Description
本発明は、ガラス粉末複合体、及びガラス粉末複合体の製造方法に関する。
従来から、機能性イオンを徐放するイオン徐放性ガラスが知られている。例えば、リンイオンやカルシウムイオンを水中に放出する無機ガラス粒子を粒状土壌改質材に用いる技術がある(特許文献1参照)。また、抗菌効果の知られている銀イオンを溶出する抗菌性ガラスを樹脂成形品に用いる技術がある(特許文献2)。また、骨修復のための材料や医療用インプラント等の医療用途でバイオガラスを用いる技術がある(特許文献3、特許文献4)。更に、イオン徐放性ガラスをデオドラント組成物に用いる技術がある(特許文献5参照)。
従来のイオン徐放性ガラスは、時間の経過に応じてイオンの徐放を変化させることが困難である。
本発明の課題は、時間の経過に応じてイオンの徐放を変化させることができるガラス粉末複合体を提供することである。
本発明の一態様に係るガラス粉末複合体は、少なくとも1種の元素を含有する第1ガラスを含み、第1期間に前記第1ガラスが溶出する第1ガラス粉末と、前記第1ガラスの前記元素と異なる少なくとも1種の元素を含有する第2ガラスを含み、前記第1期間と異なる第2期間に前記第2ガラスが溶出する第2ガラス粉末と、を含む。
本発明の一態様によれば、時間の経過に応じてイオンの徐放を変化させることができるガラス粉末複合体を提供することができる。
以下、本発明の実施の形態について、詳細に説明する。
<ガラス粉末複合体>
本実施形態に係るガラス粉末複合体は、第1ガラス粉末と第2ガラス粉末を含む。本明細書において、ガラス粉末複合体は、少なくとも2種類のガラス粉末を含むものを示す。

第1ガラス粉末は、少なくとも1種の元素を含有する第1ガラスを含む。
本明細書において、「ガラス」には、非結晶質のものの他、結晶質のものも含まれ、その一部にガラスを含むガラスセラミックも含まれる。ガラス粉末は、粉末状のガラスを示す。
なお、粉末のサイズは、粒度が0.01μm以上100μm以下であることが好ましく、0.1μm以上50μm以下であることがより好ましく、1μm以上20μm以下であることが更に好ましい。本明細書において、粒度は、レーザー回折・散乱法により測定される体積基準の累積分布が50%となるメジアン径(D50)を示す。
第1ガラスは、特に限定されないが、好ましくはリン酸塩ガラスである。リン酸塩ガラスは、リン(P)を含有することにより、ケイ酸塩ガラスやホウ酸塩ガラスに比べて網目形成成分が少ない組成でもガラス化し得る。そのため、リン酸塩ガラスは、ガラス中に多くの成分を含むことが可能であり、これらの成分が水に溶解してイオンの状態で徐々に放出されやすい(イオンを徐放しやすい又はイオン徐放性が高い)。
本明細書において、イオン徐放性は、ガラス粉末に含まれる成分が溶解してイオンの状態で徐々に放出される性質を示す。
リン酸塩ガラスは、五酸化リン(P2O5)を含有する。P2O5は、ガラス中で網目形成の役割を果たす。また、リン酸塩ガラスは、P2O5を含有することで、リン酸塩ガラスから放出されるリン酸イオン(PO4
3-)により、歯質の再石灰化や齲蝕を予防する効果を付与することができる。
リン酸塩ガラス中のP2O5の含有量は、40質量%以上であることが好ましく、より好ましくは45質量%以上、更に好ましくは50質量%以上である。リン酸塩ガラス中のP2O5の含有量が40質量%以上であることにより、ガラスが得られやすくなる。
なお、P2O5の含有量の上限は制限されないが、熔融温度が高くなりすぎずにガラスの製造を可能にする点で、90質量%以下であることが好ましく、より好ましくは80質量%以下、更に好ましくは70質量%以下である。
第1ガラスは、酸化ケイ素(SiO2)を全く含まないことが好ましく、SiO2を含む場合はSiO2の含有量が5質量%以下であることが好ましい。第1ガラスが、SiO2を全く含まない又はSiO2の含有量が5質量%以下であることで、第1ガラスにおけるイオン徐放性の低下を抑制することができる。
第1ガラスは、少なくとも1種の元素を含有する。第1ガラスに含まれる元素は、ガラスから徐放されるイオン(以下、「徐放性イオン」という)を形成する元素である。
第1ガラスに含まれる元素は、リチウム(Li)、カルシウム(Ca)、ストロンチウム(Sr)、バリウム(Ba)、イットリウム(Y)、ランタン(La)、セリウム(Ce)、プラセオジム(Pr)、ネオジム(Nd)、プロメチウム(Pm)、サマリウム(Sm)、ユウロピウム(Eu)、ガドリニウム(Gd)、テルビウム(Tb)、ジスプロシウム(Dy)、ホルミウム(Ho)、エルビウム(Er)、ツリウム(Tm)、イッテルビウム(Yb)、ルテチウム(Lu)、チタン(Ti)、ジルコニウム(Zr)、タンタル(Ta)、銅(Cu)、銀(Ag)、亜鉛(Zn)、ホウ素(B)、ガリウム(Ga)、スズ(Sn)、フッ素(F)などが挙げられる。これらの元素は、1種又は2種以上含有し得る。これらの中でも、Li、Ca、Sr、Cu、Ag、Zn、B、Ga、Fが好ましい。
これらの元素のうち、徐放したイオンが耐酸性を向上させる点で、Ca、Srが好ましく、酸の生成を抑制する点で、Znが好ましい。また、徐放性イオンが歯質の脱灰抑制効果を有する点で、Ca、Zn、Fが好ましく、骨形成を促進する点で、Ca、Srが好ましい。更に、徐放性イオンが抗菌効果を有する点で、Cu、Ag、Zn、B、Ga、Fが好ましく、徐放性イオンが抗炎症性作用を有する点で、Liが好ましい。
