Abstract
DPFではない環境下で飼育された母豚から、DPFである豚臓器を取得することができる移植材の製造方法を提供すること。 非DPF(Designated Pathogen Free)環境下で飼育された母豚から子宮を摘出すること、 摘出した前記子宮をデコンタミネーション処理後にアイソレータ内の無菌環境に収容すること、 前記子宮からの胎仔又は子豚の取出しをアイソレータ内の無菌環境で行うこと、 任意で前記胎仔又は子豚の蘇生及び飼育を含み、かつ、前記蘇生及び飼育をアイソレータ内の無菌環境で行い、及び 前記胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することを含む、前記摘出した臓器がDPFである、豚臓器又はその由来物を含む移植材の製造方法。
Description
本発明は、臓器移植等の医療用として好適な豚臓器又はその由来物を含む移植材の製造方法に関する。
豚の膵島を糖尿病患者への移植等の異種移植療法の臨床応用実現には、医療に使用し得る衛生レベルの豚(医療グレード豚)が不可欠である。異種移植に用いる豚の衛生レベルは、指定病原体フリー(designated pathogen free:DPF)であることが求められている(非特許文献1~3)。
例えば、カナダ、ヨーロッパ諸国、ニュージーランド等における異種移植の研究開発例では、臓器ドナー豚の飼育のために高額な施設を建築し、高い衛生条件を達成している。
しかしながら、上記医療グレードのDPFの衛生レベルを達成するためには、外界から隔離を行うための特殊で高度な環境を維持するために煩雑で大がかりな施設の建築を要する。このような特殊で高度な環境の維持は煩雑で莫大なコストが発生するという欠点を有しており、さらに一度ある施設を建設すると、建設された施設は、厳重に規定された条件を満たす必要があり特殊な構造をしていることから、増床等の改装は困難という問題もある。
しかしながら、上記医療グレードのDPFの衛生レベルを達成するためには、外界から隔離を行うための特殊で高度な環境を維持するために煩雑で大がかりな施設の建築を要する。このような特殊で高度な環境の維持は煩雑で莫大なコストが発生するという欠点を有しており、さらに一度ある施設を建設すると、建設された施設は、厳重に規定された条件を満たす必要があり特殊な構造をしていることから、増床等の改装は困難という問題もある。

例えば、異種移植用の医療グレードのDPF臓器の従来法による調製では、母豚の飼育、帝王切開、豚胎仔飼育、臓器摘出の全工程をDPF環境下で行わざるを得ない。特に、大きく育って飼育に広大な床面積を要する母豚の飼育を、隔離を行う等のDPF環境下で飼育することは、特殊で高度な環境を維持するための煩雑で大がかりな施設の建築を要し、莫大なコストが発生するという問題があった。
本発明は、このような従来技術の問題に鑑みてなされたものであり、DPFではない環境下で飼育された母豚から、DPFである豚臓器を取得することができる移植材の製造方法を提供することを目的とする。
本発明者らは、上記課題について鋭意検討した結果、胎仔又は子豚の出生に際し、母豚から胎仔又は子豚への感染は、経胎盤感染以外の感染は発生しないこと、及び
アイソレータ内の無菌環境で上記子宮から胎仔又は子豚の取り出しを行い、かつ当該胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することにより、DPFではない環境下で飼育された母豚から、DPFの豚臓器を取得することができることを見出した。本発明は、上記知見に基づき完成されるに至ったものである。
すなわち本発明は以下の通りである。
アイソレータ内の無菌環境で上記子宮から胎仔又は子豚の取り出しを行い、かつ当該胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することにより、DPFではない環境下で飼育された母豚から、DPFの豚臓器を取得することができることを見出した。本発明は、上記知見に基づき完成されるに至ったものである。
すなわち本発明は以下の通りである。
<1>非DPF環境下で飼育された母豚から子宮を摘出すること、
摘出した前記子宮をデコンタミネーション処理後にアイソレータ内の無菌環境に収容すること、
前記子宮からの胎仔又は子豚の取出しをアイソレータ内の無菌環境で行うこと、
任意で前記胎仔又は子豚の蘇生及び飼育を含み、かつ、前記蘇生及び飼育をアイソレータ内の無菌環境で行い、及び
前記胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することを含む、前記摘出した臓器がDPFである、豚臓器又はその由来物を含む移植材の製造方法。
<2>前記非DPF環境がSPF(Specific Pathogen Free)環境である、<1>に記載の方法。
<3>臓器の前記摘出が、前記子豚の躯体中の前記臓器に生理学的に許容される液を注入して前記臓器を膨張させた状態で行われる、<1>に記載の方法。
<4>子宮の前記摘出が、前記母豚への麻酔後に行われ、子宮の前記摘出が、前記麻酔開始後25分以内に終わる、<1>に記載の方法。
<5>前記麻酔が吸入麻酔である、<4>に記載の方法。
<6>前記母豚は、経胎盤感染性病原体による感染が陰性であり、かつそれ以外の病原体による感染が陽性又は陰性である、<1>に記載の方法。
<7>前記取出し及び蘇生と、前記飼育と、前記摘出とが、それぞれ、別個のアイソレータ内の無菌環境で行われ、
アイソレータ同士を連結したスリーブ内の無菌環境空間を通じて、子豚をアイソレータ間で移動させる、<1>に記載の方法。
<8>前記飼育のためのアイソレータが2以上存在し、前記飼育のための前記アイソレータを高さ方向に重ねて配置する、<7>に記載の方法。

本発明によれば、DPFではない環境下で飼育された母豚から、DPFである豚臓器を取得することができる移植材の製造方法を提供することができる。
また、本発明によれば、母豚のDPF施設内での飼育は要しないことから、量産化にはアイソレータの増設のみで足り、低コストでDPFの豚臓器又はその由来物を含む移植材を量産し得る。

以下、本発明の実施態様について詳細に説明するが、本発明は、以下の実施態様に何ら限定されるものではなく、本発明の目的の範囲内において、適宜変更を加えて実施することができる。
≪豚臓器を含む移植材の製造方法≫
本発明の豚臓器又はその由来物を含む移植材の製造方法は、指定病原体フリーではない環境下で飼育された母豚から子宮を摘出すること、
摘出した上記子宮をデコンタミネーション処理後にアイソレータ内の無菌環境に収容すること、
上記子宮からの胎仔又は子豚の取出しをアイソレータ内の無菌環境で行うこと、
任意で前記胎仔又は子豚の蘇生及び飼育を含み、かつ、前記蘇生及び飼育をアイソレータ内の無菌環境で行い、及び
前記胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することを含み、前記摘出した臓器がDPFである。
胎仔又は子豚の出生に際し、母豚から胎仔又は子豚への感染は、経胎盤感染以外の感染は発生しないことから、本発明の製造方法によれば、DPFではない環境下で飼育された母豚から、アイソレータ内の無菌環境で上記子宮から胎仔又は子豚の取り出しを行い、かつ当該胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することにより、DPFの豚臓器又はその由来物を取得することができる移植材の製造方法を提供することができる。
本発明の豚臓器又はその由来物を含む移植材の製造方法は、指定病原体フリーではない環境下で飼育された母豚から子宮を摘出すること、
摘出した上記子宮をデコンタミネーション処理後にアイソレータ内の無菌環境に収容すること、
上記子宮からの胎仔又は子豚の取出しをアイソレータ内の無菌環境で行うこと、
任意で前記胎仔又は子豚の蘇生及び飼育を含み、かつ、前記蘇生及び飼育をアイソレータ内の無菌環境で行い、及び
前記胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することを含み、前記摘出した臓器がDPFである。
胎仔又は子豚の出生に際し、母豚から胎仔又は子豚への感染は、経胎盤感染以外の感染は発生しないことから、本発明の製造方法によれば、DPFではない環境下で飼育された母豚から、アイソレータ内の無菌環境で上記子宮から胎仔又は子豚の取り出しを行い、かつ当該胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することにより、DPFの豚臓器又はその由来物を取得することができる移植材の製造方法を提供することができる。
日本における上記指定病原体としては以下が挙げられる。

以下、図1~5を参照して本発明の豚臓器又はその由来物を含む移植材の製造方法について説明する。
<アイソレータ>
(筐体)
図1~5に示したように、アイソレーター1は、外界(アイソレータ1の外部)からの汚染を防止するために、内部を無菌環境とされて外界と隔離された空間を創製し得る構造を有する筐体2を含む。
本明細書において、「隔離」とは、ある2つの空間があったとき、その一方の空間と、他方の空間との間で、ウイルス、細菌等の微生物、可視可能な物質及び非揮発性の化学物質の行き来ができないことをいう。隔離は、任意の空間(たとえば、外界)に対して行われる。
上記筐体2の材質としては、特に制限はないが、プラスチック(軟質プラスチック(軟質ポリ塩化ビニルなど)、硬質プラスチック)、金属(例えば、ステンレス、鉄等)などが挙げられる。
(筐体)
図1~5に示したように、アイソレーター1は、外界(アイソレータ1の外部)からの汚染を防止するために、内部を無菌環境とされて外界と隔離された空間を創製し得る構造を有する筐体2を含む。

(吸気部3、排気部4及び開閉部5)
図1に示したように、上記アイソレータ1は、無菌空気(例えば、後述するフィルターを通って微生物、ウイルス等が除去された無菌空気)を送り込む後述するフィルターを具備する吸気部3及び内部空気を送り出す排気部4、並びに、搬入及び搬出を行うための1又は2以上の開放及び密閉が可能な開閉部5(好ましくは2以上の開閉部)を有し得る。
上記アイソレータ1において、開閉部5を密閉した状態では、無菌空気を送り込む上記吸気部3及び内部空気を送り出す上記排気部4を除いて、気密性が確保されることが好ましい。
これにより、上記アイソレータ1の外部が、指定病原体フリーではない環境であっても外気が上記アイソレータ1内に流入することによる微生物、ウイルスの混入を防ぐことができる。また、制御されない上記アイソレータ1内の内部空気の外部への流出による、内部気圧の予想外の圧力低下、気密性低下に起因する、その後の外気の流入を防ぐことができる。
本明細書において「気密性」とは、区切られたある空間があったとき、その空間(アイソレータ内部)が、その空間の外側(アイソレータ外部)に対して気体が行き来することができない状態又は性質ということができる。ここでいう空間とは、1つのアイソレータの内部空間に限定されず、後述するように、複数のアイソレータを(例えば、開閉部5と着脱可能なスリーブ7を介して)連結して各アイソレータの内部空間を連通した場合、連通された内部空間に対して、外部空間から気体が行き来することができない状態又は性質も包含される。
上記気密性は、例えばマルチ環境測定機器(testo-435)にて連続測定したときに空気漏洩がないこと等により確認することができる。
図1に示したように、上記アイソレータ1は、無菌空気(例えば、後述するフィルターを通って微生物、ウイルス等が除去された無菌空気)を送り込む後述するフィルターを具備する吸気部3及び内部空気を送り出す排気部4、並びに、搬入及び搬出を行うための1又は2以上の開放及び密閉が可能な開閉部5(好ましくは2以上の開閉部)を有し得る。
上記アイソレータ1において、開閉部5を密閉した状態では、無菌空気を送り込む上記吸気部3及び内部空気を送り出す上記排気部4を除いて、気密性が確保されることが好ましい。
これにより、上記アイソレータ1の外部が、指定病原体フリーではない環境であっても外気が上記アイソレータ1内に流入することによる微生物、ウイルスの混入を防ぐことができる。また、制御されない上記アイソレータ1内の内部空気の外部への流出による、内部気圧の予想外の圧力低下、気密性低下に起因する、その後の外気の流入を防ぐことができる。

