The chewing gum consisting of natural Nano Fenchol and Nano Quercetin which is anti-Alzheimer and also anticancer in the fields of food and Medical industry. The main and common component in all diseases such as different types of Cancers and alzheimers is the existent of oxidative stress and oxygen free Radicals production. Quercetin and Fenchol not only restrain producing ROS inside the body cells but also they prevent forming Amyloid plaques in brain as well as stimulating pathways of apoptosis inside the cancer cells besides having barrier properties for cardiovascular system. These substances are primarily produced in size of Nanoparticles then are used in Natural gums chewing gum produced from various natural sources. Drug delivery is gradually done having high permeation because of Natural gums and Nano structure. Producing Nanostructured drugs can increase its penetrance as well as targeted transmission. hence, producing every kind of food supplement, types of filling materials and flavored chewing gum including fenchol and quercetin can be effective for various diseases such as cancers of tongue, esophageal, stomach as well as Alzheimer disease.
ROS (reactive oxygen species) is a common factor in all diseases, especially cancer and neurodegenerative diseases. On the other hand, due to the significant increase in smoking and non-observance of oral and dental hygiene, various types of diseases, especially tongue and esophagus cancer, have also increased. Delivering the drug to the epithelial cells of the mouth and esophagus is a fundamental problem in the treatment of this disease due to its ephemerality on the one hand and the difficulty of swallowing in various people with complications in the mouth and esophagus on the other hand. In recent years, many advances have been made in the field of drug delivery methods. Among these methods that can release the drug in a targeted manner, chewing gum can be mentioned. In this invention, chewing gum containing antioxidant and anticancer Nano fenchol and natural Nano quercetin due to its convenience and availability and on the other hand its popularity among people, as one of the suitable methods to transfer all kinds of drugs, not only in It can be used for patients with drug swallowing problems, but also for different groups of patients, in this case it can be used to solve the aforementioned problem, which is easily absorbed in epithelial cells due to its Nanostructure.
ROS are signaling molecules used in biological processes. A shift in the balance of the extent of oxygen free radicals created and the antioxidant defense, on the other hand, may cause damage to internal cellular organelles and normal physiological functions. Moreover, the biological cause of a range of diseases such as cancer, asthma, and pulmonary hypertension is a disruption in the equilibrium of oxygen free radicals and antioxidants. As a result, a comprehensive knowledge of this topic may lead to the discovery of the cause and treatment of numerous illnesses. Fenchol and Quercetin-containing gum has anti-cancer and anti-ROS properties. Because of its Nano structure, it may have a direct and entirely targeted impact on all forms of oral malignancies (Liczbiński and et al 2020, University of South Florida (USF Health), October 5, 2021).
Fenchol works in the gut on short-chain fatty acids (SCFAs). SCFAs then travel through the bloodstream to the brain, where they bind to and activate FFAR2 (free fatty acid receptor 2) receptors. When activated, this receptor protects the nervous system and prevents the formation of amyloid (Aβ) accumulations and plaques (University of South Florida (USF Health), October 5, 2021).
Extensive research has shown that Quercetin can prevent cancers such as breast, lung, nasopharyngeal, kidney, colorectal, prostate, esophageal, pancreatic, and ovarian cancer. The presence of two antioxidant pharmacophores in the structure of Quercetin accounts for its high ability to inhibit free radicals and bind to transition metal ions. Furthermore, the presence of catechol and the OH group at the C3 position of its structure provides an excellent configuration for free radical elimination. Quercetin is a pentahydroxy flavanol with 5 hydroxyl groups on carbons 3, 30, 40, 5, and 7 in its flavanol skeleton. The substitution of Quercetin's different functional groups results in its various biochemical and medicinal functions. Quercetin is a lipophilic compound that can cross cell membranes and activate a variety of intracellular signaling pathways involved in chemoprevention. Several studies have found Quercetin to have anticancer properties in a variety of cancers. One of Quercetin's unique properties is its ability to act as both a pro-oxidant and an antioxidant. Increased ROS due to oxidative stress causes DNA damage, which results in mutations. The mutation causes malignant tumor cells to grow uncontrollably. Quercetin can reduce ROS by donating electrons, resulting in less DNA damage caused by ROS. At high concentrations, Quercetin is also known to induce apoptosis. The main mechanism by which Quercetin induces apoptosis has been identified as a mitochondria-mediated pathway. Quercetin causes apoptosis by activating p53, increasing pro-apoptotic molecules like Bax and caspase-3, and decreasing anti-apoptotic factors like survivin and Bcl-2. Also, Quercetin has no toxic effect on healthy cells (Rana Shafabakhsh & Zatollah Asemi. 2019)
The steps are as follows:
- In the first phase, basil leaf or cactus fruit extracts are extracted using various solvents (any origin or plant that contains Fenchol and Quercetin compounds, or Quercetin and Fenchol itself can also be used).
This is accomplished with the use of a Soxhlet apparatus and various solvents. To separate the heavy oily hydrocarbons and waxes, ethanol is added to it and stirred for one hour before being maintained at -15 degrees for evaporation and filtration.
2. Fenchol and Quercetin are separated using Hplc and fplc according to the protocol.
For this aim, in order to separate better from the process of soaking in solvent and extracting with hexane, column chromatography was done on silica gel 60 in a column with a length of 50 cm and a diameter of 2 cm, and lastly, the HPLC spectrum was created.
