  • Cell information acquisition method 2 The cell adhesion area was evaluated as follows. After replacing the culture medium in the cell-cultured dish with a detachment solution, the cells were placed under a phase-contrast microscope (Axio Obserer 7; manufactured by Carl Zeiss) and observed under phase contrast, as described in the cell adhesion area evaluation method. The average value was calculated from the sum of the projected areas of the cells within a 40x magnification field, and designated the cell adhesion area B1. Next, the detachment operation of step [1] was performed according to the detachment method described above, and phase-contrast observation was performed using a phase-contrast microscope immediately before starting step [2]. The cell projection area was calculated as described above.

From the images thus obtained, the average brightness value of the culture surface was calculated and designated as cell brightness information C1.

  • Example 1 CHO cells on a dish were detached using detachment method 1. The detached cells were collected and then counted using a hemocytometer. The viability was calculated using a viability test using trypan blue staining. Furthermore, all cells that could not be detached by ultrasound were detached from the dish using a cell scraper, and the cell count was measured using a hemocytometer. The total number of detached cells was calculated by adding the number of cells detached by ultrasound and the number of cells subsequently detached with the cell scraper. The detachment rate was calculated as the ratio of the number of ultrasonically detached cells to the total number of detached cells. The resulting viability was 96.0%, and the detachment rate was 98.0%. A viability and detachment rate of 90% or higher was considered good. The cell types used, detachment methods, calculated viability, and detachment rates are shown in Table 6.

Description

細胞剥離方法及び細胞剥離システム
 本開示は、細胞剥離方法及び細胞剥離システムに関するものである。
 細胞医薬・再生医療の分野においては、大量の細胞が必要であり、特に、生体組織の多くを占める接着性細胞を効率的かつ安定的に供給することが求められている。
 接着性細胞の培養においては、例えばポリスチレン製ディッシュのような培養基材上での細胞の培養、培養基材からの細胞の剥離、細胞の回収及び洗浄の各工程を経て、目的とする細胞を取得する。細胞をさらに増やす際には、取得した細胞の一部を新しい培養基材に移して培養する、いわゆる継代操作を行なう。このような細胞の取得、及び継代培養の工程の中でも、細胞に対するダメージを与えずに、高い剥離率で回収するために、剥離方法の検討がされている。
 一方、せん断力を用いた細胞剥離方法が提案されている。特許文献1には、細胞と、培地又は液体とを保持する容器に対して超音波振動を付与し、細胞の少なくとも一部を容器から剥離させる細胞剥離装置の開示がある。また特許文献2には、培養液の流動による機械的な刺激により細胞を剥離させる方法の開示がある。本発明者らは上記先行技術について課題を見出した。すなわち、容器に対する接着力が大きい細胞のように、剥離が困難な細胞を取り扱う場合などに、より剥離効率が強い条件で剥離を行うと細胞の生存率が低下する場合がある。

 そこで本開示は、細胞の生存率と剥離率がともに高くなるような細胞剥離方法、及びそれを用いた細胞剥離システムを提供することを目的とする。
 本発明者らは、細胞の剥離方法を検討する中で、培養基材に超音波帯域の振動を付与した後に培養基材の培養面に対してせん断力を付与した場合に、細胞にダメージを与えることなく高効率に剥離が可能である事を見出した。
 すなわち、本開示に係る細胞剥離方法は、培養基材の上で培養された接着細胞の剥離方法であって、前記培養基材に対して、超音波帯域の振動を付与する振動付与工程、及び前記培養基材の培養面に対して、せん断力を付与するせん断力付与工程を含み、前記振動付与工程の開始が、前記せん断力付与工程の開始よりも先であり、前記せん断力は超音波の振動以外の手段によって付与されることを特徴とする。
 また、本開示に係る細胞剥離システムは、培養基材の上で培養された接着細胞を剥離する剥離部、及び制御部を有する細胞剥離システムであって、前記剥離部は、前記培養基材に対して、超音波帯域の振動を与える振動付与部、及び前記培養基材の培養面に対して、せん断力を付与する、せん断力付与部を有し、前記制御部は、前記振動付与部の駆動の開始が、前記せん断力付与部の駆動の開始よりも先となるよう制御することを特徴とする。

(細胞剥離方法)
 本開示は、培養基材の上で培養された接着細胞の剥離方法であって、前記培養基材に対して、超音波帯域の振動を付与する振動付与工程、及び前記培養基材の培養面に対して、せん断力を付与するせん断力付与工程を含み、前記振動付与工程の開始が、前記せん断力付与工程の開始よりも先であり、前記せん断力は超音波の振動以外の手段によって付与されることを特徴とする細胞の剥離方法を提供する。
 本開示の細胞剥離方法の具体例の一形態を図1A及び図1Bに示す。
 図1Aに示すように、まず、培養基材を設置して超音波帯域の振動を付与する振動付与工程(ステップS110)を開始する。
 次いで、培養基材を設置してせん断力付与工程(ステップS140)を開始する。最後に、剥離された細胞を回収する(ステップS150)。
 また、図1Bに示すように、振動付与工程(ステップS110)の開始は、せん断力付与工程(ステップS140)の開始より先に行われる。
 さらなる一形態を図2A及び図2Bに示す。
 図2Aに示すように、まず、培養基材を設置して超音波帯域の振動を付与する振動付与工程(ステップS210)を開始する。その後、培養基材の細胞情報を取得する情報取得工程(ステップS220)を実行する。取得した細胞情報に基づいて、せん断力付与工程の開始タイミングを決定するせん断力付与工程開始時刻決定工程(ステップ230)を実行する。次いで、培養基材を設置してせん断力付与工程(ステップS240)を開始する。
 最後に、剥離された細胞を回収する(ステップS250)。
 また、図2Bに示すように、振動付与工程(ステップS210)の開始は、せん断力付与工程(ステップS240)の開始より先に行われる。

 振動付与工程では、培養基材に対して超音波帯域の振動を与えることによって、培養基材から細胞を剥離しやすくする。
 本発明者らは、超音波帯域の振動を細胞が培養されている培養基材に対して与えると、細胞が培養基材から剥離する前の段階において、細胞の接着力を低下させることができることを見出し、この作用を積極的に利用することを検討した。
 本開示の超音波帯域の振動とは、10kHz以上1MHz以下の周波数をもつ振動のことを指す。また、振動付与工程の超音波帯域の振動を付与する時間としては特に限定はないが、細胞の接着力を効果的に低下させ、細胞の生存率も高める観点からは、例えば1秒以上1時間以下、好ましくは10秒以上30分以下、さらに好ましくは30秒以上15分以下である。本開示の細胞剥離方法における細胞の環境(細胞剥離液の種類や環境温度、CO濃度など)によっては、長時間にわたり超音波振動が付与されることが好ましくない場合がある。また、時間が短すぎると超音波帯域の振動によって細胞の接着力が十分に低下しない場合がある。
 振動付与工程の超音波帯域の振動の振動面の最大振幅としては、特に制限はないが、細胞生存率を維持する観点からは、10μm以下が望ましい。この範囲であれば、振動体の発熱の影響を抑え、細胞に接着力を低下させるために適した量の振動のエネルギーを伝えることができる。また、振幅を10μm以下とすることで超音波帯域の振動印加時にキャビテーションが発生しづらくなり、細胞生存率が低下しづらくなるため好ましい。
 超音波帯域の振動の方向は、培養面に対して垂直な方向の成分を含むことが好ましい。
 本開示において、超音波帯域の振動の方向とは、超音波帯域の振動を発生させる振動体の振動面と、超音波帯域の振動を伝達させる振動伝達部材とを結ぶ方向を指す。
 特に培養基材の底面より超音波帯域の振動を付与することで、培養面に対して垂直な方向の成分を有する超音波帯域の振動を付与することが可能となり、また、培養面に対して垂直な方向の成分を含むことで、細胞の接着力を低下させやすくなる。