本明細書において「フィルター」とは、病原体(例えば、細菌、ウィルスなどの微生物、好ましくは指定病原体)、塵などを通さず、気体のみを通すようなものをいう。そのようなフィルターとしては、HEPAフィルター(High Efficiency Particulate Air)、ULPA(Ultra Low Penetration Air)フィルター、高通気性除塵フィルター(三菱製紙のEPシリーズ)等が挙げられ、HEPAフィルターは、セパレーター形であってもノンセパレ一ター形であってもよい。上記フィルターとしては、微生物の行き来を遮断することができるフィルターが好ましく、また、滅菌状態を保持することができるという機能を有するフィルターが好ましい。HEPAフィルター又はULPAフィルターがより好ましい。
上記吸気部3及び排気部4は、単なる円状のような開口部であってもよく、吸気部3及び排気部4に管が接続されたものでもよい。吸気部3及び排気部4にはまた、弁が備えられていてもよい。吸気部3及び排気部4は、上記筐体2と気密可能に連結され得る構造を有することが好ましい。弁は、必要に応じて上記上記吸気部3及び排気部4に取り付けられ、気体などの出入りを調節する機能を付与する。
本明細書において「弁」とは、管などを流れる気体又は液体の出入りあるいは流れの方向を調節する装置をいう。

上記アイソレータ1内の無菌環境としては、例えば、国際標準ISO基準14644-1(JIS準拠)でISOクラス5(クラス100)以上の清浄度の環境、EU-GMPガイドライン(EUの医薬品の製造管理および品質管理に関する基準)でグレードA以上の清浄度の環境が挙げられ、ISOクラス4(クラス10)以上の清浄度の環境が好ましく、ISOクラス3(クラス1)以上の清浄度の環境がより好ましい。
本明細書において「清浄度」とは、ある空間(例えば、アイソレータの内部)の清浄のレベルをいう。そのような清浄度としては、例えば、豚にとっての病原体(例えば、特定のウィルス、細菌など)を含まない状態が挙げられ、指定病原体を含まない状態が好ましい。清浄度の設定は、上述したフィルターを使用することによって行うことができる。
本明細書において「清浄度」とは、ある空間(例えば、アイソレータの内部)の清浄のレベルをいう。そのような清浄度としては、例えば、豚にとっての病原体(例えば、特定のウィルス、細菌など)を含まない状態が挙げられ、指定病原体を含まない状態が好ましい。清浄度の設定は、上述したフィルターを使用することによって行うことができる。
上述のように、上記アイソレータ1は、搬入及び搬出を行う1又は2以上の開閉部5(好ましくは2以上の開閉部5)を有し得る。
搬入及び搬出を行う開閉部5は、後述する消毒槽(除染槽)6との連結部(好ましくは着脱可能な連結部)を兼ねることができ、また、後述するようにアイソレータ1が複数存在する場合には、各アイソレータ同士の連結部(好ましくは着脱可能な連結部)を兼ねることができる。
図1~4に示したように、各アイソレータの開閉部5は、外界からの隔離性及び/又は気密性を維持しつつ連結することができ、かつ開閉部5と着脱可能なスリーブ(連結器)7を介して他のアイソレータの開閉部と着脱可能に連結可能であることが好ましい。上記スリーブ7は各アイソレータの内部空間同士を着脱可能に連結可能である。上記スリーブ7はパスボックスの形態を兼ねていてもよい。
上記連結することにより、各アイソレータ間を成豚、胎仔又は子豚8を移動させることができる。
一方、取り外す(脱する)ことにより、各アイソレータ毎に各アイソレータの外部環境(例えば、非DPF環境又はDPF環境)を独立して選択することができ、また、各アイソレータの設置場所も各アイソレータ毎に独立して選択することができる。
上記アイソレータ内に外気が流入した際に、微生物、ウイルス等の病原体の混入を防ぐ観点から、各アイソレータ同士の取り外し(脱)、又は、上記スリーブ7の取り外し(脱)は、DPF環境下で行うことが好ましい。
本明細書において「連結可能」とは、ある2つの部分が連結され得る構造を有する場合、必要に応じてその2つの部分(例えば、開閉部5)を連結することができることをいい、隔離性及び/又は気密性を維持しつつ連結可能であることが好ましい。
上記アイソレータには、隔離性及び/又は気密性を保つためにパッキンが取り付けられ得る。そのようなパッキンは、例えば、各部分の接続部分に取り付け得るし、消毒槽(除染槽)との連結部、各アイソレータ1同士の着脱可能な連結部、上記スリーブ7との着脱可能な連結部等にも取り付け得る。
搬入及び搬出を行う開閉部5は、後述する消毒槽(除染槽)6との連結部(好ましくは着脱可能な連結部)を兼ねることができ、また、後述するようにアイソレータ1が複数存在する場合には、各アイソレータ同士の連結部(好ましくは着脱可能な連結部)を兼ねることができる。
図1~4に示したように、各アイソレータの開閉部5は、外界からの隔離性及び/又は気密性を維持しつつ連結することができ、かつ開閉部5と着脱可能なスリーブ(連結器)7を介して他のアイソレータの開閉部と着脱可能に連結可能であることが好ましい。上記スリーブ7は各アイソレータの内部空間同士を着脱可能に連結可能である。上記スリーブ7はパスボックスの形態を兼ねていてもよい。

上記開閉部5の形状としては特に制限はないが、上記アイソレータ1の使用態様に応じて、例えば、四角形、長方形、正方形、円形(例えば略円形又は正円形)、楕円形等とすることができ、管状のものが付随していてもよく、ある2つの空間(例えば、アイソレータ1の内部)を連結する機能を有する形状であることが好ましい。上記開閉部5は、それ自体が開放可能及び密閉可能であってもよく、別の手段(例えば、弁)を用いることによって開放可能及び密閉可能であってもよい。弁は、必要に応じて上記開閉部5に取り付けられ、気体などの出入りを調節する機能を付与する。
例えば、消毒槽(除染槽)6との連結部、各アイソレータ1同士の着脱可能な連結部、上記スリーブ7との着脱可能な連結部等との連結を解除する場合に、上記アイソレータ1の内部と外界との隔離を保つことを可能にする弁を備えることができる。このような弁は当該分野において周知である。
例えば、消毒槽(除染槽)6との連結部、各アイソレータ1同士の着脱可能な連結部、上記スリーブ7との着脱可能な連結部等との連結を解除する場合に、上記アイソレータ1の内部と外界との隔離を保つことを可能にする弁を備えることができる。このような弁は当該分野において周知である。
(グローブ)
以降説明する子宮摘出、胎仔又は子豚8の取出しの作業性(好ましくは、上記に加えて、蘇生、飼育、臓器摘出の作業性)の観点から、上記アイソレータは、図1及び2に示したように、右手用及び左手用グローブ9(好ましくは、軟質プラスチック製(より好ましくは、軟質ポリ塩化ビニル製)若しくはゴム製グローブ)をそれぞれ備えていることが好ましい。
(機械アーム)
上記アイソレータ1(好ましくは、飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21、臓器摘出用アイソレータ30)は内部に、外界から操作(好ましくは、遠隔操作)可能な機械アームを備えていなくてもよいが、備えていることが好ましい。上記機械アームとしては、外界から操作である限り、材質、機能、モジュール等に特に制限はない。
上記アイソレータ1(好ましくは、飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21、臓器摘出用アイソレータ30)内での上記子豚、上記成豚の固定作業(例えば、飼育時、注射時、臓器摘出時等の固定作業)に好適である。
また、上記機械アームに注射器を担持させることにより、外界からの操作で注射(好ましくは、遠隔注射)を可能にするためである。例えば、上記アイソレータ1(好ましくは、飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21、臓器摘出用アイソレータ30)内部で、上記機械アームにより麻酔注射を行うことができる。特に、上記子豚、上記成豚からの臓器14の摘出の際の麻酔注射に好適である。
また、上記機械アームに手術器具(例えば、メス)を担持させることにより、臓器14の摘出手術も行い得る。
以降説明する子宮摘出、胎仔又は子豚8の取出しの作業性(好ましくは、上記に加えて、蘇生、飼育、臓器摘出の作業性)の観点から、上記アイソレータは、図1及び2に示したように、右手用及び左手用グローブ9(好ましくは、軟質プラスチック製(より好ましくは、軟質ポリ塩化ビニル製)若しくはゴム製グローブ)をそれぞれ備えていることが好ましい。
(機械アーム)
上記アイソレータ1(好ましくは、飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21、臓器摘出用アイソレータ30)は内部に、外界から操作(好ましくは、遠隔操作)可能な機械アームを備えていなくてもよいが、備えていることが好ましい。上記機械アームとしては、外界から操作である限り、材質、機能、モジュール等に特に制限はない。
上記アイソレータ1(好ましくは、飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21、臓器摘出用アイソレータ30)内での上記子豚、上記成豚の固定作業(例えば、飼育時、注射時、臓器摘出時等の固定作業)に好適である。
また、上記機械アームに注射器を担持させることにより、外界からの操作で注射(好ましくは、遠隔注射)を可能にするためである。例えば、上記アイソレータ1(好ましくは、飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21、臓器摘出用アイソレータ30)内部で、上記機械アームにより麻酔注射を行うことができる。特に、上記子豚、上記成豚からの臓器14の摘出の際の麻酔注射に好適である。
また、上記機械アームに手術器具(例えば、メス)を担持させることにより、臓器14の摘出手術も行い得る。
(予め内部に収容して置く物)
上記アイソレータ1(好ましくは、子豚の取出し用アイソレータ、又は、臓器摘出用アイソレータ30)は内部に、予め滅菌済みの手術器具(例えば、メス)を一式、配置しておくことができる。
また、上記アイソレータ1(好ましくは、飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21)は内部に、予め、人工哺育用の、水、代用乳、人工乳、飼料10(好ましくは、滅菌済みの、水、代用乳、人工乳、飼料)などを一式、配置しておくことができる。
また、上記アイソレータ1(好ましくは、臓器摘出用アイソレータ30)は内部に、予め、開放及び密閉可能な臓器収容用容器31(好ましくは、滅菌済の開放及び密閉可能な臓器収容用容器)を配置しておくことができる。
上記アイソレータ1は使用前に、上記アイソレータ1内部を、過酸化水素、過酢酸、オゾンガス、ホルマリンガス、紫外線、放射線(γ線照射)、プラズマ等の任意の方法により滅菌し、上記アイソレータ1内部を無菌環境とすることができる。
上記アイソレータ1(好ましくは、子豚の取出し用アイソレータ、又は、臓器摘出用アイソレータ30)は内部に、予め滅菌済みの手術器具(例えば、メス)を一式、配置しておくことができる。
また、上記アイソレータ1(好ましくは、飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21)は内部に、予め、人工哺育用の、水、代用乳、人工乳、飼料10(好ましくは、滅菌済みの、水、代用乳、人工乳、飼料)などを一式、配置しておくことができる。
また、上記アイソレータ1(好ましくは、臓器摘出用アイソレータ30)は内部に、予め、開放及び密閉可能な臓器収容用容器31(好ましくは、滅菌済の開放及び密閉可能な臓器収容用容器)を配置しておくことができる。
上記アイソレータ1は使用前に、上記アイソレータ1内部を、過酸化水素、過酢酸、オゾンガス、ホルマリンガス、紫外線、放射線(γ線照射)、プラズマ等の任意の方法により滅菌し、上記アイソレータ1内部を無菌環境とすることができる。
<母豚11の飼育>
母豚11を飼育する上記非DPF環境(指定病原体フリーではない環境)としては、DPF環境ではない限り特に制限はないが、SPF(Specific Pathogen Free)環境、国際標準ISO基準14644-1(JIS準拠)でISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等が挙げられ、煩雑で大がかりなDPF施設を要することなく、母豚11の経胎盤感染性病原体による感染を回避する観点から、SPF環境、ISOクラス6(クラス1,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境が好ましく、SPF環境がより好ましい。
上記母豚11の飼育方法としては、上記非DPF環境で飼育する限り特に制限はない。
本発明において、母豚11とは、受胎から分娩までの妊娠期にある妊娠雌豚を意味し、妊娠末期の雌豚が好ましい。
母豚11を飼育する上記非DPF環境(指定病原体フリーではない環境)としては、DPF環境ではない限り特に制限はないが、SPF(Specific Pathogen Free)環境、国際標準ISO基準14644-1(JIS準拠)でISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等が挙げられ、煩雑で大がかりなDPF施設を要することなく、母豚11の経胎盤感染性病原体による感染を回避する観点から、SPF環境、ISOクラス6(クラス1,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境が好ましく、SPF環境がより好ましい。
上記母豚11の飼育方法としては、上記非DPF環境で飼育する限り特に制限はない。
本発明において、母豚11とは、受胎から分娩までの妊娠期にある妊娠雌豚を意味し、妊娠末期の雌豚が好ましい。
本発明者らは、母豚11から胎仔又は子豚8への感染は、経胎盤感染以外の感染は原則発生しないことを見出している。
上記母豚11は、経胎盤感染性病原体による感染が陰性であり、かつそれ以外の病原体による感染が陽性又は陰性であることが好ましい。
これにより、胎仔又は子豚8、成豚22への経胎盤感染を防ぎ、最終的に得られる摘出臓器のDPF化に寄与し得る。
また、経胎盤感染性以外の病原体が陽性の母豚11でも、本発明の製造方法に適用することができるから、移植材製造コストを低減することができる。
また、母豚11が経胎盤感染性以外の病原体がたとえ陽性であったとしても、胎仔又は子豚8への垂直感染が発生しないことから、母豚11について、経胎盤感染性以外の病原体感染の検査を要せず、移植材製造プロセスにおいてボトルネック(律速)となっていた上記検査の結果判明までの待機時間を省略することができる。
上記の通り、本発明の製造方法は、母豚11について、経胎盤感染性以外の病原体感染の検査を行わないことが好ましい。
上記経胎盤感染性病原体としてはブタ繁殖・呼吸症候群(PRRS)ウイルス、オーエスキー病ウイルス、豚サーコウイルス2型、パルボウイルス、日本脳炎ウイルス、豚熱ウイルス、豚痘ウイルス、レプトスピラ、ブルセラ・スイス、豚丹毒菌、トキソプラズマが挙げられる。
上記母豚11は、経胎盤感染性病原体による感染が陰性であり、かつそれ以外の病原体による感染が陽性又は陰性であることが好ましい。
これにより、胎仔又は子豚8、成豚22への経胎盤感染を防ぎ、最終的に得られる摘出臓器のDPF化に寄与し得る。
また、経胎盤感染性以外の病原体が陽性の母豚11でも、本発明の製造方法に適用することができるから、移植材製造コストを低減することができる。
また、母豚11が経胎盤感染性以外の病原体がたとえ陽性であったとしても、胎仔又は子豚8への垂直感染が発生しないことから、母豚11について、経胎盤感染性以外の病原体感染の検査を要せず、移植材製造プロセスにおいてボトルネック(律速)となっていた上記検査の結果判明までの待機時間を省略することができる。
上記の通り、本発明の製造方法は、母豚11について、経胎盤感染性以外の病原体感染の検査を行わないことが好ましい。
上記経胎盤感染性病原体としてはブタ繁殖・呼吸症候群(PRRS)ウイルス、オーエスキー病ウイルス、豚サーコウイルス2型、パルボウイルス、日本脳炎ウイルス、豚熱ウイルス、豚痘ウイルス、レプトスピラ、ブルセラ・スイス、豚丹毒菌、トキソプラズマが挙げられる。
<子宮12の摘出>
図1に示した母豚11から子宮12の摘出方法としては特に制限はなく、任意の手術台13上で行っても行わなくてもよく、常法(例えば、仰臥位にした後の常法)による、腹式若しくは膣式子宮摘出術、帝王切開による子宮摘出術などが挙げられ、常法による開腹後に子宮摘出を行ってもよい。
図1に示した母豚11から子宮12の摘出方法としては特に制限はなく、任意の手術台13上で行っても行わなくてもよく、常法(例えば、仰臥位にした後の常法)による、腹式若しくは膣式子宮摘出術、帝王切開による子宮摘出術などが挙げられ、常法による開腹後に子宮摘出を行ってもよい。
摘出前に母豚11に麻酔を施すことが好ましい。
上記麻酔としては全身麻酔であっても局所麻酔であってもよいが全身麻酔が好ましい。
本発明者らは、取り出される子豚のバイタルサイン(特に蘇生率)への麻酔による影響を見出している。
取り出される胎仔又は子豚8のバイタルサインの観点から、子宮12の上記摘出が、上記麻酔開始後25分以内に終わることが好ましく、上記麻酔開始後27分以内に上記胎仔又は子豚の取出しが終わることがより好ましい。
吸入麻酔であっても静脈麻酔(による深麻酔)であってもよいが、麻酔の胎仔又は子豚への浸透を低減して、取り出される胎仔又は子豚8の蘇生率を向上する観点から、吸入麻酔が好ましい。
取り出される胎仔又は子豚の蘇生率を向上させる観点から、任意の麻酔拮抗剤又は呼吸促進剤を任意の時期(例えば、取出し時又は蘇生時)に任意の方法(例えば、注射)により胎仔又は子豚に投与してもしなくてもよい。
上記麻酔としては全身麻酔であっても局所麻酔であってもよいが全身麻酔が好ましい。

