Abstract

本発明は、自殺遺伝子を羊膜由来間葉系幹細胞に導入し、遺伝子修飾された羊膜由来間葉系幹細胞の製造方法を提供することを課題とする。 羊膜由来間葉系幹細胞を含む細胞集団に、トランスポゾン法等のプラスミドベクターを用いる方法により自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入する工程を含む、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造方法。

Description

遺伝子修飾された間葉系幹細胞を含む細胞集団の製造方法
 本発明は、遺伝子修飾された間葉系幹細胞を含む細胞集団の製造方法に関する。
 癌は、世界的に患者数及び死者数の多い疾患であり、年間数百万人が、癌が原因で死亡している。癌は発生する臓器や発生源となる細胞によって様々な種類に分けられる。その中には未だに既存療法では治療効果が乏しいものもあり、新規治療法の研究が盛んに行なわれている。
 悪性脳腫瘍は、生命予後が特に悪い癌の一つである。悪性脳腫瘍に対する既存の治療法としては、外科的手術による腫瘍摘出とその後の放射線治療、及び/又は薬物治療が標準とされる。しかしながら、脳腫瘍の種類や発生箇所によっては外科的手術で腫瘍を摘出しきれない場合が多く、放射線治療や薬物治療を施しても癌細胞が残存し再発することが多い。癌細胞に対する標的性をさらに高めた新たな悪性脳腫瘍の治療法が求められている。
 間葉系幹細胞は、骨髄、脂肪組織、歯髄、羊膜等に存在することが報告されている体性幹細胞である。間葉系幹細胞は免疫抑制作用を有しており、他家移植も可能であることから、細胞治療における有望な細胞ソースとして注目されており、実際に、急性移植片対宿主病(GVHD)や脊髄損傷等に対する実用化が進んでいる。さらに、間葉系幹細胞は生体内で癌病変に対して特異的に遊走し集積する性質を有することが知られており、標的性を高めたがん治療法として間葉系幹細胞を利用する研究が進められている。
 間葉系幹細胞を利用した癌治療法としては、単純ヘルペスウイルスチミジンキナーゼ(HSV-TK)等の自殺遺伝子を遺伝子導入することで抗癌機能を付与した間葉系幹細胞を用いる方法がある。この際の遺伝子導入方法には、遺伝子導入効率や遺伝子発現の安定性の観点から、ウイルスベクターを用いる方法が一般的である。例えば、非特許文献1には、脂肪組織由来間葉系幹細胞にHSV-TKをレンチウイルスベクターにより遺伝子導入し、目的の細胞を製造する方法が記載されている。一方で、人工多能性幹細胞や歯髄幹細胞に自殺遺伝子をウイルスベクターにより遺伝子導入すると、細胞毒性を示し自殺遺伝子導入細胞の増殖能が低下する場合があることが報告されている(非特許文献2、非特許文献3)。
 羊膜由来間葉系幹細胞は、帝王切開分娩後の医療廃棄物として扱われる羊膜を原料とするために細胞の採取に新たなドナーの侵襲を伴わない点、胎児由来組織である羊膜を原料とするために細胞特性のドナー間差が小さい点、1ドナーの羊膜から多数の間葉系幹細胞を取得できる点を特長とする。これらの特長から、羊膜由来間葉系幹細胞は、様々な組織由来間葉系幹細胞の中でも特に細胞医療材料として優れており、癌治療分野において利用することで、低コストかつ品質の安定した細胞医療の提供が期待できる。

 本発明者らは、最初に、遺伝子導入効率が高いことで知られている手法としてレンチウイルスベクターを用いて羊膜由来間葉系幹細胞に自殺遺伝子の遺伝子導入を行った。その結果、細胞毒性により遺伝子導入細胞の増殖能が著しく低下し、目的の細胞を製造できないことがわかった。そこで、本発明の課題は、自殺遺伝子を導入した羊膜由来間葉系幹細胞の増殖能を大きく低下させることなく、遺伝子修飾された羊膜由来間葉系幹細胞を製造する方法を提供することである。
 本発明者らは、前述の課題解決のために鋭意検討を行なった。その結果、驚くべきことに、自殺遺伝子を遺伝子導入する際にトランスポゾン法等のプラスミドベクターを用いた場合、遺伝子導入細胞は、細胞毒性による増殖能の著しい低下を起こさなかった。それにより、自殺遺伝子を安定発現する羊膜由来間葉系幹細胞の製造が可能となることを見出し、本発明の完成に至った。
 すなわち、本発明は、下記[1]~[10]に関する。
[1]羊膜由来間葉系幹細胞に、プラスミドベクターを用いて自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入する工程を含む、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造方法。
[2]前記プラスミドベクターが、エレクトロポレーション法により細胞内に導入される、[1]に記載の製造方法。
[3]前記遺伝子導入工程で、前記自殺遺伝子又は自殺遺伝子の発現カセットを染色体に組み込む、[1]又は[2]に記載の製造方法。
[4]前記遺伝子導入工程において、前記自殺遺伝子又は自殺遺伝子の発現カセットをトランスポザーゼが認識する反復配列に隣接して配置したコンストラクトを含むプラスミドベクターとトランスポザーゼ発現カセットを含むプラスミドベクターの2種類のプラスミドベクターを用いる、[1]~[3]のいずれかに記載の製造方法。

 本発明によれば、細胞増殖能を低下させることなく、自殺遺伝子を発現する羊膜由来間葉系幹細胞を含む細胞集団を製造することができる。

 本発明の一実施形態について以下に説明するが、本発明はこれらに限定されるものではない。
 [1]用語の説明
 (羊膜由来間葉系幹細胞)
 本明細書における「羊膜由来間葉系幹細胞」とは、羊膜から採取することができる、下記の定義を満たす幹細胞のことをいう。
 i)標準培地での培養条件で、プラスチックに接着性を示す。
 ii)表面抗原のCD73、CD90が陽性であり、CD45、CD326が陰性である。
 ここでいう「羊膜」は、卵膜の最内層にある血管に乏しい透明薄膜のことを指し、上皮羊膜上皮層(上皮組織)、基底膜及び緻密層(間質組織)を含む。本明細書における「間葉系幹細胞(Mesenchymal stem cells:MSC)」には、「間葉系間質細胞(Mesenchymal stromal cells)」も含まれる。本明細書における羊膜の由来は特に限定されず、任意の生物に由来することができる。前記生物としては、例えば、ヒト、サル、ウシ、ウマ、イヌ、ネコ、ブタ、ヒツジ、マウス、ラット、及びウサギ等の哺乳類、インコ、オウム、及びニワトリ等の鳥類が挙げられる。本明細書における羊膜は、好ましくは、ヒト、サル、ウシ、ウマ、イヌ、ネコ、ブタ、ヒツジ、マウス、ラット、又はウサギ由来であり、より好ましくはヒト、サル、イヌ、又はネコ由来であり、さらに好ましくはヒト由来である。
 (間葉系幹細胞を含む細胞集団)
