キャピラリを狭い空間に集積させても、前方散乱光の混在を生じさせないため、複数のキャピラリ(122)にレーザ光(141)を照射するライトシート(121)と、キャピラリ(122)に対し、レーザ光(141)が照射された側とは反対側に散乱する前方散乱光(142)を受光するラインセンサ(123)と、レーザ光(141)が入射する側におけるキャピラリ(122)の整列面を含む光源部に近い位置、かつ、ライトシート(121)の方向にキャピラリ(122)の軸中心を投影した位置からずれた位置に配置される第1のレーザブロッカ(111)と、キャピラリ(122)と、ラインセンサ(123)との間、かつ、キャピラリ(122)同士の間に設置される第2のレーザブロッカ(112)と、ラインセンサ(123)の受光面において、ラインセンサ(123)の方向にキャピラリ(122)の軸中心を投影した位置に設置される第3のレーザブロッカ(113)と、を有する。
Description
本発明は、フローサイトメータ及びフローサイトメータ信号解析方法の技術に関する。
フローサイトメトリ(FCM)は細胞等といったサンプルの表面や内部の分子の発現を分析する技術である。フローサイトメトリでは、サンプルが整列して流され、1つのサンプル毎に順番にレーザ光による測定が行われる。そして、個々のサンプルから発せられた散乱光と蛍光の強度が記録される。
散乱光は、入射するレーザに対して1°~10°の方向に射出される前方散乱光(FSC: forward scatter)と、90°の方向に反射する側方散乱光(SSC:side scatter)の2種類が存在する。前方散乱光の強度はサンプルの大きさの情報を反映する。側方散乱光強度はサンプル内部構造の複雑さを反映する。
細胞表面におけるタンパク質発現計測の場合、細胞はフローサイトメトリによる計測前に観測対象となる表面抗原が蛍光標識抗体でラベルされている。そして、ユーザは、蛍光標識抗体が発する蛍光によりタンパク質発現の有無や発現強度を細胞毎に知ることができる。
このように、フローサイトメトリでは個々のサンプルの大きさ・内部構造・タンパク質発現等のパラメータを得ることができる。そして、ユーザは、それらのパラメータでサンプル集団を分類し、集団毎のポピュレーションを知ることができる。
従来のフローサイトメトリではサンプル等のサンプルを整列させる手法として流体力学的絞り込み効果が用いられてきた。流体力学的絞り込み効果はフローセル内部で、外側にシース液の流れ、内側にサンプル溶液の流れが形成されることによりサンプルが整列する効果である。
一方で、流体力学的絞り込み効果とは方式の異なるフローサイトメトリとしてキャピラリフローサイトメトリがある。キャピラリフローサイトメトリでは、サンプルより少し大きい径を有するキャピラリの中にサンプル溶液を流通させる。これにより、個々のサンプルが整列し、サンプル毎に計測が行われる。キャピラリフローサイトメトリはシース液が不要であり、且つ廃液も少ないという利点がある。加えて、キャピラリフローサイトメトリはサンプルの流通を並列化することにより、サンプル計測をN倍化することが比較的容易である。そのため、キャピラリフローサイトメトリは、特に臨床応用において多数のサンプルを短い時間で測定するアプリケーションでの活用が期待されている。
非特許文献1には、並列化したキャピラリフローサイトメトリにおいて、超音波を用いることでキャピラリ中の細胞を精度高く整列させることが記載されている。

キャピラリフローサイトメトリにおいて、装置の小型化や、部品点数削減のためにはキャピラリを狭い空間に集積させることが望ましい。しかしながら、キャピラリを狭い空間に集積させると、隣接するキャピラリによる前方散乱光と、本来計測したい前方散乱光との区別が困難であるという課題がある。つまり、隣接するフローサイトメトリ同士で信号が混在する。
前記した通り、入射するレーザに対して1°~10°の方向に射出される前方散乱光はサンプルの大きさを反映する重要な情報であるため、前方散乱光が混在しないようにする工夫が必要である。
