Abstract
Description
[1]多能性幹細胞から分化誘導されたペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む、血管内皮増殖因子(VEGF)高発現ペリサイト様細胞の製造方法。
[2]ペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程の前に以下の工程を含む、[1]に記載の製造方法:
(a)多能性幹細胞を初期中胚葉細胞に分化させる工程、及び、
(b)前記工程(a)で得られた初期中胚葉細胞をペリサイト様細胞に分化させる工程。
[3]前記工程(b)が以下の工程を含む、[2]に記載の製造方法:
(b-1)前記工程(a)で得られた初期中胚葉細胞のスフェロイドを形成させる工程、及び、
(b-2)前記スフェロイドを形成した初期中胚葉細胞をペリサイト様細胞に分化させる工程。
[4]前記工程(b-1)のスフェロイドを形成させる工程が、培養面にすり鉢状の微細な複数のウェルを隙間なく備える培養容器内で行われる、[3]に記載の製造方法。
[5]前記多能性幹細胞が、ヒト多能性幹細胞である、[1]~[4]のいずれか1つに記載の製造方法。
[6]前記多能性幹細胞が、胚性幹細胞(ES細胞)又は人工多能性幹細胞(iPS細胞)である、[1]~[5]のいずれか1つに記載の製造方法。

本発明は、多能性幹細胞から分化誘導されたペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む、VEGF高発現ペリサイト様細胞の製造方法(以下、「本発明の製造方法」とも称する)を提供する。
「ペリサイト」とは、脳、末梢又は網膜などに存在する微小血管壁又は毛細血管壁を取り巻くように存在する細胞のことで、血管周皮細胞とも称される。その機能は上述した通りであり、血管内皮細胞を被覆する細胞として血管の成熟・安定化や血液脳関門の維持など正常な血流調節に重要な役割を担っている(Daneman R et al., Nature, (2010), 468: 562-568、Armulik A et al., Dev. Cell, (2011), 21: 193-215)。ペリサイトの細胞機能に障害が生じると、ペリサイト本来の機能が損なわれ、糖尿病網膜症などの重篤な血流障害関連疾患につながる。また、骨格筋由来のペリサイトの中には筋肉や骨への分化能を持つメソアンジオブラスト(Mesoangioblast)と呼ばれる細胞の存在が明らかとなっている(Gerli MFM et al., J. Vis. Exp., (2014), 83: 50523、Gerli MFM et al., Stem Cell Rep., (2019), 12: 461-473、Shimatani K et al., Am. J. Physiol. Heart Circ. Physiol., (2021), on line: https://doi.org/10.1152/ajpheart.00470.2020)。
本明細書において「ペリサイト様細胞」とは、多能性幹細胞から分化誘導された、初代ペリサイト(下記参照)と同様の性質をもつ細胞を意味する。ペリサイト様細胞がペリサイトと同様の性質を有することは、公知の方法で確認することができる(Armulik A et al., Dev. Cell, (2011), 21: 193-215)。多能性幹細胞からペリサイト様細胞への分化誘導は、当業者に公知の方法を用いて行うことができる(WO2013/108039, WO2009/156151, US9771561, US9868939, US2015/0368609, US2017/0342384, US2019/0316094)。例えば、多能性幹細胞からペリサイト様細胞への分化誘導は、後記の〔多能性幹細胞のペリサイト様細胞への分化誘導方法〕の項に記載の方法を用いて行うことができる。
本明細書において「多能性幹細胞」とは、生体に存在する多くの性質・形態の異なる細胞に分化可能である多能性を有し、かつ、増殖能をも併せもつ幹細胞を意味する。本発明における好ましい多能性幹細胞は、ヒト多能性幹細胞である。1つの実施形態において、本発明の製造方法及び本発明の選別方法において使用される多能性幹細胞より分化誘導されたペリサイト様細胞を含む集団は、ヒト多能性幹細胞より分化誘導されたペリサイト様細胞を含む集団である。
ES細胞は、ヒトやマウスなどの哺乳動物の初期胚(例えば胚盤胞)の内部細胞塊から樹立された、分化多能性(pluripotency)と自己複製による増殖能を有する幹細胞である。

