항암화학요법제는 암세포 제거, 진행억제 및 종양크기 감소 등을 기대할 수 있으나, 대부분의 항암제는 빠르게 성장·분열하는 암세포에 작용하여 혈액세포 감소, 탈모 외 다양한 장기에 부작용을 초래할 수 있다. 이러한 부작용은 약물치료를 중단하면 사라지는 증상들로 이와 같은 부작용을 최소화하고 효능이 뛰어난 새로운 화학요법제가 다양한 분자를 대상으로 연구개발 중에 있다.
본 발명은 신규한 벤즈옥사졸아민 유도체 화합물 및 이의 제조방법을 제공하는 것을 목적으로 한다.
또한, 본 발명은 우수한 항암 용도를 가지는 벤즈옥사졸아민 유도체 화합물을 제공하는 것을 목적으로 한다.

본 발명에서 "알키닐(alkynyl)"은 탄소-탄소 삼중 결합을 1개 이상 가진 탄소수 2 내지 100의 직쇄 또는 측쇄의 불포화 탄화수소에서 유래되는 1가의 치환기를 의미한다. 이의 예로는 에티닐(ethynyl), 2-프로파닐(2-propynyl) 등을 들 수 있으나, 이에 한정되지는 않는다.
본 발명에서 "시클로알킬"은 탄소수 3 내지 100의 모노사이클릭 또는 폴리사이클릭 비-방향족 탄화수소로부터 유래된 1가의 치환기를 의미한다. 이러한 사이클로알킬의 예로는 사이클로프로필, 사이클로펜틸, 사이클로헥실, 노르보닐(norbornyl), 아다만틴(adamantine) 등을 들 수 있으나, 이에 한정되지는 않는다.
본 발명에서 "아릴"은 단독 고리 또는 2이상의 고리가 조합된 탄소수 6 내지 100의 방향족 탄화수소로부터 유래된 1가의 치환기를 의미한다. 또한, 2 이상의 고리가 서로 단순 부착(pendant)되거나 축합된 형태도 포함될 수 있다. 이러한 아릴의 예로는 페닐, 나프틸, 페난트릴, 안트릴 등을 들 수 있으나, 이에 한정되지는 않는다.
본 발명에서 "헤테로시클로알킬"은 핵원자수 3 내지 100의 비-방향족 탄화수소로부터 유래된 1가의 치환기를 의미하며, 고리 중 하나 이상의 탄소, 바람직하게는 1 내지 3개의 탄소가 N, O, S 또는 Se와 같은 헤테로 원자로 치환된다. 이러한 헤테로시클로알킬의 예로는 모르폴린, 피페라진 등을 들 수 있으나, 이에 한정되지는 않는다.
본 발명에서 "헤테로아릴"은 핵원자수 5 내지 100의 모노헤테로사이클릭 또는 폴리헤테로사이클릭 방향족 탄화수소로부터 유래된 1가의 치환기를 의미한다. 이때, 고리 중 하나 이상의 탄소, 바람직하게는 1 내지 3개의 탄소가 N, O, S 또는 Se와 같은 헤테로원자로 치환된다. 또한, 2 이상의 고리가 서로 단순 부착(pendant)되거나 축합된 형태도 포함될 수 있고, 나아가 아릴기와의 축합된 형태도 포함될 수 있다. 이러한 헤테로아릴의 예로는 피리딜, 피라지닐, 피리미디닐, 피리다지닐, 트리아지닐과 같은 6-원 모노사이클릭 고리, 페녹사티에닐(phenoxathienyl), 인돌리지닐(indolizinyl), 인돌릴(indolyl), 퓨리닐(purinyl), 퀴놀릴(quinolyl), 벤조티아졸(benzothiazole), 카바졸릴(carbazolyl)과 같은 폴리사이클릭 고리 및 2-퓨라닐, N-이미다졸릴, 2-이속사졸릴, 2-피리디닐, 2-피리미디닐 등을 들 수 있으나, 이에 한정되지는 않는다.