第1ガラスにLiが含まれる場合、第1ガラス中のLiの含有量は、特に限定されないが、酸化リチウム(Li2O)に換算した量で0質量%以上30質量%以下であることが好ましく、1質量%以上25質量%以下であることがより好ましく、5質量%以上20質量%以下であることが更に好ましい。第1ガラスがLiを含有することで、抗炎症作用を付与することができ、Li2Oの含有量が30質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第1ガラスにCaが含まれる場合、第1ガラス中のCaの含有量は、特に限定されないが、酸化カルシウム(CaO)に換算した量で0質量%以上25質量%以下であることが好ましく、0.5質量%以上10質量%以下であることがより好ましく、1質量%以上5質量%以下であることが更に好ましい。第1ガラスがCaを含有することで、耐酸性効果、歯質の脱灰抑制効果、骨形成促進作用を付与することができ、CaOの含有量が25質量%以下であることにより、ガラスが得られやすくなる。
第1ガラスにSrが含まれる場合、第1ガラス中のSrの含有量は、特に限定されないが、酸化ストロンチウム(SrO)に換算した量で0質量%以上50量%以下であることが好ましく、10質量%以上45質量%以下であることがより好ましく、20質量%以上40質量%以下であることが更に好ましい。第1ガラスがSrを含有することで、耐酸性効果、骨形成促進作用を付与することができ、SrOの含有量が50質量%以下であることにより、ガラスの熔融温度が高くなりすぎず、ガラスが得られやすくなる。
第1ガラスにCuが含まれる場合、第1ガラス中のCuの含有量は、特に限定されないが、酸化銅(CuO)に換算した量で0質量%以上10質量%以下であることが好ましく、0.01質量%以上8質量%以下であることがより好ましく、0.1質量%以上5質量%以下であることが更に好ましい。第1ガラスがCuを含有することで、抗菌効果を付与することができ、CuOの含有量が10質量%以下であることにより、ガラスが得られやすくなる。
第1ガラスにAgが含まれる場合、第1ガラス中のAgの含有量は、特に限定されないが、酸化銀(Ag2O)に換算した量で0質量%以上5質量%以下であることが好ましく、0.01質量%以上4質量%以下であることがより好ましく、0.1質量%以上3質量%以下であることが更に好ましい。第1ガラスがAgを含有することで、抗菌効果を付与することができ、Ag2Oの含有量が5質量%以下であることにより、ガラスが得られやすくなる。
第1ガラスにZnが含まれる場合、第1ガラス中のZnの含有量は、特に限定されないが、酸化亜鉛(ZnO)に換算した量で0質量%以上45質量%以下であることが好ましく、1質量%以上35質量%以下であることがより好ましく、5質量%以上30質量%以下であることが更に好ましい。第1ガラスがZnを含有することで、酸生成抑制効果、歯質の脱灰抑制効果、抗菌効果を付与することができ、ZnOの含有量が45質量%以下であることにより、ガラスが得られやすくなる。
第1ガラスにBが含まれる場合、第1ガラス中のBの含有量は、酸化ホウ素(B2O3)に換算した量で0質量%以上10質量%以下であることが好ましく、0.1質量%以上9質量%以下であることがより好ましく、1質量%以上8質量%以下であることが更に好ましい。第1ガラスがBを含有することで、抗菌効果を付与することができ、B2O3の含有量が10質量%以下であることにより、分相を抑え、ガラスが得られやすくなる。
第1ガラスにGaが含まれる場合、第1ガラス中のGaの含有量は、特に限定されないが、酸化ガリウム(Ga2O3)に換算した量で0質量%以上40質量%以下であることが好ましく、0.1質量%以上38質量%以下であることがより好ましく、1質量%以上35質量%以下であることが更に好ましい。第1ガラスがGaを含有することで、抗菌効果を付与することができ、Ga2O3の含有量が40質量%以下であることにより、ガラスが得られやすくなる。
第1ガラスにFが含まれる場合、第1ガラス中のFの含有量は、特に限定されないが、0質量%以上25質量%以下であることが好ましく、0.1質量%以上24質量%以下であることがより好ましく、1質量%以上22質量%以下であることが更に好ましい。第1ガラスがFを含有することで、歯質の脱灰抑制効果、抗菌効果を付与することができ、Fの含有量が25質量%以下であることにより、ガラスが得られやすくなる。
なお、本実施形態のガラス粉末複合体では、本発明の目的を損なわない限り、第1ガラスは、その他の元素として、ナトリウム(Na)、カリウム(K)、アルミニウム(Al)が含まれていてもよい。
第1ガラスにNaが含まれる場合、第1ガラス中のNaの含有量は、特に限定されないが、酸化ナトリウム(Na2O)に換算した量で1質量%以上40質量%以下であることが好ましく、3質量%以上35質量%以下であることがより好ましく、5質量%以上30質量%以下であることが更に好ましい。第1ガラスがNaを含有することで、ガラスの熔融温度を下げ、更にガラスの溶解性を高めることができ、Na2Oの含有量が40質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第1ガラスにKが含まれる場合、第1ガラス中のKの含有量は、特に限定されないが、酸化カリウム(K2O)に換算した量で1質量%以上40質量%以下であることが好ましく、3質量%以上35質量%以下であることがより好ましく、5質量%以上30質量%以下であることが更に好ましい。第1ガラスがKを含有することで、ガラスの熔融温度を下げ、更にガラスの溶解性を高めることができ、K2Oの含有量が40質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第1ガラスにAlが含まれる場合、第1ガラス中のAlの含有量は、特に限定されないが、酸化アルミニウム(Al2O3)に換算した量で0質量%以上20質量%以下であることが好ましく、0.5質量%以上15質量%以下であることがより好ましく、1質量%以上10質量%以下であることが更に好ましい。第1ガラスがAlを含有することで、第1ガラスの化学的耐久性が改善され、ガラスとしての安定性を高めることができ、Al2O3の含有量が20質量%以下であることにより、第1ガラスの粘性の調整が可能になる。