本発明において、ペリサイト様細胞は、前述の通り、多能性幹細胞からペリサイト様細胞に分化誘導して得ることができる。多能性幹細胞からペリサイト様細胞に分化誘導する方法は特に限定されないが、1つの実施形態において、以下の工程(a)及び(b)を含む方法によることができる:
(a)多能性幹細胞を初期中胚葉細胞に分化させる工程、及び、
(b)前記工程(a)で得られた初期中胚葉細胞をペリサイト様細胞に分化させる工程。
(b-1)前記工程(a)で得られた初期中胚葉細胞のスフェロイドを形成させる工程、及び、
(b-2)前記スフェロイドを形成した初期中胚葉細胞をペリサイト様細胞に分化させる工程。
本発明の製造方法では、ペリサイト様細胞を含む集団から「CD56(-)のペリサイト様細胞を選別する」。また、本発明の選別方法では、ペリサイト様細胞を含む集団からVEGF高発現ペリサイト様細胞の集団の選別において、「CD56(-)の細胞表面マーカーを用いる」。本発明の選別方法における「CD56(-)の細胞表面マーカーを用いる」とは、本発明の製造方法における「CD56(-)のペリサイト様細胞を選別する」と同義であり、これらを以下、まとめて「本発明の選別工程」と称する。本発明の選別工程は、VEGF高発現ペリサイト様細胞、又はVEGF高発現ペリサイト様細胞の集団の選別に用いられる。
CD56(GeneID: 4684)はneural cell adhesion molecule 1(NCAM1)、NCAM、MSK39とも呼ばれる免疫グロブリンスーパーファミリーの一員で、細胞接着分子として機能する糖タンパク質である。CD56は胚形成、発生及び神経細胞間の接触を介した相互作用に関与している。ヒトCD56には複数のアイソフォームが存在している(Lanier LL et al., J. Immunol., (1991), 146: 4421-4426、van Duijnhoven HLP et al., Int. J. Cancer, (1992), 50:118-123、Rossel RJ et al., Biochim. Biophys. Acta, (1992), 1130: 95-96)。例えば、ヒトCD56タンパク質の1型アイソフォームのNCBIアクセッション番号はNP_000606.3である。
本発明は、多能性幹細胞から分化誘導されたペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、VEGF高発現ペリサイト様細胞(以下、「本発明の細胞」とも称する)を提供する。
本発明の細胞及び本発明の細胞集団を培養・増殖する方法は、特に限定されず、当該技術分野で公知のペリサイトの培養・増殖方法を用いることができる。
本発明はまた、本発明の細胞又は本発明の細胞集団(以下、本項において纏めて「本発明の細胞等」と称す)を含む医薬組成物(「本発明の医薬組成物」とも称する)を提供する。当該医薬組成物は、当該分野において通常用いられる賦形剤、即ち、薬剤用賦形剤や薬剤用担体等を用いて、通常使用される方法によって調製することができる。医薬組成物の製剤化にあたっては、薬学的に許容される範囲で、これら剤型に応じた賦形剤、担体、添加剤等を使用することができる。1つの実施形態において、本発明の医薬組成物は、多能性幹細胞から分化誘導されたペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、VEGF高発現ペリサイト様細胞を含む。1つの実施形態において、本発明の医薬組成物は、(a)多能性幹細胞を初期中胚葉細胞に分化させる工程、(b)前記工程(a)で得られた初期中胚葉細胞をペリサイト様細胞に分化させる工程、及び(c)前記ペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、VEGF高発現ペリサイト様細胞を含む。1つの実施形態において、本発明の医薬組成物は、(a)多能性幹細胞を初期中胚葉細胞に分化させる工程、(b-1)前記工程(a)で得られた初期中胚葉細胞のスフェロイドを形成させる工程、(b-2)前記スフェロイドを形成した初期中胚葉細胞をペリサイト様細胞に分化させる工程、及び(c)前記ペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、VEGF高発現ペリサイト様細胞を含む。1つの実施形態において、本発明の医薬組成物は、(a)多能性幹細胞を初期中胚葉細胞に分化させる工程、(b-1’)培養面にすり鉢状の微細な複数のウェルを隙間なく備える培養容器内で、前記工程(a)で得られた初期中胚葉細胞のスフェロイドを形成させる工程、(b-2)前記スフェロイドを形成した初期中胚葉細胞をペリサイト様細胞に分化させる工程、及び(c)前記ペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、VEGF高発現ペリサイト様細胞を含む。