例えば、非特許文献1では、前記したように並列化したキャピラリフローサイトメトリにおいて、超音波を用いることによってキャピラリ中の細胞を精度高く整列させることに成功している。しかし、この中でも1°~10°の方向に散乱される前方散乱光は検出できておらず、前方散乱光の代わりに25°の後方散乱光データが取得されている。つまり、非特許文献1に記載の技術では、前方散乱光の混在を防止することはできない。
このような背景に鑑みて本発明がなされたのであり、本発明は、キャピラリを狭い空間に集積させても、前方散乱光の混在を生じさせないことを課題とする。
前記した課題を解決するため、本発明は、複数のキャピラリにレーザ光を照射する光源部と、前記キャピラリに対し、前記レーザ光が照射された側とは反対側に散乱する前方散乱光を受光する受光部と、光を遮蔽するレーザブロッカと、を備えるフローサイトメータであって、前記レーザブロッカとして、前記レーザ光が入射する側における前記キャピラリの整列面を含む前記光源部に近い位置、かつ、前記光源部の方向に前記キャピラリの軸中心を投影した位置からずれた位置に配置される第1のレーザブロッカと、前記キャピラリと、前記受光部との間、かつ、キャピラリ同士の間に設置される第2のレーザブロッカと、前記受光部の受光面において、前記受光部の方向に前記キャピラリの軸中心を投影した位置に設置される第3のレーザブロッカと、を有する。

本発明によれば、キャピラリを狭い空間に集積させても、前方散乱光の混在を生じさせないことが可能である。
本発明の実施形態について、図面を用いて説明する。ただし、本発明は以下に示す実施形態の記載内容に限定して解釈されるものではない。また図面等において示す各構成の位置、大きさ、形状、範囲などは、発明の理解を容易にするため、実際の位置、大きさ、形状、範囲などを表していない場合がある。このため、本発明は、必ずしも、図面等に開示された位置、大きさ、形状、範囲などに限定されない。
また、本実施形態において、フローサイトメータ1は、フローサイトメトリの1つであるキャピラリフローサイトメトリの技術を用いた装置である。
[第1実施形態]
図1は、本実施形態に係るフローサイトメータ1の構成を示す模式図である。また、図2は、レーザブロッカ110の配置位置を示す図である。

フローサイトメータ1は、光源部であるライトシート121、複数のキャピラリ122、受光部であるラインセンサ123を備える。ライトシート121はレーザ光141を発するものである。ちなみに、図1は、レーザ光141の入射方向に対して90度の方向からフローサイトメータ1を見た図である。また、キャピラリ122は、サンプル131の流通方向に対して直交する面による断面を示しており、サンプル131の流通方向は、紙面奥行き方向から紙面手前方向となる。サンプル131はキャピラリ122の内部を流れる液体とともにキャピラリ122の内部を流通する。なお、キャピラリ122の内部を流れる液体は、図示しないポンプの吐出力によって流通する。なお、キャピラリ122は石英、ソーダ石灰、ホウケイ酸といったガラスもしくはポリカーボネートといった樹脂によって構成されている。
ライトシート121は、複数のキャピラリ122にレーザ光141を照射するものである。また、ラインセンサ123は、キャピラリ122に対し、レーザ光141が照射された側とは反対側に散乱する前方散乱光142を受光するものである。なお、反対側とは、受光部であるラインセンサ123の側である。
フローサイトメータ1には、光を遮蔽するレーザブロッカ110が設けられている。レーザブロッカ110として、第1のレーザブロッカ111、第2のレーザブロッカ112、第3のレーザブロッカ113の3種類が設けられている。第2のレーザブロッカ112、第3のレーザブロッカ113は、隣接するキャピラリ122に由来する前方散乱光142をブロックするために設けられている。
図2に示すように、第1のレーザブロッカ111は、レーザ光141が入射する側におけるキャピラリの整列面151aを含むライトシート121に近い位置に配置される。そして、第1のレーザブロッカ111は、ライトシート121の方向にキャピラリ122の軸中心122aを投影した位置(矢印171)からずれた位置に配置される。具体的には、第1のレーザブロッカ111は、レーザ光141が入射する側におけるキャピラリ122の整列面151aに設置される。なお、図2に示す例では、第1のレーザブロッカ111は、レーザ光141が入射する側におけるキャピラリ122の整列面151aに設置されているが、これに限らない。例えば、キャピラリ122の整列面151aよりライトシート121に近い位置(整列面151aより上方)に設置されてもよい。これは、例えば、透明部材151の厚みを、キャピラリ122の外径より厚くすることで可能となる。
