When AAV-DJ-Postn-GFP was directly injected into the hearts of healthy, non-infarcted mice, low levels of GFP expression were detected only at the injection site. Furthermore, when AAV-DJ-Postn-Cre was systemically administered via the tail vein to healthy control mice ( Figures 16 and 17) and infarcted Tomato reporter mice ( Figures 18 and 19), Tomato expression was observed only in the infarcted area, but not in healthy myocardium or other tissues. These results suggest that the AAV-DJ-Postn vector preferentially targets activated CFs after myocardial injury, but not quiescent CFs or other organs.

Description
本発明は、心筋ダイレクトリプログラミングのための組換えアデノ随伴ウイルスベクターに関する。
心血管疾患は世界的な死亡原因の第1位である。心血管疾患の1種である心筋梗塞では、発症後に心臓線維芽細胞が活性化されて増殖し、心筋の線維化と心機能障害を引き起こすことが知られている。しかし、成人の心筋細胞は終末分化した細胞であり、ほとんど再生能力を有さないため、心臓疾患の治療方法は限定されている。
これまでに、心筋リプログラミング因子(GATA4、MEF2C、及びTBX5等)を用いて、心臓線維芽細胞から心筋細胞を誘導する方法が見出されている(特許文献1、非特許文献1~3)。レトロウイルスベクター、センダイウイルスベクター等を用いたin vivo心臓リプログラミングは、心臓線維芽細胞を誘導心筋細胞にリプログラムし、梗塞したマウスの心臓を修復できることが開示されている(非特許文献2)。また、心臓リプログラミングは、マウスの慢性心筋梗塞モデルにおいて、心筋収縮を改善し、再生効果と抗線維化効果によって線維化を抑制することが開示されている(非特許文献3)。

上記文献に開示されている方法においては、レトロウイルスベクター、センダイウイルスベクター等が用いられているが、それぞれ挿入突然変異、免疫反応等を引き起こす可能性があった。そのため、安全で効率的な遺伝子導入システムの開発が求められている。

本発明によれば、生体内での心筋ダイレクトリプログラミングを行うことのできる、新規ベクターを提供することができる。

本発明に係る「ウイルスゲノム」及び「ポリヌクレオチド」は各々、DNAの形態(例えば、cDNA又はゲノムDNA)で存在し得るが、場合によってはRNA(例えば、mRNA)の形態であってもよい。本明細書において使用されるウイルスゲノム及びポリヌクレオチドは各々、二本鎖又は一本鎖のDNAであり得る。一本鎖DNA又はRNAの場合、コード鎖(センス鎖としても知られる)であっても、非コード鎖(アンチセンス鎖としても知られる)であってもよい。特に述べられない限り、本明細書中、rAAVゲノムがコードするプロモーター、目的遺伝子、ポリアデニレーションシグナル等の遺伝子上の配置について説明される場合、rAAVゲノムがセンス鎖である場合についてはその鎖自体について、アンチセンス鎖である場合はその相補鎖について記載される。
本明細書において、「タンパク質」は、「ポリペプチド」とも換言できる。
本明細書中において使用される場合、「パッケージング」とは、1本鎖ウイルスゲノムの調製、カプシドの組み立て、及びウイルスゲノムをカプシドで包むこと等を含む事象をいう。適切なプラスミドベクター(通常、複数のプラスミド)が適切な条件下でパッケージング可能な細胞株に導入される場合、組換えウイルス粒子(すなわち、ウイルスビリオン、ウイルスベクター)が組み立てられ、培養物中に分泌される。
本明細書において、「作動可能に連結する」とは、転写、翻訳等のような所望の機能を与える核酸間の機能的連結を指す。例えば、プロモーター、シグナル配列等の核酸発現制御配列と目的のタンパク質をコードするポリヌクレオチド配列との間の機能的連結を含む。発現制御配列は目的のタンパク質をコードするポリヌクレオチド配列の転写及び/又は翻訳に影響する。
本明細書において、「活性化された心臓線維芽細胞」とは、休止状態でない心臓線維芽細胞を指す。心臓線維芽細胞は、心筋梗塞、心臓の損傷、慢性的な高血圧等のストレスにさらされると、遊走活性が増加し、増殖するようになるとともに、筋線維芽細胞へ転換される。筋線維芽細胞は、運動性の増強、平滑筋アクチンの発現、コラーゲンその他ECM成分の合成の増加、等を特徴とする。本明細書においては、このような休止状態でない心臓線維芽細胞をまとめて、「活性化された心臓線維芽細胞」と呼ぶ。
1.組換えアデノ随伴ウイルスベクター
本発明は、組換えアデノ随伴ウイルスベクター(rAAVベクター)を提供する。本発明のrAAVベクターは、ペリオスチンプロモーター配列、コラーゲン1a2プロモーター配列、軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列からなる群より選択される少なくとも1種を含むポリヌクレオチドと、AAV-DJカプシドタンパク質とを含む。本発明のrAAVベクターを遺伝子発現に用いることにより、活性化された心臓線維芽細胞(例えば、心筋梗塞後の心臓線維芽細胞等)に選択的に、目的のタンパク質を発現させることができる。
本発明は、組換えアデノ随伴ウイルスベクター(rAAVベクター)を提供する。本発明のrAAVベクターは、ペリオスチンプロモーター配列、コラーゲン1a2プロモーター配列、軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列からなる群より選択される少なくとも1種を含むポリヌクレオチドと、AAV-DJカプシドタンパク質とを含む。本発明のrAAVベクターを遺伝子発現に用いることにより、活性化された心臓線維芽細胞(例えば、心筋梗塞後の心臓線維芽細胞等)に選択的に、目的のタンパク質を発現させることができる。
(アデノ随伴ウイルス)
天然のアデノ随伴ウイルス(AAV)は、非病原性であり、免疫原性が低い。野生型AAVでは、カプシドにパッケージングされた1本鎖DNAのウイルスゲノム上に、両端の逆方向末端反復(ITR)配列に挟まれたrep遺伝子及びcap遺伝子が存在する。rep遺伝子から産生されるRepタンパク質(rep78、rep68、rep52及びrep40)は、ウイルスゲノムの複製、転写調節、染色体への組み込みに必要とされる。cap遺伝子は3つのカプシドタンパク質(VP1、VP2及びVP3)をコードする。

(カプシドタンパク質)
本発明のrAAVベクターは、AAV-DJカプシドタンパク質を含む。AAV-DJはDNAファミリーシャッフリングによって8種類の異なる天然型セロタイプ由来のハイブリッドカプシドを形成している(Grimm D.,et al.,J. Virol., 82(12), 5887~5911(2008).)。一実施形態において、本発明のrAAVベクターが含むAAV-DJカプシドタンパク質は、配列番号1に示すアミノ酸配列と少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、又は100%の同一性を有するアミノ酸配列を有し、かつ、カプシドを形成可能なタンパク質が含まれる。また、例えば、配列番号1のアミノ酸配列において、例えば、1~70個、1~60個、1~50個、1~40個、1~35個、1~30個、1~25個、1~20個、1~15個、1~14個、1~13個、1~12個、1~11個、1~10個、1~9個(1~数個)、1~8個、1~7個、1~6個、1~5個、1~4個、1~3個、1~2個又は1個のアミノ酸残基が欠失、置換、挿入及び/ 又は付加されたアミノ酸配列を含み、かつカプシドを形成可能であるタンパク質が挙げられる。これら欠失、置換、挿入及び付加のうち2種以上の組合わせを同時に含んでもよい。一実施形態において、本発明に用いるカプシドタンパク質は、カプシドを形成する機能を有する。また、当該カプシド内には、目的の遺伝子(例えばリプログラミング因子をコードする配列)等を含むポリヌクレオチドがパッケージングされる。

