Description

サンプル溶液分離デバイス、サンプル溶液分離システム、及びサンプル溶液分離方法
 本発明は、サンプル溶液を分離するサンプル溶液分離デバイス、サンプル溶液分離システム、及びサンプル溶液分離方法に関する。
 デジタルPCR(Polymerase Chain Reaction:ポリメラーゼ連鎖反応)は、核酸を高感度に検出する技術であり、従来のリアルタイム定量PCRと比較して、低頻度な遺伝子変異を検出することができる。
 デジタルPCRは、サンプル溶液を微小な体積に分割した後、分割された溶液内でPCRを実施し、計測対象のDNAを増幅し、計測する。サンプル溶液を微小な容器に入れる方法として、特許文献1には、ポートをシールして容器内を真空化し、サンプル溶液を容器内に引き込む方法が開示されている。また、非特許文献1には、容器内を脱気し、アウトレット側のバルブを閉じ、インレット側のバルブを開けて溶液を入れる方法が開示されている。特許文献2には、気液分離フィルタを用いて、溶液を無駄にすることなく吐出する構成が開示されている。

 デジタルPCRでは、検出対象のDNAを含むサンプル溶液を多数の区画に分離し、各々の区画に対してPCRを行い、各区画内に存在するDNAの種類の判別を行う。各区画内に分離した後にPCRを行うことにより、高感度に対象DNAを計測できることが特徴である。例えば、血液中に微量に存在するセルフリーDNA(cfDNA)を検出する液体生検が適した応用の一つである。セルフリーDNAには腫瘍由来のDNAであるctDNA(circlating tumor DNA)が含まれる場合があり、ctDNAの変異の種類や割合を計測することにより、がんの診断、治療選択及び治療効果のモニタリングへの応用が期待されている。
 cfDNAやctDNAは血液中に微量に存在するため、高感度に計測する必要がある。デジタルPCRでは、サンプル溶液を複数の微小体積のチャンバに分離して計測する。その際に、サンプル溶液の全てを使用できるものではなく、微小体積のチャンバに分離できず、計測することができない溶液、すなわち、デッドボリュームが生じる。高感度に計測するためには、収集したサンプル溶液を可能な限り多く計測する必要がある。すなわち、サンプル溶液のデッドボリュームを削減する必要がある。
 本発明は、このようなことに鑑みてなされたものであり、その目的の一つは、サンプル溶液のデッドボリュームを削減することが可能なサンプル溶液分離デバイス、サンプル溶液分離システム、及びサンプル溶液分離方法を提供することにある。
 上記課題を解決するために、本発明のサンプル溶液分離デバイスは、サンプル溶液を分離するサンプル溶液分離デバイスであって、複数のマイクロチャンバと、複数のマイクロチャンバを接続する流路と、サンプル溶液の流路への導入口である第一の開口部と、第一の開口部と複数のマイクロチャンバとの間に設けられるバルブと、サンプル溶液が収容される複数のマイクロチャンバ間を分離する分離液の流路への導入口であって、複数のマイクロチャンバを挟んで第一の開口部の反対側に設けられる第二の開口部と、第二の開口部と複数のマイクロチャンバとの間に設けられる固相と、を備え、固相は、通気性を有し、サンプル溶液に対して撥水性を有し、且つ分離液に対して通過性を有する。
 また、本発明のサンプル溶液分離システムは、サンプル溶液を分離するサンプル溶液分離デバイスであって、複数のマイクロチャンバと、複数のマイクロチャンバを接続する流路と、サンプル溶液の流路への導入口である第一の開口部と、第一の開口部と複数のマイクロチャンバとの間に設けられるバルブと、サンプル溶液が収容される複数のマイクロチャンバ間を分離する分離液の流路への導入口であって、複数のマイクロチャンバを挟んで第一の開口部の反対側に設けられる第二の開口部と、第二の開口部と複数のマイクロチャンバとの間に設けられる固相と、を備え、固相は、通気性を有し、サンプル溶液に対して撥水性を有し、且つ分離液に対して通過性を有する、サンプル溶液分離デバイス、複数のマイクロチャンバ及び流路内の空気を第二の開口部から固相を通じて脱気するポンプ、及びバルブを開放するバルブ制御手段と、第二の開口部から分離液を導入する分離液導入手段、を有する。
 また、本発明のサンプル溶液分離方法は、サンプル溶液を分離するサンプル溶液分離デバイスであって、複数のマイクロチャンバと、複数のマイクロチャンバを接続する流路と、サンプル溶液の流路への導入口である第一の開口部と、第一の開口部と複数のマイクロチャンバとの間に設けられるバルブと、サンプル溶液が収容される複数のマイクロチャンバ間を分離する分離液の流路への導入口であって、複数のマイクロチャンバを挟んで第一の開口部の反対側に設けられる第二の開口部と、第二の開口部と複数のマイクロチャンバとの間に設けられる固相と、を備え、固相は、通気性を有し、サンプル溶液に対して撥水性を有し、且つ分離液に対して通過性を有する、サンプル溶液分離デバイス、を準備すること、固相を介して、複数のマイクロチャンバ及び流路内の空気を脱気すること、バルブを開放し、第一の開口部からサンプル溶液を複数のマイクロチャンバ及び流路に導入すること、及び第二の開口部から固相を介して分離液を流路に導入すること、を有する。
 本発明のサンプル溶液分離デバイス、サンプル溶液分離システム、及びサンプル溶液分離方法によれば、複数のマイクロチャンバにサンプル溶液を分離する際に、サンプル溶液のデッドボリュームを削減することが可能となる。
 その他の課題と新規な特徴は、本明細書の記述及び添付図面から明らかになるであろう。

(実施例1)
 実施例1に係るサンプル溶液分離デバイス101について、図1~図4を用いて説明する。図1は、実施例1に係るサンプル溶液分離デバイス101の構成図である。サンプル溶液分離デバイス101は、サンプル溶液を複数のマイクロチャンバ105に分離して収容するデバイスである。サンプル溶液分離デバイス101は、第一の開口部102、バルブ103、流路104、複数のマイクロチャンバ105、固相106、及び第二の開口部107を備える。
 流路104は、バルブ103を介して第一の開口部102に接続される。流路104には、複数のマイクロチャンバ105が接続される。また、流路104は、固相106を介して第二の開口部107に接続される。すなわち、流路104は、第一の開口部102、複数のマイクロチャンバ105、及び第二の開口部107を接続する。
