Abstract

본 발명은 금 나노입자; 상기 금 나노입자의 표면에 개질화된 폴리에틸렌이민; 및 상기 폴리에틸렌이민으로 개질화된 금 나노입자에 코팅된 키토산을 포함하는 핵산 전달용 다중층 나노입자(Multi-Layer Nanoparticles)와 이의 제조방법, 이를 포함하는 핵산 전달용 조성물, 항균용 조성물 및 감염성 질환 예방 또는 치료용 약학 조성물에 관한 것이다. 본 발명에 따르면, 표적 균 세포 내로의 핵산(예를 들어, ASO) 유입이 극대화되며, 고효율로 탑재된 핵산을 균 내로 효과적으로 전달함으로써 표적 핵산 억제 효과를 극대화할 수 있으므로, 항균제 및 유전자 치료제로서 유용하게 사용될 수 있다.

Description

핵산 전달용 다중층 나노입자
본 발명은 핵산 전달을 위한 다중층 나노입자 및 이의 용도에 관한 것으로, 더욱 상세하게는 금 나노입자; 상기 금 나노입자의 표면에 개질화된 폴리에틸렌이민; 및 상기 폴리에틸렌이민으로 개질화된 금 나노입자에 코팅된 키토산을 포함하는 핵산 전달용 다중층 나노입자(Multi-Layer Nanoparticles)와 이의 제조방법, 이를 포함하는 핵산 전달용 조성물, 항균용 조성물 및 감염성 질환 예방 또는 치료용 약학 조성물에 관한 것이다.
유전자 치료 기술은 질환과 관련된 타겟 유전자에 따라 특이적 설계하여 이를 발현, 억제 또는 조절함으로써 희귀 유전질환, 각종 암, 중추신경계 질환, 감염병 등의 치료나 백신으로써 다양하게 적용될 수 있는 획기적인 기술이다. 유전자 치료제는 특정 유전자를 발현하는 바이러스, 플라스미드 DNA, 메신저 RNA(mRNA) 또는 이를 억제하거나 조절하는 안티센스 올리고뉴클레오티드(ASO), 소간섭 RNA(siRNA) 등 다양한 형태를 갖는다(Bulaklak, K., Gersbach, C.A., Nat Commun, 2020, 11, 5820).
유전자 치료는 전달 방법에 따라 생체 내(in vivo) 및 생체 외(ex vivo) 치료 방법으로 분류할 수 있는데, 생체 내 전달의 경우 104억 5,120만 달러, 생체 외 전달의 경우 25억 5,450만 달러에 이를 것으로 보고되었다. 현재까지 식약처에서 승인된 대표적인 유전자 치료제로 백신을 제외하면, 바이러스 벡터 중에는 Spark Therapeutics사의 Luxturna, Avexis/Novartis사의 Zolgensma 등, 그리고 비-바이러스 치료제로는 Alnylam사의 Oxlumo, Givlaari, Onpattro, Novartis사의 Leqvio, Biogen사의 Spinraza, Sarepta Therapeutics사의 Vyondys, Exondys 등이 있다.
바이러스 기반 유전자 치료제의 경우 에볼라바이러스 백신으로 Merck사의 Ervebo®, Johnson & Johnson사의 Zabdeno®/Mvabea®가 있고, COVID-19 백신으로 Johnson & Johnson사의 JNJ-78436735 제품 등이 있다.
유전자 치료제 중 바이러스 기반 전달의 경우 생체 내 전달 효율이 높지만, 체내 면역 부작용, 변이로 인한 발암성 등의 문제점을 갖는다. 반면, 비-바이러스 전달 방법은 상대적으로 안전하지만, 생체 내 빠른 분해, 낮은 조직 및 세포 투과성으로 인해 합성 캐리어를 이용한 전달체 연구가 필수적이다.
기존에 주로 플라스미드 DNA, siRNA 등 비-바이러스 유전자 전달에 사용하는 캐리어의 종류로 고분자(polymer), 지질(lipid), 세포투과성 펩타이드(cell penetrating peptide), 덴드리머(dendrimer) 등 다양하게 개발되었다. 이중 siRNA 치료제로 식약처에 허가된 캐리어는 Onpattro 제품의 지질 기반 나노입자(lipid nanoparticle, LNP)가 있다.
나노입자를 이용한 약물 및 유전체 전달은 기존 제형에 비하여 용해도 증가, 안정성 개선, 투과성 및 생체이용률 개선, 반감기 연장, 조직 표적화 및 부작용을 최소화시켜 치료 효과를 향상시킬 뿐만 아니라, 주변 환경의 수소이온 농도 또는 효소의 변화에 반응하여 치료제를 방출할 수 있게 한다.
LNP를 전달체로 개발되어 온 치료제의 경우 낭포성섬유증 치료에 Translate Bio사의 MRT5005, 흑색종 치료에 BioNTech사의 BNT111(Lipo-MERIT), 고형암 및 lymphoma 치료에 Moderna사의 mRNA-2416이 있고, 백신 치료제로는 독감 백신으로 Moderna사의 VAL-506440, 파라인플루엔자 백신으로 Moderna사의 mRNA-1653, 지카바이러스 백신으로 Moderna사의 mRNA-1325 등이 있다. 또한 최근 COVID-19 백신으로 허가된 BioNTech/Pfizer사의 Comirnaty(BNT162b2), Moderna사의 Spikevax(mRNA-1273) 제품이 있다.
박테리아의 경우 미국식품의약국에서 승인된 항생제 전달 나노입자가 51가지가 있으며, 피부 및 피하 부위 또는 폐에 박테리아에 의한 상처 감염을 금이나 은, 산화철 나노입자, 폴리락테이트-co-글라이클레이트(PLGA), 리포좀을 이용한 항생제 및 전달을 통한 치료하는 연구가 진행되었다.
