Abstract
본 발명은 본 발명은 지지체; 상기 지지체 표면에서 고정된 포획 프로브; 상기 포획 프로브와 시료를 사이에 두고 샌드위치 형태로 특이적 결합을 하는 검출 프로브; 및 상기 검출 프로브에 연결되어 기질과 반응하는 질화전이금속 나노자임으로 구성된 표지;를 포함하는 바이오센서로서, 상기 질화전이금속 나노자임은 효소로서 작용하며 기질과의 반응에 의하여 전기화학 신호 또는 시각적 신호를 발생하는 것을 특징으로 하는 바이오센서를 제공한다. 본 발명의 질화전이금속 나노자임을 활용하여 효소와 같은 역할을 할 수 있으며, 기질과의 반응에 의하여 전기화학 신호 또는 시각적 신호를 발생하게 할 수 있다.
Description
본 발명은 질화전이금속 나노자임을 포함하는 바이오센서에 관한 것으로서, 더욱 상세하게는 질화전이금속 나노자임을 포함한 표지가 효소와 같은 역할을 하여, 기질과의 반응에 의하여 시각적 신호를 발생할 수 있는 전이금속 나노자임을 포함하는 바이오센서에 관한 것이다.
단백질칩이란, 특정 표적 단백질과 반응할 수 있는 항체, 수용체, 핵산, 탄수화물 등의 포획분자를 단일칩 상에 고밀도로 고정화한 것으로서, 단백질의 분리, 확인, 정량 및 기능 해석에 이르는 일련의 단백질 분석 작업을 수행할 수 있다. 그 중 항체를 칩상에 고정화한 단백질칩이 가장 일반적이다.
단백질칩을 이용하면 단백질-단백질 간의 상호관계에 관한 데이터 생성이 가능하며, 특히 질병발생시 특이적으로 과발현되는 특정 단백질을 검출하여 질병을 진단할 수도 있고, 환경 감시, 유해성 검진 등 잠재 수요가 대단히 크기 때문에 DNA칩보다 더 큰 시장을 형성할 것으로 예상되고 있어, 단백질칩에 대한 개발이 절실히 요구되고 있다.
단백질칩의 구성을 보면, 크게 센서칩(sensor chip)과 단백질 결합 분석장치(detection system)로 구성된다. 이 중 센서칩이 단백질칩의 핵심 구성으로 단백질칩의 발전속도를 결정하고 있는 주요 요인이 되고 있는 부분이다.
센서칩 제작에서 단백질칩의 표면 작용기 도입, 포획 분자의 고정화, 표적 단백질과의 반응, 표지물질과의 반응, 표지물질 준비공정, 측정 장비의 상태 등 다양하고 복잡한 공정 기술이 사용되고, 특히 특정 표적 단백질과 반응할 수 있는 포획분자를 칩의 표면에 결합시키는 고정화 기술이 단백질칩의 민감도를 결정하는 핵심기술이다. 특히 항체를 포획분자로 사용하는 경우, Y 모양의 구조에서 항원과 결합하는 부위가 Fab에 존재하기 때문에 효과적으로 항원과 반응시키기 위해서는 항체의 Fc부위가 칩 상에 고정화되고, Fab 부위가 표적 항원을 향하도록 항체의 방향성(orientation)을 확보하면서 고정화하는 것이 절대적으로 중요하다.
최근에는 무기-유기물이 결합된 형태의 효소 모방 센서물질이 보고되었고, 특히 금속-유기 골격체(metal-organic framework; MOF)는 구조적인 특성으로 인해 합성 후 개질(post synthetic modification)을 통해 기능화 및/또는 기공도를 조절하여 촉매 활성을 부여하거나 조절할 수 있다.
따라서 금속-유기 골격체 자체만으로는 효소로 활용하기에 적절치 못한 문제점이 있어서 금속-유기 골격체의 개질을 통한 바이오센서 물질의 개발을 필요로 하는 실정이다.

상기와 같은 문제점을 해결하기 위하여, 본 발명은 전이금속 나노자임으로 구성된 표지를 기질과 반응하여 형성된 시각적 신호를 측정하여 정보를 얻게 되는 바이오센서를 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위하여 본 발명은,
지지체;
상기 지지체 표면에서 고정된 포획 프로브;
상기 포획 프로브와 시료를 사이에 두고 샌드위치 형태로 특이적 결합을 하는 검출 프로브; 및
상기 검출 프로브에 연결되어 기질과 반응하는 질화전이금속 나노자임으로 구성된 표지;를 포함하는 바이오센서로서,
상기 질화전이금속 나노자임은 효소로서 작용하며 기질과의 반응에 의하여 시각적 신호를 발생하는 것을 특징으로 하는 바이오센서를 제공한다.