「血管内皮細胞」は、血管内腔を裏打ちする一層の扁平状の細胞で、血管の緊張度や血管透過性の調節、血管新生、凝血促進など多彩な機能を有する。動・静脈レベルの大きな血管は内膜、中膜、及び外膜で構成される3層構造を取っており、それぞれ主に、血管内皮細胞、平滑筋細胞、繊維芽細胞で構成されている。一方、毛細血管レベルの小さな血管では血管内皮細胞の管腔構造がペリサイトにより囲まれている。成熟した毛細血管においては、ペリサイトは血管内皮細胞と基底膜を共有し、その中に埋まり込む形で存在している。近年、ペリサイトと血管内皮細胞が相互に細胞シグナル伝達を行うことで、分化、増殖を調節し、毛細血管の成熟、安定化、維持、及び基底膜の形成、細胞外マトリックスの沈着などに重要な役割を果たすことが知られている。
DMEM/F12培地(ナカライテスク, 11581-15)25mLにマトリゲル(登録商標)ヒトES細胞最適化マトリックス(Corning, 354277)を230μL添加し、6ウェルプレート(Iwaki, 3810-006)の3ウェルに1.5mLずつ添加し、室温で3時間静置してマトリゲルコーティングプレートを作製した。ROCK阻害剤である10μM Y-27632 (ナカライテスク, 08945)を添加したmTeSR1培地(STEMCELL Technologies, ST-85850)に懸濁したヒトES細胞(国立成育医療研究センター, SEES6)をマトリゲルコーティングプレートに各5.0×105個/ウェルで前記3ウェルに播種し、37℃、5% CO2雰囲気下で24時間培養した。その後、上清を除き、STEMdiff Mesoderm Induction Medium(STEMCELL Technologies, 05220)を前記各ウェルに3.5mL添加した。24時間後に再度上清を除き、STEMdiff Mesoderm Induction Mediumを前記各ウェルに3.5mL添加し、さらに37℃、5% CO2雰囲気下で24時間培養した。上清を除き、PBSで洗浄し、プレートから細胞を剥離するためにTrypLE Express(Thermo Fisher Scientific, 12604013)を前記各ウェルに1mL添加し、37℃プレート上で5分間静置した。次に、前記各ウェルにスフェロイド形成培地(下記参照)を5mL添加し、剥離した細胞を回収した。回収した細胞が初期中胚葉細胞へ分化しているかどうかをフローサイトメトリー法にて確認した。具体的には、初期中胚葉細胞の細胞表面マーカーとしてPDGFRα及びAPLNRを選定し、各細胞表面マーカーに対する抗体としてそれぞれ、BV421 Anti-Human CD140α (PDGFRα) antibody(BD, 562799)、APC Anti-Human APJ antibody(R&D Systems, FAB8561A-025)を使用した。回収した細胞懸濁液4mLを300g、室温、5分間の条件で遠心分離した。上清を除き、2%FBS(Sigma-Aldrich)含有PBSを1mL添加し、再度300g、室温、5分間の条件で遠心分離した。上清を除き、2%FBS含有PBSを100μL添加し、細胞を懸濁後、FcR Blocking Reagent(Miltenyi Biotec, 130-059-901)を5μL添加して氷上で30分間静置した。氷上で30分静置後の細胞懸濁液20μLに対してBV421 Anti-Human CD140α antibodyを5μL、APC Anti-Human APJ antibodyを10μL添加し、氷上にて20分間反応させた。反応後、2%FBS含有PBSで2回洗浄した。また、マトリゲルコーティングプレート上でmTeSR1培地を用いて培養していたヒトES細胞(SEES6)についても上清を除き、PBSで洗浄し、プレートから細胞を剥離するためにTrypLE Expressを1mL添加し、37℃プレート上で5分間静置した。次に、mTeSR1培地を5mL添加し、剥離した細胞を回収した。回収した細胞の懸濁液のうち、4mLを300g、室温、5分間の条件で遠心分離した。上清を除き、2%FBS含有PBSを150μL添加し、さらにFcR Blocking Reagentを5μL添加して氷上で20分間静置した。