Abstract
本発明により、医薬品として有用な式(I)で表される化合物、または医薬として許容されるその塩が提供される。さらに本発明により、シヌクレイノパチーの治療または予防のための医薬組成物が提供される。
Description
本発明は、脂肪酸結合タンパク質に結合するトリフェニルアゾール化合物に関する。さらに本発明は、該化合物を含有する医薬組成物、および該化合物を使用する治療方法または予防方法に関する。
α-シヌクレインはα-シヌクレイン遺伝子(SNCA)によってエンコードされるアミノ酸140残基からなるタンパク質であり、脳内のシナプス前末端において豊富に発現している。α-シヌクレインの異常蓄積を特徴とする神経変性疾患群はシヌクレイノパチーと呼ばれ、レビー小体型認知症(DLB)でのレビー小体・神経突起における蓄積、多系統萎縮症のグリア細胞質封入体における蓄積などが知られている。α-シヌクレインのオリゴマー化において多価不飽和脂肪酸が関与することについて報告がされている(非特許文献1)。本格的な高齢化社会を迎えている我が国においてシヌクレイノパチーに該当する疾患の患者数は近年大きく増加している。
脂肪酸結合タンパク質(Fatty Acid Binding Protein: FABP)は、分子量14~15kDaの細胞質型タンパク質であり、組織特異的に発現する。FABPは中鎖~長鎖脂肪酸をリガンドとし、脂質代謝の恒常性維持やシグナル伝達に関与すると考えられている。FABPは、分子構造が互いに類似している複数のサブタイプが知られている。
FABP3は、脳、心臓、骨格筋、乳腺、胎盤において発現しており(非特許文献2)、その機能は完全には解明されていないものの、脂質の取り込み、ミトコンドリアでのβ-酸化系への輸送などの脂質ホメオスタシスに関与しているほか、脳内においては神経の興奮と抑制のバランスを制御しているものと考えられている。
また、FABP3がα-シヌクレイン凝集を促進することについて報告がされている(非特許文献4および5)。
FABP3は、脳、心臓、骨格筋、乳腺、胎盤において発現しており(非特許文献2)、その機能は完全には解明されていないものの、脂質の取り込み、ミトコンドリアでのβ-酸化系への輸送などの脂質ホメオスタシスに関与しているほか、脳内においては神経の興奮と抑制のバランスを制御しているものと考えられている。
また、FABP3がα-シヌクレイン凝集を促進することについて報告がされている(非特許文献4および5)。
α-シヌクレイン凝集体(封入体)がパーキンソン病(PD)では黒質ドーパミン神経に発現し、レビー小体型認知症(DLB)では大脳皮質に瀰漫性に発現する。線維状シヌクレインをラット線条体に注入すると、黒質に加えて大脳皮質に凝集体が伝播して、神経細胞にシヌクレイン封入体を形成するという報告がある(非特許文献3)。
FABP3はドーパミン作動性神経細胞に高発現し、in vivo および in vitroでドーパミン神経毒である1-メチル-4-フェニル-1,2,3,6-テトラヒドロピリジン(MPTP) 処置後のα-シヌクレインオリゴマー化とドーパミン作動性神経細胞死を促進するが、FABP3欠損マウスにMPTPを投与しても神経毒作用を発現しないこと(非特許文献5)、FABP3の阻害活性を有するFABP3リガンドであるLigand 1化合物(MF1ともいう)がドーパミン作動性神経細胞をα-シヌクレインのオリゴマー化による細胞死から保護することが報告されている(特許文献1および非特許文献6)。

シヌクレイノパチーとして分類される神経変性疾患に対しては、十分な効果が得られる治療方法および予防方法が確立しているとは言えず、新規な治療剤および予防剤が求められている。
一つの側面において、本発明はシヌクレイノパチーの治療または予防に用いるための医薬組成物の提供を目的とする。さらに本発明は特定のトリフェニルアゾール化合物を用いたシヌクレイノパチーの治療方法または予防方法の提供を目的とする。
本発明者らは、上記課題を達成するために鋭意研究を進めたところ、トリフルオロメチル基等のハロC1-6アルキル基を導入することで、トリフェニルアゾール化合物がα-シヌクレインの凝集を抑制すること、運動機能障害および認知機能障害を改善することおよび神経保護作用を有することなどの有益な効果を有することを見出し、本発明を完成させた。
さらに、本発明のトリフェニルアゾール化合物は、予想外にも公知FABP3リガンドであるLigand 1化合物(特許文献1)よりもFABP3への親和性が高いことを見出した。
本明細書の開示は、以下の[1-1]~[1-31]および[2-1]~[2-31]に記載の発明を包含する。
さらに、本発明のトリフェニルアゾール化合物は、予想外にも公知FABP3リガンドであるLigand 1化合物(特許文献1)よりもFABP3への親和性が高いことを見出した。

