Abstract
Die vorliegende Erfindung beschreibt Enzyme und enzymbasierte Verfahren zur Spaltung von aromatischen Carbonaten. Die Enzyme sind besonders gut zur Spaltung von Carbonaten auf Basis von Bisphenol A geeignet und können deswegen zur Wiedergewinnung von Bisphenol A aus Polymeren genutzt werden.
Description
Neuartige Enzyme zur Spaltung aromatischer Carbonate
Die vorliegende Erfindung beschreibt Enzyme und enzymbasierte Verfahren zur Spaltung von aromatischen Carbonaten. Die Enzyme sind besonders gut zur Spaltung von Carbonaten auf Basis von Bisphenol A geeignet und können deswegen zur Wiedergewinnung von Bisphenol A aus Polymeren genutzt werden.
Durch die Reaktion von Phosgen und Bisphenol A können Polycarbonate erhalten werden, die sich durch hohe Transparenz und Schlagzähigkeit auszeichnen. Wegen dieser Eigenschaften haben derartige Polycarbonate weite Anwendung als Glasersatz, im Fahrzeugbau und als Hüllen von elektronischen Geräten, z.B. von Mobiltelefonen, gefunden. Entsprechend hoch ist die Jahresproduktion dieser Polymere.
Da Polycarbonate thermoplastisch sind, ist grundsätzlich ein Recycling durch Aufschmelzen möglich. Erschwert wird dies aber durch den Umstand, dass Polycarbonate häufig mit Additiven versetzt werden, um ihre anwendungstechnischen Eigenschaften zu verbessern. Eine Abtrennung dieser Additive kann wünschenswert sein, ist technisch aber schwierig. Zudem führt wiederholtes Aufschmelzen eines Polycarbonats zu einer teilweisen Depolymerisierung, wobei Bisphenol A freigesetzt werden kann. Wegen der gesundheitsschädlichen Eigenschaften von Bisphenol A ist dies unerwünscht und beschränkt die Anzahl der Recycling-Zyklen des Polymers. Wenn Polycarbonate in Mischungen mit anderen Thermoplasten verwendet werden, stellt sich bei der Wiederverwendung zusätzlich das Problem der Trennung dieser Gemische, wenn für die Weiterverwendung reine Polymere gewünscht sind.
Aus diesem Grund besteht ein erheblicher Bedarf nach weiteren Verfahren, mit denen Polycarbonate rezykliert werden können. Ein von anderen Polymeren gut bekannter Ansatz ist der gezielte Abbau des Polymers mit Hilfe von Enzymen, um unter schonenden Reaktionsbedingungen gut definierte Chemierohstoffe wiederzugewinnen. Dies ist z.B. bei Polyurethanen (EP 3 587 570) und Polyethylenterephthalat bereits gezeigt worden (Tournier (2020) „An engineered PET depolymerase to break down and recycle plastic bottles", Nature 580: 216-219. Auch für aliphatische Polycarbonate ist dies möglich (Suyama & Tokiwa (1997), „Enzymatic degradation of an aliphatic polycarbonate, poly(tetramethylene carbonate)", Enzyme and Microbial Technology, 20: 122-126").
Für die wegen ihrer umfangreichen Anwendung technisch und wirtschaftlich sehr wichtigen aromatischen Polycarbonate, insbesondere solche, die auf Bisphenol A basieren, sind technisch sinnvolle enzymatische Abbauverfahren bisher noch nicht beschrieben. Die Freisetzung von Bisphenol A aus solchen Verbindungen durch ein Bakterium, Pseudoxanthomonas sp., ist beschrieben worden (Yue et. al. (2021), „Biodegradation of bisphenol-A polycarbonate plastic by Pseudoxanthomonas sp.
strain NY3600" Journal of Hazardous Materials, 416: 125775). Allerdings wurde nicht gezeigt, welche Enzyme für den Abbau verantwortlich sind. Es ist noch nicht einmal klar, ob es sich ein einziges Enzym oder um einen Enzymkomplex handelt. Für die technische Anwendung eines Recyclingverfahrens sind klar definierte und isolierte Enzyme aber wegen ihrer besser kontrollierbaren Eigenschaften vorzuziehen. Es wurde beschrieben (Artham et al. (2011), „Mechanistic investigations of lipase- catalyzed degradation of polycarbonate in organic solvents", Enzyme and Microbial Technology, 48: 71-79), dass Lipasen wie sie z.B. in Candidada rugosa oder Candida antarctica gefunden werden, Bisphenol A aus Polycarbonaten freisetzen. Es hat sich allerdings gezeigt, dass diese Reaktion von den dort eingesetzten Enzymen nicht in wässrigem Medium katalysiert wird. Der enzymatische Abbau von aromatischem Polycarbonat in organischen Lösungsmitteln wurde auch an anderer Stelle berichtet (Sivalingam & Madras (2004) „Dynamics of Lipase Catalyzed Enzymatic Degradation of Poly(bisphenol- A carbonate), Journal of Polymer Science, 91: 2391-2396).
In der der vorliegenden Erfindung zugrunde liegenden Studie wurden erstmals Enzyme identifiziert, welche die Freisetzung von Bisphenol A aus aromatischen Polycarbonaten in wässrigen Medien ermöglichen. Alle Enzyme mit dieser katalytischen Fähigkeit weisen ein hohes Maß an Sequenzidentität auf. Alle getesteten Enzyme aus dieser Gruppe zeigten - wenn auch in unterschiedlichem Ausmaß - die gewünschte Aktivität.
Um die Aminosäurepositionen zu identifizieren, die für die Aktivität entscheidend sind, wurde eine Konsensussequenz der Aminosäuresequenzen aller untersuchten Enzyme erstellt. Diese ist durch SEQ ID NO.: 1 definiert. Der Schwellenwert bei der Erstellung der Konsensussequenz wurde auf 100 % gesetzt, d.h. die in der Konsensussequenz dargestellten konservierten Positionen enthalten in allen untersuchten Enzymen dieselbe Aminosäure. Angesichts der großen Anzahl getesteter Enzyme ist davon auszugehen, dass es tatsächlich diese konservierten Positionen sind, die für die erfindungsgemäße Funktion verantwortlich sind. Demgegenüber wird davon ausgegangen, dass die mit „X" als Platzhalter für beliebige Aminosäuren gekennzeichneten Positionen funktionell irrelevant sind.
In einer ersten Ausführungsform betrifft die vorliegende Erfindung die Verwendung
(a) eines Enzyms, das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 1 oder eine Variante des vorgenannten Enzyms, die durch eine Aminosäuresequenz mit wenigstens 85 % Identität zu den in SEQ ID NO.: 1 definierten konservierten Aminosäuren definiert ist; oder
(b) eines Enzyms, das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit
wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist; zur Spaltung eines aromatischen Carbonats in einem wässrigen Medium.