 本明細書における「間葉系幹細胞を含む細胞集団」とは、間葉系幹細胞を1細胞以上含む、1細胞以上で構成される細胞の集合体のことをいう。その形態は特に限定されず、例えば、組織、組織片、細胞ペレット、細胞凝集塊、細胞シート、細胞浮遊液、細胞懸濁液、及びこれらの凍結物等が挙げられる。
 (自殺遺伝子)
 本明細書における「自殺遺伝子」とは、プロドラッグを細胞毒性物質に変換する働きを有するタンパク質をコードする遺伝子であり、当該遺伝子を発現した細胞において、プロドラッグ存在下で細胞死を誘導する遺伝子のことをいう。自殺遺伝子は、例えば、単純ヘルペスウイルスチミジンキナーゼ(HSV-TK)遺伝子、シトシンデアミナーゼ(CD)遺伝子、水痘帯状疱疹ウイルスチミジンキナーゼ遺伝子、ニトロレダクターゼ遺伝子、プリンヌクレオシドホスホリラーゼ遺伝子、βガラクトシダーゼ遺伝子、シトクロムP450遺伝子、大腸菌GPT遺伝子、リナマラーゼ遺伝子、ホースラディッシュペルオキシダーゼ遺伝子、カルボキシペプチダーゼA遺伝子、及びカルボキシペプチダーゼG2遺伝子等が挙げられる。本明細書における自殺遺伝子は、好ましくは、HSV-TK遺伝子又はCD遺伝子であり、より好ましくは、HSV-TKである。
 本明細書における「プロドラッグ」は、使用する自殺遺伝子により細胞毒性を有する抗癌物質に転換され得る前駆薬物であれば特に限定されない。例えば、ガンシクロビル、5-フルオロシトシン(5-FC)、6-メトキシプリンアラビノシド(6-methoxypurine arabinoside)、CB1954、6-メチルプリンデオキシリボシド(6-methylpurine deoxyriboside)、Daun02、シクロホスファミド、イホスファミド、及びZD-2767P等が挙げられる。プロドラッグは自殺遺伝子との組み合わせが重要であり、例えばガンシクロビルはHSV-TK遺伝子との組み合わせで、5-FCはCD遺伝子との組み合わせで、6-メトキシプリンアラビノシドは水痘帯状疱疹ウイルスチミジンキナーゼ遺伝子との組み合わせで、CB1954はニトロレダクターゼ遺伝子との組み合わせで、そして6-メチルプリンデオキシリボシドはプリンヌクレオシドホスホリラーゼ遺伝子との組み合わせで、それぞれ抗癌物質に転換され得る。本明細書におけるプロドラッグは、好ましくは、ガンシクロビル又は5-FCであり、より好ましくは、ガンシクロビルである。
 (発現カセット)
 本明細書における「発現カセット」とは、1種類以上の遺伝子を含み、前記遺伝子の発現を制御する1以上の核酸領域を任意で含む、前記遺伝子の発現単位を包含する核酸領域の集合体のことをいう。遺伝子の発現を制御する核酸領域としては、特に限定されない。例えば、プロモーター、エンハンサー、ターミネーター、ポリAシグナル、及びリンカー等が挙げられる。
 (遺伝子導入)
 本明細書における「遺伝子導入」とは、細胞等の対象物に、人為的に核酸を導入することをいう。遺伝子導入の方法としては、一般的にウイルスベクターを用いる方法又は非ウイルスベクターを用いる方法が挙げられる。ウイルスベクターとしては、例えば、アデノウイルスベクター、アデノ随伴ウイルスベクター、レトロウイルスベクター、レンチウイルスベクター、バキュロウイルスベクター、ワクシニアウイルスベクター、及び単純ヘルペスウイルスベクター等が挙げられる。非ウイルスベクターとしては、例えばプラスミドベクターが挙げられる。プラスミドベクターを用いた遺伝子導入方法としては、例えば、トランスポゾン法、陽イオン性脂質を用いた方法、リン酸カルシウム共沈殿法、ジエチルアミノエチル(DEAE)ーデキストラン法、陽イオン性ポリマーを用いた方法、エレクトロポレーション法、マイクロインジェクションクション法、遺伝子銃を用いた方法、レーザーを用いた方法、及び磁性ナノ粒子を用いた方法等が挙げられる。
 本発明における「エレクトロポレーション法」とは、細胞に電気パルスを当てることで一時的に細胞膜に孔を形成し、孔を介してプラスミドベクター等の核酸を細胞内に送達する遺伝子導入法をいう。本明細書における「エレクトロポレーション法」には、「電気穿孔法」も含まれる。
 本明細書における「トランスポゾン法」は、トランスポザーゼ(Transposase)とトランスポザーゼが認識する特定の反復配列の組み合わせによる遺伝子転移の機構を利用した遺伝子導入方法をいう。前記トランスポザーゼは、特に限定されない。例えば、Sleeping Beauty、piggyBac、Tol2、Frog Prince、及びHimarI等が挙げられる。本明細書における「トランスポゾン」には、「DNAトランスポゾン」及び「レトロトランスポゾン」も含まれる。
 (遺伝子修飾された)
 本明細書における「遺伝子修飾された」細胞とは、人為的に導入された核酸を有している細胞をいう。
 (羊膜由来間葉系幹細胞を含む細胞製剤)
 本明細書における「羊膜由来間葉系幹細胞を含む細胞製剤」とは、遺伝子修飾された羊膜由来間葉系細胞を1細胞以上含む、生体外で調製される組成物のことをいう。その形態は特に限定されず、例えば、細胞浮遊液、細胞懸濁液、細胞ペレット、細胞凝集塊、シート、シール剤、及びこれらの凍結物等が挙げられる。
 本明細書における「癌」は、特に限定されない。例えば、扁平上皮癌、肺癌、腹膜癌、中皮腫、結腸癌、胆管腫瘍、鼻咽頭癌、喉頭癌、気管支癌、口腔癌、骨肉腫、軟部肉腫、胆嚢癌、腎臓癌、白血病、膀胱癌、黒色腫、脳癌、神経膠腫、脳腫瘍、皮膚癌、膵臓癌、乳癌、肝癌、骨髄癌、食道癌、大腸癌、胃癌、子宮頸癌、子宮内膜癌、前立腺癌、卵巣癌、頭頸部癌、直腸癌、及び精巣癌等が含まれる。本明細書における「癌」は、「がん」、「腫瘍」、及び「悪性腫瘍」を含む。
 [2]遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造方法
 本発明の一以上の実施形態では、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造方法を提供する。本発明における遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造方法は、羊膜由来間葉系幹細胞に、自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入する工程を含む製造方法である。
 本発明における遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造の原料となる羊膜由来間葉系幹細胞を含む細胞集団は、羊膜由来間葉系幹細胞を1細胞以上含む細胞集団であれば特に限定されず、他の細胞を含む集団であってもよい。特に限定されない。前記羊膜由来間葉系幹細胞を含む細胞集団における羊膜由来間葉系幹細胞の比率は、例えば、0.1%以上、1%以上、10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、75%以上、80%以上、85%以上、87%以上、88%以上、89%以上、90%以上、91%以上、92%以上、93%以上、94%以上、及び95%以上である。
