기존의 향미제는 흡연 매질의 장기관 보관 시 상온에서 화학 구조의 분해 가능성이 높고, 향미 성분이 휘발되어 흡연 중에 담배 맛을 증진시킬 수 있는 충분한 향미 발현이 어렵거나 흡연 시간이 경과함에 따라 향미의 지속성이 약하거나 담배 맛이 변화한다. 이에 흡연 중에 흡연 만족도를 높일 수 있는 향미제의 개발이 필요하다. 식품에서 다양한 향미를 부가하기 위해서 향미제를 적용하고 있으나, 식품의 장기간 가공 및/또는 저장될 경우에 향미가 휘발되고 방출되어 사라져 버리는 경우가 빈번하다. 이에 휘발성 향미의 방출을 방지하거나 지연시켜 저장 수명을 증가시키고, 소비자가 사용할 때 충분한 향미 발현이 이루어질 수 있는 향미제의 개발이 필요하다.
기존의 향미제 기능을 갖는 화합물이 상온(rt) 또는 이와 근접한 온도에서 화학 구조적 안정성이 낮아 구조적 변형 또는 분해가 발생하여 향미 성분이 휘발될 수 있다. 본 발명은, 이를 해결하기 위해서, 열을 가할 경우에 열분해에 의한 향미 성분이 방출되는, 신규한 향미제를 제공하는 것이다.

본 발명의 일 예로, 상기 화학식 1에서 향료 화합물은 카보네이트 연결기()로 공유 결합된 것으로, 상기 화학식 1의 화합물은 열을 가할 경우에 열분해 되어 향료 화합물 및 락톤 화합물로 분해되어 향미 성분의 발현이 이루어질 수 있다. 예를 들어, 상기 화학식 1의 화합물은 락톤계 화합물의 링 열림 메커니즘으로 향료 화합물의 하이드록실기와 반응하여 카보네이트 연결기로 통해 향료 화합물을 공유 결합시킨다. 이는 상온(rt) 및/또는 근접한 온도에서 링 닫힘에 의한 락톤 화합물로 전환되는 것을 방지하는 보호기로 작용할 수 있다. 상기 화학식 1의 화합물은 약 상온(rt) 또는 이와 근접한 온도에서 구조적 안정성을 가지며, 휘발성이 낮고, 열을 받아 링 닫힘 메커니즘으로 카보네이트 연결기가 끊어져 락톤계 화합물 및 향료 화합물로 분해되어 향미가 발현되고, 분해 과정에서 인체에 무해한 이산화탄소가 발생될 수 있다. 즉, 열에 의해서 카보네이트 연결기가 끊어져 향료 화합물로 분해되고, 이산화탄소가 생성될 수 있다. 다음으로 링 닫힘으로 락톤계 화합물로 분해되어 향미를 발현시킬 수 있다.
본 발명의 일 실시예에 따라, 상기 화학식 1에서 n은 1 또는 2의 정수일 수 잇다. R은 탄소수 1 내지 30의 직쇄 또는 분지쇄 알킬; 바람직하게는 탄소수 2 내지 10의 직쇄 또는 분지쇄 알킬일 수 있다.

본 발명의 일 실시예에 따라, 상기 조성물은, 다양한 상(phase)로 제조되고, 예를 들어, 고상(예: 분말, 크리스탈, 플레이크, 분쇄물), 슬러리, 페이스트, 젤, 액상, 에멀젼 또는 에어로졸인 것일 수 있다. 예를 들어, 상기 조성물은, 성형되거나 원하는 제품에 혼합되거나 인쇄, 침지, 분무 및/또는 코팅 등과 같은 본 발명의 기술 분야에서 알려진 방식으로 적용될 수 있으며, 본 문서에는 구체적으로 언급하지 않는다.