Das aromatische Carbonat ist bevorzugt eine Verbindung gemäß Formel (I)
Wobei
RI ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Hydroxyl und beliebigen organischen Resten; und
R2 unabhängig von RI ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, einer Verbindung gemäß Formel (I) und beliebigen organischen Resten.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung befindet sich die Carbonatgruppe, an die R2 gebunden ist, in der para-Position, stärker bevorzugt, befinden sich beide Substituenten, RI und der Sauerstoff der Carbonatgruppe, in der para-Position. Dies ist in Formel (la) gezeigt.
Der Begriff „Spaltung" bezieht sich auf die hydrolytische Spaltung der in Formel (I) dargestellten Carbonatgruppe. Hierbei wird ein Diphenylmethan-Derivat als Alkohol und ein Derivat des Carbonats als Carbonsäure freigesetzt. Bei Spaltung der Carbonatgruppe am Sauerstoffatom, das R2 trägt, entsteht ein von R2 abgeleiteter primärer Alkohol und eine von Diphenylmethan abgeleitete Carbonsäure. Wenn R2 Wasserstoff ist, so wird Kohlensäure freigesetzt. Wenn RI eine Hydroxylgruppe ist, so wird Bisphenol A freigesetzt.
Ein „wässriges Medium" ist ein Lösungsmittelgemisch, das bezogen auf das Gesamtgewicht aller anwesenden Lösungsmittel zu wenigstens 50 Gew.-% aus Wasser besteht. Die Studie, die der vorliegenden Erfindung zu Grunde liegt, hat gezeigt, dass die Zugabe eines polaren organischen Lösungsmittels zum wässrigen Medium die Abbauleistung der Enzyme erhöht. Deswegen enthält das wässrige Medium in einer bevorzugten Ausführungsform der vorliegenden Erfindung zwischen 10 und 50 Gew.-% wenigstens eines polaren organischen Lösungsmittels, stärker bevorzugt 30 bis 50 Gew.-%, wobei sich die Massenanteile aller anwesenden Lösungsmittel inklusive des polaren organischen Lösungsmittels zu 100 % addieren. Es ist besonders bevorzugt, dass das wässrige Medium bezogen auf die Gesamtmasse aller anwesenden Lösungsmittel zu 80 Gew.-%, stärker bevorzugt zu 90 Gew.-% und am stärksten bevorzugt zu 95 Gew.-% aus Wasser und optional dem polaren organischen Lösungsmittel besteht. Ein „polares organisches Lösungsmittel" ist vorzugsweise ein organisches Lösungsmittel, das keine Carbonat- oder Estergruppen enthält und einen log P-Wert von höchstens 1,3 hat. Bevorzugte polare organische Lösungsmittel sind Aceton, Acetonitril, Dichlormethan, Ethylacetat, THF und DMF Besonders bevorzugt ist DMSO als polares organisches Lösungsmittel.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung liegt das erfindungsgemäß verwendete Enzym immobilisiert vor. Zur Immobilisierung sind alle dem Fachmann geläufigen Techniken wie Adsoption, kovalente Bindung and Trägermaterialien, Affinitätsimmobilisation und Einschlussimmobilisation geeignet.
Die Konzentration des aromatischen Carbonats, insbesondere des aromatischen Polycarbonats im wässrigen Medium beträgt vorzugsweise 1 mg/l bis 1.000 g/l, stärker bevorzugt 500 mg/l bis 500 g/l, noch stärker bevorzugt 100 mg/l bis 150 g/l und am stärksten bevorzugt 2 g/l bis 70 g/l.
Die Enzymkonzentration bei der Verwendung beträgt vorzugsweise 1 pg/ml bis 2.000 pg/ml, stärker bevorzugt 10 pg/ml bis 1.500 pg/ml, noch stärker bevorzugt 30 pg/ml bis 1.000 pg/ml und am stärksten bevorzugt 70 pg/ml bis 600 pg/ml. Konzentrationen von 120 pg/ml, 250 pg/ml und 500 pg/ml sind ebenfalls denkbar.
Bevorzugt handelt es sich bei der Verbindung gemäß Formel (I) um ein Polycarbonat. Der Begriff „Polycarbonat" bezeichnet vorzugsweise Polymere, die durch Reaktion wenigstens einer Verbindung ausgewählt aus der Gruppe bestehend aus Bisphenol A, Bisphenol S, Dihydroxydiphenylsulfid, Tetramethylbisphenol A, l,l-Bis(4-hydroxyphenyl)-3,3,5-trimethylcyclohexan (BPTMC) und 1,1,1- Tris(4-hydroxyphenyl)-ethan (THPE) mit Phosgen oder Diphenylcarbonat erhältlich sind. Derartige Polycarbonate haben vorzugsweise ein zahlenmittleres Molekulargewicht zwischen 10.000 und 50.000 g/mol, bevorzugt zwischen 18.000 und 40.000 g/mol. Optional enthalten derartige Polycarbonate geringe Anteile von tert-Butylphenol und/oder Cumylphenol. Dieser werden in der Polymerisation als Kettenabbrecher verwendet.
Bevorzugte Polycarbonate sind solche, die durch Reaktion von Bisphenol A mit Phosgen oder Diphenylcarbonat erhältlich sind. Hierbei ist die Anwesenheit weiterer als der oben genannten Verbindungen nicht ausgeschlossen, aber weniger bevorzugt. Es ist bevorzugt, dass Bisphenol A wenigstens 80 Gew.-%, bevorzugt wenigstens 90 Gew.-% und besonders bevorzugt wenigstens 95 Gew.-% des Gesamtgewichts aller hydroxyfunktionellen Aufbaukomponenten des Polycarbonats ausmacht. Derartige Polycarbonate werden vorzugsweise durch Formel (II) definiert.
Eine Verbindung gemäß Formel (II) wird typischerweise durch die Reaktion von Bisphenol A und Phosgen oder durch eine Schmelzkondensation von Bisphenol A und Diphenylcarbonat erhalten.
In einer weiteren bevorzugten Ausführungsform ist das aromatische Polycarbonat ein Copolymer aus einem der oben definierten aromatischen Polycarbonate und wenigstens einem Polyester. Bevorzugt ist es ein Copolymer aus einem Polycarbonat wie in Formel (II) definiert und wenigstens einem Polyester. Da die erfindungsgemäß verwendeten Enzyme in der Lage sind, Polyester ebenso abzubauen wie Polycarbonate, ist ein Abbau eines Polycarbonat-Polyester-Copolymers in seine monomeren Bestandteile möglich. Bevorzugte Polyester zum Aufbau derartiger Copolymere sind Polyethylenterephthalat, Polybutylenterephthalat, Polypentylenterephthalat und Polyhexylenterephthalat.
