Abstract

エルゴチオネイン等の化合物を用いて、非生物的ストレス耐性の向上効果をさらに高めることができる、植物の非生物的ストレス耐性向上剤を提供する。本発明の植物の非生物的ストレス耐性向上剤は、下記式(I)で示される化合物もしくはその互変異性体、またはそれらの農学的に許容可能な塩である第1の成分と、アミノ酸、2以上10以下のアミノ酸長であるペプチド、ベタイン、有機酸またはその塩、核酸塩基、ビタミン、および、糖または糖アルコールからなる群から選択される少なくとも1である第2の成分と、を含む。(式(I)中、RおよびRは、独立して水素原子または炭素数1以上4以下のアルキル基を表し、R、RおよびRは、独立して炭素数1以上4以下のアルキル基を表す。)

Description

植物の非生物的ストレス耐性向上剤、非生物的ストレス耐性向上方法、非生物的ストレス耐性向上用製剤、非生物的ストレス耐性向上用製品、および植物
 本発明は、植物の非生物的ストレス耐性向上剤、非生物的ストレス耐性向上方法、非生物的ストレス耐性向上用製剤、非生物的ストレス耐性向上用製品、および植物に関する。
 薬剤の付与により、非生物的ストレスを緩和させる方法や、植物の成長を促進し、収穫量を高める方法が検討されている。たとえば、特許文献1には、グリシンベタインが、植物に対する灌水不足によるストレスを緩和させる作用を有すると記載されている。
  また、特許文献2には、発芽した植物体にエルゴチオネイン水溶液を付与すると、草丈が高くなり、花および実の数が増え、種子の収穫量も高くなったと記載されている。

 非生物的ストレスは、植物が潜在的に有する生産性の発現を大きく阻害していると考えられている。そのため、非生物的ストレスを取り除くバイオスティミュラントを付与することで、植物の生産量は飛躍的に増大すると期待される。
 特許文献2に記載のエルゴチオネイン等の化合物は、生産量、草丈、種子、花実および果実数の増加などの効果を有する。そのため、これらの化合物は、植物に対する有望な第1の成分であると考えられる。一方で、これらの化合物による非生物的ストレス耐性の向上効果をさらに高めて、非生物的ストレスによる生産量低下のさらなる改善効果を得る方法を見出だすことも求められている。
 本発明は上記問題に鑑みてなされたものであり、エルゴチオネイン等の化合物を用いて、非生物的ストレス耐性の向上効果をさらに高めることができる、植物の非生物的ストレス耐性向上剤、これを用いる非生物的ストレス耐性向上方法、これを含む非生物的ストレス耐性向上用製剤および製品、ならびにこれにより処理された植物を提供することを、その目的とする。
 上記課題を解決するための本発明の一実施形態は、下記[1]~[9]の植物の非生物的ストレス耐性向上剤、非生物的ストレス耐性向上方法、非生物的ストレス耐性向上用製剤、非生物的ストレス耐性向上用製品、および植物に関する。

 本発明によれば、エルゴチオネイン等の化合物を用いて、非生物的ストレス耐性の向上効果をさらに高めることができる、植物の非生物的ストレス耐性向上剤、これを用いる非生物的ストレス耐性向上方法、これを含む非生物的ストレス耐性向上用製剤および製品、ならびにこれにより処理された植物が提供される。
 本発明の一実施形態は、植物の種子を、下記式(I)で示される化合物もしくはその互変異性体、またはそれらの農学的に許容可能な塩である第1の成分と、アミノ酸、2以上10以下のアミノ酸長であるペプチド、ベタイン、有機酸またはその塩、核酸塩基、ビタミン、および、糖または糖アルコールからなる群から選択される少なくとも1つである第2の成分と、を含む、植物の非生物的ストレス耐性向上剤に関する。
 [第1の成分]
 第1の成分は、下記式(I)で示される化合物もしくはその互変異性体、またはそれらの農学的に許容可能な塩である。非生物的ストレス耐性向上剤は、第1の成分を1種のみ含んでもよいし、複数種含んでもよい。

 式(I)中、RおよびRは、独立して水素原子または炭素数1以上4以下のアルキル基を表し、R、RおよびRは、独立して炭素数1以上4以下のアルキル基を表す。
 R~Rが取り得るアルキル基は、直鎖状であっても分枝状であってもよい。R~Rが取り得るアルキル基の例には、メチル基、エチル基、プロピル基、イソプロピル基、ブチル基、イソブチル基、sec-ブチル基およびtert-ブチル基が含まれる。
 RおよびRは、少なくとも一方が水素原子であることが好ましく、いずれも水素原子であることがより好ましい。RおよびRは、アルキル基である場合には、メチル基、エチル基またはプロピル基であることが好ましく、メチル基またはエチル基であることがより好ましく、メチル基であることがさらに好ましい。
 R、RおよびRは、独立してメチル基、エチル基、またはプロピル基であることが好ましく、メチル基またはエチル基であることがより好ましく、メチル基であることがさらに好ましい。R、RおよびRは、少なくとも1つがメチル基であることが好ましく、少なくとも2つがメチル基であることがより好ましく、全てがメチル基であることがさらに好ましい。
 式(I)で示される化合物は、RおよびRのうち少なくとも一方が水素原子であるとき、互変異性体を有する。より具体的には、式(I)で示される化合物は、Rが水素原子であるとき、互変異性体である下記式(II)で示される化合物を有する。また、式(I)で示される化合物は、Rが水素原子であるとき、互変異性体である下記式(III)で示される化合物を有する。なお、本明細書において単に「互変異性体」というときは、式(II)で示される化合物および式(III)で示される化合物の両方を意味する。