第1ガラス粉末は、第1期間に第1ガラスが溶出する。本明細書において、期間は、所定の時間を示す。また、「ガラスが溶出する」とは、ガラスが水に溶解して、ガラスに含まれる元素がイオンとして徐放することを示す。
第2ガラス粉末は、少なくとも1種の元素を含有する第2ガラスを含む。
第2ガラスは、特に限定されないが、好ましくはリン酸塩ガラスである。リン酸塩ガラスは、上述のように、ガラス中で網目形成の役割を果たす五酸化リン(P2O5)を含有する。
第2ガラスにおいて、リン酸塩ガラス中のP2O5の含有量は、40質量%以上であることが好ましく、より好ましくは45質量%以上、更に好ましくは50質量%以上である。リン酸塩ガラスは、P2O5を含有することで、リン酸塩ガラスから放出されるリン酸イオン(PO4
3-)により、歯質の再石灰化や齲蝕を予防する効果を付与することができ、リン酸塩ガラス中のP2O5の含有量が40質量%以上であることにより、ガラスが得られやすくなる。
なお、第2ガラスにおいて、P2O5の含有量の上限は制限されないが、熔融温度が高くなりすぎずにガラスの製造を可能にする点で、90質量%以下であることが好ましく、より好ましくは80質量%以下、更に好ましくは70質量%以下である。
第2ガラスは、酸化ケイ素(SiO2)を全く含まないことが好ましく、SiO2を含む場合はSiO2の含有量が5質量%以下であることが好ましい。第2ガラスが、SiO2を全く含まない又はSiO2の含有量が5質量%以下であることで、第2ガラスにおけるイオン徐放性の低下を抑制することができる。
第2ガラスは、第1ガラスの元素と異なる少なくとも1種の元素を含有する。第2ガラスに含まれる元素は、徐放性イオンを形成する元素である。
本明細書において、第1ガラスの元素と異なる元素とは、第2ガラスに含まれる元素の少なくとも1つが、第1ガラスに含まれていない元素であることを示す。例えば、第1ガラスに亜鉛(Zn)が含まれ、リチウム(Li)が含まれていない場合は、第2ガラスにはリチウム(Li)が含まれ、及び/又は、亜鉛(Zn)が含まれていないことを意味する。
第2ガラスに含まれる元素は、第1ガラスの元素と異なる元素であれば特に限定されないが、例えば、Li、Ca、Sr、Ba、Y、La、Ce、Pr、Nd、Pm、Sm、Eu、Gd、Tb、Dy、Ho、Er、Tm、Yb、Lu、Ti、Zr、Ta、Cu、Ag、Zn、B、Ga、Sn、Fなどが挙げられる。これらの元素は、1種又は2種以上含有し得る。これらの中でも、Li、Ca、Sr、Cu、Ag、Zn、B、Ga、Fが好ましい。
これらの元素のうち、徐放性イオンが耐酸性を向上させる点で、Ca、Srが好ましく、酸の生成を抑制する点で、Znが好ましい。また、徐放性イオンが歯質の脱灰抑制効果を有する点で、Ca、Zn、Fが好ましく、骨形成を促進する点で、Ca、Srが好ましい。更に、徐放性イオンが抗菌効果を有する点で、Cu、Ag、Zn、B、Ga、Fが好ましく、徐放性イオンが抗炎症性作用を有する点で、Liが好ましい。
第2ガラスにLiが含まれる場合、第2ガラス中のLiの含有量は、特に限定されないが、酸化リチウム(Li2O)に換算した量で0質量%以上30質量%以下であることが好ましく、1質量%以上25質量%以下であることがより好ましく、5質量%以上20質量%以下であることが更に好ましい。第1ガラスがLiを含有することで、抗炎症作用を付与することができ、Li2Oの含有量が30質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第2ガラスにCaが含まれる場合、第2ガラス中のCaの含有量は、特に限定されないが、酸化カルシウム(CaO)に換算した量で0質量%以上25質量%以下であることが好ましく、0.5質量%以上10質量%以下であることがより好ましく、1質量%以上5質量%以下であることが更に好ましい。第2ガラスがCaを含有することで、耐酸性効果、歯質の脱灰抑制効果、骨形成促進作用を付与することができ、CaOの含有量が25質量%以下であることにより、ガラスが得られやすくなる。
第2ガラスにSrが含まれる場合、第2ガラス中のSrの含有量は、特に限定されないが、酸化ストロンチウム(SrO)に換算した量で0質量%以上50量%以下であることが好ましく、10質量%以上45質量%以下であることがより好ましく、20質量%以上40質量%以下であることが更に好ましい。第2ガラスがSrを含有することで、耐酸性効果、骨形成促進作用を付与することができ、SrOの含有量が50質量%以下であることにより、ガラスの熔融温度が高くなりすぎず、ガラスが得られやすくなる。
第2ガラスにCuが含まれる場合、第2ガラス中のCuの含有量は、特に限定されないが、酸化銅(CuO)に換算した量で0質量%以上10質量%以下であることが好ましく、0.01質量%以上8質量%以下であることがより好ましく、0.1質量%以上5質量%以下であることが更に好ましい。第2ガラスがCuを含有することで、抗菌効果を付与することができ、CuOの含有量が10質量%以下であることにより、ガラスが得られやすくなる。