 超音波帯域の振動パターンは、振動付与手段として使用する振動体や、超音波デバイスの種類によって様々な振動モードを使用することができる。その中で細胞種や剥離の条件に応じて様々な振動モードを選択することができるが、好ましくは容器全体において均一な振幅が励起される振動モードであり、例えばランジュバン型振動子のように面内均一性の高い振動体を使用する場合が挙げられる。また、円環状圧電素子をガラス板などに張り付けたリング状振動子のように曲げ振動で様々な振動モードを発生させることができる振動体を使用する場合は、複数に電極を分割することで発生させることができる進行波や搬送波など一定周期で均一な振幅を付与するモードが好ましい。培養基材器底面における面内の振幅が均一に近づくことで超音波帯域の振動を印加した際に容器の任意の位置間における振幅変化が小さくなるため、培養している細胞間の影響差を少なくすることが可能である。
 また、振動付与手段に用いる振動体の種類や駆動方法によっては、駆動中に発熱する場合がある。印加電圧や使用周波数帯、装置環境にもよるが、共振周波数での駆動を続けることで、この傾向が強まる傾向にある。それに対し、一定間隔で駆動と休止を繰り返すこと(バースト駆動)や、一定の周波数範囲内で繰り返し、かつ、連続的に駆動周波数を変化させること(スイープ駆動)、などを単独、或いは併用することで、発熱量を制御することが可能である。さらに、発熱を抑制するために、振動付与手段の周辺に冷却のための機構を備えて振動体を直接的に、或いは環境温度を介して間接的に制御することもできる。
 また、駆動中の振動体の発熱によって共振周波数が変化する場合があり、その場合は公知の、例えば電流検知による共振周波数追従制御などを行うこともできる。
 振動付与工程を行う際の環境温度としては特に限定はなく、使用する細胞種に応じて適宜設定することができる。細胞生存率を維持する観点からは、20℃以上40℃以下が好ましく、例えば哺乳動物細胞の場合、特に30.0℃以上37.5℃以下であることが好ましい。この範囲であれば、環境の温度が培養時の温度と近く、細胞への温度変化による影響を少なくでき、生存率を高く保つことができるためである。培養基材として例えば温度応答性膜を使用している公知の容器を使用する場合、振動付与工程の環境温度としては温度応答性に対応した温度域としても構わない。
 振動付与工程は細胞培養と同様にCO濃度5%の環境下で行っても構わない。振動付与工程を行う際の環境条件については細胞の種類や特性、剥離後の細胞使用方法に応じて適宜設定することができる。
(せん断力付与工程)
 せん断力付与工程では、培養基材の培養面に対してせん断力を加えることにより、培養基材から細胞を剥離させる。
 本発明者らは、さらに、培養された接着細胞は、超音波帯域の振動により培養基材に対する細胞の接着力を低下させると、せん断力の付与によって、より顕著に剥離しやすくなることを見出した。せん断力とは、細胞に対して側方から作用させる力をいう。つまり、超音波帯域の振動により接着力を低下させた後、細胞に側方から剥離させる力として、せん断力を別途加えることにより、細胞の生存率を下げることなく剥離することができるようになった。
 ここで、せん断力の付与方法としては、少なくとも培養基材へ付与される叩打(培養基材の叩打)、振とう(培養基材の振とう)、揺動(培養基材の揺動)、及び偏心回転(培養基材の偏心回転)、培養基材への液体の吐出、培養基材上での液体の撹拌などが挙げられる。なお、超音波の振動によって付与されるものは、せん断力には含めない。せん断力は超音波の振動以外の手段によって付与されるものとする。具体的には、以下の例が挙げられる。
 培養基材の叩打の例としては、適切な質量を有する部材を培養基材に衝突させる事で、培養基材に対して撃力を与えることを挙げられる。衝突させる部材は、棒状であっても良く、ハンマー状や球状であっても良いが、部材を培養基材に衝突させることで撃力を与える手段はこれに限定するものではない。また、基材へ叩打を付与する叩打部は、培養基材を平坦或いは円弧状、突起状の部材に衝突させることで撃力を付与することもできるし、これらの方法を組み合わせることもできる。これらの中でも本開示の叩打部は、適切な質量を有する部材を培養基材に衝突させて培養基材に撃力を付与する手段を用いることが装置構成を小型化する観点から好ましい。
 また、衝突させる部材は与える衝撃力に応じて、適宜重さや原材料を選ぶことができ、原材料としては金属やセラミックスなどの無機材料や、樹脂やゴムなどの有機材料、さらにはこれらの複合体など公知の材料を用いることができる。
 衝突させる部材を動かす動力源としては、例えばモーターやソレノイドといった通電によって力を発生させ得る方法を使用することができる。また、それらの動力源を直接使用しても良く、バネのようにエネルギーを貯蔵できる部材を介して使用することもできる。
 本開示の叩打部は、叩打方向の対側に微小変形部材を配置して培養基材に与える衝撃力、慣性力を制御することも可能である。微小変形部材の形状としては接点を最小化する観点より、円柱状や半球状、或いはその組み合わせが好ましく、材料としては弾性体、すなわちゴム材料が好ましい。
 培養基材の振とうの例としては、培養基材の培養面に対し平行な方向成分を含む加速度を加えることで、培養基材内の液体に流れを発生させる構成を使用することを挙げられる。
 例えば、培養基材を、モーターで回転運動させた上で、スライドレールなどを用いて一軸の往復運動させる構成を使用することできる。また、リニアモーターなどを用いて培養基材を加速させることで培養面に対し平行な方向成分や非平行な方向成分を含む加速度を加える構成を使用することができる。振とうによって培養基材の培養面に対して平行な方向成分や非平行な方向成分を含む加速度を加えることができる構成になるように、振とうの軸を適宜設計することが可能である。
 本開示の振とうの具体例について例示する。
 例えば、水平方向や垂直方向に往復運動する往復振とうが挙げられる。往復振とうはそれぞれ水平又は垂直方向成分を含んでいれば良く、水平方向や垂直方向のみに振とうしても良いし、これらを組み合わせた形で斜めに振とうすることもできる。また、振とう軸は1つである一軸に限らず、例えば二軸や多軸などの複数軸で異なる振とう方向に振とうしても構わない。
 また、他の振とう方法としては例えば、水平方向成分を含む方向に円運動をする旋回振とうが挙げられる。旋回振とうは水平方向成分を含んでいれば良く、水平方向のみに振とうしても良いし、上下方向を含む形で斜めに振とうすることもできる。また、円運動は真円状や楕円状、八の字状、あるいはこれらを組み合わせた形状となっても構わない。
 また、他の振とう方法としては例えば、シーソー振とうが挙げられる。シーソー振とうは培養基材内の一点を軸としてシーソーのように往復運動する振とう方法を指し、水平方向の往復、垂直方向の往復、又はこれらを組み合わせた往復運動などが挙げられる。
 また、他の振とう方法としては例えば、振り子のように支点を有して円弧状に振とうする、揺動運動が挙げられる。
 培養基材の振とうはこれらの具体例を単独で使用しても良いし、組み合わせて使用しても構わない。
 培養基材の偏心回転としては、撹拌動作のような回転運動の回転軸を偏心させた回転運動が挙げられる。具体的には前述したように回転運動の回転軸を偏心させた構成、さらには水平偏心震動、ボルテックス撹拌といった方法が挙げられる。
 培養基材への液体の吐出の例としては、例えば、ニードルなどから液体を吐出し培養面にあてることで培養面に沿って液を拡散させ付与する方法や、図15A及び図15Bに示し、後に説明するようにピペッターなどから液体を吐出、及び吸引を繰り返すことで培養面に沿って異なる方向から液の流れを付与する方法、さらには図16に示し、のちに説明するような培養面に沿った流路を作成しポンプやシリンジを用いて一方向もしく方向を切り替えて送液することで付与する方法などが挙げられる。
 また、液体の撹拌の例としては、例えば、図17に示すように撹拌翼を用いた撹拌機9などを用いて培養面に対して平行な流れを作りだすことで付与する方法、などが挙げられる。装置9は、図17に示されるように、せん断力付与機構を有する装置であり、培養基材10を保持する保持機構11と、せん断力付与するための、撹拌機構137を有する。
 液体の撹拌については、これら以外にも撹拌子を使用する方法など、公知の手法を用いることができる。
 これらのせん断力付与方法のうち好ましくは叩打、振とう、偏心回転、液体の吐出である。
 また、ここで例示したせん断力付与方法を単独で用いてもよいし、複数組み合わせてもよい。
 培養面に与えられるせん断力の大きさは、せん断力付与工程中に時間的に変化させることが望ましい。時間的な変化とは、例えば単調や、一定の割合や、任意の変化量でせん断力が増加又は減少したり、増加や減少を周期的、複数種の組合せで、又はランダムに繰り返すことなどが挙げられる。さらに、せん断力の大きさに対して、例えば5%以上の周期で周期的に変化してもよいし、正弦波状に変化してもよいし、せん断力をパルス状に繰り返して与えてもよい。
 これらの時間的な変化はせん断力の強度が変化すれば良く、例えばせん断力の強さそのものが変化しても良いし、単位時間当たりのせん断力の力積としてせん断力の強度を変化させても良い。
 本開示のせん断力付与工程で与えられるせん断力の大きさは、周期的に変化することが好ましい。一定の周期で変化することで、単位時間あたりに均等に一定の強度域のせん断力を付与することが可能となるため、剥離を効率化する観点で好ましい。
 本開示のせん断力付与工程において、せん断力の働く方向を変化させることが望ましい。せん断力の働く方向の変化については、時間的に変化させる場合や、異なる方向からのせん断力を同時に付与する場合などがある。本開示で例示しているせん断力の種類とその方向の具体例としては、例えば液体の吐出のように一方向からのせん断力や、往復振とうのように一つの軸で切り返す場合や二つの方向からの叩打のように二種類の方向から作用させるせん断力や、二軸ステージで旋回振とうさせたり、培養基材を回転させながら叩打させたりと多数の方向からのせん断力などが挙げられる。
 また、せん断力の働く方向と、培養面に加えるせん断力の大きさが、せん断力付与工程中に変化することが望ましい。これは、細胞の生育状態や接着状態には細胞個々でばらつきがあるため、せん断力の大きさやせん断力の働く方向を変化させることで、多面的な力を細胞にかけることができ、個々の細胞ごとに適したせん断力を付与できるためではないかと考えている。
 せん断力の働く方向は、培養面に対して平行な方向の成分を含むことが好ましい。
 培養面に対して平行な方向の成分を含むことで、振動付与工程において接着力が低下した細胞を、効果的に剥離しやすくなる。