본 발명에서 약학적으로 허용되는 염은 의약업계에서 통상적으로 사용되는 염을 의미하며, 예를 들어 염산, 질산, 인산, 브롬산, 요오드산, 과염소산, 주석산 및 황산 등으로 제조된 무기산염, 메탄설폰산, 에탄설폰산, 벤젠설폰산, p-톨루엔술폰산, 나프탈렌설폰산, 아세트산, 트리플루오로아세트산, 시트르산, 말레인산, 숙신산, 옥살산, 벤조산, 타르타르산, 푸마르산, 만데르산, 프로피온산, 구연산, 젖산, 글리콜산, 글루콘산, 갈락투론산, 글루탐산, 글루타르산, 글루쿠론산, 아스파르트산, 아스코르브산, 카본산, 바닐릭산, 하이드로 아이오딕산 등으로 제조된 유기산염, 글리신, 아르기닌, 라이신 등으로 제조된 아미노산염 등이 있으나, 이에 한정되는 것은 아니다. 이러한 약학적으로 허용되는 염은 통상의 방법, 예를들어 본 발명의 벤즈옥사졸을 과량의 상기 약제학적으로 허용되는 산이 녹아있는 수용액에 용해시키고, 이를 메탄올, 에탄올, 아세톤 또는 아세토니트릴과 같은 수 혼화성 유기용매를 사용하여 침전시켜 제조할 수 있다.
다음으로, 전술한 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 유효성분으로 포함하는 약학 조성물의 항암 용도에 관하여 구체적으로 설명한다.
본 발명의 실시예에 의한 암의 예방 또는 치료용 약학 조성물은 전술한 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 유효성분으로 포함한다.
본 발명의 실시예에 의한 암의 예방 또는 치료용 약학 조성물은 항암제에 대한 내성을 억제시킬 수 있다.
또한, 본 발명의 실시예에 의한 암의 예방 또는 치료용 약학 조성물은 항암제의 민감성을 증진시킬 수 있다.
일 예로서, 본 발명의 암의 예방 또는 치료용 약학 조성물은 암세포 마커 CD133 또는 약물저항성 마커 TUBB3(Tubulin Beta 3 Class Ⅲ) 중 어느 하나 이상의 발현을 억제함으로써, 항암 활성을 나타낼 수 있다.
또한, 일 예로서, 본 발명의 암의 예방 또는 치료용 약학 조성물은 암세포의 세포사(Apoptosis) 마커 PUMA(p53 upregulated modulator of apoptosis) 유전자의 발현을 증가시킴으로써, 암세포의 증식을 효과적으로 조절할 수 있다.
본 명세서에서 사용된 용어 "암"은 세포가 정상적인 성장한계를 무시하고 분열 및 성장하는 공격적(aggressive) 특성, 주위 조직에 침투하는 침윤적(invasive) 특성, 및 체내의 다른 부위로 퍼지는 전이적(metastatic) 특성을 갖는 세포에 의한 질병을 총칭하는 의미이다.
일 예로서, 상기 암은 유방암, 폐암, 위암, 간암, 혈액암, 뼈암, 췌장암, 피부암, 두경부암, 피부 또는 안구 흑색종, 자궁육종, 난소암, 직장암, 항문암, 대장암, 난관암, 자궁내막암, 자궁경부암, 소장암, 내분비암, 갑상선암, 부갑상선암, 신장암, 연조직종양, 요도암, 전립선암, 기관지암, 또는 골수암일 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 암의 예방 또는 치료용 약학 조성물은 개별 치료제로 투여하거나, 다른 사용중인 항암제와 병용투여하여 사용할 수 있다.
상기 암의 예방 또는 치료용 약학 조성물은 상기 화학식 1로 표시되는 화합물, 이의 약제학적으로 허용가능한 염, 수화물 또는 용매화물의 약제학적 유효량으로 포함될 수 있다.
상기 암의 예방 또는 치료용 약학 조성물의 적법한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 따라 다양한 방법으로 처방될 수 있다. 일 예로서, 상기 투여량은 1일 당 0.001-10,000 mg/kg(체중) 일 수 있다.
상기 암의 예방 또는 치료용 약학 조성물에 포함되는 유효 성분의 농도는 치료 목적, 환자의 상태, 필요 기간, 질환의 위중도 등을 고려하여 결정되며 특정 범위의 농도로 한정되지 않는다.
상기 암의 예방 또는 치료용 약학 조성물은 경구 또는 비경구로 투여할 수 있고, 비경구로 투여되는 경우, 정맥내 주입, 피하 주입, 근육 주입, 복강 주입, 경피 투여 등으로 투여할 수 있으나, 이에 한정되는 것은 아니다. 상기 암의 예방 또는 치료용 약학 조성물은 각각 통상의 방법에 따라 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있으나, 이에 한정되는 것은 아니다.