以下、本発明を更に具体的に説明する。
本発明の1つの側面によれば、式(I)または式(IX)で表される化合物、または医薬として許容されるその塩を有効成分として含有する、シヌクレイノパチーまたは障害の治療または予防のための医薬組成物が提供される。1つの態様において、式(Ia)または式(Ib)で表される化合物を有効成分として含有する、シヌクレイノパチーまたは障害の治療または予防のための医薬組成物が提供される。本明細書において、式(I)または式(IX)で表される化合物には、式(Ia)または式(Ib)で表される化合物が包含される。
本明細書において「C1-6アルキル」とは、炭素数1~6の直鎖状、分岐鎖状、環状または部分的に環状のアルキル基を意味し、例えば、メチル、エチル、n-プロピル、iso-プロピル、n-ブチル、sec-ブチル、iso-ブチル、tert-ブチル、n-ペンチル、3-メチルブチル、2-メチルブチル、1-メチルブチル、1-エチルプロピル、n-ヘキシル、4-メチルペンチル、3-メチルペンチル、2-メチルペンチル、1-メチルペンチル、3-エチルブチル、および2-エチルブチル、シクロプロピル、シクロブチル、シクロペンチル、シクロヘキシル、およびシクロプロピルメチルなどが含まれ、例えば、C1-4アルキルおよびC1-3アルキルなども含まれる。

ハロゲン原子の例としては、フッ素原子(F)、塩素原子(Cl)、臭素原子(Br)、ヨウ素原子(I)などが挙げられる。
本明細書において「ハロC1-6アルキル」とは、1つ以上のハロゲン原子に置換されたアルキルを意味し、例えば、フルオロメチル、ジフルオロメチル、トリフルオロメチル、クロロメチル、ジクロロメチル、トリクロロメチル、ヨウ化メチルなどが挙げられる。
本明細書において「ハロC1-6アルキル」とは、1つ以上のハロゲン原子に置換されたアルキルを意味し、例えば、フルオロメチル、ジフルオロメチル、トリフルオロメチル、クロロメチル、ジクロロメチル、トリクロロメチル、ヨウ化メチルなどが挙げられる。
式(I)、(Ia)、(Ib)または(IX)においてmが0の場合、R3で示される置換基は存在しない。さらにmが2以上の場合、R3で示される置換基は同一であっても異なっていてもよい。また式(I)、(Ia)または(IX)においてnが0の場合、R4で示される置換基は存在しない。さらにnが2以上の場合、R4で示される置換基は同一であっても異なっていてもよい。また式((IX)においてpが0の場合、R6で示される置換基は存在しない。さらにpが2の場合、R6で示される置換基は同一であっても異なっていてもよい。
本明細書において式(I)または式(IX)のHetは、少なくとも1つの窒素原子を有する5員含窒素芳香族ヘテロ環を表す。ここでHetに環原子として含まれる1つの窒素原子と、2つの炭素原子はフェニル基で置換されており、各フェニル基は式(I)または式(IX)で示される置換基を有していてもよい。5員含窒素芳香族ヘテロ環であるここで5員含窒素芳香族ヘテロ環の例としては、ピロール、イミダゾール、ピラゾール、およびトリアゾールが挙げられる。本明細書において式(Ia)で表される化合物は、以下の式(Ia-1)、(Ia-2)、(Ia-3)、および(Ia-4)で表される化合物または医薬として許容されるその塩を包含する。
式(I)または式(IX)の化合物または医薬として許容されるその塩が水和物などの溶媒和物を形成する場合には、本発明は該溶媒和物を用いて実施することができる。さらに本発明の化合物または医薬として許容されるその塩は、混合物、溶液、結晶多形などとして適宜実施することができる。