第2ガラスにAgが含まれる場合、第2ガラス中のAgの含有量は、特に限定されないが、酸化銀(Ag2O)に換算した量で0質量%以上5質量%以下であることが好ましく、0.01質量%以上4質量%以下であることがより好ましく、0.1質量%以上3質量%以下であることが更に好ましい。第2ガラスがAgを含有することで、抗菌効果を付与することができ、Ag2Oの含有量が5質量%以下であることにより、ガラスが得られやすくなる。
第2ガラスにZnが含まれる場合、第2ガラス中のZnの含有量は、特に限定されないが、酸化亜鉛(ZnO)に換算した量で0質量%以上45質量%以下であることが好ましく、1質量%以上35質量%以下であることがより好ましく、5質量%以上30質量%以下であることが更に好ましい。第2ガラスがZnを含有することで、酸生成抑制効果、歯質の脱灰抑制効果、抗菌効果を付与することができ、ZnOの含有量が45質量%以下であることにより、ガラスが得られやすくなる。
第2ガラスにBが含まれる場合、第2ガラス中のBの含有量は、酸化ホウ素(B2O3)に換算した量で0質量%以上10質量%以下であることが好ましく、0.1質量%以上9質量%以下であることがより好ましく、1質量%以上8質量%以下であることが更に好ましい。第2ガラスがBを含有することで、抗菌効果を付与することができ、B2O3の含有量が10質量%以下であることにより、分相を抑え、ガラスが得られやすくなる。
第2ガラスにGaが含まれる場合、第2ガラス中のGaの含有量は、特に限定されないが、酸化ガリウム(Ga2O3)に換算した量で0質量%以上40質量%以下であることが好ましく、0.1質量%以上38質量%以下であることがより好ましく、1質量%以上35質量%以下であることが更に好ましい。第2ガラスがGaを含有することで、抗菌効果を付与することができ、Ga2O3の含有量が40質量%以下であることにより、ガラスが得られやすくなる。
第2ガラスにFが含まれる場合、第2ガラス中のFの含有量は、特に限定されないが、0質量%以上25質量%以下であることが好ましく、0.1質量%以上24質量%以下であることがより好ましく、1質量%以上22質量%以下であることが更に好ましい。第2ガラスがFを含有することで、歯質の脱灰抑制効果、抗菌効果を付与することができ、Fの含有量が25質量%以下であることにより、ガラスが得られやすくなる。
なお、本実施形態のガラス粉末複合体では、本発明の目的を損なわない限り、第2ガラスは、その他の元素として、ナトリウム(Na)、カリウム(K)、アルミニウム(Al)が含まれていてもよい。
第2ガラスにNaが含まれる場合、第2ガラス中のNaの含有量は、特に限定されないが、酸化ナトリウム(Na2O)に換算した量で1質量%以上20質量%以下であることが好ましく、3質量%以上15質量%以下であることがより好ましく、5質量%以上10質量%以下であることが更に好ましい。第2ガラスがNaを含有することで、ガラスの熔融温度を下げ、更にガラスの溶解性を高めることができ、Na2Oの含有量が20質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第2ガラスにKが含まれる場合、第2ガラス中のKの含有量は、特に限定されないが、酸化カリウム(K2O)に換算した量で1質量%以上20質量%以下であることが好ましく、3質量%以上15質量%以下であることがより好ましく、5質量%以上10質量%以下であることが更に好ましい。第2ガラスがNaを含有することで、ガラスの熔融温度を下げ、更にガラスの溶解性を高めることができ、K2Oの含有量が20質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第2ガラスにAlが含まれる場合、第2ガラス中のAlの含有量は、特に限定されないが、酸化アルミニウム(Al2O3)に換算した量で0質量%以上20質量%以下であることが好ましく、0.5質量%以上15質量%以下であることがより好ましく、1質量%以上10質量%以下であることが更に好ましい。第2ガラスがAlを含有することで、第2ガラスの化学的耐久性が改善され、ガラスとしての安定性を高めることができ、Al2O3の含有量が20質量%以下であることにより、第2ガラスの粘性の調整が可能になる。
第2ガラス粉末は、第1期間と異なる第2期間に第2ガラスが溶出する。本明細書において、第1期間と異なる第2期間は、第2期間が第1期間より長くてもよく、第2期間が第1期間より短くてもよい。
例えば、第2期間が第1期間より長い場合、第2期間中の第1期間と重なる期間は、第1ガラス粉末の第1ガラスが溶出し、且つ第2ガラス粉末の第2ガラスが溶出する。また、第2期間中の第1期間が終了した後の期間は、第2ガラス粉末の第2ガラスが溶出する。
具体的には、第1ガラスがZnを含有し、第2ガラスがLiを含有する場合、第1期間中は第1ガラス粉末がZn2+を徐放し、且つ第2ガラス粉末がLi+を徐放し、第2期間中は、第2ガラス粉末がLi+を徐放する。
第1ガラスの残存率と第2ガラスの残存率との差は20%以上であることが好ましく、より好ましくは25%以上であり、更に好ましくは30%以上である。本明細書において、残存率は、溶解率ともいい、ガラスを水に浸漬させたときに、浸漬前のガラスの質量に対する浸漬後に溶解しなかったガラスの質量の割合を示し、下記の式に従って算出する。

ガラス粉末複合体に配合される第1ガラスと第2ガラスとの質量比は、特に限定されないが、例えば、10:1~1:10であり、好ましくは7:1~1:7であり、より好ましくは5:1~1:5である。