본 발명은, 본 발명에 의한 향미제를 포함하는 흡연 물품에 관한 것이다. 본 발명의 일 실시예에 따라, 상기 흡연 물품은, 상기 언급한 본 발명에 의한 화학식 1로 표시되는 향미제 화합물 중 적어도 하나 이상을 포함할 수 있다. 상기 흡연 물품의 가열 및/또는 연소 시 상기 향미제의 열분해에 의한 향미를 제공할 수 있다. 즉, 상기 흡연 물품의 가열 및/또는 연소 시 주류연 및/또는 부류연에 향미를 발현시킬 수 있다.
본 발명의 일 실시예에 따라, 상기 "흡연 물품" (smoking article)이란, 담배, 담배 파생물, 팽화처리 담배(expanded tobacco), 재생 담배(reconstituted tobacco) 또는 담배 대용물에 기반하느냐에 상관없이 흡연 가능한 임의의 제품 또는 흡연 체험을 제공할 수 있는 임의의 제품을 의미할 수 있다. 예를 들어, 상기 흡연 물품은, 궐련, 엽궐련(cigar), 작은 엽궐련(cigarillo), 전자 담배 등 같은 에어로졸을 발생시킬 수 있는 흡연 가능 물품을 의미할 수 있다. 흡연 물품은 에어로졸 발생 물질 또는 에어로졸 형성 기질을 포함할 수 있다. 또한, 흡연 물품은 판상엽 담배, 각초, 재구성 담배 등 담배 원료를 기초로 하는 고체 물질을 포함할 수 있다.
본 발명의 일 실시예에 따라, 상기 향미제는, 상기 흡연 물품에서 흡연매질 100 중량부에 대해 0.0001 중량부 이상; 0.001 중량부 이상; 0.1 중량부 이상; 1 중량부 이상; 1 중량부 내지 20 중량부로 포함될 수 있다.
본 발명의 일 실시예에 따라, 상기 향미제는, 흡연 시 상기 흡연 물품에서 흡연 매질 100 중량부에 대해 100 중량부에 대해 0.00001 중량부 이상; 0.0001 중량부 이상; 0.001 중량부 이상; 0.1 중량부 이상; 1 중량부 이상; 1 중량부 내지 20 중량부로 향미 성분, 예를 들어, 락톤류를 발현시킬 수 있다.
본 발명의 일 실시예에 따라, 상기 흡연 물품은, 궐련형 담배, 액상형 담배 또는 하이브리드형 담배이고, 연소식 궐련 또는 가열식 담배일 수 있다. 또는 전자 담배 (예 : 전자식으로 가열되는 담배) 일 수 있다.
본 발명의 일 실시예에 따라, 상기 향미제는, 궐련의 궐련지에 적용하여 담배 가열 및/또는 연소시, 특히 연기 발생(smouldering)시 열에 의해 향미 성분(예: 락톤류 및/또는 향기 성분)이 발현되어 부류연의 매캐한 냄새를 개선하는 효과를 줄일 수 있다.
본 발명의 일 실시예에 따라, 가열식 담배 스틱의 매질에 적용시 향미 성분의 맛 지속력을 부여할 수 있다. 즉 가열식 담배는 정적인 가열에 의해 매질이 품고 있는 향미 성분이 초기 퍼프(puff)에서 소진되지만 상기 향미제는 열에 의해 분해되어야지 발현되기 때문에 퍼프(puff)가 지속되더라도 향미 성분이 마지막 퍼프(puff)에서도 생성되어 담배맛을 일정하게 유지시킬 수 있다.