 第一の開口部102は、サンプル溶液の流路104への導入口である。
 バルブ103は、第一の開口部102と複数のマイクロチャンバ105との間に設けられる。
 第二の開口部107は、サンプル溶液が収容される複数のマイクロチャンバ105間を分離する分離液の流路104への導入口である。この第二の開口部107は、複数のマイクロチャンバ105を挟んで第一の開口部102の反対側に設けられる。
 固相106は、第二の開口部107と複数のマイクロチャンバ105との間に設けられる。固相106は、通気性を有し、サンプル溶液に対して撥水性を有し、且つ分離液に対して通過性を有する。
 図2A~図2Dは、実施例1に係るサンプル溶液分離デバイス101にサンプル溶液201及び分離液202を導入する様子を示した図である。
 まず、サンプル溶液201は、第一の開口部102に導入される。サンプル溶液201が第一の開口部102に導入される時点では、バルブ103は閉鎖している(図2A)。バルブ103を閉鎖したその後、第二の開口部107から、固相106を通じて、流路104及びマイクロチャンバ105の内部の空気を脱気する。すなわち、流路104及びマイクロチャンバ105を真空状態にする。
 流路104及びマイクロチャンバ105を真空状態にして、バルブ103を開けることにより、サンプル溶液201は、流路104及びマイクロチャンバ105に引き込まれる(図2B)。
 ここで、固相106は、通気性、疎水性、親油性を有する固体である。固相106の通気性により、空気を通過させることが可能となり、上記したように、流路104及びマイクロチャンバ105を真空状態にすることができる。また、固相106の疎水性により、サンプル溶液201のような水溶液を撥水させて、サンプル溶液201が通過するのを防止することができる。
 従って、流路104内に引き込まれたサンプル溶液201は、固相106に到達した時点で停止する。マイクロチャンバ105内は真空状態であるため、サンプル溶液201は、継続して引き込まれる。そして、マイクロチャンバ105内は、サンプル溶液201により満たされる(図2C)。
 サンプル溶液201が収容された複数のマイクロチャンバ105間を分離するための分離液202(図2D参照)は、サンプル溶液201と混和しない物性を有するもの(例えば、オイル)を用いる。固相106は、親油性を有することにより、この分離液202を通過させることができる。
 次に、流路104内にあるサンプル溶液201は、分離液202に置き換えられる。分離液202は、第二の開口部107より導入される。固相106は、親油性を有し、分離液202を通過させる性質を有するため、分離液202は、第二の開口部107から、固相106を通じて、流路104へと導入される。流路104内にあるサンプル溶液201は、第一の開口部102側へと押し戻され、流路104が分離液202により満たされる(図2D)。このとき、サンプル溶液201は、マイクロチャンバ105内に残るため、流路104の分離液202により、複数のマイクロチャンバ105内のサンプル溶液201が分離される。
 このように、通気性、疎水性、親油性を有する固相106を用いることにより、流路104及びマイクロチャンバ105内の空気を抜くことができ、かつ、サンプル溶液201を固相106の位置で停止させることができる。また、固相106は、分離液202を流路104に導入し、サンプル溶液201をマイクロチャンバ105内に分割して封入することができる。
 図3は、実施例1に係るサンプル溶液分離デバイス101にサンプル溶液201及び分離液202を導入し、サンプル溶液201をマイクロチャンバ105に分離する際のフロー図である。
 まず、サンプル溶液201は、第一の開口部102にセットされる(S301)。このとき、バルブ103は、流路104を閉鎖している。
 その後、第二の開口部107に接続されるポンプ403(図4参照)が駆動されることにより、流路104及び複数のマイクロチャンバ105内の空気が固相106及び第二の開口部107を介して排出される(S302)。これにより、流路104及び複数のマイクロチャンバ105が真空状態となる。
 流路104及び複数のマイクロチャンバ105が真空状態となった後、バルブ103が流路104を開放することにより、サンプル溶液201が第一の開口部102からバルブ103を介して、流路104及び複数のマイクロチャンバ105内に導入される(S303)。
 その後、第二の開口部107に接続されるポンプ405(図4参照)が駆動されることにより、分離液202が第二の開口部107及び固相106を介して、流路104に導入される(S304)。流路104内のサンプル溶液201は、分離液202によって第一の開口部102の方向に押し戻され、流路104内がサンプル溶液201から分離液202に置き換わり、複数のマイクロチャンバ105間が分離液202によって分離される。なお、分離液202を流路104に導入した後、複数のマイクロチャンバ105内のサンプル溶液201及び流路104内の分離液202が移動しないように、バルブ103を閉鎖してもよい。
 図4は、実施例1に係るサンプル溶液分離デバイス101にサンプル溶液201を分離して導入するサンプル溶液分離システム400の構成図である。サンプル溶液分離システム400は、バルブ制御機構401、開口部コンタクト部402、ポンプ403、開口部コンタクト部404、ポンプ405、移動機構406、及びサンプル溶液分離デバイス101を備える。
 バルブ制御機構401(バルブ制御手段)は、バルブ103の開閉を制御する。サンプル溶液201を複数のマイクロチャンバ105内に導入する際には、バルブ制御機構401は、バルブ103を閉鎖状態から開放状態にする。
 開口部コンタクト部402は、サンプル溶液分離デバイス101の第二の開口部107に接続される。また、開口部コンタクト部402は、流路104及び複数のマイクロチャンバ105の真空引きを行うポンプ403に接続される。移動機構406は、流路104及び複数のマイクロチャンバ105の真空引きを行う際に、開口部コンタクト部402を移動して、第二の開口部107に接続する。