また、図2に示すように、第2のレーザブロッカ112は、キャピラリ122と、ラインセンサ123との間、かつ、キャピラリ122同士の間に設置される。具体的には、第2のレーザブロッカ112は、レーザ光141が入射する側におけるキャピラリ122の整列面151aからラインセンサ123の受光面123aまで設置される。
そして、図2に示すように、第3のレーザブロッカ113は、ラインセンサ123の受光面123aにおいて、ラインセンサ123の方向にキャピラリ122の軸中心122aを投影した位置(矢印172)に設置される。
また、レーザブロッカ110は、ポリカーボネート、アクリル、ポリプロピレン等の樹脂や、ガラス、アルミ等の金属で構成される。なお、レーザブロッカ110には反射防止処理を施していることが望ましい。この処理により、レーザ光141は乱反射せずレーザブロッカ110に吸光される。もし、反射防止処理がない場合、乱反射による迷光がラインセンサ123で光検出される可能性があるため注意が必要である。反射防止処理の例としては,レーザブロッカ110の着色(黒色等),表面に微細な凹凸構造の付与,誘電体膜の表面積層等が挙げられる。なお、キャピラリ122の軸方向における、それぞれのレーザブロッカ110のサイズ(図1、図2における奥行き方向のサイズ)は、レーザ光141及び前方散乱光142をブロックできるサイズである。
キャピラリ122は、透明部材151に埋設されている。さらに、第2のレーザブロッカ112は、透明部材151及び透明部材152に埋設されている。透明部材151、152は、透明性の高い部材であり、ポリカーボネート、アクリル、ポリプロピレン等の樹脂や、ガラス等で構成される(有機材料や無機材料を問わず透明性の高い部材で構成)。なお、図1に示す例では、透明部材151及び透明部材152の2つが設けられているが、透明部材151及び透明部材152の部分が1つの透明部材で構成されてもよい。
図1に示す例では、キャピラリ122の径は30μmとする。そして、それぞれのキャピラリ122は、30μm間隔で並列に配置されている。ただし、キャピラリ122の径及びキャピラリ122の配置間隔は、30μmに限らない。
また、前方散乱光142は、キャピラリ122の直下方向に対して1°~10°の範囲でサンプル131によって散乱される光である。ちなみに、本実施形態ではライトシート121の方向を上方とし、ラインセンサ123の方向を下方とする。また、前方散乱光142が散乱する範囲(1°~10°)は、おおよその値である。図1に示す構成の場合、ラインセンサ123に到達した際における前方散乱光142の幅は、およそ30μmとなる。
また、受光部としてラインセンサ123が配置されている。図1に示す例では、前方散乱光142の強度が、透明部材152によって前方散乱光142が減衰しすぎないうちに前方散乱光142がラインセンサ123に到達できるよう、キャピラリ122から180μmの距離にラインセンサ123が配置されている。ただし、ラインセンサ123とキャピラリ122の距離は180μmに限らない。ラインセンサ123は、前方散乱光142を受光すると信号を処理装置200へ送る。ちなみに、キャピラリ122とラインセンサ123と間の距離が長くなると、前方散乱光142の幅は広くなるので、ラインセンサ123に到達する前方散乱光142の単位面積当たりの強度は弱くなる。
図3A及び図3Bは、フローサイトメータ1をレーザ光141の入射方向から見た図である。なお、図1は、図3A及び図3BをA-A矢視方向から見た図である。なお、図3A及び図3Bにおいて、レーザブロッカ110は図示省略されている。また、図1を適宜参照し、図1において説明済みの要素については説明を適宜省略する。