子宮12の上記摘出は、非DPF環境(指定病原体フリーではない環境)で行われても、DPF環境で行われてもよく、煩雑で大がかりなDPF施設を要しない観点から、非DPF環境が好ましい。摘出子宮12への不用意な病原体付着を回避する観点から、SPF環境、ISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等がより好ましい。
<デコンタミネーション処理>
図1に示した上記摘出子宮のデコンタミネーション処理は、任意の消毒媒体(除染媒体)による上記子宮の除染又は消毒等が挙げられ、より具体的には、上記子宮の上消毒媒体(除染媒体)への浸漬又は接触、上記子宮の上記消毒媒体(除染媒体)による洗浄等が挙げられる。
上記子宮摘出処理及び上記デコンタミネーション処理により、母豚から胎仔又は子豚への経産道の感染を防ぎ、最終的に得られる摘出臓器のDPF化に寄与し得る。
図1に示したように、上記消毒媒体(除染媒体)を満たした(充填した)消毒槽(除染槽)6の一端が開閉部5を介して上記アイソレータ内と連結(好ましくは上記アイソレータの気密性が維持された状態での連結)することが好ましい。この際、アイソレータ内部と、外部とが、消毒槽6に充填された消毒媒体(除染媒体)により隔離されている(すなわち、連通していない)ことがより好ましい。
摘出した上記子宮12を上記消毒槽6に投入して上記消毒槽内、及び上記連結している部分の内部を通過させ上記アイソレータ1内に上記子宮12を送達(及び収容)することが好ましい。これにより、上記消毒媒体(除染媒体)と接触後の上記子宮の外界若しくは外気との接触を回避できるからである。
上記除染媒体又は消毒媒体としては、任意の、除染液若しくは消毒液(例えば、過酢酸)、又は、除染ガス若しくは消毒ガス等が挙げられ、上記接触回避の観点から、除染液若しくは消毒液が好ましい。
図1に示した上記摘出子宮のデコンタミネーション処理は、任意の消毒媒体(除染媒体)による上記子宮の除染又は消毒等が挙げられ、より具体的には、上記子宮の上消毒媒体(除染媒体)への浸漬又は接触、上記子宮の上記消毒媒体(除染媒体)による洗浄等が挙げられる。
上記子宮摘出処理及び上記デコンタミネーション処理により、母豚から胎仔又は子豚への経産道の感染を防ぎ、最終的に得られる摘出臓器のDPF化に寄与し得る。
図1に示したように、上記消毒媒体(除染媒体)を満たした(充填した)消毒槽(除染槽)6の一端が開閉部5を介して上記アイソレータ内と連結(好ましくは上記アイソレータの気密性が維持された状態での連結)することが好ましい。この際、アイソレータ内部と、外部とが、消毒槽6に充填された消毒媒体(除染媒体)により隔離されている(すなわち、連通していない)ことがより好ましい。
摘出した上記子宮12を上記消毒槽6に投入して上記消毒槽内、及び上記連結している部分の内部を通過させ上記アイソレータ1内に上記子宮12を送達(及び収容)することが好ましい。これにより、上記消毒媒体(除染媒体)と接触後の上記子宮の外界若しくは外気との接触を回避できるからである。
上記除染媒体又は消毒媒体としては、任意の、除染液若しくは消毒液(例えば、過酢酸)、又は、除染ガス若しくは消毒ガス等が挙げられ、上記接触回避の観点から、除染液若しくは消毒液が好ましい。
上記デコンタミネーション処理を行う上記消毒媒体(除染媒体)を充填した消毒槽6を設置する環境(外部環境)としては、非DPF環境であっても、DPF環境であってもよく、煩雑で大がかりなDPF施設を要しない観点から、非DPF環境が好ましい。摘出子宮12への不用意な病原体付着を回避する観点から、SPF環境、ISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等がより好ましい。
摘出した上記子宮12の収容前に上記アイソレータ1内部を、オゾンガス、ホルマリンガス、紫外線、放射線(γ線照射)、プラズマ等の任意の方法により滅菌し、上記アイソレータ内部を無菌環境とすることができる。
<胎仔又は子豚の取出し、蘇生、飼育>
本発明において、上記子宮からの胎仔又は子豚の取出しをアイソレータ内の無菌環境で行う。さらに任意で前記胎仔又は子豚の蘇生及び飼育を含み、その場合には前記蘇生及び飼育をアイソレータ内の無菌環境で行う。
つまり、上記「任意で前記胎仔又は子豚の蘇生及び飼育を含み、かつ、前記蘇生及び飼育をアイソレータ内の無菌環境で行」うことは、アイソレータ内の無菌環境で前記胎仔又は子豚の蘇生及び飼育を行ってもよいし、アイソレータ内の無菌環境で前記胎仔又は子豚の蘇生及び飼育を行わなくてもよいことを意味し、アイソレータ内の無菌環境で前記胎仔又は子豚の蘇生及び飼育を行うことが好ましい。アイソレータ内の無菌環境で前記胎仔又は子豚の蘇生及び飼育を行わない場合であっても、下記のような無菌的な環境で前記胎仔又は子豚の蘇生及び飼育することが好ましい。
アイソレータ内で行わない場合(つまり、アイソレータ外)の無菌的な環境としては、非DPF環境又はDPF環境のいずれであってもよいが、一般的な外科手術室のクリーンクラスの環境であることが好ましく、SPF環境、ISOクラス6(クラス1,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境がより好ましく、ISOクラス5(クラス100)以上の清浄度の環境、EU-GMPガイドラインでグレードA以上の清浄度の環境が更に好ましく、ISOクラス4(クラス10)以上の清浄度の環境が好ましく、ISOクラス3(クラス1)以上の清浄度の環境が特に好ましい。
本発明において、上記子宮からの胎仔又は子豚の取出しをアイソレータ内の無菌環境で行う。さらに任意で前記胎仔又は子豚の蘇生及び飼育を含み、その場合には前記蘇生及び飼育をアイソレータ内の無菌環境で行う。
つまり、上記「任意で前記胎仔又は子豚の蘇生及び飼育を含み、かつ、前記蘇生及び飼育をアイソレータ内の無菌環境で行」うことは、アイソレータ内の無菌環境で前記胎仔又は子豚の蘇生及び飼育を行ってもよいし、アイソレータ内の無菌環境で前記胎仔又は子豚の蘇生及び飼育を行わなくてもよいことを意味し、アイソレータ内の無菌環境で前記胎仔又は子豚の蘇生及び飼育を行うことが好ましい。アイソレータ内の無菌環境で前記胎仔又は子豚の蘇生及び飼育を行わない場合であっても、下記のような無菌的な環境で前記胎仔又は子豚の蘇生及び飼育することが好ましい。
アイソレータ内で行わない場合(つまり、アイソレータ外)の無菌的な環境としては、非DPF環境又はDPF環境のいずれであってもよいが、一般的な外科手術室のクリーンクラスの環境であることが好ましく、SPF環境、ISOクラス6(クラス1,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境がより好ましく、ISOクラス5(クラス100)以上の清浄度の環境、EU-GMPガイドラインでグレードA以上の清浄度の環境が更に好ましく、ISOクラス4(クラス10)以上の清浄度の環境が好ましく、ISOクラス3(クラス1)以上の清浄度の環境が特に好ましい。
後述するように、取出し用アイソレータ、蘇生用アイソレータとは、別個のアイソレータであってもよいし、取出し用アイソレータが、蘇生用アイソレータを兼ねてもよい。
上述のように、上記アイソレータ1は、右手用及び左手用グローブ9(好ましくは、軟質プラスチック製(より好ましくは、軟質ポリ塩化ビニル製)若しくはゴム製グローブ)をそれぞれ備えていることが好ましい。これにより、右手用及び左手用グローブ9越しに、上記子宮12からの胎仔又は子豚8の取出し(及び、必要に応じて蘇生作業)を両手で行うことができる。
図1に示したように、上記子宮12からの胎仔又は子豚8の取出しは、上記アイソレータ内の無菌環境で胎仔又は子豚8を取出せ得る限り特に制限はなく、上記右手用及び左手用グローブ9越しに任意の方法(例えば、手技)で行い得る。
上記子宮からの胎仔又は子豚の取出しは上記右手用及び左手用グローブ9越しに、予め配置しておいた滅菌済みの上記手術器具(例えば、メス)を操作して行うことが好ましい。
より具体的には、上記手術器具(例えば、メス)を操作して子宮及び羊膜を(好ましくは同時に)切開して、胎仔又は子豚8を取出すことが挙げられる。
なお、指定病原体フリーではない環境下で飼育された母豚11から摘出した子宮12内であっても、子宮内は無菌的である。
上述のように、上記アイソレータ1は、右手用及び左手用グローブ9(好ましくは、軟質プラスチック製(より好ましくは、軟質ポリ塩化ビニル製)若しくはゴム製グローブ)をそれぞれ備えていることが好ましい。これにより、右手用及び左手用グローブ9越しに、上記子宮12からの胎仔又は子豚8の取出し(及び、必要に応じて蘇生作業)を両手で行うことができる。
図1に示したように、上記子宮12からの胎仔又は子豚8の取出しは、上記アイソレータ内の無菌環境で胎仔又は子豚8を取出せ得る限り特に制限はなく、上記右手用及び左手用グローブ9越しに任意の方法(例えば、手技)で行い得る。
上記子宮からの胎仔又は子豚の取出しは上記右手用及び左手用グローブ9越しに、予め配置しておいた滅菌済みの上記手術器具(例えば、メス)を操作して行うことが好ましい。
より具体的には、上記手術器具(例えば、メス)を操作して子宮及び羊膜を(好ましくは同時に)切開して、胎仔又は子豚8を取出すことが挙げられる。
なお、指定病原体フリーではない環境下で飼育された母豚11から摘出した子宮12内であっても、子宮内は無菌的である。
上記子豚の蘇生は、上記胎仔又は子豚を蘇生できる限り特に制限はなく、上記アイソレータ内の無菌環境で行うことが好ましく、上記右手用及び左手用グローブ越しに、上記胎仔(新生仔)又は子豚の体と口腔、鼻孔の羊水を拭き取る等の体表面の洗浄等の任意の方法(例えば、手技)で蘇生させることが挙げられる。
アイソレータ1の無菌環境とされた内部は、上述のように、アイソレータ1の外部から隔離されていることから、胎仔又は子豚の取出し作業を行うアイソレータ及び蘇生作業を行うアイソレータを設置する環境(すなわち、アイソレータの外部環境)は各々独立に非DPF環境又はDPF環境のいずれであってもよく、煩雑で大がかりなDPF施設を要しない観点から、非DPF環境が好ましく、アイソレータ内部への不用意な病原体の混入を回避する観点から、SPF環境、ISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等がより好ましい。
アイソレータ1の無菌環境とされた内部は、上述のように、アイソレータ1の外部から隔離されていることから、胎仔又は子豚の取出し作業を行うアイソレータ及び蘇生作業を行うアイソレータを設置する環境(すなわち、アイソレータの外部環境)は各々独立に非DPF環境又はDPF環境のいずれであってもよく、煩雑で大がかりなDPF施設を要しない観点から、非DPF環境が好ましく、アイソレータ内部への不用意な病原体の混入を回避する観点から、SPF環境、ISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等がより好ましい。
上記蘇生後の上記胎仔又は子豚の飼育、子豚が生育した成豚22の飼育は、上記アイソレータ外で飼育しても上記アイソレータ内で飼育してよいが、上記アイソレータ外で飼育する場合であっても、上述したような一般的な外科手術室のクリーンクラスの環境等の無菌的な環境で飼育することが好ましい。図2に示したように、上記蘇生後の上記胎仔又は子豚の飼育、子豚が生育した成豚22の飼育は、上記アイソレータ内の無菌環境で飼育することが好ましく、上記アイソレータ内に予め配置しておいた、水、代用乳、人工乳、飼料10(好ましくは、滅菌済みの、水、代用乳、人工乳、飼料)で人工哺育することがより好ましい。
上記のように、アイソレータ1内に予め配置して置くことによりアイソレータ1内部の外部からの隔離性を維持することができ、飼育時において、水、代用乳、人工乳、飼料等の搬入(例えば、開閉部5を介した搬入)に伴う外気流入による病原体の混入を防止し、最終的に得られる摘出臓器のDPF化に寄与し得る。
必要に応じてスリーブ7を介して他のアイソレータの開閉部5と連結可能な開閉部5の位置は、胎仔又は子豚8、成豚22を飼育する位置(例えば、下記床15)に対して、25cm以上の高さを有することが好ましく、30cm以上の高さを有することがより好ましく、35cm以上の高さを有することが更に好ましく、40cm以上の高さを有することが特に好ましく、50cm以上の高さを有することがとりわけ好ましく、60cm以上の高さを有することが最も好ましい。
これにより、胎仔又は子豚8、成豚が他のアイソレータへ自力で移動してしまうことを防止し、脱走を防止することができる。
上記高さの上限値としては特に制限はないが、例えば、160cm以下、140cm以下、120cm以下、100cm以下、90cm以下等が挙げられる。
上記のように、アイソレータ1内に予め配置して置くことによりアイソレータ1内部の外部からの隔離性を維持することができ、飼育時において、水、代用乳、人工乳、飼料等の搬入(例えば、開閉部5を介した搬入)に伴う外気流入による病原体の混入を防止し、最終的に得られる摘出臓器のDPF化に寄与し得る。
必要に応じてスリーブ7を介して他のアイソレータの開閉部5と連結可能な開閉部5の位置は、胎仔又は子豚8、成豚22を飼育する位置(例えば、下記床15)に対して、25cm以上の高さを有することが好ましく、30cm以上の高さを有することがより好ましく、35cm以上の高さを有することが更に好ましく、40cm以上の高さを有することが特に好ましく、50cm以上の高さを有することがとりわけ好ましく、60cm以上の高さを有することが最も好ましい。
これにより、胎仔又は子豚8、成豚が他のアイソレータへ自力で移動してしまうことを防止し、脱走を防止することができる。
上記高さの上限値としては特に制限はないが、例えば、160cm以下、140cm以下、120cm以下、100cm以下、90cm以下等が挙げられる。
上記アイソレータ1内に胎仔、子豚、成豚を配置ないし飼育するための床15を設けても設けなくてもよく、上記床15を境に上記アイソレータ内部を上部と下部とに分けても分けなくてもよい。上記床の材質としては特に制限はなく、木材、金属材(ステンレス、鉄等)プラスチック材等が挙げられる。
上記床15としては、糞尿を床下に落下させ、胎仔又は子豚8、成豚22を糞尿で汚染させることなく衛生管理する観点から、網目構造を有する床が好ましく、例えば、簀子、金網(エキスパンドメタル、メッシュ金網、鉄網等)が好ましい。
床下に落下した糞尿を貯留するため、上記アイソレータ内の床下に糞尿貯留槽を設けても設けなくてもよい。
飼育の観点から、特に、図2に示したように、後述する飼育用のアイソレータ20が、上記床15及び糞尿貯留槽を有することが好ましい。
特に、子豚から成豚への生育のような長期間(例えば、4週間以上、好ましくは3か月以上、より好ましくは6か月以上、更に好ましくは、1年以上、特に好ましくは1年6か月以上)の飼育における衛生管理の観点から、上記糞尿貯留槽は、上記アイソレータ1(好ましくは飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21)内の床下と管で連結されたバキューム式の糞尿貯留槽が好ましい。この際、外界に対して上記アイソレータ1(好ましくは飼育用アイソレータ(胎仔又は子豚飼育用アイソレータ)20、成豚飼育用アイソレータ21)内は陽圧に維持されていることが好ましい。これにより、上記管を介して気圧差により糞尿を吸引して上記糞尿貯留槽を運ぶことができる。
この際、上記糞尿貯留槽は、別の管を介して、外界(アイソレータ外)は陰圧に維持された外界(アイソレータ外)に連結していてもいなくてもよい。上記各管は気圧制御の観点から弁を有していてもいなくてもよい。
上記糞尿貯留槽は、バキュームブロアにより陰圧化されていてもいなくてもよい。
上記床15としては、糞尿を床下に落下させ、胎仔又は子豚8、成豚22を糞尿で汚染させることなく衛生管理する観点から、網目構造を有する床が好ましく、例えば、簀子、金網(エキスパンドメタル、メッシュ金網、鉄網等)が好ましい。
床下に落下した糞尿を貯留するため、上記アイソレータ内の床下に糞尿貯留槽を設けても設けなくてもよい。
飼育の観点から、特に、図2に示したように、後述する飼育用のアイソレータ20が、上記床15及び糞尿貯留槽を有することが好ましい。