Um die Zugänglichkeit der Verbindung gemäß Formel (I), (la) oder des aromatischen Polycarbonats, insbesondere des Polycarbonats gemäß Formel (II), für das Enzym zu erhöhen, wenn diese Verbindung als Feststoff vorliegt, können alle gängigen Verfahren eingesetzt werden, insbesondere das Mahlen.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung liegt die Verbindung gemäß Formel
(I), (la) oder das aromatische Polycarbonat, insbesondere das aromatische Polycarbonat gemäß Formel
(II) als Bestandteil einer Mischung aus wenigstens zwei unterschiedlichen Polymeren vor. Als unterschiedlich werden hier Polymere verstanden, die durch unterschiedliche funktionelle Gruppe vernetzt sind. Die Verbindung gemäß Formel (I) oder (II) liegt also in dieser Ausführungsform zusammen mit wenigstem einem Polymer vor, das kein Polycarbonat ist. Bevorzugt liegt in dieser Ausführungsform ein Polycarbonat wie oben definiert als Gemisch mit wenigstens einem Polymer ausgewählt aus der Gruppe bestehend aus Polyestern, Styrol-Acrylnitril-Copolymer und Acrylnitril- Butadien-Styrol-Copolymer vor. Hierbei sind Polyethylenterephthalat und/oder Polybutylenterephthalat als Polyester bevorzugt. Es ist besonders bevorzugt, dass ein auf Bisphenol A basierendes Polycarbonat mit wenigstens einem der vorgenannten Polymeren als Gemisch vorliegt.
So ist es möglich, das Polycarbonat gezielt enzymatisch abzubauen, während das weitere oder die weiteren Polymere im Gemisch nicht angegriffen werden. Auf diese Art lassen sich aromatische Polycarbonate gezielt aus Polymergemischen entfernen. Umgekehrt werden auf diese Weise Polymere, die keine Polycarbonate sind, aufgereinigt. Dies ermöglicht die Rückgewinnung sortenreiner Polymere aus polycarbonathaltigen Polymergemischen.

* Da die Enzyme von kommerziellen Anbietern bezogen wurden, ist nicht mit absoluter Sicherheit zu sagen, ob ihre Aminosäuresequenz zu den aus öffentlichen Datenbanken bezogenen Sequenzen identisch waren.
Enzyme mit den an anderer Stelle in dieser Anmeldung definierten Sequenzidentitäten zu den durch SEQ ID NO.: 2 bis SEQ ID NO.: 10 definierten Enzymen bzw. zu der durch SEQ ID NO.: 1 definierten Konsensussequenz können durch Hinzufügung, Austausch oder Deletion von Aminosäuren erhalten werden. Solche Enzyme mit Aminosäuresequenzen die von SEQ ID NO.: 1 bis 10 abweichen, werden auch als „Varianten" bezeichnet". Jede erfindungsgemäße Variante ist vorzugsweise dadurch gekennzeichnet, dass sie die in Formel (I) oder (la) dargestellte Carbonatgruppe spalten kann. Dies wird vorzugsweise durch Freisetzung von Bisphenol aus einem durch die Reaktion von Bisphenol A und Phosgen oder durch Schmelzkondensation von Bisphenol A und Diphenylcarbonat erhaltenen Polycarbonats nachgewiesen.
In einer bevorzugten Ausführungsform wird ein Enzym verwendet, dessen Aminosäuresequenz wie folgt definiert ist: (i) Die zu den konservierten Aminosäurepositionen gemäß SEQ ID NO.: 1 homologen Aminosäurepositionen des Enzyms weisen wenigstens 90 %, stärker bevorzugt wenigstens 95 % Sequenzidentität zu den konservierten Aminosäurepositionen aus SEQ ID NO.: 1 auf und (ii) die zu den beliebigen Aminosäurepositionen aus SEQ ID NO.: 1 homologen Aminosäurepositionen weisen wenigstens 70 %, bevorzugt wenigstens 80 % Sequenzidentität zu den homologen Aminosäurepositionen einer der Aminosäuresequenzen ausgewählt aus der Gruppe bestehend aus SEQ ID NO.: 3, 8, 2, 4, 5, 6, 7, 9 und 10 auf. Bevorzugt weisen die beliebigen Aminosäurepositionen hierbei die vorgenannten Sequenzidentitäten zu SEQ ID NO.: 3 oder 8 auf.
Mit anderen Worten ist das Enzym in dieser Ausführungsform dadurch gekennzeichnet, dass seine funktionell bedeutsamen Aminosäurepositionen eine hohe Übereinstimmung mit der Konsensussequenz aufweisen, während die nicht konservierten und funktionell weniger relevanten Positionen mit größerer Sequenzabweichung von einem der in der dieser Anmeldung zugrunde liegenden Studie als aktiv identifizierten Enzymen abgeleitet ist.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung ist die weiter oben definierte Variante eines durch SEQ ID NO.: 2 definierten Enzyms durch eine Sequenzidentität von wenigstens 74 %, bevorzugt wenigstens 80 %, stärker bevorzugt wenigstens 85 %, noch stärker bevorzugt wenigstens 90 % und am stärksten bevorzugt wenigstens 95 % gekennzeichnet.
In einer weiteren bevorzugten Ausführungsform der vorliegenden Erfindung ist die weiter oben definierte Variante eines durch SEQ ID NO.: 7 definierten Enzyms durch eine Sequenzidentität von wenigstens 90 % und bevorzugt wenigstens 95 % gekennzeichnet.
In noch einer weiteren Ausführungsform betrifft die vorliegende Erfindung die Verwendung eines Enzyms, das eine Aminosäuresequenz hat wie in SEQ ID NO.: 3, 8, 2, 4, 5, 6, 7 , 9, 10, TI, 29 oder 30 definiert oder einer Variante eines der vorgenannten Enzyme mit wenigstens 85 % Sequenzidentität zu einer der vorgenannten Sequenzen zur Spaltung eines aromatischen Carbonats in einem wässrigen Medium. Alle oben gegebenen Definitionen für das aromatische Carbonat gelten auch für diese Ausführungsform.
In einer bevorzugten Ausführungsform betrifft die vorliegende Erfindung die Verwendung eines Enzyms, das eine Aminosäuresequenz hat wie in SEQ ID NO.: 3, 8, 7, 9, 5 oder 27 definiert oder einer Variante eines der vorgenannten Enzyme mit wenigstens 85 % Sequenzidentität zu einer der vorgenannten Sequenzen zur Spaltung eines aromatischen Carbonats in einem wässrigen Medium. Alle oben gegebenen Definitionen für das aromatische Carbonat gelten auch für diese Ausführungsform.
In einer besonders bevorzugten Ausführungsform betrifft die vorliegende Erfindung die Verwendung eines Enzyms, das eine Aminosäuresequenz hat wie in SEQ ID NO.: 3 oder 8 definiert oder einer Variante eines der vorgenannten Enzyme mit wenigstens 85 % Sequenzidentität zu einer der vorgenannten Sequenzen zur Spaltung eines aromatischen Carbonats in einem wässrigen Medium. Alle oben gegebenen Definitionen für das aromatische Carbonat gelten auch für diese Ausführungsform.