상기 암의 예방 또는 치료용 약학 조성물은 당해 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제형화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기내에 내입시켜 제조될 수 있다.
상기 약제학적으로 허용되는 담체는 경구 투여 시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제로 투여 시에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있으나, 이에 한정되는 것은 아니다.
상기 암의 예방 또는 치료용 약학 조성물의 제형은 전술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 일 예로서, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서(elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제로 투여 시에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있으나, 이에 한정되는 것은 아니다. 또한, 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제형할 수 있으나, 이에 한정되는 것은 아니다.
한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말디톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있으나, 이에 한정되는 것은 아니다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 또 다른 측면으로, 본 발명의 실시예에 의한 암의 예방 또는 개선용 기능성 식품 조성물은 전술한 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 유효성분으로 포함한다.
본 발명의 식품 조성물은 건강기능식품으로서 사용될 수 있다. 상기 "건강기능식품"이라 함은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능성"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다.
상기 암의 예방 또는 개선용 기능성 식품 조성물은 전술한 화학식 1의 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물 이외에, 당 분야에서 식품 조성물에 통상적으로 이용되는 성분들을 포함할 수 있다. 일 예로서, 유기산, 인산염, 항산화제, 유당 카제인, 덱스트린, 포도당, 설탕 및 솔비톨로 이루어진 군으로부터 선택되는 하나 이상의 첨가제를 더 포함할 수 있다. 또한 여러 가지 영양제, 비타민, 광물 (전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제, 팩트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 추가로 함유할 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 식품 조성물은 암의 예방 및/또는 개선을 목적으로, 조성물 총 중량에 대하여 상기 화학식 1의 화합물을 0.01 내지 95 %, 바람직하게는 1 내지 80 % 중량 백분율로 포함할 수 있으나, 이에 한정되는 것은 아니다.
상기 암의 예방 또는 개선용 기능성 식품 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들면 정제, 과립제, 환제, 캡슐제, 액상 제제, 시럽 또는 음료의 형태로 제공될 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 실시예에 의한 암의 예방 또는 치료방법은 전술한 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 유효성분으로 포함한다.
본 발명의 실시예에 의한 암의 예방 또는 치료방법은 항암제에 대한 내성을 억제시킬 수 있다.
또한, 본 발명의 실시예에 의한 암의 예방 또는 치료방법은 항암제의 민감성을 증진시킬 수 있다.
본 발명의 실시예에 의한 암의 예방, 개선 또는 치료를 위한 약제의 제조에 사용하기 위한 용도는 전술한 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 유효성분으로 포함한다.

본 명세서에서 사용된 용어 "용매 화합물"은 분자 복합체(molecular complex)를 설명하기 위해 사용되었으며, 본 발명에 따른 화합물 및 하나 이상의 약제학적으로 허용되는 용매 분자들로 존재할 수 있고, 예를 들어, 에탄올의 화학양론적 양(stoichiometric amount)을 포함한다. 용어 "수화물"은 용매가 물일 때, 사용된다.

<중간체의 제조예>
(1) <화합물 a> 에틸 3-아미노-4-하이드록시벤조에이트, Ethyl 3-amino-4-hydroxybenzoate
에탄올 (10 mL)에 녹인 3-아미노-4-하이드록시벤조산 (0.76 g, 5.0 mmol) 용액에 진한 황산 (0.5 mL)을 질소 분위기 하에 0℃에서 천천히 첨가한다. 첨가가 완료된 후, 반응 혼합물을 서서히 승온하여 70℃에서 15시간 동안 환류 시킨다. 반응이 끝나면 온도를 상온으로 낮춰준 뒤 용매를 감압 제거하고 포화 NaHCO3 수용액을 사용하여 반응 혼합물의 pH를 8로 조정한 다음 아세트산에틸 (50 mL)로 2번 추출하고 유기층은 물 (30 mL)로 3번, 포화 염화나트륨 용액 (30 mL)으로 한번 씻어주고 무수 황산마그네슘으로 건조하여 감압 증류 후 에틸 3-아미노-4-하이드록시벤조에이트 (a) (0.81 g, 89.2 %)를 옅은 갈색 고체로 얻었다.