상기 기질은 3,3',5,5'-테트라메틸벤지딘[3,3',5,5'-tetramethylbenzidine: TMB], 3-아미노-9-에틸카바졸[3-amino-9-ethylcarbazole], o-페닐렌디아민[o-phenylenediamine], 3,3'-디아미노벤지딘[3,3'-diaminobenzidine], 2,2'-아지노비스(3-에틸벤조티아졸린-6-술폰산)[2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)], 및 10-아세틸-3,7-디하이드록시페녹사진[10-acetyl-3,7-dihydroxyphenoxazine] 등으로 이루어진 군으로부터 선택된 하나 이상을 포함하는 것이 바람직하다.
상기 질화전이금속 나노자임은 질화전이금속과 다공성 탄소를 결합하여 형성되는 것이 바람직하다.
상기 질화전이금속은 질화텅스텐(W2N), 질화바나듐(VN), 및 질화크롬(CrN) 중에서 선택된 하나인 것이 바람직하다.
상기 시료는 인간 세럼, 플라즈마, 혈액 및 소변 시료 중 어느 하나 이상인 것이 바람직하다.
상기 나노자임은 기질과의 반응에 의하여 색상이 파란색으로 변경될 수 있다.
본 발명의 질화전이금속 나노자임을 활용하여 효소와 같은 역할을 할 수 있으며, 기질과의 반응에 의하여 시각적 신호를 발생하게 할 수 있다. 이를 항체와 콘쥬게이션하여 lateral flow immunoassay 종이칩에 신호형질로 적용하여 다양한 질병을 조기에 진단할 수 있다. 따라서 암 등을 포함한 4대 중증 질환의 Point of care 진단 키트로 활용가능하다. 또한 단순성, 소형화의 용이성, 뛰어난 선택성 및 감도를 가진다는 장점이 있다.
도 1은 본 발명의 일 실시예에 따라 TMB 기질을 촉매하는 다양한 나노자임의 peroxidase 활성에 대한 정성적 분석결과를 나타낸다.
도 2a는 본 발명의 일 실시예에 따른 UV-Vis 분광분석결과를 도시한 것이다.

이하, 본 발명을 더욱 상세하게 설명한다.
본 발명은 지지체; 상기 지지체 표면에서 고정된 포획 프로브; 상기 포획 프로브와 시료를 사이에 두고 샌드위치 형태로 특이적 결합을 하는 검출 프로브; 및 상기 검출 프로브에 연결되어 기질과 반응하는 질화전이금속 나노자임으로 구성된 표지;를 포함하는 바이오센서로서, 상기 질화전이금속 나노자임은 효소로서 작용하며 기질과의 반응에 의하여 시각적 신호를 발생하는 것을 특징으로 하는 바이오센서를 제공한다.
본 발명에서 질화전이금속 나노자임 표지는 질화전이금속과 다공성 탄소를 결합하여 형성되는 것이 바람직하다. 또한 질화전이금속은 질소와 전이금속이 결합된 형태를 나타내고 이에 한정되는 것은 아니지만 예를 들어, 질화텅스텐(W2N), 질화바나듐(VN), 및 질화크롬(CrN) 중에서 선택된 하나 이상이 바람직하다. 더욱 바람직하게는 질화크롬(CrN)이다.
이러한 질화전이금속 나노자임은 기질과 반응하여 효소와 동일한 역할을 함으로써 시료의 검출에 필요한 요소인 용이한 콘쥬게이트와 강한 시각적 변화를 도출할 수 있다. 구체적으로는 본 발명에서 질화전이금속 나노자임 표지는 기질과의 반응에 의하여 나타나는 시각적 변화를 나타내는데 예를 들어, 기질 색상이 파란색으로 변화될 수 있다.
본 발명의 나노자임(nanozyme)은 천연 효소가 촉진하는 반응과 유사한 반응을 촉매할 수 있는 효소와 유사한 특성을 지닌 다양한 나노 물질을 의미한다. 기존에도 나노자임(nanozyme)을 효소 대신에 활성이 높고 선택적인 촉매로 사용하기 위한 연구가 진행되었다. 나노자임의 선택적인 반응에도 불구하고 천연 효소만큼의 반응 속도를 달성하는 것은 어려운 일이다. 많은 금속산화물 나노물질과 금속 나노물질은 산화제(H2O2, O2 등)의 존재 하에서 퍼옥시데이즈(peroxidase)와 옥시데이즈(oxidase) 효소와 유사한 나노자임으로 작용할 수 있다.