図2に示したように、上記アイソレータ1内に棚を具備することにより、例えば、上述した、人工哺育用の、水、代用乳、人工乳、飼料10(好ましくは、滅菌済みの、水、代用乳、人工乳、飼料)などを収容しておくことができる。これにより、人工哺育用の、水、代用乳、人工乳、飼料10を糞尿で汚染させることなく衛生管理することができる。
アイソレータ1の無菌環境とされた内部は、上述のように、アイソレータ1の外部から隔離されていることから、胎仔、子豚、成豚の飼育を行うアイソレータを設置する環境(すなわち、アイソレータの外部環境)は非DPF環境又はDPF環境のいずれであってもよく、煩雑で大がかりなDPF施設を要しない観点から、非DPF環境が好ましく、アイソレータ内部への不用意な病原体の混入を回避する観点から、SPF環境、ISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等がより好ましい。
図2に示したように、上記アイソレータ1内に棚を具備することにより、例えば、上述した、人工哺育用の、水、代用乳、人工乳、飼料10(好ましくは、滅菌済みの、水、代用乳、人工乳、飼料)などを収容しておくことができる。これにより、人工哺育用の、水、代用乳、人工乳、飼料10を糞尿で汚染させることなく衛生管理することができる。
アイソレータ1の無菌環境とされた内部は、上述のように、アイソレータ1の外部から隔離されていることから、胎仔、子豚、成豚の飼育を行うアイソレータを設置する環境(すなわち、アイソレータの外部環境)は非DPF環境又はDPF環境のいずれであってもよく、煩雑で大がかりなDPF施設を要しない観点から、非DPF環境が好ましく、アイソレータ内部への不用意な病原体の混入を回避する観点から、SPF環境、ISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等がより好ましい。
飼育期間としては、臓器摘出の種類、使用目的等に応じて適宜設定し得る。
前駆細胞の収集、その後の数週間の分化誘導培養等に供する臓器(例えば、膵臓)の摘出を目的とする場合、飼育期間としては、1週齢以上になるまで飼育することが好ましく、2週齢以上になるまで飼育することがより好ましく、3週齢以上になるまで飼育することが更に好ましい。
この際の飼育の期間の上限としては、2か月齢以下が好ましく、1か月齢以下がより好ましく、4週齢以下が更に好ましい。
一方、大きさ、機能等の成熟を要する臓器(例えば、心臓、腎臓)の摘出を目的とする場合(つまり、子豚が生育した成豚22からの摘出が好ましい場合)、飼育期間としては、6か月以上が好ましく、1年以上がより好ましい。この際の飼育の期間の上限としては、2年以下が好ましく、1年6か月齢以下がより好ましい。