ASO는 작용 특이성이 높고 인간 유전자 발현에 대한 위험이 적으며 가능한 모든 유전자를 표적으로 하는 쉬운 설계 및 합성의 장점이 있어 박테리아에서 유전자 치료 방법으로 사용된다. 금 나노입자, 세포투과성 펩타이드(CPP), 지질 나노입자, DNA 나노 구조 등을 포함한 다양한 비-바이러스 유전자 벡터를 이용하고 있다. 동물 감염 모델에서 CPP에 접합된 ASO의 상당한 효능이 확인되었으나, CPP는 염증 반응을 활성화하고 가려움증을 유발할 수 있는 부작용이 존재한다.
진균의 경우 아졸, 에키노칸딘, 폴리엔, 알릴아민 및 항대사물질을 항생제로 사용하고 있으며 이를 이용한 항균작용은 효과적이지만, 높은 독성과 내성으로 인해 치료 효능이 제한된다. 피부 백선 치료제로 Minia 대학에서 나노에멀젼 젤 형태와 칸디다증 치료제로 Matinas BioPharma사에서 cochleate 지질-결정 나노입자를 임상 2상까지 진행하였으며, 조갑백선증 치료제로 Celtic Pharma사에서 트랜스퍼좀(transfersome)의 약물 전달을 임상 3상까지 진행하였다.
한편, 금 나노입자의 표면을 아미노페놀로 개질하여 항생제를 모사하는 나노전달체를 제조하고 이를 여러 균주에 처리하여 항균 효과를 보고한 연구 결과가 존재하지만(Le Wang, et al., Nano Lett. 2022 May 11;22(9):3576-3582), 포유 세포 내로 유입될 경우 세포 독성이 높으며, 포유 세포 활성에 영향을 줄 수 있는 부작용이 존재한다. 또한, ASO 전달을 위한 다중층 나노전달체를 개시한 선행문헌(Marcel Janis Beha, et al., Materials Science & Engineering C 126 (2021) 112167)이 존재하지만, PEI 자체의 심각한 독성으로 인해 포유 세포에 대한 세포독성을 나타내는 부작용이 있다.
이러한 기술적 배경 하에서, 본 발명자들은 포유 세포에 대한 세포독성 없이 표적 균에 효과적으로 유전자(특히, ASO)를 전달하여 항균 효과를 극대화시키고, 다중 표적이 가능하며 구조적 안정성을 갖는 유전자 전달체를 개발하고자 예의 노력한 결과, 높은 생체적합성을 갖는 금 나노입자 및 고분자(branched PEI, chitosan)를 포함하는 다중층 나노입자를 개발하였으며, 균의 바이오필름 억제 효과 증가, 핵산 탑재 효율 증가, 안정적 구조를 통한 장기간 보관 가능, 세포 내 전달 효과 증가, 항균 효과 증가, 균주의 항생제 감응성 회복 효과, 감염된 개체에 대한 적은 독성 효과를 나타내는 것을 확인하고, 본 발명을 완성하였다.

발명의 상세한 설명 및 바람직한 구현예
다른 식으로 정의되지 않는 한, 본 명세서에서 사용된 모든 기술적 및 과학적 용어들은 본 발명이 속하는 기술분야에서 숙련된 전문가에 의해서 통상적으로 이해되는 것과 동일한 의미를 갖는다. 일반적으로 본 명세서에서 사용된 명명법은 본 기술분야에서 잘 알려져 있고 통상적으로 사용되는 것이다.
기존에 유전자 전달을 위해 개발된 여러 나노전달체들과는 달리, 본 발명에서는 생체적합성이 높은 금 나노입자와 고분자(PEI, chitosan)를 사용하여 다중층 나노입자를 제조하였으며, 특히 포유 세포의 유입을 최소화하고 이에 따른 독성을 극소화함으로써 표적 균에 선택적으로 작용하는 유전자 전달체 및 유전자 치료제로서 사용할 수 있다.
따라서 본 발명은 일 관점에서, 금 나노입자; 상기 금 나노입자의 표면에 개질화된 폴리에틸렌이민(PEI); 및 상기 폴리에틸렌이민으로 개질화된 금 나노입자에 코팅된 키토산(chitosan)을 포함하는 핵산 전달용 다중층 나노입자(Multi-Layer Nanoparticles)에 관한 것이다.
