Abstract
生物载片及其制备方法,该生物载片包括载片本体,载片本体具有多个开口位于同侧的槽结构,所述多个槽结构包括第一槽和至少一个与该第一槽距离最近且深度不同的第二槽。生物载片中距离最近的槽结构的深度不同,能减少相邻槽结构之间样本产生的光信号的互相干扰,增加了荧光光学系统的分辨率,提升了生物载片检测过程中的信噪比;且可以通过提升槽结构的密度来提升处理通量,并通过控制槽结构的深度提高信号质量,以实现高通量和高信号质量的兼容;另外,该生物载片可以重复使用,有效的降低了使用成本。
Description
本申请涉及生物样本检测技术领域,尤其涉及一种生物载片及其制备方法。
在生物样本(样品)检测技术领域中,对于微观样本(如基因测序中的DNA样本),通常需要用生物载片(如测序芯片)承载,然后采集其荧光信号。然而,现有生物载片存在光学信号相互干扰的问题。
以测序芯片为例,现有的测序芯片一般采用化学修饰表面(如APTMS(3-氨基丙基-三甲氧基硅烷)等修饰的表面)来吸附生物样品(例如,DNA(脱氧核糖核酸)分子纳米球(DNB)等),通过光刻与化学气相沉积(CVD)的方式在硅基底表面制作出化学修饰图案化阵列位点,然后利用表面的化学修饰(例如APTMS)进行生物样品的吸附。
但现有的测序芯片上相邻阵列位点上的样品存在互相干扰的风险,导致信噪比降低;另外,由于光学信号会有一定散射和衍射,受到光学系统的限制,现有测序芯片很难在不影响测序信号质量的条件下持续提升芯片密度或通量。
发明内容
为了解决以上缺陷中的至少之一,本申请有必要提出一种生物载片,该生物载片有利于降低相邻阵列位点上样品之间的相互干扰,提高信噪比;且能够在不影响检测信号质量的条件下持续提升芯片密度或通量。
另,本申请还提供了前述生物载片的制备方法。
第一方面,本申请实施例提供了一种生物载片,所述生物载片包括载片本体,所述载片本体具有多个开口位于同侧的槽结构,所述多个槽结构包括第一槽和至少一个与所述第一槽距离最近且深度不同的第二槽。
在一些可能的实施例中,所述第一槽和与所述第一槽距离最近的所述第二槽之间的深度差大于或等于20nm。
在一些可能的实施例中,所述多个槽结构还包括与所述第一槽相邻的第三槽,所述第三槽与所述第一槽之间的距离大于所述第二槽与所述第一槽之间的距离,所述第三槽与所述第一槽的深度可以相同。
在一些可能的实施例中,所述槽结构的深度为20nm~150nm。
在一些可能的实施例中,所述载片本体包括基底以及位于所述基底上的至少一吸附结构层,所述槽结构形成于所述吸附结构层上,所述第一槽和所述第二槽伸入所述吸附结构层的深度不同。
在一些可能的实施例中,所述吸附结构层包括:位于所述基底的表面上的吸附层和位于所述吸附层的表面上的隔离层,其中,所述吸附层用于吸附生物样本,所述槽结构至少贯穿所述隔离层,所述吸附层由所述槽结构露出。
在一些可能的实施例中,所有或部分所述槽结构伸入所述吸附层。
在一些可能的实施例中,所述载片本体包括多层层叠设置的所述吸附结构层,所述第一槽和所述第二槽伸入不同层的所述吸附结构层。
在一些可能的实施例中,所述槽结构的内径为100nm~300nm;和/或,相邻两个所述槽结构的中心之间的距离为200nm-1000nm。
在一些可能的实施例中,所述吸附层包含用于吸附生物样本的吸附材料,所述吸附材料包括金属氮化物、金属氧化物以及金属中的至少一种,及/或,所述隔离层的材质包括金属氮化物和金属氧化物中的至少一种。
在一些可能的实施例中,所述基底与所述吸附结构层之间还设有缓冲层,所述缓冲层的材质包括氧化物和氮化物中的至少一种。
第二方面,本申请实施例提供了一种生物载片的制备方法,该制备方法包括:
于所述生物载片的载片本体上形成开口位于同侧的多个槽结构,其中,所述多个槽结构包括第一槽和至少一个与所述第一槽距离最近且深度不同的第二槽。
在一些可能的实施例中,所述于所述生物载片的载片本体上形成开口位于同侧的多个槽结构的步骤包括:
于所述载片本体上同时形成所述第一槽和所述第二槽;或
于所述载片本体上分别形成所述第一槽和所述第二槽。
在一些可能的实施例中,所述于所述载片本体上同时形成所述第一槽和所述第二槽的步骤包括:
于所述载片本体的表面上形成具有第一蚀刻孔和第二蚀刻孔的图案化膜;以及
去除所述第一蚀刻孔对应的部分所述载片本体以在所述载片本体上形成所述第一槽,并去除所述第二蚀刻孔对应的部分所述载片本体以在所述载片本体上形成所述第二槽。
在一些可能的实施例中,所述于所述载片本体上分别形成所述第一槽和所述第二槽的步骤包括:
于所述载片本体的表面上形成具有第一蚀刻孔的图案化膜;

本申请实施例提供的生物载片及其制备方法,生物载片的槽结构中距离最近的第一槽和第二槽之间的深度设置为不同,这种具有不同深度的槽结构能够大大减少相邻槽结构之间样本的互相干扰,增加了荧光光学系统的分辨率,提升了生物载片在检测过程中的信噪比;且可以通过提升槽结构的密度来提升生化反应的通量,并通过控制槽结构的深度提高光信号质量,例如应用于测序领域,可以实现高测序通量和高测序信号质量的兼容;另外,该生物载片可以重复使用,有效的降低了使用成本。