図3Aは、直線状のキャピラリ122のそれぞれが独立して複数配置されている例である。つまり、図3Aに示すフローサイトメータ1では、それぞれのキャピラリ122において、キャピラリ122毎に導入口と排出口が設けられている(図3Aに示す矢印)。また、図3Bは、キャピラリ122が、蛇行している経路を有する1本のキャピラリ122である例を示している。図3Bは、1つのキャピラリ122が蛇行することによって、連続したひとつの流路を形成している例である。すなわち、図3Bに示すフローサイトメータ1では、1つの導入口と1つの排出口を有している(図3Bに示す矢印)。なお、図3Bにおいて、キャピラリ122の蛇行回数は5となっているが(U字部分は5つとなっているが)、これ以外の蛇行回数も可能である。
図3Aに示す例では、複数のキャピラリ122に対してレーザ光141が照射されるようライトシート121が設けられている。また、図3Bでは、キャピラリ122の複数個所にレーザ光141が照射されるようライトシート121が設けられている。なお、交点161については後記する。
レーザ光141は、キャピラリ122に対し図3A、図3Bの紙面手前方向から紙面奥側方向に向けて照射される。そして、それぞれのキャピラリ122に由来する前方散乱光142や、側方散乱光144(図4参照)、蛍光143(図4参照)が検出される。レーザ光141の照射は、一般的なレーザ光141をキャピラリ122毎に照射してもよいが、フローサイトメータ1の簡略化を達成するためには、ライトシート121のように面で一度にすべてのキャピラリ122に照射する方法が望ましい。このように、光源としてライトシート121が用いられることで、1つの光源(ライトシート121)で並列化したキャピラリ122のすべてに同一波長のレーザ光141を照射することができる。なお、一般的なレーザ光141とは、1つの照射ポイントにピンポイントでレーザ光141を照射するものである。
サンプル131の測定は、サンプル131が、それぞれのキャピラリ122においてライトシート121の照射部分を通過したときに実施される。
(蛍光143及び側方散乱光144の取得)

図4において、サンプル131(図1参照)は、キャピラリ122の内部を白抜き矢印方向に流れるものとする。
レーザ光141がキャピラリ122の内部を流通しているサンプル131に照射されると、サンプル131から前方散乱光142(図1参照)の他に、蛍光143及び側方散乱光144が発せられる。
フローサイトメータ1は、蛍光143を取得するための構成として集光用レンズ311、レーザ用ノッチフィルタ312、回折格子313、2DCCDカメラ314を備える。サンプル131から発した蛍光143は、集光用レンズ311、レーザ用ノッチフィルタ312、回折格子313を介して2DCCDカメラ314で受光される。2DCCDカメラ314は、蛍光143を受光すると、信号を処理装置200へ送る。
また、フローサイトメータ1は、側方散乱光144を取得するための構成として、集光用レンズ321、レーザ用バンドパスフィルタ322、ラインセンサ323を備える。サンプル131によって散乱する側方散乱光144は、集光用レンズ321、レーザ用バンドパスフィルタ322を介してラインセンサ323で受光される。ラインセンサ323は、側方散乱光144を受光すると信号を処理装置200へ送る。
フローサイトメータ1は、図1、図3A、図3Bに示す構成によって前方散乱光142を取得するとともに、図4に示す構成によって蛍光143及び側方散乱光144を取得する。
即ち、図4に示す構成を併せ持つことにより、複数のキャピラリ122に由来する前方散乱光142とともに、蛍光143や側方散乱光144をまとめて測定、解析することが可能である。
第1実施形態に示すフローサイトメータ1によれば、キャピラリ122を並列化したフローサイトメータ1において、図2を参照して説明した場所に。3種類のレーザブロッカ110が配置される。
これにより、隣接するキャピラリ122に由来する前方散乱光142の影響を受けることなく、それぞれのキャピラリ122に由来する前方散乱光142の取得が可能になる。つまり、隣接するキャピラリ122に由来する信号の影響を受けずにキャピラリ122毎にサンプル131に由来する前方散乱光142を取得することができる。これにより、キャピラリ122の間の距離を小さくすることができるため、キャピラリ122の集積化が可能となる。