 せん断力付与工程において、せん断力を与える時間、すなわち、せん断力付与工程を行う時間としては特に限定はないが、細胞を効果的に剥離させ、細胞の生存率も高める観点からは、例えば1秒以上1時間以下、好ましくは5秒以上30分以下、さらに好ましくは10秒以上10分以下である。せん断力を与える時間が長すぎると、せん断力によって細胞へ過剰なダメージを与える場合があり好ましくない。また、時間が短すぎると十分なせん断力を細胞に与えることができず、剥離効果が低くなる場合がある。
 せん断力付与工程を行う際の環境温度としては特に限定はないが、細胞生存率を維持する観点からは、20℃以上40℃以下が好ましく、特に30.0℃以上37.5℃以下であることが好ましい。この範囲であれば、環境の温度が培養時の温度と近く、細胞への温度変化による影響を少なくでき、生存率を高く保つことができるためである。培養基材として例えば温度応答性膜を使用している公知の容器を使用する場合、せん断力付与工程の環境温度としては温度応答性に対応した温度域としても構わない。
 また、せん断力付与工程は細胞培養と同様にCO濃度5%の環境下としても構わない。せん断力付与工程を行う際の環境条件については細胞の種類や特性、剥離後の細胞使用方法に応じて適宜設定することができる。
 また、本開示の細胞の剥離方法は、振動付与工程、又はせん断力付与工程以外のその他の工程を含んでも構わない。振動付与工程とせん断力付与工程の間にその他の工程を含んでもよいし、せん断力付与工程の実施中にその他の工程を含んでもよいし、せん断力付与工程の終了後にその他の工程を含んでもよく、また振動付与工程が開始する前に含んでもよい。
 その他の工程としては、剥離細胞回収工程、情報取得工程、せん断力付与工程開始時刻決定工程、さらには、上述の培養基材の液を細胞剥離液に置換する工程や、細胞剥離液で細胞をリンスする工程、剥離液を希釈したり均一化する工程などが挙げられる。
 振動付与工程、及びせん断力付与工程は一定の周期、又はランダムな周期で繰り返しても構わない。
 振動付与工程が実施される合計時間と、せん断力付与工程が実施される合計時間との比率は、0.01以上、100以下であることが好ましい。すなわち、振動付与工程が行われる時間をa、前記せん断力付与工程が行われる時間をbとしたとき、a及びbの関係が式5を満たすことが好ましい。

 振動付与工程とせん断力付与工程の開始の時刻差は、振動付与工程の開始が、せん断力付与工程の開始よりも先であれば、特に限定がない。細胞の生存率を高める観点からは、時間差は例えば30分以下、好ましくは10分以下、さらに好ましくは5分以下である。
 また、せん断力付与工程は、振動付与工程が終了する前に開始してもよいし、振動付与工程の終了と同時に開始してもよいし、振動付与工程の終了後に開始しても構わない。せん断力付与工程を振動付与工程が終了する前に開始することで、細胞の接着力を弱めながらせん断力を付与することができ、より効果的に細胞を剥離することができる。また、振動付与工程からせん断力付与工程に移行する判断をするために、培養基材に接着した細胞情報を測定し、その測定情報を利用してもよい。
 また、振動付与工程が開始される時刻と、せん断力付与工程が終了する時刻との差については、特に限定がないが、細胞の生存率を高める観点からは、例えば10秒以上、30分以下が好ましい。また、振動付与工程の終了後にせん断力付与工程が終了してもよいし、振動付与工程が終了する前にせん断力付与工程が終了してもよく、さらに振動付与工程とせん断力付与工程が同時に終了しても構わない。また、振動付与工程又はせん断力付与工程を終了する判断をするために、培養基材に接着した細胞情報を測定し、その測定情報を利用してもよい。
 振動付与工程が開始される際の細胞と培養基材との接着力をA1、せん断力付与工程が開始される際の細胞と培養基材との接着力をA2、振動付与工程、及びせん断力付与工程がいずれも終了した際における細胞と培養基材との接着力をA3とした際、A1、A2及びA3の関係が式1を満たすことが好ましい。

 B1とB2の関係がB2<0.5×B1(式2)を満たすことで、振動付与工程によってせん断力付与工程開始時に細胞の接着力が十分低下し、せん断力を付与しても生存率が高い状態で細胞が剥離しやすくなる。また、(式3)を満たすことで、細胞の剥離率が高くなる。
 また、振動付与工程の開始時刻に細胞の光学的な測定によって取得される輝度情報をC1、せん断力付与工程の開始時刻に前記細胞の光学的な測定によって取得される輝度情報をC2としたとき、C1とC2の関係が式4を満たすことが好ましい。

(情報取得工程)
 本開示の細胞の剥離方法は、さらに、細胞情報を取得する工程である、情報取得工程を有してもよい。そして、さらには、取得した細胞情報に基づいて、せん断力付与工程の開始タイミングを決定するせん断力付与工程開始時刻決定工程を有してもよい。
 本開示の細胞情報としては、細胞種、継代回数、培養時間のほかに、細胞の測定情報が挙げられる。
 本開示における測定情報の例としては、細胞の接着力や、細胞の面積、位相差顕微鏡画像(位相差画像とも呼ぶ)におけるハロ領域の面積、位相差画像における輝度値、斜入射照明により培養基材の細胞培養面を観察した際の輝度値などが挙げられる。
 細胞の接着力の測定情報の例としては、例えば、細胞培養面に対してせん断力が既知の水流などを噴射することで、せん断力を作用させて細胞を剥離し、細胞が剥離した領域の大きさから作用したせん断力を見積ることで、接着力とみなす方法が挙げられる。
 振動付与工程、及びせん断力付与工程での接着力の変化を評価することで、振動付与工程における超音波の強度や、せん断力付与工程の開始時刻、さらにせん断力付与工程内におけるせん断力の大きさや方向、さらには時間などの変化の制御を最適化することもできる。
 細胞の面積の測定情報の例としては、例えば、接着している細胞の位相差画像を取得し、画像処理によって、細胞一つの専有面積を計測することで、細胞の面積を数値化することを挙げられる。
 接着している細胞は位相差画像で見ると面積が大きく写り、振動付与工程及びせん断付与工程により接着力が低下している細胞は表面張力により球形に近い形をとるため、位相差画像で見ると細胞の面積が小さく写ることが知られている。これを利用して、せん断力付与工程の開始タイミングを判断してもよい。
 別の細胞情報として、位相差画像における輝度値あるいはハロ領域の面積が挙げられる。
 細胞が接着した状態の位相差画像においては全体的に小さい位相差(細胞が平たい状態)の領域しか検出されない。一方、剥離液が添加され、細胞の接着が弱くなり、細胞が培養基材から浮き上がると、浮き上がったところは位相差が大きくなり、位相差画像において、他に比べて画像輝度値が高くなるハロ領域がみられることが知られている。すなわち、細胞の接着が弱くなれば、ハロ領域の面積が増え、その値は一定に近づく。これを利用して、せん断力付与工程の開始タイミングを判断してもよいし、さらにせん断力付与工程内におけるせん断力の大きさや方向、さらには時間などの変化の制御を細胞情報の変化量に基づいて決定してもよい。
 また別の細胞情報として、斜入射照明により培養基材の細胞培養面を観察した際の輝度値が挙げられる。細胞剥離液の作用で接着力が低下した細胞は、仮足が縮み、培養基材との微視的な接着面が減少するため、培養基材と細胞との界面で光が散乱しやすくなる。よって、散乱光の比率が高くなる斜入射照明を用いて観察すると、細胞の接着状態が弱くなれば、培養基材の細胞培養面の輝度値が高くなり、その値は一定に近づく。これを利用して、振動付与工程の終了タイミングや、せん断力付与工程の開始タイミングを判断してもよいし、振動付与工程の超音波帯域の振動付与条件である振動強度や振動付与時間、さらにはせん断力付与工程のせん断力付与条件である、せん断力付与強度やせん断力付与時間、せん断力付与の変化の制御など、それぞれの条件のうち少なくとも一つを決定してもよい。
 本開示の細胞剥離方法は、振動付与工程、せん断力付与工程の条件(超音波の強度、超音波帯域の振動の付与時間、振動モード、またせん断力の種類や強度、付与時間など)を細胞種に応じて予め設定する制御部を備えてもよいし、細胞種と観察などの情報を組み合わせて制御してもよい。
(超音波帯域の振動)
 本開示において超音波帯域の振動として、10kHz以上1MHz以下の周波数をもつ振動を例示できる。振動発生手段は、細胞に超音波帯域の振動を付与することが可能な手段であれば、特に制限なく用いることができる。一例として、チタン酸ジルコン酸鉛(PZT)などの超音波発振子を振動体として用いる例を挙げることができる。
 ここで、培地が充填されて密閉された培養基材の外面に、振動体を直接当接させて、培養基材に振動を与えることができる。他には、振動付与手段を直接容器に接触させるのではなく、振動付与手段と処置対象領域との間に超音波伝達物質と記載されている振動伝達部材を介在させて、超音波を、剥離対象たる細胞に付与することもできる。
(振動体)
 本開示において、振動体は超音波帯域の振動を発生するものであれば、限定されることなく用いられるが、例えば圧電体と振動板とを接着したものが挙げられる。圧電体が円形型の場合は振動板はガラス、SUS、石英であることが好ましい。振動板をガラス、SUS、石英にすることで、振動体として破損することなく超音波帯域の比較的高い駆動周波数(振動周波数)で大きい振幅を出力することができる。振動体は超音波振動子を含むことができる。
 リング形状の圧電体の場合、振動板の外径寸法は圧電体の外径寸法と等しいことが好ましい。
 圧電体と振動板とが接着され撓み振動した際に、撓みの厚さ方向の中点すなわち引っ張り、圧縮のどちらにもならない中立面が振動板側にあるように、振動板の厚さが調整されていることが、圧電体の歪を効率良く撓みに利用するために好ましい。
 本開示の振動体には、市販のランジュバン型振動子や矩形型振動子を用いることもできる。ランジュバン型振動子は2つの金属ブロック間に圧電体を挟み込み、ボルト締めなどで締め付けて一体構造としているものが挙げられ、例えば本多電子製、富士セラミックス製などがある。
 本開示の振動体として、マイクロメートルオーダーで振幅制御が容易である、リング形状の圧電体をガラス板、又は金属板に接着したリングデバイスや、ランジュバン型振動子(ランジュバンデバイス)が好ましく用いられる。
 リングデバイスを定在波モードで使用する場合、圧電体や振動板のサイズや共振周波数にもよるが、中心部など、振動モードに応じた位置に最も強い振幅を有する振動を伝達させることができ、リングデバイスを進行波モード、搬送波モードで使用する場合、屈曲振動の腹や節の位置を周期的に移動させることが可能となり、培養基材に対してより均一な振動を付与することが可能となる。また、ランジュバンデバイスは培養基材に対して均一な振動を付与することが可能である。
