではないことを示す。本発明者らは次にこれらの考察を考慮して、細胞表面に発現されるネイティブなPD−L1分子上にコンフォメーション修飾を観察し得るかどうかを調査した。