 式(II)および(III)中、R~Rは、式(I)におけるR~Rと同一である。
 通常、溶液中では、式(I)で示される化合物と式(II)で示される化合物または式(III)で示される化合物とは平衡状態で存在し得る。式(I)で示される化合物と式(II)で示される化合物または式(III)で示される化合物との比率は、溶媒、温度またはpHなどにより変動し得る。
 式(I)で示される化合物またはその互変異性体は、エルゴチオネインであることが好ましく、L-(+)-エルゴチオネインであることがより好ましい。
 これら化合物は、市販のものを用いてもよく、当業者に周知の技術、たとえば特表2013-506706号公報および特開2006-160748号公報などに記載の手法により合成したものを用いてもよい。また、エルゴチオネインは、細菌および真菌によって産生されることが知られている。このような微生物を用いる産出方法の例には、特開2012-105618号公報、特開2014-223051号公報、国際公開第2016/104437号、国際公開第2016/121285号、国際公開第2015/168112号および国際公開第2017/150304号などに記載の方法が含まれる。エルゴチオネインは、これらの微生物から得られたエルゴチオネインを含む培養物として用いてもよいし、上記培養物を濃縮または精製して得られたエルゴチオネインを用いてもよい。
 「農学的に許容可能」とは、安全であり、非毒性であり、かつ生物学的にも他の意味でも所望されないものではなく、農薬用として、特に植物の非生物的ストレス耐性を向上させる農薬用として、許容されるものを意味する。
 式(I)で示される化合物またはその互変異性体の「農学的に許容可能な塩」とは、上記で定義されたような農学的に許容される塩であって、式(I)で示される化合物またはその互変異性体の作用および効果が得られるものを意味する。このような塩の例には、水和物、溶媒和物、酸付加塩、式(I)で示される化合物またはその互変異性体に存在する酸性プロトン(proton acide)が金属イオンにより置換されて形成される塩、および上記酸性プロトンが有機塩基もしくは無機塩基と配位して形成される塩などが含まれる。
 酸付加塩は、無機酸と形成されるものであってもよいし、有機酸と形成されるものであってもよい。無機酸の例には、塩酸、臭化水素酸、硫酸、硝酸、およびリン酸などが含まれる。有機酸の例には、酢酸、ベンゼンスルホン酸、安息香酸、カンファースルホン酸、クエン酸、エタンスルホン酸、フマル酸、グルコヘプトン酸、グルコン酸、グルタミン酸、グリコール酸、ヒドロキシナフトエ酸、2-ヒドロキシエタンスルホン酸、乳酸、マレイン酸、リンゴ酸、マンデル酸、メタンスルホン酸、ムコン酸、2-ナフタレンスルホン酸、プロピオン酸、サリチル酸、コハク酸、ジベンゾイル-L-酒石酸、酒石酸、p-トルエンスルホン酸、トリメチル酢酸、およびトリフルオロ酢酸などが含まれる。
 式(I)で示される化合物またはその互変異性体に存在する酸性プロトンと置換し得る金属イオンの例には、アルカリ金属イオン、アルカリ土類金属イオンおよびアルミニウムイオンなどが含まれる。
 式(I)で示される化合物またはその互変異性体に存在する酸性プロトンと配位し得る有機塩基の例には、ジエタノールアミン、エタノールアミン、N-メチルグルカミン、トリエタノールアミン、およびトロメタミンなどが含まれる。式(I)で示される化合物またはその互変異性体に存在する酸性プロトンと配位し得る無機塩基の例には、水酸化アルミニウム、水酸化カルシウム、水酸化カリウム、炭酸ナトリウムおよび水酸化ナトリウムなどが含まれる。
 上記第1の成分は、植物の生長を促進したり、非生物的ストレスを緩和したりする、いわゆるバイオスティミュラント(非生物的ストレス耐性向上剤)として作用すると考えられる。
 [第2の成分]
 第2の成分は、アミノ酸、2以上10以下のアミノ酸長であるペプチド、ベタイン、有機酸またはその塩、核酸塩基、ビタミン、および、糖または糖アルコールからなる群から選択される少なくとも1つの成分である。これらの第2の成分は、第1の成分との相乗効果により、植物の非生物的ストレス耐性の向上効果を顕著に高める。非生物的ストレス耐性向上剤は、第2の成分を1種のみ含んでもよいし、複数種含んでもよい。
 アミノ酸は、酸性アミノ酸、中性アミノ酸および塩基性アミノ酸のいずれであってもよい。また、アミノ酸は、極性アミノ酸であってもよいし、無極性アミノ酸であってもよい。また、アミノ酸は硫黄を含んでいてもよい。
 アミノ酸の例には、グルタミン酸、アスパラギン酸、ヒスチジン、アルギニン、リシン、グリシン、アラニン、バリン、ロイシン、イソロイシン、スレオニン、フェニルアラニン、チロシン、トリプトファン、プロリン、セリン、グルタミン、アスパラギン、5-アミノレブリン酸、γ-アミノ酪酸、システイン、メチオニン、およびオルニチンなどが含まれる。これらのうち、グルタミン酸およびアルギニンが好ましい。非生物的ストレス耐性向上剤は、アミノ酸を1種のみ含んでもよいし、複数種含んでもよい。
 ペプチドは、2以上10以下のアミノ酸長を有するペプチドであり、2以上6以下のアミノ酸長を有するペプチドであることが好ましい。ペプチドは、酸化型グルタチオンおよび還元型グルタチオンなどのグルタチオンであることが好ましい。非生物的ストレス耐性向上剤は、ペプチドを1種のみ含んでもよいし、複数種含んでもよい。
 ベタインは、アミノ酸のN-アルキル置換体であることが好ましく、アミノ酸のN-トリアルキル置換体であることがより好ましく、アミノ酸のN-トリメチル置換体であることがさらに好ましい。ベタインの例には、グルタミン酸、アスパラギン酸、ヒスチジン、アルギニン、リシン、グリシン、アラニン、バリン、ロイシン、イソロイシン、スレオニン、フェニルアラニン、チロシン、トリプトファン、プロリン、セリン、グルタミン、アスパラギン、5-アミノレブリン酸、γ-アミノ酪酸、システイン、メチオニン、およびオルニチンの、N-アルキル置換体、好ましくはN-トリアルキル置換体、よりN-トリメチル置換体、ならびにカルニチンが含まれる。ベタインの好ましい例には、グリシンベタイン、アラニンベタイン、およびグルタミン酸ベタインなどが含まれる。非生物的ストレス耐性向上剤は、ベタインを1種のみ含んでもよいし、複数種含んでもよい。