본 발명에 있어서, 상기 다중층 나노입자는 금 나노입자 및 폴리에틸렌이민이 코어-쉘 형태로 결합되고, 상기 코어-쉘 형태에 전달 대상인 핵산이 결합되어 있으며, 상기 핵산이 결합된 코어-쉘 형태에 키토산이 쉘 형태로 코팅되어 다중층 형태를 이루는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 폴리에틸렌이민은 가지형(branched) 또는 선형(linear) 폴리에틸렌이민인 것을 특징으로 할 수 있으며, 바람직하게는 가지형 폴리에틸렌이민인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 금 나노입자는 1 내지 100nm의 크기를 갖는 것을 특징으로 할 수 있으며, 바람직하게는 1 내지 80nm, 보다 바람직하게는 2 내지 50nm, 더욱 바람직하게는 3 내지 30nm, 가장 바람직하게는 4 내지 10nm의 크기를 갖는 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 금 나노입자는 표면이 작용기로 수식된 것을 특징으로 할 수 있다. 상기 작용기는 N-하이드록시숙신이미드(N-hydroxysuccinimide; NHS), 아민(-NH2), 카르복실(-COOH), 티올(-SH), 하이드록실(-OH), 말레이미드(maleimide), 염화에틸렌(ethylene dichloride; EDC), 아자이드(azide), 알킨(alkyne) 등을 포함할 수 있으나, 이에 한정되는 것은 아니다. 본 발명의 일 실시예에서, 상기 금 나노입자는 NHS ester로 수식되는 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 폴리에틸렌 이민의 분자량은 0.5 내지 100kDa일 수 있으며, 바람직하게는 0.5 내지 50kDa, 보다 바람직하게는, 1 내지 30kDa, 가장 바람직하게는 2 내지 25kDa인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 키토산의 분자량은 1 내지 1000kDa일 수 있으며, 바람직하게는 2 내지 700kDa, 보다 바람직하게는 3 내지 500kDa, 가장 바람직하게는 5 내지 310kDa인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에서, 상기 폴리에틸렌이민의 분자량은 25kDa이고, 상기 키토산의 분자량은 5kDa인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 폴리에틸렌이민과 키토산의 농도 비는 1:0.1 내지 1:20일 수 있으며, 보다 바람직하게는 1:1 내지 1:10, 보다 바람직하게는 1:1 내지 1:5, 가장 바람직하게는 1:2인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에서, 상기 금 나노입자 100pmole/ml를 기준으로, 폴리에틸렌이민의 농도는 1μg/ml이고, 키토산의 농도는 2μg/ml인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 명세서에서, 용어 핵산은 유전자와 상응하는 의미로 상호 교환적으로 사용된다.
본 발명에 있어서, 상기 핵산은 다중층 나노입자에 탑재되어 전달되어 시험관 내 및 생체 내 모두에서 표적 유전자 및 단백질의 발현을 감소시키거나 목적한 단백질의 발현을 유도하는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 핵산은 siRNA, rRNA, RNA, DNA, cDNA, plasmid, 앱타머(aptamer), mRNA, tRNA, lncRNA, piRNA, circRNA, saRNA, 안티센스 올리고뉴클레오타이드(ASO), shRNA, miRNA, 리보자임(ribozyme), PNA 및 DNAzyme으로 구성된 군에서 선택되는 어느 하나 이상인 것을 특징으로 할 수 있으나, 이에 한정되는 것은 아니다.
안티센스 올리고뉴클레오타이드(antisense oligonucleotide, ASO) 기술은 단일가닥의 RNA 또는 DNA 가닥을 통해 mRNA의 중간대사(metabolism)를 변경함으로써 유전자로부터 단백질로의 정보전달을 조절하는 기술이다. 즉, 충분히 상보적, 특이적으로 혼성화하는 염기서열을 선택하여 목표 단백질의 바람직한 발현억제가 이루어지도록 하는 것이다. ASO의 결합은 목표 유전자에 서열 특이적으로 결합하므로 목표 유전자 이외의 다른 유전자 발현에 영향을 주지 않는다. 따라서 ASO 기술은 특정 단백질의 생체 내 역할의 분석에서 유용한 도구일 뿐만 아니라, 특정 질병에 대한 유전자치료법으로 활용 가능성을 가진다(K J Scanlon, et al., FASEBJ. 1995 Oct;9(13):1288-96).
안티센스 DNA는 목표 mRNA와 결합하여 RNA/DNA 이중나선을 형성하며, 이 RNA/DNA 이중나선구조는 생체 내에 존재하는 RNase H(RNase H; 리보핵산분해효소의 한 종류로서 RNA/DNA 혼성 이중나선이 형성되어 있는 mRNA를 특이적으로 분해하는 효소)의 공격을 받아 분해가 유발된다. 안티센스 RNA는 RNA/RNA 이중나선을 형성하며, RNase L의 공격을 받아 목표 mRNA의 분해가 유발된다. RNase L은 이중나선 RNA 가닥 주변의 단일가닥 RNA를 우선적으로 분해하는 리보핵산분해효소이다(S T Crooke, Annu Rev Pharmacol Toxicol. 1992:32:329-76).
본 명세서에서, "안티센스 올리고뉴클레오타이드(antisense oligonucleotide, ASO)"는 수소결합에 의해 표적 핵산 서열과 혼성화(hybridization)할 수 있는 올리고뉴클레오타이드를 포함하는 것으로 해석된다. 안티센스 올리고뉴클레오타이드는 표적 핵산 서열과 혼성화하여 그의 발현을 조절하는 올리고뉴클레오타이드, 올리고뉴클레오타이드 유사체, 올리고뉴클레오타이드 모사체, siRNA, 단일가닥 siRNA(ss siRNA), 작은 헤어핀 RNA(short hairpin RNA, shRNA), 마이크로 RNA 모방체, 리보자임(ribozyme), 외부 가이드 서열(external guide sequence) 올리고뉴클레오타이드 및 기타 올리고뉴클레오타이드를 포함하지만, 이에 제한되지 않으며, 상기 안티센스 올리고뉴클레오타이드는 단일가닥 및 이중가닥 올리고뉴클레오타이드를 포함하는 개념으로 해석된다.
본 발명의 일 실시예에서, 5nm 크기의 금 나노입자의 표면을 가지형 폴리에틸렌이민(branched PEI)으로 개질하고, 균 표적 핵산(Anti-icaA, Anti-mecA 및 Anti-ftsZ ASO)을 탑재한 후, 최종적으로 키토산(chitosan)으로 표면을 개질하여 다중층(multi-layer) 나노입자를 제조하였으며, 상기 핵산이 나노입자에 안정적이고 고효율로 탑재되는 것을 확인하였다.