さらに、図3Bに示すように、キャピラリ122が蛇行することにより、個々のサンプル131に対して複数回ずつ前方散乱光142、側方散乱光144、蛍光143の測定を行うことが可能となる。
[第2実施形態:フローサイトメータ信号解析方法]
続いて、図5A~図9を参照して、フローサイトメータ1によってラインセンサ123、323、2DCCDカメラ314から取得した信号の解析方法を説明する。以降の説明において、図1~図4を適宜参照する。

そして、以降の説明では、図3Bに示すような蛇行流路として並列化したキャピラリ122による測定結果を用いることとする。蛇行流路として並列化したキャピラリ122を用いることにより、同一のサンプル131を複数回測定することができる。図5A~図9では、このようなフローサイトメータ1による測定結果を用いたゲーティングの例を示す。これにより、図5A~図9を参照してサンプル集団401の境界を明瞭にする方法を説明する。
第2実施形態では、図3Bに示すような蛇行するキャピラリ122中をサンプル131が流通することによって、同一のサンプル131について交点161毎に測定が行われる。そして、測定結果の平均値を基にサンプル集団401(図6B参照)が抽出される。このように、測定結果の平均値を基にサンプル集団401が抽出されることで、サンプル集団401の境界が明瞭になることを第2実施形態で示す。
このように、測定結果の平均値を基にサンプル集団401を抽出する際には、サンプル131を1つのみ流通させるキャピラリ122を有するキャピラリフローサイトメトリの技術が有用である。特に、図3Bのように蛇行するキャピラリ122を有するフローサイトメータ1が有用である。そして、図3Bのように蛇行するキャピラリ122を有するフローサイトメータ1の小型化や、部品点数削減を行う際に、図1に示す、3種類のレーザブロッカ110が有用となる。
図5Aは、それぞれのサンプル131(本実施形態に示す例では細胞)を1回ずつ測定した際における検出強度を示す図である。また、図5Bは、図5Aに示す結果を用いてゲーティングを行った結果を示す例である。
前記したように、キャピラリ122の配置は図3Bに示すものであり、光源としてライトシート121が用いられているものとする。また、前記したように、個々のサンプル131は図3Bに示すキャピラリ122とライトシート121の交点161において6回測定される。この際、隣接するキャピラリ122に由来する前方散乱光142の干渉を避けるため、図1に示すようにレーザブロッカ110が配置される。図5A及び図5Bでは、それぞれの交点161で計測された結果が、そのまま示されている。
3種類のレーザブロッカ110(第1のレーザブロッカ111、第2のレーザブロッカ112、第3のレーザブロッカ113)の配置は、図1で説明した通りである。
そして、図1に示すように、ラインセンサ123は、サンプル131によって前方1°~10°の方向に散乱する前方散乱光142を受光する。また、図4に示す構成により、処理装置200は、前方散乱光142とともに、蛍光143及び側方散乱光144による信号も取得する。
図5Aにおいて、横軸は、サンプル131(本実施形態に示す例では細胞)、縦軸は検出強度を示している。検出強度は、蛍光強度もしくは散乱光強度である。なお、強度とは、ラインセンサ123、2DCCDカメラ314、及び、ラインセンサ323が出力する信号強度である。また、以降において、散乱光とは、前方散乱光142、又は、側方散乱光144を指すものとする。つまり、散乱光強度とは、前方散乱光142の信号強度、又は、側方散乱光144の信号強度である。
フローサイトメータ1の測定では、図5Bに示すようにサンプル集団401をプロット点402でゲーティングすることが重要である。ゲーティングとは、サンプル集団401をクラスタリングすることである。
1つのサンプル131に対し、1度の測定しか行わないと、図5Aに示すように、検出強度にばらつきが生じる。このようなばらつきは、サンプル131の測定時におけるレーザ光141に対するサンプル131の位置ブレや、サンプル131の向きの違いが影響していると考えられる。
図5Bは、図5Aと同様の手法で、1つのサンプル131について測定を1度行い、その結果をプロットしたものである。図5Bにおいて、横軸及び縦軸は、蛍光強度もしくは散乱光強度を示している。つまり、図5Bにおいて、横軸及び縦軸は、蛍光強度、前方散乱光142の信号強度、及び、側方散乱光144の信号強度のうち、いずれかである。