Virol., 82(12), 5887~5911(2008).)。一実施形態において、本発明のrAAVベクターが含むAAV-DJカプシドタンパク質は、配列番号1に示すアミノ酸配列と少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、又は100%の同一性を有するアミノ酸配列を有し、かつ、カプシドを形成可能なタンパク質が含まれる。また、例えば、配列番号1のアミノ酸配列において、例えば、1~70個、1~60個、1~50個、1~40個、1~35個、1~30個、1~25個、1~20個、1~15個、1~14個、1~13個、1~12個、1~11個、1~10個、1~9個(1~数個)、1~8個、1~7個、1~6個、1~5個、1~4個、1~3個、1~2個又は1個のアミノ酸残基が欠失、置換、挿入及び/ 又は付加されたアミノ酸配列を含み、かつカプシドを形成可能であるタンパク質が挙げられる。これら欠失、置換、挿入及び付加のうち2種以上の組合わせを同時に含んでもよい。一実施形態において、本発明に用いるカプシドタンパク質は、カプシドを形成する機能を有する。また、当該カプシド内には、目的の遺伝子(例えばリプログラミング因子をコードする配列)等を含むポリヌクレオチドがパッケージングされる。
本発明に用いられるAAV-DJカプシドタンパク質をコードするポリヌクレオチドは、本発明のrAAVベクターを形成可能であるカプシドタンパク質と機能的に同等なタンパク質をコードする。このようなポリヌクレオチドは、例えば以下が挙げられる。
(i)配列番号2で表される塩基配列を含むポリヌクレオチド。
(ii)配列番号2で表される塩基配列において1又は数個の塩基が置換、付加、又は欠失した塩基配列を含むポリヌクレオチド。
(iii)配列番号2で表される塩基配列と90%以上の同一性を有する塩基配列を含むポリヌクレオチド。
(iv)配列番号2で表される塩基配列に相補的な塩基配列とストリンジェントな条件下でハイブリダイズする塩基配列を含むポリヌクレオチド。
(i)配列番号2で表される塩基配列を含むポリヌクレオチド。
(ii)配列番号2で表される塩基配列において1又は数個の塩基が置換、付加、又は欠失した塩基配列を含むポリヌクレオチド。
(iii)配列番号2で表される塩基配列と90%以上の同一性を有する塩基配列を含むポリヌクレオチド。
(iv)配列番号2で表される塩基配列に相補的な塩基配列とストリンジェントな条件下でハイブリダイズする塩基配列を含むポリヌクレオチド。
上記(i)において、配列番号2で表される塩基配列を含むポリヌクレオチドであって、本発明のrAAVベクターにおけるカプシドを形成可能なものが包含される。
上記(ii)において、置換、欠失、付加又は挿入する1又は数個の塩基の数は、1以上の整数であれば特に限定されない。例えば、1~数十個程度、好ましくは1~20個程度、より好ましくは1~15個程度、さらに好ましくは1~10個程度、特に好ましくは1~5個程度とすることができる。配列番号2で表される塩基配列において1又は数個の塩基が置換、付加、又は欠失した塩基配列を含むポリヌクレオチドであって、本発明のrAAVベクターにおけるカプシドを形成可能なものが包含される。
ある態様において、置換、付加又は欠失する1又は複数の塩基の数は、配列番号2の塩基配列との塩基配列の同一性が90%以上となるように1~220個程度、好ましくは同一性が95%以上となるように1~110個程度、より好ましくは同一性が97%以上となるように1~66個程度、さらに好ましくは同一性が98%以上となるように1~44個程度、特に好ましくは同一性が99%以上となるように1~22個程度とすることができる。
上記(iii)において、塩基配列の同一性は、90%以上、好ましくは95%以上、より好ましくは97%以上、さらに好ましくは98%以上、特に好ましくは99%以上とすることができる。塩基配列の相同性若しくは同一性は、100%未満とすることができる。塩基配列間の相同性及び同一性は、BLAST等の公知のアルゴリズムを用いて決定することができる。配列番号2で表される塩基配列と90%以上の同一性を有する塩基配列を含むポリヌクレオチドであって、本発明のrAAVベクターにおけるカプシドを形成可能なものが包含される。
上記(iv)において、「ストリンジェントな条件」とは、特異的なハイブリダイゼーションのみが生じ、非特異的なハイブリダイゼーションが生じないような条件を言う。このような条件は、「1×SSC(0.9M NaCl,0.09M クエン酸三ナトリウム)又は6×SSPE(3M NaCl,0.2M NaH2PO4, 20mM EDTA・2Na,pH7.4)中の42℃でハイブリダイズさせ、さらに42℃で0.5×SSCにより洗浄する」条件が、ストリンジェントな条件の1例として挙げられるが、これに限定されるものではない。このような条件は、例えば、M. R. Greenら,Molecular Cloning: A Laboratory Manual,Fourth Edition, Cold Spring Harbor Laboratory Press(2012)に記載されている。配列番号2で表される塩基配列に相補的な塩基配列とストリンジェントな条件下でハイブリダイズする塩基配列を含むポリヌクレオチドであって、本発明のrAAVベクターにおけるカプシドを形成可能なものが包含される。
(ポリヌクレオチド(rAAVゲノム))
本発明のrAAVベクター内にパッケージングされるポリヌクレオチド(rAAVゲノム)は、目的のタンパク質(例えば、リプログラミング因子等)を発現させるため、そのタンパク質をコードするポリヌクレオチド配列がプロモーター配列と作動可能に組み合わされる。本発明において、プロモーター配列としては、ペリオスチンプロモーター配列、コラーゲン1a2プロモーター配列、軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列からなる群より選択される少なくとも1種が用いられる。
ペリオスチンは、活性化された心臓線維芽細胞においてのみ発現する細胞外マトリックスタンパク質である。心臓線維芽細胞は、心筋梗塞、圧負荷等のストレスを受けた心筋組織において活性化することが知られている。ペリオスチンプロモーター配列を用いることにより、活性化された心臓線維芽細胞(例えば、心筋梗塞後の心臓線維芽細胞等)に選択的に、目的のタンパク質を発現させることができる。ペリオスチンプロモーター配列を用いることにより、心筋梗塞急性期(心筋梗塞の発生から10日以内)における活性化された心臓線維芽細胞に目的のタンパク質を発現させることができる。
コラーゲン1a2は、心臓線維芽細胞全体で、心筋梗塞の有無にかかわらず発現するタンパク質である。コラーゲン1a2プロモーター配列を用いることにより、心臓線維芽細胞に選択的に目的のタンパク質を発現させることができる。そのため、心筋梗塞の急性期及び慢性期における心臓線維芽細胞においても目的のタンパク質を発現させることができる。
軟骨オリゴマーマトリックスタンパク質(COMP)は、心筋梗塞慢性期(心筋梗塞の発生から10日後以降)に発現量が増加するタンパク質である。軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列を用いることにより、心筋梗塞慢性期における心臓線維芽細胞に選択的に目的のタンパク質を発現させることができる。
本発明のrAAVベクター内にパッケージングされるポリヌクレオチド(rAAVゲノム)は、目的のタンパク質(例えば、リプログラミング因子等)を発現させるため、そのタンパク質をコードするポリヌクレオチド配列がプロモーター配列と作動可能に組み合わされる。本発明において、プロモーター配列としては、ペリオスチンプロモーター配列、コラーゲン1a2プロモーター配列、軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列からなる群より選択される少なくとも1種が用いられる。

軟骨オリゴマーマトリックスタンパク質(COMP)は、心筋梗塞慢性期(心筋梗塞の発生から10日後以降)に発現量が増加するタンパク質である。軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列を用いることにより、心筋梗塞慢性期における心臓線維芽細胞に選択的に目的のタンパク質を発現させることができる。
本発明におけるペリオスチンプロモーター配列としては、下記(1-1)~(1-4)のいずれかであることが好ましい。
(1-1)配列番号3で表される塩基配列。
(1-2)配列番号3で表される塩基配列において1又は数個の塩基が置換、付加、又は欠失した塩基配列。
(1-3)配列番号3で表される塩基配列と90%以上の同一性を有する塩基配列。
(1-4)配列番号3で表される塩基配列に相補的な塩基配列とストリンジェントな条件下でハイブリダイズする塩基配列。
(1-1)配列番号3で表される塩基配列。
(1-2)配列番号3で表される塩基配列において1又は数個の塩基が置換、付加、又は欠失した塩基配列。
(1-3)配列番号3で表される塩基配列と90%以上の同一性を有する塩基配列。

上記(1-1)において、配列番号3で表される塩基配列であって、ペリオスチンプロモーターの機能を有するものが包含される。なお、配列番号3は、マウスのペリオスチンのエンハンサー配列にマウスのペリオスチンのプロモーター配列を結合した配列である(表1)。