 有機酸の例には、カルボン酸、スルホン酸、フェノールおよびチオールなどが含まれる。非生物的ストレス耐性向上剤は、有機酸を1種のみ含んでもよいし、複数種含んでもよい。
 カルボン酸は、炭素数が1以上10以下であることが好ましく、1以上6以下であることがより好ましい。カルボン酸は、モノカルボン酸であってもよく、ジカルボン酸であってもよく、トリカルボン酸であってもよい。カルボン酸の具体例には、乳酸、グリコール酸、ギ酸、酢酸、プロピオン酸、酪酸、シュウ酸、マロン酸、コハク酸、アジピン酸、クエン酸、グルタル酸、リンゴ酸、アスコルビン酸、酒石酸、フマル酸、マレイン酸、ピルビン酸、および安息香酸などが含まれる。これらのうち、酢酸、クエン酸およびマレイン酸が好ましい。
 フェノール類の例には、フェノールおよびアミノフェノールなどが含まれる。
 スルホン酸の例には、メタンスルホン酸、エタンスルホン酸、プロパンスルホン酸、ブタンスルホン酸、ペンタンスルホン酸、ヘキサンスルホン酸、ベンゼンスルホン酸、およびトルエンスルホン酸などが含まれる。
 有機酸の塩の例には、上述した有機酸の、ナトリウム塩およびリチウム塩などのアルカリ金属塩、ならびにマグネシウム塩およびカルシウム塩などのアルカリ土類金属塩などが含まれる。
 第1の成分との相乗効果を高める観点からは、有機酸またはその塩は、カルボン酸またはその塩であることが好ましく、炭素数1以上10以下のカルボン酸またはその塩であることがより好ましく、酢酸、クエン酸もしくはマレイン酸またはその塩であることがさらに好ましい。
 核酸塩基は、デオキシリボ核酸またはリボ核酸を構成するヌクレオチド分子に含まれる塩基である。核酸塩基の例には、グアニン、チミン、ウラシル、アデニン、およびシトシンなどが含まれる。これらのうち、グアニン、チミン、およびウラシルが好ましい。非生物的ストレス耐性向上剤は、核酸塩基を1種のみ含んでもよいし、複数種含んでもよい。
 ビタミンは、脂溶性ビタミンであってもよいし水溶性ビタミンであってもよい。非生物的ストレス耐性向上剤は、ビタミンを1種のみ含んでもよいし、複数種含んでもよい。複数種のビタミンを含むとき、非生物的ストレス耐性向上剤は、脂溶性ビタミンのみを複数種含んでもよいし、水溶性ビタミンのみを複数種含んでもよいし、脂溶性ビタミンおよび水溶性ビタミンの両方を含んでもよい。
 脂溶性ビタミンの例には、ビタミンA類、ビタミンD類、ビタミンE類、ビタミンK類などが含まれる。ビタミンD類の例には、ビタミンD2、ビタミンD3およびビタミンD4などが含まれる。水溶性ビタミンの例には、ビタミンB1、ビタミンB2、ビタミンB6およびビタミンB12などのビタミンB類、ならびにビタミンC類などが含まれる。これらのうち、ビタミンB1およびビタミンD2が好ましい。
 糖は、単糖であってもよいし、二糖であってもよいし、多糖であってもよい。単糖の例には、グルコース、フルクトース、およびガラクトースなどが含まれる。二糖の例には、トレハロース、ラクトース、マルトース、およびスクロースなどが含まれる。多糖の例には、セルロースおよびデンプンなどが含まれる。
 糖アルコールの例には、マンニトール、イノシトール、エリスリトール、キシリトール、およびソルビトールなどが含まれる。
 [非生物的ストレス耐性の向上]
 本実施態様に関する非生物的ストレス耐性向上剤は、有効成分としての第1の成分および第2の成分の相乗効果により、植物の非生物的ストレス耐性を顕著に向上させる。非生物的ストレスとは、害虫、病気、雑誌、雑草および間引きなどの生物的ストレスとは異なる、非生物的なストレスを意味する。非生物的ストレスの例には、高温ストレス、低温ストレス、凍結ストレス、塩ストレス、過剰養分ストレス、乾燥ストレス、過剰水ストレス、紫外線ストレス、弱光ストレス、強光ストレス、過剰刈込ストレス、踏圧ストレス、雹ストレスおよび強風ストレスなどが含まれる。非生物的ストレス耐性向上剤は、これにより処理された植物の、これらの非生物的ストレス、たとえば高温ストレスに対する耐性を向上させる。
 また、上記非生物的ストレス耐性向上剤は、非生物的ストレスに対する耐性を向上させることにより、非生物的ストレスによる、植物の収穫量の減少を抑制したり、草丈の低下や草丈の生長速度の低下を抑制したり、環境ストレスによる花数の低下を抑制したり、地上部重量および地下部重量の低下を抑制したりすることもできる。
 上記非生物的ストレス耐性向上剤は、いずれの植物の処理に使用するものでもよい。上記植物の例には、イネ、コムギ、オオムギ、ライムギ、エンバク、ライコムギ(トリチケール)、トウモロコシ、モロコシ(ソルガム)、サトウキビ、シバ、ベントグラス、バミューダグラス、フェスクおよびライグラスなどのイネ科類、ダイズ、ラッカセイ、インゲンマメ、エンドウ、アズキおよびアルファルファなどのマメ科類、サツマイモなどのヒルガオ科類、トウガラシ、ピーマン、トマト、ナス、ジャガイモおよびタバコなどのナス科類、ソバなどのタデ科類、ヒマワリなどのキク科類、チョウセンニンジンなどのウコギ科類、ナタネ、ブロッコリー、ハクサイ、カブ、キャベツ、ルッコラ、ダイコンおよびハツカダイコンなどのアブラナ科類、テンサイなどのアカザ科類、ワタなどのアオイ科類、コーヒーノキなどのアカネ科類、カカオなどのアオギリ科類、チャなどのツバキ科類、スイカ、メロン、キュウリおよびカボチャなどのウリ科類、タマネギ、ネギおよびニンニクなどのユリ科類、イチゴ、リンゴ、アーモンド、アンズ、ウメ、オウトウ、スモモ、モモおよびナシなどのバラ科類、ニンジンなどのセリ科類、サトイモなどのサトイモ科類、マンゴーなどのウルシ科類、パイナップルなどのパイナップル科類、パパイアなどのパパイア科類、カキなどのカキノキ科類、ブルーベリーなどのツツジ科類、ペカンなどのクルミ科類、バナナなどのバショウ科類、オリーブなどのモクセイ科類、ココヤシおよびナツメヤシなどのヤシ科類、みかん、オレンジ、グレープフルーツおよびレモンなどのミカン科類、ブドウなどのブドウ科類、草花(Flowers and ornamental plants)、果樹以外の樹ならびにその他の観賞用植物などが含まれる。