また、図5Bに示す図において、プロット点402の1つ1つはサンプル131(本実施形態で示す例では細胞)を示している。そして、図5Bではゲーティングが行われることにより、類似している性質をもつサンプル131をサンプル集団401としてグループ分け(クラスタリング)している。
図5Bに示す図では、サンプル集団401の一部が重複している。このような状態は、サンプル131の測定を1度しか行わない場合、しばしば生じる。このような、サンプル集団401の重複は、図5Aに示すような検出強度のばらつきによるものと考えられる。
図6Aは、図3Bに示すフローサイトメータ1を使用することにより、1つのサンプル131を交点161(図3B参照)毎に測定した結果を示す図である。本実施形態に示す例において、1つのサンプル131に対する測定回数は6回となる。
図6Aでは、サンプル131に交点161毎に測定された結果を平均し、その平均値と、標準偏差が示されている。なお、平均値及び標準偏差は、測定結果のうち、ユーザが外れ値を除外した上で、処理装置200が平均値及び標準偏差を算出してもよい。
図6Bは、図6Aで示す方法で算出した平均値をプロットし、サンプル集団401のゲーティングを行った例を示す図である。
図6Bにおいても、図5Aと同様、プロット点402の1つ1つはサンプル131を示している。しかし、図6Bでは、図3Bに示すフローサイトメータ1を用いて、同じサンプル131を複数回測定し、その結果を平均化し、その平均値をプロットしている。その上で、プロット点402のゲーティングが行われている。
図6Bに示すように、同じサンプル131を複数回測定し、その結果を平均化した値を用いることにより、サンプル集団401が狭い範囲となる。これにより、サンプル集団401の境界が明瞭になる(サンプル集団401間で重複が生じない)。このように、同じサンプル131を複数回測定し、その結果を平均化した値を用いることにより、ゲーティングの精度を向上させることができる。
次に、実際に同じサンプル131を複数回測定することにより、信号のばらつきが抑えられることを検証したデータを図7~図9に示す。
図7~図9のそれぞれは、33個の蛍光ビーズを水の入った微細な1つの流路に整列させ、それぞれにビーズに由来する蛍光強度を蛍光顕微鏡で測定した結果である。なお、図7~図9の結果は、キャピラリ122を模した流路に蛍光ビーズを整列させて行われた実験によって得られたものである。つまり、実際にフローサイトメータ1のキャピラリ122に蛍光ビーズを流通させたものではない。
測定は、10回繰り返し実施され、それぞれの測定の間で蛍光ビーズを流路内で順番が変わらぬように静かに動かした。ただし、蛍光ビーズは流れていない状態で、動かされている。なお、図7~図9では、蛍光143が用いられているが、蛍光143を前方散乱光142に置き換えることで、前方散乱光142についても同様の議論を行うことができる。
33個の蛍光ビーズは、同じサンプル集団401に属すサンプル131を模している。それぞれの蛍光ビーズは、基本的には同程度の蛍光強度を示すが、蛍光ビーズ毎ごとに多少のばらつきがある。また、蛍光ビーズを流路内で動かすことは、以下のことを模している。つまり、フローサイトメータ1において、キャピラリ122を移動しているサンプル131が、レーザ光141に対して測定回毎に微妙に異なる位置や角度で測定されることを模している。
図7は、1~10回までの各測定回における、33個の蛍光ビーズに由来する蛍光143の強度(蛍光強度)をプロットした箱ひげ図である。図7に示すひとつひとつのプロット点411は、蛍光ビーズによる蛍光強度を表している。ただし、同じ数値の結果が複数存在するため、図7におけるプロット数は33より少なく示されている。なお、箱ひげ図における「×」印は平均値を示している。
一方、図8は、それぞれの蛍光ビーズについて10回の測定値を平均した結果を用いた箱ひげ図である。図8において、ひとつひとつのプロット点412は、それぞれの蛍光ビーズにおける蛍光強度の10回平均値を表している。図7と同様、同じ数値の結果が存在するため、図7におけるプロット数は10より少なく示されている。