ある態様において、置換、付加又は欠失する1又は複数の塩基の数は、配列番号3の塩基配列との塩基配列の同一性が90%以上となるように1~140個程度、好ましくは同一性が95%以上となるように1~69個程度、より好ましくは同一性が97%以上となるように1~42個程度、さらに好ましくは同一性が98%以上となるように1~28個程度、特に好ましくは同一性が99%以上となるように1~14個程度とすることができる。

本発明のrAAVベクター内にパッケージングされるポリヌクレオチドには、目的のタンパク質をコードするポリヌクレオチドが組み込まれる。コードするタンパク質は、1種類であっても2種以上であってもよいが、目的のタンパク質をコードするポリヌクレオチドを含めて、rAAVベクター内にパッケージングされるポリヌクレオチドの全長は野生型ゲノムと同等程度であることが好ましい。すなわち、全長が野生型の全長である5kbと同程度、例えば約2~6kb、好ましくは約4~6kbであることが好ましい。また、目的のタンパク質をコードするポリヌクレオチドは、好ましくは長さが約0.01~3.7kb、より好ましくは長さが約0.01~2.5kb 、さらに好ましく約0.01~2kbであるが、これに限定されない。
一実施形態において、上記目的のタンパク質としては、リプログラミング因子があげられる。リプログラミング因子としては、Gata4、Hand2、Tbx5、Mef2c、Mef2c-TAD、MYOCD(ミオカルディン)、ASCL1(Achaete-scute ファミリーbHLH転写因子1)、miR-133(マイクロRNA)等があげられる。これらは単独で用いてもよく、2種以上を組み合わせて用いてもよい。本発明のrAAVベクター内にパッケージングされるポリヌクレオチド中にコードされるリプログラミング因子は、Gata4、Hand2、Tbx5、Mef2c、Mef2c-TAD、MYOCD、ASCL1、及びmiR-133からなる群より選択される少なくとも1種であることが好ましく、2種以上であることがより好ましく、3種以上であることがさらに好ましく、4種以上であることが特に好ましい。一実施形態において、本発明のrAAVベクター内にパッケージングされるポリヌクレオチドは、Gata4とHand2とTbx5とをコードし、さらにMef2c又はMef2c-TADをコードすることが好ましく、Gata4とHand2とTbx5とMef2c-TADをコードすることがより好ましい。

Gata4ポリペプチドは多くの遺伝子のプロモーター領域に存在するGATAモチーフを認識して結合するGATAファミリー亜鉛フィンガー転写因子のメンバーである。様々な種に由来するGata4ポリペプチドに関するアミノ酸配列及びGata4ポリペプチドをコードするヌクレオチド配列が当該分野で知られている。Gata4ポリペプチドに関するアミノ酸配列のアクセッション番号としては、例えば、NP_002043(Homo sapiens)、NP_032118(Mus musculus)、NP_653331(Rattus norvegicus)等があげられる。Gata4ポリペプチドをコードするヌクレオチド配列のアクセッション番号としては、例えば、NM_002052(Homo sapiens)、NM_008092(Mus musculus)、NM_144730(Rattus norvegicus)等があげられる。

