Abstract
菌体培養槽(10)は、メタン(M)を生成するメタン生成菌を培養する培養液(S)を上昇させる構造を有する上昇槽(11)と、上昇槽(11)から越流した培養液(S)を下降させる構造を有する下降槽(12)と、上昇槽(11)および下降槽(12)の培養液(S)から放出されたメタン(M)を回収する回収部(13)と、を備える。
Description
本発明は、菌体培養槽および菌体培養方法に関する。
微生物(適宜、「菌体」)を利用したメタネーション(methanation)は、二酸化炭素と水素を培養液中に導入し、培養液内に存在するメタン生成菌(適宜、「メタン菌」)によってメタンを生産する手法である。
しかしながら、メタン生成菌の培養によるメタンの生産効率を向上させることは難しい。例えば、従来技術では、生成したメタンの微細気泡が培養液とともに排出され、菌体培養槽の外でメタンが培養液から脱離するので、メタンの回収効率が低下することがある。また、従来技術では、培養液の上昇速度が遅く、メタン生成菌の流動が少ないので、水素とメタン生成菌の接触効率が低下し、メタンの生成効率が低下することがある。
本発明は、上記に鑑みてなされたものであって、菌体が生成する気体の生産効率を向上させることを目的とする。
本発明の一実施形態に係る菌体培養槽は、気体を生成する菌体を培養する培養液を上昇させる構造を有する上昇槽と、前記上昇槽から越流した前記培養液を下降させる構造を有する下降槽と、前記上昇槽および前記下降槽の前記培養液から放出された前記気体を回収する回収部と、を備える。
本発明の一実施形態に係る菌体培養方法は、菌体培養槽によって実行される菌体培養方法であって、気体を生成する菌体を培養する培養液を上昇させる上昇工程と、前記上昇工程によって越流した前記培養液を下降させる下降工程と、前記上昇工程および前記下降工程の前記培養液から放出された前記気体を回収する回収工程と、を含む。
本発明によれば、菌体が生成する気体の生産効率を向上させることができるという効果がある。
以下に、本発明の一実施形態に係る菌体培養槽および菌体培養方法を、図面を参照して詳細に説明する。なお、本発明は、以下に説明する実施形態により限定されるものではない。
以下に、実施形態に係る菌体培養槽10の概要、菌体培養槽10の構成および機能について順に説明し、最後に実施形態の効果について説明する。
〔1.菌体培養槽10の概要〕
図1を用いて、実施形態に係る菌体培養槽10の概要について説明する。図1は、実施形態に係る菌体培養槽10の概要図の一例を示す図である。以下では、メタン生成菌のメタン生成反応、菌体培養槽10の基本構成例、および菌体培養槽10の効果について説明する。

(1-1.メタン生成菌のメタン生成反応)
メタン生成菌は、下記(1)式で表わされる反応によって、メタンMを生成する。なお、下記(1)式において、「CO2」が二酸化炭素C、「H2」が水素H、「CH4」がメタンM、「H2O」が水である。また、常温常圧の条件下において、二酸化炭素C、水素H、およびメタンMが気体であり、水が液体である。
メタン生成菌は、下記(1)式で表わされる反応によって、メタンMを生成する。なお、下記(1)式において、「CO2」が二酸化炭素C、「H2」が水素H、「CH4」がメタンM、「H2O」が水である。また、常温常圧の条件下において、二酸化炭素C、水素H、およびメタンMが気体であり、水が液体である。