 上記第1の成分および第2の成分は、有効成分として上記第1の成分および第2の成分を含み、さらにその他の補助剤またはその他の有効成分と共に、各種剤型の製剤として市場に流通させることができる。上記活性成分そのものを製剤としてもよいし、その他の補助剤またはその他の有効成分と共に、各種剤型の製剤としてもよい。剤型は特に限定されず、処理方法に応じて選択すればよい。剤型の例には、粉剤、粒剤、粉粒剤、水和剤、水溶剤、乳剤、液剤、油剤、エアゾル、マイクロカプセル剤、ペースト剤、塗布剤、くん煙剤、くん蒸剤および微量散布剤などが含まれる。

 その他の有効成分の例には、バイオスティミュラント、植物生長調節剤、殺菌剤、殺虫剤、殺ダニ剤、殺線虫剤および除草剤に含まれる有効成分を挙げることができる。なお、上記第1の成分および第2の成分に他のバイオスティミュラントを併用することで、非生物的ストレスへの耐性をより高めたり、生長の促進効果をより高めたりすることもできる。

 上記微生物由来昆虫中腸内膜破壊剤の例には、Bacillus thuringiensis subsp. israelensis、Bacillus thuringiensis subsp. aizawai、Bacillus thuringiensis subsp. kurstaki、Bacillus thuringiensis subsp. tenebrionis、B.t.作物に含まれるタンパク質(Cry1Ab、Cry1Ac、Cry1Fa、Cry1A.105、Cry2Ab、Vip3A、mCry3A、Cry3Bb、Cry34Ab1/Cry35Ab1)およびBacillus sphaericusなどが含まれる。
 上記ミトコンドリアATP合成酵素阻害剤の例には、ジアフェンチウロン、アゾシクロチン、水酸化トリシクロヘキシルスズ(シヘキサチン)、酸化フェンブタスズ、BPPS(プロパルギット)およびテトラジホンなどが含まれる。
 上記プロトン勾配を攪乱する酸化的リン酸化脱共役剤の例には、クロルフェナピル、DNOCおよびスルフルラミドなどが含まれる。
 上記ニコチン性アセチルコリン受容体(nAChR)チャネルブロッカーの例には、ベンスルタップ、カルタップ塩酸塩、チオシクラム、チオスルタップナトリウム塩およびモノスルタップなどが含まれる。