他の実施形態において、目的のタンパク質をコードするポリヌクレオチドとしては、マーカー遺伝子があげられる。マーカー遺伝子を組み込んだポリヌクレオチドをパッケージングしたrAAVベクターを被験動物に投与し、当該マーカー遺伝子を発現する細胞を確認することにより、遺伝子導入された細胞数及び分布を確認することができる。上記マーカー遺伝子としては、公知のものを特に制限なく用いることができる。このようなマーカー遺伝子としては、例えば、緑色蛍光タンパク質(GFP)遺伝子、LacZ遺伝子、発光タンパク質遺伝子(ホタルルシフェラーゼ等)等が挙げられる。
その他、目的のタンパク質としては、例えば、Creリコンビナーゼ等があげられるが、特に限定されない。
目的のタンパク質をコードするポリヌクレオチドを取得する方法として、PCR等の核酸増幅法を用いる方法を挙げることができる。例えば、当該ポリヌクレオチドのcDNAのうち、5’側及び3’側の配列(又はその相補配列)の中からそれぞれプライマーを調製し、これらプライマーを用いてゲノムDNA又はcDNA等を鋳型にしてPCR等を行い、両プライマー間に挟まれるDNA領域を増幅する。これによって、本発明に係るポリヌクレオチドを含むDNA断片を大量に取得できる。
本発明のrAAVベクター内にパッケージングされるポリヌクレオチドは、AAVゲノムの5’側及び3’側に位置するITR配列の間に位置する内部領域(すなわち、rep遺伝子及びcap遺伝子の一方又は両方)のポリヌクレオチドを、目的のタンパク質をコードするポリヌクレオチド(例えば、リプログラミング因子等)及びペリオスチンプロモーター配列、コラーゲン1a2プロモーター配列、軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列からなる群より選択される少なくとも1種を含む遺伝子カセットで置換したポリヌクレオチドであることが好ましい。当該ポリヌクレオチドの5’末端及び3’末端には、それぞれITR配列を有することがより好ましい。
アデノ随伴ウイルスのITRは、おおよそ145塩基の鎖長の領域であり、複製開始点等として機能する。本発明において、当該ポリヌクレオチドのITR配列は、複製開始点としての機能や宿主細胞への遺伝子挿入等の、本来のITRの機能の少なくとも一つを有するものである限り、いずれのセロタイプのAAV由来のものであってもよいが、好ましくはAAV2に由来するものである。
一実施形態において、5’側ITR配列は、配列番号6に示す塩基配列と少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、又は100%の配列同一性を有する塩基配列を含む。一実施形態において、3’側ITR配列は、配列番号7に示す塩基配列と少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、又は100%の配列同一性を有する塩基配列を含む。一般的に、ITR部分は容易に相補配列が入れ替わった配列をとるため、本発明のrAAVベクター内にパッケージングされるポリヌクレオチドに含まれるITRは、5’と3’の方向が逆転していてもよい。
2.rAAVベクターの調製
本発明のrAAVベクターを調製する方法としては、公知の方法を利用することができる。このような方法としては、例えば、AAVヘルパープラスミド(プラスミド1)、及びrAAVベクタープラスミド(プラスミド2)を、培養細胞にトランスフェクトする工程を含む。当該方法はさらに、アデノウイルスヘルパープラスミドを培養細胞にトランスフェクトする工程、又は、アデノウイルスを培養細胞に感染させる工程を含むことができる。さらに、上記のトランスフェクトされた培養細胞を培養する工程、及び培養上清より組換えアデノ随伴ウイルスベクターを収集する工程を含むことができる。
本発明のrAAVベクターを調製する方法としては、公知の方法を利用することができる。このような方法としては、例えば、AAVヘルパープラスミド(プラスミド1)、及びrAAVベクタープラスミド(プラスミド2)を、培養細胞にトランスフェクトする工程を含む。当該方法はさらに、アデノウイルスヘルパープラスミドを培養細胞にトランスフェクトする工程、又は、アデノウイルスを培養細胞に感染させる工程を含むことができる。さらに、上記のトランスフェクトされた培養細胞を培養する工程、及び培養上清より組換えアデノ随伴ウイルスベクターを収集する工程を含むことができる。
上記AAVヘルパープラスミド(プラスミド1)は、AAV-DJカプシドタンパク質をコードするポリヌクレオチド、及びRepタンパク質をコードするポリヌクレオチドを含む。AAV-DJカプシドタンパク質、及びRepタンパク質は、1種、又は2種以上のプラスミドに組込まれていてもよい。また、これらのAAV-DJカプシドタンパク質及びRepタンパク質のうちの1種以上がrAAVゲノムに含まれてもよい。本発明において、AAV-DJカプシドタンパク質及びRepタンパク質は全て、1種のポリヌクレオチドにコードされ、AAVヘルパープラスミドとして用いられることが好ましい。また、上記ポリヌクレオチドは、培養細胞において作動可能である公知のプロモーター配列に作動可能に結合されることが好ましい。このようなプロモーター配列としては、例えば、サイトメガロウイルス(CMV)プロモーター、EF-1αプロモーター、SV40プロモーター等を適宜使用することができる。さらに、AAVヘルパープラスミドは、公知のエンハンサー配列、Kozak配列、ポリA付加シグナル配列等を適宜含み得る。
本発明において用いられるRepタンパク質は、ITR配列を認識してその配列に依存してゲノム複製を行う機能、ウイルスベクター内へとrAAVゲノム(又は野生型AAVゲノム)をリクルートしてパッケージングする機能、本発明のrAAVベクターを形成する機能等公知の機能を野生型AAVのRepタンパク質と同程度に有する限り、野生型AAVのRepタンパク質のアミノ酸配列に対して、少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、又は100%の配列同一性を有するアミノ酸配列を含んでもよい。一実施形態において、AAV-DJのRepタンパク質のアミノ酸配列(配列番号8)に対して、少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、又は100%の配列同一性を有するアミノ酸配列を含んでもよい。本発明において、好ましくは、公知のAAV-DJ由来のRepタンパク質が使用される。
本発明において用いられるRepタンパク質をコードするポリヌクレオチドは、ITR配列を認識してその配列に依存してゲノム複製を行う機能、ウイルスベクター内へとrAAVゲノム(又は野生型AAVゲノム)をリクルートしてパッケージングする機能、本発明のrAAVベクターを形成する機能等公知の機能を野生型AAVのRepタンパク質と同程度に有するRepタンパク質をコードする限り、野生型AAVのRepタンパク質をコードするヌクレオチド配列に対して、少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、又は100%の配列同一性を有するヌクレオチド配列を含んでもよい。一実施形態において、AAV-DJのRepタンパク質をコードするヌクレオチド配列に対して、少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、又は100%の配列同一性を有するヌクレオチド酸配列を含んでもよい。本発明において、好ましくは、公知のAAV-DJ由来のRep遺伝子が使用される。
rAAVベクタープラスミド(プラスミド2)は、2つのITR配列の間に配置された目的のタンパク質をコードするヌクレオチド配列を含むプラスミドである。プラスミド2は、ペリオスチンプロモーター、コラーゲン1a2プロモーター、軟骨オリゴマーマトリックスタンパク質(COMP)プロモーターからなる群より選択される少なくとも1種と作動可能である位置に目的のタンパク質をコードするポリヌクレオチド配列を含む。さらに、公知のエンハンサー配列、Kozak配列、ポリA付加シグナル配列等を適宜含み得る。
アデノウイルスヘルパープラスミドは、アデノウイルス由来因子をコードするプラスミドである。AAVはヘルパー依存性ウイルスであるため、本発明のrAAVベクターを調製するためには、培養細胞に感染する際に、アデノウイルス等のヘルパーウイルスとの共感染を必要とする。ヘルパーウイルスとの共感染がない場合、AAVは、ウイルスゲノムを宿主細胞染色体に挿入するが、挿入されたウイルスゲノムに由来する感染性AAVベクターは生成されない。その挿入されたウイルスゲノムを有する宿主がヘルパーウイルスに感染される場合、組み込まれたゲノムに由来する感染性AAVベクターを生じ得る。
本発明のrAAVベクターの調製において、アデノウイルスヘルパープラスミドが、上記プラスミド1及びプラスミド2と同時に培養細胞に導入され得る。本発明において、好ましくは、アデノウイルスヘルパープラスミドは、培養細胞と同じ種のウイルスに由来する。例えば、ヒト培養細胞293Tを用いる場合、ヒトAdV由来のアデノウイルスヘルパープラスミドを用いることができる。このようなアデノウイルスヘルパープラスミドとして、市販品を使用することができる。
本発明のrAAVベクターの調製において、上記の1種以上のプラスミドを培養細胞にトランスフェクションする方法は、例えば、リン酸カルシウム法、リポフェクション法、エレクトロポレーション法等、公知の種々の方法を使用することができる。このような方法は、例えばMolecular Cloning 3rd Ed.、Current Protocols in Molecular Biology, John Wiley & Sons 1987-1997等に記載されている。
3.本発明のrAAVベクターを含む医薬組成物
別の実施形態において、本発明のrAAVベクターを含む医薬組成物を提供する。
一実施形態において、本発明のrAAVベクターは、リプログラミング因子をコードするポリヌクレオチドを含む。このようなrAAVベクターを含む医薬組成物は、種々の心臓障害(例えば、心筋梗塞、心不全、不整脈等)に対する治療に有用である。当該医薬組成物は、心機能改善剤として用いることができる。リプログラム因子としては、上記に記載のものを好ましく用いることができる。
別の実施形態において、本発明のrAAVベクターを含む医薬組成物を提供する。
一実施形態において、本発明のrAAVベクターは、リプログラミング因子をコードするポリヌクレオチドを含む。このようなrAAVベクターを含む医薬組成物は、種々の心臓障害(例えば、心筋梗塞、心不全、不整脈等)に対する治療に有用である。当該医薬組成物は、心機能改善剤として用いることができる。リプログラム因子としては、上記に記載のものを好ましく用いることができる。
本発明の医薬組成物における、上記のrAAVベクターの含有量は、医薬組成物100質量部に対して、0.001~100質量部程度とすることができる。すなわち、上記のrAAVベクターそのものを、本発明に係る医薬組成物としてもよい。
本発明の医薬組成物には、薬学分野の組成物を製造する際に使用される薬学的に許容可能な担体又は添加物を配合してもよい。この様な担体あるいは添加物は公知であり、その具体例として、任意の担体、希釈剤、賦形剤、懸濁剤、潤滑剤、アジュバント、媒体、送達システム、乳化剤、錠剤分解物質、吸収剤、保存剤、界面活性剤、着色剤、香料、又は甘味料等が挙げられる。
本発明の医薬組成物は上記の担体又は配合物を適宜組み合わせてあらゆる剤形とすることができる。具体的には、具体的な剤形としては、静脈注射剤、輸液剤、埋め込み注射剤、持続性注射剤等の注射剤;素錠、糖衣錠、フィルムコート錠、腸溶錠、口腔内崩壊錠、チュアブル錠、発泡錠、分散錠、溶解錠等の錠剤;硬カプセル錠、軟カプセル錠等のカプセル剤;発泡顆粒剤、徐放性顆粒剤、腸溶性顆粒剤等を含む顆粒剤;散剤;エリキシル剤、懸濁剤、乳剤、リモナーデ剤等の経口液剤;シロップ用剤等のシロップ剤;経口ゼリー剤等が挙げられる。
本発明の医薬組成物の使用対象動物は、生体であればヒトに限らずあらゆる動物個体を対象とできる。例えば、マウス、ラット、ウサギ、ハムスター、モルモット、サル、チンパンジー等の実験動物;イヌ、ネコ等の愛玩動物;その他保護を必要とするあらゆる動物種等が挙げられる。
本発明の医薬組成物の投与方法は、特に限定されず、上記の投与対象、剤形等を適宜勘案して公知の投与方法を採用すればよい。具体的には、心臓組織内、静脈内、経口、筋肉内、動脈内等へ投与する方法が挙げられ、心臓組織内又は静脈内への投与が好ましく、心臓組織内へ直接投与することがより好ましい。一実施形態において、本発明の医薬組成物の投与方法は、本発明の医薬組成物を個体の線維芽細胞に接触させることを包含する。「接触」は、本発明の医薬組成物を、治療部位又はその近傍(例えば心臓の中又は周囲)において、個体に投与することを含む。投与の方法としては、例えば、末端の動脈に挿入したカテーテルを心臓患部近傍まで導き、カテーテルの先端から線維化した患部組織に本発明の医薬組成物を注入して接触させる方法が挙げられる。
本発明の医薬組成物の投与量は特に限定されず、疾患の種類、患者の年齢、症状、投与経路、治療の目的、併用薬剤の有無等の種々の条件に応じて適切な投与量を選択することが可能である。本発明の医薬組成物の投与量は、例えば、ヒトであれば、通常は0.1~50mg/kg程度とすればよく、投与対象動物がマウスであれば、通常は0.1~50mg/kg程度とすればよい。また、投与単位としてvg(vector genome)を利用する場合、例えば、体重1kgあたり、109~1014vg、好ましくは1010~1013vg、さらに好ましくは1010~1012vgの範囲の投与量を選択することが可能であるが、これらに限定されない。その他の投与対象動物であれば、上記のヒト及びマウスにおける投与量を基に適宜設定することができる。また、本発明の医薬組成物の投与は、上記の量を一日に一度に投与してもよく、数回に分けて投与してもよい。また、治療効果を有する範囲において、投与間隔は、毎日、隔日、毎週、隔週、2~3週毎、毎月、隔月又は2~3ヶ月毎でもよい。
4.本発明のrAAVベクターを用いた遺伝子発現
一実施形態において、本発明のAAVベクターを用いた遺伝子発現方法を提供する。当該方法は、本発明のrAAVベクターを投与することを含む。投与の対象としては、個体であってもよく、組織、細胞等であってもよい。本発明のAAVベクターは、活性化された心臓線維芽細胞において目的の遺伝子を発現させることができる。活性化された心臓線維芽細胞は、心筋梗塞後の心臓線維芽細胞であることが好ましい。一実施形態において、個体は非ヒト哺乳動物である。

