Abstract
본 발명은 초임계 유체 추출 공정을 이용하여 제조된 탈세포화된 무세포 진피조직에 관한 것이다. 보다 상세하게는, 초임계 유체 추출 공정을 이용하여 세포외기질 보존 정도 개선 및 임계하중, 탄성 복원력 등 최적화된 기계적 물성을 갖는 무세포 진피조직에 관한 것이다. 본 발명에 따른 무세포 진피조직은 계면활성제 처리 없이 탈세포화되어 기계적 물성이 유지될 뿐 아니라, 잔존 계면활성제에 따른 독성이 존재하지 않고, 이식거부반응을 최소화하여 피부조직 손상 환자 치료에 유용하게 활용될 수 있다.
Description
본 발명은 초임계 유체 추출 공정을 이용하여 제조된 무세포 진피조직에 관한 것이다. 보다 상세하게는, 초임계 유체 추출 공정을 이용하여 세포외기질 보존 정도 개선 및 임계하중, 탄성 복원력 등 최적화된 기계적 물성을 갖는 무세포 진피조직에 관한 것이다.
인체의 피부조직은 가장 바깥쪽을 이루는 표피층과 그 아래층의 진피층, 그리고 피하조직으로 이루어진다. 이 중 표피층은 표피층과 진피층이 단단하게 결합할 수 있도록 하는 기저막으로부터 여러 층으로 분화된 상피세포와 그 밖에 멜라닌세포, 면역세포로 이루어진다. 표피층 아래의 진피층은 주로 섬유아세포와 콜라겐 및 엘라스틴 등으로 구성되는 세포외기질(Extracellular matrix)로 이루어진다.
피부조직 또는 내부 장기조직은 화상, 외상, 궤양 등으로 인해 조직의 일부가 손상될 수 있는데, 이 경우 손상된 조직의 치유를 목적으로 하거나 또는 재건성형을 목적으로 본인 피부조직 또는 내부 장기조직으로부터 이식하는 방법을 사용한다. 이러한 경우에는 이식받는 자가 자신의 피부조직 혹은 장기조직을 추가적으로 적출하는 수술적 부담이 있어 이식받는 사람의 건강상태가 양호하지 않은 경우에는 위험할 수가 있다. 그 외에 이종조직이나 합성 생체재료(Synthetic biomaterial)를 사용하여 이식하는 방법이 있는데, 이 경우에는 면역거부 반응이 발생하여 장기간 이식시 염증반응을 초래하여 재수술하는 결과를 초래할 수도 있다.
상기 문제점을 해결하기 위해, 공여자로부터 적출된 피부조직으로부터 이식용 무세포 진피층을 제조하는 방법이 한국등록특허 제10-0469661호 및 제10-0791502호에 개시되어 있으나, 이러한 방법은 계면활성제 처리 공정을 따르고 있다. 계면활성제 처리에 의해 콜라겐과 같은 단백질의 변성, 성장인자의 파괴 등의 문제가 야기될 수 있다. 또한, 구조적 형태, 조직학적 형태의 유지가 어렵고, 임계하중, 탄력도와 같은 기계적 물성이 약화될 수 있다. 더욱이, 잔존 계면활성제의 제거가 쉽지 않고, 이의 제거가 완전히 되지 않는 경우 독성이 있을 수 있다.
또한, 무세포 진피를 제조하기 위해 면역원성 세포 제거 방법으로 트립신 또는 디스파제 같은 단백질 분해 효소를 사용하거나, 반복적인 동결-해동, NaCl과 SDS 처리 방법 등이 사용되어 왔다. 그러나, 이러한 공정으로 만들어진 무세포 진피는 많은 항원 성분을 지니고 있어 수여부에 이식시 면역거부반응을 일으켜 결과적으로 낮은 생착률을 초래하였다(R.J. Walter et.al, Burns, 24: 104-113, 1998).
이에 본 발명자들은 이식거부반응 및 비용적 측면의 문제점을 개선함과 동시에 화학적 처리 방법에 의한 탈세포화로 인해 야기된 기계적 물성 약화 문제점이 개선된 무세포 진피 이식소재를 개발하고자 연구하였다. 그 결과, 계면활성제 처리 없이 최적화된 초임계 유체 처리 공정을 구축하고, 이에 따른 기계적 물성 및 세포외기질 보존 정도가 개선된 무세포 진피조직을 수득함으로써 본 발명을 완성하였다.
상기 목적 달성을 위해, 본 발명의 일 측면은, 임계하중(Critical load)이 11 내지 19 N이고, 영률(Young's modulus)은 0.5 내지 1.2 MPa인, 무세포 진피조직 이식체를 제공한다.
본 발명의 다른 측면은, a) 개체로부터 분리된 피부조직을 표피층 및 진피층으로 분리하는 단계; b) 상기 분리된 진피층을 초임계 유체로 추출하는 단계; 및 c) 초임계 유체로 추출된 진피층을 인산염 완충액으로 세척하는 단계;를 포함하는, 무세포 진피조직 이식체의 제조 방법을 제공한다.
본 발명에 따른 무세포 진피조직은 계면활성제 처리 없이 탈세포화되어 임계하중, 탄력도 등과 같은 기계적 물성이 유지되면서, 동시에 잔존 계면활성제에 따른 독성이 존재하지 않으므로, 화상, 외상 등의 피부조직 손상 환자에게 이식하기에 적합하다. 따라서, 최적화된 초임계 유체 처리 공정을 통해 수득한 본 발명의 무세포 진피조직은 독성 및 이식거부반응이 제거되어 피부조직 손상 환자 치료에 유용하게 활용될 수 있다.
도 1은 본 발명의 일 실시예에 따른 무세포 진피 시료의 외형을 관찰한 사진이다. 여기에서, RTU(Ready-To-Use) 타입은 일반 식염수를 포함하여 진피 시료를 이중 포장 후 멸균하여 준비한 것이다. FD(Feeze-Drying) 타입은 진피 시료를 동결건조 후 이중 포장하여 멸균하여 준비한 것이다. 또한, Native는 표피를 제거하지 않은 미가공 천연 피부조직 시료이며, SCR, SCF, A, B, C, D, E, F는 하기 표 1에 나타낸 바와 같다. 진피 시료 크기는 1×1 cm2이다.

도 3은 본 발명의 일 실시예에 따른 초임계 유체 추출 공정 미실시 진피조직(Native), 초임계 유체 추출 공정 후 탈세포화된 진피조직(SCR, SCF) 및 표 1의 세부사양에 따른 종래 공지된 방법에 따라 제조된 무세포 진피조직(A, B, C, D, E, F)의 DNA 잔존량을 확인한 1% 아가로스 젤 전기영동 이미지이다. 여기서, SCR, A, B, C는 RTU(Ready-To-Use) 타입의 시료이고, SCF, D, E, F는 FD(Feeze-Drying) 타입의 시료이다.
도 4a는 본 발명의 일 실시예에 따른 초임계 유체 추출 공정 후 수득한 무세포 진피 시료의 잔존 DNA 정량 분석 결과를 나타낸 도면이다. 이때, 잔존 DNA 정량분석 결과는 무세포 진피조직의 건조 중량을 기준으로 나타낸 것이다. 여기서, Native는 무처리군으로서 원조직을 지시하고, SCR, SCF는 초임계 유체를 처리하여 탈세포한 진피조직 시험군, A, B, C, D, E, F는 표 1의 세부사양에 따른 탈세포한 진피조직 양성 대조군이다.

무세포 진피조직 이식체
본 발명의 일 측면은, 하기 특징을 갖는 무세포 진피조직 이식체를 제공한다:
상기 무세포 진피조직의 임계하중은 11 내지 19 N이고, 영률(Young's modulus)은 0.5 내지 1.2 MPa이다.
본 명세서에서 사용된 용어, "진피(dermis)"는 콜라겐 단백질로 만들어진 교원섬유가 주요 구성 성분이고, 그 사이에 엘라스틴 단백질로 이루어진 탄성섬유가 그물 모양으로 짜여 있다. 진피는 피부의 대부분을 차지하며, 표피에 영양분을 공급하여 표피를 지지하고, 외부의 손상으로부터 몸을 보호한다. 또한 수분을 저장하는 능력과 체온 조절의 기능이 있으며, 감각에 대한 수용체 역할을 하고, 표피와 상호작용에 의해 피부를 재생하는 기능도 한다.