 本発明における羊膜由来間葉系幹細胞を含む細胞集団は、前培養操作の有無、及び凍結操作の有無を問わない。具体的には、個体から採取した組織又は細胞、又はこれらの凍結物であってもよいし、個体から採取した組織又は細胞の培養物、又はこれらの凍結物であってもよい。
 本発明は、羊膜を酵素処理することにより、原料となる羊膜由来間葉系幹細胞を含む細胞集団を取得する、細胞集団取得工程を含むものでもよい。前記細胞集団取得工程は、羊膜を帝王切開により取得する工程をさらに含む工程でもよい。
 本発明の一以上の実施形態によれば、羊膜由来間葉系幹細胞を含む細胞集団の製造方法は、好ましくは、羊膜を含む試料を酵素処理して得た細胞集団である。前記酵素処理は、好ましくは、上皮細胞層を分解することなく羊膜の細胞外基質層に含まれる間葉系幹細胞を遊離することができる酵素による処理である。前記活性を有する酵素としては、例えば、コラゲナーゼ及び金属プロテイナーゼを挙げることができる。金属プロテイナーゼとしては、非極性アミノ酸のN末端側を切断する金属プロテイナーゼであるサーモリシン及びディスパーゼを挙げることができるが、特に限定されない。また、前記活性を有する酵素を2種類以上組み合わせて使用してもよい。
 本発明は、羊膜由来間葉系幹細胞を含む細胞集団を培養する工程をさらに含んでもよい。この時の羊膜由来間葉系幹細胞を含む細胞集団の培養方法は、後述する遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の培養方法と同様の方法を用いることができる。
 本発明は、羊膜由来間葉系幹細胞を含む細胞集団に含まれる羊膜由来間葉系幹細胞に、自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入する工程を含む。自殺遺伝子導入細胞の製造効率の観点から、自殺遺伝子又は自殺遺伝子の発現カセットは、遺伝子修飾された細胞の染色体に組み込まれることが好ましい。これにより、染色体外ベクターを有する遺伝子修飾された細胞と比べて、遺伝子修飾された細胞をより安定的に産生することが可能となる。
 導入遺伝子を標的細胞の染色体に組み込む遺伝子導入方法としては、ウイルスベクターを用いる方法が一般的に用いられる。ところが、羊膜由来間葉系幹細胞は、ウイルスベクターを用いる方法により自殺遺伝子又は自殺遺伝子の発現カセットの遺伝子導入を施すと、細胞増殖能が低下し、遺伝子修飾された細胞の製造効率が著しく低下することが本発明者により初めて確認された。そのため、本発明では、プラスミドベクターを用いて遺伝子導入を施すことを特徴とする。
 本発明におけるプラスミドベクターは、少なくとも1種類のプラスミドベクターを用いればよく、2種類以上のプラスミドベクターを用いてもよい。特に限定されない。例えば、自殺遺伝子又は自殺遺伝子の発現カセットを構造に持つプラスミドベクターと、自殺遺伝子の標的細胞ゲノムへの組み込みや自殺遺伝子の発現を補助又は制御するプラスミドベクターを組み合わせて用いてもよい。
 本発明におけるプラスミドベクターの細胞内への導入方法は、例えば、陽イオン性脂質を用いた方法、リン酸カルシウム共沈殿法、ジエチルアミノエチル(DEAE)―デキストラン法、陽イオン性ポリマーを用いた方法、エレクトロポレーション法、マイクロインジェクションクション法、遺伝子銃を用いた方法、レーザーを用いた方法、及び磁性ナノ粒子を用いた方法等が挙げられる。本発明におけるプラスミドベクターの細胞内への導入方法は、導入効率及び操作の簡便性の観点から、好ましくは、陽イオン性脂質を用いた方法、リン酸カルシウム共沈殿法、ジエチルアミノエチル(DEAE)―デキストラン法、陽イオン性ポリマーを用いた方法、又はエレクトロポレーション法であり、さらに好ましくは陽イオン性脂質を用いた方法、陽イオン性ポリマーを用いた方法、又はエレクトロポレーション法であり、最も好ましくはエレクトロポレーション法である。
 本発明におけるプラスミドベクターを用いた遺伝子導入方法は、好ましくはトランスポゾン法である。トランスポゾン法は、導入遺伝子又は導入遺伝子の発現カセットをトランスポザーゼが認識する反復配列に隣接して配置したコンストラクトを標的細胞に導入し、さらに標的細胞内でトランスポザーゼを発現させることで、導入遺伝子が標的細胞の染色体に安定的に組み込まれ得るため、導入遺伝子のゲノム組み込み効率が比較的高い遺伝子導入方法である。羊膜由来間葉系幹細胞に自殺遺伝子又は自殺遺伝子の発現カセットの遺伝子導入を施す際にトランスポゾン法を用いると、驚くべきことに、ウイルスベクターを用いた場合に起こる細胞増殖能の低下が起こらず、効率的に遺伝子修飾された細胞を製造することが可能となる。したがって、本発明は、自殺遺伝子又は自殺遺伝子の発現カセットをトランスポザーゼが認識する反復配列に隣接して配置したコンストラクトを構造に持つプラスミドベクターと、トランスポザーゼ発現カセットを構造に持つプラスミドベクターの2種類のプラスミドベクターを用いることが好ましい。又は、自殺遺伝子又は自殺遺伝子の発現カセットをトランスポザーゼが認識する反復配列に隣接して配置したコンストラクトとトランスポザーゼ発現カセットの双方を構造に持つプラスミドベクターを用いることもできる。
 本発明における導入遺伝子は、1種類以上の自殺遺伝子を含んでいればよく、2種類以上の自殺遺伝子を含んでもよい。また、必要に応じて1種類以上の他の遺伝子を含んでもよい。
 前記他の遺伝子又はその発現カセットの遺伝子導入は、自殺遺伝子又は自殺遺伝子の発現カセットの遺伝子導入の前に施されてもよく、自殺遺伝子又は自殺遺伝子の発現カセットの遺伝子導入の後に施されてもよく、又は自殺遺伝子又は自殺遺伝子の発現カセットの遺伝子導入と同時に施されてもよい。また、前記他の遺伝子は、自殺遺伝子の発現カセットに構成要素として含まれていてもよい。この時、他の遺伝子の遺伝子導入は自殺遺伝子又は自殺遺伝子の発現カセットの遺伝子導入と同時に施され得る。
 前記他の遺伝子としては、自殺遺伝子以外の遺伝子であれば特に限定されない。例えば、抗生物質耐性遺伝子、レポーター遺伝子、タグペプチド、未分化性に関わる遺伝子、及び細胞分化に関わる遺伝子等が挙げられる。前記抗生物質耐性遺伝子としては、例えば、ピューロマイシン、ネオマイシン、ハイグロマイシン、及びブラストサイジン等に対する耐性を付与する遺伝子が挙げられる。前記レポーター遺伝子としては、例えば、GFP、EGFP、RFP、Venus、DsRed、β-ガラクトシダーゼ、β-グルクロニターゼ、及びルシフェラーゼ等が挙げられる。前記タグペプチドとしては、タンパク質を標識化することのできる十数アミノ酸~数十アミノ酸からなる短ペプチドであれば特に限定されず、例えば、FLAG、HA、His、及びmyc等が挙げられる。前記未分化性に関わる遺伝子としては、例えば、Nanog、Oct4、Sox2、及びKlf4等が挙げられる。前記細胞分化に関わる遺伝子としては、例えば、Runx2、CBF-1α、Sox9、PPARγ、GATA4、GATA6、及びMyoD等が挙げられる。