 ポンプ403は、流路104及びマイクロチャンバ105内の空気を第二の開口部107から固相106を通じて脱気し、流路104及び複数のマイクロチャンバ105の真空引きを行う。
 開口部コンタクト部404は、サンプル溶液分離デバイス101の第二の開口部107に接続される。また、開口部コンタクト部404は、流路104に分離液202を導入するポンプ405に接続される。
 移動機構406(切替手段)は、流路104及び複数のマイクロチャンバ105から空気を脱気する際に、開口部コンタクト部402を第二の開口部107に接続する。また、移動機構406は、流路104に分離液202を導入する際に、開口部コンタクト部404を移動して、第二の開口部107に接続する。すなわち、移動機構406は、第二の開口部107の接続先をポンプ403又はポンプ405に切り替える。
 ポンプ405(分離液導入手段)は、第二の開口部107から固相106を通じて流路104に分離液202を導入する。
 真空引きを行うポンプ403は、大気圧から、例えば、5kPa~0.01kPa程度まで減圧可能なポンプを用いる。真空度が高ければ、マイクロチャンバ105内からより多くの空気を逃がすことができ、より多くのサンプル溶液201を入れることができる。
 また、分離液202を導入するポンプ405は、例えば、シリンジポンプが用いられる。シリンジポンプは、あらかじめ設置されている分離液202を押し出して、分離液202を流路104へと導入する。なお、ポンプ405は、あらかじめ設置されている容器内の分離液202を吸引し、吸引した分離液202を流路104に押し出すものであってもよい。ポンプ405は、加圧して分離液202を流路へ導入するものであってもよく、例えば、定量吐出ポンプ、ダイヤフラムポンプ、ロータリポンプなどであってもよい。分離液202を流路104に導入する際に分離液202を加圧することにより、固相106の抵抗や流路抵抗がある状態でも分離液202を押し入れることが可能となる。
 固相106は、具体的には、多孔質膜が用いられる。例えば、固相106は、PTFE(ポリテトラフルオロエチレン)が用いられる。固相106は、多孔質で通気性を有するため空気を通過させることができ、撥水性を有するためサンプル溶液201を停止させることができ、親油性を有するため分離液202を通過させることができる。また、固相106は、PFA(パーフルオロアルコキシアルキレン)、FEP(フルオロエチレンプロピレン)、ETFE(エチレンテトラフルオロエチレン)などや、フッ素系撥水膜、シリコン系撥水膜、ポリマー系撥水膜、ナノ多孔質膜を用いてもよい。また、固相106は、膜でなくてもよく、疎水性を持つビーズを流路104に詰めたものであってもよい。ビーズは、疎水性を有するため、サンプル溶液201を撥水させて停止させることができる。多孔質の膜として、水との接触角が、例えば、80度以上の疎水性の膜を用いてもよい。また、固相106は、分離液202を通すことができるように、親油性であり、分離液202が染み込み通過できる材質のものを用いる。
 従来は、例えば、非特許文献1に開示されている方法のように、上記した固相106ではなく、バルブを用いていた。非特許文献1では、バルブを閉鎖するとサンプル溶液を停止させることができ、開放すると空気を抜くことや分離液を導入することができる。しかしながら、例えば、当該バルブがチューブをつぶすようなピンチバルブである場合、流路からチューブまでの部分及びチューブにサンプル溶液が通過する体積が必要となる。このチューブに残るサンプル溶液は、計測に用いることができないため、デッドボリュームとなる。このデッドボリュームによって、計測可能な対象DNAが減り、検出感度の低下につながる。一方で、実施例1の固相106を用いることにより、固相106をマイクロチャンバ105の近くに配置することができ、サンプル溶液201のデッドボリュームを削減することができる。
 上記したように、実施例1の固相106を用いることにより、流路104上に固相106を実装することができるため、バルブのようなデッドボリュームを抑制することができる。流路104に残ったサンプル溶液201は、デッドボリュームとなりうるものの、流路104の断面積を小さく設計及び加工することにより、流路104に残ったサンプル溶液201の体積を最小限に抑えることができ、サンプル溶液201のデッドボリュームを低減できる。
 また、実施例1では、サンプル溶液201を流路104に導入する前に、バルブ103を用いる。バルブ103は、真空引きをしている際には閉鎖状態となり、サンプル溶液201を導入する際には開放状態となる。バルブ103には、サンプル溶液201を流路104に導入する際に、一度だけ開放されるワンタイムバルブを使用することもできる。具体的には、例えば、樹脂のフィルムをバルブとして用いて、通常時は閉鎖状態とし、サンプル溶液201を導入するときに当該フィルムを破ることにより、開放状態とする。このようにすることで、低コストかつ小型なデバイスを提供することが可能となる。なお、バルブ103としては、電磁バルブや、チューブを押しつぶすピンチバルブを用いてもよい。
 分離液202としては、サンプル溶液201と混和しない物性を有する液体が用いられる。具体的には、例えば、分離液202は、シリコーンオイル、ミネラルオイル等のオイル、パラフィンワックス、光硬化樹脂などを用いることができる。
 分離液202は、光硬化樹脂であってもよい。光硬化樹脂を用いてサンプル溶液201を分離する場合、光硬化樹脂を液体の状態で流路104に導入する。その後、紫外線などの光照射により、光硬化樹脂を固化させ、サンプル溶液201が収容された複数のマイクロチャンバ105内に分離して密閉することができる。また、分離液は1種類でなく、複数種類を用いてもよい。具体的には、例えば、第二の開口部107より、光硬化樹脂とオイルをこの順に導入する。光硬化樹脂はバルブ103付近まで到達し、流路104はオイルにて満たされる。その後、光硬化樹脂を硬化することにより、流路104の端を光硬化樹脂にて密閉し、各マイクロチャンバ105間はオイルにて分離する構成とすることができる。
 サンプル溶液201は、例えば、計測対象のDNA、ポリメラーゼ、バッファ、プライマー、プローブを含む溶液である。その溶液量は、10μL程度から50μL程度に調整される。