式(I)または式(IX)の化合物の「医薬として許容されるその塩」とは、医薬品として使用可能な塩であれば特に限定されない。本発明化合物が塩基と形成する塩としては、ナトリウム、カリウム、マグネシウム、カルシウム、アルミニウムなどの無機塩基との塩;メチルアミン、エチルアミン、エタノールアミン等の有機塩基との塩などが挙げられる。
本発明の1つの態様において、式(I)または式(IX)の化合物、そのエナンチオマー、そのジアステレオマー、または医薬として許容されるその塩は、プロドラッグとして投与され、生体内において活性化合物に変換される。
本発明におけるシヌクレイノパチーとは、α-シヌクレインの異常蓄積を特徴とする神経変性疾患を意味し、例えば、PD、DLB、または多系統萎縮症が挙げられ、本発明の医薬組成物および方法は、例えば、シヌクレイノパチーの進行の抑制のために使用することができる。
本発明の医薬組成物および方法において、処置するシヌクレイノパチーの進行度、重症度および病態については特に限定されない。PD重症度分類はヤールI~V度が用いられる。DLBには初期、中期、後期の段階があり、α-シヌクレイン凝集体の発現部位から脳幹優位型、辺縁型、新皮質型に分けられる。さらに、多系統萎縮症の重症度分類ではmodified Ranking Scale 1~6が用いられ、病態により、Shy Drager 症候群、オリーブ橋小脳萎縮症、線条体鉱質変性症などに分類される。いずれもα-シヌクレイン凝集体が神経細胞あるいはグリア細胞に発現する疾患群であり、本発明の医薬組成物および方法はこれらの疾患群から選択される疾患の治療または予防に使用することができる。
前頭皮質あるいは海馬において、神経細胞死がおこることで認知症が発症すると考えられる。本発明ではFABP3リガンドがα-シヌクレインの凝集を抑制することを提唱する。その結果、脳内でのα-シヌクレイン凝集体による神経細胞死が抑制される。
本発明の医薬組成物および方法は、特にPDおよびDLBの治療、および予防に用いることができる。
また、本発明の医薬組成物および方法は、運動機能障害、認知機能障害、自閉症、および統合失調症などの精神疾患の改善および進行抑制をすることができる。
運動機能障害としては、PDにおける振戦(手や足のふるえ)、無動(動きが遅くなる)、固縮(筋肉がかたくなってこわばり、関節の曲げ伸ばしに抵抗がある)、姿勢反射障害(体のバランスがとりにくくなる)、DLBにおいても、パーキンソン症状が、 多系統萎縮症においては、パーキンソン症状に加え、自律神経障害(排尿障害、勃起障害、起立性低血圧、発汗低下など)、小脳性運動失調(失調性歩行と構音障害、四肢の運動失調、もしくは小脳性眼球運動障害)などが挙げられる。
認知機能障害としては、DLBにおいては、記憶障害、前頭葉・頭頂葉機能障害(注意障害、視空間障害、構成障害、実行機能障害など)、認知機能の動揺、幻視などが挙げられる。PDにおいては、初期では、遂行機能(プランニング、セットの変換・保持、問題解決など)、作動記憶、手続き記憶などの記憶機能,視空間機能の高次脳機能障害が見られる場合があり、進行すると幻視や視空間認知障害、思考緩慢等、DLBと類似した症候が見られる場合がある。多系統萎縮症においては、物忘れなど認知症が見られる場合、などが挙げられる。
また、本発明の医薬組成物および方法は神経変性疾患の治療、および予防に用いることができる。
神経変性疾患としては、筋萎縮性側索硬化症(ALS)、PD、アルツハイマー型認知症、DLB、ギランバレー症候群、慢性炎症性脱髄性多発神経炎、多発性硬化症、視神経脊髄炎などが挙げられる。

また、本発明の医薬組成物および方法は神経細胞保護作用を有するため、脳梗塞、脳出血、くも膜下出血などの脳血管障害、頭部外傷の予防および治療に用いることができる。

本発明の医薬組成物は、種々の剤形、例えば、経口投与のためには、錠剤、カプセル剤、散剤、顆粒剤、丸剤、液剤、乳剤、懸濁液、溶液剤、酒精剤、シロップ剤、エキス剤、エリキシル剤とすることができ、非経口剤としては、例えば、皮下注射剤、静脈内注射剤、筋肉内注射剤、腹腔内注射剤などの注射剤;経皮投与または貼付剤、軟膏またはローション;口腔内投与のための舌下剤、口腔貼付剤;ならびに経鼻投与のためのエアゾール剤とすることができるが、これらには限定されない。これらの製剤は、製剤工程において通常用いられる公知の方法により製造することができる。
当該医薬組成物は、一般に用いられる各種成分を含みうるものであり、例えば、1種以上の薬学的に許容され得る賦形剤、崩壊剤、希釈剤、滑沢剤、着香剤、着色剤、甘味剤、矯味剤、懸濁化剤、湿潤剤、乳化剤、分散剤、補助剤、防腐剤、緩衝剤、結合剤、安定剤、コーティング剤等を含みうる。また本発明の医薬組成物は、持続性または徐放性剤形であってもよい。
本発明の医薬組成物の投与量は、投与経路、患者の体型、年齢、体調、疾患の度合い、発症後の経過時間等により、適宜選択することができ、本発明の医薬組成物は、治療有効量および/または予防有効量の上記式(I)または式(IX)の化合物を含むことができる。本発明において上記式(I)または式(IX)の化合物は、一般に1μg~1000mg/日/成人、例えば、1μg~200mg/日/成人、具体的には5μg~100mg/日/成人、より具体的には10μg~50mg/日/成人の用量で使用されうる。当該医薬組成物の投与は、単回投与または複数回投与であってもよい。
本発明の医薬組成物は、必要に応じ、従来公知の着色剤、保存剤、香料、風味剤、コーティング剤、抗酸化剤、ビタミン、アミノ酸、ペプチド、タンパク質、およびミネラル分(鉄、亜鉛、マグネシム、ヨードなど)などの成分を含有していてもよい。本発明の医薬組成物は、経口投与に適した形態、例えば顆粒剤(ドライシロップを含む)、カプセル剤(軟カプセル剤、硬カプセル剤)、錠剤(チュアブル剤などを含む)、散剤(粉末剤)、丸剤などの各種の固形製剤、または内服用液剤(液剤、懸濁剤、シロップ剤を含む)などの液状製剤などの形態で調製してもよい。
製剤化のための添加物としては、例えば、賦形剤、滑沢剤、結合剤、崩壊剤、流動化剤、分散剤、湿潤剤、防腐剤、粘稠剤、pH調整剤、着色剤、矯味矯臭剤、界面活性剤、溶解補助剤が挙げられる。また、液剤の形態にする場合は、ペクチン、キサンタンガム、グアガムなどの増粘剤を配合することができる。また、コーティング剤を用いてコーティング錠剤にしたり、ペースト状の膠剤とすることもできる。さらに、他の形態に調製する場合であっても、従来の方法に従えばよい。
本発明の治療方法または予防方法は上記記載に基づいて実施されうる。式(I)または式(IX)の化合物または医薬として許容されるその塩が投与される対象としては、哺乳動物、例えばヒトが例示される。
(製造法)
以下に、本発明の化合物の製造法について、例を挙げて説明するが、本発明はもとよりこれに限定されるものではない。