一実施形態において、当該遺伝子発現方法は、リプログラミング因子をコードするポリヌクレオチドを含むrAAVベクターを投与することを含む。リプログラミング因子をコードするポリヌクレオチドとしては、上記に記載のものが好ましく用いられる。このようなrAAVベクターを用いた当該遺伝子発現方法により、活性化された心臓線維芽細胞から心筋細胞が誘導される。
5.本発明のrAAVベクターの調製キット
本発明は別の実施形態において、本発明のrAAVベクターを調製するためのキットを提供する。このようなキットは、例えば、上記のプラスミド1、及びプラスミド2を含むことができる。また、本発明のrAAVベクターを調製するためのキットは、本明細書中に記載されるいずれの構成(例えば、アデノウイルスヘルパープラスミド等)もさらに含むことができる。
本発明は別の実施形態において、本発明のrAAVベクターを調製するためのキットを提供する。このようなキットは、例えば、上記のプラスミド1、及びプラスミド2を含むことができる。また、本発明のrAAVベクターを調製するためのキットは、本明細書中に記載されるいずれの構成(例えば、アデノウイルスヘルパープラスミド等)もさらに含むことができる。
以下、実施例を挙げて本発明を具体的に説明するが、これらの実施例は本発明の範囲を限定するものではない。

[材料及び方法]
(AAVベクターの作製)
AAVベクターは、Kurosaki et al., 2017及びMizukami et al., 2006に記載された方法により作製した。組換えAAVベクタープラスミドは、GFP、Cre、GATA4、HAND2、TBX5、MEF2C、及びMEF2C-TADのコード領域をPCRで増幅することによって作製した。増幅した上記DNAをAAV-CMV(Agilent Technologies)及びAAV-Postn(ペリオスチン)(Piras et al., 2016) にサブクローニングした。また、増幅したGFPのDNAをAAV-COMP及びAAV-Col1a2にサブクローニングした。AAV-DJ Rep-CapはCell Biolabs, Inc.から購入した。パッケージングプラスミド(AAV1~10, DJ)とヘルパープラスミド(pHelper for AAV1~10:Agilent Technologies、pHelper for AAV-DJ:Cell Biolabs Inc.)を、リン酸カルシウム沈殿を用いてHEK293細胞に共導入した。ウイルス含有培地を3日後に回収し、細胞をPBSに再懸濁した。AAVベクターは、ポリエチレングリコール沈殿と塩化セシウムを用いた超遠心分離を用いて精製した。各ベクターの力価(VG)は、リアルタイムPCR(Thermal Cycler Dice(登録商標) Real Time System III, Takara Bio Inc.)で測定した。なお、本実施例にて使用したAAV-DJカプシドタンパク質のアミノ酸配列を配列番号1に示す。AAV-DJカプシドタンパク質をコードするポリヌクレオチド配列を配列番号2に示す。ペリオスチンプロモーターのポリヌクレオチド配列を配列番号3に示す。Mef2c-TADポリペプチドのアミノ酸配列を配列番号4に示す。Mef2c-TADポリペプチドをコードするポリヌクレオチド配列を配列番号5に示す。軟骨オリゴマーマトリックスタンパク質(COMP)プロモーターのポリヌクレオチド配列を配列番号9に示す。コラーゲン1a2プロモーターのポリヌクレオチド配列を配列番号10に示す。

(マウス)
遺伝子改変Tcf21 iCre/Tomatoマウスは、Tcf21 iCreマウスとR26R-tdTomatoマウスとの交配により得た。また、遺伝子改変Postn MCM/Tomatoマウスは、Postn MCMマウスとR26R-tdTomatoマウスとの交配により得た。ICR(Jcl:ICR)マウスは日本クレア株式会社から購入した。トランスジェニックマウスに免疫不全及び他の健康問題は観察されず、本実験開始まで他実験及び薬物未使用の状態であった。すべての動物を、12時間/12時間の明暗サイクルを持つ専用の飼育SPF(Specific pathogen free)施設にて群飼いし、餌と水に自由にアクセスできる状態で、毎日モニターした。SPFグレードを維持するために定期的に健康状態をチェックした。外科処置の対象となった動物は、同じ条件下でSPF施設に移した。

実験は8週齢の雌雄マウスを無作為に割り付けて行った。麻酔の導入と維持にはイソフルランを使用した。低体温を防ぐためにホットマットを使用した。経口気管挿管を行い、皮膚と心臓上の肋間切開から胸部アプローチを行った。心臓を露出し、7-0絹縫合糸を用いて左冠動脈を結紮した。冠動脈結紮後すぐに、60μLのAAVベクター含有溶液(1×1011 VG/マウス)を29ゲージの注射針で梗塞と境界域の境界に3箇所注射した。全身投与では、冠動脈結紮直後に尾静脈からAAVベクター含有溶液(1×1012 VG/マウス)200μLを注入した。マウスを操作する人は、この試験について盲検化されていた。心筋梗塞後の死亡率は10%未満と記録された。心エコー検査は術後2日目に行い、駆出率(EF)が45%以上のマウスは解析から除外した。

(ラット初代培養心筋細胞及びラット初代培養心臓線維芽細胞)
出生後のSprague-Dawleyラット(P1-3)由来の細胞を、パーコール勾配遠心法を用いて行った遠心分離に基づいて心筋細胞と心臓線維芽細胞に分け、プラスチックディッシュに付着させて心筋細胞を得た (Haginiwa et al., 2019)。細胞を10%FBS添加DMEM/Medium199に再懸濁し、37℃、5%CO2で培養し、培地は2-3日ごとに交換した。ラット心筋細胞及び心臓線維芽細胞は、翌日に上記ベクター(2.5×105 VG/細胞)で形質導入し、さらに1週間培養した。
出生後のSprague-Dawleyラット(P1-3)由来の細胞を、パーコール勾配遠心法を用いて行った遠心分離に基づいて心筋細胞と心臓線維芽細胞に分け、プラスチックディッシュに付着させて心筋細胞を得た (Haginiwa et al., 2019)。細胞を10%FBS添加DMEM/Medium199に再懸濁し、37℃、5%CO2で培養し、培地は2-3日ごとに交換した。ラット心筋細胞及び心臓線維芽細胞は、翌日に上記ベクター(2.5×105 VG/細胞)で形質導入し、さらに1週間培養した。
(マウス胎児線維芽細胞 (MEF))
妊娠12.5~13.5日のマウス胎児を回収し、無菌的に実体顕微鏡下で頭部、心臓、その他の内臓を取り除き、残った胴体部を3~5個体分集めてハサミで細かく刻んだ。得られた組織片を、あらかじめ37℃に予温した0.25%トリプシン/EDTA溶液15 mLに移し、37℃で20分間インキュベートし、反応停止のために同量のFBS(15 mL)を加え、ピペッティングを数回行って組織を解離した。解離後の懸濁液を遠心し、得られたペレットを10 mLのMEF培地に再懸濁し、再懸濁した細胞を100 mmディッシュに播種し、5% CO2、37℃の条件下で培養した。
なお、培地は、Dulbecco's Modified Eagle Medium(DMEM)に10%ウシ胎児血清(FBS)、1%ピルビン酸ナトリウム、1%非必須アミノ酸(NEAA)、1%抗生物質・抗真菌剤混合液(PSA)を加えて調製した。
妊娠12.5~13.5日のマウス胎児を回収し、無菌的に実体顕微鏡下で頭部、心臓、その他の内臓を取り除き、残った胴体部を3~5個体分集めてハサミで細かく刻んだ。得られた組織片を、あらかじめ37℃に予温した0.25%トリプシン/EDTA溶液15 mLに移し、37℃で20分間インキュベートし、反応停止のために同量のFBS(15 mL)を加え、ピペッティングを数回行って組織を解離した。解離後の懸濁液を遠心し、得られたペレットを10 mLのMEF培地に再懸濁し、再懸濁した細胞を100 mmディッシュに播種し、5% CO2、37℃の条件下で培養した。
なお、培地は、Dulbecco's Modified Eagle Medium(DMEM)に10%ウシ胎児血清(FBS)、1%ピルビン酸ナトリウム、1%非必須アミノ酸(NEAA)、1%抗生物質・抗真菌剤混合液(PSA)を加えて調製した。
(タモキシフェン投与)
Tcf21 iCre/Tomatoマウスに、クエン酸タモキシフェンを5日間連続で腹腔内投与した(2 mg/日;Millipore Sigma, T5648)。タモキシフェンは90%ピーナッツ油(Millipore Sigma, P2144)と10%エタノールに溶解し、50mg/mLの濃度とした。MI手術はタモキシフェン投与最終日の7日後に行った。Postn MCM /Tomatoマウスでは、タモキシフェン含有フードペレットを心筋梗塞手術後2週間継続して投与した。フードペレットは、クエン酸タモキシフェン0.4g(T2510、東京化成工業)及び標準マウスチャウ(MF、オリエンタル酵母工業)1kgあたりショ糖49.6gを添加して調製した。
Tcf21 iCre/Tomatoマウスに、クエン酸タモキシフェンを5日間連続で腹腔内投与した(2 mg/日;Millipore Sigma, T5648)。タモキシフェンは90%ピーナッツ油(Millipore Sigma, P2144)と10%エタノールに溶解し、50mg/mLの濃度とした。MI手術はタモキシフェン投与最終日の7日後に行った。Postn MCM /Tomatoマウスでは、タモキシフェン含有フードペレットを心筋梗塞手術後2週間継続して投与した。フードペレットは、クエン酸タモキシフェン0.4g(T2510、東京化成工業)及び標準マウスチャウ(MF、オリエンタル酵母工業)1kgあたりショ糖49.6gを添加して調製した。
(試験管内でのAAVベクターの感染)
一次培養した新生児ラットの心筋細胞及び心臓線維芽細胞を指定のAAVベクター(2.5×105 VG/細胞)で形質導入し、さらに1週間培養した。細胞をFACS及び免疫蛍光で分析し、AAVの細胞トロピズムを決定した。