(圧電体)
 本開示において振動体に用いられる圧電体はリング形状や円環形状の圧電体が好ましいが、振動体を撓ませる圧電体の形状は円板形状であっても構わない。情報取得工程において、細胞の情報を直接観察する場合は、圧電体は、培養基材下部の視野を確保できる円環形状が好ましい。
 圧電体は、分極処理を施すことで圧電特性を発揮するが、図3の通り、電極パターンに応じて分極の極性を変えて構わない。図3では、電極α1、α2、α3、β1、β2、β3がパターニングされた部分は変形に寄与する駆動相301であり、電極α1とβ1に挟まれた領域は変形度合いを検知するためのセンサ相302である。分極処理した圧電体は、入力した交流電圧の位相を電極パターン毎に制御することで、定在波モード、進行波モードの振動モードを励振することができる。プラスに分極した電極(α1、α3、β1、β3)に対して、マイナスに分極した電極(α2、β2)に180度の位相差を付けた交流電圧を印加することで定在波が発生する。これに対し、図3のように、一本鎖線の左右にA相303(電極α1、α2、α3)、B相304(電極β1、β2、β3)と分けたとき、A相とB相の位相差を90度にした交流電圧を印加すると、2相駆動の進行波が発生する。図3でGNDはグランドを指し、裏面に配置された電極の接地に用いる。
 電極パターンは、Agを印刷法で形成する例を挙げられるが、電極材料にはAgの他、Au、Pt、Pdなどの貴金属やCuなどの卑金属でも構わない。形成方法は印刷法、メッキ法、スパッタ法であっても構わない。
 振動体に用いた圧電体の材料組成はPbZrTiO(PZT)を用いたが、環境規制を考慮して実質的にPbを用いない非鉛圧電材料であることが好ましい。非鉛圧電材料には、主成分がBaTiO(BT)、NaNbO、BiNaTiO、BiFeOなどが挙げられ、それぞれの組み合わせや、主成分に金属元素が添加されていても構わない。特に、BaTiO(BT)系材料で、PbZrTiO(PZT)と同等の振動性能を確認した。また、セラミックス以外の材料として、単結晶材料、高分子系圧電材料でも構わない。
 圧電体と振動板とが接着され撓み振動した際に、撓みの厚さ方向の中点すなわち引っ張り、圧縮のどちらにもならない中立面が振動板側にあるように、振動板の厚さが調整されていることが、圧電体の歪を効率良く撓みに利用するために好ましい。圧電体と振動板の厚さの組み合わせは、圧電体の硬さと振動板の硬さに基づいて算出した。
 図4に培養基材に対して、圧電体を利用して、超音波帯域の振動を付与する例を示す。
 図4において、振動を付与する機構4102は、振動体4101と振動伝達部材413とカバー415からなり、振動体4101は振動板411と圧電体412からなっている。
 培養基材421は、振動伝達部材413の上に設置される。この例においては、培養基材421は、培養基材上蓋422で覆われ、その上に、重り414が置かれた例を示している。このように培養基材上蓋422で覆い、さらに、重り414を置くことで、超音波帯域の振動を効率的に付与することができる場合がある。
(振動源振幅評価)
 超音波帯域の振動付与部の振動源の振幅はレーザードップラー振動計(グラフテック社製、AT7200)で計測ることができる。計測手法は、所望の共振周波数、電圧で超音波帯域の振動付与部を駆動し、振動子表面をレーザーでXY方向の2次元的にスキャンすると、各点の振動速度が得られ、計算よりZ方向の振幅を算出することができる。この結果、振動子の振動波形を3次元的に計測することができ、駆動時の振動源の最大振幅を把握することができる。振動源の振幅とは、図5Aに示すような電極E1、E2をそれぞれ挟んで配置された2つの円環形状の圧電素子P1、P2をさらに2つの金属ブロックM1、M2で挟み込み、軸方向にボルト500で締結されたボルト締めランジュバン型振動子では、振幅はM1、M2の表面の振幅である。また、図5Bに示すような、圧電素子をガラス板や金属板などに接着剤などで張り付けた平板状の振動子(張り付け型振動子)などでは、振幅はガラス板501や金属板502の振幅である。
(細胞)
 本開示における細胞としては、インビトロで培養器材に接着培養可能なものであれば特に限定されるものではない。例えばチャイニーズハムスター卵巣由来CHO細胞やマウス結合組織L929細胞、マウス骨格筋筋芽細胞(C2C12細胞)、ヒト胎児肺由来正常二倍体線維芽細胞(TIG-3細胞)、ヒト胎児腎臓由来細胞(HEK293細胞)、ヒト肺胞基底上皮腺癌由来A549細胞やマウスマクロファージ様細胞(RAW264.7)やヒト子宮頸癌由来HeLa細胞などの種々の培養細胞株に加え、例えば生体内の各組織、臓器を構成する上皮細胞や内皮細胞、収縮性を示す骨格筋細胞、平滑筋細胞、心筋細胞、神経系を構成するニューロン細胞、グリア細胞、繊維芽細胞、生体の代謝に関与する肝実質細胞、肝非実質細胞や脂肪細胞、分化能を有する細胞として、誘導多能性幹(iPS)細胞、胚性幹(ES)細胞、胚性生殖(EG)細胞、胚性癌(EC)細胞、間葉系幹細胞、肝幹細胞、膵幹細胞、皮膚幹細胞、筋幹細胞、生殖幹細胞などの各種幹細胞、又は各組織の前駆細胞、さらにはそれらから分化誘導した細胞などが挙げられる。また本開示の細胞の剥離方法はシート状の細胞に適用してもよい。本開示の細胞の剥離方法は、これらの中でも細胞間の結合が強い細胞や、培養基材への接着力が高い細胞や、トリプシン感受性の高い細胞において好適である。細胞の大量培養のニーズから、例えば、タンパク質生産用に用いられるCHO細胞や、細胞治療で利用可能な間葉系幹細胞などに対して特に好適である。
(培養基材)
 本開示における培養基材は、細胞培養に用いられる培養基材のことを意味する。培養基材は、細胞接着性の培養基材であれば特に制限はなく、例えば、フラスコ、組織培養用フラスコ、ディッシュ、ペトリディッシュ、組織培養用ディッシュ、マルチディッシュ、マイクロプレート、マルチウェルプレート、マルチプレート、シャーレ、培養バッグ、ボトルなどが挙げられる。
 本開示における培養基材の材質は、化学的に安定であり、所望の細胞を培養可能な材質であればよく、例えば、ポリエチレン、ポリプロピレン、ポリカーボネート、ポリスチレン、ポリ塩化ビニル、ナイロン、ポリウレタン、ポリウレア、ポリ乳酸、ポリグリコール酸、ポリビニルアルコール、ポリ酢酸ビニル、ポリ(メタ)アクリル酸、ポリ(メタ)アクリル酸誘導体、ポリアクリロニトリル、ポリ(メタ)アクリルアミド、ポリ(メタ)アクリルアミド誘導体、ポリスルホン、セルロース、セルロース誘導体、ポリシリコーン、ポリメチルペンテン、ガラス、金属などが挙げられる。この中でもポリスチレンが好ましい。
(細胞剥離液)
 本開示において細胞剥離液は、本開示の細胞剥離方法で用いる、細胞を剥離させるための溶液である。
 本開示における細胞剥離液のpHは中性領域であることが好ましい。中性領域は細胞培養に適しており、細胞の生存率を安定的に高く保つことができるためである。pHは塩酸や水酸化ナトリウムなどで適宜調整可能である。また、pHを安定的に保つために各種緩衝液が好適に用いられる。
 本開示における細胞剥離液の粘度は1.80mPa・s以下であることが好ましい。これは、粘度がこの範囲であれば、超音波帯域の振動によって発生する剥離液の流動が妨げられず、剥離効率を高く保つことができるためである。細胞剥離液の粘度はポリマーや糖類の添加などによって適宜調整可能である。
 本開示における細胞剥離液は、タンパク質分解酵素を含んでもよいが、好ましくは細胞剥離液の全質量に対するタンパク質分解酵素の量は0.0005質量%以下であり、より好ましくは、タンパク質分解酵素を含まない。これは、タンパク質分解酵素は細胞の一部を分解するため剥離効率が高くできる半面、細胞の品質を低下させる可能性があるからである。
 本開示においてタンパク質分解酵素とは、例えば、細胞の一部を分解し、細胞を培養基材から剥離しやすくするものである。例えば、トリプシン、アキュターゼ、コラゲナーゼ、天然プロテアーゼ、キモトリプシン、エラスターゼ、パパイン、プロナーゼ、もしくはそれらの組み換え体などが挙げられる。
 本開示における細胞剥離液としては、金属イオンキレート剤(以後、キレート剤と呼ぶことがある)を含む溶液が、特に好ましい。キレート剤を含む細胞剥離液を用いることによって、細胞を超音波帯域の振動により効果的に剥離できるからである。
 本開示におけるキレート剤としては、特に限定はないが、例えば、エチレンジアミン四酢酸(以後、EDTAと呼ぶことがある)、エチレンジアミン、エチレンジアミンテトラメチレンホスホン酸、グリコールエーテルジアミン四酢酸、ニトリロトリ酢酸、ジエチレントリアミン五酢酸、イミノ二酢酸、ジヒドロキシエチルグリシン、ジカルボキシメチルグルタミン酸、エチレンジアミンジコハク酸、エチドロン酸、クエン酸、グルコン酸、ホスホノブタン三酢酸などを挙げることができる。この中でも二価の陽イオンとキレートを形成するキレート剤が好ましく、Ca2+及びMg2+とキレートを形成するキレート剤が特に好ましく、エチレンジアミン四酢酸が最も好ましい。キレート剤としてエチレンジアミン四酢酸を用いた場合、細胞剥離液のpHは7.0以上8.0以下であることが好ましい。これは細胞生存率を高く保つことができる中性領域の中でも高めのpHであることにより、エチレンジアミン四酢酸のキレート能を高めることができ、剥離効率をより高くすることが可能であるためである。なおキレート剤は単独で用いても、或いは2種類以上を複合して用いてもよい。
 キレート剤の含有量としては0.01mM以上5.0mM以下であることが好ましい。
 この範囲であることにより、キレート効果を確実に得ることができ、過剰なキレート剤の存在による活性低下も抑えることができる。
 本開示における細胞剥離液として、ポリアルキレングリコール構造を含む親水性ポリマーを含んでいてもよい。ポリアルキレングリコールは、超音波を用いた細胞剥離方法において、細胞の生存率を高くすることができる。ポリアルキレングリコール構造を含む親水性ポリマーの例として、ポリエチレングリコールが挙げられる。親水性ポリマーは、ゲルパーミエーションクロマトグラフィーにより測定されるピーク分子量Mpが800以上50000以下であることが好ましく、1200以上20000以下であることがさらに好ましい。これは、ポリマーの細胞への影響が少なく、かつ、ポリマーによる培地の増粘効果が抑制できるためである。
(緩衝液)
 本開示における緩衝液は、中性領域を保つことができれば制限なく使用できる。例えば、Tris-HCl緩衝液などのTris緩衝液、リン酸緩衝液、HEPES緩衝液、クエン酸-リン酸緩衝液、グリシルグリシン-水酸化ナトリウム緩衝液、Britton-Robinson緩衝液、GTA緩衝液などが挙げられる。この中でも、生体内の環境に近いリン酸緩衝液が好ましく、このリン酸緩衝液に、細胞内液と等張になるよう調整したリン酸緩衝生理食塩水(PBS)がより好適に用いられる。