나노자임에 비해 천연 효소의 한 가지 단점은 가혹한 조건에서 안정성이 제한된다는 점이다. 효소는 종종 pH, 온도 및 화학적 환경의 변화에 민감하여 활성과 효과가 저하될 수 있다. 반면 나노자임은 안정성, 비용 효율성, 조정 가능한 촉매 특성 등 천연 효소에 비해 장점이 있어 다양한 연구 및 응용 분야에서 매력적인 대안이 되고 있다.
본 발명에서 생체특이적 결합은 포획 프로브(prob)가 지지체(substrate)에 고정되고, 검출 프로브가 표지와 연결되어, 포획 프로브와 검출 프로브가 시료를 사이에 두고 샌드위치 형태로 특이적 결합할 수 있다. 또는 포획 프로브가 지지체에 고정되고, 시료와 표지가 결합되거나 시료가 포획 프로브에 경쟁적으로 결합할 수 있다.
본 발명의 시료 검출 바이오센서에 있어서, 기질은 효소 역할을 하는 질화전이금속 나노자임과의 반응 후 시각적 신호를 발생하는 것이 특징이다. 이러한 시각적 신호를 발생하도록 하는 것은 나노자임과 반응하게 되는 기질에 따른 것이므로 기질의 종류에 따라서 적절하게 활용할 수 있다.
본 발명에서 시각적 신호를 발생시키는 기질의 예로서, 이에 한정되는 것은 아니지만 3,3',5,5'-테트라메틸벤지딘[3,3',5,5'-tetramethylbenzidine: TMB], 3-아미노9-에틸카바졸[3-amino-9-ethylcarbazole], o-페닐렌디아민[o-phenylenediamine], 3,3'디아미노벤지딘[3,3' diaminobenzidine], 2,2'-아지노비즈[3-에틸벤조티아졸린-6-술폰산][2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)], 10-아세틸-3,7-디하이드록시페녹사진[10-acetyl-3,7-dihydroxyphenoxazine] 등으로 이루어진 군으로부터 선택된 어느 하나일 수 있다. 바람직하게는 기질은 3,3',5,5'-테트라메틸벤지딘(TMB) 및 과산화수소인 것이다.
본 발명의 시료는 인간 세럼, 플라즈마, 혈액 및 소변에서 추출할 수 있으며, 질병 진단에 사용되는 시료라면 사용에 제한이 없다. 각종 질환을 대표할 수 있는 바이오마커가 존재한다면 질환의 종류를 제한하지 않는다. 본 발명의 바이오센서는 정상 대조군에 비하여 질환이 발생한 대상체의 혈액, 조직 또는 세포에서 증가 또는 감소 양상을 보이는 단백질 또는 핵산, 지질, 당지질, 당단백질 등과 같은 유기 생체 분자 등을 포함할 수 있으며, 바람직하게는 단백질이다.
본 발명의 다른 일측면에 따르면, 바이오센서를 적용한 진단용 키트를 제공한다. 본 발명의 키트는 크게 진단 스트립, 하부 케이스, 상부 케이스로 구성되며, 진단 스트립은 멤브레인, 검체 패드, 및 흡수패드를 포함한다. 본 발명의 하부 케이스는 진단 스트립의 일면을 지지하며, 상부 케이스는 하부 케이스에 결합되어 분석 스트립의 다른 일면을 덮으며, 검체 패드에 대응하는 검체 투입구와 멤브레인에 대응하는 결과 표시창 및 상기 흡수 패드에 대응하는 문자 표시구를 포함한다.
멤브레인은 검출 결과를 나타내는 부분으로서 조성물이 고정된 검사선 및 대조선이 위치하는데, 통상의 진단 스트립의 재료로 사용되는 것들을 이용할 수 있으며, 예를 들어 니트로셀룰로오스, 셀룰로오스, 셀룰로오스 아세테이트, 폴리에틸렌 등의 각종 합성 중합체에서 선택되는 1종이 될 수 있다.