본 명세서에서 사용된 용어, "무세포 진피(Acellular dermal matrix, ADM)"는 인간 또는 동물 피부로부터 무세포화 기술을 통해 얻어지는 진피층 기질로서, 콜라겐 및 엘라스틴 등으로 구성되는 세포외기질(Extracellular Matrix, ECM) 형태의 생체유래 피부대체재를 의미한다.
상기 무세포 진피는 개체로부터 분리된 피부에서 면역반응을 일으킬 수 있는 세포들을 제거한 생체소재로서, 화상, 교통사고, 궤양 등으로 인한 피부결손 환자에게 이식해 피부를 복원시키는 데 이용될 수 있다. 또한, 인체이식용 피부조직인 무세포 진피는 전층 피부, 비중격, 뇌척수 경막결손창의 재건뿐만 아니라 함몰 반흔 교정, 반안면 위축 교정, 유두 재건, 입술 확대의 재건성형 및 미용성형에도 확대되어 이용될 수 있다.
상기 무세포 진피는 피부조직 내 진피층의 3차원적 구조를 손상시키지 않고, 다양한 구조 단백질 및 성분들을 유지한 상태에서 면역반응의 대상인 세포성 항원만을 선별적으로 제거되어야 한다.
또한, 무세포 진피는 손상 조직 또는 손상 피부에 적용되어 손상 부위를 복원시키는 동안, 적용된 시술 부위의 움직임에도 안정적으로 유지될 수 있도록 적절한 신축성과 임계하중을 가져야 한다. 무세포 진피는 시술 부위 주변 정상 조직이 손상되지 않는 재료이어야 하고 시술 용이성을 가져야 한다.
본 명세서에서 상기 무세포 진피는 무세포 진피조직 이식체, 무세포 진피조직, 탈세포화된 진피, 탈세포화된 진피조직 또는 무세포 진피조직으로 혼용하여 사용될 수 있다.
본 명세서에서 사용된 용어, "탈세포화(decellularization)"는 목적으로 하는 이식 조직 또는 기관의 원래의 구조를 유지하면서 전체 장기로부터 세포를 제거하여 인공 지지체를 생산하는 새로운 방법이다. 탈세포화 과정에서, 조직으로부터 세포 성분들이 제거되지만, 세포외기질 및 일부 성장인자 단백질들은 보존된다. 따라서, 탈세포화 조직 내 보존된 콜라겐, 피브로넥틴(fibronectin) 및 엘라스틴을 포함하는 다양한 세포외기질 성분들은 온전한 조직 내와 유사한 삼차원적인 미세환경을 제공함으로써 배양된 세포의 생존, 증식 및 분화를 향상시킬 수 있다.
본 발명에 있어서, 탈세포화는 이에 제한되지는 않으나, 계면활성제 처리 없이, 초임계 유체 추출에 의해 수행될 수 있다.
본 명세서에서 사용된 용어, "초임계 유체 추출(supercritical fluid extraction)" 또는 "초임계 추출(supercritical extraction)"은 임계점 즉, 임계 온도 및 임계 압력 이상에 존재하는 기체와 액체의 중간 성격을 갖는 초임계 유체를 사용하여 물질을 분리하는 방법이다. 상기 초임계 유체 추출은 추출 원료와 초임계 유체의 용해도 차이에 의해 원료 중에 함유된 가용 성분이 초임계 유체로 용해되는 용매 추출 원리와 원료 중에 함유된 용질 분자가 고밀도 응축상으로부터 저밀도 팽창상인 초임계 유체로 이행하는 증발 현상인 증류 원리를 복합적으로 이용한 것이다.
본 명세서에서 사용된 용어, "초임계 유체"는 일반적인 조건에서는 기체 상태이나 임계 온도와 임계 압력 이상에서는 유체인 것을 말한다. 본 발명에서 사용하기에 적합한 초임계 유체로는 특별히 한정되지 않으나, 예를 들어, 이산화탄소, 질소, 아산화질소, 메탄, 에틸렌, 프로판 및 프로필렌이 있다. 바람직하게는 임계온도가 31℃이고 임계 압력이 72.8기압인 이산화탄소를 사용할 수 있다.
본 발명에 있어서, 탈세포화는 초임계 유체 추출 시 초임계 유체 이외에 '공용매(co-solvent)'를 첨가할 수 있다. 상기 공용매는 초임계 유체의 추출능 증가 및 용해도 향상 등의 목적으로 첨가될 수 있으며, 이에 제한되지는 않으나, 에탄올, 메탄올, 석유 에테르, 아세토니트릴, 헥산 등을 공용매로서 사용할 수 있다. 이때, 바람직하게는 공용매는 에탄올 일 수 있다.
본 발명의 탈세포화된 진피조직은 개체로부터 분리된 피부조직으로부터 유래할 수 있다. 상기 개체는 상기 진피조직을 이식하려는 대상인 개체와 동종 또는 이종에 속하는 개체일 수 있고, 구체적으로 인간, 마우스, 래트, 쥐, 원숭이, 침팬지, 오랑우탄, 말, 소, 돼지, 고양이, 개, 및 토끼 등을 포함하는 포유류일 수 있으나, 이에 제한되지 않는다.
본 발명의 무세포 진피조직은 상기 개체로부터 분리된 원조직과 비교, 즉 탈세포화를 수행하지 않은 조직과 비교하여 세포가 90% 이상, 91% 이상, 92% 이상, 93% 이상, 94% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 99% 이상 또는 100% 제거된 것일 수 있다.
본 명세서에서 사용된 용어, "임계하중(Critical load)"은 구조물이나 부재가 파괴 또는 과도한 변형과 같은 상태가 되는 한계 하중을 의미한다. 즉, 인덴터를 통해 하중을 가하여 이루어지는 압입 평가시, 시료편의 변형이나 균열이 발생하는 시작점의 하중을 의미한다. 임계하중에 의해서 시료편의 변형이나 균열이 발생할 때까지의 최대응력으로, 파단(rupture)까지의 최대하중을 시험편의 원래의 단면적으로 나눈 값을 의미한다. 임계하중이 클수록 지지체의 파단까지 큰 힘이 필요함을 의미하며, 이는 진피조직이 손상 피부 또는 손상 조직에 적용될 때는 물론 유통 및 보관 중에도 형태 유지가 잘 될 수 있음을 아울러 의미할 수 있다.
본 발명에서 측정하는 임계하중은 만능 인장 시험기(Universal Tensile Machine)를 사용하여 측정될 수 있다. 또한, 본 발명에서 무세포 진피조직의 임계하중은 멸균 증류수에 젖어 있는 상태, 즉 습윤 상태의 조건하에서 측정되는 습윤 임계하중일 수 있으나, 이에 한정되지는 않는다.
본 발명에 있어서, 상기 탈세포화된 진피조직은 습윤 상태의 조건하에서, 진피조직 시료의 크기가 1×1 cm2를 기준으로 11 내지 19 N의 임계하중을 가질 수 있다.
일 구체예에 있어서, 동결건조를 실시하지 않은 RTU(Ready-To-Use) 타입의 진피조직 시료는 11.0 N, 11.1 N, 11.2 N, 11.3 N, 11.4 N, 11.5 N, 11.6 N, 11.7 N, 11.8 N, 11.9 N, 또는 12.0 N의 임계하중을 가질 수 있으나, 이에 제한되지는 않는다. 이때, 임계하중 측정은 진피조직 시료의 크기 1×1 cm2를 기준으로 한다.
또한, 일 구체예에 있어서, 동결건조를 실시한 FD(Freeze-Drying) 타입의 진피조직 시료는 16.0 내지 19.0 N, 16.5 내지 19.0 N, 17.0 내지 19.0 N, 17.5 내지 19.0 N, 18.0 내지 19.0 N, 18.1 N, 18.2 N, 18.3 N, 18.4 N, 18.5 N, 18.6 N, 18.7 N, 18.8 N의 임계하중을 가질 수 있으나, 이에 제한되지는 않는다. 이때, 임계하중 측정은 진피조직 시료의 크기 1×1 cm2를 기준으로 한다.