 前記自殺遺伝子以外の他の遺伝子又はその発現カセットの遺伝子導入方法は、染色体への組み込みの有無を問わず、ウイルスベクターを用いる方法でもよく、非ウイルスベクターを用いる方法でもよい。前記ウイルスベクターとしては、例えば、アデノウイルスベクター、アデノ随伴ウイルスベクター、レトロウイルスベクター、レンチウイルスベクター、バキュロウイルスベクター、ワクシニアウイルスベクター、及び単純ヘルペスウイルスベクター等が挙げられるが、特に限定されない。前記非ウイルスベクターを用いる方法としては、例えば、トランスポゾン法、陽イオン性脂質を用いた方法、リン酸カルシウム共沈殿法、ジエチルアミノエチル(DEAE)ーデキストラン法、陽イオン性ポリマーを用いた方法、エレクトロポレーション法、マイクロインジェクションクション法、遺伝子銃を用いた方法、レーザーを用いた方法、及び磁性ナノ粒子を用いた方法等が挙げられるが、特に限定されない。なお、前記他の遺伝子又はその発現カセットの遺伝子導入が自殺遺伝子又は自殺遺伝子の発現カセットの遺伝子導入と同時に施される場合は、他の遺伝子又はその発現カセットの遺伝子導入は、非ウイルスベクターを用いる方法により施され、好ましくはトランスポゾン法により施される。
 本発明における導入遺伝子は、プロモーター、エンハンサー又はこれらの組み合わせが作動可能に連結されていてもよい。前記プロモーターは、特に限定されないが、例えば、サイトメガロウイルス(Cytomegalovirus;CMV)プロモーター、ポリペプチド鎖伸長因子遺伝子(Elongation Factor 1 alpha;EF1α)プロモーター、シミアンウイルス40(Simian Virus 40;SV40)プロモーター、ホスホグリセリン酸キナーゼ(phosphoglycerate kinase;PGK)プロモーター、及びCAGプロモーター等が挙げられる。
 本発明における導入遺伝子は、2種類以上の遺伝子がリンカーで連結されていてもよい。前記リンカーは、2種類以上の遺伝子を連結する核酸又はペプチドであれば特に限定されないが、例えば、IRES(internal ribosome entry site)、Ubx-IRES、2Aペプチド(P2A、T2A、E2A、F2A等)、及びGSリンカー等が挙げられる。
 本発明は、遺伝子修飾された羊膜由来間葉系幹細胞を選別する工程をさらに含むものでもよい。遺伝子修飾された羊膜由来間葉系幹細胞を選別する方法としては、例えば、前述した他の遺伝子(抗生物質耐性遺伝子、レポーター遺伝子、タグペプチド等)を利用する方法が挙げられる。より具体的な例としては、抗生物質を含有した培地中で遺伝子導入した細胞集団を培養して抗生物質耐性遺伝子を発現した細胞を選択する方法、セルソーターを用いてレポーター遺伝子を発現した細胞を選別する方法、及びタグペプチドに結合する抗体を用いてタグペプチドを発現した細胞をクロマトグラフィーにより分離する方法等が挙げられるが、特に限定されない。また、2種類以上の選別方法を組み合わせることもできる。
 本発明における遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団は、遺伝子修飾された羊膜由来間葉系幹細胞を1細胞以上含む細胞集団であれば特に限定されず、遺伝子修飾されていない羊膜由来間葉系幹細胞やその他の細胞を含む集団であってもよい。特に限定されないが、前記遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団における遺伝子修飾された羊膜由来間葉系幹細胞の比率は、例えば、0.01%以上、0.1%以上、1%以上、10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、75%以上、80%以上、85%以上、87%以上、88%以上、89%以上、90%以上、91%以上、92%以上、93%以上、94%以上、又は95%以上である。
 本発明は、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団を培養する工程をさらに含むものでもよい。羊膜由来間葉系幹細胞を含む細胞集団を培養する工程の培地は、任意の動物細胞培養用液体培地を基礎培地とし、必要に応じて他の成分を適宜添加することにより調製することができる。前記他の成分としては、例えば、アルブミン、血清、血清代替試薬、血小板溶解物、サイトカイン、及び抗生物質等が挙げられるが、特に限定されない。また、2種類以上の他の成分を組み合わせて添加してもよい。
 前記基礎培地としては、例えば、BME培地、BGJb培地、CMRL1066培地、Glasgow MEM培地、Improved MEM Zinc Option培地、IMDM培地(Iscove’s Modified Dulbecco’s Medium)、Medium 199培地、Eagle MEM培地、αMEM(Alpha Modification of Minimum Essential Medium Eagle)培地、DMEM培地(Dulbecco’s Modified Eagle’s Medium)、ハムF10培地、ハムF12培地、RPMI 1640培地、Fischer’s培地、及びこれらの混合培地(例えば、DMEM/F12培地(Dulbecco’s Modified Eagle’s Medium/Nutrient Mixture F-12 Ham))等の培地を使用することができるが、特に限定されない。また、市販の各種の無血清培地も使用できる。
 遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団を培養する工程の播種密度は、特に限定されない。例えば500~100,000細胞/cmの密度で播種することができる。前記播種密度の下限としては、例えば500細胞/cm以上、1,000細胞/cm以上、2,000細胞/cm以上、3,000細胞/cm以上、4,000細胞/cm以上、又は5,000細胞/cm以上が好ましい。また、前記播種密度の上限としては、細胞培養の効率性の観点から、100,000細胞/cm以下を用いることが一般的である。他にも、例えば50,000細胞/cm以下、30,000細胞/cm以下、20,000細胞/cm以下、又は15,000細胞/cm以下が使用できる。
 前記遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団を培養する工程は、継代工程を含んでもよいし、異なる培養条件で複数回培養を繰り返す工程を含んでもよい。