(培地)
 培地の種類に特に制限はなく、例えば、ダルベッコ改変イーグル培地(Dulbecco’s Modified Eagle’s Medium;DMEM)、ハムF12培地(Ham’s Nutrient Mixture F12)、DMEM/F12培地、マッコイ5A培地(McCoy’s 5A medium)、イーグルMEM培地(Eagle’s Minimum Essential Medium;EMEM)、αMEM培地(alpha Modified Eagle’s Minimum Essential Medium;αMEM)、MEM培地(Minimum Essential Medium)、RPMI1640培地、イスコフ改変ダルベッコ培地(Iscove’s Modified Dulbecco’s Medium;IMDM)、MCDB131培地、ウィリアム培地E、IPL41培地、Fischer’s培地、StemSpan H3000(ステムセルテクノロジー社製)、StemSpanSFEM(ステムセルテクノロジー社製)、StemlineII(シグマアルドリッチ社製)、Endothelial Cell Growth Medium 2 Kit(プロモセル社製)、Mesenchymal Stem Cell Growth Medium 2(プロモセル社製)、MSCGM Bullet Kit(ロンザ社製)、mTeSR1或いは2培地(ステムセルテクノロジー社製)、リプロFF或いはリプロFF2(リプロセル社製)、NutriStem培地(バイオロジカルインダストリーズ社製)、MF-Medium間葉系幹細胞増殖培地(東洋紡株式会社製)などが挙げられる。
 これらの中から、それぞれの細胞の培養に適した培地を用いるのが好ましい。
(血清)
 上記の培地には、血清や抗生物質を添加してもよい。血清の例としては、牛胎児血清(Fetal Bovine Serum:FBS)、児牛血清、成牛血清、ウマ血清、ヒツジ血清、ヤギ血清、ブタ血清、ニワトリ血清、ウサギ血清、ヒト血清を挙げられるが、入手の容易さから一般的にFBSがよく用いられる。また、未処理又は未精製の血清をいずれも含まず、精製された血液由来成分又は動物組織由来成分(増殖因子など)を含有する無血清培地であってもよい。
(抗生物質)
 培地に添加される抗生物質の例としては、ペニシリン、ストレプトマイシン、アンピシリン、カルベニシリン、テトラサイクリン、ブレオマイシン、アクチノマイシン、カナマイシン、アクチノマイシンD、アンホテリシンBなどが挙げられる。
(細胞の培養条件)
 細胞の培養条件は、培養する細胞に従って適宜選択し得る。一般的には、培養基材内に適切な培地を添加し、そこに1.0×10~5.0×10cells/cm程度の細胞を播種し、37℃、CO濃度5%の環境下で培養する。この際、培養基材中の細胞占有面積率が7~8割程度、いわゆるサブコンフルエントの状態になるまで培養するのが好ましい。
(細胞剥離システム)
 本開示は、さらなる実施形態として、細胞剥離システムを提供するものである。
 本開示の細胞剥離システムは、培養基材の上で培養された接着細胞を剥離する剥離部、及び制御部を有する細胞剥離システムであって、前記剥離部は、前記培養基材に対して、超音波帯域の振動を与える振動付与部、及び前記培養基材の培養面に対して、せん断力を付与する、せん断力付与部を有し、前記制御部は、前記振動付与部の駆動の開始が、前記せん断力付与部の駆動の開始よりも先となるよう制御することを特徴とする細胞剥離システムである。
 また、本開示の細胞剥離システムは、前記せん断力付与部は、前記培養基材へ付与される叩打、振とう、揺動、偏心回転から選択される少なくとも一つの手段により、前記培養面に対してせん断力を付与することを特徴とする。
 本開示の細胞剥離システムの、前記制御部は、前記せん断力付与部を制御する、せん断力制御部を有し、該せん断力制御部は、前記せん断力の大きさを経時的に変化するように前記せん断力付与部を制御することを特徴とする。
 本開示の細胞剥離システムの前記制御部は、前記せん断力付与部を制御する、せん断力制御部を有し、該せん断力制御部は、前記せん断力の大きさを周期的に変化するように前記せん断力付与部を制御することを特徴とする。
 本開示の細胞剥離システムの制御部は、前記せん断力付与部を制御する、せん断力制御部を有し、該せん断力制御部は、前記細胞の大きさ、形状、接着面積、及び輝度値からなる群から選択される少なくとも1つに基づき付与するせん断力の方向を変化させるように前記せん断力付与部を制御することを特徴とする。
 なお、制御部は、一体となって、せん断力制御部を有してもよく、すなわち、振動付与部の駆動の開始が、せん断力付与部の駆動の開始よりも先となるよう制御し、かつ、せん断力付与部を制御してよい。あるいは、制御部は、せん断力制御部を、振動付与部の駆動の開始が、せん断力付与部の駆動の開始よりも先となるよう制御する部分と、せん断力制御部が分かれて存在してもよい。
 例えば、振動付与部の駆動の開始が、せん断力付与部の駆動の開始よりも先となるよう制御する情報処理装置とは別に、せん断力制御部が、せん断力付与部に備わった状態であってもよい。
 図6及び図7に本開示の細胞剥離システムの例を示す。
 図6に例示する細胞剥離システム61000においては、培養基材621の上で培養された接着細胞を剥離する剥離部61001、及び制御部61002を有する細胞剥離システムである。剥離部61001は、培養基材621に対して、超音波帯域の振動を与える振動付与部6102、及び前記培養基材の培養面に対して、せん断力を付与する、せん断力付与部6103を有し、制御部61002は、振動付与部6102の駆動の開始が、せん断力付与部6103の駆動の開始よりも先となるよう制御する。図6の例においては、剥離部61001を構成する、振動付与部6102は、振動体6101と振動伝達部材613とカバー615からなり、振動体6101は振動板611と圧電体612からなっている。また、せん断力付与部6103は、叩打部617と叩打部材616からなる。制御部61002は、振動付与部6102を制御する振動制御部619、せん断力付与部を制御する、せん断力制御部618と、主制御部620からなっている。なお、振動制御部619、せん断力制御部618と、主制御部620は一体となっていても、分かれていても構わない。図6では、さらに、培養基材621は、培養基材上蓋622で覆われ、その上に、重り614が置かれた例を示している。
 図7に例示する細胞剥離システム7001においては、この例においてはディッシュである、培養基材721の上で培養された接着細胞を剥離する剥離部71001、及び制御部71002、細胞情報取得部71003を有する細胞剥離システムである。
 剥離部71001は、保持部材722で保持した培養基材721に対して、超音波帯域の振動を与える振動付与部7102、及び培養基材の培養面に対して、せん断力を付与する、せん断力付与部7103を有する。この例において、せん断力付与部7103は、二軸振とう機構である。
 細胞情報取得部71003は、培養基材に接着した細胞に関する細胞情報を取得する機構を有する。細胞情報取得部71003によって取得された細胞情報は、制御部71002に処理されてもよく、細胞情報に基づいて制御部71002が剥離部71001を制御することも可能である。取得した細胞情報は、情報取得工程(ステップS220)の実行に用いられ、例えば、振動付与工程からせん断力付与工程に移行する判断、あるいは、振動付与工程又はせん断力付与工程を終了する判断のために利用することができる。
 また、この例においては、制御部71002は、演算及び記憶を行うコンピュータとしての機能を有する。
 制御部71002は、演算及び記憶を行うコンピュータとしての機能を実現するため、CPU(Central Processing Unit)706、RAM(Random Access Memory)707、ROM(Read Only Memory)708及びHDD(Hard Disk Drive)709を備える。また、制御部71002、通信I/F(インターフェース)710、表示装置711、及び入力装置712を備える。CPU706、RAM707、ROM708、HDD709、通信I/F710、表示装置711、及び入力装置712は、バス713を介して相互に接続される。なお、表示装置711及び入力装置712は、これらの装置を駆動するための不図示の駆動装置を介してバス713に接続されてもよい。
 図7では、制御部71002を構成する各部が一体の装置として図示されているが、これらの機能の一部は外付け装置により構成されていてもよい。例えば、表示装置711及び入力装置712は、CPU706などを含むコンピュータの機能を構成する部分とは別の外付け装置であってもよい。
 CPU706は、RAM707、HDD709などに記憶されたプログラムに従って所定の動作を行うとともに、制御部71002の各部を制御する機能をも有する。RAM707は、揮発性記憶媒体から構成され、CPU706の動作に必要な一時的なメモリ領域を提供する。ROM708は、不揮発性記憶媒体から構成され、情報処理システム705の動作に用いられるプログラムなどの必要な情報を記憶する。HDD709は、不揮発性記憶媒体から構成され、個別独立分離区画の数や位置に関する情報、蛍光強度などの記憶を行う記憶装置である。
 通信I/F710は、Wi-Fi(登録商標)、4Gなどの規格に基づく通信インターフェースであり、他の装置との通信を行うためのモジュールである。表示装置711は、液晶ディスプレイ、OLED(Organic Light Emitting Diode)ディスプレイなどであって、動画、静止画、文字などの表示に用いられる。入力装置712は、ボタン、タッチパネル、キーボード、ポインティングデバイスなどであって、利用者が情報処理システム705を操作するために用いられる。表示装置711及び入力装置712は、タッチパネルとして一体に形成されていてもよい。
 なお、図7に示されているハードウェア構成は例示であり、これら以外の装置が追加されていてもよく、一部の装置が設けられていなくてもよい。また、一部の装置が同様の機能を有する別の装置に置換されていてもよい。さらに、一部の機能がネットワークを介して他の装置により提供されてもよく、本実施形態を構成する機能が複数の装置に分散されて実現されるものであってもよい。例えば、HDD709は、フラッシュメモリなどの半導体素子を用いたSSD(Solid State Drive)に置換されていてもよく、クラウドストレージに置換されていてもよい。