また、メタン生成菌は、嫌気性菌であるので、酸素が存在する環境下では上記(1)式のメタン生成反応によってメタンMを生成することができない。
(1-2.菌体培養槽10の基本構成例)
図1に示すように、菌体培養槽10は、上昇槽11、下降槽12、回収部13、上面14、および底面15で構成される。また、菌体培養槽10は、培養液Sに含まれるメタン生成菌を培養することによって、二酸化炭素Cおよび水素Hを原料としてメタンMを生産する培養槽である。
図1に示すように、菌体培養槽10は、上昇槽11、下降槽12、回収部13、上面14、および底面15で構成される。また、菌体培養槽10は、培養液Sに含まれるメタン生成菌を培養することによって、二酸化炭素Cおよび水素Hを原料としてメタンMを生産する培養槽である。
(1-2-1.上昇槽11)
上昇槽11は、外部壁11a、供給部11b(11b-1、11b-2)、および発生部11c(11c-1、11c-2)で構成される。また、上昇槽11は、円筒形の外部壁11aおよび円筒形の内部壁12aによって構成される二重円筒形の外径側の構造である。
上昇槽11は、外部壁11a、供給部11b(11b-1、11b-2)、および発生部11c(11c-1、11c-2)で構成される。また、上昇槽11は、円筒形の外部壁11aおよび円筒形の内部壁12aによって構成される二重円筒形の外径側の構造である。
上昇槽11は、供給部11bによって、メタン生成菌が含まれ、二酸化炭素Cが溶解した培養液Sを上昇させる。例えば、上昇槽11は、供給部11bによって一定量の培養液Sを連続的に供給することによって、上昇槽11内部の培養液Sの液面の高さを上昇させる。また、上昇槽11は、発生部11cによって、水素Hを培養液Sに微細気泡として発生させる。このとき、メタン生成菌は、培養液Sにおいて、上記(1)式で表わされる代謝反応によって、二酸化炭素Cおよび水素HからメタンMを生成する。
ここで、上昇槽11は、供給部11bが供給する培養液Sの単位時間当たりの供給量を調整することによって、培養液Sの上昇速度を調整することができる。このとき、上昇槽11は、供給部11bによって連続的に供給する培養液Sを増加させることによって、培養液Sの上昇速度を増加させることができる。一方、上昇槽11は、供給部11bによって連続的に供給する培養液Sを減少させることによって、培養液Sの上昇速度を減少させることができる。例えば、上昇槽11は、培養液Sの上昇速度をメタン生成菌の沈降速度以上にすることによって、メタン生成菌を培養液Sとともに上昇させることができる。ここで、沈降速度とは、培養液Sに流れがない場合のメタン生成菌が沈降していく速度である。一方、上昇槽11は、培養液Sの上昇速度をメタン生成菌の沈降速度未満にすることによって、メタン生成菌を沈降させ、培養液Sと分離させることができる。
なお、図1では、供給部11bは、2つで構成されているが、培養液Sを上昇槽11の内部に均一に供給できるのであれば、1つであってもよいし、3つ以上であってもよい。また、図1では、発生部11cは、2つで構成されているが、水素Hを上昇槽11の内部に均一に発生できるのであれば、1つであってもよいし、3つ以上であってもよい。
(1-2-2.下降槽12)

ここで、下降槽12は、後述する段状構造である下降段P(不図示)が内部壁12aに設置されている場合には、培養液Sの内部壁12aへの接触時間や接触回数が増加することによって、培養液Sに溶解したメタンMの放出を促進することができる。また、下降槽12は、後述する捕集膜F(不図示)が内部壁12aに設置されている場合には、下降する培養液Sからメタン生成菌を捕集することができる。このとき、捕集膜Fの上側からメタン生成菌を引き抜いて上昇槽に戻すことによって、メタン生成菌の外部への流出を防ぐことができる。
(1-2-3.回収部13)
回収部13は、上面14の中央に設置され、上昇槽11および下降槽12の培養液Sから放出されるメタンMを回収する。ここで、回収部13は、回収したメタンMを、配管を通じて気密性を有する保管タンクに送出する。

以下では、メタネーションの概要、参考技術の概要および問題点について説明した上で、実施形態に係る菌体培養槽10の概要および効果について説明する。

(1-3-1.メタネーションの概要)
鉄鋼、化学、自動車、電気等の製造工程や、石炭や天然ガスを用いる発電プラント等から排出される二酸化炭素Cは地球温暖化防止の観点から排出量を削減する必要に迫られている。その背景で、温室効果ガスとなる二酸化炭素Cの排出量の削減のために、二酸化炭素Cを資源として有効活用する研究開発が盛んに行われており、藻類を用いた光合成によるバイオ燃料の合成、微生物を用いた化学原料の合成、二酸化炭素Cおよび水素Hを原料としたメタンMの合成(メタネーション)等が行われている。工場の排ガス中の二酸化炭素Cを活用してメタンMを合成することにより、二酸化炭素Cを資源として循環させることができる。メタンMは、都市ガスの主成分であり、体積エネルギー密度が水素Hやアンモニアと比較して高く、また、既存のパイプラインやガス管を通して供給できる。燃料として主流の天然ガスでは、燃焼した際に二酸化炭素Cが発生するので、カーボンニュートラルとなる合成ガスが望まれている。