(2) <화합물 b> 에틸 4-아미노-3-하이드록시벤조에이트, Ethyl 4-amino-3-hydroxybenzoate
에탄올 (12 mL)에 녹인 4-아미노-3-하이드록시벤조산 (0.92 g, 6.0 mmol) 용액에 진한 황산 (0.61 mL)을 질소 분위기 하에 0℃에서 첨가한다. 첨가가 완료된 후, 반응 혼합물을 서서히 승온하여 70℃에서 16시간 동안 환류 시킨다. 반응이 끝나면 온도를 상온으로 낮춰준 뒤 용매를 감압 제거하고 포화 NaHCO3 수용액을 사용하여 반응 혼합물의 pH를 8로 조정한 다음 아세트산에틸 (60 mL)로 2번 추출하고 유기층은 물 (30 mL)로 3번, 포화 염화나트륨 용액 (30 mL)으로 한번 씻어주고 무수 황산마그네슘으로 건조하여 감압 증류 후 에틸 4-아미노-3-하이드록시벤조에이트 (b) (1.01 g, 93.5 %)를 옅은 갈색 고체로 얻었다.

(3) <화합물 c> 2,4-디하이드록시-5-니트로벤조산, 2,4-Dihydroxy-5-nitrobenzoic acid
아세트산 (17 mL)에 질산 (69 %, 14 mL)을 첨가하고 0℃로 냉각시킨 뒤, 2,4-디하이드록시벤조산 (3.85g, 25.0mmol)을 15-20분에 걸쳐 천천히 첨가한다. 첨가 완료 후, 반응 혼합물을 13시간 동안 교반 한다. (발열반응이기 때문에 반응용기의 온도가 올라가지 않도록 주의한다.) 반응 완료 후, 고체를 여과하고 최소의 물을 사용하여 세척하고 건조하고 아세트산에틸 (130 mL)에 용해시키고 무수 황산마그네슘으로 건조하여 감압 증류 후 2,4-디하이드록시-5-니트로벤조산 (c) (2.89 g, 58.1 %)을 연분홍색 고체로서 수득하고, 추가 정제없이 다음 단계에 사용하였다.

(4) <화합물 d> 에틸 2,4-디하이드록시-5-니트로벤조에이트, Ethyl 2,4-dihydroxy-5-nitrobenzoate
에탄올 (18 mL)에 녹인 2,4-디하이드록시-5-니트로벤조산 (1.42 g, 7.13 mmol) 용액에 진한 황산 (0.4 mL)을 질소 분위기 하에 0℃에서 천천히 첨가한다. 첨가 완료 후, 반응 혼합물을 서서히 승온하여 70℃에서 60시간 동안 환류 시킨다. 반응이 끝나면 온도를 상온으로 낮춰준 뒤 용매를 감압 제거하고 아세트산에틸 (180 mL)로 혼합물을 용해시키고 물 (30 mL)로 3번, 포화 염화나트륨 용액 (30 mL)으로 한번 씻어주고 무수 황산나트륨으로 건조하여 감압 증류 후 컬럼크로마토그래피 (아세트산에틸/헥세인 = 1/8)로 분리하여 에틸 2,4-디하이드록시-5-니트로벤조에이트 (d) (1.01 g, 62.3 %)를 흰색 고체로 얻었다.

(5) <화합물 e> 에틸 5-아미노-2,4-디하이드록시벤조에이트, Ethyl 5-amino-2,4-dihydroxybenzoate
1,4-다이옥세인/증류수 (3/1, 10 mL)에 녹아 있는 에틸 2,4-디하이드록시-5-니트로벤조에이트 (d) (0.22 g, 1.0 mmol), 활성화된 아연가루 (0.52g, 8.0mmol, 8.0 eq) 용액에 염화암모늄 (0.42g, 8.0 mmol, 8.0 eq)을 0℃에서 천천히 첨가한다. 반응 온도를 서서히 실온으로 가온 하고 3시간 동안 교반 한다. 반응 완료 후, 규죠토® 패드를 통해 여과하고 아세트산에틸 (45 mL)로 세척한다. 여액의 용매를 감압 제거하고 물 (10 mL)을 첨가하여 아세트산에틸 (50 mL)로 3번 추출하고 유기층은 물 (20 mL)로 2번, 포화 염화나트륨 용액 (20 mL)으로 한번 씻어주고 무수 황산나트륨으로 건조하여 감압 증류 후 컬럼크로마토그래피 (아세트산에틸/헥세인 = 1/2 to 1/1)로 분리하여 에틸 5-아미노-2,4-디하이드록시벤조에이트 (e) (0.02 g, 11.4 %)를 얻었다.