 CPU706は、ROM708などに記憶されたプログラムをRAM707にロードして実行することにより、振動付与部の駆動の開始が、前記せん断力付与部の駆動の開始よりも先となるよう制御を実現するとともに、振動付与部及びせん断力付与部の制御を実現する。また、CPU706は、表示装置711を制御することにより表示部711の機能を実現する。また、CPU706は、HDD709を制御することにより記憶部の機能を実現する。
 以下、実施例、比較例をもって本開示をさらに詳細に説明するが、これは本発明を何ら限定するものではない。
 本開示の実施例で用いられる細胞はそれぞれ次に示す培養条件に従って得られた。
(CHO細胞の培養基材上の培養)
 CHO-K1(Chinese Hamster Ovary)細胞を培養基材の一例であるΦ60ポリスチレンディッシュ(コーニング社製)に10,000cells/cmの密度で播種し、37℃、CO 濃度5%の環境下で培養した。培地はHam’s F12(サーモフィッシャーサイエンティフィック製)にFetal Bovine Serum(シグマアルドリッチ製)を10%、ペニシリン-ストレプトマイシン(10000U/ml、サーモフィッシャーサイエンティフィック製)を1%添加したものを用いた。培養は48時間行ない、位相差顕微鏡で細胞の様子を観察し、細胞接着ならびに増殖を確認した。培養基材の細胞占有面積率は8割程度であった。
(A549細胞の培養基材上の培養)
 ヒト肺胞基底上皮腺癌細胞であるA549細胞を培養基材の一例であるΦ60ポリスチレンディッシュ(コーニング社製)に10,000cells/cmの密度で播種し、37℃、CO濃度5%の環境下で培養した。培地はDMEM(サーモフィッシャーサイエンティフィック社製)にFetal Bovine Serum(シグマアルドリッチ社製)を10%、ペニシリン-ストレプトマイシン(10000U/ml、サーモフィッシャーサイエンティフィック社製)を1%添加したものを用いた。培養は48時間行ない、位相差顕微鏡で細胞の様子を観察し、細胞接着ならびに増殖を確認した。培養基材の細胞占有面積率は8割程度であった。
(hMSC細胞の培養基材上の培養)
 ヒト間葉系幹細胞であるhMSC細胞を培養基材の一例であるΦ60ポリスチレンディッシュ(コーニング社製)に4,000cells/cmの密度で播種し、37℃、CO濃度5%の環境下で培養した。培地はMesenchymal Stem Cell Growth Medium 2(プロモセル社製)にFetal Bovine Serum(シグマアルドリッチ社製)を10%、ペニシリン-ストレプトマイシン(10000U/ml、サーモフィッシャーサイエンティフィック社製)を1%添加したものを用いた。48時間毎に培地を交換しながら培養を7日間行ない、位相差顕微鏡で細胞の様子を観察し、細胞接着ならびに増殖を確認した。培養基材の細胞占有面積率は8割程度であった。
(マウスマクロファージ様細胞の培養基材上の培養)
 マウスマクロファージ様細胞(RAW264.7)を培養基材の一例であるΦ60ポリスチレンディッシュ(コーニング社製)に20,000cells/cmの密度で播種し、37℃、CO濃度5%の環境下で培養した。培地はDMEM(サーモフィッシャーサイエンティフィック社製)にFetal Bovine Serum(シグマアルドリッチ社製)を10%、ペニシリン-ストレプトマイシン(10000U/ml、サーモフィッシャーサイエンティフィック社製)を1%添加したものを用いた。培養は48時間行ない、位相差顕微鏡で細胞の様子を観察し、細胞接着ならびに増殖を確認した。培養基材の細胞占有面積率は8割程度であった。
(剥離液への置換)
 培養基材内の培地を除去し、剥離液としてリン酸緩衝生理食塩水(PBS(-)、サーモフィッシャーサイエンティフィック社製)を、培養基材内に添加した。
(接着力評価方法)
 シリコンチューブ1701、チューブポンプ1702、リザーバータンク1703、三方弁1704、ニードル1705、ノズル1707を図8のように組み合わせて接着力評価装置1700を作製した。内径1.04mmのノズル1707の先端と培養面の距離が17mmとなるように細胞を培養した培養基材1706を配置し、ノズル1707から0.65mL/sで培養面に垂直に3秒間PBSを噴射してせん断力を付与した。噴射後に培養基材からPBSを除去した後、噴射により剥離した細胞の境界となる位置(剥離円の大きさ)を観察し、その位置で生じるせん断力をAnsys Fluentを用いて算出し、接着力とした。
(細胞接着面積評価方法)
 培養基材を位相差顕微鏡にセットして、倍率40倍において位相差像を観察した。位相差像の視野内の細胞の投影面積をそれぞれ求め、その平均値を細胞の接着面積とした。
(細胞の輝度情報評価方法)
 培養基材をステージ上にセットして、斜入射照明により観察されるディッシュ培養面の画像より平均の輝度値を算出し、細胞の輝度情報とした。
(各装置)
 振動付与、及びせん断力付与には、それぞれ、装置1から装置8の振動付与機構又は、振とう付与、叩打付与、揺動付与、ボルテックス撹拌、噴流付与のいずれかの、せん断力付与機構を用いた。
 それぞれに用いた装置1から9について、図9から図17に示す。
 装置1は図9に示されるように、振動付与機構を有する装置であり、培養基材10を保持する保持機構11と超音波帯域の振動付与機構12を有した。超音波帯域の振動付与機構12の詳細は表1に示す。
 装置2は図10に示されるように、せん断力付与機構を有する装置であり、培養基材10を保持する保持機構11と二軸振とう機構131を有した。
 装置3は図11に示されるように、振動付与機構とせん断力付与機構を有する装置であり、培養基材10を保持する保持機構11と超音波帯域の振動付与機構12と二軸振とう機構131を有した。超音波帯域の振動付与機構12の詳細は表1に示す。
 装置4は図12に示されるように、せん断力付与機構を有する装置であり、培養基材10を保持する保持機構11とせん断力付与するための、回転機構132と叩打機構133を有した。
 装置5は図13に示されるように、振動付与機構とせん断力付与機構を有する装置であり、培養基材10を保持する保持機構11と超音波帯域の振動付与機構12と、せん断力付与するための、回転機構132と叩打機構133を有した。超音波帯域の振動付与機構12の詳細は表1に示す。
 装置6は図14A及び図14Bに示されるように、せん断力付与機構を有する装置であり、培養基材10を保持する保持機構11と、揺動機構134を有した。
 装置7は、図15A及び図15Bに示されるように、せん断力付与機構を有する装置であり、培養基材10を保持する保持機構11と、せん断力付与するための、噴流機構135と、X-Y移動ステージ14と、Z移動ステージ15を有した。ステージ移動を利用し、図15Bに示すように、培養基材上に10mm間隔で、噴流を付与した。
 装置8は、図16に示されるように、せん断力付与機構を有する装置であり、培養基材10を保持する保持機構11と、せん断力付与するための、フロー機構136を有した。
(超音波帯域の振動付与方法)
 装置1(図9)、装置3(図11)、装置5(図13)の、それぞれの超音波帯域の振動付与機構には表1で示す超音波デバイスをそれぞれ使用し、超音波デバイス上に設置された保持機構に培養基材をセットした。その後、環境温度37℃において、表1に示すそれぞれの超音波デバイス及び振動付与条件で駆動させ、超音波帯域の振動を付与した。
 本実施例においてリングデバイスを定在波モードで使用する場合、使用した周波数帯域において、培養基材の中心部に最大振幅が作用するような振動モードの屈曲振動を付与することができ、リングデバイスを進行波モードで使用する場合、使用した周波数帯域において、培養基材の外径に対し、中心部から2/3の位置に最大振幅が作用するような振動モードの屈曲振動を付与することができる。さらに進行波モードとすることで、これらの屈曲振動が中心部を軸に周方向に回転するような振動を付与することができる。ランジュバンデバイスを使用する場合、培養基材の全域に対して中心部が最大振幅となるほぼ平坦な振動分布を有する振動を付与することができる。
 以下表1に、装置1、3、5の超音波帯域の振動付与機構で用いた条件である、振動付与条件1~8を示す。
(振とうによるせん断力付与方法)
 装置2(図10)及び装置3(図11)は、せん断力付与機構として、二軸振とう機構131を有した。装置2及び3は、それぞれ独立にx方向、y方向に動く自動ステージによって振とうによるせん断力付与が可能な二軸振とう機構上に培養基材を保持する保持機構、又は保持機構を設置済みの超音波帯域の振動付与機構を設置した。培養基材を保持機構にセットし、環境温度37℃において、二軸振とう機構を表2に示す駆動条件で動作させ、せん断力を付与した。
 以下表2に、装置2及び3の二軸振とう機構で用いた条件である、移動条件1~4を示す。
(叩打によるせん断力付与方法)
 装置4(図12)、又は装置5(図13)は、せん断力付与機構として、回転機構132と叩打機構133を有した。
 装置4及び装置5は、叩打によるせん断力付与が可能な叩打機構と培養基材の保持機構、及び保持機構を回転させることが可能な回転機構、さらに装置5は超音波帯域の振動付与機構とを組み合わせた装置だった。この装置を使用して、培養基材を保持機構にセットした後、環境温度37℃において、培養基材の外周部に表3に示すような、力と周波数、及び力の付与位置を変更して、せん断力を付与した。
 以下表3に、装置4及び5の回転機構の駆動条件である、駆動条件1~3を示す。
(その他のせん断力付与方法)
 その他のせん断力付与機構として、装置6は、図14A及び図14Bに示すように、揺動機構を有し、装置7は図15A及び図15Bに示すように、噴流機構135を有し、装置8は図16に示すようにフロー機構136を有した。
 せん断力の付与をそれぞれ以下の方法によって行った。
 揺動については、装置6により、図14Aに示すように中心を支点として円弧上に移動するステージを用いた揺動付与機構上に、培養基材保持機構を設置した装置を使用した。
 揺動付与時のステージの移動イメージを図14Bに示す。この装置を使用し、培養基材を保持機構にセットした後、環境温度37℃において、表4の動作条件1に示す速度、及び動作でせん断力を付与した。
 偏心回転については、ボルテックス撹拌を使用した。装置はボルテックスミキサーとして、ボルテックスジェニー2(SI-0286;サイエンティフィックインダストリーズ社製)のサンプル設置部を培養基材の保持が可能な形状に変更した培養基材の保持機構を使用した。保持機構に培養基材をセットして固定し、環境温度37℃において、表4の動作条件2に示す速度、及び動作でせん断力を付与した。速度はミキサー上の積載重量などによって変化するため、予め保持機構上に同一液量を入れた培養基材をセットしておき、高速度カメラで観察して設定速度となるようにミキサーの駆動条件を決定した。