(インビボでのAAVベクターの感染)
心臓への直接注射では、1×1011 VGのAAVベクターを心筋梗塞後又は無傷のマウスの心臓に注射した。全身投与では、1×1012 VGのAAVベクターをマウスの尾静脈に注射した。

(蛍光活性化セルソーティング(FACS)分析)
マウスをCO2吸入で安楽死後、心臓を直ちにカニュレーションして冷却PBS (50 mL) で灌流した。心臓を摘出後、心房と弁を切離して心室を分離し、心室心筋を氷上の滅菌シャーレ上で1 mm程度のサイズに切断した。酵素溶液 3 mL を含む 10 mL チューブに移し、37℃のウォーターバスで 45 分間インキュベートし、15 分ごとにピペッティングした。酵素液には、2 mg/mL collagenase type IV (Worthington Biochemical,、CLS-4) と 1.2 U/mL Dispase II (Sigma,、255-914-4) が含まれていた。最終ピペッティング後、細胞懸濁液を 40 μm セルストレーナーで濾過した。Debris removal solution(Miltenyi Biotec,、130-109-398)による遠心分離でデブリを除去後、RBC lysis buffer(pluriSelect,、60-00050-11)を用いて赤血球を除去した。
その後、GFP発現を検出するために、フローサイトメーター(Beckman Coulter, CytoFLEX S)とFlowJoソフトウェア(BD Biosciences, Franklin Lakes, NJ, USA)を用いて細胞を解析した。

細胞を4%PFAで25℃、15分間固定し、5%正常ヤギ血清ブロッキング液(Vector Laboratories, S-1000)でブロックし、0.2%TritonX-100(Sigma)で10分間透過処理し、GFPに対する一次抗体とインキュベートした。その後、Alexa Fluor 488を結合させた二次抗体とインキュベートした。観察は、オールインワン蛍光顕微鏡(Keyence, BZX810)を用いて行った。
細胞を4%PFAで25℃、15分間固定し、5%正常ヤギ血清ブロッキング液(Vector Laboratories, S-1000)でブロックし、0.2%TritonX-100(Sigma)で10分間透過処理し、GFPに対する一次抗体とインキュベートした。その後、Alexa Fluor 488を結合させた二次抗体とインキュベートした。観察は、オールインワン蛍光顕微鏡(Keyence, BZX810)を用いて行った。
(免疫組織化学)
マウスの心臓、肝臓、肺、骨格筋を4%PFAで一晩固定した後、20%スクロース溶液で置換し、OCTに包埋し、液体窒素で凍結した。凍結切片は7μmの厚さで切り取った。切片をα-アクチニン(Sigma, A7811)、CD31(Abcam, ab28364)、CD45(Thermo, 30-F11)、COL1(Millipore, Millipore Sigma, AB765P)、COMP(PGI, 28369-1-AP)、cTnT(Thermo, MS-295-P1)、GFP(MBLライフサイエンス, 598)、及びSMMHC(Biomedical Technologies, Madrid, Spain, BT562)に対する一次抗体で染色した。Alexa 546又はAlexa 488を結合させた二次抗体とインキュベートし、DAPI(Thermo, D21490)を用いて細胞核を染色した。共焦点顕微鏡はLSM800顕微鏡(Carl Zeiss)を用いて行い、標準プロトコールに従ってZスタック画像を収集した。測定と計算は盲検下で行った。Comp+ の面積はAdobe Photoshopの解析ツールを用いて測定した。免疫陽性細胞の比率は、セクションごとに無作為に選んだ6フィールドで測定した。各マウスのデータは30から50の切片をもとに計算し、1群につき5匹のマウスを観察した。各マウスについて合計36,000-60,000個の細胞をカウントした。
マウスの心臓、肝臓、肺、骨格筋を4%PFAで一晩固定した後、20%スクロース溶液で置換し、OCTに包埋し、液体窒素で凍結した。凍結切片は7μmの厚さで切り取った。切片をα-アクチニン(Sigma, A7811)、CD31(Abcam, ab28364)、CD45(Thermo, 30-F11)、COL1(Millipore, Millipore Sigma, AB765P)、COMP(PGI, 28369-1-AP)、cTnT(Thermo, MS-295-P1)、GFP(MBLライフサイエンス, 598)、及びSMMHC(Biomedical Technologies, Madrid, Spain, BT562)に対する一次抗体で染色した。Alexa 546又はAlexa 488を結合させた二次抗体とインキュベートし、DAPI(Thermo, D21490)を用いて細胞核を染色した。共焦点顕微鏡はLSM800顕微鏡(Carl Zeiss)を用いて行い、標準プロトコールに従ってZスタック画像を収集した。測定と計算は盲検下で行った。Comp+ の面積はAdobe Photoshopの解析ツールを用いて測定した。免疫陽性細胞の比率は、セクションごとに無作為に選んだ6フィールドで測定した。各マウスのデータは30から50の切片をもとに計算し、1群につき5匹のマウスを観察した。各マウスについて合計36,000-60,000個の細胞をカウントした。
(線維化領域の測定)
マッソン三色染色は、標準的な手順に従ってパラフィン包埋切片に対して行った。瘢痕の大きさを測定するために、Adobe Photoshopを用いて、左心室内の3つのレベル(心尖部、中間部、基部)にまたがる横断切片の瘢痕面積(青色)と健常域面積(赤色)を測定した。各レベルで3枚の組織切片(合計9切片)を測定し、1群につき5匹のマウスを観察した。測定と計算は盲検下で行った。

(心エコー検査)
心機能は心エコー(Vevo 2100, Visual Sonics)を用いて評価した。心エコーはイソフルラン麻酔下で行った。麻酔深度は心拍数が約500/分になるように調整した。左室短軸像を描出し、乳頭筋レベルでMモードを測定して左室拡張末期径と収縮末期径を求め、駆出率(EF)を算出した。駆出率(EF)と短縮率(FS)はTeichholtzの式を用いて算出した。