벤즈옥사졸아민 유도체 화합물 1-10의 합성
(1) <화합물-1> 에틸 2-((4-메톡시페닐)아미노)벤조[d]옥사졸-5-카복실레이트, Ethyl 2-((4-methoxyphenyl)amino)benzo[d]oxazole-5-carboxylate
무수 메탄올 (6 mL)에 녹아 있는 에틸 3-아미노-4-하이드록시벤조에이트 (a) (0.18 g, 1.0 mmol) 용액에 4-메톡시페닐이소티오시아네이트 (0.14mL, 1.0mmol)를 질소 분위기 하에 첨가하고 혼합물을 36시간 동안 교반 한다. 반응이 완료된 후 용매를 감압 제거하고 헥세인 용액으로 씻어주고 진공하에 건조하여 싸이오요소 유도체를 얻는다. 이 화합물을 추가 정제없이 다음 단계에 바로 사용한다.

이하, 본 발명의 각 실시예에 의한 조성물의 항암 용도를 입증하는 실험에 관한 것이다.
<세포 배양>
본 연구에 사용된 세포주(폐암세포주;A549, 대장암세포주;HT29, 백혈병세포주;Molt-4, 자궁경부암세포주;HeLa)는 10% 우태아 혈청을 포함한 Dulbecco's Modified Eagle Medium(DMEM, Hyclone, USA) 또는 RPMI1640 배지를 사용하여 37℃, 5% CO2로 조정된 CO2 배양기에서 배양하였다. 세포가 90% 증식이 되었을 때 실험에 사용하였으며, 20 passage가 넘지 않도록 조절하였다. 배양된 세포는 0.25% trypsin-EDTA로 부유시킨 후 혈구계산기로 세포 수를 산정하고, 유전자 발현 실험을 위해 12 well plate에 5Х106 cells/dish이 되도록 분주하여 약물 처리 20시간 후 유전자를 추출하였다.
<유전자 분석>
유전자 발현 분석을 위해 Trizol Reagent(Life Technologies, CA, USA)를 이용하여 각 세포로부터 총 RNA를 추출하였다. 즉, Trizol 용액 1 ml를 첨가하여 세포를 용해시키고 실온에서 5분 동안 방치 후 클로로포름 200 μL를 첨가하여 13,500 rpm에서 15분 동안 원심분리 하였다. 투명한 상층액(500 μl)을 취하여 새로운 튜브로 옮기고 동량의 이소프로필 알코올을 첨가한 후 13,500 rpm에서 10분 동안 원심분리 하여 RNA를 침강시켰다. RNA 침전물을 diethyl pyrocarbonate(DEPC, Sigma-Aldrich) 처리한 증류수로 희석한 70% 에탄올 0.75 ml 로 세척한 후 공기 중에서 건조해 역전사 샘플(reverse transcription sample)로 사용하였다. 1차 가닥(first strand) cDNA 합성은 추출된 총 RNA 1 μg을 사용하여 수행되었고, TOPscript Reverse Transcription system(enzynomics, Daejeon, Korea)과 oligo dT primer를 사용하여 역전사 반응을 수행하였다. qPCR 분석은 Rotor-Gene 6000(Qiagen, CA, USA)을 사용하였으며, CD133, TUBB3 및 PUMA 유전자 증폭을 위한 프라이머를 이용하여 유전자의 발현을 정량적으로 측정하였고 housekeeping 유전자인 β-actin에 정상화(normalization) 시킨 상대 수치를 비교 분석하였다.

도 1을 참조하면, 특히 화합물-2, 5, 7, 8, 9, 10에 의해 HT29 세포주에서 암세포 마커 유전자인 CD133 유전자의 발현이 유의한 수준으로 감소하였다.
도 2를 참조하면, 특히 화합물-1, 2, 5, 6, 8에 의해 Molt-4 세포주에서 약물 저항성 마커 유전자인 TUBB3 유전자 발현이 유의한 수준으로 감소하였다.