 噴流については、装置7を用い、図15Aに示すように培養基材保持機構をX-Y方向に移動可能なX-Y移動ステージ上に設置し、さらに電動ピペッターを使用した噴流機構を設置したZ移動ステージを設置した装置を使用した。この装置を使用して、保持機構に培養基材をセットし、環境温度37℃において、図15Bに示す培養基材を上面視した図に対応した培養基材の各位置に対して、ピペット容量を1mLに設定して、表4の動作条件3に示す、速度、及び動作を行ってせん断力を付与した。各位置の動作は3秒間で吸引と吐出を2回ずつ繰返し、1秒かけて次の位置に移動し、さらに同じ動作を繰り返した。
 例えば、図15Bに示す視野には、上記に対応する位置が、全部で21存在する。これらのすべてに噴流を付与すると、せん断力付与工程が行われる時間は83秒となる。
 フローについては、装置8を用い、図16に示すように培養基材に設置可能なフロー機構136を使用し、フロー速度を設定可能なポンプと接続して液体をフローさせる装置を使用した。この装置を使用して、保持機構に培養基材をセットし、環境温度37℃において、表4の動作条件4に示す、速度、及び動作を行ってせん断力を付与した。
 以下表4に、動作条件1(装置6の揺動)、動作条件2(ボルテックスによる偏心回転)、動作条件3(装置7の噴流)、動作条件4(装置8によるフロー)のそれぞれの動作を示す。
(剥離方法)
 剥離方法1~33は、上記の装置1~8を用いて、それぞれ以下の表5で示す条件で、工程[1]及び工程[2]を設定した。工程[1]は工程[2]より先に開始される。それぞれの剥離方法の詳細については、以下に述べる。
[剥離方法1、8~12、15~17、20~27]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、超音波帯域の振動付与装置すなわち装置1の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5の条件、付与時間でディッシュに超音波帯域の振動を付与した。その後、工程[2]の剥離操作のため、表5に示すそれぞれのせん断力付与装置の機構における保持機構上にディッシュをセットする際に揺らし、工程[1]の剥離操作では細胞剥離が完了していないことを確認した。
 次に、工程[2]の剥離操作として、表5に示すそれぞれのせん断力付与装置の機構上において、超音波帯域の振動付与が終了して30秒後から、表5に示す各条件でせん断力を付与した。
[剥離方法2、5~7、13、18、19]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、超音波帯域の振動付与装置とせん断力付与装置が一体化した、装置3又は装置5の、それぞれの装置の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5の条件、付与時間でディッシュに超音波帯域の振動の付与を開始した。
 次いで、工程[2]開始前に目視で工程[1]の剥離操作では細胞剥離が完了していないことを確認してから、工程[2]の剥離操作として、超音波帯域の振動付与が終了する30秒前より、表5に示すそれぞれのせん断力付与装置を、それぞれの条件で駆動させてせん断力を付与した。
[剥離方法3]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、超音波帯域の振動付与装置とせん断力付与装置が一体化した装置3の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5の条件、付与時間でディッシュに超音波帯域の振動の付与を開始した。本方法では工程[1]の振動付与終了後、直ちに工程[2]の剥離操作を、表5に示す条件で駆動させてせん断力を付与した。工程[2]の剥離操作終了後は工程[1]の剥離操作を行い、これらを合計6回繰り返した。
[剥離方法4]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、超音波帯域の振動付与装置とせん断力付与装置が一体化した装置3の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5の条件、付与時間でディッシュに超音波帯域の振動の付与を開始した。本剥離方法は工程[1]の振動付与をし続けながら工程[2]の剥離操作を繰り返す剥離方法である。工程[1]の剥離操作開始20秒後から、工程[2]の剥離操作を、表5に示す条件で駆動させてせん断力を付与した。
[剥離方法14]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、工程[1]の前操作として、図14A及び図14Bの装置6の揺動機構を使用して、室温である25℃環境下で30秒間揺動させて剥離液を良くなじませた。次いで、表5に示す超音波帯域の振動付与装置、すなわち装置1の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5の条件、付与時間でディッシュに超音波帯域の振動を付与した。その後、工程[2]の剥離操作のため、表5に示すせん断力付与装置の機構における保持機構上にディッシュをセットする際に揺らし、工程[1]の剥離操作では細胞剥離が完了していないことを確認した。
 次に、工程[2]の剥離操作として、表5に示すせん断力付与装置の機構上において、超音波帯域の振動付与が終了して30秒後から、表5に示す各条件でせん断力を付与した。
[剥離方法28]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、工程[1]の前操作として、37℃インキュベーター内に2分間静置させて剥離液を良くなじませた。次いで、表5に示す超音波帯域の振動付与装置、すなわち装置1の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5の条件、付与時間でディッシュに超音波帯域の振動を付与した。その後、工程[2]の剥離操作のため、表5に示すせん断力付与装置、すなわち装置2の保持機構上にディッシュをセットする際に揺らし、工程[1]の剥離操作では細胞剥離が完了していないことを確認した。
 次に、工程[2]の剥離操作として、表5に示すせん断力付与装置すなわち装置2の機構上において、超音波帯域の振動付与が終了して30秒後から、表5に示す各条件でせん断力を付与した。
[剥離方法29]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、せん断力付与装置である装置2の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5の条件、付与時間でディッシュにせん断力を付与した。その後、工程[2]の剥離操作のため、表5に示す超音波帯域の振動付与装置すなわち、装置1の保持機構上にディッシュをセットする際に揺らし、工程[1]の剥離操作で一部の細胞が剥離していることを確認した。
 次に、工程[2]の剥離操作として、表5に示す超音波帯域の振動付与機構、すなわち装置1上において、せん断力付与が終了して30秒後から、表5に示す各条件で超音波帯域の振動を付与した。
[剥離方法30、31]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、それぞれのせん断力付与装置、すなわち、装置2又は装置4の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5のそれぞれの条件、付与時間でディッシュにせん断力を付与した。工程[2]は行わなかった。
[剥離方法32、33]
 細胞を培養したディッシュ内の培養液を剥離液に置換し、次いで表5に示すように、超音波帯域の振動付与装置、すなわち装置1の保持機構上にディッシュをセットし、工程[1]の剥離操作として、表5のそれぞれの条件、付与時間でディッシュに超音波帯域の振動を付与した。工程[2]は行わなかった。
[細胞情報取得方法1]
 細胞の接着力評価は次の方法で行った。同一条件で細胞を培養したディッシュを細胞情報取得用に別に3枚用意し、ディッシュ内の培養液を剥離液に置換した。そのうち1枚を前述の接着力評価方法の通り図8で示す接着力評価装置1700を使用して、前述の条件でPBSを噴射してせん断力を付与した。噴射後に培養基材からPBSを除去した後、噴射により剥離した細胞の境界となる位置を観察し、その位置で生じるせん断力をAnsys Fluentを用いて算出することで、接着力A1とした。接着力評価装置で噴射することでディッシュ培養面の細胞は噴射位置を中心におよそ円形に剥離する。この際に生じる、剥離した細胞の境界となる位置は円形の境界となることから、剥離円とも呼ぶことができる。本開示の実施例においては得られた剥離円の上下方向の直径の平均をとり、せん断力の算出を行った。次に、残る2枚のディッシュについて、それぞれ前述の剥離方法に従って工程[1]の剥離操作を行った。工程[2]を開始する直前に、そのうち1枚のディッシュについて接着力評価装置を使用して前述と同様に接着力を評価した。この時の接着力をA2とした。
 残る1枚のディッシュについて、工程[2]の剥離操作を行った後、接着力評価装置を使用して前述と同様に接着力を評価した。この時の接着力をA3とした。
 本実施例における接着力は前述の条件において、1~80Paの範囲で評価可能である。接着力測定時にPBSを噴射した後、噴射により生じた剥離円の境界が検出されなかった場合や、剥離円の径が小さいことで評価した接着力が80Pa以上の数値となる場合には接着力を80Pa以上とみなし、剥離円の径が大きくなり、接着力が1Pa未満の値として算出された場合は一律に1Pa未満とした。また、1Pa未満となった場合の接着力の大小関係については、噴射により生じた剥離円の直径が大きいほど接着力が小さいと判断した。
 得られた接着力A1、A2及びA3より、A1>A2>A3の関係を満たすかどうかを確認した。
[細胞情報取得方法2]
 細胞の接着面積評価は次の方法で行った。細胞を培養したディッシュ内の培養液を剥離液に置換した後、前述の細胞接着面積評価方法の通り位相差顕微鏡(Axio Obserer 7;カールツァイス社製)にセットして位相差観察を行い、倍率40倍の視野内の細胞の投影面積の総和から平均値を算出し、細胞の接着面積B1とした。次いで、前述の剥離方法に従って工程[1]の剥離操作を行い、工程[2]を開始する直前に位相差顕微鏡によって位相差観察を行い、前述と同様に細胞の投影面積を算出した。この値を細胞の接着面積B2とした。次に、工程[2]の剥離操作を行った後、位相差顕微鏡によって位相差観察を行い、前述と同様に細胞の投影面積を算出した。この際に輪郭がぼやけている細胞については接着しておらず浮遊しているものとみなし接着面積を0とみなして平均を取った。この値を細胞の接着面積B3とした。