본 명세서에서 사용된 용어, "영률(Young's modulus)"은 '탄성률' 또는 '탄성계수'로도 일컬어지며, 물체의 변형에 대한 물체의 압력(응력)의 비율을 의미한다. 영률은 압력을 가할 때, 물체의 변형되는 정도를 나타내는 탄성률로, 영률 측정의 기본 원리는 물체가 압축되거나 확장될 때 탄성 변형을 겪고 하중이 제거되면 원래 모양으로 되돌아가는 성질을 이용한 것이다. 일례로, 뻣뻣한 물체에 비해 유연한 물체에서 더 많은 변형이 발생하게 되며, 영률 값이 높음은 비탄성이거나 뻣뻣함을 의미한다.
본 발명에서 측정하는 영률은 만능 인장 시험기(Universal Tensile Machine)를 사용하여 측정될 수 있다. 또한, 본 발명에서 진피조직의 영률은 이에 제한되지는 않으나, 습윤 조건하에서 측정될 수 있다.
본 발명에 있어서, 상기 탈세포화된 진피조직은 습윤 상태의 조건하에서, 진피조직 시료의 크기가 1×1 cm2를 기준으로 0.5 내지 1.2 MPa의 영률을 가질 수 있다.
일 구체예에 있어서, 동결건조를 실시하지 않은 RTU(Ready-To-Use) 타입의 진피조직 시료는 0.50 MPa, 0.51 MPa, 0.52 MPa, 0.53 MPa, 0.54 MPa, 0.55 MPa, 0.56 MPa, 0.57 MPa, 0.58 MPa, 0.59 MPa, 또는 0.60 MPa의 영률을 가질 수 있으나, 이에 제한되지는 않는다. 이때, 영률 측정은 진피조직 시료의 크기 1×1 cm2를 기준으로 한다.
또한, 일 구체예에 있어서, 동결건조를 실시한 FD(Freeze-Drying) 타입의 진피조직 시료는 1.00 MPa, 1.01 MPa, 1.02 MPa, 1.03 MPa, 1.04 MPa, 1.05 MPa, 1.06 MPa, 1.07 MPa, 1.08 MPa, 1.09 MPa, 또는 1.10 MPa의 영률을 가질 수 있으나, 이에 제한되지는 않는다. 이때, 영률 측정은 진피조직 시료의 크기 1×1 cm2를 기준으로 한다.
본 발명에 따른 무세포 진피조직은 이에 제한되지는 않으나, 건조중량을 기준으로 하기 그룹에서 선택되는 어느 하나의 특징을 가질 수 있다:
DNA 잔존량이 80 내지 110 ng/mg;
콜라겐 함량이 400 내지 700 μg/mg; 및
엘라스틴 함량이 13 내지 19 μg/mg.
본 발명에 있어서, 상기 진피조직은 건조중량을 기준으로 DNA 잔존량이 110 ng/mg 이하 포함된 것일 수 있다. 바람직하게는, 80 내지 110 ng/mg, 81 내지 109 ng/mg, 82 내지 108 ng/mg, 83 내지 107 ng/mg, 84 내지 106 ng/mg, 85 내지 105 ng/mg, 86 내지 104 ng/mg, 또는 87 내지 103 ng/mg 포함된 것일 수 있으나, 이에 제한되지는 않는다.
본 발명에 있어서, 상기 진피조직은 건조중량을 기준으로 콜라겐 함량이 400 μg/mg 이상 포함된 것일 수 있다. 바람직하게는 400 내지 700 μg/mg, 402 내지 698 μg/mg, 404 내지 696 μg/mg, 406 내지 694 μg/mg, 408 내지 692 μg/mg, 410 내지 690 μg/mg, 411 내지 688 μg/mg, 412 내지 686 μg/mg, 413 내지 684 μg/mg, 414 내지 682 μg/mg, 415 내지 680 μg/mg, 416 내지 679 μg/mg, 417 내지 678 μg/mg, 또는 418 내지 677 μg/mg 포함된 것일 수 있으나, 이에 제한되지는 않는다.
본 발명에 있어서, 상기 진피조직은 건조중량을 기준으로 엘라스틴 함량이 13 μg/mg 이상 포함된 것일 수 있다. 바람직하게는 13.0 내지 19.0 μg/mg, 13.1 내지 18.9 μg/mg, 13.2 내지 18.8 μg/mg, 13.3 내지 18.7 μg/mg, 13.4 내지 18.6 μg/mg, 13.5 내지 18.5 μg/mg, 13.6 내지 18.4 μg/mg, 13.7 내지 18.3 μg/mg, 13.8 내지 18.2 μg/mg, 또는 13.9 내지 18.1 μg/mg 포함된 것일 수 있으나, 이에 제한되지는 않는다.
본 발명에 따른 무세포 진피조직은 이에 제한되지는 않으나, 원조직 대비 하기 그룹에서 선택되는 어느 하나의 특징을 가질 수 있다:
DNA 잔존량이 5 내지 8%;
콜라겐 함량이 55 내지 99%;
엘라스틴 함량이 65 내지 95%;
임계하중은 90 내지 165%; 및
영률(Young's modulus)은 80 내지 200%.
본 발명에 있어서, 상기 진피조직은 원조직 대비, 즉 탈세포화 전 또는 탈세포화를 수행하지 않은 진피조직 대비 DNA 잔존량이 8% 이하 포함된 것일 수 있다. 바람직하게는, 5 내지 8%, 5.1 내지 7.8%, 5.2 내지 7.6%, 5.3 내지 7.6%, 5.4 내지 7.4%, 5.5 내지 7.4%, 5.6 내지 7.2%, 5.7 내지 7.0%, 또는 5.8 내지 6.8% 포함된 것일 수 있으나, 이에 제한되지는 않는다.
즉, 상기 진피조직은 원조직 대비 세포가 93% 이상 제거된 것일 수 있다. 바람직하게는, 93 내지 98%, 94 내지 98%, 95 내지 98%, 96 내지 98%, 또는 97 내지 98% 제거된 것일 수 있으나, 이에 제한되지는 않는다.
본 발명에 따른 진피조직은 세포외기질 성분, 예를 들어 콜라겐, 엘라스틴 등이 원조직 대비 소정의 함량으로 유지되어 있는 것일 수 있다.
구체적으로, 본 발명에 있어서, 상기 진피조직은 원조직 대비, 콜라겐 함량이 55% 이상 포함된 것일 수 있다. 바람직하게는 55 내지 99%, 56 내지 98%, 57 내지 97%, 또는 58 내지 96% 포함된 것일 수 있으나, 이에 제한되지는 않는다.
본 발명에 있어서, 상기 진피조직은 원조직 대비, 엘라스틴 함량이 65% 이상 포함된 것일 수 있다. 바람직하게는 65 내지 95%, 66 내지 94%, 67 내지 93%, 68 내지 92%, 69 내지 91%, 또는 70 내지 90% 포함된 것일 수 있으나, 이에 제한되지는 않는다.
본 발명에 있어서, 상기 진피조직은 원조직 대비, 임계하중이 90 내지 165% 수준을 나타내는 것일 수 있으나, 이에 제한되지는 않는다.
본 발명에 있어서, 상기 진피조직은 원조직 대비, 영률(Young's modulus)이 80 내지 200% 수준을 나타내는 것일 수 있으나, 이에 제한되지는 않는다.
본 발명에 따른 무세포 진피조직은 이에 제한되지는 않으나, 원조직 대비 하기 그룹에서 선택되는 어느 하나의 특징을 가질 수 있다:

본 발명의 예시적인 구현예들에서, 진피조직은 계면활성제를 포함하지 않는다. 종래 계면활성제 처리에 의해 탈세포화된 진피조직의 경우, 계면활성제의 처리에 따른 콜라겐과 같은 단백질의 변성과 성장인자의 파괴 등의 문제가 있었다. 또한, 잔존 계면활성제의 제거가 쉽지 않았으며, 이의 제거가 완전히 되지 않는 경우 독성이 있을 수 있는 문제가 있었다.
아울러, 계면활성제로 인해 진피조직의 임계하중, 탄력도와 같은 기계적 특성이 약화되고, 계면활성제가 진피조직에 잔존하는 경우 독성물질로 인해 이식거부반응을 초래하는 어려움이 있었다.
