Abstract

POSTUPAK IZOLACIJE RESTRIKCIONOG FRAGMENTNOG BRISAČA U VIBRIO CHOLERAE I NJEGOVI PROIZVODI, ovaj pronalazak se odnosi na postupke izolovanja mutanata brisanja Vibrio cholerae. U jednom postupku, brisanje je unapred odredjeno razaranjem sa restrikcionim endonukleaza poznate specifičnosti. Brisanja su ubačena u Vibrio cholerae hromozom in vivo rekombinacijom izmedju plazmida koji nosi željeno brisanje, sa susednim flankirajućim sekvenciama i Vibrio cholerae hromozom. U drugom postupku početna in vivo rekombinacija homolognih sekvenci iz rekombinatnog plazmida u hromozomu obezbedjuje selektibilni marker na ovom mestu. Druga in vivo rekombinacija izmedju homolognih flankirajućih sekvenci dovodi do isecanja proficijentnih gena iz hromozoma sa krajnjim proizvodom koji je mutacija brisanja. Pronalazak uključuje izolovanje i karakterizaciju nove Vibrio cholerae vrste koja ima brisanje u ctx genu, kao što je definisano pomoću Acc I, Xba I, Cla I i/ili restrikcionih endonukleaznih mesta i koja dalje ima brisanje u genu koji kodira zonula occludens toksin.