따라서, 본 발명은 다른 관점에서, (a) 금 나노입자에 폴리에틸렌이민(PEI)을 반응시켜 금 나노입자의 표면을 폴리에틸렌이민으로 개질시키는 단계; (b) 핵산을 탑재하는 단계; 및 (c) 키토산을 코팅시켜 다중층 나노입자를 제조하는 단계를 포함하는 핵산 전달용 다중층 나노입자의 제조방법에 관한 것이다.
본 발명에 있어서, 상기 금 나노입자, 폴리에틸렌이민, 키토산 및 유전자는 상술한 바와 같으며, 중복된 내용은 기재를 생략한다.
본 발명의 일 실시예에서, 상기 (a) 단계는 금 나노입자 100pmole/ml를 기준으로 분자량 25kDa의 폴리에틸렌이민 1μg/ml를 반응시키는 것을 특징으로 할 수 있으며, 상기 (c) 단계는 금 나노입자 100pmole/ml를 기준으로 분자량 5kDa의 키토산 2μg/ml를 코팅시키는 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에서, 상기 제조된 다중층 나노입자는 핵산 전달체로서 표적 균 세포 내로의 핵산 유입을 극대화하고, 균 내로 효과적으로 전달하여 표적 핵산 억제 효과를 극대화할 수 있음을 확인하였다. 또한, 포유 세포 내로의 유입은 최소화하여 이에 대한 독성을 극소화하는 것을 확인하였다. 결과적으로, 균 내로 전달된 핵산에 의한 극대화된 항균 효과를 획득할 수 있으며, 포유세포에 대한 유입 최소화 및 매우 적은 독성 효과를 나타냄을 확인하였다.
따라서, 본 발명은 또 다른 관점에서, 상기 다중층 나노입자를 포함하는 핵산 전달용 조성물에 관한 것이다.
본 명세서에서, 상기 핵산 전달용 조성물은 핵산 전달체와 상응하는 의미로 상호 교환적으로 사용된다.

본 발명에 있어서, 상기 다중층 나노입자에 탑재될 수 있는 핵산의 종류에 따라 표적 균주가 달라질 수 있어 모든 종류의 균에 대한 항균 효과를 나타냄을 예상할 수 있으므로, 상기 나열된 병원성 세균 또는 진균들에 한정되는 것은 아니다.
본 발명의 항균용 조성물은 단일제로도 사용할 수 있으며, 공인된 항균 효과를 가진다고 알려진 조성물을 추가로 포함하여 복합제제로 제조하여 사용할 수 있다.

본 발명에 있어서, 감염성 질환은 병원체에 의해 감염되어 발병하는 질환으로, 세균성 질환, 바이러스성 질환, 진균성 질환, 원생생물성 질환 등을 포함하지만 이에 한정되는 것은 아니다. 바람직하게는, 진균성 질환인 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니다.
본 명세서에서, 용어 “예방”은 본 발명의 조성물의 투여로 병원성 균에 의해 유발되는 질환을 억제시키거나 진행을 지연시키는 모든 행위를 의미하며, “치료”는 병원성 균에 의해 유발되는 질환의 발전의 억제, 증상의 경감 또는 제거를 의미한다.
본 발명에 따른 약학 조성물은 약학적으로 유효한 양의 상기 다중층 나노입자를 단독으로 포함하거나, 하나 이상의 약학적으로 허용되는 담체, 부형제 또는 희석제를 포함할 수 있다. 상기에서 약학적으로 유효한 양이란 대상 질환을 예방, 개선 및 치료하기에 충분한 양을 말한다.
상기 “약학적으로(제약상) 허용되는”이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 위장 장애, 현기증과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 것을 의미한다. 상기 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 또한 상기 약학 조성물은 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제 및 방부제 등을 추가로 포함할 수 있다.
용어 “담체(carrier)”는 세포 또는 조직 내로의 핵산 구조체의 부가를 용이하게 하는 화합물로 정의된다. 예를 들어, 디메틸술폭사이드(DMSO)는 생물체의 세포 또는 조직 내로의 많은 유기 화합물들의 투입을 용이하게 하는 통상 사용되는 담체이다.
용어 “희석제(diluent)”는 대상 화합물의 생물학적 활성 형태를 안정화시킬 뿐만 아니라, 화합물을 용해시키게 되는 물에서 희석되는 화합물로 정의된다. 버퍼 용액에 용해되어 있는 염은 당해 분야에서 희석제로 사용된다. 통상 사용되는 버퍼 용액은 포스페이트 버퍼 식염수이며, 이는 인간 용액의 염 상태를 모방하고 있기 때문이다. 버퍼 염은 낮은 농도에서 용액의 pH를 제어할 수 있기 때문에, 버퍼 희석제가 화합물의 생물학적 활성을 변형하는 일은 드물다.
본 발명에 따른 다중층 나노입자를 포함하는 약학 조성물은, 환자에게 그 자체로서 또는 병용 요법(combination therapy)에서와 같이 다른 활성 성분들과 함께 또는 적당한 담체나 부형제와 함께 혼합된 의약 조성물로서 투여될 수 있다.
상기 약학 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences(19th ed., 1995)에 상세히 기재되어 있다.
본 발명에 따른 약학 조성물은 수용액, 현탁액, 유탁액 등과 같은 주사용 제형 또는 동결건조(lyophilized)된 형태의 제형으로 제제화 할 수 있지만 이에 한정되는 것은 아니다. 더 나아가 당 분야의 적정한 방법으로 또는 Remington's Pharmaceutical Sciences에 개시되어 있는 방법을 이용하여 각 질병에 따라 또는 성분에 따라 바람직하게 제제화 할 수 있다.