Eine Variante wird aus den erfindungsgemäßen Aminosäuresequenzen bevorzugt durch Hinzufügung, Deletion oder Austausch des jeweils definierten Anteils der im jeweiligen Enzym enthaltenen Aminosäuren erhalten. Basis für die Berechnung der Sequenzidentität ist bevorzugt jeweils die durch SEQ ID NO.: 3, 8, 2, 4, 5, 6, 7, 9, 10, TI, 29 oder 30 definierte Aminosäuresequenz. Dem Fachmann ist bewusst, dass Fusionsproteine von Enzymen mit anderen Proteinen gebildet werden, ohne dass dies die Aktivität des Enzyms beeinträchtigt. Deswegen umfasst der Begriff „Variante" auch solche von den durch SEQ ID NO.: 3, 8, 2, 4, 5, 6, 7, 9, 10, TI , 29 oder 30 definierten Polypeptiden abgeleiteten Aminosäuresequenzen, die am N-Terminus und/oder am C-Terminus mit weiteren Proteinen, z.B. dem Green Fluorescent Protein, fusioniert sind.
Besonders bevorzugte Varianten der erfindungsgemäßen Enzyme werden durch Hinzufügung, Deletion oder Austausch von bis zu 20, bevorzugt bis zu 10 und noch stärker bevorzugt bis zu 5 Aminosäuren der offenbarten Sequenzen erhalten. Grundsätzlich können die vorgenannten Modifikationen an jeder beliebigen Stelle des betreffenden Polypeptids kontinuierlich oder diskontinuierlich erfolgen. Bevorzugt erfolgen sie aber nur am N-Terminus und/oder am C-Terminus des Polypeptids.
Um eine effektive Spaltung der Carbonatgruppe zu gewährleisten werden die Enzyme zusammen mit der Verbindung gemäß Formel (I) in dem weiter oben in dieser Anmeldung definierten wässrigen Medium vorzugsweise bei einer Temperatur zwischen 15 und 80 °C, stärker bevorzugt zwischen 20 und 75 °C und besonders bevorzugt zwischen 30 und 70 °C inkubiert. Der pH liegt dabei vorzugsweise zwischen 5 und 10. Das durch SEQ ID NO.: 3 definierte Polypeptid und seine Varianten zeigt zwei pH- Optima, eines bei pH 7 und eines bei pH 9. Nähere Details zu den für die erfindungsgemäß verwendeten Enzyme besonders geeigneten Reaktionsbedingungen sind in den Ausführungsbeispielen angegeben.
In einer weiteren Ausführungsform betrifft die vorliegende Erfindung ein Verfahren zur Spaltung einer Carbonatbindung in einer Verbindung nach Formel (I) enthaltend die Verfahrensschritte a) Bereitstellung eines wässrigen Mediums enthaltend ein Gemisch, das wenigstens eine Verbindung gemäß Formel (I) enthält und
(i) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.:
1 oder eine Variante des vorgenannten Enzyms, die durch eine Aminosäuresequenz mit wenigstens 85 % Identität zu den in SEQ ID NO.: 1 definierten konservierten Aminosäuren definiert ist; oder
(ii) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.:
2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist; und b) Inkubation des Gemisches unter Bedingungen, unter denen das Enzym aromatische Carbonate spaltet.
Dieses Verfahren ist besonders für die Spaltung der durch Formel (II) charakterisierten Polycarbonate geeignet. Hierbei werden diese Verbindungen in Fragmente mit geringerem Molekulargewicht gespalten. Vorzugsweise werden sie zu Kohlensäure und Bisphenol A abgebaut. Es ist besonders bevorzugt, dass Verfahrensschritt b) durchgeführt wird bis wenigstens 30 Gew.-%, bevorzugt wenigstens 50 Gew.-%, stärker bevorzugt wenigstens 70 Gew.-% und besonders bevorzugt wenigstens 90 Gew.-% der Gesamtmasse der zu Beginn des Verfahrensschritts b) vorliegenden Verbindung nach Formel (II) zu Bisphenol A und Kohlensäure abgebaut sind.
Beim Abbau einer Verbindung gemäß Formel (II) durch das oben definierte Verfahren wird Bisphenol A freigesetzt. Deswegen betrifft die vorliegende Erfindung in einer weiteren Ausführungsform ein Verfahren zur Herstellung von Bisphenol A enthaltend die Schritte a) Bereitstellung eines wässrigen Mediums enthaltend ein Gemisch, das wenigstens eine Verbindung gemäß Formel (I) enthält und
(i) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.:
1 oder eine Variante des vorgenannten Enzyms, die durch eine Aminosäuresequenz mit wenigstens 85 % Identität zu den in SEQ ID NO.: 1 definierten konservierten Aminosäuren definiert ist; oder
(ii) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.:
2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist; und b) Inkubation des Gemisches unter Bedingungen, unter denen das Enzym Esteraseaktivät zeigt.
Hierbei wird in Verfahrensschritt b) Bisphenol A freigesetzt. Somit handelt es sich um ein Verfahren zur Rückgewinnung von Bisphenol A aus Polycarbonaten, also ein Recyclingverfahren.
Der Verfahrensschritt b) wird vorzugsweise durchgeführt, bis wenigstens 5, bevorzugt wenigstens 10, stärker bevorzugt wenigstens 50 und noch stärker bevorzugt wenigstens 70 Gew.-% des zu Beginn in der eingesetzten Gesamtmasse der Verbindung gemäß Formel (II) gebundenen Bisphenol A freigesetzt sind.
In einer bevorzugten Ausführungsform enthält das oben definierte Verfahren zur Herstellung von Bisphenol A einen weiteren Verfahrensschritt der Abtrennung von Bisphenol A aus dem durch die Inkubation in Verfahrensschritt b) erhaltenen Stoffgemisch. Dies kann vorteilhaft durch die Extraktion des freigesetzten Bisphenol A mit einem organischen Lösungsmittel, vorzugsweise Toluol erfolgen. Das Bisphenol A kann dann nach erfolgter Phasentrennung leicht durch Absenkung der Temperatur auskristallisiert werden.
In noch einer weiteren Ausführungsform betrifft die vorliegende Erfindung die Verwendung eines Enzyms
(i) das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 1 oder eine Variante des vorgenannten Enzyms, die durch eine Aminosäuresequenz mit wenigstens 85 %
Identität zu den in SEQ ID NO.: 1 definierten konservierten Aminosäuren definiert ist; oder
(ii) das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist; zur Aktivierung einer Prodrug gemäß Formel (I) durch Spaltung der Carbonatgruppe.
In noch einer weiteren Ausführungsform betrifft die vorliegende Erfindung eine Zusammensetzung enthaltend a)
(i) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 1 oder eine Variante des vorgenannten Enzyms, die durch eine Aminosäuresequenz mit wenigstens 85 % Identität zu den in SEQ ID NO.: 1 definierten konservierten Aminosäuren definiert ist; oder
(ii) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist; b) wenigstens ein aromatisches Carbonat; und c) wenigstens ein wässriges Medium.
Das aromatische Carbonat ist bevorzugt eine Verbindung gemäß Formel (I)

(ii) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist; und b) wenigstens ein aromatisches Carbonat.
In einer besonders bevorzugten Ausführungsform enthält das Kit zusätzlich wenigstens ein wässriges Medium.