본 발명의 일 실시예에 따라, 상기 흡연 물품은, 상기 향미제 또는 상기 향미제 조성물을 포함하거나 제조된 것일 수 있다. 예를 들어, 상기 흡연 물품의 구성 성분 및/또는 부품에 해당될 수 있다. 바람직하게는 흡연 물품에서 가열되는 영역의 구성 성분 및/또는 부품일 수 있다. 예를 들어, 흡연 매질(예: 액상, 젤, 고상, 슬러리, 페이스트), 지관, 튜브, 필터(예: 튜브 필터, 섬유 필터, 직조물 필터, 종이 필터, 캡슐 필터), 권지, 궐련지, 팁페이퍼, 래퍼, 카트리지 (예: 가열 카트리지) 등일 수 있으며, 이들은 본 발명의 목적을 벗어나지 않는다면, 본 발명의 기술 분야에서 알려진 구성성분을 포함하며, 본 문서에는 구체적으로 언급하지 않는다.
본 발명의 일 실시예에 따라, 상기 향미제는, 상기 흡연 물품의 제조 시 그 자체로 기질 또는 기재와 혼합되거나 상기 향미제를 포함하는 조성물을 이용하여 기질 또는 기재와 혼합, 인쇄, 침지(또는, 함침), 코팅 및/또는 분사되어 적용될 수 있다.
본 발명의 일 실시예에 따라, 흡연 매질, 예를 들어, 향미제와 담배 원료(예: 매질 원료, 담배잎)를 포함하거나 첨가물을 더 포함할 수 있다. 다른 예로, 상기 향미제는 흡연 물품의 구성 성분 및/또는 부품 제조 시 향미제로 첨가되고, 흡연 물품에 적용 가능한 베이스 물질, 용매, 가향물질, 흡연 매질 물질 등과 혼합될 수 있다. 또는 상기 흡연 매질은 액상, 젤, 또는 고상일 수 있다.
이하, 실시예 및 비교예에 의하여 본 발명을 더욱 상세히 설명하고자 한다. 단, 하기 실시예는 본 발명을 예시하기 위한 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
실시예 1
1. 소듐(4-멘틸카르보닐옥시)헵타노에이트[Sodium(4-mentylcarbonyloxy)heptanoate, 5a]의 합성
(1-1)에틸 4-하이드록시헵타노에이트[Ethyl 4-hydroxyheptanoate, 2a]의 합성
γ-헵타락톤 20 g(0.15 mol)을 메탄올100 mL에 녹이고 교반하면서 KOH 11.17 g(0.16 mol, 1.05 eq.)을 서서히 넣고 실온에서 12시간 동안 반응시켰다. 반응액을 감압농축시킨 다음 DMF 80 mL를 넣고 교반하면서 브로모에탄 17 g(0.15 mol, 1 eq.)을 넣고 12시간 동안 반응시켰다. 반응액에 물 100 mL를 넣고 에틸 아세테이트로 추출한 다음, 물과 소금물로 씻어주었다. 유기층을 MgSO4로 건조한 다음 감압농축하여 18.1 g(66.7%, 2 steps)의 목적물 2a를 얻었다.