本実施形態のガラス粉末複合体では、第1ガラスに含まれる元素の少なくとも1つが、第2ガラスに含まれる元素の少なくとも1つと同一であってもよい。例えば、第1ガラスに亜鉛(Zn)が含まれている場合に、第2ガラスにも亜鉛(Zn)が含まれていてもよい。
溶解率は、ガラスを酢酸-酢酸ナトリウム緩衝液に溶解させ、下記の式に従って算出する。

本実施形態に係るガラス粉末複合体は、更に第3ガラス粉末を含んでいてもよい。
第3ガラス粉末は、少なくとも1種の元素を含有する第3ガラスを含む。
第3ガラスは、特に限定されないが、好ましくはリン酸塩ガラスである。リン酸塩ガラスは、上述のように、ガラス中で網目形成の役割を果たす五酸化リン(P2O5)を含有する。
第3ガラスにおいて、リン酸塩ガラス中のP2O5の含有量は、40質量%以上であることが好ましく、より好ましくは45質量%以上、更に好ましくは50質量%以上である。リン酸塩ガラスは、P2O5を含有することで、リン酸塩ガラスから放出されるリン酸イオン(PO4
3-)により、歯質の再石灰化や齲蝕を予防する効果を付与することができ、リン酸塩ガラス中のP2O5の含有量が40質量%以上であることにより、ガラスが得られやすくなる。
なお、第3ガラスにおいて、P2O5の含有量の上限は制限されないが、熔融温度が高くなりすぎずにガラスの製造を可能にする点で、90質量%以下であることが好ましく、より好ましくは80質量%以下、更に好ましくは70質量%以下である。
第3ガラスは、酸化ケイ素(SiO2)を全く含まないことが好ましく、SiO2を含む場合はSiO2の含有量が5質量%以下であることが好ましい。第3ガラスがSiO2を全く含まない又はSiO2の含有量が5質量%以下であることで、第3ガラスにおけるイオン徐放性の低下を抑制することができる。
第3ガラスは、第1ガラスの元素及び第2ガラスの元素と異なる少なくとも1種の元素を含有する。第3ガラスに含まれる元素は、徐放性イオンを形成する元素である。
本明細書において、第1ガラスの元素及び第2ガラスの元素と異なる元素とは、第3ガラスに含まれる元素の少なくとも1つが、第1ガラス及び第2ガラスの何れにも含まれていない元素であることを示す。例えば、第1ガラスには亜鉛(Zn)は含まれるが、ストロンチウム(Sr)は含まれておらず、第2ガラスにはリチウム(Li)が含まれるが、ストロンチウム(Sr)が含まれていない場合は、第3ガラスにはストロンチウム(Sr)が含まれるが、亜鉛(Zn)及びリチウム(Li)が含まれていないことを意味する。
第3ガラスに含まれる元素は、第1ガラスの元素及び第2ガラスの元素と異なる元素であれば特に限定されないが、例えば、Li、Ca、Sr、Ba、Y、La、Ce、Pr、Nd、Pm、Sm、Eu、Gd、Tb、Dy、Ho、Er、Tm、Yb、Lu、Ti、Zr、Ta、Cu、Ag、Zn、B、Ga、Sn、Fなどが挙げられる。これらの元素は、1種又は2種以上含有し得る。これらの中でも、Li、Ca、Sr、Cu、Ag、Zn、B、Ga、Fが好ましい。
これらの元素のうち、徐放性イオンが耐酸性を向上させる点で、Ca、Srが好ましく、酸の生成を抑制する点で、Znが好ましい。また、徐放性イオンが歯質の脱灰抑制効果を有する点で、Ca、Zn、Fが好ましく、骨形成を促進する点で、Ca、Srが好ましい。更に、徐放性イオンが抗菌効果を有する点で、Cu、Ag、Zn、B、Ga、Fが好ましく、徐放性イオンが抗炎症性作用を有する点で、Liが好ましい。
第3ガラスにLiが含まれる場合、第3ガラス中のLiの含有量は、特に限定されないが、酸化リチウム(Li2O)に換算した量で0質量%以上30質量%以下であることが好ましく、1質量%以上25質量%以下であることがより好ましく、5質量%以上20質量%以下であることが更に好ましい。第1ガラスがLiを含有することで、抗炎症作用を付与することができ、Li2Oの含有量が30質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第3ガラスにCaが含まれる場合、第3ガラス中のCaの含有量は、特に限定されないが、酸化カルシウム(CaO)に換算した量で0質量%以上25質量%以下であることが好ましく、0.5質量%以上10質量%以下であることがより好ましく、1質量%以上5質量%以下であることが更に好ましい。第3ガラスがCaを含有することで、耐酸性効果、歯質の脱灰抑制効果、骨形成促進作用を付与することができ、CaOの含有量が25質量%以下であることにより、ガラスが得られやすくなる。
第3ガラスにSrが含まれる場合、第3ガラス中のSrの含有量は、特に限定されないが、酸化ストロンチウム(SrO)に換算した量で0質量%以上50量%以下であることが好ましく、10質量%以上45質量%以下であることがより好ましく、20質量%以上40質量%以下であることが更に好ましい。第3ガラスがSrを含有することで、耐酸性効果、骨形成促進作用を付与することができ、SrOの含有量が50質量%以下であることにより、ガラスの熔融温度が高くなりすぎず、ガラスが得られやすくなる。
第3ガラスにCuが含まれる場合、第3ガラス中のCuの含有量は、特に限定されないが、酸化銅(CuO)に換算した量で0質量%以上10質量%以下であることが好ましく、0.01質量%以上8質量%以下であることがより好ましく、0.1質量%以上5質量%以下であることが更に好ましい。第3ガラスがCuを含有することで、抗菌効果を付与することができ、CuOの含有量が10質量%以下であることにより、ガラスが得られやすくなる。