검체 패드는 멤브레인 한쪽 끝 상부에 일부 중첩되게 위치하여 질화전이금속 나노자임 표지를 포함하는 검체 전개액을 흡수한다. 검체 전개액은 채취한 시료, 복합체가 혼합된 것으로, 질환이 있다면 검체 전개액에는 시료 내의 바이오마커와 질화전이금속 나노자임 표지가 결합한 상태로 존재한다. 이 검체 전개액을 검체 패드 상에 떨어트리면 바이오마커와 질화전이금속 나노자임이 결합한 채로 스트립 상에서 흘러 검사선(test line)에 위치하는 상기 바이오마커와 특이적으로 결합하는 고정 항체와 면역 샌드위치 복합체를 형성한다. 이로써 멤브레인 상의 대조선(control line)과 검사선(test line)에 모두 두 줄의 갈색선이 나타나고, 이는 질병이 있다고 판단한다. 이때 대조선은 질화전이금속 나노자임 표지만 결합하며, 복합체와 결합하는 대조선 상의 고정 항체는 교차 반응을 유발하지 않으면, 그 종류가 특별히 제한되지는 않는다.
이하, 바람직한 실시예를 들어 본 발명을 더욱 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 범위가 이에 의하여 제한되지 않는다는 것은 당업계의 통상의 지식을 가진 자에게 자명할 것이다.
어느 하나의 실시예에 포함된 구성요소와, 공통적인 기능을 포함하는 구성요소는, 다른 실시예에서 동일한 명칭을 사용하여 설명하기로 한다. 반대되는 기재가 없는 이상, 어느 하나의 실시예에 기재한 설명은 다른 실시예에도 적용될 수 있으며, 중복되는 범위에서 구체적인 설명은 생략하기로 한다.
<실시예>
1.1. 질화전이금속기반 나노자임 합성법
실시예 1
W2N@PDC-1의 제조
W2N@PDC-1은 질소를 함유하는 폴리아닐린(polyanilnine, PANI)에 포스포텅스텐산(Phosphotungstic acid, PTA)을 첨가한 후 열분해하여 합성하였으며 그 방법은 다음과 같다. PANI는 2.6 g의 염화 아닐리늄(aniline hydrochloride)을 100 ml의 물에 첨가한 용액과 11.4 g의 과황산암모늄(ammonium persulfate)을 100 ml의 물에 녹인 용액을 혼합하고 4시간 실온에서 반응 후 여과와 건조를 거쳐 약 2 g을 준비하였다. 0.2 g의 PTA를 20 ml의 에탄올에 녹이고 2 g의 PANI를 첨가한 후 30분간 초음파를 조사하여 최대한 균일한 반응물이 되도록 하였다. 증발기로 에탄올을 증발시킨 후 100 ℃ 오븐에서 약 2시간 건조하고 800 ℃에서 12시간 열분해하여 질화텅스텐(W2N)이 도핑된 PANI 유래 카본 (PANI-derived carbon, PDC) W2N@PDC-1를 합성하였다.
실시예 2
W2N@PDC-1의 제조
W2N@PDC-2는 실시예 1의 W2N@PDC-1과 동일한 방법으로 준비하였다. 0.4 g의 PTA를 20 mL의 에탄올에 녹이고 2 g의 PANI를 첨가한 후 30분간 초음파를 조사하여 최대한 균일한 반응물이 되도록 하였다. 증발기로 에탄올을 증발시킨 후 100 ℃ 오븐에서 약 2시간 건조하고, 800 ℃에서 12시간 열분해하여 질화텅스텐(W2N)이 도핑된 PANI 유래 카본 W2N@PDC-2를 합성하였다.
실시예 3
VN@PDC-1의 제조
VN@PDC-1는 PANI에 바나듐(V) 산화물 (Vanadium pentoxide, V2O5)를 첨가한 후 열분해하여 합성하였으며 그 방법은 다음과 같다. 2 ml의 염산 (HCl, 35%)을 첨가한 증류수에 0.2 g의 V2O5를 녹인 후 1 g의 PANI를 첨가하고 3시간 교반하였다. 증발기를 이용하여 염산과 증류수를 증발시킨 후 100 ℃에서 약 2시간 건조하고 1100 ℃에서 5시간 열분해하여 질화바나듐(VN)이 도핑된 PANI 유래 카본 VN@PDC를 합성하였다.
실시예 4
VN@PDC-1의 제조
VN@PDC-2는 VN@PDC-1과 같은 방법으로 준비하였으며 자세한 설명은 다음과 같다. 2 ml의 염산 (HCl, 35%)을 첨가한 증류수에 0.3 g의 V2O5를 녹인 후 1 g의 PANI를 첨가하고 3시간 교반하였다. 증발기를 이용하여 염산과 증류수를 증발시킨 후 100 ℃에서 약 2시간 건조하고 1100 ℃에서 5시간 열분해하여 질화바나듐(VN)이 도핑된 PANI 유래 카본 VN@PDC-2를 합성하였다.