下記製造法における中間体および目的化合物は、その官能基を適宜変換することによって、本発明に含まれる別の化合物へ変換することもできる。保護基としては、文献(T.W.Greene and P.G.M.Wuts,”Protective Groups in Organic Synthesis“,3rd Ed.,John Wiley and Sons,inc.,New York(1999))等に記載されている通常の保護基を用いることができ、保護基の導入および除去は有機合成化学で常用される方法(例えば、上記文献に記載の方法等)またはそれに準じた方法により行うことができる。具体的にはアミノの保護基としては、例えば、ベンジルオキシカルボニル、tert-ブトキシカルボニル、アセチル、ベンジル等を、またヒドロキシの保護基としては、例えば、トリアルキルシリル、アセチル、ベンジル等をそれぞれ挙げることができる。
下記各製造法における出発原料および中間体は、市販品を購入、公知文献より合成、あるいは公知化合物から公知の方法により合成することにより入手できる。また、出発原料および中間体は、必要に応じてそれらの塩を用いてもよい。
下記製造法における不活性溶媒とは、反応で用いる原料、試薬、塩基、酸、触媒、配位子等と反応しない溶媒を意味する。
また、式(I)または式(IX)の化合物のプロドラッグは、上記保護基と同様、原料から中間体へ至る段階で特定の基を導入、あるいは得られた式(I)の化合物を用いてさらに反応を行なうことで製造できる。反応は通常のエステル化、アミド化、脱水等、当業者に公知の方法を適用することにより行うことができる。
また、式(I)または式(IX)の化合物のプロドラッグは、上記保護基と同様、原料から中間体へ至る段階で特定の基を導入、あるいは得られた式(I)の化合物を用いてさらに反応を行なうことで製造できる。反応は通常のエステル化、アミド化、脱水等、当業者に公知の方法を適用することにより行うことができる。

(第一工程)
本工程は、式(II)の化合物と式(III)の化合物との環化反応により式(IV)の化合物を得る工程である。この反応では、式(II)の化合物と式(III)の化合物を当量あるいはいずれかを過剰量用い、これらの混合物を、反応に不活性な溶媒中又は無溶媒下、冷却下から加熱還流下、例えば室温~120℃において、通常1時間~3日間攪拌する。ここで用いられる溶媒の例としては、特に限定はされないが、ジエチルエーテル、THF、1,4-ジオキサン、1,2-ジメトキシエタン等のエーテル類、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール等のアルコール類、水、酢酸、ピリジン、アセトニトリル、NMP、DMF、DMSO及びこれらの混合物が挙げられる。トリエチルアミン、N,N-ジイソプロピルエチルアミン等の有機塩基、又は炭酸カリウム、炭酸ナトリウム、炭酸セシウム等の無機塩基の存在下で反応を行うのが、反応を円滑に進行させる上で有利な場合がある。

[第1工程](E)-1-(5-フルオロ-2-ヒドロキシフェニル)-3-(2-(トリフルオロメチル)フェニル)プロパ-2-エン-1-オンの調製
[第1工程](E)-1-(5-フルオロ-2-ヒドロキシフェニル)-3-(2-(トリフルオロメチル)フェニル)プロパ-2-エン-1-オンの調製
エタノール(700mL)に水酸化カリウム(37g)を加え溶解させた。該溶液に2-アセチル-4-フルオロフェノール(20g)を室温で加えた後、2-(トリフルオロメチル)ベンズアルデヒド(25g)をさらに加えた。得られた混合物を室温にて3時間撹拌後、氷冷し、1M塩酸水溶液(440mL)を滴下した。析出した固体をろ過により回収し、水洗した。得られた固体を40℃で減圧乾燥し、表題の橙色の化合物を(33g、収率81%)で得た。

(E)-1-(5-フルオロ-2-ヒドロキシフェニル)-3-(2-(トリフルオロメチル)フェニル)プロパ-2-エン-1-オン(23g)のDMF(120mL)の溶液に炭酸カリウム(13g)を加え、室温にて撹拌した。得られた混合物に4-ブロモブタン酸エチル(12mL)を加え、室温で3時間撹拌した。反応混合物に水(230mL)を加え、酢酸エチルで抽出した。さらに水層を2回酢酸エチルで抽出し、有機層をあわせて、水洗した。有機層を脱水、減圧濃縮して、表題の化合物(33g)を得た。これを精製せずに次工程に使用した。