 実施例1では、従来のように流路の両側にバルブを用いる構成と比較し、サンプル溶液201のデッドボリュームを削減することができる。そのため、調整したサンプル溶液201の大部分を計測に使用することができ、高感度な検出が可能となる。
 なお、第一の開口部102にサンプル溶液201を導入する際に、サンプル溶液201と連続してオイルなどの別の液体(分離液)を入れてもよい。このようにすることにより、オイルでサンプル溶液201をマイクロチャンバ105に押し込むことが可能となる。これにより、流路104内をオイルなどの分離液で満たすことができるので、流路104内のサンプル溶液201のデッドボリュームをさらに削減することができる。また、分離液202を導入した際に、第一の開口部102に押し戻される液体がオイル(分離液)となり、蓋の役割を果たすため、サンプル溶液201中の核酸が大気中に拡散することを防ぐことが可能となる。
 このようにして、サンプル溶液201が多数のマイクロチャンバ105に分離して導入されたサンプル溶液分離デバイス101は、PCRのサーマルサイクルが実施される。サーマルサイクルによって、対象DNAは、複数のマイクロチャンバ105内でPCRにより増幅される。サーマルサイクルは、例えば、高温95℃を20秒、低温60℃を40秒のように設定され、高温時に二本鎖が解離され、低温時にアニーリング、エクステンションされて、PCR増幅される。増幅産物を蛍光計測することにより、複数のマイクロチャンバ105に分離導入された対象DNAを検出することができる。
 なお、マイクロチャンバ105の体積は、例えば、数pLから数nL程度であり、マイクロチャンバ105の数は、数千個から数百万個程度である。デジタルPCRでは、多数のマイクロチャンバ105に対象DNAを分割することにより、バックグラウンドとなるDNAの量を減らし、対象DNAを高感度に検出することが可能となる。
 なお、本実施例では、移動機構406(切替手段)を用い、開口部コンタクト部402及び開口部コンタクト部404を切り替えたが、電磁弁などで流路を切り替えてもよい。
 マイクロチャンバ105と流路104とは、メインの流路から分岐した細い接続用流路により接続される。この細い接続用流路により、複数のマイクロチャンバ105間がロバストに分離される。流路104に分離液202が導入される際には、マイクロチャンバ105内に導入されるサンプル溶液201は、メイン流路を流れる。接続用流路は、流路断面積がメイン流路よりも小さい。また、接続用流路の先のマイクロチャンバ105にはサンプル溶液201が満たされているため、分離液202は、接続用流路からマイクロチャンバ105へは入り込まず、メイン流路を流れる。また、サンプル溶液201と分離液202との間には、界面張力が働くため、サンプル溶液201がマイクロチャンバ105内で液滴状になり、分離液202により複数のマイクロチャンバ105間が分離される。このように、メイン流路よりも断面積の小さい接続用流路を複数のマイクロチャンバ105に接続することにより、分離液202は、マイクロチャンバ105内には入らず、流路104を通過し、サンプル溶液201を分離することができる。
(実施例2)
 実施例1では、固相106が、流路104の一か所に配置されている例を説明したが、本発明は、これに限るものではない。図5は、実施例2に係るサンプル溶液分離デバイス501の構成図である。サンプル溶液分離デバイス501は、第一の開口部102、バルブ103、流路104、複数のマイクロチャンバ105、複数の固相506a~506d、及び第二の開口部107を備える。なお、実施例1と同様の説明は適宜省略する。流路104は、多数のマイクロチャンバ105に接続するため分岐される。メイン流路から分岐した複数の分岐流路104a~104dは、多数のマイクロチャンバ105に接続された後に、再度合流し、第二の開口部107に接続される。空気を通過させ、サンプル溶液201を停止させ、且つ分離液202を通過させる固相501a~501dは、マイクロチャンバ105と第二の開口部107の間の複数個所に設置することができる。図5の例では、流路104の複数の分岐流路104a~104dのそれぞれに、固相501a~501dが設けられる。複数の分岐流路104a~104dの個数及び固相501a~501dの個数は、4個に限定されず、2~3個であってもよいし、5個以上であってもよい。
 実施例2のように、実施例1と比較して、固相501a~501dをよりマイクロチャンバ105に接近させて配置することにより、流路104に残るサンプル溶液201を減らすことができ、サンプル溶液201のデッドボリュームを削減することができる。
(実施例3)
 実施例3のサンプル溶液分離デバイス601について、図6A、図6B及び図7を用いて説明する。図6Aは、実施例3に係るサンプル溶液分離デバイス601の上面図であり、図6Bは、実施例3に係るサンプル溶液分離デバイス601の側面図である。図6A及び図6Bに示すように、サンプル溶液分離デバイス601は、第一の開口部602、バルブ603、流路604、複数のマイクロチャンバ605、固相606、及び第二の開口部607を備える。なお、実施例1及び実施例2と同様の説明は適宜省略する。流路604及びマイクロチャンバ605は、基板608に加工される。また、流路604の一方端及び他方端には、基板608を貫通した貫通孔610及び611が形成される。貫通孔610は、バルブ603を通じて第一の開口部602と接続され、貫通孔611は、固相606を通じて第二の開口部607に接続される。また、基板608の複数のマイクロチャンバ605及び流路604を加工した側は、フィルム609により封止される。
 すなわち、実施例3のサンプル溶液分離デバイス601の流路604は、複数のマイクロチャンバ605が接続され、水平方向に延びる水平方向流路604aと、水平方向流路604aに接続され、垂直方向に延びる垂直方向流路604b(貫通孔610及び611)と、を有する。そして、固相606は、垂直方向流路604b(貫通孔611)の上端部に設けられる。
 基板608又はフィルム609の材質は、樹脂が用いられるが、本発明は、これに限るものではない。例えば、基板608又はフィルム609の材質は、自家蛍光の低いCOP(シクロオレフィンポリマー)であってもよいし、COC(シクロオレフィンコポリマー)であってもよい。また、基板608又はフィルム609の材質は、ポリカーボネート、ポリプロピレン、PMMA(メタクリル樹脂)などであってもよい。また、基板608又はフィルム609の一部は、熱伝導性の高いアルミなどの金属、光の反射を抑制できるカーボンなどの材料であってもよい。