Die nachfolgenden Ausführungsbeispiele dienen nur dazu, die Erfindung zu illustrieren. Sie sollen den Schutzbereich der Patentansprüche in keiner Weise beschränken.
Ausführungsbeispiele
Enzyme
Kommerziell erhältliche Enzyme mit den Aminosäuresequenzen gemäß SEQ. ID NO.: 15 bis 27 wurden von Sigma-Aldrich (Steinheim, Deutschland), Strem Chemicals (Bischheim, Frankreich) oder Fisher Scientific, (Rockford, USA) bezogen.
Plasmid-Konstruktion
Gene von Interesse wurden von Eurofins Genomics (Ebersberg, Deutschland) synthetisiert und mittels Restriktionsklonierung mit Xhol und Ndel in den pET26b(+)-Vektor von Merck (Darmstadt, Deutschland) oder mit anderen geeigneten Restriktionsenzymen der multiple cloning site in geegnete Escherichia coli (E. coli) Vektoren kloniert. Für die Vermehrung des Plasmids wurde eine Transformation chemisch kompetenter E. coli DH5a-Zellen durchgeführt. Gewachsene Einzelkolonien wurden zur Inokulation von 4,5 ml LB-Medium mit 50 pg/ml Kanamycin verwendet und entweder in 25 % Glycerol-Stocks konserviert oder gemäß den Herstellerangaben für die Sequenzierung bei GeneWiz (Leipzig, Deutschland) vorbereitet.
Heterologe Proteinexpression
Die heterologe Expression der Enzyme erfolgte in mit dem Plasmid-Konstrukt transformierten E. coli BL21 (DE3)-Zellen. 4,5 ml LB-Medium mit 1 % (w/v) Glucose und 50 pg/ml Kanamycin wurden aus einer Glycerol-Stammkultur beimpft und über Nacht bei 37 °C und 200 rpm inkubiert. 200 ml ZYP-5052- Autoinduktionsmedium mit 50 pg/ml Kanamycin wurden mit 0,1 % frisch gewachsenen Zellen beimpft und bei 37 °C und 200 rpm für 5 h in Kolben mit Schikanen inkubiert. Die Expression erfolgte über Nacht bei 20 °C und 200 rpm. Die Zellen wurden bei 4 °C für 10 min bei 8000 x g geerntet.
Zelllyse
Die geernteten Zellen für das initiale Screening wurden in 6 ml Ammoniumacetatpuffer (20 mM Ammoniumacetat, 0,4 % (w/v) n-Dodecyl-ß-maltosid, 1 % (w/v) Lysozym und 1 pl/ml Benzonase) resuspendiert und 30 min unter Schütteln bei Raumtemperatur inkubiert, um die Zellen zu lysieren. Geerntete Zellen mit exprimierter BhrPETase, DuraPETase, ThcCutl-ACCG, SvCutl90*SS, LCC-ICCG und LCC-WCCG für das initiale Screening wurden in 10 ml 20 mM KPi-Puffer pH 7,5 resuspendiert. Zellen für die Enzymreinigung wurden in 15 ml Lysepuffer (50 mM NaH2PO4, 300 mM NaCI und 10 mM Imidazol) resuspendiert. Die Zellen wurden zweimal mit Ultraschall (Bandelin Sonoplus HD, Sonotrode MS73, Berlin, Deutschland) unter konstanter Kühlung in einem Eisbad für 2 min (Zyklus 50 %) aufgeschlossen. Die resultierende Suspension wurde bei 10.000 x g und 4 °C für 20 min zentrifugiert. Der Überstand wurde mit einem 0,2 pm PES-Filter filtriert.
Lyophilisierung des Zelllysats
Der Überstand der Zellen mit exprimierter BhrPETase, DuraPETase, ThcCutl-ACCG, SvCutl90*SS, LCC- ICCG und LCC-WCCG sowie das Lysat der anderen Zellen für das initiale Screening, das alle Zellbestandteile enthält, wurden bei -80 °C eingefroren und 3 Tage lang bei -68 °C und 0,1 mBar lyophilisiert. Die lyophilisierten Zellen wurden bei -4 °C gelagert.
Enzymreinigung
Die Enzyme mit His-Tag wurden mittels IMAC -Affinitätschromatographie gereinigt. Die Säulen wurden mit dem erforderlichen Säulenvolumen Ni-NTA (HisPurTM Ni-NTA Resin, Fisher Scientific, Rockford, USA) gepackt und mit 8 Säulenvolumina (CV) Lysepuffer äquilibriert. Das filtrierte Lysat wurde auf die Säule geladen. Nach dem Aufträgen des Lysats wurden 10 CV Waschpuffer (50 mM NaHjPC , 300 mM NaCI, 20 mM Imidazol) auf die Säule gegeben. Das spezifisch gebundene His-getaggte Enzym wurde im Elutionspuffer (50 mM NaHjPC , 300 mM NaCI und 250 mM Imidazol pH 8) eluiert. Falls erforderlich, wurden die Proteine mit Amicon® Ultra-Zentrifugalfiltern (10 kDa MWCO) auf 2,5 ml konzentriert. Die Proteine wurden unter Verwendung des Gravity-Protokolls für PD-10-Säulen nach Herstelleranweisungen entsalzt und in 100 mM Kaliumphosphatpuffer pH 7,5 gelagert.
Natriumdodecylsulfat-Polyacrylamidgelelektrophorese (SDS-PAGE)
Die Enzymexpression und Proteinreinheit wurden mittels Natriumdodecylsulfat- Polyacrylamidgelelektrophorese (SDS-PAGE) untersucht. 14 pl der lysierten Zellsuspension, des Überstands nach der Zentrifugation oder der gereinigten Enzym-Lösung wurden mit 7 pl 3x Reducing Blue Protein Loading Dye gemischt. Die Lösungen wurden 5 min bei 95 °C inkubiert. 10 pl der Proben wurden zusammen mit 3 pl EZ-Run® Prestained Rec Protein Ladder auf ein vorgegossenes NuPAGE 4- 12% Bis-Tris-Gel aufgetragen. Die Proteine wurden bei konstanter Spannung von 200 V für 35-40 min elektrophoretisch aufgetrennt. Die Gele wurden 1 h mit 30-50 ml Roti BlueQuick gebrauchsfertiger Färbelösung angefärbt.
Bestimmung der Proteinkonzentration
Die Proteinkonzentration wurde kolorimetrisch mit dem Bradford-Assay bestimmt. Dazu wurde eine BSA-Standardkurve von 125-1000 pg/ml hergestellt. Das Enzym wurde bei Bedarf mit Kaliumphosphatpuffer pH 7,5 verdünnt. Zu 5 pl BSA- und Enzymlösung wurden 250 pl Quick Start Bradford Reagenz, lx, hinzugegeben. Nach 5 min Inkubation wurde die Absorption bei 595 nm gemessen.