 [含有量]
 製剤中の第1の成分の含有量は、剤型や植物への付与量等に応じて任意に定めることができる。たとえば、製剤は、第1の成分を、その全質量に対して0.01質量%以上90質量%以下含むことが好ましく、0.1質量%以上50重量%以下含むことがより好ましい。また、製剤は、第2の成分を、その全質量に対して0.00001質量%以上90質量%含むことが好ましく、0.0001質量%以上50質量%含むことがより好ましい。
 また、第1の成分の全質量に対する第2の成分の質量比(第2の成分/第1の成分)は、0.001以上1000以下とすることができ、0.01以上100以下であることが好ましく、0.1以上10以下であることがより好ましく、0.5以上5以下であることがさらに好ましい。
 上記製剤は、そのまま処理剤として使用してもよいし、任意に、上述した補助剤またはその他の有効成分と混合等して調製した処理剤として使用してもよい。
 [製造方法]
 非生物的ストレス耐性向上剤は、第1の成分および第2の成分を混合して調製することができる。このとき、固体担体または液体担体を用いて、所定の剤型を有する製剤を調製してもよい。なお、第1の成分は、これを含む微生物、植物または海藻の抽出物を使用してもよい。また、第2の成分は、これを含む微生物、植物または海藻の抽出物を使用してもよい。また、第1の成分および第2の成分を含む、微生物、植物または海藻の抽出物を、非生物的ストレス耐性向上剤として使用してもよい。
 なお、第1の成分および第2の成分は、混合された状態で保管および輸送されてもよいし、第1の容器に第1の成分を収容し、第2の容器に第2の成分を収容して、混合されていない別個独立の状体で保管および輸送されてもよい。また、第1の成分を収容した第1の容器と、第2の成分を収容した第2の容器とをパッケージ化(キット化)した製品としてもよい。
 [植物の非生物的ストレス耐性向上方法]
 上述した非生物的ストレス耐性向上剤またはこれを含む製剤は、予め用意した植物にこれを付与することで、当該植物の非生物的ストレス耐性を向上させるために使用することができる。
 付与の方法は特に限定されず、茎葉散布、給水への混入、土壌への散布、注入機を用いた下層土までの注入、種子、球根および塊茎など(繁殖体)への処理、ならびに植物への直接施肥など、いかなる方法で付与してもよい。
 給水に混入させるときは、たとえば、作物への給水または水田の田面水に、粒剤や処理液として上記製剤を投与すればよい。たとえば、作物への給水中の第1の成分の濃度は、0.1mg/L以上とすることができ、1mg/L以上とすることが好ましい。また、田面水に投与するときは、第1の成分の投与量は、水田10aあたり0.1g以上とすることができ、1g以上とすることが好ましい。
 茎葉散布または土壌への散布を行うときは、たとえば、苗の移植時などに植穴またはその周辺に粒剤などを投与したり、種子、植物体または植物体の周囲の土壌に粒剤および水和剤などを投与したりすればよい。また、土壌への散布の後、土と製剤とを撹拌してもよい。茎葉散布または土壌表面へ散布するときの第1の成分の投与量は、農園芸地1mあたり0.1mg以上とすることができ、1mg以上とすることが好ましい。
 繁殖体の処理を行うときは、たとえば、水和剤および粉剤などを繁殖体と混合して攪拌したり、希釈した水和剤などに繁殖体を浸漬したりすればよい。また、固体担体を含む製剤により、繁殖体をコーティングしてもよい。繁殖体の処理を行うときの第1の成分の使用量は、種子100kgに対して0.005g以上とすることができ、0.05g以上とすることが好ましい。
 処理された植物は、当該植物を生育させるための通常の条件で育成することができる。このようにして育成した植物は、ある程度生長させた移植苗を他の土壌中や培地に移植してさらに育成してもよい。
 [その他の実施形態]
 なお、上述の実施形態は、本発明の例示的な実施形態であり、本発明は、その中核となる技術思想の範囲内において上述の実施形態以外の実施態様を含み得ることはいうまでもない。
 本発明を実施例に基づき詳細に説明するが、本発明はこれらの実施例に限定されない。
 〔評価例1〕高温ストレスに対する耐性効果の比較
 以下の表1に記載の濃度になるように、実施例1、2および比較例1~3、5~7の非生物的ストレス耐性向上剤を調整した。エルゴチオネイン(EGT)、アルギニン(Arg)およびグルタミン酸(Glu)は市販品を使用し、溶媒として純水を使用した。
 直径60mm、高さ55mmのプラスチックポットに、育苗培土(タネのタキイ社製)90mLを入れ、1ポットあたり1個体のシロイヌナズナ(Col-0)を播種した。直径160mm、高さ28mmのプラスチック製深皿を用意し、2ポット設置した。
 室温22℃に設定した人工気象器内で、明期16時間、暗期8時間とした。光条件は、蛍光灯照射下の中心部において光強度5000lxとなるように設定した。給水は下部給水とし、水位を約5mmとした。播種後92日目に、給水に替えて、非生物的ストレス耐性向上剤40mLを添加し、その2日後に45℃環境下に1時間さらし、さらにその3日後に42℃環境下に3時間さらすことで高温ストレスを与えた。
 高温ストレスを与えたシロイヌナズナの萎れた葉の数を計数し、生理障害の指標として葉の萎れ率を評価した。評価結果を表1に示す。葉の萎れ率、非生物的ストレス抑制率(評価値)は以下の式の通り算出した。