 バルブ603は、貫通孔610を塞ぐように配置されるノーマリークローズのシールであって、穴が空くことによって閉鎖状態から開放状態となるワンタイムバルブである。このバルブ603の上部には、サンプル溶液201を受け入れることができる容積を持った第一の開口部602が配置される。また、固相606は、膜状であり、貫通孔611を塞ぐように配置される。固相606の上部には、分離液202を受け入れることができる容積を持つ第二の開口部607が配置される。このような構成にすることにより、基板608上に、バルブ603及び固相606を容易に配置できる。また、固相606を流路604の近くに配置できるため、サンプル溶液291のデッドボリュームを削減することができる。
 図7は、実施例3に係るサンプル溶液分離デバイス601にサンプル溶液201を分離して導入するサンプル溶液分離システム700の構成図である。サンプル溶液分離システム700は、真空ポンプ701、圧力センサ702、フィルタ703、電磁弁704、移動機構705、開口部コンタクト部706、開口部コンタクト部707、電磁弁708、液体ポンプ709、容器710、フィルタ711、圧力センサ712、加圧ポンプ713、バルブ制御機構714、開口部用蓋715、光源717、温調器718、コントローラ719、及びサンプル溶液分離デバイス601を備える。
 サンプル溶液201は、あらかじめ第一の開口部602に充填され、バルブ603の上部に接するように設置されている。このとき、バルブ603は閉鎖状態である。第一の開口部602には、開口部用蓋715が設けられている。開口部用蓋715の上面内側には、先端が尖っており、シール(バルブ603)を破ることができる尖頭部品716が設置されている。開口部用蓋715の上面は、伸縮性のある材質、例えば、エラストマーやゴムでできている。
 サンプル溶液分離デバイス601の第二の開口部607と開口部コンタクト部706とは、移動機構705(切替手段)を制御することにより、接続される。また、電磁弁704は、真空ポンプ701と開口部コンタクト部706とを接続する、又は非接続にする。真空ポンプ701は、サンプル溶液分離デバイス601の流路604及び複数のマイクロチャンバ605を真空引きする。流路604及び複数のマイクロチャンバ605が十分に減圧された後に、バルブ制御機構714(バルブ制御手段)が開口部用蓋715の上部を押すことにより、尖頭部品716は、バルブ603を破る。これにより、バルブ603が開放状態となる。ここで、バルブ603は、上記したように、アルミや樹脂等の薄膜のシールで形成されており、尖頭部品716により破ることができる。流路604及び複数のマイクロチャンバ605が減圧されているため、サンプル溶液201が流路604及び複数のマイクロチャンバ605に引き込まれ、流路604及び複数のマイクロチャンバ605にサンプル溶液201が満たされる。固相606が撥水性を有するため、流路604を通過するサンプル溶液201は、固相606で停止する。また、固相606は通気性を有するため、サンプル溶液201の先端が固相606に到達するまで、流路604及びマイクロチャンバ605内を真空ポンプ701により陰圧化することができ、真空度を上げることができる。
 電磁弁704は、流路604の密閉、真空ポンプ701と開口部コンタクト部706との接続、及び大気開放に用いる。第二の開口部607を大気に開放することにより、サンプル溶液分離デバイス601の流路604は、大気圧に開放される。その後、移動機構705は、開口部コンタクト部707と第二の開口部607とを接続する。電磁弁708は、開口部コンタクト部707と液体ポンプ709とを接続する。液体ポンプ709は、容器710内に予め設置された分離液202を吸引し、開口部コンタクト部707に向けて吐出する。分離液202は、第二の開口部607及び固相606を通じて、流路604へと導入される。
 また、電磁弁708は、第二の開口部607との接続先を、液体ポンプ709から加圧ポンプ713に切り替える。加圧ポンプ713は、分離液202を加圧し、流路604内に押し込む。加圧ポンプ713は、流路604の流路抵抗や固相606を通過する際の抵抗を超える圧力で加圧することにより、分離液202を流路604に導入する。加圧する際の圧力は、分離液202、流路604のサイズ、固相606の性質等により決まり、例えば、10kPaから200kPa程度である。流路604にあるサンプル溶液201は、第一の開口部602側へと押し戻され、流路604が分離液202で満たされる。このようにして、サンプル溶液201が各マイクロチャンバ605内に残り、分離液202により分離される。また、加圧して分離液202を流路604に導入することにより、何等かの理由により残存した気泡の膨張を抑制して分離液202を流路604に導入することができる。
 また、分離液202は、光硬化樹脂であってもよい。サンプル溶液201と混和しない光硬化樹脂を用いることにより、サンプル溶液201を各マイクロチャンバ605内に分離することができる。この場合、液体状態の光硬化樹脂を流路604に導入した後に、光源717を用いて、光硬化樹脂を硬化させる。これにより、サンプル溶液201をマイクロチャンバ605内に密閉することができる。
 また、サンプル溶液201や分離液202を導入する際に、温調器718を用いて、サンプル溶液分離デバイス601の温度を制御することも可能である。一般に、サンプル溶液201や分離液202は、高温にすると粘性が低くなる。従って、室温と比べて加温することにより、サンプル溶液201や分離液202の粘性が下がり、サンプル溶液201や分離液202は、流路604やマイクロチャンバ605に入りやすくなる。これにより、溶液(サンプル溶液201及び分離液202)導入時間の短縮が可能となり、且つ全てのマイクロチャンバ605にロバストにサンプル溶液201を入れることが可能となる。温調器718による制御は、例えば、35℃~70℃程度に加温することが好適である。