<胎仔、子豚、又は子豚が生育した成豚からの臓器摘出>
本発明は、胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することを含む。
臓器の上記摘出はアイソレータ内の無菌環境で行っても、行わなくてもよいが、アイソレータ内の無菌環境で行うことが好ましい。
臓器の上記摘出をアイソレータ内で行わない場合(つまり、アイソレータ外)であっても、上述した無菌的な環境で行うことが好ましい。上記無菌的な環境としては、非DPF環境又はDPF環境のいずれであってもよいが、一般的な外科手術室のクリーンクラスであることが好ましく、SPF環境、ISOクラス6(クラス1,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境がより好ましく、ISOクラス5(クラス100)以上の清浄度の環境、EU-GMPガイドラインでグレードA以上の清浄度の環境が更に好ましく、ISOクラス4(クラス10)以上の清浄度の環境が好ましく、ISOクラス3(クラス1)以上の清浄度の環境が特に好ましい。
本発明は、胎仔、子豚、又は子豚が生育した成豚から臓器を摘出することを含む。
臓器の上記摘出はアイソレータ内の無菌環境で行っても、行わなくてもよいが、アイソレータ内の無菌環境で行うことが好ましい。
臓器の上記摘出をアイソレータ内で行わない場合(つまり、アイソレータ外)であっても、上述した無菌的な環境で行うことが好ましい。上記無菌的な環境としては、非DPF環境又はDPF環境のいずれであってもよいが、一般的な外科手術室のクリーンクラスであることが好ましく、SPF環境、ISOクラス6(クラス1,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境がより好ましく、ISOクラス5(クラス100)以上の清浄度の環境、EU-GMPガイドラインでグレードA以上の清浄度の環境が更に好ましく、ISOクラス4(クラス10)以上の清浄度の環境が好ましく、ISOクラス3(クラス1)以上の清浄度の環境が特に好ましい。
図3に示したように、上記胎仔、子豚8、又は子豚が生育した成豚22からの臓器14の上記摘出を上記アイソレータ内で行う場合、上記右手用及び左手用グローブ9越しに、上記アイソレータ内に予め配置しておいた滅菌済みの上記手術器具(例えば、メス)を操作して行うことが好ましい。
より具体的には、麻酔していてもいなくてもよい上記胎仔又は子豚8、成豚22を、例えば仰臥位にした後、上記手術器具(例えば、メス)を操作して開腹手術を行い、任意の臓器14を摘出することが挙げられる。
上記摘出は、上記胎仔又は子豚8の躯体及び上記臓器14がいずれも数センチメートル程度の細かい作業となる場合、上記摘出は、上記胎仔又は子豚8、成豚22の躯体中の上記臓器14に生理学的に許容される液を注入して上記臓器14を膨張させた状態で行われることが好ましい。上記臓器14を膨張させることにより作業性を向上するためである。
生理学的に許容される上記液としては、ETK液(ET-Kyoto液)、HTK液(histidine-tryptophan-ketoglutarate液)、生理食塩水、UW液(University of Wisconsin液)、輸液(細胞外液)、水、リン酸緩衝液等が挙げられ、ETK液が好ましい。
より具体的には、麻酔していてもいなくてもよい上記胎仔又は子豚8、成豚22を、例えば仰臥位にした後、上記手術器具(例えば、メス)を操作して開腹手術を行い、任意の臓器14を摘出することが挙げられる。
上記摘出は、上記胎仔又は子豚8の躯体及び上記臓器14がいずれも数センチメートル程度の細かい作業となる場合、上記摘出は、上記胎仔又は子豚8、成豚22の躯体中の上記臓器14に生理学的に許容される液を注入して上記臓器14を膨張させた状態で行われることが好ましい。上記臓器14を膨張させることにより作業性を向上するためである。
生理学的に許容される上記液としては、ETK液(ET-Kyoto液)、HTK液(histidine-tryptophan-ketoglutarate液)、生理食塩水、UW液(University of Wisconsin液)、輸液(細胞外液)、水、リン酸緩衝液等が挙げられ、ETK液が好ましい。
必要に応じてスリーブ7を介して他のアイソレータの開閉部5と連結可能な開閉部5の位置は、上記胎仔又は子豚8、成豚22を飼育する位置(例えば、下記床15)に対して、25cm以上の高さを有することが好ましく、30cm以上の高さを有することがより好ましく、35cm以上の高さを有することが更に好ましく、40cm以上の高さを有することが特に好ましく、50cm以上の高さを有することがとりわけ好ましく、60cm以上の高さを有することが最も好ましい。
上記胎仔又は子豚8、成豚22が他のアイソレータへ自力で移動してしまうことを防止し、脱走を防止することができる。

上記のように、上記アイソレータ内部に、予め配置しておいた開放及び密閉可能な臓器収容用容器31(好ましくは、滅菌済の開放及び密閉可能な臓器収容用容器)に、摘出された臓器14を収容し、容器31を密閉後、上記アイソレータ外部に(例えば、開閉部5を介して)搬出することができる。この場合、開閉部5はパスボックスの形態であってもよい。
これにより、アイソレータの外部環境が非DPF環境であっても、臓器14が汚染されず、臓器14のDPF化に寄与し得る。
上記臓器14としては、内臓(例えば、膵臓、腎臓、尿管、膀胱、肝臓、心臓、胃、腸等)、生殖器(例えば、卵巣、精巣等)、骨髄(例えば、造血器官)、脳、眼、鼻、口、皮膚、神経が挙げられ、内臓、生殖器が好ましく、内臓、生殖器がより好ましく、膵臓(例えば、膵島)、腎臓(例えば、後腎、特に、後腎、尿管及び膀胱を含む泌尿器)がさらに好ましい。

上記のように、上記アイソレータ内部に、予め配置しておいた開放及び密閉可能な臓器収容用容器31(好ましくは、滅菌済の開放及び密閉可能な臓器収容用容器)に、摘出された臓器14を収容し、容器31を密閉後、上記アイソレータ外部に(例えば、開閉部5を介して)搬出することができる。この場合、開閉部5はパスボックスの形態であってもよい。
これにより、アイソレータの外部環境が非DPF環境であっても、臓器14が汚染されず、臓器14のDPF化に寄与し得る。