그러나, 본 발명에 따른 초임계 유체 공정에 따라 수득한 탈세포화된 진피조직은 종래 계면활성제를 처리하여 탈세포화된 진피조직과 동등한 수준으로 탈세포화가 이루어지고, 조직학적 형태 및 기계적 물성이 유지되는 특성을 지닌다. 특히, 초임계 유체 공정에 따른 무세포 진피조직은 계면활성제를 처리한 진피조직에 비해 세포외기질(ECM)인 콜라겐, 엘라스틴 등의 진피조직 내 보존 정도가 현저하게 우수한 이점을 지니고 있다.
따라서, 본 발명에 따른 무세포 진피조직은 조직학적 형태 및 기계적 특성을 유지하면서, 세포외기질 손실 없이 탈세포화되어 이식재로서 활용 가능하다.
무세포 진피조직 이식체의 제조
본 발명에 따른 무세포 진피조직 이식체는, 개체로부터 분리된 진피조직을 초임계 유체로 추출하는 단계를 포함하는 제조 방법에 의해 제조될 수 있다.
상기 "탈세포화", "개체", "진피조직" 및 "초임계 유체"는 상술한 바와 같다.
본 발명에 있어서, 초임계 유체는 용해도를 바탕으로 개체로부터 분리된 진피조직 내 지질 성분, 구체적으로 세포막의 주성분인 인지질 성분을 추출하여 탈세포화시킴으로써 탈세포화된 진피조직을 제조할 수 있다.
상기 초임계 유체는 이산화탄소 가스, 암모니아 가스, 질소 가스, 일산화질소(NO) 가스, 이산화질소(NO2) 가스, 아산화질소(N2O) 가스, 이산화황 가스, 수소 가스, 수증기, 포화탄화수소, 불포화탄화수소, 방향족 화합물 및 이들의 혼합 가스로 이루어진 군에서 선택된 것일 수 있다. 구체적으로, 이산화탄소 가스일 수 있으나, 진피조직의 기계적 특성뿐만 아니라 구조적 형태를 유지하면서, 진피조직의 세포를 대부분 제거하여 효율적으로 진피조직을 제조할 수 있는 초임계 유체라면 그 종류는 이에 제한되지 않는다. 상기 초임계 유체로 이산화탄소 가스를 이용 시, 이산화탄소는 낮은 임계 온도(31℃)와 임계 압력(73 bar)을 가지고 있어 쉽게 초임계 조건으로 조정이 가능하며, 자연계에 널리 존재하고 무색, 무취하며 인체에 무해하고 화학적으로 안정한 장점이 있다.
본 발명에 있어서, 상기 초임계 추출 단계는 0 내지 1000 bar, 30 내지 900 bar, 60 내지 800 bar, 90 내지 700 bar, 120 내지 600 bar, 150 내지 500 bar, 또는 200 내지 400 bar의 압력 조건 하에서 수행될 수 있다.
구체적으로, 초임계 추출 단계의 압력은 이에 제한되지는 않으나, 0 bar 이상, 50 bar 이상, 100 bar 이상, 150 bar 이상, 200 bar 이상, 250 bar 이상, 300 bar 이상, 350 bar 이상, 400 bar 이상, 450 bar 이상, 500 bar 이상, 550 bar 이상, 600 bar 이상, 650 bar 이상, 700 bar 이상, 750 bar 이상, 800 bar 이상, 850 bar 이상, 900 bar 이상 또는 950 bar 이상일 수 있다.
또한, 초임계 추출 단계의 압력은 이에 제한되지는 않으나, 1000 bar 이하, 950 bar 이하, 900 bar 이하, 850 bar 이하, 800 bar 이하, 750 bar 이하, 700 bar 이하, 650 bar 이하, 600 bar 이하, 550 bar 이하, 500 bar 이하, 450 bar 이하, 400 bar 이하, 350 bar 이하, 300 bar 이하, 250 bar 이하, 200 bar 이하, 150 bar 이하, 100 bar 이하 또는 50 bar 이하일 수 있다.
상기 초임계 추출 단계의 압력 조건은, 진피조직의 기계적 특성 및 조직의 구조적 형태를 보존하면서, 진피조직의 세포를 대부분 제거하여 효율적으로 진피조직을 제조할 수 있는 조건이라면 그 범위는 이에 제한되지 않는다.
상기 초임계 추출 단계에 있어서, 초임계 유체 이외에 공용매를 더 포함할 수 있다. 상기 공용매는 에탄올, 물, 메탄올, 헥산, 석유 에테르, 아세토니트릴, 아세톤, 에틸아세테이트 및 메틸렌클로라이드로 이루어진 군으로부터 선택된 하나 이상의 용매일 수 있다. 바람직하게는, 공용매로서 에탄올을 더 포함할 수 있다.
상기 공용매는 초임계 유체의 추출능 증가 및 용해도 향상 등의 목적으로 첨가되어 분리된 진피조직 내의 지질, 구체적으로 세포막의 인지질을 추출하여 진피조직의 세포를 대부분을 제거하되, 진피조직의 기계적 특성 및 조직의 구조적 형태를 보존하는 한, 그 종류는 특별히 제한되지 않는다.
본 발명에 있어서, 상기 초임계 추출 단계는 이에 제한되지는 않으나, 31℃ 내지 40℃, 31℃ 내지 39℃, 32℃ 내지 38℃, 33℃ 내지 37℃, 34℃ 내지 36℃, 또는 35℃의 온도 조건하에서 수행될 수 있다.
본 발명에 있어서, 상기 초임계 추출 단계는 이에 제한되지는 않으나, 3시간 이내 동안 수행될 수 있다. 바람직하게는, 60분 내지 180분, 70분 내지 170분, 80분 내지 160분, 90분 내지 150분, 100분 내지 140분, 110분 내지 130분, 또는 120분 동안 수행될 수 있으나, 이에 제한되지는 않는다.
본 발명에 따른 무세포 진피조직은, 이에 제한되지는 않으나, 상기 초임계 유체로 추출하는 단계 전에 표피층과 진피층을 분리하는 단계를 포함하여 제조될 수 있다.
상기 표피층과 진피층의 분리는 당업계에 공지된 방법을 통해 수행될 수 있다. 일반적으로, 표피층과 진피층의 분리는 여러가지 단백질 분해 효소, 예컨대, 디스파아제, 터몰리신, 트립신 등을 사용하여 수행될 수 있다.
또한, 용액의 이온 강도를 변화시켜 표피층과 진피층을 분리할 수 있다. 구체적으로, 1 M 이상의 염화나트륨(NaCl) 용액 또는 20 mM의 EDTA 용액으로 37℃에서 14시간 내지 32시간 동안 처리하면 표피층과 진피층을 분리할 수 있다.
일 구체예에 있어서, 상기 표피층 및 진피층의 분리는 이에 제한되지는 않으나, 37℃의 온도 조건하에서, 1M NaCl을 24시간 동안 처리하여 수행될 수 있다.
본 발명에 따른 무세포 진피조직은, 이에 제한되지는 않으나, 상기 초임계 유체로 추출하는 단계 후에 하기 단계 중 어느 하나의 단계를 더 포함하여 제조될 수 있다:
인산염 완충액으로 진피조직을 세척하는 단계;
진피조직을 동결건조하는 단계; 및
진피조직을 멸균하는 단계.
상기 인산염 완충액을 이용한 세척을 통해 초임계 유체 추출 이후 진피조직에 존재하는 잔존 용액 및 불순물을 세척할 수 있다.
본 발명의 일 실시예에 있어서, 초임계 유체 추출 후에 PBS(phosphate bufferd saline)로 탈세포화된 진피조직을 16시간 동안 상온에서 세척하여 잔존 용액 및 불순물을 제거하였다.
상기 초임계 유체 추출 이후 수득된 진피조직은 동결건조를 통해 사용시까지 보존될 수 있다.
상기 동결건조시, 이에 제한되지는 않으나, 동결보호제가 추가로 포함될 수 있다. 동결보호제는 진피층 조직의 구조 변화 방지를 비롯하여, 진피층 조직의 물리, 화학적 손상을 막을 수 있다.