 また、真空ポンプ701で真空引きをしている際の圧力は、圧力センサ702でモニタして制御を行う。加圧ポンプ713で加圧している際の圧力は、圧力センサ712でモニタして制御を行う。また、フィルタ703及びフィルタ711は、サンプル溶液201や分離液202にゴミ等が混入するのを防ぐ。
 上記した一連の動作の制御は、コントローラ719により行われる。コントローラ719は、プロセッサ720、メモリ721、及びインターフェース722を有する。プロセッサ720は、メモリ721上で各種プログラムを実行し、インターフェース722を介して上記した動作の制御を行うための制御信号を出力する。
 なお、バルブ制御機構714は、例えば、ソレノイドであり、電流が流れることにより移動し、バルブ603の開放を行う。また、移動機構705は、例えば、水平方向と垂直方向の2軸のモータ駆動機構である。
 通気性、疎水性、親油性を持つ固相606を用いることにより、バルブを用いることなく、流路604の真空引き、サンプル溶液201の停止、分離液202の通過を制御することが可能となる。また、固相606をマイクロチャンバ605の近くに配置することが可能となり、サンプル溶液201を無駄にすることなく、サンプル溶液201をマイクロチャンバ605に分割して入れることができる。
 また、バルブ603として、薄膜に穴をあける構成のワンタイムバルブを用いることにより、シンプルな構成のバルブを実装できる。バルブ603には、サンプル溶液201が接するため、キャリーオーバーを防ぐためには、使い捨てにする必要があるため、ワンタイムバルブが有用である。また、バルブ603は、シンプルな構成のバルブであるので、低コストでバルブを提供することができる。また、分離液202として光硬化樹脂を用いて、液状で流路604に入れた後に、固化することにより、マイクロチャンバ605を密閉できる。これにより、第一の開口部602及び第二の開口部607を閉じる必要もなくなり、構成を簡略化することが可能となる。
 なお、バルブ603は、ピペットチップで穴をあけてもよい。実施例3に係るサンプル溶液分離デバイス601は、核酸の抽出、試薬の混合、分注などを行うサンプル前処理と接続するような自動化システムにも適用可能である。例えば、採血管に採取された検体を遠心分離して血漿を取り出し、血漿を精製することにより血漿内の核酸を抽出する。その核酸を試薬と混合し、デジタルPCRにより対象の核酸を検出する。この際、混合したサンプル溶液をピペットチップにより吸引して吐出する分注システムを用い、第一の開口部602にサンプル溶液201を注入する。その際に、ピペットチップの先端でバルブを破り、サンプル溶液201が流路に導入されるようにしてもよい。
 また、第一の開口部602から、サンプル溶液201と連続してオイル(分離液)を導入することにより、サンプル溶液201のデッドボリュームをより削減することができる。具体的には、陰圧によりサンプル溶液201がマイクロチャンバ605に導入された後、連続してオイル(分離液)が流路604内に入るため、流路604内に残存するサンプル溶液201を減らすことができる。この状態で第二の開口部607から分離液202をサンプル溶液201とは逆向きに入れることにより、流路604内のサンプル溶液201を完全に分離液202に置き換え、複数のマイクロチャンバ605間を分離することができる。また、このサンプル溶液201と連続して導入したオイル(分離液)は、サンプル溶液201内の核酸がエアロゾルとなり大気中に放出されることを防ぐ蓋の役割を持つ。このようにすることにより、第一の開口部602からサンプル溶液201のみを導入する場合と比べて、よりサンプル溶液201のデッドボリュームをより削減することができる。
 なお、本実施例では、容器710内の分離液202を第二の開口部607を通じて流路604内に入れる説明をしたが、本発明はこれに限るものではない。例えば、分離液202があらかじめサンプル溶液分離デバイス601内に充填されており、このサンプル溶液分離デバイス601内に充填された分離液202を流路604内に導入してもよい。
(実施例4)
 上記した実施例3では、基板608の貫通孔611の上端部に薄膜の固相606を設置したが、本発明はこれに限るものではない。図8は、実施例4に係るサンプル溶液分離デバイス801の構成図である。実施例4に係るサンプル溶液分離デバイス801の固相806は、通気性、疎水性、親油性を有する複数の微粒子である。複数の微粒子は、貫通孔611(垂直方向流路)に詰められて、固相806を形成する。微粒子は、例えば、0.1μmから10μmのサイズを持つビーズである。微粒子は、膜よりも流路604内へと導入しやすい。また、微粒子の間を空気が通過することができるため、真空引きが可能となる。また、微粒子は、撥水性を有するため、サンプル溶液201を停止することができる。また、微粒子は、親油性を有するため、分離液202を通過することができる。
 また、実施例3では、バルブ603がワンタイムバルブであるとしたが、本発明はこれに限るものではない。例えば、実施例4に係るサンプル溶液分離デバイス800のバルブ803は、伸縮性のあるチューブであってもよい。チューブは、基板608の貫通孔610と接続するために延ばされ、バルブ803を形成する。例えば、バルブ803は、シリコーンチューブである。シリコーンチューブは、圧縮されることにより、チューブ内のフローを停止するピンチバルブを形成する。ピンチバルブを用いることにより、開閉を複数回行うことができる。真空引きする際にはバルブ803を閉じ、サンプル溶液201を入れる際にはバルブ803を開ける。また、分離液202の導入後は、バルブ803を閉じて、サンプル溶液201及び分離液202を密閉する。
(実施例5)
 本発明のサンプル溶液分離デバイスは、デジタルPCRに用いることができる。図9は、実施例5に係るデジタルPCRシステム900の構成図である。デジタルPCRシステム900(サンプル溶液分離システム)は、サンプル溶液分離デバイス601、バルブ制御機構901、ポンプ902、液体ポンプ903、温調器913、光学系914(計測手段)、及び解析部912を備える。光学系914は、光源904、レンズ905、レンズ908、レンズ910、バンドパスフィルタ906、バンドパスフィルタ909、ダイクロイックミラー907、及びCMOSセンサ911を有する。