도 3을 참조하면, 특히 화합물-1, 8, 9, 10에 의해 A549 세포주에서 세포의 사멸을 유도하는 PUMA 유전자의 발현이 증가하였다.
도 4를 참조하면, 특히 화합물-2, 3, 4, 5, 7, 8, 10에 의해 HeLa 세포주에서 세포의 사멸을 유도하는 PUMA 유전자의 발현이 증가하였다.
위와 같은 결과는 본 발명에서 제시하는 벤즈옥사졸아민 유도체 화합물이 암세포 증식억제, 내성 조절 및 세포사를 효과적으로 조절함으로써, 항암제제로서 잠재성이 높음을 의미한다.
Claims (21)
- 하기 화학식 1 로 표시되는화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물:[화학식 1][화학식 2][화학식 3]상기 화학식 1에서, R1 내지 R9는 서로 동일하거나 상이하며, 각각 독립적으로 수소, 중수소, 히드록시기, 니트로기, 니트릴기, 할로겐, 아미노기, 이미드기, 에스터기, 카보닐기, 치환 또는 비치환된 C1~C100의 알킬기, 치환 또는 비치환된 C2~C100의 알케닐기, 치환 또는 비치환된C2~C100의 알키닐기, 치환 또는 비치환된C3~C100의 시클로알킬기, 치환 또는 비치환된C6~C100의 아릴기, 치환 또는 비치환된 핵원자수 3 내지 100개의 헤테로시클로알킬기, 치환 또는 비치환된 핵원자수 5 내지 100의 헤테로아릴기, 치환 또는 비치환된C1~C100의 알콕시기 및 치환 또는 비치환된 C6~C100의 아릴옥시기 중에서 선택되고,R1 내지 R4중 적어도 어느 하나 이상은 반드시 상기 화학식 2 또는 화학식 3으로 표시되는 치환기이고,상기 화학식 2에서, R10은 수소, C1~C99의 알킬, C2~C99의 알케닐기, C2~C99의 알키닐기, C3~C99의 시클로알킬기, C6~C99의 아릴기 중에서 선택되는 어느 하나이고,상기 화학식 3에서, R11은 수소, C1~C99의 알킬, C2~C99의 알케닐기, C2~C99의 알키닐기, C3~C99의 시클로알킬기, C6~C99의 아릴기 중에서 선택되는 어느 하나이다.
- 제 1 항에 있어서,상기 화학식 1에서 R1 내지 R4는 상기 화학식 2 또는 화학식 3으로 표시되는 치환기 이외에는 수소, 히드록시기, C1~C100의 알콕시기 및 니트로기 중에서 선택되는 어느 하나인,화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물.로 허용가능한 이들의 염, 수화물 또는 용매화물.
- 제 1 항에 있어서,상기 화학식 1에서 R5 내지 R9는 수소, 히드록시기, C1~C100의 알콕시기, 니트로기, 할로겐 및 할로겐이 치환된 C1~C100의 알킬기 중에서 선택되는 어느 하나인,화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물.
- 제 4 항 내지 제 7 항 중 어느 한 항에 있어서,상기 R5 내지 R9는 수소, 히드록시기, C1~C100의 알콕시기, 니트로기, 할로겐 및 할로겐이 치환된 C1~C100의 알킬기 중에서 선택되는 어느 하나인,화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물.
- 제 4 항 내지 제 7 항 중 어느 한 항에 있어서,R5 내지 R9중 어느 하나는 -OMe, -CF3, -Br 및 -NO2 중에서 선택되는 어느 하나이고,상기 -OMe, -CF3, -OMe, -Br 및 -NO2 중에서 선택되는 어느 하나의 치환기 이외에는 모두 수소인,화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물.