 得られた細胞の接着面積B1、B2及びB3より、B2<0.5×B1及び、0≦B3<B2の関係を満たすかどうかを確認した。
[細胞情報取得方法3]
 細胞の輝度情報評価は次の方法で行った。細胞を培養したディッシュ内の培養液を剥離液に置換した後、前述の細胞の輝度情報評価方法の通り、光学的な測定を行った。すなわち、ディッシュをステージ上にセットして、斜入射照明をディッシュ上面に、カメラをディッシュ下面にセットしてディッシュの培養面の画像を取得した。このよう得られた画像より培養面の平均の輝度値を算出し、細胞の輝度情報C1とした。次いで、前述の剥離方法に従って工程[1]の剥離操作を行い、工程[2]を開始する直前に前記と同様にディッシュをステージ上にセットして、斜入射照明により観察されるディッシュ培養面の画像から平均の輝度値を算出した。この値を細胞の輝度情報C2とした。
 得られた細胞の輝度情報C1及びC2より、C1<C2の関係を満たすかどうかを確認した。
[実施例1]
 剥離方法1を用いて、ディッシュ上のCHO細胞を剥離させた。剥離した細胞は回収した後、血球計算盤を用いて細胞数の測定及びトリパンブルー染色による生死判別法を用いて生存率を算出した。また、超音波剥離後のディッシュに対して、セルスクレーパーを用いて超音波で剥離できなかった細胞も全て剥離し、血球計算盤で細胞数を測定した。超音波で剥離した細胞数とその後セルスクレーパーで剥離した細胞数を合わせて総剥離細胞数とし、総剥離細胞数に対する超音波剥離細胞数の割合を剥離率と定義し、算出した。結果、生存率は96.0%、剥離率は98.0%であった。なお、生存率、剥離率ともに90%以上を良好と判断した。使用した細胞種、剥離方法、算出した生存率、及び剥離率は表6に示す。
 また、実施例1において前述の細胞情報のうち、接着力、接着面積、輝度情報を得た。
 接着力については前述の細胞情報取得方法1に示す方法に従って取得した。接着力A1はディッシュ内の培養液を剥離液に置換した後に前述の接着力評価装置を用いて評価し、噴射によって細胞が剥離した剥離円が見られなかったためA1>80Paとした。接着力A2は工程[2]を開始する直前に評価し、噴射による剥離円の直径が大きいためA2<1Paとした。接着力A3は工程[2]終了後に評価し、前述と同様に剥離円の直径が大きいためA3<1Paとした。A2及びA3評価時における剥離円の直径の大きさはA3>A2であったため、接着力についてはA2>A3の関係となると判断した。以上より、実施例1における接着力の関係はA1>A2>A3の関係を満たすことを確認した。
 細胞の接着面積については前述の細胞情報取得方法2に示す方法に従って視野内の平均面積を取得した。接着面積B1はディッシュ内の培養液を剥離液に置換した後に評価し、B1=528μmであった。接着面積B2は工程[2]を開始する直前に評価し、192μmであった。同様に、接着面積B3は工程[2]終了後に評価し、大半の細胞が剥離していることを考慮し、平均をとると2μmとなった。これらの値はB2<0.5×B1及び、0≦B3<B2の関係を満たすことを確認した。
 細胞の輝度情報については前述の細胞情報取得方法3に示す方法に従って取得した。輝度情報C1はディッシュ内の培養液を剥離液に置換した後に評価し、C1=78.5であった。輝度情報C2は工程[2]を開始する直前に評価し、108.1であった。これらの値はC1<C2の関係を満たすことを確認した。
[実施例2~34、比較例1~6]
 実施例2~34、比較例1~6では、細胞種と剥離方法の組み合わせを変更し、剥離後の操作は実施例1と同様に評価を行った。結果を表6に示す。
 また、本開示の実施例のうち代表的な実施例について、それぞれで得られた細胞情報の結果を表7に示す。これらの結果より、せん断力を付与する工程に進むことを判断する細胞情報の閾値を決定することができる。例えば、細胞情報のうち、簡便な光学系で取得可能な輝度情報の閾値については、表7の輝度情報のC2の欄のうち最小となる数値を参考に閾値を決定することができ、本実施例においては、細胞種がCHO細胞の場合は輝度情報の閾値を105と設定することができる。設定した閾値により、経時的に輝度情報の数値をモニターすることで、せん断力を付与する工程に移るタイミングを判断することが可能になる。輝度情報の閾値は、細胞種や培養条件に応じて異なることがあるため、実験データを取得して、予め閾値を設定することができる。また、輝度情報の時間当たりの変化量を閾値として設定することもできる。また、輝度情報の時間変化をもとに、超音波の強度を決定することもできる。
 本開示によれば、細胞の生存率と剥離率がともに高くなるような、細胞剥離方法及びそれを用いた細胞剥離システムを提供することができる。
 本発明は上記実施の形態に制限されるものではなく、本発明の精神及び範囲から離脱することなく、様々な変更及び変形が可能である。従って、本発明の範囲を公にするために以下の請求項を添付する。

Claims (25)

  1.  培養基材の上で培養された接着細胞の剥離方法であって、
     前記培養基材に対して、超音波帯域の振動を付与する振動付与工程、及び
     前記培養基材の培養面に対して、せん断力を付与するせん断力付与工程
    を含み
     前記振動付与工程の開始が、前記せん断力付与工程の開始よりも先であり、
     前記せん断力は超音波の振動以外の手段によって付与されることを特徴とする細胞の剥離方法。
  2.  前記せん断力は前記培養基材へ付与される叩打、振とう、揺動、及び偏心回転、並びに前記培養基材への液体の吐出からなる群から選択される少なくとも一つにより付与されることを特徴とする請求項1に記載の細胞剥離方法。
  3.  前記振動付与工程において付与される前記超音波帯域の振動は、最大振幅が10μm以下の振動源によってもたらされることを特徴とする請求項1又は2に記載の細胞剥離方法。
  4.  前記振動付与工程の開始時刻における前記細胞と前記培養基材との接着力をA1
     前記せん断力付与工程の開始時刻における前記細胞と前記培養基材との接着力をA2

  9.  前記せん断力付与工程において、付与されるせん断力の方向が経時的に変化することを特徴とする請求項1から6のいずれか1項に記載の細胞剥離方法。
  10.  前記せん断力付与工程は、互いに方向の異なる複数のせん断力を付与する工程を含む請求項1から9のいずれか1項に記載の細胞剥離方法。
  11.  前記せん断力付与工程において、細胞の情報に基づいて、付与するせん断力の方向を決定することを特徴とする請求項1から10のいずれか1項に記載の細胞剥離方法。
  12.  前記培養基材に接着した状態の細胞の情報を取得する細胞情報取得工程、及び
     前記細胞の情報に少なくとも基づいて、前記せん断力付与工程の開始時刻を決定する、せん断力付与工程開始時刻決定工程を含み、
     前記細胞情報取得工程及び前記せん断力付与工程開始時刻決定工程は、いずれも、前記振動付与工程の開始より先に実行されることを特徴とする請求項1から11のいずれか1項に記載の細胞剥離方法。
  13.  前記細胞の情報は、前記細胞の大きさ、形状、接着面積、及び輝度値で構成される群から選択される少なくとも1つを含むことを特徴とする請求項1から12のいずれか1項に記載の細胞剥離方法。
  14.  前記せん断力付与工程において、
     前記細胞の情報に応じて前記せん断力を付与する条件を決定することを特徴とする請求項12に記載の細胞剥離方法。
  15.  前記振動付与工程が行われる時間は1秒以上、1時間以下であることを特徴とする請求項1から14のいずれか1項に記載の細胞剥離方法。
  16.  前記せん断力付与工程が行われる時間は1秒以上1時間以下であることを特徴とする請求項1から15のいずれか1項に記載の細胞剥離方法。
  17.  前記振動付与工程が行われる時間をa、前記せん断力付与工程が行われる時間をbとしたとき、a及びbの関係が式5を満たすことを特徴とする請求項1から16のいずれか1項に記載の細胞剥離方法。
     0.01≦a/b≦100(式5)
  18.  前記振動付与工程が開始される時刻と、前記せん断力付与工程が開始される時刻との差は1秒以上1時間以下であることを特徴とする請求項1から17のいずれか1項に記載の細胞剥離方法。
  19.  前記振動付与工程が開始される時刻と、前記せん断力付与工程が終了する時刻との差は10秒以上30分以下であることを特徴とする請求項1から18のいずれか1項に記載の細胞剥離方法。
  20.  培養基材の上で培養された接着細胞を剥離する剥離部、及び制御部を有する細胞剥離システムであって、
     前記剥離部は、前記培養基材に対して、超音波帯域の振動を与える振動付与部、及び前記培養基材の培養面に対して、せん断力を付与する、せん断力付与部を有し、
     前記制御部は、前記振動付与部の駆動の開始が、前記せん断力付与部の駆動の開始よりも先となるよう制御することを特徴とする細胞剥離システム。
  21.  前記せん断力付与部は、前記培養基材へ付与される叩打、振とう、揺動、偏心回転から選択される少なくとも一つの手段により、前記培養面に対してせん断力を付与することを特徴とする請求項20に記載の細胞剥離システム。
  22.  前記制御部は、前記せん断力付与部を制御する、せん断力制御部を有し、該せん断力制御部は、前記せん断力の大きさを経時的に変化するように前記せん断力付与部を制御することを特徴とする請求項20又は21に記載の細胞剥離システム。
  23.  前記制御部は、前記せん断力付与部を制御する、せん断力制御部を有し、該せん断力制御部は、前記せん断力の大きさを周期的に変化するように前記せん断力付与部を制御することを特徴とする請求項20から22のいずれか1項に記載の細胞剥離システム。
  24.  前記制御部は、前記せん断力付与部を制御する、せん断力制御部を有し、該せん断力制御部は、前記せん断力の方向を経時的に変化するように前記せん断力付与部を制御することを特徴とする請求項20から23のいずれか1項に記載の細胞剥離システム。
  25.  前記制御部は、前記せん断力付与部を制御する、せん断力制御部を有し、該せん断力制御部は、前記細胞の大きさ、形状、接着面積、及び輝度値からなる群から選択される少なくとも1つに基づき付与するせん断力の方向を変化させるように前記せん断力付与部を制御することを特徴とする請求項20から24のいずれか1項に記載の細胞剥離システム。