(E)-4-(4-フルオロ-(2-(3-(2-トリフルオロメチル)フェニル)プロパ-2-エノイル)フェノキシ)ブタン酸エチル(33g)のエタノール(310mL)の溶液にフェニルヒドラジン(11mL)を加え、2時間加熱還流下撹拌した。反応混合物を氷冷し、析出した固体をろ過により回収した。冷却したエタノールで得られた固体を洗浄し、表題の黄白色の化合物(18g、収率47%)を得た。

4-(4-フルオロ-2-(1-フェニル-5-(2-(トリフルオロメチル)フェニル)-1H-ピラゾール-3-イル)フェノキシ)ブタン酸エチル(12g)にエタノール(60mL)、1M水酸化ナトリウム水溶液(60mL)を加え、室温で終夜撹拌した。1M塩酸水溶液(5mL)を滴下して、析出した固体をろ過にて回収した。冷却したエタノールで洗浄、乾燥した。表題の白色化合物(10g、収率91%)を得た。

実施例2:4-(2-(1-フェニル-5-(2-(トリフルオロメチル)フェニル)-1H-ピラゾール-3-イル)フェノキシ)ブタン酸
[第1工程](E)-1-(2-ヒドロキシフェニル)-3-(2-(トリフルオロメチル)フェニル)プロパ-2-エン-1-オンの調製
エタノール(70mL)に水酸化カリウム(4.12g)を加え溶解させた。該溶液に2-ヒドロキシアセトフェノン(2g)を室温で加えた後、さらに2-(トリフルオロメチル)ベンズアルデヒド(2.81g)を加えた。得られた混合物を室温で17時間撹拌後、氷冷し、2M塩酸水溶液を滴下した。析出した固体をろ過にて回収し、水洗後、40℃で減圧乾燥し、橙色の(E)-1-(2-ヒドロキシフェニル)-3-(2-(トリフルオロメチル)フェニル)プロパ-2-エン-1-オン(3.2g(収率75%)で得た。

(E)-1-(2-ヒドロキシフェニル)-3-(2-(トリフルオロメチル)フェニル)プロパ-2-エン-1-オン(0.5g)のDMF(2.5mL)の溶液に炭酸カリウム(0.3g)を加え、室温で撹拌した。得られた混合物に4-ブロモブタン酸エチル(0.4mL)を加え、室温で17時間撹拌した。水(5mL)を加え、酢酸エチルで抽出した。さらに水層を2回酢酸エチルで抽出し、有機層をあわせて、水洗した。有機層を脱水、減圧濃縮して、表題の化合物として白色固体(0.7g)を得た。これを精製せずに次工程に使用した。

4-(2-(1-フェニル-5-(2-(トリフルオロメチル)フェニル)-1H-ピラゾール-3-イル)フェノキシ)ブタン酸エチル(2.9g)にエタノール(60mL)、および1M水酸化ナトリウム水溶液(60mL)を加え、得られた混合物を室温で終夜撹拌した。反応混合物に1M塩酸水溶液5mLを滴下して、析出した固体をろ過煮て回収し、冷却したエタノールで洗浄、乾燥した。白色の固体として表題の化合物(2.5g、収率90%)を得た。

実施例3:4-(2-(1-フェニル-5-(2-(トリフルオロメチル)フェニル)-1H-1,2,4-トリアゾール-3-イル)フェノキシ)ブタン酸
[第1工程]N-フェニル-2-(トリフルオロメチル)ベンゼンカルボキシイミダミドの調製
[第1工程]N-フェニル-2-(トリフルオロメチル)ベンゼンカルボキシイミダミドの調製
アニリン(2.7g)のDMSO(25mL)の溶液に55%水素化ナトリウム(油性、1.5g)を加え、室温で5分撹拌した。得られた混合物に2-トリフルオロメチルシアノベンゼン(10g)を加え、終夜撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を水洗後、減圧濃縮した。濃縮物をシリカゲルカラムクロマトグラフィ(クロロホルム/メタノール=100:1)で精製し、表題の化合物(3.5g、収率45%)を得た。

トルエン(100mL)中の2-シアノフェノール(4.3g)、N-フェニル-2-(トリフルオロメチル)ベンゼンカルボキシイミダミド(2.4g)、1,10-フェナントロリン(41mg)、酢酸銅(41mg)、および炭酸ナトリウム(3.9g)の混合物を17時間加熱還流下撹拌した。放冷後、反応混合物に水を加え、酢酸エチルで2回抽出した。有機層を合わせて水洗し、溶媒を減圧留去し、黄色の油状物を得た。カラムクロマトブラフィ(クロロホルム/メタノール=100:1)により精製し、2-(1-フェニル-5-(2-(トリフルオロメチル)フェニル)-1H-1,2,4-トリアゾール-3-イル)フェノール(120mg)を得た。