본 발명의 용어 "투여"는 어떠한 적절한 방법으로 환자에게 본 발명의 약학 조성물을 도입하는 것을 의미하며, 본 발명의 약학 조성물은 경구투여 또는 비경구로 투여할 수 있고, 예컨대 주입(infusion), 정맥내 투여(intravenous injection), 근육내 투여(intramuscular injection), 피하 투여(subcutaneous injection), 복강내 투여(intraperitoneal injection), 직장내 투여(Intrarectal administration), 국소 투여(topical administration), 비내 투여(intranasal injection) 등으로 투여될 수 있지만, 이에 한정되는 것은 아니다.
본 발명의 약학 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하며, 보통으로 숙련된 의사는 소망하는 치료 또는 예방의 의도된 목적을 달성하기에 유효한 양을 용이하게 결정 및 처방할 수 있다. 더욱 구체적으로, 치료적 유효량은 치료될 객체의 생존을 연장하거나, 질환의 증상을 방지, 경감 또는 완화시키는데 유효한 양을 의미한다. 치료적 유효량의 결정은, 특히, 여기에 제공된 상세한 개시 내용 측면에서, 통상의 기술자의 능력 범위 내에 있다.
본 발명에서, “개체”는 본 발명에 따른 다중층 나노입자를 투여하여 경감, 억제 또는 치료될 수 있는 상태 또는 질환을 앓고 있거나 그러한 위험이 있는 포유동물을 의미하며, 바람직하게는 사람을 의미한다.
본 발명에 따른 약학 조성물은 종래의 치료제와 병용하여 사용하는 용도로 사용될 수 있다. 즉, 본 발명에 따른 다중층 나노입자 및 이를 포함하는 약학 조성물은 기존의 항진균제 등의 치료제와 동시에 투여되거나, 순차적 또는 역순으로 투여될 수 있음을 의미하는 것으로, 통상의 기술자의 범위 내 적절한 유효량의 조합으로 투여될 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예 1: 유전자(ASO)를 탑재한 유전자 전달체의 제작 및 물성 분석
실시예 1-1: 유전자(ASO) 전달체(MLGNP) 제작
5nm 크기의 금 나노입자를 core 소재로 균 표적 유전자(ASO)를 전달하기 위해 여러 겹의 고분자가 개질된 '다중층 코팅 금 나노입자'(multilayer-coated gold nanoparticle, MLGNP)를 제조하였다.
먼저, 크기가 5nm이며 표면이 N-hydroxysuccinimide (NHS) ester(5nm, ~1 NHS group/nm2, polyethylene glycol(PEG) spacer 5,000Da)로 기능화된 금 나노입자(100pmole in 1ml)에 분자량이 25,000Da인 가지형 폴리에틸렌이민(branched PEI) 고분자를 1μg/ml의 농도로 상온에서 1시간 동안 반응시켜 1mM NaCl 수용액에서 수식하고, 100kDa의 amicon filter를 사용하여 원심분리방법(4000g, 10min)을 통해 10mM NaCl 수용액으로 여러 번 정제하였다.
다음으로, branched PEI가 수식된 나노입자에 균 표적 내성 유전자 및 생장에 필수적인 유전자에 대한 ASO(Anti-ftsZ, Anti-mecA 및 Anti-icaA ASO)를 합성하고, 0.1μM, 1μM, 10μM, 50μM, 100μM의 농도 조건으로 준비하여 10mM NaCl 수용액 내에서 상온에서 30분간 탑재한 후, 100kDa의 amicon filter를 사용하여 원심분리방법(4000g, 10min)을 통해 10mM NaCl 수용액으로 여러 번 정제하였다. 본 발명에 사용된 ASO의 서열은 다음과 같다:
마지막으로 키토산(Chitosan; 5kDa)을 0.2μg/ml, 1μg/ml, 2μg/ml, 10μg/ml 등의 농도로 0.01M 아세트산이 포함된 10mM NaCl 수용액 내에서 상온에서 1시간 반응시킴으로써 MLGNP를 제조하고, 100kDa의 amicon filter를 사용하여 원심분리방법(4000g, 10min)을 통해 10mM NaCl 수용액으로 여러 번 정제하였다.
개질에 사용되는 고분자(branched PEI, Chitosan)의 농도와 양을 조절하고, 반응 시간과 온도 등을 조절하여 최적의 물성과 안정성을 가지는 유전자 전달체(MLGNP)를 개발하였다(도 1).
실시예 1-2: 유전자(ASO) 전달체(MLGNP)의 최적화
선정된 최적의 조건으로는 금 나노입자(100pmole in 1ml) 기준으로 분자량이 25,000Da인 가지형 폴리에틸렌이민(branched PEI) 고분자를 1μg/ml의 농도로 수식하고, 유전자(ASO)를 원하는 농도로 탑재한 후, 최종적으로 분자량이 5kDa인 키토산(chitosan)을 2μg/ml의 농도로 개질한 제형이다(도 2).
특히, 최적의 개질 고분자의 종류와 농도 조건을 확인하기 위해, 배양한 S. aureus 균주에 유전자 전달체를 24시간 처리한 후 바이오필름 형성능을 시간에 따라 Microtiter plate assay와 Congo-red assay로 확인하였다. 표면이 PEI로 개질되어 있는 기존 개발된 유전자 전달체(MLGNP(PEI))와 표면이 Chitosan으로 개질되어 있는 본 발명에 따른 신규 유전자 전달체(MLGNP(Chitosan))를 비교한 결과, 본 발명에 따른 유전자 전달체가 억제 효과의 폭이 가장 크며 최대 48시간까지 장기간 억제 효과 또한 가장 큰 것으로 나타났다(도 3A). 또한, congo-red assay로 정성적으로 분석했을 때에도 신규 유전자 전달체가 장기간 동안 가장 좋은 바이오필름 억제 효과가 나타나는 것이 확인되었다(도 3B).