メタネーションの方式として、触媒方式、微生物方式等がある。微生物を用いたメタネーションは、二酸化炭素Cおよび水素Hを培養液Sに導入し、培養液S内に存在するメタン生成菌によりメタンMを合成する手法である。微生物を用いた方式では、100℃以下の比較的緩やかな環境で合成が可能であり、複雑な反応制御が不要となるので、保守メンテナンスが容易であり、装置の大型化が可能になる。上記のように地球温暖化防止を背景に二酸化炭素Cを資源として有効活用するために、微生物を用いたメタネーションや化学原料の合成、バイオ燃料の合成等に活用できる技術開発が急務になっている。
(1-3-2.参考技術の概要)
参考技術では、以下のように一槽構造の菌体培養槽10Pを用いてメタンMを生産する。第1に、参考技術では、メタン生成菌が存在する菌体培養槽10Pに二酸化炭素Cを溶解した培養液Sを供給する。第2に、参考技術では、培養液Sにおいて、バブリング方式で水素Hの微細気泡を発生させることでメタン生成菌にメタンMを生成させる。第3に、参考技術では、生成され、気体として放出されたメタンMを回収する。
参考技術では、以下のように一槽構造の菌体培養槽10Pを用いてメタンMを生産する。第1に、参考技術では、メタン生成菌が存在する菌体培養槽10Pに二酸化炭素Cを溶解した培養液Sを供給する。第2に、参考技術では、培養液Sにおいて、バブリング方式で水素Hの微細気泡を発生させることでメタン生成菌にメタンMを生成させる。第3に、参考技術では、生成され、気体として放出されたメタンMを回収する。
(1-3-3.参考技術の問題点)

菌体培養槽10は、上記のように二槽構造であって、上昇槽11、下降槽12、回収部13等で構成される。第1に、上昇槽11は、メタン生成菌が含まれ、二酸化炭素Cが溶解した培養液Sを上昇させる。このとき、上昇槽11は、供給部11bによって一定量の培養液Sを連続的に供給することによって、上昇槽11内部の培養液Sの液面の高さを上昇させる。また、上昇槽11は、供給する培養液Sの単位時間当たりの供給量を調整することによって、培養液Sの上昇速度を調整することができる。このとき、上昇槽11は、供給部11bによって連続的に供給する培養液Sを増減させることによって、培養液Sの上昇速度を増減させることができる。第2に、上昇槽11は、水素Hを培養液Sに微細気泡として発生させ、メタン生成菌にメタンMを生成させる。第3に、下降槽12は、上昇槽11から越流した培養液Sを下降させる。このとき、下降槽12は、上昇槽11の供給部11bによって一定量の培養液Sを連続的に供給することによって液面の高さが上昇した培養液Sであって、液面の高さが内部壁12aを超過した培養液S、すなわち越流した培養液Sを下降槽12内部に下降させる。第4に、回収部13は、上昇槽11および下降槽12の培養液Sから放出されるメタンMを回収する。

(1-3-5.効果)
第1に、菌体培養槽10は、上昇槽11の液面より下降槽12の液面を低くすることによって培養液Sの越流、接触、落下等により気液分離を促進し、メタンMを気中へ放出させることでメタンMの回収効率を向上させる。第2に、菌体培養槽10は、培養液Sの上昇速度を増加させ、メタン生成菌の流動を多くするので、水素Hとメタン生成菌との接触効率を向上させることでメタンMの生成効率を向上させる。第3に、菌体培養槽10は、メタン生成菌の沈降性を活用して菌体培養槽10からのメタン生成菌の流出を防ぎ、菌体培養槽10の菌体密度を維持することができるので、メタンMの生成効率を向上させる。