 前記培養工程の培養期間中は、培地を交換する工程を含んでもよい。交換先の培地は、交換元の培地と同一の組成の培地であってもよいし、異なる組成の培地であってもよい。培地を交換する頻度は、特に限定されない。例えば1日毎、2日毎、3日毎、4日毎、5日毎、6日毎、及び7日毎が挙げられる。
 前記遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団を培養する工程後のコンフルエント率は、特に限定されない。より多くの細胞を得る観点から、例えば50%以上、好ましくは60%以上、より好ましくは70%以上、さらに好ましくは80%以上、さらに好ましくは90%以上、最も好ましくは95%以上である。他にも、1%以上、10%以上、30%以上、及び100%が挙げられる。
 本発明においては、前記培養工程にて培養した細胞を回収する工程をさらに含んでもよい。培養した細胞の回収方法は、特に限定されないが、例えば以下のようにして行うことができる。まず、所定のコンフルエント率となるまで培養した細胞を、細胞剥離手段にて処理して培養容器から剥離させる。次に、得られた細胞懸濁液を遠心分離し、上清を除去し、得られた細胞ペレットを培地又は任意の溶媒にて懸濁する。必要に応じて細胞懸濁液をさらに遠心分離し、洗浄液を用いて洗浄する工程を含んでもよい。
 前記の細胞剥離手段として、例えば、細胞剥離剤を使用してもよい。細胞剥離剤は、少なくとも接着細胞を培養容器から剥離する作用を有する薬剤であれば特に限定されず、例えば、トリプシン、コラゲナーゼ、ディスパーゼ、エチレンジアミン四酢酸(EDTA)等が挙げられる。細胞剥離剤として、市販の細胞剥離剤を用いてもよい。例えば、トリプシン-EDTA溶液(Thermo Fisher Scientific社製)、TrypLE Select(Thermo Fisher Scientific社製)、Accutase(Stemcell Technologies社製)、及びAccumax(Stemcell Technologies社製)等が挙げられるが、これらに限定されない。また、細胞剥離手段として、物理的な細胞剥離手段を使用してもよく、例えば、セルスクレーパー(コーニング社製)を使用することができるが、これに限定されない。細胞剥離手段は、単独で使用してもよく、複数を組み合わせて使用してもよい。
 前記洗浄液としては、特に限定されないが、細胞に与えるダメージを低減する観点から、緩衝液や等張液がより好ましい。例えば、PBS、HBSS(-)、リンゲル液、乳酸リンゲル液、ブドウ糖液、各種輸液、生理食塩液、培養液、基礎培地、アルブミン溶液、血液由来成分、及びそれらの混合物等が挙げられるが、これらに限定されない。
 本発明においては、前記回収工程を経て回収した遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団を、凍結保存する工程をさらに含んでも良い。前記凍結保存の手段は、特に限定されない。例えば、プログラムフリーザー、ディープフリーザー、液体窒素での凍結保存等が挙げられる。上記の凍結保存手段としてプログラムフリーザーを用いた場合、凍結する際の温度は、好ましくは-30℃以下、-40℃以下、-50℃以下、-80℃以下、-90℃以下、-100℃以下、-150℃以下、-180℃以下、又は-196℃(液体窒素温度)以下である。プログラムフリーザーを用いた場合、凍結する際の好ましい凍結速度は、例えば、15℃/分以下、11℃/分以下、10℃/分以下、9℃/分以下、5℃/分以下、2℃/分以下、又は1℃/分以下である。上記の凍結保存手段としてプログラムフリーザーを用いた場合、例えば、少なくとも室温から-10℃の間は、1℃/分以上、2℃/分以下の凍結速度となるように設定した上で、それ以外は適宜冷却速度を変更し最終的には目的とする凍結温度(例えば-80℃から-150℃)に到達させるのが好ましい。また、上記の凍結手段として液体窒素を用いた場合、例えば、-196℃まで急速に温度を下げて凍結させた後、液体窒素(気相)中で保存することができる。また液体窒素(液相)中で保存することもできる。
 前記の凍結手段により凍結する際、凍結する細胞は任意の保存容器に入った状態で凍結されてよい。前記保存容器としては、例えば、クライオチューブ、クライオバイアル、凍結用バッグ、及び輸注バッグ等が挙げられるが、これらに限定されない。
 前記の凍結手段により凍結する際、凍結する細胞は、任意の凍結保存液中で凍結されてもよい。前記凍結保存液としては、市販の凍結保存液を用いてもよい。例えば、BAMBANKER(リンフォテック社製)、STEM-CELLBANKER(日本全薬工業社製)、CP-1(極東製薬工業社製)、ReproCryo RM(リプロセル社製)、CryoNovo(Akron Biotechnology社製)、MSC Freezing Solution(Biological Industries社製)、及びCryoStor(HemaCare社製)等が挙げられるが、これらに限定されない。
 前記凍結保存液は、所定濃度の多糖類を含有することができる。多糖類の好ましい濃度は、例えば、1質量%以上、2質量%以上、4質量%以上、又は6質量%以上である。また、多糖類の好ましい濃度は、例えば、20質量%以下、18質量%以下、16質量%以下、14質量%以下、又は13質量%以下である。多糖類としては、例えば、ヒドロキシルエチルデンプン(HES)やデキストラン(Dextran40等)等を挙げることができるが、これらに限定されない。
 前記凍結保存液は、所定濃度のジメチルスルホキシド(DMSO)を含有することができる。DMSOの好ましい濃度は、例えば、1質量%以上、2質量%以上、3質量%以上、4質量%以上、又は5質量%以上である。また、DMSOの好ましい濃度は、例えば、20質量%以下、18質量%以下、16質量%以下、14質量%以下、12質量%以下、又は10質量%以下である。
 前記凍結保存液は、0質量%より多い所定濃度のアルブミンを含有するものでもよい。アルブミンの好ましい濃度は、例えば、1質量%以上、2質量%以上、3質量%以上、又は4質量%以上である。また、アルブミンの好ましい濃度は、例えば、30質量%以下、20質量%以下、10質量%以下、又は9質量%以下である。アルブミンとしては、例えば、ウシ血清アルブミン(BSA)、マウスアルブミン、及びヒトアルブミン等を挙げることができるが、これらに限定されない。
 [3]遺伝子導入した羊膜由来間葉系幹細胞を含む細胞製剤
 本発明の別の実施態様では、自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入した羊膜由来間葉系幹細胞を含む細胞製剤を提供する。