실시예 5
CrN@MDC-1의 제조
CrN@MDC는 Metal Azolate Framework-6 (MAF-6)에 산화크롬(Cr2O3)과 멜라민(Melamine)을 도핑하여 준비한(Cr2O3-melamine)@MAF-6를 열분해하여 합성하였다. 수산화아연(Zn(OH)2) 2 g을 녹인 400 mL의 수산화암모늄(aqueous ammonia solution 25%), 2-에틸이미다졸(2-ethylimidazole) 3.85g을 녹인 220 mL의 에탄올(ethanol)-사이클로헥세인(cyclohexane) 혼합용액(에탄올 200 mL, 사이클로헥세인 20 mL), 그리고 에탄올 20 mL씩 담겨 있는 두 비커에 0.1 g의 Cr2O3와 0.6 g의 멜라민을 각각 첨가한 용액들을 혼합하고 30분간 실온에서 교반 후 여과와 건조(80 ℃)를 거쳐 (Cr2O3-melamine)@MAF-6를 준비하였다. 그 후 1000 ℃에서 6시간 열분해하여 CrN@MDC-1를 합성하였다.
실시예 6
CrN@MDC-2의 제조
CrN@MDC-2은 CrN@MDC-1과 같은 방법으로 준비하였으며 상세한 설명은 다음과 같다. 수산화아연(Zn(OH)2) 2 g을 녹인 400 mL의 수산화암모늄(aqueous ammonia solution 25%), 2-에틸이미다졸(2-ethylimidazole) 3.85 g을 녹인 220 mL의 에탄올(ethanol)-사이클로헥세인(cyclohexane) 혼합용액(에탄올 200 mL, 사이클로헥세인 20 mL), 그리고 에탄올 20 mL씩 담겨 있는 두 비커에 0.15 g의 Cr2O3와 0.9 g의 멜라민을 각각 첨가한 용액들을 혼합하고 30분간 실온에서 교반 후 여과와 건조(80 ℃)를 거쳐 (Cr2O3-melamine)@MAF-6를 준비하였다. 그 후 1000 ℃에서 6시간 열분해하여 CrN@MDC-2를 합성하였다.
비교예 1
Pt@ZDC의 제조
Pt@ZDC는 Zeolitic imidazolate Frameworks-8 (ZIF-8)에 염화테트라아민플래티넘(II)(Tetraamineplatinum (II) chloride, TAP)을 첨가한 후 열분해하여 합성하였다.
질산아연 수화물(Zn(NO3)2·6H2O) 2.9 g을 녹인 100 mL의 메탄올(methanol)과 2-메틸이미다졸(2-methyl-imidazole) 6.9 g을 녹인 100 mL의 메탄올을 혼합하고 2시간 동안 30 ℃에서 반응시킨 후 여과와 건조를 거쳐 ZIF-8을 준비하였다. 20 mL의 증류수에 0.2 g의 TAP을 녹이고 2 g의 ZIF-8을 첨가한 후 초음파를 30분 조사하였다. 증발기를 이용하여 증류수를 증발시키고 100 ℃에서 건조한다음 1100 ℃에서 3시간 동안 열분해하여 Pt@ZDC를 합성하였다.
<효소활성 측정방법>
합성한 질화전이금속 기반 나노자임의 활성은 각 나노자임을 기질인 3,3´,5,5´-tetramethylbenzidine (TMB)와 반응시켜 peroxidase와 유사한 활성을 나타내는지를 정성적으로 조사하였다. 처음에는 각각 1mg의 나노 효소를 0.2 M 아세테이트 완충액 1mL(pH 4.3)에 분산시킨 3병의 바이알을 준비하였다. 그 후, 0.2 mL의 TMB, H2O2 및 TMB/ H2O2 혼합물(1:1)을 각 튜브에 개별적으로 첨가하였다. 그런 다음 pH 4.3의 아세테이트 완충액을 추가하여 최종 부피가 2mL가 되도록 하여 TMB와 H2O2모두 최종 농도가 5 mM가 되도록 하였다. 나노자임이 TMB를 촉매하는 데 성공했다는 것은 TMB의 산화를 나타내는 푸른색 용액이 형성됨으로써 입증되었다. 이 TMB 산화반응은 매분마다 기록되었다.