DMF(5mL)中の2-(1-フェニル-5-(2-(トリフルオロメチル)フェニル)-1H-1,2,4-トリアゾール-3-イル)フェノール(100mg)、炭酸カリウム(120mg)、および4-ブロモブタン酸エチル(200μL)の混合物を90℃で2時間撹拌した。反応混合物を酢酸エチルで2回抽出し、有機層をあわせて水洗、脱水後、溶媒を留去した。
得られた生成物にエタノール(5mL)、2M水酸化ナトリウム水溶液(5mL)を加え、室温で24時間撹拌した。反応混合物を2M塩酸水溶液で中和し、酢酸エチルで2回抽出し、有機層をあわせて水洗、脱水後、溶媒を留去した。濃縮物をエタノールに溶解させ、活性炭で脱色した。水を滴下して析出した固体をろ過にて回収し、洗浄、乾燥して、表題の化合物(40mg)を得た。

試験例1:ANSを用いたFABPリガンドの親和性評価試験
大腸菌[BL21(DE3)株]にGST-FABP3、GST-FABP4、GST-FABP5およびGST-FABP7をそれぞれ発現させたのち、グルタチオンカラムを用いてアフィニティー精製を行った。
大腸菌[BL21(DE3)株]にGST-FABP3、GST-FABP4、GST-FABP5およびGST-FABP7をそれぞれ発現させたのち、グルタチオンカラムを用いてアフィニティー精製を行った。
GST-FABP3、GST-FABP4、GST-FABP5およびGST-FABP7ベクターのクローニングは以下の方法で行った。マウス心臓から単離したmRNAをcDNAに逆転写したのち、PCR法によりFABP3遺伝子(RefSeqID:NM_010174.1)およびFABP4遺伝子(RefSeqID:NM_024406.2)をそれぞれ増幅させた。これらのcDNAをそれぞれDNA ligation kit(タカラバイオ、草津)およびIn-fusion kit(タカラバイオUSA、CA,USA)を用いて、pGEX-2Tベクター(GEヘルスケアジャパン、東京)のBamHI/EcoRI切断部位の間に挿入することで、ベクターを作製した。
図1にシーケンシングを行ったGST-FABP3、GST-FABP4、GST-FABP5、およびGST-FABP7の塩基配列を記載する(最初と最後のGGATTCおよびGAATTC が制限酵素切断部位である)。
GST結合タンパク質の精製は、GST purification kit(タカラバイオUSA)を用いて行った。大腸菌を遠心により回収したのち、付属のExtractionバッファーと酸化アルミニウム粉末を加え、乳鉢で破砕した。遠心した上清を付属のグルタチオンカラムに加え、氷上で30分間静置した。その後、カラム内の上清を捨て、Extractionバッファーで洗浄を行ったのち、グルタチオンを含む溶出バッファーで溶出した。溶出産物のタンパク質濃度を280nmの吸光度から算出したのち、ANSアッセイに用いた。
FABPタンパク質と結合し蛍光を発する1-アニリノナフタレン-8-スルホン酸(ANS,最終濃度4mM、10mM KH2PO4、40mM KCl、pH7.4の溶液中)とFABPタンパク質(最終濃度0.4mM)、さらに各種リガンドを0nM、100nM、1000nM、2000nM、4000nM(最終)の濃度で2分間インキュベートしたのち、ANSの蛍光を測定した(Ex/Em=355nm/460nm)。各リガンド濃度における蛍光強度をリガンド非存在下におけるANS蛍光強度に対する相対値(%)に変換したのち、以下の等式を用いて非回帰分析により解離定数Kd(nM)を算出した。

実施例1~5の化合物(それぞれ実施例化合物1~実施例化合物5という)、公知の化合物であるLIGAND 1(WO2017/171053号公報記載)をリガンドとして使用した。
測定結果に基づいて算出した解離定数Kdを表1に示す。特にFABP3に対し、実施例化合物1~5が、LIGAND 1よりも小さな解離定数Kdをもつことが確認された。
上記表のKd値は3回の測定の平均±SEとして表示されている。Kd値が1000000以上の場合は「結合せず」と表示した。なお、表中の実施例化合物1から5は、それぞれ化合物1から5ということもある。
試験例2:PDモデル動物を用いた認知機能評価試験
ドーパミン神経毒である1-メチル-4-フェニル-1,2,3,6-テトラヒドロピリジン(MPTP、25mg/kg,i.p.、Sigma-Aldrich社(St Louis, MO)より購入)を8-10週齢の雄性 C57BL6 N マウスに一日一回五日間連続投与し(Day1~5)、試験開始20日(Day20)目のマウスがPD様を呈する。当該モデル動物として試験開始から20日後(Day20)から溶媒またはFABP3リガンド(実施例1の化合物(実施例化合物1)、0.05、0.1および0.3mg/kg,p.o.、各群n=7)を一日一回二週間投与した。試験開始から34日後(Day34)に受動回避試験により認知機能を評価した。受動回避試験(Passive Avoidance task)は、訓練試行では明室にマウスを入れ、暗室にマウスが入った際に電気刺激(0.3mA,2秒)を与えた。24時間後にもう一度マウスを明室にいれ、暗室に入るまでの時間(図2の縦軸Latency(s)に対応)を測定した。結果を図2に示す。リガンドを投与したマウス群では濃度依存的に待機時間の延長が確認された。さらに、当該vehicle(veh)のLatencyは、当該salineと比較し、有意差があることが確認されているため、本試験例の方法は妥当であるといえる。