 本発明における細胞製剤は、自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入した羊膜由来間葉系幹細胞を有効成分として含み、任意選択可能な構成成分として、製薬上許容し得る溶媒及び/又はその他添加物を含む。前記有効成分は、[2]遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造方法に記載した製造方法で製造した、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の構成成分であってよい。
 前記有効成分の含有量は特に限定されず、細胞製剤の剤形、溶媒や担体の種類、投与量、投与回数、投与経路、使用目的、及び患者又は被験者の年齢、体重及び症状等によって適宜決定することができる。
 前記溶媒は、薬学的に許容可能な溶媒であれば特に限定されない。例えば、水、生理食塩水、ブドウ糖又はその他の補助剤を含む等張液、リン酸塩緩衝液、酢酸ナトリウム緩衝液が挙げられる。前記その他の補助剤としては、例えば、D-ソルビトール、D-マンノース、D-マンニトール、塩化ナトリウム、低濃度の非イオン性界面活性剤、及びポリオキシエチレンソルビタン脂肪酸エステル類等が挙げられる。
 前記その他添加物は、薬学的に許容可能な添加物であれば特に限定されない。例えば、賦形剤、結合剤、崩壊剤、充填剤、乳化剤、流動添加調節剤、滑沢剤、ヒト血清アルブミン、及びプロドラッグ等が挙げられる。
 本発明における細胞製剤は、プロドラッグと組み合わせて用いられるものでもよい。前記プロドラッグは、細胞製剤を投与する前に対象に投与されてもよく、細胞製剤と同時に対象に投与されてもよく、又は細胞製剤を投与した後に対象に投与されてもよい。
 本発明における細胞製剤は、例えば、癌、免疫関連疾患、虚血性疾患、下肢虚血、脳血管虚血、腎臓虚血、肺虚血、神経性疾患、移植片対宿主病(GVHD)、炎症性腸疾患、クローン病、潰瘍性大腸炎、放射線腸炎、全身性エリテマトーデス、紅斑性狼瘡、膠原病、脳卒中、脳梗塞、脳内血腫、脳血管麻痺、肝硬変、アトピー性皮膚炎、多発性硬化症、乾癬、表皮水疱症、糖尿病、菌状息肉腫(Alibert-Bazin症候群)、強皮症、軟骨等の結合組織の変性及び/又は炎症から起こる疾患、関節軟骨欠損、半月板損傷、離弾性骨軟骨症、無腐性骨壊死、変形性膝関節症、炎症性関節炎、関節リウマチ、眼疾患、血管新生関連疾患、虚血性心疾患、冠動脈性心疾患、心筋梗塞、狭心症、心不全、心筋症、弁膜症、創傷、上皮損傷、線維症、肺疾患、筋ジストロフィー、又は慢性膵炎、慢性腎炎から選択される疾患の治療剤として使用することができる。本発明における細胞製剤は、好ましくは癌の治療剤として使用され得る。
 前記癌は、例えば、扁平上皮癌、肺癌、腹膜癌、中皮腫、結腸癌、胆管腫瘍、鼻咽頭癌、喉頭癌、気管支癌、口腔癌、骨肉腫、軟部肉腫、胆嚢癌、腎臓癌、白血病、膀胱癌、黒色腫、脳癌、神経膠腫、脳腫瘍、皮膚癌、膵臓癌、乳癌、肝癌、骨髄癌、食道癌、大腸癌、胃癌、子宮頸癌、子宮内膜癌、前立腺癌、卵巣癌、頭頸部癌、直腸癌、及び精巣癌等が挙げられる。本発明における細胞製剤は、好ましくは脳腫瘍の治療剤として使用され得る。
 本発明の一以上の実施形態の細胞製剤の投与方法は、特に限定されない。例えば、皮下注射、リンパ節内注射、静脈内注射、腹腔内注射、胸腔内注射、局所への直接注射、及び局所に直接移植すること等が挙げられる。
 本発明の一以上の実施形態の細胞製剤は、注射用製剤、細胞塊又はシート状構造の移植用製剤、又は任意のゲルと混合したゲル製剤として用いることも可能である。
 本発明の一以上の実施形態の細胞製剤が対象とする患者又は被験者は、典型的にはヒトであるが、他の動物であってもよい。他の動物としては、サル、ウシ、ウマ、イヌ、ネコ、ブタ、ヒツジ、マウス、ラット、及びウサギ等の哺乳類、インコ、オウム、及びニワトリ等の鳥類が挙げられる。
 [4]癌治療用キット
 本発明の別の実施態様では、
(i)自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入した羊膜由来間葉系幹細胞を含む細胞集団、及び
(ii)プロドラッグを含む癌治療用キットを提供する。
 本発明における癌治療用キットは、自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入した羊膜由来間葉系幹細胞を含む細胞集団とプロドラッグの併用処理の目的のために製造されるものである。治療を要する対象に、プロドラッグを投与する前に、プロドラッグ投与と同時に、プロドラッグを投与した後に、又は複数それらの2以上の組み合わせで、自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入した羊膜由来間葉系幹細胞を含む細胞集団を投与するように製造されたキットである。
 前記細胞集団は、[2]遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造方法に記載した製造方法で製造した、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団であってよい。
 前記対象は、典型的にはヒトであるが、他の動物であってもよい。他の動物としては、サル、ウシ、ウマ、イヌ、ネコ、ブタ、ヒツジ、マウス、ラット、ウサギ等の哺乳類、インコ、オウム、及びニワトリ等の鳥類が挙げられる。前記対象は、ヒトを含む哺乳類であることが好ましく、癌治療を要する患者であることがさらに好ましい。前記癌治療を要する患者としては、癌治療中の患者、癌治療を受けた事がある患者、癌治療を受ける必要がある患者をいずれも含み、癌治療のために癌を摘出する外科的手術を施した患者も含まれ得る。
 前記癌は、「前記[3]遺伝子導入した羊膜由来間葉系幹細胞を含む細胞製剤」に記載の癌に準ずる。
 本発明における癌治療用キットは、第1区画と第2区画で構成されてもよく、第1区画に自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入した羊膜由来間葉系幹細胞を含む細胞集団を含んでもよく、第2区画にプロドラッグを含んでもよい。前記キットは、便利性及び携帯性を高めるために、それぞれ1回の投与量に分けて一つの容器内に含有する、又は相違するいくつかの小容器内に含有してもよい。
 本発明のキットは、必要に応じて、使用に必要な装備及び各成分の投与方法を記載した指示資料等を含んでもよい。
 以下の実施例にて本発明を具体的に説明するが、本発明は実施例によって限定されるものではない。
 (比較例1)
 レンチウイルスベクターを用いたVenus発現羊膜由来間葉系幹細胞(羊膜MSC)の作製