<특정활성도 (Specific activity) 결정방법>
0.2M 아세테이트 완충액, pH 4.3에 다양한 양(0~50㎍)의 퍼옥시다아제 효소역할을 모방하는 각 나노자임이 포함된 용액 1040μL를 큐벳에 넣었다. 그 후, 100μL의 TMB(50mM)와 100μL의 H2O2 (50mM) 용액을 추가하고 완전히 혼합하였다. 비색 반응은 최대 400초 동안 6초마다 653nm에서 흡광도를 측정하는 동적 모드에서 UV-Vis로 순차적으로 측정했으며, TMB와 H2O2를 첨가하지 않은 샘플은 백그라운드(바탕신호) 흡광도 측정으로 사용하였다. 653nm에서의 결과 흡광도를 반응 시간에 대해 플롯하여 흡광도 대 시간 곡선을 얻었다. 653nm에서 얻어진 결과 흡광도값에서 바탕 신호 흡광도를 뺀 후 그 값을 다음의 수학식 1에 대입하여 나노 효소 활성을 계산하였다:

여기서, b 는 단위로 표시된 나노자임의 촉매 활성이다.
1 단위(unit)는 37℃에서 분당 1 μmol의 생성물을 촉매적으로 생성하는 나노자임의 양으로 정의되며, V 는 반응 용액의 총 부피(μL), ε는 비색 기질의 몰 흡광 계수로, 일반적으로 653nm에서 39,000 M-1 cm-1 의 최대값을 갖는다(TMB 경우).
l 은 큐벳에서 이동하는 빛의 경로 길이(cm), A는 바탕신호값을 뺀 후의 흡광도, ΔA/Δt는 653nm/분에서 흡광도의 초기 변화율입니다. 그런 다음 나노자임의 특정 활성(U/mg)은 나노자임 무게[m]에 대한 플롯 활성(b)의 기울기로서 mg 단위로 구했다(도 2b 참조).

나노 효소(b)의 촉매 활성은 질량에 선형적으로 비례하므로, [m]에 대한 b의 촉매 활성을 플롯하고 결과 직선의 기울기를 측정하여 특정 활성도 값을 수학식 2를 이용하여 결정할 수 있다.
<Steady-state kinetic 상수 측정>
각 나노자임 50μg을 아세테이트 완충액 0.2 M, pH 4.3의 1440 μL에 튜브안에 넣어서 녹인후 이에 다양한 양(0~72 μL)의 TMB 스톡 용액(DMSO 50 mM)을 첨가하였다. 그 후 최종 농도 5mM에 도달하도록 200μL의 H2O2를 혼합하였다. 혼합한 전체 샘플을 UV-Vis 큐벳에 넣고 UV-Vis 분광기로 측정하였다. 653nm에서 흡광도의 초기 변화율을 3분 동안 기록하였다(간격= 6초).
다음으로 "흡광도 대 시간" 플롯을 사용하여 각 반응의 초기 시점(초기 기울기)에서의 기울기를 구했다. 기질 농도에 대한 흡광도 증가율을 μM/분 단위로 변환하고 이를 기질 농도에 대해 플롯하였다. 그 다음 Michaelis Menten의 비선형 방정식 (수학식 3)에 핏팅하여 νmax 및 Km과 같은 knetic 파라미터를 구하였다.

수학식 3에서, νo는 초기 반응 속도이고, νmax는 포화 기질 농도에서 관찰되는 최대 반응 속도이다. [S]는 기질의 농도이고, Km은 Michaelis 상수이다. Km은 나노 효소의 기질에 대한 친화도를 반영하며 최대 속도의 절반에 해당하는 기질 농도로 정의된다. 다른 방법으로 이중 상호 플롯(Linewaver-Burk 플롯- 수학식 4)에서 Km과 νmax를 구할 수 있다. 그 결과를 도 3b에 나타내었다:

결과 및 평가
Mimicking enzyme-like activity
Fe3O4 NPs에 내재된 peroxydase활성이 처음 발견된 이후 resembling peroxidase, oxidase, haloperoxidase, 또는 superoxide dismutase와 유사한 촉매 활성을 보이는 다양한 종류의 나노자임이 개발되었다.
본 발명에서는 질화전이금속 기반 나노자임(예: W2N@PDC(10%), W2N@PDC(20%), VN@PDC (20%), VN@PDC (30%), CrN-1@MDC, 및 CrN-2@MDC) 5가지를 합성하였다. peroxydase 활성을 모방할 수 있는 나노자임의 효소 성능을 평가하였다. Horse radisch peroxidase (HRP)와 같은 peroxydase 유형 효소가 생의학 응용 분야에서 가장 널리 사용되는 효소라는 점을 고려해서 상기 나노자임을 합성하였다.
그 결과 본 발명의 실시예 1 내지 실시예 6에 따른 질화전이금속 나노자임은 비교예 1에 따른 Pt@ZDC와 비교되었다.