図7及び図8に示す結果を比較すると、蛍光強度を複数回測定に基づく平均値とした場合(図8)、単回測定の場合(図7)よりも、蛍光ビーズ間の蛍光強度のばらつきが小さくなっていることがわかる。
また、図9は、図7及び図8と同様の実験において、サンプル131(蛍光ビーズ)に由来する蛍光強度のばらつきが、測定回を重ねるごとに減少していく様子を示した結果である。
図9において、縦軸には、33個の蛍光ビーズ間での蛍光強度のばらつきとして変動係数(CV:Coefficient of Variation)が示されている。横軸は、ばらつきの計算に用いた積算測定回数を示している。例えば、積算測定回数が3回の場合は、それぞれの蛍光ビーズについて1回目~3回目の測定における蛍光強度の平均値を用いて、蛍光ビーズ間での蛍光強度のばらつきが計算されている。
図9によれば、6回程度の測定により、サンプル131に由来する蛍光強度のばらつきが最小化している。前記したように、フローサイトメータ1のサンプル131(細胞)同様、それぞれの蛍光ビーズに元々備わっている、蛍光強度の差(ばらつき)がある。そのため、測定回数を増やし続けても、ばらつき抑制の効果は頭打ちとな。
つまり、図7~図9に示す実験では、フローサイトメータ1で同一のサンプル131を複数回測定し、その平均値をサンプル131それぞれの信号値として扱うことが模擬されている。これにより、レーザ光141の照射に対するサンプル131の位置ブレや、向きの違いによるデータばらつきを抑えることができる。これにより、フローサイトメータ1の解析においてサンプル集団401の分離を明瞭化することができる。

処理装置200は、演算装置201、記憶装置202、入力装置203、出力装置204、通信装置205、メモリ210等を備える。
演算装置201は、CPU(Central Processing Unit)や、GPU(Graphic User Interface)等で構成される。記憶装置202は、HDD(Hard Disk Drive)や、SSD(Solid State Drive)等で構成される。入力装置203は、キーボードや、マウス等で構成される。出力装置204は、ディスプレイ等で構成される。通信装置205は、図1、図4で示すラインセンサ123、323や、2DCCDカメラ314から信号を受け取る。メモリ210は、RAM(Random Access Memory)等の揮発性メモリで構成される。
そして、記憶装置202に格納されているプログラムがメモリ210にロードされ、ロードされたプログラムが演算装置201によって実行される。これにより、計算部211、ゲーティング部212、判定部213が具現化する。
計算部211は、図5A~図9で説明したように、N回測定された結果、得られる測定値の平均値を算出する。ゲーティング部212は、計算部211が計算した平均値を基に測定結果をゲーティングし、サンプル集団401を抽出する。判定部213は、隣接するサンプル集団401が明瞭に分かれているか否かを判定する。

図11は、本実施形態に係るフローサイトメータ信号解析方法の手順を示すフローチャートである。適宜、図1、図10を参照する。

まず、それぞれのサンプル131がフローサイトメータ1でN回(図5A~図9に示す例では、N=6あるいは10)測定される(S1)。ステップS1は、複数のサンプル131に対し、それぞれのサンプル131について複数回測定を行う測定ステップである。
続いて、計算部211は、それぞれのサンプル131について、散乱光強度及び蛍光強度のN回平均値を算出する(S2)。ステップS2は、測定ステップ(S1)による測定結果に対して統計処理を行う統計処理ステップである。ステップS2において、計算部211は、それぞれのサンプル131について、前記複数回測定の結果の値の平均値を算出する。
そして、ゲーティング部212は、ステップS2で算出された、それぞれのサンプル131のN回平均値を図5B、図6Bに示すようなプロット図にプロットする。そして、ゲーティング部212は、プロット点402(図5B、図6B参照)に対してゲーティング(クラスタリング)を行うことで、サンプル集団401を抽出する(S3)。ステップS3は、統計処理ステップ(S2)の結果を基に、サンプル集団401を抽出するゲーティングを行うゲーティングステップである。ステップS3において、ゲーティング部212は、k-means法や、サポートベクトルマシン等を用いてゲーティングを行う。ステップS3において、ゲーティング部212は、平均値に基に、サンプル集団401を抽出する