또한, 개질 고분자에 따른 입도 크기와 작용을 비교 분석하여 최고의 효율을 가지는 제형을 분석한 결과, 여러가지 양전하성 고분자 중 분자량이 5kDa의 Chitosan으로 표면을 개질한 경우 가장 좋은 바이오필름 억제 효과를 나타냄이 확인되었다(도 4).
실시예 1-3: 유전자(ASO) 전달체(MLGNP)의 성공적 제조 확인
실시예 1-2에 따라 개발된 유전자 전달체(MLGNP)의 다중층 코팅 과정에 따른 입도 크기를 Dynamic light scattering(DLS)으로 분석한 결과, 50μM의 다중표적 ASO를 탑재했을 때 나노입자의 지름이 53.7nm, 106.0nm, 143nm로 측정되어 순차적으로 나노입자의 지름이 증가함을 확인하였다(도 5A).
다음으로, MLGNP의 다중층 코팅 과정에 따른 표면 전하(제타전위(Zeta potential))를 측정한 결과, 50μM의 다중표적 ASO를 탑재한 경우에 표면 전하가 순차적으로 +4.12mV, -9.37mV, +37mV로 나타난 것으로 확인되어 각 코팅 과정이 성공적으로 이루어짐을 검증하였다(도 5B).
또한, ASO 탑재에 따른 탑재 효율을 분석한 결과, 대조군인 금 나노입자의 탑재 효율이 약 15.2%인 것에 비해, MLGNP의 경우 65.6%로 4.3배 증가한 높은 탑재 효율을 나타내는 것을 확인하였다(도 5C).
특히, 주사전자현미경(SEM)을 통해 입자의 형태를 분석한 결과, DLS 결과와 마찬가지로 입도 크기가 약 140nm 정도로 균일하게 분포함을 확인하여 성공적으로 다중 표적 ASO 나노전달체(MLGNP)가 제조됨을 확인하였다(도 5D).
실시예 1-4: 유전자(ASO) 전달체(MLGNP)의 안정성 확인
유전자(ASO)를 탑재한 MLGNP 나노입자의 입자 안정성을 확인하기 위해, 입자 합성 조건인 0.01M 아세트산 용액과 PBS 용액에서 각각 7일 동안 4℃에서 입자를 보관하였다.
0.01M 아세트산 용액에서 7일간 보관한 경우, 합성 후 1일 째에는 입자의 크기가 소폭 증가하였지만, 이후 7일까지의 나노입자의 크기는 ASO를 10μM 탑재하였을 때에는 약 110nm, 100μM 탑재하였을 때에는 약 142nm로 오차범위 내에서 유지되는 것이 확인되어, 안정적인 것으로 나타났다(도 6A).
PBS 용액에서 제조된 유전자 전달체(MLGNP) 입자를 7일간 보관한 경우, 제조 직후부터 1일째까지는 입자의 크기가 어느 정도 유지되었으나, 2일째부터는 입자의 크기가 ASO를 10μM 탑재한 경우는 58.2nm, 100μM 탑재한 경우는 80.9nm로 작아진 채 유지되는 것을 확인하였다(도 6B).
따라서, 0.01M 아세트산 용액에 보관할 경우, 제조된 유전자 전달체(MLGNP)를 장기간 보관 가능하다는 결론을 얻을 수 있었다.
실시예 2: 균 표적 유전자 전달체(MLGNP)의 균 내 유입 효율 분석
실시예 1에서 개발된 유전자 전달체(MLGNP)의 유입 효율을 분석하기 위해 대표적인 균종들을 선정하여 유입 정도를 비교 분석하였다.
유전자 전달체(MLGNP)에 탑재될 ASO의 표적 유전자로는, 박테리아의 증식과 생장에 직접적인 역할을 하는 ftsZ, 항생제 내성 바이오마커인 mecA, 박테리아 바이오필름 EPS(세포 외 고분자 물질)를 형성하는 데 주요한 유전자인 icaA를 사용하였다(Anti-ftsZ ASO(서열번호 1), Anti-mecA ASO(서열번호 2) 및 Anti-icaA ASO(서열번호 3)).
배양한 균주에 5' 말단에 FAM 형광 표지가 되어있는 ASO가 탑재되고 분자량이 5kDa인 Chitosan으로 표면이 개질된 MLGNP를 4시간 처리하여 칸디다 종(Candida albicans, Candida glabrata, Candida tropicalis, Candida krusei) 및 여러 균종(Methicillin-sensitive Staphylococcus aureus(MSSA), Methicillin-resistant Staphylococcus aureus(MRSA), Staphylococcus epidermidis, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa)으로의 세포 유입 효율을 공초점 현미경을 통해 분석한 결과, 대조군에 비해 상당히 높은 형광 신호를 관찰함으로써 높은 효율로 균 내로 유입됨을 확인하였다(도 7).
다음으로, 개발된 유전자 전달체(MLGNP)의 효능을 비교 평가하기 위해, 유전자 전달체로 보편적으로 많이 사용되고 있는 여러 종류의 나노전달체를 배양한 균주에 처리하여 세포 유입 효율을 비교하였다.
비교를 위한 나노전달체로는 은 나노입자를 core로 한 나노전달체(MLSNP), 지질기반 나노전달체(Liposome, MC3 Lipid nanoparticle) 그리고 gold core 없이 polymer로만 이루어진 복합체(CS+PEI NP) 등을 개발한 유전자 전달체(MLGNP)와 비교 평가하였다.
배양한 칸디다 균주(C. albicans)에 5'FAM으로 형광 표지된 ASO를 탑재한 각 나노전달체를 2시간 처리한 결과, 다른 나노전달체에 비해 MLGNP의 경우가 가장 높은 효율로 세포에 유입됨을 확인하였다(도 8).