 を用いた方法により、2種の化合物の協力効果の判定を行った。
 なお、上記式中、αおよびβは、それぞれの化合物の単独添加時の非生物的ストレス抑制率を示している。

 表1に示す通り、高温ストレスにより供試したシロイヌナズナの葉の67%に萎れの生理障害が見られた(比較例4)。実施例1および2の非生物的ストレス耐性向上剤は、ArgまたはGluのみを含む非生物的ストレス耐性向上剤(比較例2、3)、および、EGTのみを含む非生物的ストレス耐性向上剤(比較例1)と比較して、葉の萎れ率が低く、非生物的ストレス抑制率が優れていた。また、実施例1および2の非生物的ストレス耐性向上剤は、混合時の理論値よりも大きく、相乗効果が確認された。
 〔評価例2〕紫外線ストレスに対する耐性効果の比較
 以下の表2に記載の濃度になるように、実施例3および比較例9、10、12、13の非生物的ストレス耐性向上剤を調整した。EGTおよびGluは市販品を使用し、溶媒として純水を使用した。
 ろ紙を敷いた9cmのシャーレに純水5mLを添加し、1シャーレあたり10粒のコムギの種子を播種した。室温22℃に設定した人工気象器内で管理し、明期16時間、暗期8時間とした。光条件は、蛍光灯照射下の中心部において光強度5000lxとなるように設定した。
 播種後8日目にシャーレ内の純水を抜き取った後に、シャーレに非生物的ストレス耐性向上剤5mLを添加し、その1日後に、紫外線ランプ(東芝社製、GL-15)照射下において254nmの波長の紫外線放射強度が535μW/cm-2となるように設定し、1時間さらすことで紫外線ストレスを与えた。
 紫外線ストレスを与えてから5日後の枯死した葉を計数し、生理障害の指標として葉の枯死率を評価した。評価結果を表10に示す。葉の枯死率および非生物的ストレス抑制率(評価値)は以下の式の通り算出した。

 表2に示す通り、紫外線ストレスによりコムギにクロロシスなどの生理障害が見られ、供試したコムギの葉の48%が枯死した(比較例11)。実施例3の非生物的ストレス耐性向上剤は、Gluのみを含む非生物的ストレス耐性向上剤(比較例10)、および、EGTのみを含む非生物的ストレス耐性向上剤(比較例9)と比較して、葉の枯死率が低く、非生物的ストレス抑制率が優れていた。また、実施例3の非生物的ストレス耐性向上剤は、混合時の理論値よりも大きく、相乗効果が確認された。
 〔評価例3〕過剰水ストレスに対する耐性効果の比較
 以下の表3に記載の濃度になるように、実施例4~7および比較例15~17、19~21の非生物的ストレス耐性向上剤を調整した。EGTおよびGluは市販品を使用し、溶媒として純水を使用した。
 ろ紙を敷いた9cmのシャーレに純水5mLを添加し、1シャーレあたり10粒のナタネの種子を播種した。室温22℃に設定した人工気象器内で管理し、明期16時間、暗期8時間とした。光条件は、蛍光灯照射下の中心部において光強度5000lxとなるように設定した。
 播種後8日目にシャーレ内の純水を抜き取った後に、シャーレに非生物的ストレス耐性向上剤5mLを添加し、その1日後に、シャーレに純水50mLを添加することで過剰水ストレスを与えた。
 過剰水ストレスを与えてから10日後の生存した植物を計数し、生理障害の指標として植物の枯死率を評価した。評価結果を表3に示す。枯死率および非生物的ストレス抑制率(評価値)は以下の式の通り算出した。

 表3に示す通り、過剰水ストレスによりナタネにクロロシスなどの生理障害が見られ、供試したナタネの84%が枯死した(比較例18)。実施例4、5、6および7の非生物的ストレス耐性向上剤は、ArgまたはGluのみを含む非生物的ストレス耐性向上剤(比較例16、17)、および、EGTのみを含む非生物的ストレス耐性向上剤(比較例15)と比較して、枯死率が低く、非生物的ストレス抑制率が優れていた。また、実施例4および6の非生物的ストレス耐性向上剤は、混合時の理論値よりも大きく、相乗効果が確認された。
 〔評価例4〕乾燥ストレスに対する耐性効果の比較
 以下の表4に記載の濃度になるように、実施例8~15および比較例23~31の非生物的ストレス耐性向上剤を調整した。エルゴチオネイン(EGT)、トレハロース、メチオニン(Met)、酸化型グルタチオン(GSSG)、γ-アミノ酪酸(GABA)、クエン酸、アスコルビン酸(VitC)およびソルビン酸カリウム(ソルビン酸)は市販品を使用し、溶媒として純水を使用した。
 60mm角、高さ66mmのプラスチックポットに、粒状培土(くみあい園芸培土)150mLを入れ、1ポットあたり1個体のダイズを播種した。直径160mm、高さ28mmのプラスチック製深皿を用意し、4ポット設置した。
 室温25℃に設定した温室内で管理した。播種後31日目に、給水に替えて、非生物的ストレス耐性向上剤50mLを深皿に添加し、その後5日間、給水を止めることで乾燥ストレスを与えた。
 乾燥ストレスを与えたダイズの枯死した葉の数を計数し、生理障害の指標として葉の枯死率を評価した。評価結果を表4に示す。葉の枯死率、非生物的ストレス抑制率(評価値)は以下の式の通り算出した。