300g、室温、5分間の条件で遠心分離後に上清を除き、2%FBS含有PBSを60μL添加し細胞を懸濁した。この細胞懸濁液20μLに対してBV421 Anti-Human CD140α antibodyを5μL、APC Anti-Human APJ antibodyを10μL添加し、氷上にて20分間反応させた。反応後、2%FBS含有PBSで2回洗浄した。2種類の抗体と反応させた上記2種類の細胞をAttune NxT Flow Cytometer (Invitrogen)を用いて解析した結果、STEMdiff Mesoderm Induction Mediumを用いて培養する前のヒトES細胞はPDGFRα(+)かつAPLNR(+)の細胞がほとんど存在しなかったのに対して(図1左)、STEMdiff Mesoderm induction Mediumで48時間培養後の細胞は、55%以上の細胞がPDGFRα(+)かつAPLNR(+)を示した(図1右)。この結果は、STEMdiff Mesoderm Induction Mediumを用いた培養によって、ES細胞から初期中胚葉細胞への分化誘導が進んだことを示している。前記、ES細胞をSTEMdiff Mesoderm Induction Mediumを用いて培養した細胞を以下で「初期中胚葉細胞」と称する。
初期中胚葉細胞のスフェロイドを形成させるためのスフェロイド形成培地を文献(Vodyanik MA et al., Cell Stem Cell, (2010), 7: 718-729)記載の方法を改良し、作製した。具体的には、40% MegaCell Dulbecco’s Modified Eagle’s Medium (Sigma-Aldrich, M3942)、25% StemSpan SFEM (STEMCELL Technologies, 09650)、25% Human Endothelial SFM (Thermo Fisher Scientific, 11111044)、10% BIT 9500 Serum Substitute (STEMCELL Technologies, 09500)になるようにそれぞれを混合した。さらに、そこに1% GlutaMax (Thermo Fisher Scientific, 35050061)、0.1% Ex-CYTE(登録商標)NZ Growth Enhancement Media Supplement (Merck, 81150N)、100μM Monothioglycerol(富士フイルム和光純薬, 195-15791)、50μg/mL L-アスコルビン酸 2-リン酸 セスキマグネシウム塩 水和物(Sigma-Aldrich, A8960)、20ng/mL Animal-free Recombinant Human FGF basic-TS(Proteintech, HZ-1285)になるようにそれぞれを添加し、スフェロイド形成培地を調製した。
実施例1にて取得した初期中胚葉細胞1.7×105個にスフェロイド形成培地を10mL添加し、EZSPHERE(登録商標)ディッシュ(Iwaki, 11-0434)に播種し、37℃、5% CO2、5% O2雰囲気下で培養した。培養開始から4日後にスフェロイド形成培地10mLを徐々に添加した。培養開始から6、8、11日後に培養上清を10mL除去し、新たにスフェロイド形成培地10mLを徐々に添加した。培養開始から12日後にスフェロイドを含む培養液をFalcon(登録商標)100μmセルストレイナー(Corning, 352360)に通し、100μm以上のサイズのスフェロイドを回収した。
最終濃度3μg/mL Fibronectin(Corning, 356008)、及び10μg/mL Human Type1 Collagen(協和ファーマケミカル, Y-1)を含むPBSをFalcon(登録商標)60mm 細胞培養用ディッシュ(Corning, 353002)に2mL添加し、37℃で3時間静置した。実施例2にて回収したスフェロイド全てをペリサイト分化誘導培地(下記参照)12mLに懸濁後、前記ディッシュ2枚に播種して37℃、5% CO2、5% O2雰囲気下で培養した。培養開始から3日後に前記ディッシュの底面に単層状への細胞増殖が観察された。この時点で上清を除き、PBSで洗浄後、細胞を剥離するために前記ディッシュにAccutase(Innovative Cell Technologies, AT104)を1mL添加し、37℃のプレート上で5分間静置した。前記ディッシュにペリサイト増殖培地(下記参照)を4mL添加して細胞懸濁液を回収し、300g、室温、5分間の条件で遠心分離した。上清を除き、ペリサイト増殖培地を2mL添加し、細胞を2×105個/ディッシュとなるように60mmコラーゲンコーティングディッシュ(Corning, 354401)に播種し37℃、5% CO2、5% O2雰囲気下で培養した。ペリサイト分化誘導培地で培養し、続けてペリサイト増殖培地で培養、回収した細胞はペリサイト様細胞であり、以下の実験で用いる。