 ヒト羊膜MSC(継代数2)を20,000cells/cmの播種密度で6ウェルプレート(コーニング社製)に播種し、ヒト血小板溶解物を終濃度5%含有するαMEM中で2日間培養した。培養後の細胞の培地を除去し、Venus発現カセットを搭載したレンチウイルスベクター含有液を添加して0.5時間培養し、レンチウイルスベクター含有液と等量の培地を添加して18時間培養した。次いで、ウイルス含有液を除去し、培地を添加して24時間培養したのち、培養細胞をTrypLE Select(サーモフィッシャーサイエンティフィック社製)を用いて剥離し、培地を用いて希釈し、遠心分離によりVenus発現羊膜MSCを含む細胞集団を回収した。
 前記Venus発現カセットを搭載したレンチウイルスベクター含有液は、以下のようにして調製した。まず、CMVプロモーター制御下にVenus遺伝子を連結した核酸配列をレンチウイルスベクター作製プラスミドに導入し、Venus発現カセット搭載レンチウイルスベクター作製プラスミドを作製した。本プラスミド及びレンチウイルスパッケージングプラスミドをレンチウイルスパッケージング用細胞株であるLenti-X 293T(タカラバイオ社製)にLipofectamine 3000(サーモフィッシャーサイエンティフィック社製)を用いてトランスフェクションし、48時間培養した。その後、Lenti-X 293T培養上清を回収し、0.8μmフィルターで濾過して、Venus発現カセットを搭載したレンチウイルスベクター含有液を調製した。
 Venus発現羊膜MSCを大量製造するために、前記回収したVenus発現羊膜MSCを含む細胞集団に含まれるVenus発現羊膜MSCの増殖を試みた。具体的には、前記回収したVenus発現羊膜MSCを含む細胞集団を10,000cells/cmの播種密度でT25フラスコ(住友ベークライト社製)に播種し、ヒト血小板溶解物を終濃度5%含有するαMEM中で培養した。培養3日目に、顕微鏡下で細胞培養状態の明視野観察(×40倍率、露光時間1/25秒)及び緑色蛍光観察(×40倍率、露光時間1秒)を行った(図1A)。さらに7日間培養した培養10日目に、細胞培養状態を再度顕微鏡下で明視野観察(×40倍率、露光時間1/25秒)及び緑色蛍光観察(×40倍率、露光時間1秒)した(図1B)。その結果、明視野観察画像で細胞増殖を認め、かつ緑色蛍光観察画像において蛍光シグナルを発するVenus発現細胞が増えていたことから、播種した細胞集団中に含まれるVenus発現羊膜MSCが増殖していたことがわかった。すなわち本結果は、レンチウイルスベクターを用いて自殺遺伝子以外の遺伝子を遺伝子導入した場合、羊膜由来間葉系幹細胞の増殖能に問題はないことを示す。
 (比較例2)
 レンチウイルスベクターを用いたHSV-TK発現羊膜MSCの作製
 ヒト羊膜MSC(継代数2)を20,000cells/cmの播種密度で6ウェルプレート(コーニング社製)に播種し、ヒト血小板溶解物を終濃度5%含有するαMEM中で2日間培養した。培養後の細胞の培地を除去し、HSV-TK発現カセットを搭載したレンチウイルスベクター含有液を添加して0.5時間培養し、レンチウイルスベクター含有液と等量の培地を添加して18時間培養した。次いで、ウイルス含有液を除去し、培地を添加して24時間培養したのち、比較例1と同様の方法でHSV-TK発現羊膜MSCを含む細胞集団を剥離・回収した。
 前記HSV-TK発現カセットを搭載したレンチウイルスベクター含有液は、以下のようにして調製した。まず、EF-1αプロモーター制御下にHSV-TK遺伝子を配置し、さらにHSV-TK遺伝子の下流にIRESを介してVenus遺伝子を連結した核酸配列をレンチウイルスベクター作製プラスミドに導入し、HSV-TK発現カセット搭載レンチウイルスベクター作製プラスミドを作製した。HSV-TK遺伝子の下流にIRESを介してVenus遺伝子を連結することで、HSV-TK発現細胞をVenus蛍光シグナルにより判別できるようにした。本プラスミド及びレンチウイルスパッケージングプラスミドをレンチウイルスパッケージング用細胞株であるLenti-X 293T(タカラバイオ社製)にLipofectamine 3000(サーモフィッシャーサイエンティフィック社製)を用いてトランスフェクションし、48時間培養した。その後、Lenti-X 293T培養上清を回収し、0.8μmフィルターで濾過して、HSV-TK発現カセットを搭載したレンチウイルスベクター含有液を調製した。
 HSV-TK発現羊膜MSCを大量製造するために、前記回収したHSV-TK発現羊膜MSCを含む細胞集団に含まれるHSV-TK発現羊膜MSCの増殖を試みた。具体的には、前記回収したHSV-TK発現羊羊膜MSCを含む細胞集団を10,000cells/cmの播種密度でT25フラスコ(住友ベークライト社製)に播種し、ヒト血小板溶解物を終濃度5%含有するαMEM中で培養した。培養3日目に、顕微鏡下で細胞培養状態の明視野観察(×40倍率、露光時間1/25秒)及び緑色蛍光観察(×40倍率、露光時間2.5秒)を行った(図2A)。さらに7日間培養した培養10日目に、細胞培養状態を再度顕微鏡下で明視野観察(×40倍率、露光時間1/25秒)及び緑色蛍光観察(×40倍率、露光時間5秒)した(図2B)。その結果、明視野観察画像では細胞増殖を認めたが、緑色蛍光観察画像では蛍光シグナルを発するHSV-TK発現細胞の増殖は認められなかった。このことから、HSV-TK発現羊膜MSCの増殖に失敗したことがわかった。明視野観察画像においてHSV-TK未導入細胞の増殖は認められたことから、HSV-TKの遺伝子導入及び発現により羊膜MSCの増殖能が大きく低下ことが示された。
 (実施例1)
 トランスポゾン法を用いたHSV-TK発現羊膜MSCの作製
 トランスポゾン法を用いてHSV-TK発現羊膜MSCを作製するに当たり、HSVTK発現羊膜MSCの視認性を高めるために、まずはレンチウイルスベクターを用いて羊膜MSCにVenus遺伝子を導入した。具体的には、比較例1と同様の方法でVenus発現羊膜MSCを含む細胞集団を回収し、セルソーターFACSAriaIII(BD社製)を用いて、蛍光シグナルを指標にしてVenus発現羊膜MSCを選別し、回収した。
 Venus発現羊膜MSCを10,000cells/cmの播種密度で15cmディッシュ(住友ベークライト社製)に播種し、ヒト血小板溶解物を終濃度5%含有するαMEM中で3日間培養した。培養後の細胞をTrypLE Select(サーモフィッシャーサイエンティフィック社製)を用いて剥離し、培地を用いて希釈し、遠心分離により回収した。Cell Line Nucleofector Kit L(Ronza社製)及びNucleofector II/2b(Ronza社製)を用いたエレクトロポレーション法を用いて、HSV-TK発現カセットをトランスポザーゼが認識する反復配列に隣接して配置したコンストラクトを構造に持つプラスミドベクター、及びトランスポザーゼ発現カセットを構造に持つプラスミドベクター(計2種類のプラスミドベクター)を回収細胞にトランスフェクションした。