〔2.菌体培養槽10の構成および機能〕
図2を用いて、図1に示した菌体培養槽10の構成および機能について説明する。図2は、実施形態に係る菌体培養槽10の構成例を示すブロック図である。菌体培養槽10は、上昇槽11、下降槽12、回収部13、上面14、および底面15で構成される。以下では、菌体培養槽10の構成例および機能例について、菌体培養槽10の構造例および菌体の具体例について説明した上で、上昇槽11、下降槽12、回収部13、上面14、底面15の順に詳細に説明する。

(2-2.菌体の具体例)
菌体は、例えば、気体としてメタンMを生成するメタン生成菌であるが、気体や菌体の種類については特に限定されない。また、メタン生成菌は、例えば、メタノバクテリウム属(Methanobacterium)、メタノコックス属(Methanococcus)等に分類される嫌気条件でメタンMを合成する古細菌であるが、メタン生成菌の種類については特に限定されない。また、菌体は、気体の基質として、例えば、二酸化炭素Cおよび水素Hを用いるが、気体の基質については特に限定されない。また、菌体が生成する気体はメタンMに限定されず、揮発性の炭化水素(例:エタン、エチレン、アセチレン)等であってもよい。

培養液Sを供給する速度について説明すると、〔3.菌体培養槽10の具体例〕(3-6.菌体培養槽10の具体例5)で後述するように、供給部11bは、菌体の沈降速度以上の上昇速度で培養液Sを供給する。すなわち、供給部11bは、メタンMの生成効率を向上させたい場合には、メタン生成菌の流動を多くすることによって、水素Hとメタン生成菌との接触効率を向上させ、培養液Sの上昇速度をメタン生成菌の沈降速度以上になるように単位時間当たりの培養液Sの供給量を増加させる調整をする。また、〔3.菌体培養槽10の具体例〕(3-7.菌体培養槽10の具体例6)で後述するように、供給部11bは、菌体の沈降速度未満の上昇速度で培養液Sを供給する。すなわち、供給部11bは、メタン生成菌の外部への流出を防ぎたい場合や死滅したメタン生成菌を除去したい場合には、培養液Sの上昇速度をメタン生成菌の沈降速度未満になるように単位時間当たりの培養液Sの供給量を減少させる調整をする。このとき、供給部11bは、培養液Sの上昇速度をメタン生成菌の沈降速度未満であって適切な上昇速度を保つように調整することによって、培養液Sに溶解している二酸化炭素Cの槽内滞留時間を増加させ、二酸化炭素Cとメタン生成菌との接触機会を増加させ、メタン生成菌の下降槽12への流出を減少させ、菌密度を保つことができるので、メタンMの生成効率を向上させることもできる。
菌体が基質として用いる二酸化炭素Cの由来について説明すると、供給部11bは、炭素を含有する燃料の燃焼によって生成された二酸化炭素Cを含む培養液Sを供給する。すなわち、供給部11bは、例えば、石油や石炭等の燃焼によって生成された二酸化炭素CをメタンMに変換するために利用することができる。また、供給部11bは、菌体が生成したメタンMの燃焼によって生成された二酸化炭素Cを含む培養液Sを供給する。すなわち、供給部11bは、メタン生成菌から生産したメタンMをボイラー等の装置に供給し、当該装置内でメタンMの燃焼によって生成された二酸化炭素Cを回収し、再びメタンMに変換するために利用することができる。
(2-3-3.発生部11c)
発生部11cは、例えば、微細孔を有し、菌体培養槽10の内部への送出機能を有する送出口および配管で実現される。また、発生部11cは、例えば、外部壁11aの下部に設置されるが、上昇槽11の領域に対応する底面15に設置されてもよく、設置位置については特に限定されない。

基質として用いる水素Hの由来について説明すると、発生部11cは、再生可能エネルギーを用いた電気分解によって生成された水素Hを気泡として発生する。例えば、発生部11cは、太陽光によって発電した電気エネルギーを用いた電気分解によって生成された水素HをメタンMに変換するために利用することができる。また、発生部11cは、製造工程で副次的に生成された水素Hを気泡として発生する。例えば、発生部11cは、苛性ソーダの製造工程で副次的に生成された水素HをメタンMに変換するために利用することができる。また、発生部11cは、都市ガス改質によって生成された水素HをメタンMに変換するために利用することもできる。