組成は以下の通りである:
・50% Stemline(登録商標)II Hematopoietic Stem Cell Expansion Medium(Sigma-Aldrich, S0192)

[ペリサイト増殖培地]
組成は以下の通りである:
・80% MegaCell Dulbecco’s Modified Eagle’s Medium
・20% FBS
・1% GlutaMax
・1% MEM Non-essential Amino Acid Solution(Sigma-Aldrich, M7145)
・1% Penicillin-Streptmycin(Sigma-Aldrich, P0781)
・100μM 2-Mercaptethanol(Thermo Fisher Scientific, 21985023)
・5ng/mL Animal-free Recombinant Human FGF basic-TS
実施例3にて取得したペリサイト様細胞をペリサイト増殖培地存在下で2×105個/ディッシュとなるように10cmコラーゲンコーティングディッシュ(Corning, 356450)5枚に播種し37℃、5% CO2、5% O2雰囲気下で培養した。4日後に上清を除き、PBSで洗浄後、細胞を剥離するために前記ディッシュにAccutaseを2mL添加し、37℃のプレート上で5分間静置した。前記各ディッシュにペリサイト増殖培地を8mL添加して細胞懸濁液を回収し、300g、室温、5分間の条件で遠心分離した。上清を除き、ペリサイト増殖培地を10mL添加した。この細胞懸濁液から1×107個の細胞を別チューブに取り、300g、室温、5分間の条件で遠心分離した。上清を除き、PBSを100μL添加し、さらにFcR Blocking Reagentを20μL添加して氷上で15分間静置した。続けてAnti-CD56 PE antibody(Miltenyi Biotec、130-090-755)を30μL添加し、氷上で20分間静置した。PBSで2回洗浄した後、再度PBSに懸濁し、セルソーターSH800S(ソニー)を用いてCD56(+)ペリサイト様細胞及びCD56(-)ペリサイト様細胞を分取した(図2上)。分取後の各細胞はペリサイト増殖培地に懸濁して10cmコラーゲンコーティングディッシュに播種して培養した。
実施例4で分取した3×105個のCD56(-)ペリサイト様細胞、及び3×105個のCD56(+)ペリサイト様細胞のそれぞれを、ペリサイト増殖培地10mLを用いて10cmコラーゲンコーティングディッシュ上で培養し、3日後に培養上清を回収した。イムノアッセイキット(Human VEGF immunoassay kit, PerkinElmer, AL201C)を用いて、回収した培養上清中のVEGF濃度を測定した。VEGFの濃度の測定はキット添付のプロトコルに従った。その結果、CD56(-)ペリサイト様細胞は、CD56(+)ペリサイト様細胞に比べてVEGFの発現量が有意に高かった(図2下)。統計解析検定として、2群間でt検定を実施した。
実施例4にて分取したCD56(-)ペリサイト様細胞をペリサイト増殖培地中で培養し、複数の細胞表面抗原の発現を、以下のようにAttune NxT Flow Cytometerを用いて調べた。
4.8×105個のCD56(-)ペリサイト様細胞を10cmコラーゲンコーティングディッシュ上で、10mLのペリサイト増殖培地を用いて培養した。数日後にコンフルエントな状態まで増殖が確認された時点で上清を除き、PBSで洗浄後、Accutaseを2mL添加し、37℃プレート上で5分間静置した。前記ディッシュにペリサイト増殖培地を8mL添加して細胞懸濁液を回収し、300g、室温、5分間の条件で遠心分離した。上清を除き、ペリサイト増殖培地を3mL添加し、懸濁液中の細胞数を計測した。その後、1.5×105乃至3.0×105個のCD56(-)ペリサイト様細胞を、新しい10cmコラーゲンコーティングディッシュ上で、10mLのペリサイト増殖培地を用いて培養する手順を繰り返すことで継代培養を行った。