 表4に示す通り、乾燥ストレスによりダイズにネクロシスなどの生理障害が見られ、供試したダイズの葉の56%が枯死した(比較例32)。実施例8、9、10、11、12、13、14および15の非生物的ストレス耐性向上剤は、トレハロース、Met、GSSG、GABA、クエン酸、VitCまたはソルビン酸のみを含む非生物的ストレス耐性向上剤(比較例24、25、26、27、28、29、30、31)、および、EGTのみを含む非生物的ストレス耐性向上剤(比較例23)と比較して、葉の枯死率が低く、非生物的ストレス抑制率が優れていた。また、実施例8、9、10、11、12、13、14および15の非生物的ストレス耐性向上剤は、混合時の理論値よりも大きく、相乗効果が確認された。
 〔評価例5〕低温ストレスに対する耐性効果の比較
 以下の表5に記載の濃度になるように、実施例16~17および比較例34~36の非生物的ストレス耐性向上剤を調整した。エルゴチオネイン(EGT)、ヒスチジン(His)およびウラシルは市販品を使用し、溶媒として純水を使用した。
 ろ紙を敷いた9cmのシャーレに純水5mLを添加し、1シャーレあたり20粒のルッコラの種子を播種した。室温22℃に設定した人工気象器内で管理し、明期16時間、暗期8時間とした。光条件は、蛍光灯照射下の中心部において光強度5000lxとなるように設定した。
 播種後6日目にシャーレ内の純水を抜き取った後に、シャーレに非生物的ストレス耐性向上剤5mLを添加し、その24時間後に、4℃環境下に48時間さらすことで低温ストレスを与えた。
 低温ストレスを与えてから2日後の萎れた葉の数を計数し、生理障害の指標として葉の萎れ率を評価した。評価結果を表5に示す。葉の萎れ率、非生物的ストレス抑制率(評価値)は以下の式の通り算出した。

 表5に示す通り、低温ストレスにより供試したルッコラの葉の94%に萎れの生理障害が見られた(比較例37)。実施例16および17の非生物的ストレス耐性向上剤は、Hisまたはウラシルのみを含む非生物的ストレス耐性向上剤(比較例35、36)、および、EGTのみを含む非生物的ストレス耐性向上剤(比較例34)と比較して、葉の萎れ率が低く、非生物的ストレス抑制率が優れていた。また、実施例16および17の非生物的ストレス耐性向上剤は、混合時の理論値よりも大きく、相乗効果が確認された。
 〔評価例6〕凍結ストレスに対する耐性効果の比較
 以下の表6に記載の濃度になるように、実施例18および比較例39、40の非生物的ストレス耐性向上剤を調整した。エルゴチオネイン(EGT)およびリンゴ酸は市販品を使用し、溶媒として純水を使用した。
 ろ紙を敷いた9cmのシャーレに純水5mLを添加し、1シャーレあたり20粒のブロッコリー(緑積)の種子を播種した。室温22℃に設定した人工気象器内で管理し、明期16時間、暗期8時間とした。光条件は、蛍光灯照射下の中心部において光強度5000lxとなるように設定した。
 播種後6日目にシャーレ内の純水を抜き取った後に、シャーレに非生物的ストレス耐性向上剤5mLを添加し、その24時間後に、-20℃環境下に30分さらし、更にその48時間後に-20℃環境下に30分さらすことで凍結ストレスを与えた。
 凍結ストレスを与えてから1日後の生存した植物を計数し、生理障害の指標として植物の枯死率を評価した。評価結果を表6に示す。枯死率および非生物的ストレス抑制率(評価値)は以下の式の通り算出した。

 表6に示す通り、凍結ストレスによりブロッコリーに茎の折れまたはネクロシスなどの生理障害が見られ、供試したブロッコリーの44%が枯死した(比較例41)。実施例18の非生物的ストレス耐性向上剤は、リンゴ酸のみを含む非生物的ストレス耐性向上剤(比較例40)、および、EGTのみを含む非生物的ストレス耐性向上剤(比較例39)と比較して、枯死率が低く、非生物的ストレス抑制率が優れていた。また、実施例18の非生物的ストレス耐性向上剤は、混合時の理論値よりも大きく、相乗効果が確認された。
 〔評価例7〕過剰養分ストレスに対する耐性効果の比較
 以下の表7に記載の濃度になるように、実施例19~23および比較例43~48の非生物的ストレス耐性向上剤を調整した。エルゴチオネイン(EGT)、プロリン(Pro)、5-アミノレブリン酸(5-ALA)、グリシンベタイン(GB)および酢酸は市販品を使用し、溶媒として純水を使用した。
 60mm角、高さ66mmのプラスチックポットに、粒状培土(くみあい園芸培土)150mLを入れ、1ポットあたり1個体のダイズを播種した。直径160mm、高さ28mmのプラスチック製深皿を用意し、2ポット設置した。
 室温25℃に設定した温室内で管理した。播種後31日目に、給水に替えて、非生物的ストレス耐性向上剤50mLを深皿に添加し、その1日後に、液体肥料(HYPONeX、ハイポネックスジャパン社製)を、5倍希釈で施肥することで過剰養分ストレスを与えた。
 過剰養分ストレスを与えてから1日後のダイズの枯死した葉の数を計数し、生理障害の指標として葉の枯死率を評価した。評価結果を表7に示す。葉の枯死率、非生物的ストレス抑制率(評価値)は以下の式の通り算出した。