第3ガラスにBが含まれる場合、第3ガラス中のBの含有量は、酸化ホウ素(B2O3)に換算した量で0質量%以上10質量%以下であることが好ましく、0.1質量%以上9質量%以下であることがより好ましく、1質量%以上8質量%以下であることが更に好ましい。第3ガラスがBを含有することで、抗菌効果を付与することができ、B2O3の含有量が10質量%以下であることにより、分相を抑え、ガラスが得られやすくなる。
第3ガラスにGaが含まれる場合、第3ガラス中のGaの含有量は、特に限定されないが、酸化ガリウム(Ga2O3)に換算した量で0質量%以上40質量%以下であることが好ましく、0.1質量%以上38質量%以下であることがより好ましく、1質量%以上35質量%以下であることが更に好ましい。第3ガラスがGaを含有することで、抗菌効果を付与することができ、Ga2O3の含有量が40質量%以下であることにより、ガラスが得られやすくなる。
第3ガラスにFが含まれる場合、第3ガラス中のFの含有量は、特に限定されないが、0質量%以上25質量%以下であることが好ましく、0.1質量%以上24質量%以下であることがより好ましく、1質量%以上22質量%以下であることが更に好ましい。第3ガラスがFを含有することで、歯質の脱灰抑制効果、抗菌効果を付与することができ、Fの含有量が25質量%以下であることにより、ガラスが得られやすくなる。
なお、本実施形態のガラス粉末複合体では、本発明の目的を損なわない限り、第3ガラスは、その他の元素として、ナトリウム(Na)、カリウム(K)、アルミニウム(Al)が含まれていてもよい。
第3ガラスにNaが含まれる場合、第3ガラス中のNaの含有量は、特に限定されないが、酸化ナトリウム(Na2O)に換算した量で1質量%以上20質量%以下であることが好ましく、3質量%以上15質量%以下であることがより好ましく、5質量%以上10質量%以下であることが更に好ましい。第3ガラスがNaを含有することで、ガラスの熔融温度を下げ、更にガラスの溶解性を高めることができ、Na2Oの含有量が20質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第3ガラスにKが含まれる場合、第3ガラス中のKの含有量は、特に限定されないが、酸化カリウム(K2O)に換算した量で1質量%以上20質量%以下であることが好ましく、3質量%以上15質量%以下であることがより好ましく、5質量%以上10質量%以下であることが更に好ましい。第3ガラスがNaを含有することで、ガラスの熔融温度を下げ、更にガラスの溶解性を高めることができ、K2Oの含有量が20質量%以下であることにより、水への溶解性の高すぎないガラスが得られやすくなる。
第3ガラスにAlが含まれる場合、第3ガラス中のAlの含有量は、特に限定されないが、酸化アルミニウム(Al2O3)に換算した量で0質量%以上20質量%以下であることが好ましく、0.5質量%以上15質量%以下であることがより好ましく、1質量%以上10質量%以下であることが更に好ましい。第3ガラスがAlを含有することで、第3ガラスの化学的耐久性が改善され、ガラスとしての安定性を高めることができ、Al2O3の含有量が20質量%以下であることにより、第3ガラスの粘性の調整が可能になる。
第3ガラス粉末では、第1期間及び第2期間と異なる第3期間に第3ガラスが溶出する。本明細書において、第1期間及び第2期間と異なる第3期間は、第3期間が第1期間及び第2期間より長くてもよく、第3期間が第1期間及び第2期間より短くてもよい。
例えば、第2期間が第1期間より長く、さらに第3期間が第2期間より長い場合、第3期間中の第1期間及び第2期間と重なる期間は、第1ガラス粉末の第1ガラスが溶出し、第2ガラス粉末の第2ガラスが溶出し、且つ第3ガラス粉末の第3ガラスが溶出する。また、第3期間中の第1期間が終了した後の期間は、第2ガラス粉末の第2ガラスが溶出し、且つ第3ガラス粉末の第3ガラスが溶出する。さらに、第3期間中の第2期間が終了した後の期間は、第3ガラス粉末の第3ガラスが溶出する。
具体的には、第1ガラスがZnを含有し、第2ガラスがLiを含有し、第3ガラスがSrを含有する場合、第1期間中は、第1ガラス粉末がZn2+を徐放し、第2ガラス粉末がLi+を徐放し、且つ第3ガラス粉末がSr2+を徐放し、第2期間中は、第2ガラス粉末がLi+を徐放し、且つ第3ガラス粉末がSr2+を徐放し、第3期間中は、第3ガラス粉末がSr2+を徐放する。
第2ガラスの残存率と第3ガラスの残存率との差は20%以上であることが好ましく、より好ましくは25%以上であり、更に好ましくは30%以上である。
本実施形態のガラス粉末複合体では、上述のように、第1期間に第1ガラスが溶出する第1ガラス粉末と、第1期間と異なる第2期間に第2ガラスが溶出する第2ガラス粉末とを含むことで、時間の経過に応じてイオンの徐放を変化させることができる。
また、本実施形態のガラス粉末複合体では、上述のように、第1ガラスの残存率と第2ガラスの残存率との差が20%以上であると、時間の経過に応じてイオンの徐放を変化させる効果が顕著になる。
本実施形態のガラス粉末複合体では、上述のように、第1ガラス及び/又は第2ガラスがリン酸塩ガラスであることで、第1ガラス及び/又は第2ガラスに含まれる元素のイオン徐放性を高めることができる。
本実施形態のガラス粉末複合体では、上述のように、第1ガラス及び/又は第2ガラスがナトリウム(Na)を含有することで、ガラスの熔融温度を下げ、更にガラスの溶解性(イオン徐放性)を高めることができる。