은 나노입자가 core인 MLSNP의 경우 세포 내 유입은 거의 나타나지 않았고, 지질기반 나노전달체인 Liposome 및 MC3 lipid nanoparticle의 경우 부분적으로 세포 내 유입이 나타난 경우가 있으나, MLGNP에 비하여 유의미한 세포 유입을 나타내지는 않았다. 또한, 금 나노입자 core 없이 polymer로만 이루어진 복합체(CS+PEI NP)를 처리한 경우에는 유전자(ASO)를 단독으로 처리한 대조군과 비슷하게 거의 세포 내 유입이 이루어지지 않은 것으로 보아, 세포 유입에 MLGNP의 금 나노입자 core와 사용된 polymer 조합이 중요한 역할을 하는 것을 확인할 수 있다.
실시예 3: 균 표적 유전자 전달체(MLGNP)의 균 내 전달 효과 및 항균 효과 분석
실시예 3-1: 표적 유전자 억제 효과 분석
유전자 전달체(MLGNP)에 의한 표적 유전자 억제 효과를 관측하기 위해, MRSA(Methicillin-resistant Staphylococcus aureus) 균주를 배양하고 실시예 1에 따라 제조한 유전자 전달체(MLGNP)를 균주에 처리하여 표적 유전자의 억제 효율을 RT-qPCR로 분석하였다.
유전자 전달체(MLGNP)에 탑재된 ASO의 표적 유전자로는, 박테리아의 증식과 생장에 직접적인 역할을 하는 ftsZ, 항생제 내성 바이오마커인 mecA, 그리고 박테리아 바이오필름 EPS(세포 외 고분자 물질)를 형성하는 데 주요한 유전자인 icaA를 사용하였다(Anti-ftsZ ASO(서열번호 1), Anti-mecA ASO(서열번호 2) 및 Anti-icaA ASO(서열번호 3)).
박테리아 증식과 생장에 직접적인 역할을 하는 ftsZ를 표적으로 하는 ASO를 탑재한 유전자 전달체(MLGNP)를 MRSA 균주에 처리한 경우, 균주의 ftsZ 발현량이 약 31% 감소하며 억제되었다(도 9A). 항생제 내성 바이오마커인 mecA를 표적으로 하는 ASO를 탑재한 유전자 전달체(MLGNP)를 MRSA 균주에 처리하여 mecA의 발현량을 확인했을 때, mecA 발현량이 약 60% 감소하며 억제됨을 확인하였다(도 9B). 또한, 박테리아 바이오필름 EPS를 형성하는 데 주요한 유전자인 icaA를 표적으로 하는 ASO를 탑재한 유전자 전달체(MLGNP)를 균주에 처리한 경우에는, MRSA 균주의 icaA 발현량이 약 51% 감소하며 억제됨을 확인하였다(도 9C).
결과적으로, 유전자 전달체(MLGNP)가 균 내로 효과적으로 전달되어 작용하는 것을 확인하였다.
실시예 3-2: 항생제 감응성 회복 평가
유전자 전달체(MLGNP)에 의한 항생제 감응성 회복을 평가하기 위해, MRSA의 메티실린 내성과 관련된 대표적인 유전자인 mecA를 표적으로 하는 ASO를 유전자 전달체(MLGNP)에 탑재하였다.
배양한 MRSA 균주에 제조한 유전자 전달체(MLGNP)를 처리하여 항생제 감응성 회복 정도를 WST-8 분석법을 통해 확인하였다. WST-8 분석법은 색도 분석법으로, WST-8 시약이 살아있는 세포의 탈수소효소(Dehydrogenase)와 반응하여 오렌지색의 포르마잔(Formazan)을 형성하여 450nm에서 흡광도를 나타내기 때문에, 시료 내에 살아있는 세포를 정량할 수 있는 분석법으로 널리 사용되고 있다.
배양한 MRSA 균주를 메타실린계 항생제인 옥사실린이 포함된 배지에서 배양했을 때, 항생제 감응성의 회복 정도를 WST-8 분석으로 평가하였다(도 10).
옥사실린이 0μg/ml 포함된 배지의 경우, 모든 실험군에서 항균 효과가 나타나지 않았고, MRSA의 최소 억제 농도(Minimal inhibition concentration)인 512μg/ml가 포함된 배지의 경우, 모든 실험군에서 항균 효과가 나타났다. 최소 억제 농도의 1/16인 32μg/ml의 옥사실린이 포함된 배지에서 유전자 전달체(MLGNP)가 처리된 균주를 배양한 경우, 대조군에 비해 유전자 전달체(MLGNP)가 처리된 균주의 항생제 감응성이 약 43.7% 회복되었다. 또한, 최소 억제 농도의 1/8인 64μg/ml의 옥사실린이 포함된 배지의 경우, 대조군에 비해 유전자 전달체가 처리된 균주의 항생제 감응성이 28.6% 회복되는 결과가 나타났다.
결과적으로, 유전자 전달체(MLGNP)가 메티실린 내성 균주에 효과적으로 작용해 균주의 항생제 감응성이 회복됨을 확인할 수 있다.
실시예 3-3: 바이오필름 억제 효과 분석
유전자 전달체(MLGNP)에 의한 바이오필름 억제 효과를 확인하기 위한 방법으로, Microtiter plate assay와 Confocal 현미경 관측 등의 분석법을 이용하였다. Microtiter plate assay의 경우, 바이오필름 시료를 crystal violet 염색 후 570nm의 흡광도를 측정해 시료 내 바이오필름의 전체 바이오매스를 정량할 수 있는 방법이다. 유전자 전달체(MLGNP)에 탑재된 ASO의 표적 유전자로는 박테리아 바이오필름 EPS(세포 외 고분자물질)을 형성하는 데 주요한 역할을 하는 icaA를 사용하였다.