コントロールとしてヒト生体由来の初代ペリサイトを用いた。ヒト生体由来の初代ペリサイトは、以下の方法で取得した。即ち、3×104個の初代ペリサイトを60mmコラーゲンコーティングディッシュ上で、5mLのペリサイト増殖培地を用いて培養した。数日後にコンフルエントな状態まで増殖が確認された時点で上清を除き、PBSで洗浄後、Accutaseを1mL添加し、37℃プレート上で5分間静置した。前記ディッシュにペリサイト増殖培地を4mL添加して細胞懸濁液を回収し、300g、室温、5分間の条件で遠心分離した。上清を除き、ペリサイト増殖培地を3mL添加し、懸濁液中の細胞数を計測した。その後、1.5×105乃至5.0×105個の初代ペリサイトを10cmコラーゲンコーティングディッシュ上で、10mLのペリサイト増殖培地を用いて培養する手順を繰り返すことで継代培養を行った。CD56(-)ペリサイト様細胞及び初代ペリサイトについて、継代日数を横軸に、細胞の累計分裂回数を縦軸にプロットした(図4)。CD56(-)ペリサイト様細胞は初代ペリサイトよりも高い増殖能を示した。
ヒト臍帯静脈内皮細胞(Human Umbilical Vein Endothelial Cells;HUVEC, PromoCell, C-12200)を、内皮細胞増殖培地(PromoCell, C-22111)を用いて37℃、5% CO2雰囲気下で培養した。同時に、実施例4と同様の手順で選別したCD56(-)ペリサイト様細胞、及び比較対象としてCD56(+)ペリサイト様細胞を10cmコラーゲンコーティングディッシュ上に、ペリサイト増殖培地を用いて培養した。前記3種類の細胞のそれぞれについて、コンフルエントな状態まで増殖が確認された時点で、実施例7と同じ手順で(但し、HUVECの懸濁には内皮細胞増殖培地を使用した)細胞懸濁液を回収し、300g、室温、5分の条件で遠心分離して上清を除いた後、HUVECは内皮細胞増殖培地に、前記2種類のペリサイト様細胞はペリサイト増殖培地に懸濁して、懸濁液中の細胞数を計測した。1.5mLチューブ9本にHUVECを1本あたり5.5×105個添加した。そのうちの3本には、さらにCD56(-)ペリサイト様細胞を、別の3本にはCD56(+)ペリサイト様細胞を、それぞれ5.5×105個添加して混和した。各チューブを300g、4℃、5分間の条件で遠心分離し、上清を除き、さらにPBSで1回洗浄後、再度同じ条件で遠心分離し、上清を除いた。次にそれぞれのチューブに細胞外マトリクス(Matrigel(登録商標)Growth factor reduced, Corning, 356231、以下「マトリゲル」と呼ぶ)を400μL加え、氷上で混和後に、細胞を含むマトリゲルを25ゲージ針付きシリンジで吸引した。
実施例8の手順にて回収した各マトリゲルを2mL チューブ(Eppendorf, 0030120.094)に入れ、解剖バサミで数回切断した。そこにステンレススチールビーズ(Qiagen, 69989)を1個添加し、さらに水で希釈した0.1% Brij(登録商標)L23溶液(Sigma-Aldrich, B4184)を350μL添加した。TissueLyzer II(Qiagen, 85300)を用いてマトリゲルを破砕し、10,000g、4℃、5分間の条件で遠心分離後、上清を回収した。QuantiChrom Hemoglobin Assay Kit(BioAssay Systems, DIHB-250)を用いて、回収した上清のヘモグロビン濃度を測定した。測定方法は製品のプロトコルに従った。統計解析検定として、HUVECとCD56(+)ペリサイト様細胞(CD56(+)/HUVEC)、及びHUVECとCD56(-)ペリサイト様細胞(CD56(-)/HUVEC)の2群間でt検定を実施した。その結果、CD56(-)ペリサイト様細胞を含むマトリゲルは、CD56(+)ペリサイト様細胞を含むマトリゲルよりも有意に高いヘモグロビン濃度を示した(図6)。
Claims (26)
- 多能性幹細胞から分化誘導されたペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、VEGF高発現ペリサイト様細胞。
- (a)多能性幹細胞を初期中胚葉細胞に分化させる工程、
(b)前記工程(a)で得られた初期中胚葉細胞をペリサイト様細胞に分化させる工程、及び
(c)前記ペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、請求項1に記載のVEGF高発現ペリサイト様細胞。 - (a)多能性幹細胞を初期中胚葉細胞に分化させる工程、