 サンプル溶液分離デバイス601には、第一の開口部602からサンプル溶液201がセットされる。ポンプ902の駆動により、流路604及びマイクロチャンバ605内の空気が第二の開口部607を通じて脱気され、陰圧化される。バルブ制御機構901は、バルブ603を開放し、サンプル溶液201が流路604及びマイクロチャンバ605内に導入される。その後、液体ポンプ903の駆動により、分離液202が第二の開口部607経由で流路604に導入される。このようにすることにより、サンプル溶液201を複数のマイクロチャンバ605内に分離して導入できる。
 ここで、サンプル溶液201は、検出対象であるDNA、ポリメラーゼ、バッファ、プライマー、プローブを含むPCR反応液である。PCR反応液であるサンプル溶液201が多数のマイクロチャンバ605に分離して入れられたデバイスに対し、サーマルサイクル処理が行われる。サーマルサイクル処理では、温調器913がディネーチャ、アニーリング、エクステンションが行われる温度帯に制御される。温調器913は、ヒーター、ペルチェ素子等から構成されるが、本発明はこれに限るものではない。サーマルサイクル処理により、計測対象のDNAがマイクロチャンバ605内に存在する場合、対象DNAが増幅される。
 光学系914(計測手段)は、PCR増幅がなされたサンプル溶液201内の対象の核酸を計測する。光源904から照射された光は、レンズ905によってコリメートされ、バンドパスフィルタ906で所定の波長の光を透過させ、ダイクロイックミラー907で反射され、レンズ908を通過してサンプル溶液分離デバイス601に照射される。サンプル溶液分離デバイス601のマイクロチャンバ605からの蛍光は、レンズ908を通り、ダイクロイックミラー907、バンドパスフィルタ909、レンズ910を通過して、CMOSセンサ911にて撮像される。解析部912は、CMOSセンサ911で撮像された画像に基づいて解析を実施し、各マイクロチャンバ内の対象DNAを検出する。
 図10は、実施例5に係るデジタルPCRシステム900の動作フロー図である。
 バルブ制御機構901は、バルブ603を開放し、第一の開口部602にセットされたサンプル溶液201をサンプル溶液分離デバイス601に導入する(S1001)。
 液体ポンプ903は、分離液202を第二の開口部607からサンプル溶液分離デバイス601に導入する(S1002)。これにより、サンプル溶液201が複数のマイクロチャンバ605に分離して導入される。
 次に、温調器913は、サンプル溶液分離デバイス601をサーマルサイクルにかける(S1003)。
 光学系914は、サンプル溶液分離デバイス601に光を照射し、各マイクロチャンバ605の蛍光強度を計測する(S1004)。
 そして、解析部915は、光学系914によって計測された蛍光強度を解析して、サンプル溶液201内のターゲットDNAを検出する(S1005)。
 なお、サンプル溶液201や分離液202を導入する際に、サンプル溶液201や分離液202を加温してもよい。サンプル溶液201や分離液202の粘性が変わり、流路604への導入が容易になる。また、サンプル溶液201や分離液202を導入する時間を短縮することができる。また、サンプル溶液201や分離液202を加温する温調器は、サーマルサイクルを実施する温調器913であってもよいし、温調器913とは別の温調器であってもよい。
 PCR反応液として、フォワード側のプライマーとリバース側のプライマーの濃度を変えて、非対称PCRを行い、増幅産物の内、片方の一本鎖DNAが多く増幅されるようにしてもよい。このようにして、分子ビーコンにより一本鎖DNAを検出する。また、蛍光計測時にサンプル溶液分離デバイスの温度を制御し、融解曲線解析を行う。蛍光計測は、複数のフィルタを用い、複数の色で計測する。各マイクロチャンバの蛍光の色、蛍光強度と融解温度を用いて、対象のDNAを特定することにより、高マルチプレックス、かつ、高感度な計測が実現できる。
(サンプル溶液分離方法)
 ここで、サンプル溶液201を複数のマイクロチャンバ105に分離する方法を説明する。サンプル溶液分離方法は、
 上記したサンプル溶液分離デバイス101を準備こと、
 固相106を介して、複数のマイクロチャンバ105及び流路104内の空気を脱気すること、
 バルブ103を開放し、第一の開口部102からサンプル溶液201を複数のマイクロチャンバ105及び流路104に導入すること、及び
 第二の開口部107から固相106を介して分離液202を流路104に導入すること、を有する。
 なお、準備するサンプル溶液分離デバイス101は、サンプル溶液分離デバイス501、601又は801であってもよい。
 また、サンプル溶液分離方法は、
 複数のマイクロチャンバ105にサンプル溶液201が分離されたサンプル溶液分離デバイス101に対してPCR(ポリメラーゼ連鎖反応)のサーマルサイクルを実施すること、
 複数のマイクロチャンバ105の蛍光強度を計測すること、及び
 蛍光強度を解析し、サンプル溶液201内のターゲットDNAを検出すること、をさらに有する。
 なお、上記した第一の開口部102からサンプル溶液201を複数のマイクロチャンバ105及び流路104に導入することは、第一の開口部102からサンプル溶液201、及びサンプル溶液201に続くオイル(分離液)を導入することを含む。
(変形例)
 本開示は、前述した実施形態に限定されるものではなく、様々な変形例が含まれる。例えば、前述した実施形態は本開示を分かりやすく説明するために詳細に説明したものであり、必ずしも説明した全ての構成を備えるものに限定されるものではない。また、ある実施形態の構成の一部を他の実施形態の構成に置き換えることが可能であり、また、ある実施形態の構成に他の実施形態の構成を加えることも可能である。また、各実施形態の構成の一部について、他の構成の追加・削除・置換をすることが可能である。
 例えば、上記した実施例では、複数のマイクロチャンバ105間を分離するために分離液を用いたが、本発明はこれに限らず、分離気体であってもよい。この場合、固相は、分離気体を通過させる物性をもっていればよい。

Claims (16)

  1.  サンプル溶液を分離するサンプル溶液分離デバイスであって、
     複数のマイクロチャンバと、