상기 동결보호제는 당업계에 공지된 동결보호제를 채택할 수 있다. 예시적인 동결보호제는 당 및 이에 상응하는 당 알콜일 수 있다. 구체적인 당 및 이에 상응하는 당 알콜의 일례로서 수크로스, 말티톨, 글루시톨, 락티톨 및 이소-말툴로스 등이 있다. 이외에, 현재 많이 사용되는 동결보호제로는 DMSO, 덱스트란, 프로필렌글리콜, 글리세롤, 트레할로스, 폴리에틸렌글리콜, 혈청알부민 등이 있다.
상기 동결보호제는 이에 제한되지는 않으나, 동결건조 전에 진피조직 내 충분히 침투될 수 있도록 처리할 수 있다. 구체적으로, 상기 동결보호제가 처리된 조직은 약 -70℃ 이하, 바람직하게는 -40℃ 내지 -70℃의 초저온 냉동 조건하에서 보관될 수 있다. 또한, 동결보호제가 처리된 조직은 4시간 이상, 바람직하게는 4 내지 48시간 동안 보관될 수 있다.
상기 동결건조는 이에 제한되지는 않으나, 동결건조기를 이용하여 24시간 내지 48시간 동안 수행될 수 있다.
본 발명의 일 실시예에 있어서, 초임계 유체 추출 후에 수득된 진피조직은 동결건조를 위해 말티톨을 처리하여 침투시키고, 약 -70℃ 이하의 온도에서 최소 4시간 이상 냉동 보관하였다. 냉동 후, 동결건조기로 약 24시간 내지 48시간 동결 건조시키고, 사용시까지 보존하였다.
상기 초임계 유체 추출 이후 수득된 진피조직은 방사선을 조사하여 멸균을 수행할 수 있다. 방사선의 조사 범위는 이에 제한되지는 않으나 10 내지 30 kGy일 수 있다.
본 발명의 일 실시예에 있어서, 초임계 유체 추출 후에 수득된 진피조직은 포장이 완료된 후 감마선 10 내지 30 kGy를 조사하여 멸균을 진행하였다.
무세포 진피조직 이식체의 용도
본 발명에 따른 무세포 진피조직은 계면활성제 처리 없이 탈세포화되어 임계하중, 탄력도 등과 같은 기계적 물성이 유지되면서, 동시에 잔존 계면활성제에 따른 독성이 존재하지 않는다. 따라서, 최적화된 초임계 유체 처리 공정을 통해 수득한 본 발명의 진피조직은 독성 및 이식거부반응이 제거되어 심각한 화상, 외상을 비롯하여, 함몰 반흔 교정, 반안면 위축 교정, 유두 재건, 입술 확대의 재건성형 및 미용성형 등에 유용하게 활용될 수 있다.
무세포 진피조직 이식체를 제조하는 방법
본 발명의 다른 측면은, a) 개체로부터 분리된 피부조직을 표피층 및 진피층으로 분리하는 단계; b) 상기 분리된 진피층을 초임계 유체로 추출하는 단계; 및 c) 초임계 유체로 추출된 진피층을 인산염 완충액으로 세척하는 단계;를 포함하는, 무세포 진피조직 이식체의 제조 방법을 제공한다.
상기 "개체" 및 "초임계 유체"는 상술한 바와 같다.
상기 a) 단계는 개체로부터 분리된 피부조직을 표피층 및 진피층으로 분리하는 단계이다. 구체적으로, 표피층과 진피층의 분리는 당업계에 공지된 방법을 통해 수행될 수 있다. 일반적으로, 표피층과 진피층의 분리는 여러가지 단백질 분해 효소, 예컨대, 디스파아제, 터몰리신, 트립신 등을 사용하여 수행될 수 있다. 또한, 용액의 이온 강도를 변화시켜 표피층과 진피층을 분리할 수 있다. 구체적으로, 1 M 이상의 염화나트륨(NaCl) 용액 또는 20 mM의 EDTA 용액으로 37℃에서 14시간 내지 32시간 동안 처리하면 표피층과 진피층을 분리할 수 있다.
상기 b) 단계는 표피층이 분리된 진피층을 초임계 유체로 추출하는 단계이다. 구체적으로, 진피층으로부터 세포외기질을 제외한 다른 세포 성분, 예를 들어, 면역반응을 유발하는 세포막의 주성분인 인지질 성분을 초임계 유체로 추출함으로써 탈세포화시킬 수 있다.
상기 초임계 유체의 종류, 초임계 유체 추출의 압력 조건, 온도 조건, 및 수행 시간은 '무세포 진피조직 이식체의 제조'에서 상술한 바와 같다.
또한, 상기 b) 단계에서 초임계 유체는 이에 제한되지는 않으나, 공용매를 더 포함할 수 있다. 상기 공용매는 초임계 유체의 추출능 증가 및 용해도 향상 등의 목적으로 첨가되어 분리된 진피층으로부터 세포외기질을 제외한 다른 세포 성분을 추출하여 진피조직의 세포를 대부분 제거할 수 있다.
상기 공용매는 진피조직의 기계적 특성 및 조직의 구조적 형태를 보존하는 한, 그 종류는 특별히 제한되지 않으며, 예를 들어, 에탄올, 물, 메탄올, 헥산, 석유 에테르, 아세토니트릴, 아세톤, 에틸아세테이트 및 메틸렌클로라이드로 이루어진 군으로부터 선택된 하나 이상의 용매일 수 있다. 바람직하게는, 공용매로서 에탄올을 더 포함할 수 있다.
상기 c) 단계는 초임계 유체로 추출된 진피층을 인산염 완충액으로 세척하는 단계로, 상기 인산염 완충액을 이용한 세척을 통해 초임계 유체 추출 이후 진피조직에 존재하는 잔존 용액 및 불순물을 세척할 수 있다.
또한, 상기 세척이 완료된 진피조직은 동결건조, 멸균 및 밀봉 포장 중 어느 하나가 추가로 수행될 수 있으나, 이에 제한되지는 않는다.
이하, 본원 발명을 하기 실시예에 의하여 더욱 상세하게 설명한다. 단, 하기 실시예는 본원 발명을 예시하기 위한 것일 뿐, 본원 발명의 범위가 이들만으로 한정되는 것은 아니다.
I. 초임계 유체 추출 공정을 이용한 무세포 진피의 제조
실시예 1. 초임계 유체 추출 공정을 이용한 무세포 진피의 제조
피부조직의 탈세포화를 위해 초임계 유체 추출 공정을 수행하였다.
먼저, 기증받은 환자의 인체 피부조직(IRB No. 20201305, 서울아산병원)에서 지방층을 제거하였다. 지방층 제거 후, 37℃의 온도 조건 하에서 1M NaCl(Sigma Aldrich, Cat No. S9888)을 24시간 동안 처리하여, 표피를 제거하였다. 표피 제거 후, 분리된 진피층을 멸균된 PBS(biowest, Cat No. L0615-500)로 1시간 동안 세척하였다.
세척 후, 수득한 진피조직을 초임계 추출 장치(Supercritical Extraction System, SES)의 추출조에 투입하고, 초임계 유체 이산화탄소와 공용매 에탄올을 함께 추출조 내부로 주입하였다. 이후, 72.8 기압(bar)의 압력 조건 및 31℃의 온도 조건 하에서 1~3시간 초임계 처리하여 진피조직을 탈세포화시켰다.
이후, 탈세포화된 진피조직을 멸균된 PBS로 상온에서 24시간 동안 세척하였다. 세척이 완료된 RTU(Ready-To-Use) 타입의 무세포 진피조직은 멸균 생리식염수와 함께 밀봉 포장하였다. 또한, FD(Freeze-Drying) 타입의 무세포 진피조직은 말티톨을 처리 및 침투하도록 하고, -80℃에서 4시간 이상 동결시켰다. 이후, 동결건조기를 이용하여 24시간 동안 동결건조 후 밀봉 포장하였다. 포장이 완료된 무세포 진피조직은 감마 멸균(그린피아, 15 kGy)을 수행하여 멸균하고, 멸균 후 실온에서 사용시까지 보관하였다.