도 1은 본 발명의 일 실시예에 따라 TMB 기질을 촉매하는 다양한 나노자임의 peroxidase 활성에 대한 정성적 분석결과를 나타낸다. 일반적으로 W2N@PDC를 제외한 거의 모든 물질이 TMB와 반응할 때 파란색 변화를 유도하였다. H2O2만 첨가하면 파란색이 나타나지 않았다. 그러나 TMB와 H2O2를 모두 첨가하면 파란색의 진화가 가속화되었다. 이 경우 H2O2는 TMB의 산화에 필요한 산소 원자를 제공함으로써 효소 반응에서 중요한 역할을 할 수 있다.
초기 반응 시간(1~3분)에서 알 수 있듯이 5mM의 TMB/H2O2를 1mg의 VN@PDC (20%), VN@PDC (30%), CrN-1@MDC, 및 CrN-2@MDC로 촉매 처리했을 때 파란색 생성은 현저히 빨랐다. 초기 반응에서 파란색 생성물이 더 빨리 생성될수록 나노 효소가 기질 TMB의 산화를 촉진하는 데 효과적이라는 것을 나타내며, 이는 분석의 판독에 더욱 필수적이다.
나노자임의 정량적 정보를 얻기 위해 나노자임 반응의 결과로 생성되는 653nm의 청색 TMB의 흡수 특성을 조사하기 위해 UV-Vis 분광분석을 하였다.
도 2a는 본 발명의 일 실시예에 따른 UV-Vis 분광분석결과를 도시한 것이다. 도 2a를 참조하면, 모든 합성 나노효소 중 653nm에서 가장 높은 흡수 피크는 CrN-1@MDC와 CrN-2@MDC를 활용했을 때 관찰되었으며, 그 다음으로는 VN@PDC(30%)가 그 뒤를 이었다. 또한 VN@PDC(20%)의 경우 653nm에서 흡광도 피크가 Pt@ZDC와 유사한 결과를 나타냈다. 반면, W2N@PDC(10%) 및 W2N@PDC(20%)는 TMB/H2O2와 반응하는 능력이 더 낮게 나타났다.
또한, 도 2b는 본 발명의 일 실시예에 따른 나노자임의 질량에 대한 촉매 활성의 선형 관계의 기울기로 표시되는 특정 활성을 도시한 것이다. 도 2b를 참조하면, CrN-2@MDC는 특정 활성의 면에서 모든 합성 물질 중 특정 반응을 촉매하는 데 가장 효과적인 나노자임이라는 것을 확인할 수 있다. 이 비교에는 촉매 효율, 기질 특이성, 안정성 및 비용 효율성과 같은 요소가 포함될 수 있다. 가장 높은 특정 활성을 아는 것은 품질 관리 목적에 필수적이며, 상업적 사용을 위해 대량으로 생산되는 나노 효소의 성능의 일관성을 보장하는 것이 필요하다.
또한 Michaelis-Menten 방정식에 따라 반응 속도와 기질 농도를 연관시키는 나노 효소의 steady-state kinetics을 입증하는 것도 중요하다. 촉매 kinetic 상수를 구하기 위해서는 다양한 TMB 농도([S])에서의 초기 반응 속도(νo)를 미카엘리스-멘텐 방정식에 맞춰야 한다. 이렇게 얻은 동역학 파라미터를 사용하여 특정 기질에 대한 나노 효소의 기질 특이성(Km), 촉매 속도 상수(kcat) 및 최대 반응 속도(νmax)를 정량적으로 비교할 수 있다.
도 3a는 본 발명의 일 실시예에 따른 상이한 나노자임에 의하여 활성화된 TMB 기질의 Michaelis-Menten 결과를 도시한 것이다. 도 3a를 참조하면, 반응 속도의 초기 급격한 증가와 기질 농도가 증가함에 따라 정체되는 쌍곡선 곡선은 W2N@PDC(10%), VN@PDC(30%), CrN-1@MDC, CrN-2@MDC, 및 Pt@ZDC에 의해 달성되었다. 매우 낮은 기판 농도에서는 곡선이 선형으로 나타나고 초기 속도는 기판 농도에 정비례한다. 기질 농도가 높으면 나노자임이 포화 상태가 되어 모든 나노자임 활성 부위가 점유되면서 최대 속도(νmax)가 일정해 진다. 그러나 W2N@PDC(20%)와 VN@PDC(20%)는 선형 플롯을 보여줌으로써 Michaelis-Menten 방정식 피팅에 부합하지 않는다.