请参阅图1A、图1B与图2所示,本申请实施例提供了一种生物载片100,所述生物载片100包括载片本体10,所述载片本体10具有多个开口位于同侧的槽结构4,所述多个槽结构4包括第一槽4a和至少一个与第一槽4a距离最近且深度不同的第二槽4b。该槽结构4可以用于容置生物样本,以为生物样本提供反应场所,即,每个槽结构4可以作为生物载片100的一个反应区域,生物样本在槽结构4内进行生化反应,之后可以检测产物样本所发出的光信号,实现对产物的检测。具体地,该生物载片100可以用于基因测序,例如可以是测序芯片,可以理解的,该生物载片100还可以用于其他生化用途。
其中,生物载片100中每一个第一槽4a都和与之距离最近的第二槽4b具有不同的深度,从而使得每个第一槽4a与周围距离该第一槽4a最近的第二槽4b内的生物样本位于不同的平面内,这样能降低位于距离最近的不同反应区域内样本之间互相干扰(如荧光信号的干扰)的风险,有利于提高检测(例如测序)过程中的信噪比。
在一些实施例中,所述多个槽结构4还包括与第一槽4a相邻的第三槽4g,第三槽4g与第一槽4a之间的距离大于第二槽4b与第一槽4a之间的距离,第三槽4g与第一槽4a的深度可以相同。如图1A所示,第一槽4a的周围存在多个第二槽4b和多个第三槽4g,呈阵列方式排布,多个第二槽4b大致位于第一槽4a的四边,第三槽4g大致位于第一槽4a的斜对角的位置,第三槽4g与第一槽4a的距离相较于第二槽4b要远一些,此时,第三槽4g与第一槽4a内的生物样本反应所产生的光信号不易发生干扰,第三槽4g的深度可以与第一槽4a的深度相同。可以理解的,第三槽4g的深度也可以与第一槽4a的不同。
在一些实施例中,可以通过一预设距离来限定相邻两个槽结构4之间的深度关系,
即,当任意相邻的两个槽结构4之间的距离(即相邻两个槽的中心之间的距离)小于或等于该预设距离时,这两个相邻的槽结构4具有不同的槽深,当任意相邻的两个槽结构4之间的距离大于该预设距离时,这两个相邻的槽结构4可以具有相同或者不同的槽深。而这个预设距离是相邻两个槽结构4内的样本之间光信号发生干扰的最大距离,即,相邻两个槽结构4之间的距离小于或等于这个预设距离时,两个槽结构4内的光信号就会发生干扰,相邻两个槽结构4之间的距离大于预设距离,两个槽结构4内的光信号不会发生干扰或者干扰很弱。如图1A所示,所述预设距离例如可以为相邻的第二槽4b与第一槽4a之间的距离L,当在第二槽4b与第一槽4a之间需要进一步形成其他槽结构,以增加槽结构4的密度时,槽的深度便需要设置的与第一槽4a和第二槽4b不同,以降低光信号的干扰。由此可见,所述预设距离可以根据实际不同槽结构4内样本发生光信号干扰的情况而定。
另外,通过将生物载片100上的不同槽结构4设计成具有不同的槽深,减小相邻荧光信号之间散射和衍射的干扰,这样可以在光信号干扰的范围内通过调整槽深来增加槽结构4的数量,进而增加槽结构4的密度,而且还能获得较高的信号质量,实现槽结构4的密度与信号质量的兼容,有利于提升生物载片100的在生化处理过程中的通量。
如图1A与图1B所示的生物载片100具有两种深度的槽结构4,其中第一槽4a和第三槽4g之间的间距较大,可以是同一深度的槽,第二槽4b的深度与第一槽4a和第三槽4g不同。如图2所示的生物载片100具有四种深度的槽结构,槽结构4之间的间距缩小,槽结构4的密度增加,当图1A与图2两种生物载片100的载片本体10的尺寸基本一致的情况下,通过调整相邻槽结构4的槽深,可以有效增加槽结构4的密度,因此,可以通过槽结构4的槽深以及槽间距的设计,进而兼顾生物载片100的通量和检测信号质量的双重提高,例如在测序过程中可以实现高测序通量和高测序信号质量的兼容。
在一些实施例中,第一槽4a与的第二槽4b之间的深度之差可以大于或等于20nm,通过控制槽深之差在20nm以上,可以进一步提高距离最近的相邻反应区域内样本的发光面之间的高度差,从而进一步降低互相干扰的风险,提高信噪比。可以理解的,具体两个距离最近的槽结构4的槽深之差可以根据实际情况进行设计,例如可以根据槽结构4的密度需求来设计深度差。例如,生物载片100的通量需求较高时,可以将槽结构4的密度设计的大一些,此时,相邻槽结构4之间的距离较近,光信号干扰的程度较大,为了降低光信号干扰,可以将相邻槽结构4的深度差设计的大一些;当生物载片100的通量需求不高时,可以将槽结构4的密度设计的小一些,此时,相邻槽结构4之间的距离较远,光信号干扰的程度相对较低,可以将相邻槽结构4的深度差设计的小一些。
在一些实施例中,所述槽结构4的深度可以为20nm~150nm,进一步为30nm~100nm。示例性的,槽结构4的深度可以为20nm、30nm、40nm、50nm、60nm、70nm、80nm、90nm、100nm、110nm、120nm、130nm、140nm或150nm等。前述槽结构4的深度范围较大,通过控制不同槽结构4槽深均在以上范围内,且控制距离最近的两个槽结构4的深度之差大于或等于20nm,能够设计出更多种不同深度的槽结构4,有利于在确保具有较高信号质量的前提下,进一步提高槽结构4的密度,提高处理通量,实现高通量与高信号质量的兼容。