Knuer's Well Chrom 2000 HPLC apparatus with the Stark 1000 Maxi pump and the Eurospher 100 C18 detector with a diameter of 4 mm and a length of 25 cm was utilized for this.
Methanol/water/acetic acid in 5/45/50 proportions were utilized as the mobile phase. Each analysis took 60 minutes to complete.
The oven temperature was 30 degrees Celsius, and the solvent flow rate in the column was 1 ml/min. In addition, the amount of sample injected was 20 microliters.
3. After its approval, fenchol and quercetin are added to it along with other permitted compounds.
Nano Fenchol and Nano Quercetin are two of the components employed in this product (total 0.5 to 1 percent).
Starch, sorbitol, lecithin, peppermint essence, allowed and natural food additives, Beneh tree Saghez (with standard percentage).
4. Fenchol and Quercetin are synthesized in Nano size form by ultrasonication and the resultant extract is utilized to create chewing gum. The second aspect of slow release is the tiny size, which is contained in the gum polymer and arranged in countless numbers. The ingredients are blended in a predetermined size. The Fenchol gum is then packaged in sterile packaging.
5. We bring the container to a temperature of 30-38 degrees Celsius. The manufacturing process starts by melting the gum base texture for 10 minutes, then the relevant sugars are added to it and mixed together for 10 minutes. Then add the rest of the ingredients including fenchol and quercetin, and mix completely for 5 to 10 minutes. Rhys buffering method will be used to transfer fenchol inside the chewing gum polymer. Finally, after forming a homogeneous texture, it is transferred to the machine for producing 5 gram molds.
Mixture test:
This research was conducted on Alzheimer's model mice.
- Alzheimer's model mice were created using manganese Nano powder (60 mg/mouse for two weeks as an intraperitoneal injection) and were validated using molecular tests and histopathology.
- Alzheimer's rats model were split into three groups: gavage treatment, oral treatment, and intraperitoneal injection treatment.
- To apply the treatment in the form of intraperitoneal injection in doses of 0.5, 1 and 2 cc (3 mg per cc) for one week.
- In accordance with medical ethics protocols, the samples were anesthetized with ether and the spinal cord was cut. Then, the rats' brains were surgically removed and samples were taken from the hippocampus.
- Molecular and histopathology tests were performed on hippocampus samples.
Because pharmaceutical forms and other combinations of substances containing Fenchol and Quercetin are not easily available and usable, chewing gum to administer Fenchol can easily solve this problem. Nanostructures, on the other hand, have the properties of gradual release as well as high penetration. Saghez (Natural gum) chewing gum derived from the Bene plant is combined with Nano Fenchol and Nano Quercetin in this invention. In addition to Nano Fenchol and Quercetin, various permitted herbal flavorings, such as mint or other flavorings, are added to this invention for flavoring.
The gum substance itself has the property of gradual drug release (Ughade and et al 2012), whereas the presence of Nanostructures is the second factor of gradual drug release and also has the ability to penetrate deeper into cells.
Gradual release and treatment of cells that are at risk of increased ROS levels.
Due to the ease of use, no restriction of use for different ages and accessibility.
Direct effect using drug delivery technique for targeted drug delivery.
Use of natural sugars such as sorbitol due to the absence of tooth decay.
Transfer correctly and in the right amount of fennel and quercetin.
No toxicity in high doses of fencol and quercetin.
Benefiting from the beneficial effects of fennel and quercetin for the body.
Compliance with the standards of the amount of effective substance in each gum.
Synergistic effect of quercetin and fencol together without creating negative interaction.
Direct effect on mouth, esophagus and stomach cancers.
Examples
In the first step, basil leaf and cactus fruit extracts are extracted using various solvents. The Fenchol and Quercetin are then separated using Hplc and fplc according to the protocol. After it has been approved, Fenchol and Quercetin are added, along with other permitted ingredients. Ultrasonication is used to prepare Fenchol and Quercetin in Nano size form, and the resulting extract is used to make chewing gum. The second factor of gradual release is the small size, which is encapsulated in the gum polymer and placed in countless numbers. The materials are combined in a predetermined size. Then the Fenchol gum is packed in sterile packages. We heat the container to between 30-38 degrees Celsius. The manufacturing process begins with 10 minutes of melting the gum base texture, followed by 10 minutes of mixing in the relevant sugars. Then add the remaining compounds, including Fenchol, and thoroughly mix for 5-10 minutes. The Fenchol within the gum polymer will be used via the weaving thread method. Finally, it is transferred to the machine for producing 5-gram molds after forming a homogeneous texture.

Gelis | The anti-cancer, cancer prevention and antimetastatic potential of curcumin has been established through multiple animal studies and cancer cells cultures. Curcumin is a multipotent natural compound that induces apoptosis, inhibits survival signals, scavenges reactive oxidative species (ROS), and reduces the inflammatory cancer microenvironment depending on the study context. Curcumin induces apoptosis in upper aerodigestive tract cancer cell lines (including the lung and bronchus, larynx, pharynx and oral cavity) which is mediated by the modulation of multiple pathways such as induction of p73, and inhibition of p-AKT and Bcl-2 | |