배양한 MRSA 균주에 유전자 전달체(MLGNP)를 처리하고, Crystal violet 염색 후 시료 내의 전체 바이오매스를 확인한 결과, icaA가 탑재된 유전자 전달체(MLGNP)의 경우에서 최대 80.5% 감소하였다. 특히, 대조군에 비해 매우 유의미하게 바이오매스가 감소함을 확인했으며, 억제 효과가 최대 72시간까지 지속됨을 확인하였다(도 11A).
또한, 배양한 균주에 유전자 전달체(MLGNP)를 여러 농도로 처리하여 바이오필름의 억제 효과를 확인한 결과, 전달체를 10nM, 25nM, 그리고 50nM 처리했을 때, 최대 바이오매스 감소 효과가 각각 약 45.4%, 75.4%, 그리고 86.2%로 관측되어 고농도로 처리할수록 바이오매스 감소 효과가 큰 것을 확인하였다(도 11B).
다음으로, 시료 내의 세포와 세포 외 고분자 물질(EPS)의 감소 정도를 정성적으로 확인하기 위해 Confocal 현미경을 이용해 관측하였다.
배양한 MRSA 균주에 anti-icaA ASO가 탑재된 유전자 전달체(MLGNP)를 처리하고, 시료 내의 세포 DNA에 결합하는 DAPI(4′,6-diamidino-2-phenylindole)와 세포 외 고분자 물질(EPS) 중 다당류에 결합하는 Sypro Ruby 형광을 시료에 염색하여 시료 내의 세포와 세포 외 고분자 물질(EPS)의 감소 정도를 육안으로 관찰하였다.
각 대조군을 처리한 균주에서의 세포와 세포 외 고분자 물질(EPS)은 각각 강한 형광 신호를 보이는 데에 비해, anti-icaA ASO가 탑재된 유전자 전달체(MLGNP)를 처리한 실험군에서는 세포와 세포 외 고분자물질(EPS)의 형광 신호가 유의미하게 줄어드는 결과를 보였다(도 12).
따라서, 개발한 유전자 전달체(MLGNP)가 효과적으로 균 내 유입되어 MRSA 바이오필름을 억제하는 것이 정량적, 그리고 정성적 결과로 확인되었다.
실시예 4: 유전자 전달체(MLGNP)의 포유 세포 독성 평가
신규 개발된 유전자 전달체(MLGNP)를 생체 내에 투여하였을 때 나타날 수 있는 독성을 예측하기 위해, 포유 세포주로 NIH-3T3에 MLGNP를 처리하여 cell counting kit-8(CCK-8) assay를 통해 세포 생존율을 측정하고, 포유 세포 내 유입 정도를 확인하였다.
먼저, 유전자(ASO)가 탑재된 전달체(MLGNP)를 배양한 포유 세포주(NIH-3T3)에 처리하여 세포 생존율을 측정한 결과, 최대 72시간까지 최저 95.2%의 세포 생존율을 보였으며 이를 통해 포유세포에 대한 MLGNP의 독성이 매우 적은 것을 확인하였다(도 13A).
다음으로, ASO가 표적하는 항생제 내성 유전자가 포유 세포의 유전체상에 homology가 있는 경우 균뿐만 아니라 포유 세포의 유전자 발현을 억제하는 off-target 효과가 나타날 수 있으므로, 개발된 유전자 전달체(MLGNP)의 포유 세포 유입 정도를 공초점 현미경(confocal microscopy)을 통해 확인하였다.
5'말단에 FAM 형광 표지가 되어있는 유전자(ASO)를 MLGNP에 탑재하고, 이를 포유 세포주(NIH-3T3)에 처리하여 세포 유입 정도를 공초점 현미경을 통해 분석한 결과, 6시간 이후에도 유의미한 세포 유입이 나타나지 않는 것을 확인하였다(도 13B).
이에 따라, 개발된 유전자 전달체(MLGNP)는 포유 세포에 대한 독성이 매우 적고, 포유 세포에 유의미하게 유입되지 않는 것으로 보아 off-target 효과가 매우 적은 것이 검증되었다.
본 발명은 금 나노입자를 코어(core)로 고분자 물질(PEI 및 chitosan)을 개질하며 이들의 함량과 비율 등을 조절하여 제작한 다중층 나노입자에 관한 것으로, 표적 균 세포 내로의 유전자(예를 들어, ASO) 유입이 극대화되며, 고효율로 탑재된 유전자를 균 내로 효과적으로 전달함으로써 표적 유전자 억제 효과를 극대화할 수 있으므로, 항균제 및 유전자 치료제로서 유용하게 사용될 수 있다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.

  1. 금 나노입자; 상기 금 나노입자의 표면에 개질화된 폴리에틸렌이민(PEI); 및 상기 폴리에틸렌이민으로 개질화된 금 나노입자에 코팅된 키토산(chitosan)을 포함하는 핵산 전달용 다중층 나노입자(Multi-Layer Nanoparticles).
  2. 제1항에 있어서, 상기 금 나노입자 및 폴리에틸렌이민이 코어-쉘 형태로 결합되고, 상기 코어-쉘 형태에 전달 대상인 핵산이 결합되어 있으며, 상기 핵산이 결합된 코어-쉘 형태에 키토산이 쉘 형태로 코팅되어 다중층 형태를 이루는 것을 특징으로 하는 다중층 나노입자.
  3. 제1항에 있어서, 상기 폴리에틸렌이민은 가지형(branched) 또는 선형(linear) 폴리에틸렌이민인 것을 특징으로 하는 다중층 나노입자.