多个槽结构4可以呈阵列排布,确保每个槽结构4周围的距离最近的其他槽结构4与其具有不同的深度即可。可以根据生物载片100中每个反应区域所需的容量来设计所述槽结构4的形状以及槽结构4沿径向方向的尺寸等。在一些实施例中,所述槽结构4可以为圆槽,内径可以为100nm~300nm,进一步为120nm~250nm,进一步为150nm~200nm。示例性的,槽结构4的内径可以为100nm、110nm、120nm、130nm、140nm、150nm、160nm、170nm、180nm、190nm、200nm、210nm、220nm、230nm、240nm、250nm、260nm、270nm、280nm、290nm或300nm等。
在一些实施例中,相邻两个所述槽结构4的槽中心之间的距离(即槽间距)可以为200nm~1000nm,进一步为250nm~800,进一步为300nm~600。示例性的,相邻两个所述槽结构4的槽中心之间的距离可以为200nm、250nm、300nm、350nm、400nm、450nm、500nm、550nm、600nm、650nm、700nm、750nm、800nm、850nm、900nm、950nm或1000nm等。可以通过调控槽间距和槽深,提高槽结构4的密度,提高生化处理通量。另外,由于相邻的槽结构4具有不同的深度,可以保证在槽间距较小的情况下,生物载片100依然具有较高的信号质量,因此,可以最大程度地提高槽结构4的密度,以制备出更高通量的生物载片,实现高通量与高信号质量的兼容。
在载片本体10上形成具有不同深度的槽结构4时,可以采用分步成型的方式,例如针对图1A中槽结构4的结构形式,即第一槽4a和第二槽4b的深度不同,第一槽4a和第三槽4g的深度相同,此时采用两种类型的掩膜版即可实现,如图3A所示的第一掩膜版具有第一蚀刻孔,对应一种深度的槽结构4(例如第一槽4a和第三槽4g),如图3B所示的第二掩膜版具有第二蚀刻孔,对应另一种深度的槽结构4(例如第二槽4b)。可以通过第一掩膜版和第二掩膜版这两种排布方式不同的掩膜版,并采用分次图案化的方式实现如图1A所示具有两种深度的槽结构4的成型。可以理解的,在其他实施例中,如图4A至图4D所示的第三掩膜版至第六掩膜版这四种槽排布方式不同的掩膜版,并采用分次图案化的方式,实现如图2所示的具有四种不同深度的槽结构4的成型。还可以理解
的,可以采用更多类型的掩膜版实现更多深度不同的槽结构4的成型,以提高槽结构4的排布密度。还可以理解的,不同深度的槽结构4的成型不局限于以上分布成型的方式,还可以采用具有多种蚀刻孔的掩膜版一步形成。
请参阅图5与图6所示,生物载片100中的所述载片本体10包括基底1、以及层叠设于基底1的表面上的至少一层吸附结构层20,槽结构4形成于吸附结构层20上,其中,第一槽4a与第二槽4b伸入吸附结构层20的深度不同。
在一些实施例中,载片本体10包括一层吸附结构层20。所述吸附结构层20包括位于基底1的表面上的吸附层2以及位于吸附层2的表面上的隔离层3。其中,基底1例如可以是硅基底(硅材料板)。所述吸附层2用于吸附生物样本,例如,吸附层2可以通过能够吸附生物样本的吸附材料形成。所述槽结构4至少贯穿所述隔离层3,以使所述吸附层2能够由所述槽结构4露出,即吸附层2构成所述槽结构4的底部,进而实现对生物样本的吸附固定。
在一些实施例中,当同一层隔离层3的不同位置厚度相等时,即隔离层3具有均一的厚度,此时为了形成不同深度的槽结构4,所有或部分所述槽结构4需要延伸至所述吸附层2内,但需要留一部分吸附层2来吸附生物样本。可以理解的,也可以将隔离层3的厚度设置为非均一的,这样可以通过隔离层3的厚度差异实现距离最近的槽结构4之间深度差的设计。
在一些实施例中,如图5所示,可以将第一槽4a仅贯穿隔离层3,而第二槽4b贯穿隔离层3之后再延伸至一定厚度的吸附层2内,这样便可以形成不同深度的槽结构4。可以理解的,延伸至吸附层2内的这部分槽结构4可以根据需要使槽结构4延伸至吸附层2的不同厚度位置,以形成多种深度类型的槽结构4。如图6所示,所有槽结构4均伸入吸附层2,例如第一槽4c和第二槽4d均延伸至吸附层2,且根据深度差使第一槽4c和第二槽4d延伸至吸附层2的不同深度,以形成不同槽深的槽结构4。
在一些实施例中,所述吸附材料可以包括金属氮化物(如氮化钛)、金属氧化物(如氧化钛、氧化锆、氧化锌)以及金属(如银)等中的至少一种。例如,所述吸附材料可以是金属氮化物(例如氮化钛)或金属氧化物(例如氧化钛)。