[比較例1]CMVプロモーターを用いたin vitro及びin vivoスクリーニング(in vitro)
AAVカプシドの心臓線維芽細胞トロピズムを調べるため、10種類の天然型(AAV1~10)と1種類のキメラ型(AAV-DJ)をスクリーニングした。構成的CMVプロモーターの制御下でGFPを発現する11種類のAAVベクター血清型(AAV-CMV-GFP)を感染多重度2.5×105 VG/cell(ベクターゲノム/細胞)でin vitroで初代培養ラットCF及びCMに導入した。導入効率は、1週間後に蛍光活性化セルソーティング(FACS)と免疫蛍光(IF)を用いて決定した(図1)。FACS解析の結果、AAV1、AVV6、AVV-DJが11種類の血清型の中で心臓線維芽細胞において高い形質導入効率を示した(図2)。心筋細胞の遺伝子治療に広く用いられているAAV2とAAV9は、in vitroでは心臓線維芽細胞での導入効率が低かった。対照的に、AAV-DJを含むほとんどのAAVベクターは、in vitroで心筋細胞において中程度の導入効率(~40%)を示した(図3)。免疫蛍光染色により、AAV1、AAV6、及びAAV-DJは心臓線維芽細胞において強いGFP発現を示し、AAV1とAAV4は培養中の心筋細胞において強いGFP発現を示した(図4)。

[実施例1]AAV-DJ-Postnベクターを用いたin vivoでの心臓線維芽細胞での遺伝子発現 ペリオスチン(Postn)プロモーター駆動型Creリコンビナーゼ(AAV-Postn-Cre)を含むAAVコンストラクトを作製し、AAV6、9、DJカプシドにパッケージした(図6)。AAV6-Postn-Creベクター、AAV9-Postn-Creベクター、AAV-DJ-Postn-Creベクター(以後、単にAAV6-Postn-Cre、AAV9-Postn-Cre AAV-DJ-Postn-Creと称する場合がある)を心筋梗塞後のTomatoレポーターマウスの心臓に直接注入し、導入遺伝子の発現を測定した(図7)。AAV6-Postn-Cre, AAV9-Postn-Cre, AAV-DJ-Postn-Creでは、Tomatoレポーターは主に梗塞領域と境界領域で発現したが、遠隔領域では発現しなかった(図8)。特に、AAV-DJ-Postn-Creは3つの血清型の中で最も高い導入効率を示した(図9)。心筋細胞(cTnT)、心筋線維芽細胞(COL1)、平滑筋細胞(SMC; SMMHC)、内皮細胞(EC; CD31)に対する複数の抗体を用いた共免疫染色により、Tomatoが主に常在CFで発現していることが示され、AAV-DJ-Postn-Cre注入による導入効率は約90%に達した(図10)。Tomato+細胞は、心筋梗塞後4週間、境界部と梗塞部で維持された。
[実施例2]AAV-DJ-Postnベクターを用いたヒト初代培養心臓線維芽細胞での遺伝子発現 ヒト心臓線維芽細胞における遺伝子導入効率を調べるために、ヒト心臓由来心臓線維芽細胞を培養し、AAV-DJ-Postn-GFPを構築し、遺伝子導入を行った。感染多重度5x104 vg/cellの条件で遺伝子導入を行い、導入効率は導入1週間後に免疫蛍光(IF)を用いて決定した。その結果、AAV-DJ-Postnベクターを用いた場合、ヒト心臓線維芽細胞においてもin vitroで遺伝子発現が確認された(データ非図示)。
[実施例3]Postnプロモーターの制御下での導入遺伝子発現の時間経過の確認
Postnプロモーターの制御下での導入遺伝子発現の時間経過を調べるため、AAV-DJ-Postn-GFPを構築し、心筋梗塞後のマウス心臓に直接注入した(図11)。心筋梗塞後の心臓では、心筋梗塞1週間後と比較して、2週間後と4週間後にGFP発現が低下した(図12)。
Postnプロモーターの制御下での導入遺伝子発現の時間経過を調べるため、AAV-DJ-Postn-GFPを構築し、心筋梗塞後のマウス心臓に直接注入した(図11)。心筋梗塞後の心臓では、心筋梗塞1週間後と比較して、2週間後と4週間後にGFP発現が低下した(図12)。
[実施例4]AAV-DJ-Postnベクターの特異性
免疫組織化学染色及びFACS解析により、1週間後、GFP+細胞が心臓で検出されたが、肝臓、肺、骨格筋等の他の組織では検出されなかったことが確認された(図13~15)。AAV-DJ-Postn-GFPを健康で梗塞のないマウスの心臓に直接注入すると、注入部位でのみ低レベルのGFP発現が検出された。さらに、AAV-DJ-Postn-Creを尾静脈から健常対照マウス(図16,17)及び梗塞Tomatoレポーターマウス(図18,19)に全身投与したところ、Tomatoは梗塞部でのみ発現し、健常心筋及びその他の組織では発現しなかった。これらの結果は、AAV-DJ-Postnベクターが心筋損傷後の活性化CFを優先的に標的とするが、静止CF及び他の臓器は標的としないことを示唆している。

[実施例5]AAV-DJ-Postnベクターを用いたin vivoでの心筋線維芽細胞の心筋リプログラミング(1)
AAV-DJ-Postnベクターを用いた遺伝子導入により、生体内に常在する心臓線維芽細胞を誘導心筋細胞にリプログラミングできるかどうかを調べた。まず、GATA4、HAND2、TBX5、MEF2C、及びMEF2C-TAD(M-TAD)を含むリプログラミング因子を発現するAAV-DJ-Postnベクターを作製した(図20)。インビボでの心臓リプログラミングを試験するために、
(i)リン酸緩衝生理食塩水(PBS)(コントロール、Ctrl)、
(ii)PBSに懸濁したAAV-DJ-Postn-GATA4、AAV-DJ-Postn-HAND2、AAV-DJ-Postn-TBX5とAAV-DJ-Postn-MEF2C(AAV-DJ-Postn-GHT/M)、又は
(iii)PBSに懸濁したAAV-DJ-Postn-GATA4、AAV-DJ-Postn-HAND2、AAV-DJ-Postn-TBX5とAAV-DJ-Postn-MEF2C-TAD(AAV-DJ-Postn-GHT/M-TAD)
上記(i)~(iii)のいずれかを心筋梗塞直後のPostn MCM/Tomatoマウス心臓に直接注入し、活性化されたCF系譜を追跡した(図21,22)。マウスには心筋梗塞後2週間タモキシフェンを継続投与し、2週間後に心臓を採取した(Kanisicakら、2016)。免疫組織化学染色により、AAV-DJ-Postn-GHT/M及びAAV-DJ-Postn-GHT/M-TADの両方が、Tomato+細胞にα-アクチニン発現を誘導することが明らかになった(図23)。α-アクチニン+/Tomato+ 誘導心筋細胞は明瞭なサルコメア構造を示し、三次元解析により細胞が両方のタンパク質を発現していることが確認された。重要なことは、AAV-DJ-Postn-GHT/M-TADは、境界領域においてAAV-DJ-Postn-GHT/Mよりも2倍多くの誘導心筋細胞を生成したことであり、活性化MEF2Cがin vivoでの心臓リプログラミングを促進することを示唆している(図24)。

[実施例6]AAV-DJ-Postnベクターを用いたin vivoでの心筋線維芽細胞の心筋リプログラミング(2)
Tcf21 iCre/Tomatoマウスは、心筋梗塞前にタモキシフェンを投与することで、静止期の心臓線維芽細胞を特異的に標識することができるため、心筋細胞におけるリーク発現なしに心臓線維芽細胞の系統を追跡することができる。Tcf21 iCre/Tomatoマウスは、心筋梗塞前にTcf21発現心臓線維芽細胞を標識するために5日間タモキシフェンで処理され、最後のタモキシフェン処理から7日後に冠動脈結紮が行われた。Tcf21 iCre/Tomatoマウスの心筋梗塞4週後の境界/梗塞部には、Postn MCM/Tomatoマウスと同様に、多数のTomato+心臓線維芽細胞が認められた(図25~27)。心筋梗塞直後のTcf21 iCre/Tomatoマウスの心臓にPBS又はAAV-DJ-Postn-GHT/M-TADを直接注入した。免疫組織化学染色の結果、各群5匹ずつ計36,000~60,000個の細胞をカウントしたところ、AAV-DJ-Postn-GHT/M-TAD群では境界部のTomato+細胞の約1.8%がα-アクチニンを発現していたのに対し、Ctrl群では0.3%がα-アクチニンを発現していた(図28)。誘導心筋細胞は明確なサルコマー構造を示した。これらの結果は、活性化MEF2CとGHTを発現するAAV-DJ-Postnベクターが、心筋梗塞後の常在心臓線維芽細胞を誘導心筋細胞に効率的にリプログラミングすることを示している。