준비예 1. 무세포 진피 시료의 준비 및 외형 관찰
상기 실시예 1에서 제조한 무세포 진피(Acellular Dermal Matrix, ADM) 시료에 대한 비교군으로서 하기 표 1의 세부사양에 따른 종래 공지된 방법에 따라 제조된 인체 진피 ADM 시료를 준비하였다.
상기 준비된 각 시료에 대해 두께를 측정하고(표 2), 외형을 관찰하였다(도 1). 이때, 두께 측정은 버니어 캘리퍼스를 사용하여 경쟁사 제품의 서로 다른 세 부분의 두께를 측정하였다.

구체적으로, Native 조직을 포함한 모든 진피조직은 감마선 멸균(15kGy) 처리하였다. 이때, FD(Freeze-Drying) 타입의 Native 시료는 포함되지 않았으며, RTU 타입의 Native 조직을 모든 분석의 미처리 대조군(control) 시료로 이용하였다. 또한, FD 타입 시료의 분석 결과에 포함된 Native 값은 RTU(Ready-To-Use) 타입의 Native 시료의 값으로 표시하였다.
RTU 타입은 일반 식염수(Normal saline)를 포함하여 진피 시료를 이중 포장 후 멸균하여 준비하였다. 또한, FD 타입은 진피 시료를 동결건조 후 이중 포장하여 멸균하여 준비하였다. FD 타입 무세포 진피 시료들은 일반 식염수에서 약 100분간 침지하여 수화시켜 외형 관찰을 수행하였다(도 1).
도 1에 나타낸 바와 같이, 외형상 상기 실시예 1에서 제조한 무세포 진피 시료(SCR, SCF)의 색상은 종래 공지된 방법에 따라 제조된 인체 진피 ADM 제품과 유사함을 확인하였다.
II. 초임계 유체 추출 공정에 따른 무세포 진피의 생화학적 특성 평가
실험예 1. 탈세포화된 진피조직의 조직학적 분석
상기 실시예 1에서 제조한 탈세포화된 진피조직의 조직학적 분석을 위해, H&E(Hematoxylin & Eosin) 염색을 수행하여 조직의 조직학적 형태 및 탈세포 정도를 분석하였다. 즉, 탈세포 성능 지표 중 하나인 Hematoxylin(파란색) & Eosin(붉은색) 염색 수행을 통해 세포핵의 유무를 확인하였다. 세포의 DNA에 결합되는 Hematoxylin은 DNA 정량 결과와 연관이 있으므로, 탈세포 정도의 분석이 가능하다.
H&E 염색 시, 음성 대조군으로는 표피를 제거하지 않은 초임계 유체 처리를 미실시한 원조직 시료(Native)를 사용하였다. 시험군으로는 표피를 제거한 초임계 유체 처리를 실시한 무세포 진피 시료(SCR, SCF)를 사용하였다. 한편, 비교군으로는 표 1의 세부사양에 따른 인체 진피 ADM 시료(A, B, C, D, E, F)를 사용하였다.
염색 결과, 초임계 유체 공정에 따라 탈세포화된 진피 시료 SCR, SCF의 경우 핵이 완전히 제거됨을 확인하였다(도 2). 또한, 인체 진피 ADM 시료 중 C 및 F 시료에서 핵이 관찰되지 않았다. 이에 반해, A, B, D, E 시료에서는 Hematoxylin 염색 부분이 확인되어 탈세포화가 완전히 이루어지지 않음을 확인하였다.
한편, 조직 내부 구조 보존 정도는 A 및 D 시료가 가장 우수함을 확인하였다. 구체적으로, RTU 타입의 경우, A>SCR(초임계 유체 공정 처리 시료)>C>B 순으로 조직 내부 구조 보존 정도가 우수하였다. 또한, FD 타입의 경우, D>SCF(초임계 유체 공정 처리 시료)>E=F 순으로 조직 내부 구조 보존 정도가 우수하였다.
실험예 2. 탈세포화된 진피조직 내 잔존 DNA 함량 확인
상기 실시예 1에서 제조한 탈세포화된 진피조직(SCR, SCF)에 대해 탈세포화 정도를 확인하기 위해, 탈세포화된 진피 시료 내 DNA 함량을 측정하였다. 이때, 음성 대조군으로는 초임계 유체 처리를 미실시한 원조직(Native)을, 양성 대조군으로는 종래 방법에 따라 제조된 표 1의 무세포 진피조직 시료를 사용하였다. DNA 함량 측정은 RTU(Ready-To-Use) 타입 및 FD(Feeze-Drying) 타입의 무세포 진피 시료에 대해 수행하였다.
구체적으로, 각 시료는 Dneasy Blood & tissue kit(QIAGEN, Cat#69506)를 사용하여 각 진피조직의 gDNA를 추출하였다. 이후, 이를 1% 아가로스(agarose) 젤을 이용하여 전기영동을 실시하였다(도 3). 전기영동 시, ExcelBand™ 1KB Plus(0.1-10 kb) DNA Ladder(SMOBIO사, Cat#DM3200)를 사이즈 마커로 사용하였다.
전기영동 수행 결과, RTU 및 FD 두 타입 모두 초임계 유체 처리 공정에 따른 탈세포화된 진피 시료(SCR, SCF)에서 잔존 dsDNA 양이 시판 무세포 진피 시료(A, B, C, D, E, F) 대비 낮음을 확인하였다. 종래 방법에 따라 화학적 처리 또는 효소적 처리 공정에 따라 제조된 무세포 진피 시료 중 B에서 DNA 밴드가 약하게 나타났으나, 나머지 A, D, E 무세포 진피 시료에서는 명확한 DNA 밴드가 확인되었다. 이에 반해, SCR, SCF에서는 DNA 밴드가 전혀 관찰되지 않음을 확인하였다(도 3).
또한, dsDNA의 특정 부위에 직접 결합하는 형광 Dye를 추출된 DNA에 넣어 형광 강도를 측정하는 방법을 채용하는 Qubit dsDNA BR assay kit(Thermo, Cat#Q32853)를 사용하여 무세포 진피 시료의 잔존 dsDNA 함량을 정량 분석하였다(도 4a 및 도 4b). 이때, 모든 시료는 동결건조하여 고형분 질량 기준으로 분석하였다.
분석 결과, DNA 정량 값은 전기영동의 단편화된 DNA의 합과 유사한 결과로 측정되었다. 즉, 전기영동 결과와 마찬가지로, RTU 및 FD 두 타입 모두 초임계 유체 처리 공정에 따른 탈세포화된 진피 시료(SCR, SCF)에서 잔존 dsDNA 양이 종래 방법에 따라 제조된 무세포 진피 시료(A, B, C, D, E, F) 대비 낮음을 확인하였다.
이를 통해, 종래 무세포 진피 제조에 적용되는 계면활성제를 사용하는 화학적 처리 또는 효소적 처리 공정에 비해 초임계 유체 처리시 DNA 제거 효과가 우수함을 알 수 있었다. 이와 더불어, 원조직 90% 이상의 탈세포화가 확인된 바, 이식 거부반응 없이 이식재로서 활용 가능한 우수한 효과를 지님을 알 수 있었다.
실험예 3. 탈세포화된 진피조직 내 면역원성 단백질의 발현 정도 확인
상기 실시예 1에서 제조한 탈세포화된 진피조직(SCR, SCF)에 대해 이식 거부반응 정도를 확인하기 위해, 무세포 진피 시료 내 면역원성 단백질 MHC1의 발현 정도를 확인하였다. 이때, 양성 대조군으로는 종래 화학적 처리 또는 효소적 처리 공정에 의해 제조된 무세포 진피조직 시료(A, B, C, D, E, F)를 사용하였으며, 시험은 RTU 타입 및 FD 타입의 무세포 진피 시료에 대해 수행하였다.
구체적으로, 각 시료는 RIPA 용해 버퍼(lysis buffer)를 사용하여 각 진피조직의 단백질을 추출하였다. 이후, BCA(bicinchoninic acid) 방법을 채용한 Pierce BCA Protein Assay Kit(Thermo, Cat#23225)를 사용하여 단백질 정량을 수행하였다. 단백질 정량 후, MHC1(SANTA CRUZ Biotechnology, Inc., Cat#sc-55582) 항체를 이용하여 웨스턴 블롯을 실시하였다(도 5).