在一些实施例中,所述吸附层2的厚度可以为20nm~500nm,进一步为30nm~300nm,进一步为50nm~100nm。示例性的,所述吸附层2的厚度可以为20nm、30nm、40nm、50nm、60nm、70nm、80nm、90nm、100nm、120nm、150nm、180nm、200nm、250nm、300nm、350nm、400nm、450nm或500nm等。当需要设计多种槽深的槽结构4时,可以将吸附层2的厚度设计的厚一些。
在一些实施例中,所述隔离层3的材质包括氧化硅、氮化硅、或其他氧化物或其他氮化物。
在一些实施例中,所述隔离层3的厚度可以为20nm~150nm,可以根据实际槽结构4的深度来设计隔离层3的厚度。
在一些可实施例中,所述基底1与所述吸附层2之间还设有缓冲层5,所述缓冲层5的材质包括氧化硅、氮化硅、或其他氧化物或其他氮化物。在一些实施例中,可以在基底1的表面先沉积一层20nm~1000nm的缓冲层5,例如SiO2层。通过在基底1的表面上增加缓冲层5,可以提高吸附层2在基底1上的附着力,提升吸附层2的稳定性,提升生物载片100的质量。
本申请实施例提供的生物载片100的结构进行了全面优化,生物载片100中距离最近的第一槽4a和第二槽4b具有不同的深度,这种具有不同深度的槽结构4能够大大减少相邻反应区域之间样本的光信号之间互相干扰,增加了荧光光学系统的分辨率,提升了生物载片100在生化处理(例如测序)过程中的信噪比;而且可以通过调整不同槽结构4的深度和间距,进而提升槽结构4的密度,有利于制备更高通量的生物载片100,实现较高的信号质量与较高的处理通量的兼容;另外,该生物载片100可以重复使用,有效的降低了使用成本。
请参阅图7所示,本申请实施例提供了另一种生物载片200,该生物载片200与前述实施例中的生物载片100的结构基本相同,区别在于:所述生物载片200的所述基底1上层叠设置有多层所述吸附结构层20和20’,所述第一槽4e和所述第二槽4f伸入不同层的所述吸附结构层20和20’,具体地,第一槽4e伸入吸附结构层20’,第二槽4f伸入吸附结构层20,从而实现不同深度的第一槽4e与第二槽4f的成型。
具体地,每层所述吸附结构层20和20’上的槽结构4均贯穿位于最外层的所述隔离层3’,以使不同层的吸附层2和2’由对应的槽结构4露出。也就是,每层吸附结构层20对应的槽结构4均贯穿位于该吸附结构层20远离基底1一侧的其他所有吸附结构层20’。如图7所示,即第一槽4e和第二槽4f的开口均位于最外层的所述隔离层3’远离所述基底1的表面。
本实施例中的生物载片200,通过增加吸附结构层的层数,有利于形成多种不同深度的槽结构4,尤其是槽深差距较大的槽结构4,以实现更高密度的槽结构4的设计,进一步提高生化处理的通量。与此同时,由于位于不同层吸附结构层上的槽结构4之间具有较大的深度差,也进一步提高了检测过程中信号抗干扰的能力,使信号质量得到进一步提升。
请参阅图8所示,本申请实施例还提供了前述生物载片的制备方法,具体包括以下步骤:
步骤S1,于生物载片的载片本体上形成开口位于同侧的多个槽结构,其中,所述多个槽结构包括第一槽和至少一个与所述第一槽距离最近且深度不同的第二槽。
步骤S2,于所述载片本体上形成与所述第一槽相邻的第三槽,所述第三槽与所述第一槽之间的距离大于所述第二槽与所述第一槽之间的距离,所述第三槽与所述第一槽的深度可以相同。
其中,在步骤S1中,根据第一槽和第二槽同步成型和分步成型,可以包括以下两种具体的实现方式。
同步成型法具体为:于载片本体上同时形成第一槽和第二槽。可以理解的,当需要成型第三槽时,第三槽也可以与第一槽和第二槽同时成型,当需要成型更多类型槽时,也可以采用同步成型的方式成型。
请参阅图9所示,于载片本体上同时形成第一槽和第二槽具体包括以下步骤:
步骤S11,于载片本体的表面上形成具有第一蚀刻孔和第二蚀刻孔的图案化膜。
步骤S12,去除所述第一蚀刻孔对应的部分所述载片本体以在所述载片本体上形成第一槽,并去除所述第二蚀刻孔对应的部分所述载片本体以在所述载片本体上形成第二槽。
分步成型法具体为:于载片本体上分别形成第一槽和第二槽。
请参阅图10所示,于载片本体上分别形成第一槽和第二槽具体包括以下步骤:
步骤S11’,于载片本体的表面上形成具有第一蚀刻孔的图案化膜。
步骤S12’,去除所述第一蚀刻孔对应的部分所述载片本体,以在所述载片本体上形成第一槽。
步骤S13’,于所述载片本体的表面上形成具有第二蚀刻孔的图案化膜。

步骤S101,于基底的表面上形成至少一层吸附结构层。
具体地,所述吸附结构层包括位于所述基底的表面上的吸附层和位于所述吸附层的表面上的隔离层,所述吸附层用于吸附生物样本。
步骤S102,于所述吸附结构层上形成多个槽结构,并使多个槽结构中的第一槽和第
二槽伸入吸附结构层的不同深度。
具体地,当吸附结构层包括层叠设置的吸附层和隔离层时,所述槽结构至少贯穿所述隔离层,以使所述吸附层由所述槽结构露出。具体地,第一槽仅贯穿隔离层,第二槽贯穿隔离层后延伸至部分吸附层。
请参阅图12所示,在其他实施例中,当基底上形成多层吸附结构层时,生物载片的制备方法包括以下步骤:
步骤S101’,于基底的表面上形成多层吸附结构层。