도 3b는 본 발명의 일 실시예에 따른 kinetic 변수를 보다 정확하게 분석하기 위해 비선형 Michaelis-Menten 곡선을 선형인 Linewaver-Burk 플롯으로 변환한 것을 도시한 것이다. 도 3b를 참조하면, 이러한 선형화는 데이터 분석을 간소화하고 νmax 및 Km와 같은 매개변수를 더 쉽게 결정할 수 있게 해 준다. 가장 높은 νmax 결과는 최대 44.25 μM/min의 CrN-2@MDC에 도달했으며, 이는 모든 나노자임 활성 부위가 TMB 기질로 포화되었을 때 반응의 이론적 최대 속도를 나타낸다.
CrN-2@MDC의 νmax값은 대조군 Pt@ZDC보다 6배 높았다. 이는 또한 CrN-2@MDC 나노 효소가 TMB 기질을 파란색으로 전환하는 촉매 작용에 매우 효율적이라는 것을 시사한다. 이는 회전율이 높아 단위 시간당 나노자임이 기질 분자를 처리할 수 있는 용량이 더 크다는 것을 확인할 수 있다. 이러한 결과는 표지 물질로서 질화전이금속을 사용한 결과에 기인하는 것이라는 것을 알 수 있다.
νmax는 매개변수로서 중요한 의미를 갖지만, 일반적으로 Km와 같은 다른 동역학 인자와 함께 평가될 수 있다. Km은 반응 속도가 νmax의 절반에 도달하는 기질 농도를 나타낸다. Km 값이 낮을수록 효소의 기질에 대한 친화력이 높다는 것을 나타낸다.
결과는 다음과 같은 순서로 Km 값이 가장 낮은 것부터 가장 높은 것까지 다양하다. W2N@PDC(10%), Pt@ZDC, VN@PDC(30%), CrN-2@MDC, CrN-1@MD, VN@PDC(20%), W2N@PDC(20%) 순이다. W2N@PDC(10%) 및 Pt@ZDC가 가장 낮은 Km 값을 보였지만, νmax는 여전히 CrN@C보다 낮다.
CrN-2@MDC가 653nm에서 가장 높은 흡광도, 가장 높은 비활성도, 가장 높은 νmax 값을 보였고, 비슷한 Km 값을 보인다는 점을 고려하면 CrN-2@MDC가 모든 합성 물질 중에서 우수한 나노 효소라는 것을 확인할 수 있다.
전술한 내용은 후술할 발명의 청구범위를 더욱 잘 이해할 수 있도록 본 발명의 특징과 기술적 장점을 다소 폭넓게 상술하였다. 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명이 그 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허청구범위에 의하여 나타내어지며, 특허청구범위 그리고 그 균등 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (4)
- 지지체;상기 지지체 표면에서 고정된 포획 프로브;상기 포획 프로브와 시료를 사이에 두고 샌드위치 형태로 특이적 결합을 하는 검출 프로브; 및상기 검출 프로브에 연결되어 기질과 반응하는 질화전이금속 나노자임으로 구성된 표지;를 포함하는 바이오센서로서,상기 질화전이금속은 질화텅스텐(W2N), 질화바나듐(VN), 및 질화크롬(CrN) 중에서 선택된 하나이고,상기 질화전이금속 나노자임은 질화전이금속과 탄소를 결합하여 형성되고, 효소로서 작용하며 기질과의 반응에 의하여 시각적 신호를 발생하는 것을 특징으로 하는 바이오센서.
- 제 1 항에 있어서,상기 기질은 3,3',5,5'-테트라메틸벤지딘[3,3',5,5'-tetramethylbenzidine: TMB], 3-아미노-9-에틸카바졸[3-amino-9-ethylcarbazole], o-페닐렌디아민[o-phenylenediamine], 3,3'-디아미노벤지딘[3,3'-diaminobenzidine], 2,2'-아지노비스(3-에틸벤조티아졸린-6-술폰산)[2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)], 및 10-아세틸-3,7-디하이드록시페녹사진[10-acetyl-3,7-dihydroxyphenoxazine] 등으로 이루어진 군으로부터 선택된 하나 이상을 포함하는 것을 특징으로 하는 바이오센서.
- 제1항에 있어서,상기 시료는 인간 세럼, 플라즈마, 혈액 및 소변 시료 중 어느 하나 이상인 것을 특징으로 하는 바이오센서.
- 제2항에 있어서,상기 나노자임은 기질과의 반응에 의하여 색상이 파란색으로 변경되는 것을 특징으로 하는 바이오센서.