アイソレータ1の無菌環境とされた内部は、上述のように、アイソレータ1の外部から隔離されていることから、臓器14の摘出を行うアイソレータを設置する環境(すなわち、アイソレータの外部環境)は非DPF環境又はDPF環境のいずれであってもよく、煩雑で大がかりなDPF施設を要しない観点から、非DPF環境が好ましく、アイソレータ内部への不用意な病原体の混入を回避する観点から、SPF環境、ISOクラス7(クラス10,000)以上の清浄度の環境、EU-GMPガイドラインでグレードB以上の清浄度の環境等がより好ましい。
上記摘出した臓器14として医療グレードのDPF(指定病原体フリー)の臓器又はその由来物を得ることができる。
上記摘出した臓器14は、子豚胎仔由来である場合、免疫原性が低く、また、指定病原体フリーであることから、移植材(好ましくは、異種移植材)として好適である。
一方、上記摘出した臓器14が、成豚由来である場合、大きさ、機能等の成熟が好適であり、また、指定病原体フリーであることから、移植材(好ましくは、異種移植材)として好適である。
上記摘出した臓器14は、子豚胎仔由来である場合、免疫原性が低く、また、指定病原体フリーであることから、移植材(好ましくは、異種移植材)として好適である。
一方、上記摘出した臓器14が、成豚由来である場合、大きさ、機能等の成熟が好適であり、また、指定病原体フリーであることから、移植材(好ましくは、異種移植材)として好適である。
<アイソレータの複数化>
上記子宮からの胎仔又は子豚8の取出し、蘇生、飼育、上記胎仔、子豚又は成豚22からの臓器の上記摘出は、内部を無菌環境に維持した単一のアイソレータ内で一貫して行っても行わなくてもよいが、上記子宮からの胎仔又は子豚の取出し作業、蘇生作業、飼育作業、上記胎仔、子豚又は成豚からの臓器の上記摘出作業は、それぞれ、内部を無菌環境に維持した2以上の別個のアイソレータ内で行うことが好ましい。これにより、上記各作業に対応する各アイソレータを設置する環境(すなわち、各アイソレータの外部環境)を非DPF環境又はDPF環境に上記各作業毎(各アイソレータ毎)に独立して選択することができる。また、各アイソレータの設置場所も各アイソレータ毎に独立して選択することができる。
さらに、所要時間が相違する上記各作業を別個のアイソレータ内で行うことにより作業効率及び管理性を向上し得る。
図1~4に示したように、上記子宮からの子豚の取出し(子豚の取出し用アイソレータ1)と、上記蘇生と、上記飼育(飼育用アイソレータ20)と、上記摘出(臓器摘出用アイソレータ30)とが、それぞれ、内部を無菌環境に維持した別個のアイソレータ内で行うことがより好ましい。
特に、飼育用アイソレータ20は、図4に示したように、生育状況に応じて内部の床15の高さ、水、飼料10等を調整した、胎仔又は子豚飼育用アイソレータ20、成豚飼育用アイソレータ21等の別個の複数種のアイソレータとしてもしなくてもよい。
上記子宮からの胎仔又は子豚8の取出し、蘇生、飼育、上記胎仔、子豚又は成豚22からの臓器の上記摘出は、内部を無菌環境に維持した単一のアイソレータ内で一貫して行っても行わなくてもよいが、上記子宮からの胎仔又は子豚の取出し作業、蘇生作業、飼育作業、上記胎仔、子豚又は成豚からの臓器の上記摘出作業は、それぞれ、内部を無菌環境に維持した2以上の別個のアイソレータ内で行うことが好ましい。これにより、上記各作業に対応する各アイソレータを設置する環境(すなわち、各アイソレータの外部環境)を非DPF環境又はDPF環境に上記各作業毎(各アイソレータ毎)に独立して選択することができる。また、各アイソレータの設置場所も各アイソレータ毎に独立して選択することができる。
さらに、所要時間が相違する上記各作業を別個のアイソレータ内で行うことにより作業効率及び管理性を向上し得る。
図1~4に示したように、上記子宮からの子豚の取出し(子豚の取出し用アイソレータ1)と、上記蘇生と、上記飼育(飼育用アイソレータ20)と、上記摘出(臓器摘出用アイソレータ30)とが、それぞれ、内部を無菌環境に維持した別個のアイソレータ内で行うことがより好ましい。
特に、飼育用アイソレータ20は、図4に示したように、生育状況に応じて内部の床15の高さ、水、飼料10等を調整した、胎仔又は子豚飼育用アイソレータ20、成豚飼育用アイソレータ21等の別個の複数種のアイソレータとしてもしなくてもよい。
上記子宮からの胎仔又は子豚の取出し、蘇生、飼育、上記胎仔又は子豚8、成豚22からの臓器14の上記摘出は、それぞれ、内部を無菌環境に維持した別個のアイソレータを使用する場合、
各アイソレータは、外界からの隔離性及び気密性を維持しつつ連結することができ、かつ開閉部5と着脱可能なスリーブ(連結器)7を介して着脱可能に連結可能であることが好ましい。上記スリーブ(連結器)7は各アイソレータの内部空間同士を着脱可能に連結可能である。
上記連結することにより、各アイソレータ間を胎仔又は子豚8、成豚22を移動させることができる。
一方、取り外す(脱する)ことにより、各アイソレータ毎に各アイソレータの外部環境(例えば、非DPF環境又はDPF環境)を独立して選択することができ、また、各アイソレータの設置場所も各アイソレータ毎に独立して選択することができる。
上記アイソレータ内に外気が流入した際に、微生物、ウイルス等の病原体の混入を防ぐ観点から、各アイソレータ同士の取り外し(脱)、又は、上記スリーブ7の取り外し(脱)は、DPF環境下で行うことが好ましい。
上記スリーブ7(好ましくは、子豚移動用スリーブ)は、内部を過酸化水素、過酢酸、オゾンガス、ホルマリンガス、紫外線、放射線(γ線照射)、プラズマ等の任意の方法により滅菌してから使用することが好ましい。
これにより、各アイソレータ同士を連結したスリーブ内の無菌環境空間を通じて、胎仔又は子豚8、成豚22をアイソレータ間で移動させることができ、例えば、最終的には、上記胎仔又は子豚8、成豚22からの臓器14の上記摘出するためのアイソレータの内部空間へ移動させることができる。
上記スリーブ7の材質としては、特に制限はないが、プラスチック(軟質プラスチック(軟質ポリ塩化ビニルなど)、硬質プラスチック)、金属(例えば、ステンレス、鉄等)などが挙げられる。
各アイソレータは、外界からの隔離性及び気密性を維持しつつ連結することができ、かつ開閉部5と着脱可能なスリーブ(連結器)7を介して着脱可能に連結可能であることが好ましい。上記スリーブ(連結器)7は各アイソレータの内部空間同士を着脱可能に連結可能である。
上記連結することにより、各アイソレータ間を胎仔又は子豚8、成豚22を移動させることができる。
一方、取り外す(脱する)ことにより、各アイソレータ毎に各アイソレータの外部環境(例えば、非DPF環境又はDPF環境)を独立して選択することができ、また、各アイソレータの設置場所も各アイソレータ毎に独立して選択することができる。
上記アイソレータ内に外気が流入した際に、微生物、ウイルス等の病原体の混入を防ぐ観点から、各アイソレータ同士の取り外し(脱)、又は、上記スリーブ7の取り外し(脱)は、DPF環境下で行うことが好ましい。

これにより、各アイソレータ同士を連結したスリーブ内の無菌環境空間を通じて、胎仔又は子豚8、成豚22をアイソレータ間で移動させることができ、例えば、最終的には、上記胎仔又は子豚8、成豚22からの臓器14の上記摘出するためのアイソレータの内部空間へ移動させることができる。
上記スリーブ7の材質としては、特に制限はないが、プラスチック(軟質プラスチック(軟質ポリ塩化ビニルなど)、硬質プラスチック)、金属(例えば、ステンレス、鉄等)などが挙げられる。
飼育用アイソレータ20、21とは別個の胎仔又は子豚の取出し用アイソレータ1、又は、臓器摘出用アイソレータ30内部に、予め滅菌済みの手術器具(例えば、メス)を配置しておくことにより、胎仔又は子豚8、成豚22の飼料、糞尿等による上記手術器具の汚染を回避し得る。
また、図2に示したように、胎仔又は子豚の取出し用アイソレータ1等とは別個の飼育用のアイソレータ20内部に予め、人工哺育用の、水、代用乳、人工乳、飼料10(好ましくは、滅菌済みの、水、代用乳、人工乳、飼料)などを一式、配置しておくことにより、胎仔又は子豚8取出し時の、血液、羊水等の体液等による人工哺育用の、水、代用乳、人工乳、飼料10の汚染を回避し得る。
また、図3に示したように、飼育用アイソレータ20、21とは別個の臓器摘出用アイソレータ30内部に、予め、開放及び密閉可能な臓器収容用容器31を配置しておくことにより、胎仔又は子豚8、成豚22の飼料、糞尿等による上記容器31の汚染を回避し得る。
また、図2に示したように、胎仔又は子豚の取出し用アイソレータ1等とは別個の飼育用のアイソレータ20内部に予め、人工哺育用の、水、代用乳、人工乳、飼料10(好ましくは、滅菌済みの、水、代用乳、人工乳、飼料)などを一式、配置しておくことにより、胎仔又は子豚8取出し時の、血液、羊水等の体液等による人工哺育用の、水、代用乳、人工乳、飼料10の汚染を回避し得る。
また、図3に示したように、飼育用アイソレータ20、21とは別個の臓器摘出用アイソレータ30内部に、予め、開放及び密閉可能な臓器収容用容器31を配置しておくことにより、胎仔又は子豚8、成豚22の飼料、糞尿等による上記容器31の汚染を回避し得る。
上記子宮からの胎仔又は子豚の取出し作業、蘇生作業、飼育作業、上記胎仔、子豚又は成豚22からの臓器の上記摘出作業を、それぞれ、2以上の別個のアイソレータ内で行う場合、各々の作業を並行して行い量産性を向上させる観点から、各作業について2以上の複数個のアイソレータを併用してもよい。例えば、胎仔又は子豚の取出し用アイソレータ1を複数個併用してもよいし、蘇生用アイソレータを複数個併用してもよいし、飼育用アイソレータ20を複数個併用してもよいし、臓器摘出用アイソレータ30を複数個併用してもよい。
胎仔又は子豚の取出し作業及び蘇生作業、並びに、臓器摘出作業の所要時間は数時間であるのに対して、飼育作業の所要時間は数週間を要し律速作業となり得る。換言すると、胎仔又は子豚の取出し用アイソレータ1、蘇生用アイソレータ並びに、臓器摘出用アイソレータ30に対して、飼育用アイソレータ20、21は、再使用の回転速度が極端に低く、移植材の製造のボトルネックになり得る。
飼育作業の上記律速性を回避する観点から、飼育用アイソレータ20、21を複数個併用することが好ましい。
各作業について2以上の複数個のアイソレータを併用する場合、各作業についてのアイソレータの併用個数同士は、同数であっても、異数であってもよい。例えば、胎仔又は子豚の取出し用アイソレータ1を2個併用してもよいし、蘇生用アイソレータを2個併用してもよいし、飼育用アイソレータ20、21を3個以上併用してもよいし、臓器摘出用アイソレータ30を2個併用してもよい。