步骤S102’,于每层所述吸附结构层上形成槽结构,且每个所述槽结构贯穿最外层的所述吸附结构层中的隔离层,以使每层所述吸附结构层中的吸附层均由对应的所述槽结构露出。
即,多个槽结构贯穿位于不同的层中的隔离层,以使位于不同层的吸附层均由对应的槽结构露出。
本申请实施例提供的生物载片100(200)的制备方法,在制备工艺上进行了全面优化,通过在基底1上沉积氮化钛等吸附层2(2’)以及氧化硅等隔离层3(3’),然后通过掩膜版设计,采用分次曝光显影刻蚀的方法,对隔离层3(3’)进行图案化以形成槽深不同的槽结构4的阵列,使相邻的两个槽结构4具有不同的槽深,从而提高了检测的信号质量,同时能够制作出槽密度更大的生物载片,进而提高处理通量。该制备方法的工艺简单,不同槽深的槽结构4的尺寸精度高,从而提升了生物载片100(200)的生化处理过程(例如测序过程)的稳定性和质量。
以下通过具体实施例对前述生物载片的制备方法进行进一步说明。
实施例一
请结合参阅图13A至图13F,一并结合参阅图5,当需要形成一层吸附结构层20时,生物载片100的制备方法包括以下步骤:
步骤S11a,如图13A所示,于基底1的表面上形成吸附层2。
在本步骤中,例如可以提供8英寸或12英寸的硅基底(晶圆),例如厚度约为725μm的硅基底。当然本实施例并不具体限定基底类型及尺寸,均可根据实际需求进行相应的选择及调整。
在本步骤中,在形成所述吸附层2之前,所述制备方法还包括:
于所述基底1的表面上形成缓冲层5,其中,所述吸附层2位于所述缓冲层5的表面上。
在本实施例中,所述缓冲层5通过在所述基底1的表面沉积缓冲材料的方式形成,所述缓冲材料包括氧化硅、氮化硅、或其他氧化物或其他氮化物。在本步骤中,作为可选实施方式,在硅基底1的表面上采用化学气相沉积的方式沉积氧化硅以形成厚度范围为20nm~1000nm的缓冲层5,例如沉积35nm左右厚的二氧化硅作为缓冲层5,可根据实际需求进行相应的选择及调整。可以理解的,也可以不在基底1上形成缓冲层5。
在本步骤中,所述吸附层2的形成方法包括:
将用于吸附生物样本的吸附材料沉积于所述基底1的表面上,以形成所述吸附层2。其中,所述吸附材料包括氮化钛、氧化钛、银、氧化锆、氧化锌等中的至少一种,例如可以是金属氮化物(氮化钛)或金属氧化物(氧化钛)。在本实施例中,吸附材料可以为TiN,吸附层为TiN层,但并不仅限于此,可根据实际需求或可能出现的需求进行相应的调整及选择。
在本步骤中,在缓冲层5的表面上沉积用于吸附生物样品的吸附材料以形成厚度范围为20nm~500nm的用于吸附生物样品的吸附层2。例如,利用物理气相沉积的方式在缓冲层5上沉积一层60nm左右厚的氮化钛吸附材料。
在本实施例中,吸附层可以以吸附薄膜的形式体现,该吸附薄膜的厚度太薄则难以实现其沉积工艺,存在均匀性问题,若厚度太厚则会影响到生化反应(例如测序)的质量,因此可将吸附层2的厚度控制在上述可选厚度范围之内,从而保证工艺稳定性的同时保证生化反应质量。
在本实施例中,生物样品例如可以为DNA纳米球等,但不仅限于此,可根据实际需求进行相应的选择及调整。
步骤S12a,如图13A所示,于所述吸附层2的表面上形成隔离层3。
在本步骤中,所述隔离层3通过在所述吸附层2的表面上沉积隔离材料的方式形成,所述隔离材料包括氧化硅、氮化硅、或其他氧化物或其他氮化物。
在一些实施例中,在用于吸附生物样品的吸附层2上沉积氧化硅材料以形成厚度范围为20nm~150nm的隔离层3。例如,利用化学气相沉积的方式沉积45nm左右厚度的二氧化硅作为隔离层3。隔离层3的厚度太薄则会在后续反应过程中容易被腐蚀,导致影响整个工艺稳定性,且不易制备不同槽深的槽结构;隔离层3的厚度太厚则会影响荧光亮度导致信号弱且会影响槽内试剂的替代,影响到生化反应(例如测序)质量,因此可将隔离层3的厚度控制在上述可选厚度范围之内,从而保证工艺稳定性的同时保证生化反应的质量。
步骤S13a,如图13A与图13B所示,于所述隔离层3的表面上形成具有第一蚀刻
孔61的第一图案化膜60。
其中第一图案化膜60的图案化方法有两种,分别为蚀刻法和压印法,所谓图案化,是指多个槽按照特定图案形式排布,图案化膜即形成有特定图案排布形式的槽结构的膜层。
采用蚀刻法形成第一图案化膜60的方法具体包括以下步骤:
第一步,如图13A所示,于所述隔离层3的表面上形成抗反射层40。
所述抗反射层40通过在所述隔离层3的表面上沉积抗反射材料的方式形成,所述抗反射材料包括氮氧化硅或底部抗反射涂层(Bottom Anti-Reflection Coating,BARC)。
在一些实施例中,在隔离层3上沉积抗反射材料以形成厚度范围为50nm~70nm的抗反射层40。抗反射材料可以为SiON材料,抗反射层40可以为SiON层,当然本实施例并不具体限定抗反射材料层类型及厚度,均可根据实际需求进行相应的选择及调整。
第二步,如图13A所示,于所述抗反射层40的表面上形成光刻胶层50。