- 제 1 항에 있어서,상기 화합물은 하기 화합물 중에서 선택되는 어느 하나 이상인,화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물:에틸 2-((4-메톡시페닐)아미노)벤조[d]옥사졸-5-카복실레이트 (Ethyl 2-((4-methoxyphenyl)amino)benzo[d]oxazole-5-carboxylate),에틸 2-((4-(트리플루오로메틸)페닐)아미노)벤조[d]옥사졸-5-카복실레이트 (Ethyl 2-((4-(trifluoromethyl)phenyl)amino)benzo[d]oxazole-5-carboxylate),에틸 2-((2-메톡시페닐)아미노)벤조[d]옥사졸-5-카복실레이트 (Ethyl 2-((2-methoxyphenyl)amino)benzo[d]oxazole-5-carboxylate),에틸 2-((4-메톡시페닐)아미노)벤조[d]옥사졸-6-카복실레이트 (Ethyl 2-((4-methoxyphenyl)amino)benzo[d]oxazole-6-carboxylate),에틸 2-((4-(트리플루오로메틸)페닐)아미노)벤조[d]옥사졸-6-카복실레이트 (Ethyl 2-((4-(trifluoromethyl)phenyl)amino)benzo[d]oxazole-6-carboxylate),에틸 2-((2-메톡시페닐)아미노)벤조[d]옥사졸-6-카복실레이트 (Ethyl 2-((2-methoxyphenyl)amino)benzo[d]oxazole-6-carboxylate),에틸 6-하이드록시-2-((2-메톡시페닐)아미노)벤조[d]옥사졸-5-카복실레이트 (Ethyl 6-hydroxy-2-((2-methoxyphenyl)amino)benzo[d]oxazole-5-carboxylate),에틸 2-((2-브로모페닐)아미노)벤조[d]옥사졸-5-카복실레이트 (Ethyl 2-((2-bromophenyl)amino)benzo[d]oxazole-5-carboxylate),에틸 2-((4-니트로페닐)아미노)벤조[d]옥사졸-5-카복실레이트 (Ethyl 2-((4-nitrophenyl)amino)benzo[d]oxazole-5-carboxylate), 및에틸 2-((3-니트로페닐)아미노)벤조[d]옥사졸-5-카복실레이트 (Ethyl 2-((3-nitrophenyl)amino)benzo[d]oxazole-5-carboxylate).
- 제 1 항의 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 유효성분으로 포함하는,암의 예방 또는 치료용 약학 조성물.
- 제 10 항의 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 유효성분으로 포함하는,암의 예방 또는 치료용 약학 조성물.
- 제 11 항에 있어서,상기 조성물은 항암제에 대한 내성을 억제시키는,암의 예방 또는 치료용 약학 조성물.
- 제 11 항에 있어서,상기 조성물은 항암제의 민감성을 증진시키는,암의 예방 또는 치료용 약학 조성물.
- 상기 암은 유방암, 폐암, 위암, 간암, 혈액암, 뼈암, 췌장암, 피부암, 두경부암, 피부 또는 안구 흑색종, 자궁육종, 난소암, 직장암, 항문암, 대장암, 난관암, 자궁내막암, 자궁경부암, 소장암, 내분비암, 갑상선암, 부갑상선암, 신장암, 연조직종양, 요도암, 전립선암, 기관지암, 또는 골수암 중에서 선택되는 어느 하나 이상인,암의 예방 또는 치료용 약학 조성물.
- 제 1 항의 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 유효성분으로 포함하는,암의 예방 또는 개선용 식품 조성물.
- 제 1 항 내지 제 10 항 중 어느 하나의 항의 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 대상체에게 투여하는 단계를 포함하는 암의 예방 또는 치료방법.
- 제 17 항에 있어서,상기 방법은 항암제에 대한 내성을 억제시키는,암의 예방 또는 치료방법.
- 제 17 항에 있어서,상기 방법은 항암제의 민감성을 증진시키는,암의 예방 또는 치료방법.
- 제 17 항에 있어서,상기 암은 유방암, 폐암, 위암, 간암, 혈액암, 뼈암, 췌장암, 피부암, 두경부암, 피부 또는 안구 흑색종, 자궁육종, 난소암, 직장암, 항문암, 대장암, 난관암, 자궁내막암, 자궁경부암, 소장암, 내분비암, 갑상선암, 부갑상선암, 신장암, 연조직종양, 요도암, 전립선암, 기관지암, 또는 골수암 중에서 선택되는 어느 하나 이상인, 방법.
- 제 1 항 내지 제 10 항 중 어느 하나의 항의 화합물, 약학적으로 허용가능한 이들의 염, 수화물 또는 용매화물을 암의 예방, 개선 또는 치료를 위한 약제의 제조에 사용하기 위한 용도.