(b-1)前記工程(a)で得られた初期中胚葉細胞のスフェロイドを形成させる工程、
(b-2)前記スフェロイドを形成した初期中胚葉細胞をペリサイト様細胞に分化させる工程、及び
(c)前記ペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、請求項1に記載のVEGF高発現ペリサイト様細胞。 - (a)多能性幹細胞を初期中胚葉細胞に分化させる工程、
(b-1’)培養面にすり鉢状の微細な複数のウェルを隙間なく備える培養容器内で、前記工程(a)で得られた初期中胚葉細胞のスフェロイドを形成させる工程、
(b-2)前記スフェロイドを形成した初期中胚葉細胞をペリサイト様細胞に分化させる工程、及び
(c)前記ペリサイト様細胞を含む集団からCD56(-)のペリサイト様細胞を選別する工程を含む製造方法で得られる、請求項1に記載のVEGF高発現ペリサイト様細胞。 - 前記多能性幹細胞がヒト多能性幹細胞である、請求項1~4のいずれか1項に記載のVEGF高発現ペリサイト様細胞。
- 前記多能性幹細胞がES細胞又はiPS細胞である、請求項1~5のいずれか1項に記載のVEGF高発現ペリサイト様細胞。
- 多能性幹細胞から分化誘導されたペリサイト様細胞を含む集団からCD56(-)の細胞表面マーカーを用いて選別された、VEGF高発現ペリサイト様細胞の集団。
- (a)多能性幹細胞を初期中胚葉細胞に分化させる工程、及び
(b)前記工程(a)で得られた初期中胚葉細胞をペリサイトに分化させる工程、を含む方法で分化誘導されたペリサイト様細胞を含む集団から、CD56(-)の細胞表面マーカーを用いて選別された、請求項7に記載のVEGF高発現ペリサイト様細胞の集団。 - (a)多能性幹細胞を初期中胚葉細胞に分化させる工程、
(b-1)前記工程(a)で得られた初期中胚葉細胞のスフェロイドを形成させる工程、及び
(b-2)前記スフェロイドを形成した初期中胚葉細胞をペリサイト様細胞に分化させる工程、を含む方法で分化誘導されたペリサイト様細胞を含む集団から、CD56(-)の細胞表面マーカーを用いて選別された、請求項7に記載のVEGF高発現ペリサイト様細胞の集団。 - (a)多能性幹細胞を初期中胚葉細胞に分化させる工程、
(b-1’)培養面にすり鉢状の微細な複数のウェルを隙間なく備える培養容器内で、前記工程(a)で得られた初期中胚葉細胞のスフェロイドを形成させる工程、及び
(b-2)前記スフェロイドを形成した初期中胚葉細胞をペリサイト様細胞に分化させる工程、を含む方法で分化誘導されたペリサイト様細胞を含む集団から、CD56(-)の細胞表面マーカーを用いて選別された、請求項7に記載のVEGF高発現ペリサイト様細胞の集団。 - 前記多能性幹細胞がヒト多能性幹細胞である、請求項7~10のいずれか1項に記載のVEGF高発現ペリサイト様細胞の集団。
- 前記多能性幹細胞がES細胞又はiPS細胞である、請求項7~11のいずれか1項に記載のVEGF高発現ペリサイト様細胞の集団。
- 請求項1~6のいずれか1項に記載のVEGF高発現ペリサイト様細胞を含む、血管新生療法のための医薬組成物。
- 血管新生療法が重症下肢虚血の治療である、請求項13に記載の医薬組成物。
- 血管内皮細胞と併用される、請求項13又は請求項14に記載の医薬組成物。
- 請求項1~6のいずれか1項に記載のVEGF高発現ペリサイト様細胞及び血管内皮細胞を組み合わせてなる、血管新生療法のための医薬組成物。
- 血管新生療法が重症下肢虚血の治療である、請求項16に記載の医薬組成物。
- 血管新生療法に使用するための、請求項1~6のいずれか1項に記載のVEGF高発現ペリサイト様細胞。
- 重症下肢虚血の治療に使用するための、請求項1~6のいずれか1項に記載のVEGF高発現ペリサイト様細胞。
- 請求項7~12のいずれか1項に記載のVEGF高発現ペリサイト様細胞の集団を含む、血管新生療法のための医薬組成物。
- 血管新生療法が重症下肢虚血の治療である、請求項20に記載の医薬組成物。
- 血管内皮細胞と併用される、請求項20又は21に記載の医薬組成物。
- 請求項7~12のいずれか1項に記載のVEGF高発現ペリサイト様細胞の集団及び血管内皮細胞を組み合わせてなる、血管新生療法のための医薬組成物。
- 血管新生療法が重症下肢虚血の治療である、請求項23に記載の医薬組成物。
- 血管新生療法に使用するための、請求項7~12のいずれか1項に記載のVEGF高発現ペリサイト様細胞の集団。