Test der Enzymaktivität mit BPA-PC
BPA-PC (Makrolon 2408, Covestro Deutschland AG) wurde vor der Verwendung auf 500 pm gemahlen. Das initiale Screening auf Hydrolyse von BPA-PC wurde mit einer Spatelspitze lyophilisiertem Enzym (kommerziell oder intern heterolog exprimiert) durchgeführt, das in 200 pl Kaliumphosphatpuffer (50 mM KPi, 100 mM NaCI pH 7,5) und einer Spatelspitze BPA-PC gelöst wurde. Die Reaktionen wurden
bei 40 °C, 50 °C, 60 °C und 70 °C und 800 rpm für 146 h inkubiert. Für schlecht exprimierte Enzyme im Lyophilisat wurde gereinigtes Enzym verwendet. Zu 10 mg/ml BPA-PC wurde 100 pg/ml Enzym bis zu einem Endvolumen von 200 pl in 100 mM Kaliumphosphatpuffer pH 5-8 hinzugegeben. Die Reaktionen wurden bei 40-70 °C und 800 rpm für 384 h inkubiert.
Alle Enzyme mit detektierbarer BPA-PC-Hydrolyse wurden gereinigt. Die Reaktionsbedingungen mit höchster BPA-PC-Hydrolyse aus dem vorherigen Test für jedes Enzym wurden verwendet, um die Umwandlung zu BPA zu quantifizieren. Die 4 besten Enzyme wurden ausgewählt und die Umwandlung zu BPA wurde bei Zugabe von 50-600 pg/ml Enzym zu 1, 10 und 50 mg/ml BPA-PC in 100 mM Kaliumphosphatpuffer, CHES-Puffer oder 1,3-Bis(tris(hydroxymethyl)methylamino)propan bei pH 5-10 gemessen. Die Reaktion lief bei 50-90 °C und 800 rpm für 72 h. Anschließend wurde DMSO im Verhältnis 1:1 zur Reaktion hinzugegeben. Die Proben wurden 10 s gevortext und 5 min bei 11.000 x g zentrifugiert. Der Überstand wurde 1:10 verdünnt und mit einem PVDF-Filter (0,2 pm) filtriert.
HPLC-Messungen
Die HPLC-Messungen wurden an einem Agilent 1260 Infinity Il-System (Santa Clara, USA) durchgeführt. Die LC war mit einem Multisampler (G7167A), einem DAD (Diodenarray-Detektor) für UV und den sichtbaren Lichtbereich und einer Zorbax Eclipse Plus C18-Säule (4,6 mm x 150 mm, 5 pm, Agilent, Santa Clara, USA) ausgestattet. Für alle Methoden wurden 5 pl Probe injiziert und die Säule auf 40 °C beheizt. Der Fluss betrug 1,0 ml/min. Die für die Analyse und Quantifizierung der BPA-PC-Hydrolyse Reaktion entwickelte Methode ist in Tabelle 2 aufgeführt. Die Daten wurden mit der Software Chromeieon Chromatography Data System 7.3.1 (Thermo Scientific, Waltham, USA) analysiert.

Konstruktion des phylogenetischen Baums und Konsensussequenz
Sequenzen verschiedener Hydrolasen-Vertreter aus unterschiedlichen Organismen wurden aus der PDB- und Uniprot-Datenbank abgerufen und mit den für diese Arbeit interessanten Sequenzen kombiniert. Für Aspergillus niger und Burkholderia cepacia sind 3 Hydrolase-Sequenzen beschrieben, die aus der Uniprot-Datenbank stammen und in die Analyse einbezogen wurden. In der MEGA11- Software wurde eine ClustalW-Sequenzalignment durchgeführt. Die phylogenetische Analyse erfolgte durch Auswahl der Standardeinstellungen und der Maximum-Likelihood-Methode. Der Bootstrap-
Konsensus-Baum repräsentiert die evolutionäre Geschichte der analysierten Taxa. Ursprüngliche Bäume für die heuristische Suche wurden automatisch durch Anwendung der Neighbor-Join- und BioNJ-Algorithmen auf eine Matrix von paarweisen Distanzen erzeugt, die mit einem Jones-Taylor- Thornton (JTT)-Modell und derTopologie mit dem überlegenen Log-Likelihood-Wert geschätzt wurden. Die Analyse umfasste 25 Aminosäuresequenzen. Die Sequenz der alkalischen Phosphatase aus E. coli wurde als Ausreißer Sequenz verwendet.
Ein ClustalW-Alignment der Aminosäuresequenzen von BPA-PC-hydrolysierenden Enzymen wurde verwendet, um Konsensusssequenzen mit 100 % und 85 % Schwellenwert zu berechnen. Zwei phylogenetischen Gruppen von BPA-PC-hydrolysierenden Enzymen wurde verwendet, um die Sequenzähnlichkeit zu bestimmen.
Ergebnisse
Die Hydrolyse von BPA-PC zu BPA wurde mit den in der Tabelle aufgeführten Enzymen getestet. Von 26 getesteten Enzymen hydrolysierten 10 BPA-PC. Enzyme mit den Nummern 15-27 sind kommerzielle Enzyme. Weiterhin zeigte die Cutinase aus Humicola insolens, die als Novozym® 51032 bezogen wurde eine Hydrolyse von BPA-PC. Dieses Enzym hat eine Aminosäuresequenz wie durch SEQ. ID NO.: 27 definiert (Ternström et al. (2005) „Unfolding and inactivation of cutinases by AOT and guanidine hydrochloride", Biochimica et Biophysika Acta, 1748: 74-83). Mit den Lipasen aus Candida antarctica (CAL), Candida rugosa (CRL) und der Pankreas-Lipase vom Schwein (PPL), die zuvor veröffentlicht wurden (Artham et al. (2011), „Mechanistic investigations of lipase-catalyzed degradation of polycarbonate in organic solvents", Enzyme and Microbial Technology, 48: 71-79), wurde keine Umwandlung zu BPA detektiert.

*Die bei den Abbauversuchen eingesetzten Enzyme wurden von kommerziellen Anbietern bezogen, die im Sequenzprotokoll angeführten Aminosäuresequenzen sind öffentlichen Datenbanken entnommen und wurden für die Sequenzanalyse genutzt.
Die BPA-PC hydrolysierenden Enzyme wurden gereinigt und auf die Umwandlung von BPA-PC analysiert (Tabelle 3). Bei 70 °C in 100 mM Bis(tris(hydroxymethyl)methylamino)propan pH 7 mit ThcCutl-ACCG oder pH 9 für LCC, LCC-ICCG und LCC-WCCG wurden Umsetzungen von 16,8-33,2 % bestimmt. Die Zugabe von DMSO wurde mit den 2 besten Varianten ThcCutl-ACCG und LCC-ICCG getestet. Eine Zugabe von 20-50 % DMSO zur Reaktion mit ThcCutl führte zu 100 % Umwandlung zu BPA. Mit LCC-ICCG war die Umwandlung zu 92,9 % BPA bei Zugabe von 30 % DMSO am höchsten. Ohne DMSO-Zugabe führte das 4-malige Ersetzen der wässrigen Phase nach 72 h durch eine frische Enzym- Pufferlösung zu 94 % Umwandlung zu BPA mit ThcCutl-ACCG und 51 % mit LCC-ICCG. Dafür wurde nach 72 h die Reaktion 3 min bei 13300 rpm zentrifugiert, der Überstand in ein neues Gefäß überführt und das gleiche Volumen Enzym-Pufferlösung zum BPA-PC gegeben. Die Reaktion lief erneut bei 70 °C und 800 rpm für 72 h. Nach 3-maligem Ersetzen der Enzym-Pufferlösung wurde DMSO im Verhältnis 1:1 zur Reaktion oder zum Überstand gegeben. Die Messung der Proben erfolgte wie beschrieben.