本実施形態のガラス粉末複合体では、上述のように、第1ガラス及び/又は第2ガラスがアルミニウム(Al)を含有することで、第1ガラス及び/又は第2ガラスの化学的耐久性が改善され、ガラスとしての安定性を高めることができる。
本実施形態のガラス粉末複合体では、上述のように、第1ガラス又は第2ガラスが、リチウム(Li)を含有することで、抗炎症作用を付与することができる。
本実施形態のガラス粉末複合体では、上述のように、第1ガラス又は第2ガラスが亜鉛(Zn)を含有することで、酸生成抑制効果、歯質の脱灰抑制効果、抗菌効果を付与することができる。
本実施形態のガラス粉末複合体では、上述のように、第1ガラス又は第2ガラスがストロンチウム(Sr)を含有することで、耐酸性効果、骨形成促進作用を付与することができる。
本実施形態のガラス粉末複合体では、上述のように、第1期間及び第2期間と異なる第3期間に第3ガラスが溶出する第3ガラス粉末を更に含むことで、時間の経過に応じてイオンの徐放を変化させる効果のバリエーションを増やすことができる。
本実施形態のガラス粉末複合体の用途としては、特に限定されない。例えば、時間の経過により抗菌性を発揮する樹脂や塗膜、土壌改質材が挙げられる。また、時間の経過により変化が起こる骨破壊又は骨形成に適応する骨補填材や、口腔内の病態の変化に適応する歯科材料が挙げられる。
ガラス粉末複合体の用途の一例として、本実施形態のガラス粉末複合体を歯科材料に用いる場合の歯科用組成物について説明する。本実施形態に係る歯科用組成物は、上述のガラス粉末複合体を含有する。本明細書において、歯科用組成物とは、歯科材料に用いられる組成物を示す。
具体的には、歯科用組成物に用いられるガラス粉末複合体が、少なくとも1種の元素を含有する第1ガラスを含み、第1期間に第1ガラスが溶出する第1ガラス粉末と、第1ガラスの元素と異なる少なくとも1種の元素を含有する第2ガラスを含み、第1期間と異なる第2期間に第2ガラスが溶出する第2ガラス粉末とを含む。
例えば、第2期間が第1期間より長い場合、第2期間中の第1期間と重なる期間は、口腔内で第1ガラス粉末の第1ガラスが溶出し、且つ第2ガラス粉末の第2ガラスが溶出する。また、第2期間中の第1期間が終了した後の期間は、口腔内で第2ガラス粉末の第2ガラスが溶出する。
具体的には、第1ガラスがZnを含有し、第2ガラスがLiを含有する場合、口腔内で第1期間中は第1ガラス粉末がZn2+を徐放し、且つ第2ガラス粉末がLi+を徐放し、第2期間中は、第2ガラス粉末がLi+を徐放する。
また、歯科用組成物に用いられるガラス粉末複合体は、第1ガラスの残存率と第2ガラスの残存率との差は20%以上であることが好ましく、より好ましくは25%以上であり、更に好ましくは30%以上である。
本実施形態の歯科用組成物では、上述のガラス粉末複合体を含むことで、上述のガラス粉末複合体の効果が得られる。すなわち、歯科用組成物に含まれるガラス粉末複合体が、第1期間に第1ガラスが溶出する第1ガラス粉末と第1期間と異なる第2期間に第2ガラスが溶出する第2ガラス粉末とを含むことで、口腔内での時間の経過に応じてイオンの徐放を変化させることができる。
また、本実施形態の歯科用組成物では、上述のように第1ガラスの残存率と第2ガラスの残存率との差が20%以上であるガラス粉末複合体を含むことで、口腔内で時間の経過に応じてイオンの徐放を変化させる効果が顕著になる。
なお、本実施形態の歯科用組成物は、上述のガラス粉末複合体以外の他の成分を含有していてもよい。本実施形態の歯科用組成物に含まれるガラス粉末複合体以外の他の成分としては、特に限定されないが、例えば、ポリアクリル酸等の高分子を含有することができる。歯科用組成物は、このようなガラス粉末複合体とポリアクリル酸等の高分子とを混合することで、アイオノマーセメントを構成することができる。
また、本実施形態の歯科用組成物は、本発明の目的を損なわない限り、その他の任意の成分として、必要に応じて塩酸塩、硫酸塩等の硬化促進剤、炭化水素類、高級脂肪酸類、エステル類等の油性成分、各種の無機あるいは有機の着色剤、抗菌材、香料等を含有してもよい。
本実施形態の歯科用組成物は、各種歯科材料に用いることができる。本実施形態の歯科用組成物の用途は、特に限定されないが、例えば、歯科用セメント、歯科用接着剤、歯科用仮封材、歯科用仮着材、歯科用プライマー、歯科用コート剤、根面被覆材、歯科用コンポジットレジン、歯科用硬質レジン、歯科切削加工用レジン材料、歯科用暫間修復材、歯科用充填剤、覆髄材、骨補填材、骨セメント、義歯床材料、人工歯、歯磨剤などが挙げられる。
本実施形態に係るガラス粉末複合体の製造方法は、上述のガラス粉末複合体を実質的に製造する方法である。具体的には、少なくとも1種の元素を含有する第1ガラスを含み、第1期間に第1ガラスが溶出する第1ガラス粉末と、第1ガラスの元素と異なる少なくとも1種の元素を含有する第2ガラスを含み、第1期間と異なる第2期間に第2ガラスが溶出する第2ガラス粉末とを混合する。
本実施形態に係るガラス粉末複合体の製造方法における第1ガラス粉末と第2ガラス粉末とを混合する工程では、上述のガラス粉末複合体に含まれる第1ガラス粉末及び第2ガラス粉末が用いられる。そのため、本実施形態に係る製造方法で得られたガラス粉末複合体は、上述のガラス粉末複合体の効果が得られる。
すなわち、得られるガラス粉末複合体は、少なくとも1種の元素を含有する第1ガラスを含み、第1期間に第1ガラスが溶出する第1ガラス粉末と、第1ガラスの元素と異なる少なくとも1種の元素を含有する第2ガラスを含み、第1期間と異なる第2期間に第2ガラスが溶出する第2ガラス粉末とを含有し、第1ガラス粉末及び第2ガラス粉末が、何れもイオン徐放性を有するガラス粉末複合体である。そのため、本実施形態に係る製造方法によれば、時間の経過に応じてイオンの徐放を変化させることができるガラス粉末複合体を得ることができる。