無菌環境を維持するための巨大な空調設備を要する高規格飼育施設の床面積を低減する観点、及び、設備維持費の大幅な削減の観点から、
図5に示したように、上記飼育用アイソレータ20、21が2以上存在する場合、2以上の上記飼育用アイソレータ20、21を高さ方向に重ねて配置することが好ましい。また、上記アイソレータ20、21は、少なくとも高さ方向に関して区画化する(幅方向に延びる)複数の棚板を有するラック23(棚)に収容することが好ましい。より好ましくは、幅方向に関して区画化する(高さ方向に延びる)棚板をさらに有するラックを使い、区画化された各空間に、単一のアイソレータ又は同一母豚由来の子豚を飼育する複数のアイソレータを収容する。
無菌環境を維持するための巨大な空調設備を要する高規格飼育施設の床面積を低減する観点、及び、設備維持費の大幅な削減の観点から、
図5に示したように、上記飼育用アイソレータ20、21が2以上存在する場合、2以上の上記飼育用アイソレータ20、21を高さ方向に重ねて配置することが好ましい。また、上記アイソレータ20、21は、少なくとも高さ方向に関して区画化する(幅方向に延びる)複数の棚板を有するラック23(棚)に収容することが好ましい。より好ましくは、幅方向に関して区画化する(高さ方向に延びる)棚板をさらに有するラックを使い、区画化された各空間に、単一のアイソレータ又は同一母豚由来の子豚を飼育する複数のアイソレータを収容する。
<アイソレータのその他構成>
上記アイソレータの寸法は、胎仔、子豚、成豚の収容数に応じて適宜、設計することができ、胎仔、子豚、成豚を飼育するに十分な空間を有することが好ましい。例えば、収容される豚総計のサイズの約30倍以上の容積を有することが挙げられる。
例えば100~1000cm、150~500cm、又は200~300cm等の長さ、例えば100~1000cm、150~500cm、又は200~300cm等の幅、例えば50~500cm、100~300cm、又は150~250cm等の高さを有し得るが、寸法はこれに限らない。
上記アイソレータの寸法は、胎仔、子豚、成豚の収容数に応じて適宜、設計することができ、胎仔、子豚、成豚を飼育するに十分な空間を有することが好ましい。例えば、収容される豚総計のサイズの約30倍以上の容積を有することが挙げられる。
例えば100~1000cm、150~500cm、又は200~300cm等の長さ、例えば100~1000cm、150~500cm、又は200~300cm等の幅、例えば50~500cm、100~300cm、又は150~250cm等の高さを有し得るが、寸法はこれに限らない。
上記アイソレータは、1又は2以上の観察窓を有していてもいなくてよく、2以上の観察窓を有していることが好ましく、例えば2つ、3つ、又は4つしていることがより好ましい。
観察窓は上記アイソレータの上部(上面)及び/又は側部(側面)に設けることができる。上記アイソレータの上部及び側部の一部を傾斜させ、該傾斜面(傾斜部ともいう)に観察窓を設けることができる。観察窓は、上記アイソレータの上部、側部、及び/又は傾斜部に設けることができる。それぞれの観察窓の寸法、形状、及び材料は、同一でも異なってもよい。
観察窓は上記アイソレータの上部(上面)及び/又は側部(側面)に設けることができる。上記アイソレータの上部及び側部の一部を傾斜させ、該傾斜面(傾斜部ともいう)に観察窓を設けることができる。観察窓は、上記アイソレータの上部、側部、及び/又は傾斜部に設けることができる。それぞれの観察窓の寸法、形状、及び材料は、同一でも異なってもよい。
観察窓は、例えば、観察窓枠及び硬質窓材料を有する。
観察窓及び観察窓枠は、上記アイソレータの使用態様に応じて、四角形、長方形、正方形、円形(例えば略円形又は正円形)、楕円形等とすることができる。硬質窓材料は硬質フィルム、例えばビニール板であり得る。観察窓及び観察窓枠の寸法は特に制限はなく、例えば50cm2~8000cm2、300cm2~5000cm2程度の面積(枠の内側の面積)を有する楕円形、四角形、長方形、又は正方形の枠であり得るがこれに限らない。ある実施形態において、硬質窓材料、例えばビニール板、例えば透明硬質塩化ビニール板は、例えば、0.05mm~5mmの厚みを有するものであり得る。
観察窓は、硬質窓材料として透明硬質塩化ビニール(0.1mm厚の薄い板)を、接着剤で、観察窓枠に接着して構成する。観察窓枠は上述したパッキンとしても機能し得る。
観察窓及び観察窓枠は、上記アイソレータの使用態様に応じて、四角形、長方形、正方形、円形(例えば略円形又は正円形)、楕円形等とすることができる。硬質窓材料は硬質フィルム、例えばビニール板であり得る。観察窓及び観察窓枠の寸法は特に制限はなく、例えば50cm2~8000cm2、300cm2~5000cm2程度の面積(枠の内側の面積)を有する楕円形、四角形、長方形、又は正方形の枠であり得るがこれに限らない。ある実施形態において、硬質窓材料、例えばビニール板、例えば透明硬質塩化ビニール板は、例えば、0.05mm~5mmの厚みを有するものであり得る。
観察窓は、硬質窓材料として透明硬質塩化ビニール(0.1mm厚の薄い板)を、接着剤で、観察窓枠に接着して構成する。観察窓枠は上述したパッキンとしても機能し得る。
曇りによる支障なく細かい作業を遂行する観点から、上記硬質窓材料の表面が任意の曇り止め加工(例えば、撥水加工、超撥水加工等)されていてもいなくてもよいが、されていることが好ましい。
光の反射の支障なく細かい作業を遂行する観点から、上記硬質窓材料は、波打つことのできる軟質プラスチック材料と比較して乱反射が少ないことが好ましく、例えば透明硬質塩化ビニールが示す程度に乱反射が少ないことが挙げられる。
硬質窓材料は透明塩化ビニール板であり得るがこれに限らない。
光の反射の支障なく細かい作業を遂行する観点から、上記硬質窓材料は、波打つことのできる軟質プラスチック材料と比較して乱反射が少ないことが好ましく、例えば透明硬質塩化ビニールが示す程度に乱反射が少ないことが挙げられる。
硬質窓材料は透明塩化ビニール板であり得るがこれに限らない。
図1に示したように、上記アイソレータ(例えば吸気部3)は、アイソレータ内の温度、湿度、酸素濃度、二酸化炭素濃度よりなる群から選択される少なくとも1つのパラメータの能動的な制御を行うエアコンディショナー16を具備していてもいなくてもよい。
アイソレータ内温度は例えば5℃から38℃までの範囲内に保持され、好ましくは10℃から35℃までの範囲内に保持される。アイソレータ内湿度は例えば0%から100%(相対湿度)までの範囲内に保持され、好ましくは40%から70%までの範囲内に保持される。アイソレータ内酸素濃度は、通常の新鮮な空気における濃度であることが好ましい。豚に悪影響を与えないため、低酸素(例えば、5%)状態となるような条件は避けるべきである。アイソレータ内二酸化炭素濃度は、新鮮な空気における濃度であることが好ましい。豚に悪影響を与えないため、高二酸化炭素(例えば、約50%以上)となるような条件は避けるべきである。
アイソレータ内温度は例えば5℃から38℃までの範囲内に保持され、好ましくは10℃から35℃までの範囲内に保持される。アイソレータ内湿度は例えば0%から100%(相対湿度)までの範囲内に保持され、好ましくは40%から70%までの範囲内に保持される。アイソレータ内酸素濃度は、通常の新鮮な空気における濃度であることが好ましい。豚に悪影響を与えないため、低酸素(例えば、5%)状態となるような条件は避けるべきである。アイソレータ内二酸化炭素濃度は、新鮮な空気における濃度であることが好ましい。豚に悪影響を与えないため、高二酸化炭素(例えば、約50%以上)となるような条件は避けるべきである。
以下に本発明の実施例を示し、本発明をさらに具体的に説明するが、本発明はこれらに限定されるものではなく、本発明の技術的思想を逸脱しない範囲内で種々の応用が可能である。
<参考例1>
DPFではない環境下で飼育された妊娠母豚から自然分娩によって得られた子豚について口腔内、消化管内、体表等に病原体を保有する個体の存在を検査した。結果を下記表1及び2に示す。
上記表1から明らかなように、母豚には豚呼吸器・繁殖障害症候群ウイルス等の病原体に感染されている個体が存在した。
上記表2から明らかなように、DPFではない環境下で飼育された妊娠母豚から自然分娩によって得られた子豚は、産道の通過及び母豚からの哺乳によって口腔内、消化管内、体表等に様々な菌、真菌、ウイルス等の病原体を保有するに至っていることが分かる。
DPFではない環境下で飼育された妊娠母豚から自然分娩によって得られた子豚について口腔内、消化管内、体表等に病原体を保有する個体の存在を検査した。結果を下記表1及び2に示す。
<参考例2>
帝王切開によれば、母豚からの経産道感染は免れ得る。
DPFではない環境で飼育した妊娠母豚から帝王切開により出生した子豚3頭について、2頭は内部を滅菌後の無菌環境を維持されたアイソレータ内で2週齢まで飼育した。
残りの1頭は、アイソレータ外飼育に相当する態様として内部が未滅菌のアイソレータ内で2週齢まで飼育した。
3頭それぞれの口腔、皮膚及び糞便について細菌の存在を検査した。
結果を下記表3に示す。
上記表3に示した結果から明らかなように、アイソレータ外飼育に相当する態様として内部が未滅菌のアイソレータ内で飼育した子豚は、肺炎桿菌(Klebsiella pneumoniae)、黄色葡萄球菌(Staphyrococcus aureus)、溶血性連鎖球菌(α-Sreptococcus)、コリネバクテリウム(Corynebacterium spp.)及びエンテロコッカス(Enterococcus spp.)が陽性であった。
なお、内部が無菌環境に維持されたアイソレーター内で飼育した子豚は、細菌の存在は検出されなかった。
帝王切開によれば、母豚からの経産道感染は免れ得る。
DPFではない環境で飼育した妊娠母豚から帝王切開により出生した子豚3頭について、2頭は内部を滅菌後の無菌環境を維持されたアイソレータ内で2週齢まで飼育した。
残りの1頭は、アイソレータ外飼育に相当する態様として内部が未滅菌のアイソレータ内で2週齢まで飼育した。
3頭それぞれの口腔、皮膚及び糞便について細菌の存在を検査した。
結果を下記表3に示す。
なお、内部が無菌環境に維持されたアイソレーター内で飼育した子豚は、細菌の存在は検出されなかった。
<参考例3>
抗体検査により経胎盤感染性病原体(豚サーコウイルス)を保有することを確認した妊娠母豚を用意し、SPF環境下で飼育した後、吸入麻酔を行い、常法により開腹後に腹式子宮摘出を行った。
図1に示したように、上記摘出した複数の子宮を消毒液を満たした消毒槽に投入、洗浄及び通過させてデコンタミネーションし、予め内部が滅菌済で無菌環境に維持された子豚取出し用アイソレータ内に収容した。
子豚取出し用アイソレータ内に予め配置しておいた滅菌済みのメス等をグローブ越しに操作して子宮及び羊膜を同時に切開して子豚を取出し常法により蘇生した(子豚3頭)。
取り出し蘇生された新生子豚3頭について血液を採取し、豚サーコウイルスの核酸に選択的に(好ましくは特異的に)結合するプライマー対を使用して豚サーコウイルスの感染の有無をPCR法により検査した。結果を下記表4に示す。
上記表4に示した結果から明らかなように、母豚が保有する豚サーコウイルスが子豚2頭に経胎盤感染したことが確認された。
本発明の製造方法に適用する母豚は、経胎盤感染性病原体による感染が陰性であることが好ましいと言える。
抗体検査により経胎盤感染性病原体(豚サーコウイルス)を保有することを確認した妊娠母豚を用意し、SPF環境下で飼育した後、吸入麻酔を行い、常法により開腹後に腹式子宮摘出を行った。
図1に示したように、上記摘出した複数の子宮を消毒液を満たした消毒槽に投入、洗浄及び通過させてデコンタミネーションし、予め内部が滅菌済で無菌環境に維持された子豚取出し用アイソレータ内に収容した。
子豚取出し用アイソレータ内に予め配置しておいた滅菌済みのメス等をグローブ越しに操作して子宮及び羊膜を同時に切開して子豚を取出し常法により蘇生した(子豚3頭)。
取り出し蘇生された新生子豚3頭について血液を採取し、豚サーコウイルスの核酸に選択的に(好ましくは特異的に)結合するプライマー対を使用して豚サーコウイルスの感染の有無をPCR法により検査した。結果を下記表4に示す。
本発明の製造方法に適用する母豚は、経胎盤感染性病原体による感染が陰性であることが好ましいと言える。
<実施例>
SPF環境下で飼育した複数の妊娠母豚のうち、経胎盤感染性病原体が陰性であることを確認した約20頭の妊娠母豚に吸入麻酔を行い、常法により開腹後に腹式子宮摘出を行った。上記妊娠母豚は、経胎盤感染性病原体以外の病原体の感染検査を行わなかったが、上記参考例1に記載の通りの傾向で陽性であるものと推定される。
図1に示したように、上記摘出した複数の子宮を消毒液を満たした消毒槽に投入、洗浄及び通過させてデコンタミネーションし、予め内部が滅菌済で無菌環境(ISOクラス5(クラス100)又はEU-GMPガイドライングレードA程度の清浄度の環境)に維持された子豚取出し用アイソレータ内に収容した。上記アイソレータの吸気部に高性能HEPAフィルターを設置し内部を無菌環境に維持した。
子豚取出し用アイソレータ内に予め配置しておいた滅菌済みのメス等をグローブ越しに操作して子宮及び羊膜を同時に切開して子豚を取出し常法により蘇生した。
SPF環境下で飼育した複数の妊娠母豚のうち、経胎盤感染性病原体が陰性であることを確認した約20頭の妊娠母豚に吸入麻酔を行い、常法により開腹後に腹式子宮摘出を行った。上記妊娠母豚は、経胎盤感染性病原体以外の病原体の感染検査を行わなかったが、上記参考例1に記載の通りの傾向で陽性であるものと推定される。
図1に示したように、上記摘出した複数の子宮を消毒液を満たした消毒槽に投入、洗浄及び通過させてデコンタミネーションし、予め内部が滅菌済で無菌環境(ISOクラス5(クラス100)又はEU-GMPガイドライングレードA程度の清浄度の環境)に維持された子豚取出し用アイソレータ内に収容した。上記アイソレータの吸気部に高性能HEPAフィルターを設置し内部を無菌環境に維持した。
子豚取出し用アイソレータ内に予め配置しておいた滅菌済みのメス等をグローブ越しに操作して子宮及び羊膜を同時に切開して子豚を取出し常法により蘇生した。
上記複数を3つの群に分け、1つの群について下記試験Aを行い、もう1つの群について下記試験Bを行い、残りの群については、後述するように飼育し、最終的には臓器摘出まで行った。
(試験A)麻酔開始後子宮摘出までの所要時間と、取り出される子豚の蘇生率との相関試験
上記吸入麻酔開始から子豚の取出しまで所要時間を計測し、上記所要時間と子豚蘇生率との相関を試験した。結果を図6に示す。
図6は、麻酔開始後子宮摘出までの所要時間と、取り出される子豚の蘇生率との相関を示す図である。
(試験Aの結果)
図6に示した結果から明らかなように、子宮摘出が吸引麻酔開始から25分を超えるNo.1~3の群は、それぞれ、蘇生率75%(蘇生個体3頭/取り出した個体4頭)、75%(蘇生個体3頭/取り出した個体4頭)、60%(蘇生個体3頭/取り出した個体5頭)であった。
一方、子宮摘出が吸引麻酔開始から25分以内のNo.4~6の群はいずれも、蘇生率100%であった(蘇生個体9頭/取り出した個体9頭、蘇生個体7頭/取り出した個体7頭、蘇生個体6頭/取り出した個体6頭)。
子宮摘出が吸引麻酔開始から25分以内が蘇生率の観点から好ましいと言える。
(試験A)麻酔開始後子宮摘出までの所要時間と、取り出される子豚の蘇生率との相関試験
上記吸入麻酔開始から子豚の取出しまで所要時間を計測し、上記所要時間と子豚蘇生率との相関を試験した。結果を図6に示す。
図6は、麻酔開始後子宮摘出までの所要時間と、取り出される子豚の蘇生率との相関を示す図である。
(試験Aの結果)
図6に示した結果から明らかなように、子宮摘出が吸引麻酔開始から25分を超えるNo.1~3の群は、それぞれ、蘇生率75%(蘇生個体3頭/取り出した個体4頭)、75%(蘇生個体3頭/取り出した個体4頭)、60%(蘇生個体3頭/取り出した個体5頭)であった。
一方、子宮摘出が吸引麻酔開始から25分以内のNo.4~6の群はいずれも、蘇生率100%であった(蘇生個体9頭/取り出した個体9頭、蘇生個体7頭/取り出した個体7頭、蘇生個体6頭/取り出した個体6頭)。
子宮摘出が吸引麻酔開始から25分以内が蘇生率の観点から好ましいと言える。
(試験B)取出し直後(出生直後)の新生子豚の病原体(ウイルス、原虫及び細菌)の存在(感染)の有無の試験
取り出された新生子豚6頭について血液を採取し、下記ウイルス67種、下記原虫1種及び細菌の存在(感染)の有無をPCR法により試験した。
具体的には、様々なウイルス(豚サーコウイルス1型、豚サーコウイルス2型、豚リンパ球向性ヘルペスウイルス1型、豚リンパ球向性ヘルペスウイルス2型、豚サイトメガロウイルス、脳心筋炎ウイルス、豚パルボウイルス、豚ロタウイルスA、豚ロタウイルスBアメリカ型、豚ロタウイルスBベトナム型、豚ロタウイルスC、哺乳類レオウイルス、豚エンテロウイルス、豚赤血球凝集性脳脊髄炎ウイルス、豚繁殖・呼吸障害症候群ウイルス、E型肝炎ウイルス、豚伝染性胃腸炎ウイルス、豚インフルエンザウイルスH1N1型、豚インフルエンザウイルスH3N2型、豚呼吸器コロナウイルス、豚熱ウイルス、豚ポックスウイルス、アフリカ豚熱ウイルス、オーエスキー病ウイルス、ゲタウイルス、日本脳炎ウイルス、豚流行性下痢ウイルス、豚水疱病ウイルス、豚水疱疹ウイルス、水疱性口炎ウイルスnew jersey、水疱性口炎ウイルスindiana、口蹄疫ウイルス、リッサ(狂犬病)ウイルス1、リッサウイルス2、リッサウイルス3、リッサウイルス4、リッサウイルス5、リッサウイルス6、リッサウイルス7、豚アデノウイルス、豚アストロウイルス1、豚アストロウイルス3、豚アストロウイルス4、豚アストロウイルス5、メナングルウイルス、ニパウイルス、西部ウマ脳炎ウイルス、ボルナウイルス、アポイウイルス、ウシウイルス性下痢ウイルス1型、ウシウイルス性下痢ウイルス2型、ウシウイルス性下痢ウイルス3型、ウシ伝染性鼻気管炎ウイルス、豚トルクテノウイルス1型、豚トルクテノウイルス2型、サポウイルス3型、サポウイルス6型、サポウイルス7a型、サポウイルス7b型、サポウイルス7c型、サポウイルス8型、サポウイルス9型、サポウイルス10型、ノロウイルスA型、ノロウイルスB型、豚ルブラウイルス、豚内在性レトロウルス)に対して各ウイルスの核酸に特異的に結合するプライマー対を使用して上記様々なウイルスによる感染の有無をPCR試験した(特開2021-023243号)。
取り出された新生子豚6頭について血液を採取し、下記ウイルス67種、下記原虫1種及び細菌の存在(感染)の有無をPCR法により試験した。