에틸 4-하이드록시헵타노에이트(Ethyl 4-hydroxyheptanoate, 2a) 18 g(0.1 mol)을 THF 120 mL에 녹이고, 피리딘 16 g(0.2 mol, 2 eq.)을 넣고 얼음물로 냉각하고 교반하면서, 멘틸 클로로포르메이트 23 g(0.1 mol, 1 eq.) THF 20 mL 용액을 천천히 dropping 시켰다. 1시간 후 반응액을 실온까지 올리고, 밤새 반응시킨 다음, 물을 넣고 에틸 아세테이트로 추출하였다. 유기 층을 각각 묽은 염산, 탄산수소나트륨 포화용액 및 소금물로 씻어 준 다음 MgSO4로 건조하고, 감압농축하여 30 g(수율 81%)의 목적물 3a을 노란색 액체로 얻었다.

4-(멘틸카르보닐옥시)헵타노익 엑시드 2.5g(4-(Mentylcarbonyloxy)hepanoic acid, 7.5 mmol)을 95% 에탄올 20 mL에 녹이고 98% NaOH 0.29 g(0.95 eq)를 넣고 실온에서 2 시간 동안 교반하였다. Azotrope 현상을 이용하여 물과 에탄올을 날리고 톨루엔을 추가하여 물을 제거한 다음, 헥세인(hexane), 에틸 아세테이트를 넣어 여과하여 흰색 고체를 얻었다.
2. 소듐 4-(메틸카보닐옥시)노나노에이트[4-(Mentylcarbonyloxy)nonanoate, 5b]의 합성
(2-1)에틸 4-하이드록시노나노에이트[Ethyl 4-hydroxynonanoate, 2b]의 합성
γ-노나락톤 20 g (γ-Nonalactone, 0.13 mol)을 메탄올 100 mL에 녹이고 교반하면서 KOH 9.18 g (0.14 mol, 1.05 eq.)을 서서히 넣고 실온에서 12시간 동안 반응시켰다. 반응액을 감압농축시킨 다음 DMF 80 mL를 넣고 교반하면서 브로모에탄 14 g (0.13 mol, 1 eq.)을 넣고 12시간 동안 반응시켰다. 반응액에 물 100 mL를 녛고 에틸 아세테이트로 추출한 다음, 물과 소금물로 씻어주었다. 유기층을 MgSO4로 건조한 다음 감압농축하여 24 g (93%, 2 steps)의 목적물 2b를 얻었다.
(2-2)에틸 4-(메틸카보닐옥시)노나노에이트[Ethyl 4-(mentylcarbonyloxy)nonanoate, 3b]의 합성
에틸 4-하이드록시노나노에이트(2b) 24 g (0.12 mol)을 THF 120 mL에 녹이고, 피리딘 18 g (0.42 mol, 2 eq.)을 넣고 얼음물로 냉각하고 교반하면서, 멘틸 클로로포르메이트 26 g (0.12 mol, 1 eq.) THF 30 mL 용액을 천천히 적가(dropping) 시켰다. 1시간 후 반응액을 실온까지 올리고, 밤새 반응시킨 다음, 물을 넣고 에틸 아세테이트로 추출하였다. 유기 층을 각각 묽은 염산, 탄산수소 나트륨 포화용액 및 소금물로 씻어 준 다음 MgSO4로 건조하고, 감압농축하여 34 g (수율 74.5%)의 목적물 3b의 노란색 액체로 얻었다.

4-(메틸카보닐옥시)노나노익 엑시드 2.5 g (4-(Mentylcarbonyloxy)nonanoic acid, 4b, 7.5 mmol)을 95% 에탄올 20 mL에 녹이고 98% NaOH 0.29 g(0.95 eq)를 넣고 실온에서 2 시간 동안 교반하였다. Azotrope 현상을 이용하여 물과 에탄올을 날리고 톨루엔을 추가하여 물을 제거한 다음, 헥세인 (hexane), 에틸 아세테이트를 넣어 여과하여 흰색 고체를 얻었다.
3. 소듐 5-(메틸카보닐옥시)데카노에이트 5-(Mentylcarbonyloxy)decanoate, 5c]
(3-1) 에틸 5-하이드록시데카노에이트(Ethyl 5-hydroxydecanoate, 2c)의 합성
δ-데카락톤 10 g (δ-Decalactone, 58.7 mmol)을 메탄올 50 mL에 녹이고 교반하면서 KOH 4.2 g (64.7 mmol, 1.05 eq.)을 서서히 넣고 실온에서 12시간 동안 반응시켰다. 반응액을 감압농축시킨 다음 DMF 40 mL를 넣고 교반하면서 브로모에탄 6.4 g (58.7 mmol, 1 eq)을 넣고 12시간 동안 반응시켰다.
반응액에 물 100 mL를 넣고 에틸아세테이트(ethylacetate)로 추출한 다음, 물과 소금물로 씻어주었다. 유기층을 MgSO4로 건조한 다음 감압농축하여 7.6 g (60%, 2 steps)의 목적물 2c를 얻었다.
(3-2)에틸 5-(메틸카보닐옥시)데카노에이트[Ethyl 5-(mentylcarbonyloxy)decanoate, 3c]의 합성
에틸 4-하이드록시노나노에이트 (2c) 7.5 g (34.6 mmol)을 THF 50 mL에 녹이고, 피리딘 5.3 g (69.2 mmol, 2 eq.)을 넣고 얼음물로 냉각하고 교반하면서, 멘틸 클로로포르메이트 8.3 g (37.9 mmol, 1.1 eq.) THF 20 mL 용액을 천천히 적가(dropping) 시켰다. 1시간 후 반응액을 실온까지 올리고, 밤새 반응시킨 다음, 물을 넣고 에틸 아세테이트로 추출하였다. 유기 층을 각각 묽은 염산, 탄산수소 나트륨 포화용액 및 소금물로 씻어 준 다음 MgSO4로 건조하고, 감압 농축하였다. 혼합물을 n-헥세인과 에틸 아세테이트 혼합용매 (7:1)를 사용하여 실리카 겔 컬럼 크로마토그래피하여 4.5 g (수율 32.6%)의 목적물 3c를 얻었다.