 前記コンストラクトは、EF-1αプロモーター制御下にHSV-TK遺伝子を配置し、さらにHSV-TK遺伝子の下流にT2Aを介してピューロマイシン耐性遺伝子を連結した発現カセットの両端にトランスポザーゼが認識する反復配列を配置した。HSV-TK遺伝子の下流にT2Aを介してピューロマイシン耐性遺伝子を連結することで、HSV-TK発現細胞を薬剤選択により選別できるようにした。
 トランスフェクション後の細胞をT75フラスコに播種して24時間培養した後、ピューロマイシンを終濃度1μg/mLで添加し、培養した。培養3日目に、顕微鏡下で細胞培養状態の明視野観察(×40倍率、露光時間1/25秒)及び緑色蛍光観察(×40倍率、露光時間3秒)を行った(図3A)。さらに7日間培養した培養10日目に、細胞培養状態を再度顕微鏡下で明視野観察(×40倍率、露光時間1/25秒)及び緑色蛍光観察(×40倍率、露光時間1秒)した(図3B)。その結果、明視野観察画像及び緑色蛍光観察画像においてピューロマイシン耐性を示すHSV-TK発現細胞が増えていたことから、播種した細胞集団中に含まれるHSV-TK発現羊膜MSCが増殖していたことがわかった。
 上記結果より、ウイルスベクターを用いてHSV-TKを遺伝子導入した羊膜MSCは増殖が阻害される一方、プラスミドベクターを用いてトランスポゾン法によりHSV-TKを遺伝子導入した羊膜MSCは、増殖能が低下することなく効率的に製造することが可能であることが示された。
 (実施例2)
 トランスポゾン法を用いて作製したHSV-TK発現羊膜MSCの脳腫瘍に対する抗癌効果の評価
 HSV-TK発現カセットの遺伝子導入にトランスポゾン法を用いて作製したHSVTK発現羊膜MSC(TK-AMSC)が、脳腫瘍に対して抗癌効果を有することを確認するための実験を行った。まず、実施例1に記載の方法で作製し、ピューロマイシン耐性を指標にして選別したTK-AMSCを、比較例1と同様の方法で剥離・回収した。回収後のTK-AMSCを1,000cells/cmの播種密度で15cmディッシュ(住友ベークライト社製)に播種し、ヒト血小板溶解物を終濃度5%含有するαMEM中で7日間培養した。培養後のTK-AMSCを比較例1と同様の方法で剥離・回収し、2×10cellsのTK-AMSCを50%マトリゲル(コーニング社製)を含有した生理食塩液50μLに懸濁し、TK-AMSC投与液を調製した。また、2×10cellsのヒト脳腫瘍由来細胞株であるU-87MGを50%マトリゲル(コーニング社製)を含有した生理食塩液50μLに懸濁し、U-87MG投与液を調製した。TK-AMSC投与液及び/又はU-87MG投与液を生後5週齢の免疫不全ヌードマウス(Balb/c-nu/nu)の右腹側部皮下に移植した。細胞移植の翌日より7日間、ガンシクロビル(GCV)を50mg/kgの容量で1日1回腹腔内に投与した。細胞移植後1日、3日、7日、10日、14日、17日、21日、24日、28日、31日に腫瘍の長径及び短径を計測し、計算式(腫瘍体積(mm)=長径(mm)×短径(mm)×短径(mm)÷2)をもとに腫瘍体積を算出してグラフ化した(図4)。その結果、TK-AMSCの移植及びGCV投与処置を施した実験群(TK-AMSC(+),GCV(+))は、無処置群(TK-AMSC(-),GCV(-))及びガンシクロビル投与処置のみを施した実験群(TK-AMSC(-),GCV(+))と比較して顕著に腫瘍の成長が抑制された。本結果は、HSV-TK発現羊膜由来間葉系幹細胞移植及びガンシクロビル投与を施した試験群において、腫瘍の成長阻害効果を認めたことを示す。
 以上の結果より、実施例に記載の方法で作製したHSV-TK発現羊膜MSCが優れた抗癌効果を有し、癌治療用の細胞製剤として非常に有用であることが確認された。

Claims (19)

  1.  羊膜由来間葉系幹細胞に、プラスミドベクターを用いて自殺遺伝子又は自殺遺伝子の発現カセットを遺伝子導入する工程を含む、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団の製造方法。
  2.  前記プラスミドベクターが、エレクトロポレーション法により細胞内に導入される、請求項1に記載の製造方法。
  3.  前記遺伝子導入工程で、前記自殺遺伝子又は自殺遺伝子の発現カセットを染色体に組み込む、請求項1に記載の製造方法。
  4.  前記遺伝子導入工程において、前記自殺遺伝子又は自殺遺伝子の発現カセットを、トランスポザーゼが認識する反復配列に隣接して配置したコンストラクトを含むプラスミドベクターと、トランスポザーゼ発現カセットを含むプラスミドベクターの2種類のプラスミドベクターを用いる、請求項1に記載の製造方法。
  5.  前記遺伝子導入工程において、前記自殺遺伝子又は自殺遺伝子の発現カセットを、トランスポザーゼが認識する反復配列に隣接して配置したコンストラクト、及びトランスポザーゼ発現カセットを含むプラスミドベクターを用いる、請求項1に記載の製造方法。
  6.  前記自殺遺伝子が、単純ヘルペスウイルスチミジンキナーゼ遺伝子又はシトシンデアミナーゼ遺伝子である、請求項1に記載の製造方法。
  7.  前記自殺遺伝子が、単純ヘルペスウイルスチミジンキナーゼ遺伝子である、請求項1に記載の製造方法。
  8.  請求項1~7のいずれか一項に記載の製造方法により製造される、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞製剤。
  9. 羊膜由来間葉系幹細胞を含む細胞製剤であって、前記羊膜由来間葉系幹細胞が、
    (i)自殺遺伝子又は自殺遺伝子の発現カセット、及び
    (ii)トランスポザーゼが認識する反復配列
    が染色体に組み込まれている、細胞製剤。
  10.  前記細胞製剤がプロドラッグと組み合わせて使用される、請求項8又は9に記載の細胞製剤。
  11.  前記プロドラッグがガンシクロビル又は5-フルオロシトシンである、請求項10に記載の細胞製剤。
  12.  前記プロドラッグがガンシクロビルである、請求項11に記載の細胞製剤。
  13.  前記細胞製剤が癌治療用である、請求項8又は9に記載の細胞製剤。
  14.  前記細胞製剤が脳腫瘍治療用である、請求項12に記載の細胞製剤。
  15.  (i)請求項1~7のいずれか一項に記載の製造方法により製造される、遺伝子修飾された羊膜由来間葉系幹細胞を含む細胞集団、及び
     (ii)プロドラッグ
    を含む癌治療用キット。
  16.  請求項9に記載の細胞製剤、及びプロドラッグを含む癌治療用キット。
  17.  前記プロドラッグがガンシクロビル又は5-フルオロシトシンである、請求項15又は16に記載の癌治療用キット。
  18.  前記プロドラッグがガンシクロビルである、請求項17に記載の癌治療用キット。
  19.  前記癌が脳腫瘍である、請求項15又は16に記載のキット。