 表7に示す通り、過剰養分ストレスによりダイズにクロロシスなどの生理障害が見られ、供試したダイズの葉の76%が枯死した(比較例49)。実施例19、20、21、22および23の非生物的ストレス耐性向上剤は、Pro、5-ALA、GBまたは酢酸のみを含む非生物的ストレス耐性向上剤(比較例45、46、47、48)、および、EGTのみを含む非生物的ストレス耐性向上剤(比較例43、44)と比較して、葉の枯死率が低く、非生物的ストレス抑制率が優れていた。また、実施例19、20、21、22および23の非生物的ストレス耐性向上剤は、混合時の理論値よりも大きく、相乗効果が確認された。
 〔評価例8〕高温ストレスに対する耐性効果の比較
 以下の表8に記載の濃度になるように、実施例24、25および比較例51~54の非生物的ストレス耐性向上剤を調整した。エルゴチオネイン(EGT)、酢酸およびプロリン(Pro)は市販品を使用し、溶媒として純水を使用した。
 直径60mm、高さ55mmのプラスチックポットに、育苗培土(タネのタキイ社製)90mLを入れ、1ポットあたり1個体のトマト(レジナ)を播種した。直径160mm、高さ28mmのプラスチック製深皿を用意し、4ポット設置した。
 室温22℃に設定した人工気象器内で、明期16時間、暗期8時間とした。光条件は、蛍光灯照射下の中心部において光強度5000lxとなるように設定した。給水は下部給水とし、水位を約5mmとした。播種後30日目に、給水に替えて、非生物的ストレス耐性向上剤50mLを添加し、その2日後に45℃環境下に1時間さらし、さらにその後に55℃環境下に30分間さらすことで高温ストレスを与えた。
 高温ストレスを与えたトマトの萎れた葉の数を計数し、生理障害の指標として葉の萎れ率を評価した。評価結果を表8に示す。葉の萎れ率、非生物的ストレス抑制率(評価値)は以下の式の通り算出した。

 表8に示す通り、高温ストレスにより供試したトマトの葉の62%に萎れの生理障害が見られた(比較例55)。実施例24および25の非生物的ストレス耐性向上剤は、酢酸またはProのみを含む非生物的ストレス耐性向上剤(比較例53、54)、および、EGTのみを含む非生物的ストレス耐性向上剤(比較例51、52)と比較して、葉の萎れ率が低く、非生物的ストレス抑制率が優れていた。また、実施例24および25の非生物的ストレス耐性向上剤は、混合時の理論値よりも大きく、相乗効果が確認された。
 本出願は、2023年11月21日出願の特願2023-197530号の優先権を主張する。当該出願の出願当初の明細書、および請求の範囲に記載された事項は、参照により本出願に援用される。
 本発明によれば、植物の非生物的ストレス耐性を高め、植物の生産量を容易に増加させることができる。

Claims (9)

  1.  下記式(I)で示される化合物もしくはその互変異性体、またはそれらの農学的に許容可能な塩である第1の成分と、
     アミノ酸、2以上10以下のアミノ酸長であるペプチド、ベタイン、有機酸またはその塩、核酸塩基、ビタミン、および、糖または糖アルコールからなる群から選択される少なくとも1である第2の成分と、を含む、植物の非生物的ストレス耐性向上剤。

     (式(I)中、RおよびRは、独立して水素原子または炭素数1以上4以下のアルキル基を表し、R、RおよびRは、独立して炭素数1以上4以下のアルキル基を表す。)
  2.  前記式(I)で示される化合物は、エルゴチオネインである、
     請求項1に記載の非生物的ストレス耐性向上剤。
  3.  植物への付与により、高温ストレス、低温ストレス、凍結ストレス、塩ストレス、過剰養分ストレス、乾燥ストレス、過剰水ストレス、紫外線ストレス、弱光ストレス、強光ストレス、過剰刈込ストレス、踏圧ストレス、雹ストレスおよび強風ストレスからなる群から選択される少なくとも1つである非生物的ストレスに対する前記植物の耐性を向上させる、
     請求項1または2に記載の非生物的ストレス耐性向上剤。
  4.  植物への付与により、高温ストレス、紫外線ストレスおよび過剰水ストレスからなる群から選択される少なくとも1つである非生物的ストレスに対する前記植物の耐性を向上させる、
     請求項1~3のいずれか1項に記載の非生物的ストレス耐性向上剤。
  5.  前記第2の成分は、アミノ酸を含み、
     前記アミノ酸は、グルタミン酸またはアルギニンである、
     請求項1~4のいずれか1項に記載の非生物的ストレス耐性向上剤。
  6.  植物を用意する工程と、
     前記植物を、請求項1~5のいずれか1項に記載の非生物的ストレス耐性向上剤で処理する工程と、を含む、
     植物の非生物的ストレス耐性向上方法。
  7.  下記式(I)で示される化合物もしくはその互変異性体、またはそれらの農学的に許容可能な塩である第1の成分と、
     アミノ酸、2以上10以下のアミノ酸長であるペプチド、ベタイン、有機酸またはその塩、核酸塩基、ビタミン、および、糖または糖アルコールからなる群から選択される少なくとも1である第2の成分と、
     液体担体または固体担体と、を含む、
     植物の非生物的ストレス耐性向上用製剤。