以下、本発明について、更に実施例を用いて説明する。各種の試験及び評価は、下記の方法に従う。なお、以下において、単位のない数値又は「%」は、特に断りのない限り、質量基準である。
<ガラス粉末の調製>
ガラス粉末(ガラス1~9)を、表1に示す組成で調製した。まず、原料を秤量し、乳鉢で10分混合し、白金るつぼに入れ、1100℃で1時間熔融し、融液をアイロンプレスにより冷却した。これをボールミルで30分粉砕し(エタノール湿式粉砕、40mmアルミナボール、100rpm)、さらにボールミルで30分粉砕(エタノール湿式粉砕、5mmアルミナボール、100rpm)した。その後、遠心分離によりガラス粉末を回収し、残ったエタノールを減圧乾燥(ゲージ圧:-0.1MPa、40℃)で飛ばし、ガラス粉末(ガラス1~9)を得た。
ガラス粉末(ガラス1~9)を、表1に示す組成で調製した。まず、原料を秤量し、乳鉢で10分混合し、白金るつぼに入れ、1100℃で1時間熔融し、融液をアイロンプレスにより冷却した。これをボールミルで30分粉砕し(エタノール湿式粉砕、40mmアルミナボール、100rpm)、さらにボールミルで30分粉砕(エタノール湿式粉砕、5mmアルミナボール、100rpm)した。その後、遠心分離によりガラス粉末を回収し、残ったエタノールを減圧乾燥(ゲージ圧:-0.1MPa、40℃)で飛ばし、ガラス粉末(ガラス1~9)を得た。
<ガラス粉末の組成>
得られたガラス粉末(ガラス1~9)の組成を確認した。蛍光X線分析装置(リガク社製、ZSX Primus IV)を用いて、ガラス粉末(塩ビリングで成形)を分析し、ガラス粉末の組成を求めた。結果を表1に示す。

<ガラス粉末の粒度>
得られたガラス粉末(ガラス1~9)の粒度を測定した。ガラス粉末の粒度は、レーザー回折・散乱法により測定される体積基準の累積分布が50%となるメジアン径(D50)である。ガラス粉末をエタノール中に分散させ、レーザー回折・散乱型粒度分布計(堀場製作所社製、Partica(登録商標) LA-960V2)を用いて測定した。何れのガラス粉末もD50が10±2μmであることを確認した。
得られたガラス粉末(ガラス1~9)の粒度を測定した。ガラス粉末の粒度は、レーザー回折・散乱法により測定される体積基準の累積分布が50%となるメジアン径(D50)である。ガラス粉末をエタノール中に分散させ、レーザー回折・散乱型粒度分布計(堀場製作所社製、Partica(登録商標) LA-960V2)を用いて測定した。何れのガラス粉末もD50が10±2μmであることを確認した。
<ガラスの溶解率>
50mgのガラス粉末を10mLの蒸留水(pH7)に浸漬し、37℃で保存し、その後、0.2μmシリンジフィルターでろ過し、ろ過後のガラスをシリンジフィルターとともに60℃下で48時間乾燥した。その後、シリンジフィルターおよびガラスの重量を測定し、ガラスの溶解量および溶解率(残存率)を算出した。ガラス粉末(ガラス1~9)の残存率を図1及び表1に示す。
50mgのガラス粉末を10mLの蒸留水(pH7)に浸漬し、37℃で保存し、その後、0.2μmシリンジフィルターでろ過し、ろ過後のガラスをシリンジフィルターとともに60℃下で48時間乾燥した。その後、シリンジフィルターおよびガラスの重量を測定し、ガラスの溶解量および溶解率(残存率)を算出した。ガラス粉末(ガラス1~9)の残存率を図1及び表1に示す。
ガラスの溶解率は、ガラス粉末複合体を蒸留水に1日浸漬後のガラス残存率と定義し、このガラス残存率からガラス粉末複合体を構成するガラス粉末の溶解速度を下記(1)~(3)の3種類に分類した。
(1)fast:残存率5%未満(数時間で溶解)
(2)normal:残存率5%以上60%未満(数日で溶解)
(3)slow:残存率60%以上(数週で溶解)
(1)fast:残存率5%未満(数時間で溶解)
(2)normal:残存率5%以上60%未満(数日で溶解)
(3)slow:残存率60%以上(数週で溶解)
<ガラス粉末複合体の溶解性>
表1から、ガラス粉末を2種類以上組み合わせて、段階的に1種類以上の異なるイオンが溶出するガラス粉末複合体を調製した。上記(1)~(3)のガラス粉末を組み合わせたガラス粉末複合体(実施例1~5、比較例1~5)を表2に示し、ガラス粉末複合体の残存率を図2~図11に示す。
表1から、ガラス粉末を2種類以上組み合わせて、段階的に1種類以上の異なるイオンが溶出するガラス粉末複合体を調製した。上記(1)~(3)のガラス粉末を組み合わせたガラス粉末複合体(実施例1~5、比較例1~5)を表2に示し、ガラス粉末複合体の残存率を図2~図11に示す。
表1、表2より、溶解速度が異なる(残存率の差が20%以上の)ガラス粉末を2種類以上組み合わせたガラス粉末複合体は、1種類以上の異なるイオンを時間差で溶出できることが分かる(実施例1~5)。
一方、溶解速度が同程度の(溶解率の差が20%未満の)ガラス粉末を2種類以上組み合わせたガラス粉末複合体は、1種類以上の異なるイオンを時間差で溶出することはできないことが分かる(比較例1~5)。
以上より、少なくとも1種の元素を含有する第1ガラスを含み、第1期間に第1ガラスが溶出する第1ガラス粉末と、第1ガラスの元素と異なる少なくとも1種の元素を含有する第2ガラスを含み、第1期間と異なる第2期間に第2ガラスが溶出する第2ガラス粉末と、を含むガラス粉末複合体は、時間の経過に応じてイオンの徐放を変化させることができる。