실험 결과, 세포막 단백질로서 면역원성 단백질 MHC1은 모든 무세포 진피 시료에서 음성인 것으로 확인되었다.
이를 통해, 종래 방법에 따라 제조된 무세포 진피조직 시료와 마찬가지로 초임계 유체 처리시 면역원성 단백질 MHC1의 발현이 음성임을 확인한 바, 이식 거부반응 없이 이식재로서 활용 가능한 우수한 효과를 지님을 알 수 있었다.
실험예 4. 탈세포화된 진피조직의 세포외기질(ECM) 보존 정도 확인
상기 실시예 1에서 제조한 탈세포화된 진피조직(SCR, SCF) 내 세포외기질 성분, 즉, 콜라겐, 엘라스틴 및 sGAG 등의 보존 정도를 확인하였다.
실험예 4.1. 탈세포화된 진피조직 내 콜라겐 함량 확인
상기 실시예 1에서 제조한 탈세포화된 진피조직(SCR, SCF) 내 단백질의 손실이 없음을 확인하기 위해, Sircol insoluble Collagen assay kit(Biocolor, Cat#S2000)를 사용하여 콜라겐 함량을 측정하였다. 이때, 음성 대조군으로는 초임계 유체 처리를 미실시한 원조직(Native)을 사용하였다. 양성 대조군으로 계면활성제를 사용하는 화학적 처리 또는 효소적 처리 공정이 적용된 진피조직 시료(A, B, C, D, E, F)를 사용하였다. 각 시료는 동결건조하여 약 5 mg을 사용하여 콜라겐 함량을 측정하였다.
측정 결과, 초임계 유체 공정에 따라 탈세포화된 진피조직 내 콜라겐 보존율은 원조직 기준으로 종래 방법에 따라 제조된 무세포 진피조직 시료 대비 RTU, FD 두 타입 시료 모두에서 가장 우수함을 확인하였다. 구체적으로, 콜라겐 함량은 시료의 건조 중량을 기준으로 RTU 타입 시료의 경우 418 μg/mg이고, FD 타입 시료의 경우 677 μg/mg인 것으로 확인되었다. 이는 원조직 대비 각각 58.8% 및 95.2% 수준으로, 초임계 유체 공정에 따라 탈세포화된 진피조직 내 단백질 손실을 최소화함을 알 수 있었다(도 6a 및 도 6b).
이에 반해, 종래 화학적 처리 또는 효소적 처리 공정에 의해 탈세포화된 진피조직 내 콜라겐 함량은 시료의 건조 중량을 기준으로 RTU 타입 A, B 및 C 시료의 경우 각각 16 μg/mg, 23 μg/mg, 164 μg/mg이고, FD 타입 D, E 및 F 시료의 경우 각각 317 μg/mg, 370 μg/mg, 375 μg/mg인 것으로 확인되었다. 이는 원조직 대비 RTU 타입 A, B 및 C 시료의 경우 각각 2.25%, 3.23% 및 23.1% 수준이고, FD 타입 D, E 및 F 시료의 경우 각각 44.6%, 52.0% 및 52.7% 수준에 불과하였다. 즉, 종래 화학적 처리 또는 효소적 처리 공정에 의해 탈세포화하는 경우, 탈세포화 효과는 우수하더라도 진피조직 내 단백질의 손실이 현저하게 발생함을 알 수 있었다.
상기 결과를 통해, 초임계 유체 공정 처리에 의해 탈세포화시킨 진피조직은 조직학적 형태를 유지하고, 단백질 손실을 최소화하면서 탈세포화가 효과적으로 이루어짐을 알 수 있었다. 이에 따라, 이식재로서 활용 가능한 우수한 효과가 있음을 알 수 있었다.
실험예 4.2. 탈세포화된 진피조직 내 엘라스틴 함량 확인
상기 실시예 1에서 제조한 탈세포화된 진피조직(SCR, SCF) 내 엘라스틴의 손실이 없음을 확인하기 위해, Fastin Elastin assay kit(Biocolor, Cat#F2000)를 사용하여 엘라스틴 분석을 수행하였다. 이때, 음성 대조군으로는 초임계 유체 처리를 미실시한 원조직(Native)을 사용하였다. 양성 대조군으로 계면활성제를 사용하는 화학적 처리 또는 효소적 처리 공정이 적용된 진피조직 시료(A, B, C, D, E, F)를 사용하였다. 각 시료는 동결건조하여 약 5 mg을 사용하여 엘라스틴 함량을 측정하였다.
분석 결과, 초임계 유체 공정에 따라 탈세포화된 진피조직 내 엘라스틴 보존율은 원조직 기준으로 종래 방법에 따라 제조된 무세포 진피조직 시료 대비 RTU, FD 두 타입 제품 모두에서 동등 이상 수준으로 유지 보존됨을 확인하였다. 구체적으로, 엘라스틴 함량은 시료의 건조 중량을 기준으로 RTU 타입 시료의 경우 18 μg/mg이고, FD 타입 시료의 경우 14 μg/mg인 것으로 확인되었다. 이는 원조직 대비 각각 90% 및 70% 수준으로, 초임계 유체 공정에 따라 탈세포화된 진피조직 내 엘라스틴 손실이 일어나지 않음을 알 수 있었다(도 7a 및 도 7b).
이에 반해, 종래 화학적 처리 또는 효소적 처리 공정에 의해 탈세포화된 진피조직 내 엘라스틴 함량은 시료의 건조 중량을 기준으로 RTU 타입 A, B 및 C 시료의 경우 각각 10 μg/mg, 19 μg/mg, 9 μg/mg이고, FD 타입 D, E 및 F 시료의 경우 각각 15 μg/mg, 11 μg/mg, 8 μg/mg인 것으로 확인되었다. 이는 원조직 대비 RTU 타입 A, B 및 C 시료의 경우 각각 50%, 95% 및 45% 수준이고, FD 타입 D, E 및 F 시료의 경우 각각 75%, 55% 및 40% 수준으로, B 및 D 시료 제외 엘라스틴 손실이 현저하게 발생함을 알 수 있었다.
상기 결과를 통해, 초임계 유체 공정 처리에 의해 탈세포화시킨 진피조직은 조직학적 형태를 유지하면서, 엘라스틴 손실이 최소화되어, 이식재로서 활용 가능한 우수한 효과가 있음을 알 수 있었다.
실험예 5. 탈세포화된 진피조직의 성장인자 보존 정도 확인
상기 실시예 1에서 제조한 탈세포화된 진피조직(SCR, SCF)의 성장인자 보존 정도를 확인하기 위해, Proteome Profiler Array, Human XL Cytokine Array Kit, 105 Human Cytokine Array(ARY022B; R&D Systems USA)를 사용하여 두드러지게 발현되는 사이토카인 수 및 발현 정도를 확인하였다(도 8a 내지 도 8f). 이때, 음성 대조군으로는 초임계 유체 처리를 미실시한 원조직(Native)을 사용하였다. 양성 대조군으로 계면활성제를 사용하는 화학적 처리 또는 효소적 처리 공정이 적용된 진피조직 시료(A, B, C, D)를 사용하였다.
구체적으로, 무세포 진피조직 시료를 RIPA 용해 버퍼를 사용하여 각 시료에 대한 단백질을 추출 분리 정제하였다. 이후, BCA 방법이 채용된 Pierce BCA Protein Assay Kit(Thermo, Cat#23225)를 사용하여 단백질 정량을 실시하였다. 각 시료 당 100 mg의 단백질을 각각의 어레이 멤브레인[Proteome Profiler Array, Human XL Cytokine Array Kit(ARY022B), R&D Systems USA]에 동일하게 로딩하여 분석하였다.
분석 결과는 ImageJ 소프트웨어를 사용하여 Dot intensity 값을 측정한 후, Arbitrary intensity 20 이상의 사이토카인 수를 카운팅하고, 이를 하기 표 3 및 표 4에 정리하여 나타내었다.

사이토카인 어레이 수행 결과, 성장인자 보존율이 종래 방법에 따라 제조된 무세포 진피조직 D 시료와 동등한 수준임을 확인하였다. 특히, RTU 타입 시료 중에서 본 발명의 초임계 유체 공정에 따른 무세포 진피 시료(SCR)의 성장인자 보존율이 가장 우수함을 알 수 있었다.