Inkubationsbedingungen bei denen die höchste BPA-Freisetzung gemessen wurde sind in Klammern dargestellt.

Phylogenetische Analyse
Die erhaltene Phylogenie zeigt die enge Verwandtschaft der Enzyme. Der Cluster der BPA-PC hydrolysierenden Enzyme teilt sich einen gemeinsamen Vorfahren mit IsPETase, PET2 und PET6. Bei letzteren Enzymen war keine BPA-PC-Hydrolyse detektierbar. Daher hat sich diese Funktion später entwickelt. Innerhalb des Clusters der BPA-PC hydrolysierenden Enzyme können zwei Gruppen unterschieden werden. Eine Gruppe enthält ThcCutl, ThcCutl-ACCG, TfCut2, SvCutl90*SS und PesHl (Gruppe 1). Die andere Gruppe enthält LCC, LCC-ICCG, LCC WGGC und die BhrPETase (Gruppe 2).
Die kommerziellen Enzyme (Nr. 15-27) sind nur entfernt miteinander und dem Cluster der BPA- hydrolysierenden Enzyme verwandt, wie durch die längeren Äste angezeigt. Die Mehrheit der angegebenen Beziehungen innerhalb des Clusters der BPA-hydrolysierenden Enzyme wird durch die Bootstrap-Werte validiert. Für früh abzweigende Vorfahren deuten Bootstrap-Werte auf eine weniger ausgeprägte Beziehung zwischen den Sequenzen hin.
Basierend auf dem Alignment der Sequenzen von BPA-PC hydrolysierenden Enzymen wurde die Konsensussequenz mit 100 % (SEQ ID NO.: 1) und 85 % Schwellenwert (SEQ ID NO: 28) berechnet. Die Aminosäuresequenz zeigt im Vergleich zum N-Terminus eine höhere Sequenzvariabilität am C- Terminus.

Für die Berechnung der Sequenzähnlichkeit wurde die Zahl der identischen und ähnlich Aminosäurepositionen addiert. Dann wurde der Prozentsatz dieser Summe bezogen auf die Gesamtzahl der Aminosäurepositionen des Alignments berechnet.
Folgende Aminosäuren wurden als ähnlich definiert:
Kleine/alipatische Reste: Glycin, Alanin, Valin, Leucin, Isoleucin
Aromatische Reste: Phenylalanin, Tyrosin, Tryptophan
Schwefelhaltige Reste Cystein, Methionin
OH-funktionelle oder basische Reste: Serin, Threonin, Lysin, Arginin, Histidin
Saure, amidhaltige Reste und Prolin: Asparaginsäure, Glutaminsäure, Asparagin, Glutamin,
Prolin
Für die Berechnung der Sequenzidentität wurde der Prozentsatz identischer Aminosäurepositionen auf die Gesamtzahl der Aminosäurepositionen des Alignments berechnet.
Test weiterer Enzyme
Um zu überprüfen, ob alle Aminosäurepositionen der durch SEQ ID NO.: 1 definierten Konsensussequenz essentiell für die Enzymfunktion sind, wurden zwei weitere Cutinasen getestet. Es handelt sich um Kubu-P M12 (SEQ ID NO.: 29 ) und Mipa-P M19 (SEQ ID NO.: 30). Diese Enzyme wurden in Seo et al. (2025), „Landscape profiling of PET depolymerases using a natural sequence cluster framework", Science, Vol. 387, eadp 5637 beschrieben.
Die Enzyme wurden heterolog exprimiert wie oben beschrieben. Als Testsubstrat wurde das oben beschriebene Polycarbonat verwendet. Die Quantifizierung des Polycarbonats erfolgte durch Nachweis von BPA durch HPLC wie oben beschrieben.
Kubu-P M12 und die durch SEQ ID NO.: 1 definierte Konsensussequenz haben nach den oben definierten Kriterien eine Sequenzähnlichkeit von 87 %. Die nach den oben definierten Kriterien bestimmte Sequenzidentität liegt bei 24 %. Nach 120 Stunden bei 70° C wurde in einem Gemisch von Wasser und DMSO, das bezogen auf das Gesamtgewicht des Gemisches 30 Vol.-% DMSO enthielt, ein Umsatz 10,1 % beobachtet.
Mipa-P M19 und die durch SEQ ID NO.: 1 definierte Konsensussequenz haben nach den oben definierten Kriterien eine Sequenzähnlichkeit von 87 %. Die nach den oben definierten Kriterien bestimmte Sequenzidentität liegt bei 26 %. Nach 120 Stunden bei 70° C wurde in einem Gemisch von Wasser und DMSO, das bezogen auf das Gesamtgewicht des Gemisches 30 Vol.-% DMSO enthielt, ein Umsatz 20,6 % beobachtet.
Die Ergebnisse zeigen, dass auch Cutinasen mit sehr geringer Sequenzidentität zur Konsensussequenz in der Lage sind BPA-basiertes Polycarbonat zu depolymerisieren. Es kann also davon ausgegangen werden, dass ein Austausch von bis zu 15 % der durch SEQ ID NO.: 1 definierten Aminosäurepositionen möglich ist, ohne die erfindungsgemäße Enzymfunktion zu zerstören.
Nachweis des Abbaus niedermolekularer aromatischer Carbonate
0,4 mg lyophilisiertes Rohlysat (kommerziell erworben oder heterolog exprimiertes Enzym) wurde in 400 pl KPi-Puffer (50 mM Kaliumphosphat, 100 mM NaCI pH 7,5) gelöst. 5 mg/ml Substratlösungen von Dimethylcarbonat (DMC), Benzyl-(4-nitrophenyl)-carbonat (BNPC) und Diphenylcarbonat (DPC) wurden in DMSO hergestellt.
Für die Reaktion mit DMC wurden 100 pl der Enzymlösung mit 250 pl 25 mM KPi-Puffer mit etwa 0,1 mg/ml Phenolrot, 100 pl H2O und 50 pl Substratlösung gemischt. Die Proben wurden für 22 h bei 40 °C und 800 rpm inkubiert. Bei der Hydrolyse von Dimethylcarbonat wird Kohlensäure gebildet. Die pH- Wert-Abnahme durch die Kohlensäurebildung wird mit Phenolrot nachgewiesen und ist durch einen Farbumschlag von rot nach gelb sichtbar.