其中,在形成光刻胶层50之前,还需于所述抗反射层40的表面上形成增粘涂层(图未示),所述光刻胶层50位于所述增粘涂层的表面上。
具体地,先在抗反射层40的表面上旋涂一层HMDS作为增粘涂层,之后再在增粘涂层的表面旋涂光刻胶以形成光刻胶层50。
在本步骤中,作为可选实施方式,光刻胶层50的厚度范围可以为250nm~350nm。
第三步,如图13B所示,在所述光刻胶层50形成第一蚀刻孔61,得到第一图案化膜60。
具体地,采用具有对应第一槽的掩膜版(如图3A所示的第一掩膜版的结构),通过DUV光刻机进行曝光显影等操作在光刻胶层50上形成了规则排布的圆槽阵列图案(即第一蚀刻孔61)。
采用压印法形成第一图案化膜的方法具体包括以下步骤:
第一步,于所述隔离层的表面上形成压印胶层。
第二步,在所述压印胶层形成第一蚀刻孔,得到第一图案化膜。
步骤S14a,如图13C所示,至少去除所述第一蚀刻孔61对应的所述隔离层3,以露出所述吸附层2,从而在所述吸附结构层20上形成第一槽4a。
本实施例中,仅去除第一蚀刻孔61对应的隔离层3,使吸附层2露出,则形成第一槽4a。因本实施例中,第一槽4a通过仅去除隔离层3形成,为了得到合适的槽深,隔离层3的厚度可以根据实际需要沉积的适当厚一些。
可以理解的,当采用光刻胶形成第一图案化膜60时,在去除隔离层3时还需去除增
粘涂层和抗反射层40。本实施例中采用干法蚀刻的方式去除增粘涂层、抗反射层40和隔离层3。
在本步骤中,作为可选实施方式,对抗反射层40及隔离层3(主要用于形成槽结构4的侧壁)进行干法蚀刻以在抗反射层40及隔离层3上形成预设槽径的若干槽结构4。具体地,槽结构4的预设内径的范围可以为100nm~300nm,可根据实际需求进行相应的调整及设定。
步骤S15a,如图13D与图13E所示,于所述隔离层3的表面上形成具有第二蚀刻孔31的第二图案化膜30,并使所述第二图案化膜30延伸至所述第一槽4a。
在制作第二次蚀刻时,第二图案化膜30需将第一槽4a填充上,这样可以保护第一槽4a不受影响。
其中,第二图案化膜30的形成方法与第一图案化膜60的形成方法相同,请参见前述步骤S13a,此处不做过多赘述。区别在于:形成第二图案化膜30采用的掩膜版上的槽对应第二槽,如图3B所示的第二掩膜版的结构。
步骤S16a,如图13F所示,去除所述第二蚀刻孔31对应的所述隔离层3和部分所述吸附层2,以露出所述吸附层2,从而在所述吸附结构层20上形成第二槽4b。
步骤S17a,结合参阅图5所示,去除所述第二图案化膜30,其中,所述第一槽4a和所述第二槽4b的槽深不同,所述第一槽4a和所述第二槽4b构成了所述多个槽结构4。
具体地,先采用干法蚀刻和湿法蚀刻的方式把第二图案化膜30和第一图案化膜60去除,然后再用干法蚀刻的方式把抗反射层40蚀刻掉。可以理解的,也可以在步骤S15之前,先将第一图案化膜60去除,为了简化工艺,本实施例选择将两个图案化膜在最后一起去除。
在本实施例中,该第一槽4a和第二槽4b的槽径大致相同,大小可为200nm左右,第一槽4a和第二槽4b之间的距离是715nm,第一槽4a的深度可为45nm,第二槽4b的深度可为65nm,槽的底部是氮化钛用作吸附DNA纳米球,槽的间隔为氧化硅不特异性吸附DNA纳米球。
以上步骤形成了具有两种槽深的槽结构4,可以理解的,当需要形成多于两种槽深的槽结构时,重复步骤S16a至步骤S18a即可。
实施例二
下面详细说明采用压印法制备生物载片的方法,具体包括以下步骤:
步骤S11b,提供基底。
步骤S12b,在基底的表面上沉积用于吸附生物样品的吸附材料以形成吸附层。
本实施例中,在基底的表面上可以不沉积缓冲层,直接沉积吸附层,该步骤与前述步骤S11a基本相同,具体请参阅前述步骤S11a,此处不作过多赘述。可以理解的,本步骤也可以在基底上形成缓冲层。具体可参照前述步骤S11a,此处不作过多赘述。
步骤S13b,在吸附层的表面上沉积氧化硅材料以形成隔离层。
该步骤与前述步骤S12a基本相同,具体请参阅前述步骤S12a,此处不作过多赘述。
步骤S14b,在隔离层的表面上旋涂一层增粘剂,然后再在上面旋涂一层压印胶层。
步骤S15b,采用提前准备好的工作模具压印在压印胶层上,再经过固化和脱模工艺后,将工作模具的图案转移到压印胶层上,从而形成具有第一蚀刻孔的第一图案化膜。
步骤S16b,利用干法蚀刻的方式把第一蚀刻孔内的残胶和隔离层刻掉,形成第一槽,并使第一槽底部的吸附层露出。
步骤S17b,于隔离层的表面上形成具有第二蚀刻孔的第二图案化膜,并使所述第二图案化膜延伸至所述第一槽。
第二图案化膜的形成方法与前述制作第一图案化膜的形成方法相同,请参见前述步骤S14b和步骤S15b,此处不做过多赘述。区别在于:形成第二图案化膜采用的掩膜版上的槽对应第二槽。
步骤S18b,去除所述第二蚀刻孔对应的所述隔离层和部分所述吸附层,以露出所述吸附层,从而在所述吸附结构层上形成第二槽,最后去除第一图案化膜和第二图案化膜,得到的所述第一槽和所述第二槽的槽深不同,所述第一槽和所述第二槽构成了所述多个槽结构。