なお、上記のモデルマウス作成において、MPTPの代わりに食塩水を投与した正常マウス群を図においてsalineと表示し、FABP3リガンドの代わりに0.5%カルボキシメチルセルロースを投与した群をvehicle(veh)と表示する。
試験例3:PDモデル動物を用いた認知機能評価試験
FABP3のリガンドとして実施例2の化合物(実施例化合物2;0.03、0.1および0.3mg/kg)を投与した以外は試験例2と同じ処置をしたモデルマウスにおいて試験開始から34日後(Day34)に新規物体認識試験(Novel object recognition task)により認知機能を評価した。新規物体認識試験は、訓練試行では同じ形の物体を置きマウスに記憶させ、試験試行では片方の物体を新規物体に代え、両物体に対する接触割合(図3の縦軸Discrimination index(%)に対応)を評価した。結果を図3に示す。新規物体認識試験では、リガンド投与群において新規物体と既知物体の間で接触割合に大きな差があり、認知機能の改善傾向が確認された。

試験例5:PDモデル動物を用いたドーパミン神経保護評価試験
FABP3のリガンドとして実施例化合物2(0.1、0.3および1.0mg/kg)またはLIGAND 1(0.3mg/kg)を投与した以外は試験例2と同じ処置をしたモデルマウスにおいて試験開始から34日後(Day34)にマウスを灌流固定し、50μmの厚さで黒質領域(SNpc)を含む脳切片、および腹側被蓋野(VTA)を含む脳切片を作製した。
TH抗体(Immunostar社製、mouse monoclonal antibody 22941、1:1000)、α-シヌクレイン抗体(Santa Cruz社製、rabbit polyclonal anitibody SC-70110R、1:200)、及びFBAP3抗体(確認中)と反応させ、蛍光標識された二次抗体(Alexa 594 anti-mouse IgG (Jackson ImmunoResearch社製、1:500))で検出した。黒質領域の結果を図6、図7、図10、図11、図14および図15に、腹側被蓋野の結果を図8、図9、図12、図13、図16および図17にそれぞれ示す(図6及び図8の縦軸Number of TH+ cellsは、ドーパミン作動性神経細胞の数に対応。図11および図13の縦軸Number of double-positive cells/TH+ positive cells(%)は、ドーパミン作動性神経細胞のうちリン酸化α―シヌクレインを含むものの割合に対応。図15および図17の縦軸Triple-positive cells/TH-positive cellsは、ドーパミン作動性神経細胞のうち、FABP3とリン酸化α-シヌクレインをともに含むものの割合に対応)。
FABP3のリガンドとして実施例化合物2(0.1、0.3および1.0mg/kg)またはLIGAND 1(0.3mg/kg)を投与した以外は試験例2と同じ処置をしたモデルマウスにおいて試験開始から34日後(Day34)にマウスを灌流固定し、50μmの厚さで黒質領域(SNpc)を含む脳切片、および腹側被蓋野(VTA)を含む脳切片を作製した。