한편, 감마 멸균 수행 여부에 차이가 있으나, 동결건조 타입의 종래 방법에 따라 제조된 D 시료와 본 발명에 따른 SCF 시료 간의 성장인자 보존율을 비교해 본 결과, 동일한 동결건조 타입의 시료일지라도, 초임계 유체 처리 공정에 의해 탈세포화된 본 발명의 SCF 무세포 진피의 성장인자 보존율이 보다 우수함을 알 수 있었다.
실험예 6. 세포 독성 평가
상기 실시예 1에서 제조한 탈세포화된 진피조직(SCR, SCF)의 세포 독성을 평가하였다. 구체적으로, 세포 독성 평가는 당업계에 공지된 L929 마우스 섬유아세포주를 사용하고, 1×104 cells/well의 세포 수로 실시하였다. 또한, 세포 독성 평가는 조직을 37℃에서 세포 배양액에 24시간 담궈 조직 내부 물질을 배양액으로 용출시키는 조직용출법(KCL 공인시험기관 프로토콜 적용)에 따라 수행하였다. 이때, 상기 공인시험기관 기준에 따라 생존율이 공시험액의 70% 미만으로 감소되는 경우, 이는 잠재적으로 세포독성이 있는 것으로 평가하였다.
세포 독성 평가 결과, 종래 화학적 처리 또는 효소적 처리 공정으로 제조된 무세포 진피조직 시료(A, B, C, D, E, F) 대비 본 발명의 초임계 유체 공정에 따른 무세포 진피 시료(SCR, SCF)의 세포 독성이 현저히 낮음을 알 수 있었다(도 9).
상기 결과를 통해, 초임계 유체 공정 처리에 의해 탈세포화시킨 진피조직은 조직학적 형태를 유지하면서, 세포 독성 없이 이식재로서 활용 가능한 우수한 효과가 있음을 알 수 있었다.
III. 초임계 유체 추출 공정에 따른 무세포 진피의 물리화학적 특성 평가
실험예 7. 초임계 유체 추출 공정을 통해 탈세포화된 진피조직의 임계하중 및 탄성 측정
상기 실시예 1에서 제조한 무세포 진피조직(SCR, SCF)에 대해 임계하중 및 탄성을 측정하였다. 초임계 유체 추출 공정을 통해 탈세포화된 진피조직의 임계하중 및 탄성은 만능 인장 시험기(Universal Tensile Machine; EZ-x 모델 (Shimadzu co.))를 사용하여 측정하였다. 이때, 1×1 cm2 크기의 시료를 대상으로 시험하였다. 시험 시료는 음성 대조군으로 초임계 유체 처리를 미실시한 원조직(Native)을 사용하였다. 양성 대조군으로는 계면활성제를 사용하는 화학적 처리 또는 효소적 처리 공정이 적용된 진피조직 시료(A, B, C, D, E, F)를 사용하였다. 한편, 임계하중 및 탄성 측정에 앞서 각 시료는 PBS로 100분 동안 재수화하고, 측정 직전에 조직 표면의 수분을 거즈로 제거하여 측정하였다.
결과적으로, RTU 타입의 초임계 유체 공정에 따라 탈세포화된 진피조직(SCR)의 경우 임계하중이 11.6 N로, 종래 방법에 따라 제조된 무세포 진피조직 시료 A 9.8 N, 시료 B 8.9 N 및 시료 C 6.8 N과 달리 임계하중이 원조직 대비 거의 완벽하게 유지됨을 확인하였다. 특히, FD 타입의 초임계 유체 공정에 따라 탈세포화된 진피조직(SCF)의 경우에는 임계하중이 18.8 N로 원조직 11.6 N 기준 약 1.6배 높으며, 종래 방법에 따라 제조된 무세포 진피조직 시료 D 15.6 N, 시료 E 8.5 N 및 시료 F 17.4 N과 동등 이상의 임계하중 수준을 나타냄을 확인하였다(도 10a 및 도 10b).
본 발명에 따른 RTU 타입의 SCR 시료의 영률 또한 원조직 시료와 동일한 영률 0.6 MPa로 측정되어, 원조직 대비 탄성이 거의 완벽하게 유지됨을 확인하였다. 아울러, FD 타입의 SCF 시료의 경우에도 영률이 1.1 MPa로 원조직 시료의 영률 0.6 MPa 대비 영률이 약 1.8배 높으며, 종래 방법에 따라 제조된 무세포 진피조직 시료 D 1.0 MPa, 시료 E 0.2 MPa 및 시료 F 1.1 MPa와 비교하여 동등 이상의 영률 수준을 나타냄을 확인하였다(도 11a 및 도 11b).
Claims (14)
- 무세포 진피조직 이식체에 있어서,상기 무세포 진피조직의 임계하중(Critical load)은 11 내지 19 N이고,영률(Young's modulus)은 0.5 내지 1.2 MPa인, 무세포 진피조직 이식체.
- 제1항에 있어서,상기 무세포 진피조직은 건조중량을 기준으로 하기 그룹에서 선택되는 어느 하나의 특징을 가지는 것인, 무세포 진피조직 이식체:DNA 잔존량이 80 내지 110 ng/mg;콜라겐 함량이 400 내지 700 μg/mg; 및엘라스틴 함량이 13 내지 19 μg/mg.
- 제1항에 있어서,상기 무세포 진피조직은 원조직 대비 하기 그룹에서 선택되는 어느 하나의 특징을 가지는 것인, 무세포 진피조직 이식체:DNA 잔존량이 5 내지 8%;콜라겐 함량이 55 내지 99%;엘라스틴 함량이 65 내지 95%;임계하중은 90 내지 165%; 및영률(Young's modulus)은 80 내지 200%.
- 제1항에 있어서,상기 무세포 진피조직은 원조직 대비 하기 그룹에서 선택되는 어느 하나의 특징을 가지는 것인, 무세포 진피조직 이식체:아디포넥틴 함량이 22 내지 44%;아포리포단백질 A1 함량이 84 내지 94%;안지오제닌 함량이 19 내지 38%;안지오포이에틴-2 함량이 90 내지 99%;뇌유래 신경영양인자(BDNF) 함량이 73 내지 99%;보체 성분 C5/C5a 함량이 9 내지 19%;CD30 함량이 59 내지 99%;CD40 리간드 함량이 6 내지 28%;CD26 함량이 2 내지 5%;엠프린(Emmprin) 함량이 36 내지 44%;CD105 함량이 41 내지 99%;Fas 리간드 함량이 2 내지 45%;FGF-2 함량이 15 내지 64%;FGF19 함량이 56 내지 98%;IFN-γ 함량이 63 내지 99%;IL-1A(alpha) 함량이 65 내지 99%;IL-1RA(alpha) 함량이 12 내지 43%;IL-17 함량이 82 내지 99%;칼리크레인(KLK) 3 함량이 92 내지 99%;MIF(Macrophage Migration Inhibitory Factor) 함량이 2 내지 9%;MMP(Matrix Metalloproteinase Protein) 9 함량이 83 내지 99%;펜트라신(PTX) 3 함량이 44 내지 91%;레티놀 결합 단백질(RBP) 4 함량이 8 내지 36%;비타민 D 결합 단백질(VDBP) 함량이 6 내지 16%; 및CD31 함량이 67 내지 99%.
- 제1항에 있어서,상기 무세포 진피조직은,초임계 유체로 추출하는 단계를 포함하는 제조 방법으로 제조된 것인, 무세포 진피조직 이식체.
- 제5항에 있어서,상기 초임계 유체로 사용되는 용매는 이산화탄소인 것인, 무세포 진피조직 이식체.
- 제5항에 있어서,상기 초임계 유체는 공용매로서 에탄올이 더 포함되는 것인, 무세포 진피조직 이식체.
- 제5항에 있어서,상기 추출은 30℃내지 40℃의 온도 조건하에서 수행되는 것인, 무세포 진피조직 이식체.
- 제5항에 있어서,상기 추출은 50 bar 내지 400 bar의 압력 조건하에서 수행되는 것인, 무세포 진피조직 이식체.
- 제5항에 있어서,상기 추출은 1 내지 3시간 동안 수행되는 것인, 무세포 진피조직 이식체.