在本实施例中,该第一槽和第二槽的槽径大致相同,大小可为200nm左右,第一槽和第二槽之间的距离是715nm,第一槽的深度可为45nm,第二槽的深度可为65nm,槽的底部是氮化钛用作吸附DNA纳米球,槽的间隔为氧化硅不特异性吸附DNA纳米球。
在本实施例中,生物载片的结构及尺寸可参考前述实施例一生物载片的制备方法中涉及到的结构及尺寸,故不再一一赘述。
实施例三
请参阅图14A至图14E,一并结合参阅图6,当需要形成一层吸附结构层20,且第一槽也伸入吸附层2时,生物载片100的制备方法包括以下步骤:
步骤S21,如图14A所示,本步骤与前述步骤S11a的制作方法相同,请参见前述步骤S11a,此处不做过多赘述。
步骤S22,如图14A所示,本步骤与前述步骤S12a的制作方法相同,请参见前述步
骤S12a,此处不做过多赘述。区别在于:由于后续第一槽会伸入部分吸附层2中,本步骤隔离层3的厚度无需沉积的过厚,本实施例中,隔离层3的厚度可以为25nm。
步骤S23,如图14A与图14B所示,本步骤与前述步骤S13a的制作方法相同,请参见前述步骤S13a,此处不做过多赘述。
步骤S24,如图14C所示,该步骤与前述步骤S14a的区别在于:本步骤需要将隔离层3去除,同时还需去除部分厚度的吸附层2,使第一槽4c延伸至吸附层2。
在本实施例中,蚀刻掉隔离层3后,还需将吸附层2也蚀刻掉20nm左右。
步骤S25,如图14D与图14E所示,本步骤与前述步骤S15a的制作方法相同,请参见前述步骤S15a,此处不做过多赘述。
步骤S26,如图14F所示,本步骤与前述步骤S16a的制作方法相同,请参见前述步骤S16a,此处不做过多赘述。可以理解的,本步骤形成的第二槽4d与第一槽4c的槽深不同,第一槽4c和第二槽4d构成了多个槽结构4。
在本实施例中,第二槽4d的形成过程中,将隔离层3蚀刻掉后,还需蚀刻掉至少40nm的吸附层2。
步骤S27,一并结合参阅图6所示,本步骤与前述步骤S17a的制作方法相同,请参见前述步骤S17a,此处不做过多赘述。
在本实施例中,该第一槽4c和第二槽4d的内径大致相同,大小可为200nm左右,第一槽4c和第二槽4d之间的距离是500nm,第一槽4a的深度可为45nm,第二槽4b的深度可为65nm,槽的底部是氮化钛用作吸附DNA纳米球,槽的间隔为氧化硅不特异性吸附DNA纳米球。
实施例四
请结合参阅图15A至图15F,当需要形成多层(例如两层)吸附结构层20时,生物载片200的制备方法包括以下步骤:
步骤S31,如图15A所示,于基底1的表面上形成吸附层2。
此步骤与前述步骤S11a制作方法相同,请参阅前述步骤S11a,此处不做过多赘述。
在本实施例中,沉积一层35nm的二氧化硅作为缓冲层5,并在缓冲层5上沉积一层60nm厚的氮化钛作为吸附层2。
步骤S32,如图15A所示,于所述吸附层2的表面上形成隔离层3,吸附层2与隔离层3构成第一层吸附结构层20。
此步骤与前述步骤S11a制作方法相同,请参阅前述步骤S11a,此处不做过多赘述。
在本实施例中,隔离层3可以是一层25nm的二氧化硅层。
步骤S33,如图15A所示,重复步骤S31和步骤S32,于隔离层3的表面上形成另一吸附层2’和另一层隔离层3’,吸附层2’和隔离层3’构成了另一层吸附隔离层20’。
在本实施例中,吸附层2’可以是20nm厚的氮化钛层,隔离层3’可以是45nm厚的二氧化硅层。
步骤S34,如图15A与图15B所示,于所述隔离层3’的表面上形成具有第一蚀刻孔61的第一图案化膜60。
此步骤与前述步骤S13a制作方法相同,请参阅前述步骤S13a,此处不做过多赘述。
步骤S35,如图15C所示,至少去除所述第一蚀刻孔61对应的所述隔离层3’,以露出所述吸附层2’,从而在所述吸附结构层20’上形成第一槽4e。
此步骤与前述步骤S14a制作方法相同,请参阅前述步骤S14a,此处不做过多赘述。
步骤S36,如图15D与图15E所示,于所述隔离层3’的表面上形成具有第二蚀刻孔31的第二图案化膜30,并使所述第二图案化膜30延伸至所述第一槽4e。
此步骤与前述步骤S15a制作方法相同,请参阅前述步骤S15a,此处不做过多赘述。
步骤S37,如图15F所示,去除所述第二蚀刻孔31对应的所述隔离层3,以露出所述吸附层2,从而在所述吸附结构层20上形成第二槽4f。
此步骤与前述步骤S16a制作方法接近,区别在于:由于增加了一层吸附结构层20’,两种槽结构有更充分的空间调整槽深,因此本步骤不需去除部分吸附层2,具体制作方法还请参阅前述步骤S16a,此处不做过多赘述。
可以理解的,本步骤也可以同时去除部分吸附层2,以制作出更深的第二槽4f。
步骤S38,一并结合参阅图7所示,去除所述第一图案化膜60和所述第二图案化膜30,其中,所述第一槽4e和所述第二槽4f的槽深不同,所述第一槽4e和所述第二槽4f构成了所述多个槽结构4。