[実施例7]AAV-DJ-Postnベクターを用いた心筋リプログラミングによる線維化抑制の確認
AAV-DJ-Postn-GHT/M-TADによる心臓リプログラミングによって誘導される分子変化を調べるために、心筋梗塞の4週間後にCtrl-及びAAV-DJ-Postn-GHT/M-TADを注入したマウスの心臓でRNAシークエンス解析を行った。遺伝子セットの濃縮解析から、GHT/M-TAD心臓で発現上昇した遺伝子は、心収縮又は代謝過程等心機能に関連するクラスターに濃縮されていたのに対し、発現低下した遺伝子は線維芽細胞シグネチャー又は炎症反応に濃縮されていた(図29)。
次に、Ctrlマウス、AAV-DJ-Postn-GFP-マウス、及びAAV-DJ-Postn-GHT/M-TAD-注射マウスの心機能を、心筋梗塞の2日後及び2週後と4週後に心エコー図を用いて評価した。AAV-DJ-Postn-GHT/M-TAD投与は、4週間後に左室駆出率、分画短縮を有意に改善した(図30)。
また、4週後の瘢痕の大きさを定量化するために、マッソン三色染色を用いて組織学的解析を行った。AAV-DJ-Postn-GHT/M-TADを投与した心臓では、線維化領域がCtrl及びAAV-DJ-Postn-GFPを投与した心臓よりも有意に減少した(図31)。
このように、AAV-DJ-Postn-GHT/M-TADを用いたin vivo心臓リプログラミングは、心筋梗塞後の心機能を改善し、線維症を減少させた。
AAV-DJ-Postn-GHT/M-TADによる心臓リプログラミングによって誘導される分子変化を調べるために、心筋梗塞の4週間後にCtrl-及びAAV-DJ-Postn-GHT/M-TADを注入したマウスの心臓でRNAシークエンス解析を行った。遺伝子セットの濃縮解析から、GHT/M-TAD心臓で発現上昇した遺伝子は、心収縮又は代謝過程等心機能に関連するクラスターに濃縮されていたのに対し、発現低下した遺伝子は線維芽細胞シグネチャー又は炎症反応に濃縮されていた(図29)。

このように、AAV-DJ-Postn-GHT/M-TADを用いたin vivo心臓リプログラミングは、心筋梗塞後の心機能を改善し、線維症を減少させた。
[実施例8]AAV-DJ-Col1a2ベクターおよびAAV-DJ-COMPベクターを用いたマウス胎児線維芽細胞、心臓線維芽細胞での遺伝子発現
マウス胎児線維芽細胞及び遺伝子導入効率を調べるために、これらの細胞を培養し、AAV-DJ-Col1a2-GFPベクター及びAAV-DJ-COMP-GFPベクターを構築し(図32)、遺伝子導入を行った。感染多重度5x104 vg/cellの条件で遺伝子導入を行い、導入効率は導入1週間後に免疫蛍光(IF)を用いて決定した。その結果、AAV-DJ-Col1a2ベクターを用いた場合、AAV-DJ-CMVベクター及びAAV-DJ-Postnベクターには効率が劣るものの、マウス胎児線維芽細胞及び心臓線維芽細胞においてin vitroで遺伝子発現が確認された(図33)。また、AAV-DJ-COMP-GFPベクターを用いた場合、導入効率は低いものの、マウス胎児線維芽細胞及び心臓線維芽細胞においてin vitroで遺伝子発現が確認された(図33)。
一方、心筋細胞においても同様に検討した結果、いずれのベクターにおいても心筋細胞では発現が見られず(図34)、非心筋細胞への選択性が確認できた。
マウス胎児線維芽細胞及び遺伝子導入効率を調べるために、これらの細胞を培養し、AAV-DJ-Col1a2-GFPベクター及びAAV-DJ-COMP-GFPベクターを構築し(図32)、遺伝子導入を行った。感染多重度5x104 vg/cellの条件で遺伝子導入を行い、導入効率は導入1週間後に免疫蛍光(IF)を用いて決定した。その結果、AAV-DJ-Col1a2ベクターを用いた場合、AAV-DJ-CMVベクター及びAAV-DJ-Postnベクターには効率が劣るものの、マウス胎児線維芽細胞及び心臓線維芽細胞においてin vitroで遺伝子発現が確認された(図33)。また、AAV-DJ-COMP-GFPベクターを用いた場合、導入効率は低いものの、マウス胎児線維芽細胞及び心臓線維芽細胞においてin vitroで遺伝子発現が確認された(図33)。
一方、心筋細胞においても同様に検討した結果、いずれのベクターにおいても心筋細胞では発現が見られず(図34)、非心筋細胞への選択性が確認できた。
[実施例9]AAV-DJ-Col1a2ベクターを用いたin vivoでの心臓線維芽細胞での遺伝子発現
AAV-DJ-Col1a2-GFPベクターを心筋梗塞後のICRマウスの心臓に直接注入し、1週間後、2週間後、4週間後での導入遺伝子の発現を免疫組織化学染色で観察した(図35)。
その結果、心筋梗塞後のいずれの期間においても有意な遺伝子発現が観察された(図36)。AAV-DJ-Col1a2ベクターでは、心筋梗塞の急性期から慢性期まで広く遺伝子発現が可能であることが明らかとなった。
AAV-DJ-Col1a2-GFPベクターを心筋梗塞後のICRマウスの心臓に直接注入し、1週間後、2週間後、4週間後での導入遺伝子の発現を免疫組織化学染色で観察した(図35)。
その結果、心筋梗塞後のいずれの期間においても有意な遺伝子発現が観察された(図36)。AAV-DJ-Col1a2ベクターでは、心筋梗塞の急性期から慢性期まで広く遺伝子発現が可能であることが明らかとなった。
Claims (7)
- ペリオスチンプロモーター配列、コラーゲン1a2プロモーター配列、軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列からなる群より選択される少なくとも1種を含むポリヌクレオチドと、AAV-DJカプシドタンパク質とを含む、組換えアデノ随伴ウイルスベクターであって、
前記コラーゲン1a2プロモーター配列は、以下の(2-1)~(2-4)のいずれかであり、前記軟骨オリゴマーマトリックスタンパク質(COMP)プロモーター配列は、以下の(3-1)~(3-4)のいずれかである、組換えアデノ随伴ウイルスベクター:
(2-1)配列番号9で表される塩基配列、
(2-2)配列番号9で表される塩基配列において1又は数個の塩基が置換、付加、又は欠失した塩基配列、
(2-3)配列番号9で表される塩基配列と90%以上の同一性を有する塩基配列、
(2-4)配列番号9で表される塩基配列に相補的な塩基配列とストリンジェントな条件下でハイブリダイズする塩基配列、
(3-1)配列番号10で表される塩基配列、
(3-2)配列番号10で表される塩基配列において1又は数個の塩基が置換、付加、又は欠失した塩基配列、
(3-3)配列番号10で表される塩基配列と90%以上の同一性を有する塩基配列、
(3-4)配列番号10で表される塩基配列に相補的な塩基配列とストリンジェントな条件下でハイブリダイズする塩基配列。 - 前記組換えアデノ随伴ウイルスベクターが、Gata4、Hand2、Tbx5、及びMef2cからなる群より選択される少なくとも1つのリプログラミング因子をコードするポリヌクレオチドを含む、請求項1に記載の組換えアデノ随伴ウイルスベクター。
- 請求項1又は2に記載の組換えアデノ随伴ウイルスベクターを含む、医薬組成物。
- 請求項2に記載の組換えアデノ随伴ウイルスベクターを含む、心機能改善剤。
- 請求項1又は2に記載の組換えアデノ随伴ウイルスベクターを投与することを含む、活性化された心臓線維芽細胞における遺伝子発現方法。
- 活性化された心臓線維芽細胞が、心筋梗塞後の心臓線維芽細胞を含む、請求項5に記載の遺伝子発現方法。
- 請求項1又は2に記載の組換えアデノ随伴ウイルスベクターを投与することを含む、心臓線維芽細胞から心筋細胞を誘導する方法。