続いて、判定部213が、ステップS3で抽出されたサンプル集団401の境界は明瞭であるか否かを判定する(S4)。判定部213は、隣接するサンプル集団401の境界が重複していないか否かを判定することで、サンプル集団401の境界は明瞭であるか否かを判定する。判定部213は、隣接するサンプル集団401の境界が重複していない場合、サンプル集団401の境界は明瞭であると判定する。
サンプル集団401の境界が明瞭である場合(S4→Yes)、解析は終了となる。
サンプル集団401の境界が明瞭ではない場合(S4→No)、N回測定値の中から外れ値の除外が行われる(S5)。外れ値の除外は、ユーザがプロット図を基に手作業で行ってもよいし、マハラノビス距離や、箱ひげ図等を用いてゲーティング部212が行ってもよい。
ステップS5で外れ値の除外が行われた後、処理装置200はステップS2へ処理を戻す。これにより、処理装置200は、外れ値の除外が行われたデータを用いてN回平均値、ゲーティングを行う。複数回測定の平均値によってサンプル集団401が抽出されることにより、ステップS5における外れ値の除外の回数を低減することができる。
第2実施形態によれば、複数回の測定結果を解析することにより、単回測定の場合と比較してプロット点402(図5B及び図6B参照)における集団の広がりを小さく抑えることができる。この結果、ゲーティングにおいてサンプル集団401の境界を明瞭にすることができる。
本発明は前記した実施形態に限定されるものではなく、様々な変形例が含まれる。例えば、前記した実施形態は本発明を分かりやすく説明するために詳細に説明したものであり、必ずしも説明したすべての構成を有するものに限定されるものではない。また、ある実施形態の構成の一部を他の実施形態の構成に置き換えることが可能であり、ある実施形態の構成に他の実施形態の構成を加えることも可能である。また、各実施形態の構成の一部について、他の構成の追加・削除・置換をすることが可能である。
また、フローサイトメータ1は、アナライザでもセルソータでもよい。
また、前記した各構成、機能、計算部211~判定部213、記憶装置202等は、それらの一部又はすべてを、例えば集積回路で設計すること等によりハードウェアで実現してもよい。また、図10に示すように、前記した各構成、機能等は、CPU等のプロセッサがそれぞれの機能を実現するプログラムを解釈し、実行することによりソフトウェアで実現してもよい。各機能を実現するプログラム、テーブル、ファイル等の情報は、HDに格納すること以外に、メモリや、SSD(等の記録装置、又は、IC(Integrated Circuit)カードや、SD(Secure Digital)カード、DVD(Digital Versatile Disc)等の記録媒体に格納することができる。
また、各実施形態において、制御線や情報線は説明上必要と考えられるものを示しており、製品上必ずしもすべての制御線や情報線を示しているとは限らない。実際には、ほとんどすべての構成が相互に接続されていると考えてよい。

- 複数のキャピラリにレーザ光を照射する光源部と、
前記キャピラリに対し、前記レーザ光が照射された側とは反対側に散乱する前方散乱光を受光する受光部と、
光を遮蔽するレーザブロッカと、
を備えるフローサイトメータであって、
前記レーザブロッカとして、
前記レーザ光が入射する側における前記キャピラリの整列面を含む前記光源部に近い位置、かつ、前記光源部の方向に前記キャピラリの軸中心を投影した位置からずれた位置に配置される第1のレーザブロッカと、
前記キャピラリと、前記受光部との間、かつ、キャピラリ同士の間に設置される第2のレーザブロッカと、
前記受光部の受光面において、前記受光部の方向に前記キャピラリの軸中心を投影した位置に設置される第3のレーザブロッカと、
を有するフローサイトメータ。 - 前記第1のレーザブロッカは、前記レーザ光が入射する側における前記キャピラリの整列面に設置され、