在本实施例中,该第一槽4e和第二槽4f的槽径大致相同,大小可为200nm左右,第一槽4e和第二槽4f之间的距离是715nm,第一槽4e的深度可为45nm,第二槽4f的深度可为90nm,槽的底部是氮化钛用作吸附DNA纳米球,槽的间隔为氧化硅不特异性吸附DNA纳米球。
最后应说明的是,以上实施例仅用以说明本申请的技术方案而非限制,尽管参照较佳实施例对本申请进行了详细说明,本领域的普通技术人员应当理解,可以对本申请的技术方案进行修改或等同替换,而不脱离本申请技术方案的精神和范围。
Claims (19)
- 一种生物载片,其特征在于,包括载片本体,所述载片本体具有多个开口位于同侧的槽结构,所述多个槽结构包括第一槽和至少一个与所述第一槽距离最近且深度不同的第二槽。
- 如权利要求1所述的生物载片,其特征在于,所述第一槽和与所述第一槽距离最近的所述第二槽之间的深度差大于或等于20nm。
- 如权利要求1所述的生物载片,其特征在于,所述多个槽结构还包括与所述第一槽相邻的第三槽,所述第三槽与所述第一槽之间的距离大于所述第二槽与所述第一槽之间的距离,所述第三槽与所述第一槽的深度可以相同。
- 如权利要求1至3中任意一项所述的生物载片,其特征在于,所述槽结构的深度为20nm~150nm。
- 如权利要求1至3中任意一项所述的生物载片,其特征在于,所述载片本体包括基底以及位于所述基底上的至少一吸附结构层,所述槽结构形成于所述吸附结构层上,所述第一槽和所述第二槽伸入所述吸附结构层的深度不同。
- 如权利要求5所述的生物载片,其特征在于,所述吸附结构层包括:位于所述基底的表面上的吸附层和位于所述吸附层的表面上的隔离层,其中,所述吸附层用于吸附生物样本,所述槽结构至少贯穿所述隔离层,所述吸附层由所述槽结构露出。
- 如权利要求6所述的生物载片,其特征在于,所有或部分所述槽结构伸入所述吸附层。
- 如权利要求5所述的生物载片,其特征在于,所述生物载片包括多层层叠设置的所述吸附结构层,所述第一槽和所述第二槽伸入不同层的所述吸附结构层。
- 如权利要求1所述的生物载片,其特征在于,所述槽结构的内径为100nm~300nm;和/或,相邻两个所述槽结构的中心轴之间的距离为200nm-1000nm。
- 如权利要求6所述的生物载片,其特征在于,所述吸附层包含用于吸附生物样本的吸附材料,所述吸附材料包括金属氮化物、金属氧化物以及金属中的至少一种,及/或,所述隔离层的材质包括金属氮化物和金属氧化物中的至少一种。
- 如权利要求5所述的生物载片,其特征在于,所述基底与所述吸附结构层之间还设有缓冲层,所述缓冲层的材质包括氧化物和氮化物中的至少一种。
- 一种生物载片的制备方法,其特征在于,包括:于所述生物载片的载片本体上形成开口位于同侧的多个槽结构,其中,所述多个槽 结构包括第一槽和至少一个与所述第一槽距离最近且深度不同的第二槽。
- 如权利要求12所述的生物载片的制备方法,其特征在于,所述于所述生物载片的载片本体上形成开口位于同侧的多个槽结构的步骤包括:于所述载片本体上同时形成所述第一槽和所述第二槽;或于所述载片本体上分别形成所述第一槽和所述第二槽。
- 如权利要求13所述的生物载片的制备方法,其特征在于,所述于所述载片本体上同时形成所述第一槽和所述第二槽的步骤包括:于所述载片本体的表面上形成具有第一蚀刻孔和第二蚀刻孔的图案化膜;以及去除所述第一蚀刻孔对应的部分所述载片本体以在所述载片本体上形成所述第一槽,并去除所述第二蚀刻孔对应的部分所述载片本体以在所述载片本体上形成所述第二槽。
- 如权利要求13所述的生物载片的制备方法,其特征在于,所述于所述载片本体上分别形成所述第一槽和所述第二槽的步骤包括:于所述载片本体的表面上形成具有第一蚀刻孔的图案化膜;去除所述第一蚀刻孔对应的部分所述载片本体,以在所述载片本体上形成所述第一槽;于所述载片本体的表面上形成具有第二蚀刻孔的图案化膜;以及去除所述第二蚀刻孔对应的部分所述载片本体,以在所述载片本体上形成所述第二槽。
- 如权利要求12所述的生物载片的制备方法,其特征在于,所述制备方法还包括:于所述载片本体上形成与所述第一槽相邻的第三槽,所述第三槽与所述第一槽之间的距离大于所述第二槽与所述第一槽之间的距离,所述第三槽与所述第一槽的深度可以相同。
- 如权利要求12所述的生物载片的制备方法,其特征在于,所述于所述载片本体上形成开口位于同侧的多个槽结构的步骤包括:于基底的表面上形成至少一层吸附结构层;以及于所述吸附结构层上形成所述槽结构,并使所述第一槽和所述第二槽伸入所述吸附结构层的深度不同。
- 如权利要求17所述的生物载片的制备方法,其特征在于,所述于基底的表面上形成至少一层吸附结构层的步骤包括:于所述基底的表面上形成吸附层,所述吸附层用于吸附生物样本;以及于所述吸附层的表面上形成隔离层,在所述于所述吸附结构层上形成所述槽结构的步骤中,所述槽结构至少贯穿所述隔离层,以使所述吸附层由所述槽结构露出。
- 如权利要求12所述的生物载片的制备方法,其特征在于,所述第一槽和与所述第一槽距离最近的所述第二槽之间的深度差大于或等于20nm。