原虫(トキソプラズマ)に対して各原虫の核酸に特異的に結合するプライマー対を使用して上記原虫による感染の有無をPCR試験した(特開2021-023243号)。
16SリボソームRNA(rRNA)は全ての細菌が共有し、細菌種を超えて保存性が高い領域(例えば、保存領域C1~C9)を有する。
様々な細菌(エルシニア菌、気管支敗血症菌、クロストリジウム、結核(ヒト型、ウシ型、鳥型)、サルモネラ、大腸菌、炭疽菌、豚丹毒菌、パスツレラ菌、豚赤痢菌、ヘモフィルス菌、ブドウ球菌、ブルセラ菌、ヘモパルトネラ、マイコプラズマ、リステリア菌、アクチノバチルス菌、連鎖球菌、緑膿菌、豚クチノミセス、キャンピロバクター属、アクチノバチルス属、クラミジア、コクシエラ、ローソニア、レプトスピラ(レプトピラ)、豚ペロスロゾーン等)間の細菌16S rRNAの保存領域に含まれる共通塩基配列に特異的に結合するプライマー対を使用して様々な細菌による感染の有無をPCR試験した(特開2021-023243号)。

(試験Bの結果)
試験Bの結果を下記表5に示す。
上記表5に示した試験Bの結果から明らかなように、SPF環境下で飼育した妊娠母豚から摘出しデコンタミネーションした子宮から無菌環境下で取り出した子豚は、病原体(ウイルス、原虫及び細菌)の存在(感染)は検出されないことが確認された。
試験Bの結果を下記表5に示す。
(飼育)
取り出して蘇生した複数の子豚のうち、上記試験A及びBに供していない残りの1群については、上記子豚取出し用アイソレータと、新たに用意した滅菌済の無菌環境(ISOクラス5(クラス100)又はEU-GMPガイドライングレードA程度の清浄度の環境)に維持された飼育用アイソレータとを子豚移動用スリーブを介して連結し、スリーブ内の無菌環境空間を通じて、子豚を飼育用アイソレータに移動させた。上記飼育用アイソレータの吸気部に高性能HEPAフィルターを設置し内部を無菌環境に維持した。
飼育用のアイソレータ内部に、予め、人工哺育用の滅菌済みの、水、代用乳、人工乳、飼料により、3週齢(臓器摘出適期)になるまで複数の子豚を飼育した。

飼育用のアイソレータ内部に、予め、人工哺育用の滅菌済みの、水、代用乳、人工乳、飼料により、3週齢(臓器摘出適期)になるまで複数の子豚を飼育した。
(一般細菌及び真菌の培養検査)
3週齢(臓器摘出適期)の子豚5頭、及びそれが収容されていた飼育用のアイソレータについて一般細菌及び真菌(例えばトリコフィトン属他皮膚糸状菌)の培養検査を行った。結果を下記表6に示す。
上記表6に示した検査結果から明らかなように、本発明の製造方法において3週齢まで飼育された子豚は病原体に感染しておらず、DPF状態を維持しているといえる。
3週齢(臓器摘出適期)の子豚5頭、及びそれが収容されていた飼育用のアイソレータについて一般細菌及び真菌(例えばトリコフィトン属他皮膚糸状菌)の培養検査を行った。結果を下記表6に示す。
(子豚から膵臓摘出)
3週齢の上記子豚について、上記飼育用アイソレータと、新たに用意した滅菌済の無菌環境(ISOクラス5(クラス100)又はEU-GMPガイドライングレードA程度の清浄度の環境)に維持された臓器摘出用アイソレータとを子豚移動用スリーブを介して連結し、スリーブ内の無菌環境空間を通じて、子豚を臓器摘出用アイソレータに移動させた。上記臓器摘出用アイソレータの吸気部に高性能HEPAフィルターを設置し内部を無菌環境に維持した。
図3に示したように、臓器摘出用アイソレータ内に予め配置しておいた滅菌済みのメス等をグローブ越しに操作して開腹手術を行い、膵臓を摘出した。
上記摘出に際し、上記子豚の躯体中の上記臓器にETK液を注入して上記臓器を膨張させた状態で行うことにより作業性が大幅に向上した。
3週齢の上記子豚について、上記飼育用アイソレータと、新たに用意した滅菌済の無菌環境(ISOクラス5(クラス100)又はEU-GMPガイドライングレードA程度の清浄度の環境)に維持された臓器摘出用アイソレータとを子豚移動用スリーブを介して連結し、スリーブ内の無菌環境空間を通じて、子豚を臓器摘出用アイソレータに移動させた。上記臓器摘出用アイソレータの吸気部に高性能HEPAフィルターを設置し内部を無菌環境に維持した。
図3に示したように、臓器摘出用アイソレータ内に予め配置しておいた滅菌済みのメス等をグローブ越しに操作して開腹手術を行い、膵臓を摘出した。
上記摘出に際し、上記子豚の躯体中の上記臓器にETK液を注入して上記臓器を膨張させた状態で行うことにより作業性が大幅に向上した。
(摘出膵臓及び血液の病原体(ウイルス、原虫及び細菌)の存在(感染)の有無の試験)
上記摘出膵臓(及び血液)について、下記ウイルス61種、下記原虫1種及び細菌の存在(感染)の有無をPCR法により試験した。

上記摘出膵臓(及び血液)について、下記ウイルス61種、下記原虫1種及び細菌の存在(感染)の有無をPCR法により試験した。

Claims (8)
- 非DPF(Designated Pathogen Free)環境下で飼育された母豚から子宮を摘出すること、
摘出した前記子宮をデコンタミネーション処理後にアイソレータ内の無菌環境に収容すること、
前記子宮からの胎仔又は子豚の取出しをアイソレータ内の無菌環境で行うこと、
任意で前記胎仔又は子豚の蘇生及び飼育を含み、かつ、前記蘇生及び飼育をアイソレータ内の無菌環境で行い、及び
前記胎仔、子豚、又は子豚が生育した成豚からの臓器を摘出することを含む、
前記摘出した臓器がDPFである、
豚臓器又はその由来物を含む移植材の製造方法。 - 前記非DPF環境がSPF(Specific Pathogen Free)環境である、請求項1に記載の方法。
- 臓器の前記摘出が、前記子豚の躯体中の前記臓器に生理学的に許容される液を注入して前記臓器を膨張させた状態で行われる、請求項1に記載の方法。
- 子宮の前記摘出が、前記母豚への麻酔後に行われ、子宮の前記摘出が、前記麻酔開始後25分以内に終わる、請求項1に記載の方法。
- 前記麻酔が吸入麻酔である、請求項4に記載の方法。
- 前記母豚は、経胎盤感染性病原体による感染が陰性であり、かつそれ以外の病原体による感染が陽性又は陰性である、請求項1に記載の方法。
- 前記取出し及び蘇生と、前記飼育と、前記摘出とが、それぞれ、別個のアイソレータ内の無菌環境で行われ、
アイソレータ同士を連結したスリーブ内の無菌環境空間を通じて、子豚をアイソレータ間で移動させる、請求項1に記載の方法。 - 前記飼育のためのアイソレータが2以上存在し、前記飼育のための前記アイソレータを高さ方向に重ねて配置する、請求項7に記載の方法。