     前記複数のマイクロチャンバを接続する流路と、
     前記サンプル溶液の前記流路への導入口である第一の開口部と、
     前記第一の開口部と前記複数のマイクロチャンバとの間に設けられるバルブと、
     前記サンプル溶液が収容される前記複数のマイクロチャンバ間を分離する分離液の流路への導入口であって、前記複数のマイクロチャンバを挟んで前記第一の開口部の反対側に設けられる第二の開口部と、
     前記第二の開口部と前記複数のマイクロチャンバとの間に設けられる固相と、を備え、
     前記固相は、通気性を有し、前記サンプル溶液に対して撥水性を有し、且つ前記分離液に対して通過性を有する
     ことを特徴とするサンプル溶液分離デバイス。
  2.  前記流路は、前記マイクロチャンバがそれぞれ接続される複数の分岐流路を有し、
     前記固相は、前記複数の分岐流路のそれぞれに設けられる
     ことを特徴とする請求項1に記載のサンプル溶液分離デバイス。
  3.  前記流路は、
     前記複数のマイクロチャンバが接続され、水平方向に延びる水平方向流路と、
     前記水平方向流路に接続され、垂直方向に延びる垂直方向流路と、を有し、
     前記固相は、前記垂直方向流路の上端部に設けられる
     ことを特徴とする請求項1に記載のサンプル溶液分離デバイス。
  4.  前記流路は、
     前記複数のマイクロチャンバが接続され、水平方向に延びる水平方向流路と、
     前記水平方向流路に接続され、垂直方向に延びる垂直方向流路と、を有し、
     前記固相は、前記垂直方向流路内に設けられた複数の微粒子である
     ことを特徴とする請求項1に記載のサンプル溶液分離デバイス。
  5.  前記バルブは、前記サンプル溶液を前記流路に導入する際に1度だけ開放されるワンタイムバルブである
     ことを特徴とする請求項1に記載のサンプル溶液分離デバイス。
  6.  前記固相は、前記サンプル溶液に対して撥水性を有し、且つ前記分離液に対して通過性を有する多孔質膜である
     ことを特徴とする請求項1に記載にサンプル溶液分離デバイス。
  7.  前記多孔質膜の材料は、PTFE(ポリテトラフルオロエチレン)である
     ことを特徴とする請求項6に記載のサンプル溶液分離デバイス。
  8.  前記分離液は、光硬化樹脂であって、
     前記分離液は、前記流路内で光照射によって硬化する
     ことを特徴とする請求項1に記載のサンプル溶液分離デバイス。
  9.  前記分離液は、前記サンプル溶液と混和しない物性を有するオイルである
     ことを特徴する請求項1に記載のサンプル溶液分離デバイス。
  10.  サンプル溶液を分離するサンプル溶液分離デバイスであって、複数のマイクロチャンバと、前記複数のマイクロチャンバを接続する流路と、前記サンプル溶液の前記流路への導入口である第一の開口部と、前記第一の開口部と前記複数のマイクロチャンバとの間に設けられるバルブと、前記サンプル溶液が収容される前記複数のマイクロチャンバ間を分離する分離液の流路への導入口であって、前記複数のマイクロチャンバを挟んで前記第一の開口部の反対側に設けられる第二の開口部と、前記第二の開口部と前記複数のマイクロチャンバとの間に設けられる固相と、を備え、前記固相は、通気性を有し、前記サンプル溶液に対して撥水性を有し、且つ前記分離液に対して通過性を有する、サンプル溶液分離デバイス、
     前記複数のマイクロチャンバ及び前記流路内の空気を前記第二の開口部から前記固相を通じて脱気するポンプ、
     前記バルブを開放するバルブ制御手段、及び
     前記第二の開口部から分離液を導入する分離液導入手段、を有する
     ことを特徴とするサンプル溶液分離システム。
  11.  前記第二の開口部の接続先を前記ポンプ又は前記分離液導入手段に切り替える切替手段、をさらに有する
     ことを特徴とする請求項10に記載のサンプル溶液分離システム。
  12.  前記分離液を加圧して、前記流路に導入する加圧ポンプ、をさらに有する
     ことを特徴とする請求項10に記載のサンプル溶液分離システム。
  13.  前記マイクロチャンバ内でPCR(ポリメラーゼ連鎖反応)が実施された前記サンプル溶液内の対象の核酸を計測する計測手段、をさらに有する
     ことを特徴とする請求項10に記載のサンプル溶液分離システム。
  14.  サンプル溶液を分離するサンプル溶液分離デバイスであって、複数のマイクロチャンバと、前記複数のマイクロチャンバを接続する流路と、前記サンプル溶液の前記流路への導入口である第一の開口部と、前記第一の開口部と前記複数のマイクロチャンバとの間に設けられるバルブと、前記サンプル溶液が収容される前記複数のマイクロチャンバ間を分離する分離液の流路への導入口であって、前記複数のマイクロチャンバを挟んで前記第一の開口部の反対側に設けられる第二の開口部と、前記第二の開口部と前記複数のマイクロチャンバとの間に設けられる固相と、を備え、前記固相は、通気性を有し、前記サンプル溶液に対して撥水性を有し、且つ前記分離液に対して通過性を有する、サンプル溶液分離デバイス、を準備すること、
     前記固相を介して、前記複数のマイクロチャンバ及び前記流路内の空気を脱気すること、
     前記バルブを開放し、前記第一の開口部から前記サンプル溶液を前記複数のマイクロチャンバ及び前記流路に導入すること、及び
     前記第二の開口部から前記固相を介して前記分離液を流路に導入すること、を有する
     ことを特徴とするサンプル溶液分離方法。
  15.  前記複数のマイクロチャンバに前記サンプル溶液が分離された前記サンプル溶液分離デバイスに対してPCR(ポリメラーゼ連鎖反応)のサーマルサイクルを実施すること、
     前記複数のマイクロチャンバの蛍光強度を計測すること、及び
     前記蛍光強度を解析し、前記サンプル溶液内のターゲットDNAを検出すること、をさらに有する
     ことを特徴とする請求項14に記載のサンプル溶液分離方法。
  16.  前記第一の開口部から前記サンプル溶液を前記複数のマイクロチャンバ及び前記流路に導入することは、前記第一の開口部から前記サンプル溶液、及び前記サンプル溶液に続く分離液を導入することを含む