図6~9が示す結果から、実施例化合物2がMPTP投与マウスにおけるドーパミン作動性神経細胞の減少を抑制することが確認された。一方、LIGAND 1投与群においては改善がみられなかった。また、図10~13が示す結果から、MPTP投与マウスのドーパミン作動性神経細胞において増加したα-シヌクレインのリン酸化レベルが、実施例化合物2の投与によって減少することが確認された。さらに、図14~17が示す結果から、マウスへのMPTP投与により生じた、ドーパミン作動性神経細胞におけるFABP3とリン酸化α-シヌクレインの局在化が、実施例化合物2の投与により減少することが確認された。α-シヌクレインのリン酸化によりα-シヌクレインの凝集が増加すること(Samuel, F. et al., J. Biol. Chem. 291 (2016) 4374-4385)、及び、FABP3とα-シヌクレインが共存するとα-シヌクレインが凝集すること(Fukui, N. et al., J. Biol. Chem. 296 (2021) 100663)を踏まえると、これらの結果は実施例化合物2にα-シヌクレインの凝集を改善する効果があることを示しており、パーキンソン病改善に有用である(図25)。なお、α―シヌクレイン凝集体が伝播することを示す報告(Lee, V. M. Y. et al., J. Exp. Med. Vol. 209 No. 5 975-986)を踏まえると、実施例化合物2はα-シヌクレインの凝集を改善することにより、α―シヌクレイン凝集体の伝播を抑制する効果をもつ可能性が有る。さらに、リン酸化されたα-シヌクレインが野生型α-シヌクレインよりも伝播しやすいという報告(Li, Y-M. et al., Sci Rep. 2016; 6, 37130)を踏まえると、実施例化合物2はα-シヌクレインのリン酸化レベルを減少させることにより、α-シヌクレインの伝播を抑制する効果をもつ可能性が有る。以上より、実施例化合物2はMPTPによって誘導されるリン酸化α-シヌクレインからドーパミン作動性神経細胞を保護したといえる。さらに、ドーパミン作動性ニューロンにおけるリン酸化α-シヌクレインとFABP3の相互作用は実施例化合物2によって阻害されたといえる。
試験例6:局所脳虚血モデルを用いた脳神経保護評価試験
雄のICRマウス(5週齢、25-30g)を日本エスエルシー株式会社(日本、静岡県)より購入した。動物は恒温恒湿の条件下で飼育し、12時間の明暗サイクル(点灯:09:00-21:00)で飼育し、不断給餌とした。すべてのマウスは0.3mg/kgメデトミジン、4.0mg/kgミダゾラム、および5.0mg/kgブトルファノールの組み合わせで麻酔された。右外頸動脈から内頸動脈にシリコンコーティングした6-0縫合糸(Doccol Corporation, USA)を中大脳動脈起始部まで挿入し,2時間放置した後,縫合糸を除去して再灌流を行い局所脳虚血モデル(tMCAO)のモデルマウスを作成した。偽手術群のマウスは,縫合糸を挿入する以外は同様の処置を受けた.再灌流後、マウスは指定時間に屠殺した。I/R操作中は,恒温式毛布を用いてマウスの深部体温を37℃に維持した.局所脳血流(rCBF)をレーザードップラー流速計(FLOC1,OMEGAWAVE,Tokyo,Japan)によりモニターし,右半球が虚血状態であるか否かを確認した.手術は,前述のようにCBFが約70-90%減少した時点で成功とした.24時間または7日間の再灌流後、神経学的障害の等級付システムを用いて神経学的スコアを算出した後、マウスを断頭し、脳を迅速に取り出し、-30℃に10分間冷却した。脳を2mm厚の5枚に切り出し,1% 2,3,5-トリフェニルテトラゾリウムクロライド(TTC,Wako,Japan)中,37℃で20分間インキュベートし,4%パラホルムアルデヒド(PFA;Sigma,USA)中に一晩浸漬させた.非梗塞部は灰色を示し,梗塞部は白色を示した.梗塞体積はImageJソフトウェアを用いて測定し、全半球に対する割合で表した(図19のB、図21のE、図23のHに対応。なお、図19のC、図21のF、図23のIの縦軸Neurological scoreは神経学的欠損を表す。図24の縦軸Survival rate(%)はマウス生存率に対応)。

実施例1の化合物は0.5%カルボキシメチルセスロース(CMC)に懸濁し、コントロール群には相当量の0.5%CMCを経口投与した。
マウスにtMCAOを2時間行い、再灌流30分後に実施例化合物1を異なる濃度(0.5,0.1,0.3,1.0mg/kg)で投与した。結果を図18Aおよび図19BおよびCに示す。図18Aおよび図19BおよびCは、再灌流後24時間におけるTTC染色(A),梗塞体積の定量分析(B),神経障害(C)の代表画像である(n=7-9)。再灌流後0.5,1,2時間で実施例化合物1(0.3mg/kg)を投与した。結果を図20Dおよび図21EおよびFに示す。図20Dおよび図21EおよびFは、再灌流24時間後のTTC染色(D),梗塞体積の定量分析(E),神経障害(F)の代表画像である(n=7-8)。再灌流30分後に実施例1化合物を異なる用量(0.5,0.1,0.3mg/kg)で投与した。図22Gおよび図23HおよびIに示す。図22Gおよび図23HおよびIは再灌流後7日目におけるTTC染色(G),梗塞体積の定量分析(H),神経障害(I)の代表画像である。各群の1日あたりのマウス生存数を記録し、生存率(図24J)を算出した。(n=9-12)。特に実施例化合物1を0.3mg/kg投与したマウスにおいて、脳梗塞体積がVehicleマウスよりも減少し、マウス生存率がVehicleマウスよりも向上した。

- シヌクレイノパチーの治療または予防において使用するための、請求項10に記載の医薬組成物。
- シヌクレイノパチーが、パーキンソン病、レビー小体型認知症、または多系統萎縮症である、請求項11に記載の医薬組成物。
- ドーパミン神経機能障害に起因する疾患の治療または予防において使用するための、請求項10に記載の医薬組成物。
- 脳梗塞、脳出血、くも膜下出血などの脳血管障害、頭部外傷の治療および予防において使用するための、請求項10に記載の医薬組成物。
- 自閉症や統合失調症などの精神疾患の治療または予防において使用するための、請求項10に記載の医薬組成物
- 経口投与用である、請求項10~15のいずれか1項に記載の医薬組成物。