Die BNPC- und DPC-Hydrolyse wurde mit 100 pl des gelösten Lysats bestimmt, gemischt mit 350 pl KPi- Puffer und 50 pl Substratlösung. Für DPC wurde etwa 0,1 mg/ml Phenolrot zugegeben. Die Proben wurden für 20 h bei 20 °C oder bei 20 °C gefolgt von 40 °C, 800 rpm, für BNPC bzw. DPC inkubiert. Das Hydrolyseprodukt von BNPC ist p-Nitrophenol (pNP) und wurde spektrophotometrisch bei 410 nm
nachgewiesen. Eine Standardkurve von pNP wurde als Referenz verwendet. Für die Analyse der DPC Proben wurde 500 pl Acetonitril nach er Reaktion zugegeben, um die Reaktion zu stoppen, und die Proben wurden für HPLC-Messungen verwendet. Ein Phenol-Standard wurde als Referenz verwendet. Die gleiche HPLC-Methode wie für PC wurde zur Bestimmung von Phenol verwendet.
Eine Farbänderung wurde als Indiz für Abbau gewertet, ohne diesen zu quantifizieren.

Das Ergebnis zeigt, dass die getestete Auswahl an Enzymen in der Lage war, auch niedermolekulare Carbonate zu spalten. Dieses Ergebnis ist vermutlich auch auf alle anderen Enzyme übertragbar, die Polycarbonat abbauen können, da die niedermolekulare Substrate für das Enzym besser zugänglich sind als Polymere.
Claims
1. Verwendung eines Enzyms,
(a) das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 1 oder eine Variante des vorgenannten Enzyms, die durch eine Aminosäuresequenz mit wenigstens 85 % Identität zu den in SEQ ID NO.: 1 definierten konservierten Aminosäuren definiert ist; oder
(b) das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist. zur Spaltung eines aromatischen Carbonats in einem wässrigen Medium.
2. Die Verwendung nach Anspruch 1, wobei das Enzym wie folgt definiert ist:
(i) Die zu den konservierten Aminosäurepositionen gemäß SEQ ID NO.: 1 homologen Aminosäurepositionen des Enzyms weisen wenigstens 90 % Sequenzidentität zu den konservierten Aminosäurepositionen aus SEQ ID NO.: 1 auf; und
(ii) Die zu den beliebigen Aminosäurepositionen aus SEQ ID NO.: 1 homologen Aminosäurepositionen weisen wenigstens 70 % Sequenzidentität zu den homologen Aminosäurepositionen einer der Aminosäuresequenzen ausgewählt aus der Gruppe bestehend aus SEQ ID NO.: 3, 8, 2, 4, 5, 6, 7, 9 und 10 auf.
3. Die Verwendung nach Anspruch 1, wobei das Enzym wenigstens 80 % Sequenzidentität zu SEQ ID NO.: 2 oder wenigstens 90 % Sequenzidentität zu SEQ ID NO.: 7 aufweist.
4. Die Verwendung nach einem der Ansprüche 1 bis 3, wobei das aromatische Carbonat durch Formel (I) definiert ist
Und wobei
RI ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Hydroxyl und beliebigen organischen Resten; und
R2 unabhängig von RI ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, einer Verbindung gemäß Formel (I) und beliebigen organischen Resten.
5. Die Verwendung nach einem der Ansprüche 1 bis 4, wobei das aromatische Carbonat ein Polycarbonat ist.
6. Die Verwendung nach Anspruch 5, wobei das Polycarbonat erhalten oder erhältlich ist durch Reaktion wenigstens einer Verbindung ausgewählt aus der Gruppe bestehend aus Bisphenol A, Bisphenol S, Dihydroxyfephenylsulfid, Tetramethylbisphenol A, l,l-Bis(4-hydroxyphenyl)- 3,3,5-trimethylcyclohexan (BPTMC) und l,l,l-Tris(4-hydroxyphenyl)-ethan (THPE) mit Phosgen oder Diphenylcarbonat.
7. Die Verwendung nach Anspruch 6, wobei das Polycarbonat durch Formel (II) definiert ist
8. Die Verwendung nach einem der Ansprüche 5 bis 7 , wobei das aromatische Polycarbonat als Copolymer mit wenigstens einem Polyester vorliegt.
9. Die Verwendung nach einem der Ansprüche 5 bis 7 , wobei das Polycarbonat als Gemisch mit wenigstens einem weiteren Polymer vorliegt.
10. Verfahren zur Spaltung einer Esterbindung in einer Verbindung nach Formel (I)
wobei
RI ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Hydroxyl und beliebigen organischen Resten; und
R2 unabhängig von RI ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, einer Verbindung gemäß Formel (I) und beliebigen organischen Resten; enthaltend die Verfahrensschritte a) Bereitstellung eines wässrigen Mediums enthaltend ein Gemisch, das wenigstens eine Verbindung gemäß Formel (I) enthält und
(i) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.:
1 oder eine Variante des vorgenannten Enzyms, die durch eine Aminosäuresequenz mit wenigstens 85 % Identität zu den in SEQ ID NO.: 1 definierten konservierten Aminosäuren definiert ist; oder
(ii) wenigstens ein Enzym, das durch eine Aminosäuresequenz gemäß SEQ ID NO.:
2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist; und
- TI - b) Inkubation des Gemisches unter Bedingungen, unter denen das Enzym aromatische Carbonate spaltet.
11. Das Verfahren nach Anspruch 10, wobei die Verbindung durch Formel (II) definiert ist.
12. Das Verfahren nach Anspruch 11, wobei die Verbindung gemäß Formel (II) als Gemisch mit wenigstens einem Polymer ausgewählt aus der Gruppe bestehend aus Polyestern, Styrol- Acrylnitril-Copolymer und Acrylnitril-Butadien-Styrol-Copolymer vorliegt.
13. Das Verfahren nach Anspruch 12, wobei der Polyester ausgewählt ist aus der Gruppe bestehend aus Polyethylenterephthalat und Polybutylenterephthalat.
14. Das Verfahren nach einem der Ansprüche 11 bis 13, wobei Verfahrensschritt b) solange durchgeführt wird, bis wenigstens 5 Gew.-% der zu Beginn des Verfahrensschrittes b) vorliegenden Gesamtmasse der Verbindung nach Formel (II) abgebaut sind.
15. Verwendung eines Enzyms
(i) das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 1 oder eine Variante des vorgenannten Enzyms, die durch eine Aminosäuresequenz mit wenigstens 85 % Identität zu den in SEQ ID NO.: 1 definierten konservierten Aminosäuren definiert ist; oder
(ii) das durch eine Aminosäuresequenz gemäß SEQ ID NO.: 2 oder 7 oder eine Variante eines der vorgenannten Enzyme, die durch eine Aminosäuresequenz mit wenigstens 74 % Identität zu SEQ ID NO.: 2 oder wenigstens 85 % Sequenzidentität zu SEQ ID NO.: 7 definiert ist; zur Aktivierung einer Prodrug gemäß Formel (I)

Und wobei
RI ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Hydroxyl und beliebigen organischen Resten; und R2 unabhängig von RI ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, einer