에틸 4-(메틸카보닐옥시)노나노에이트 (3c, 2.7 g (6.8 mmol)을 THF 20 mL 및 증류수 10 mL에 녹이고, 수산화리튬 1수화물 0.42 g (10.2 mmol, 1.5 eq.)을 넣고 실온에서 12 시간 동안 반응시켰다. 증류수 10 mL를 추가하고 에터로 추출하였다. 물 층을 진한 염산을 넣어 pH 3으로 조정한 다음 에틸 아세테이트로 추출하였다. 유기 층을 소금물로 씻어 준 다음 MgSO4로 건조하고, 감압농축하여 2.1 g (수율 78%)의 목적물 4b를 노란색 액체로 얻었다.

(4-1)에틸 4-하이드록시운데카노에이트[Ethyl 4-hydroxyundecanoate, 2d]의 합성
γ-운데카락톤 10 g(γ-Undecalactone, 54.2 mmol)을 메탄올 50 mL에 녹이고 교반하면서 KOH 3.9 g(56.9 mmol, 1.05 eq.)을 서서히 넣고 실온에서 12시간 동안 반응시켰다. 반응액을 감압농축시킨 다음 DMF 50 mL를 넣고 교반하면서 브로모에탄(bromoethane) 5.9 g(54.2 mmol, 1 eq.)을 넣고 12시간 동안 반응시켰다. 반응액에 물 80 mL를 녛고 에틸아세테이트(ethylacetate)로 추출한 다음, 물과 소금물로 씻어주었다. 유기층을 MgSO4로 건조한 다음 감압농축하여 10.7 g(85.6%, 2 steps)의 목적물 2d를 얻었다.

에틸 4-하이드록시운데카노에이트(Ethyl 4-hydroxyundecanoate, 2d) 11 g(47.7 mmol)을 THF 60 mL에 녹이고, 피리딘(pyridine) 6.8 g(95.5 mmol, 2 eq.)을 넣고 얼음물로 냉각하고 교반하면서, 멘틸 클로로포르메이트(mentyl chloroformate) 10.5 g(47.7 mmol, 1 eq.) THF 20 mL 용액을 천천히 dropping 시켰다. 1시간 후 반응액을 실온까지 올리고, 밤새 반응시킨 다음, 물을 넣고 에틸아세테이트(ethylacetate)로 추출하였다. 유기 층을 각각 묽은 염산, 탄산수소나트륨(sodium bicarbonate) 포화용액 및 소금물로 씻어 준 다음 MgSO4로 건조하고, 감압농축하여 8.3 g(수율 42.1%)의 목적물 3d을 노란색 액체로 얻었다.

에틸 4-(멘틸카르보닐옥시)운데카노에이트(Ethyl 4-(mentylcarbonyloxy)undecanoate)(3d) 8.3 g (19.4 mmol)을 THF 30 mL 및 증류수 20 mL에 녹이고, 수산화리튬 일수화물(lithium hydroxide monohydrate) 1.2 g (29.1 mmol, 1.5 eq.)을 넣고 실온에서 12 시간 동안 반응시켰다. 증류수 20 mL를 추가하고 에테르로 추출하였다. 물 층을 진한 염산을 넣어 pH 3으로 조정한 다음 에틸아세테이트로 추출하였다. 유기 층을 소금물로 씻어 준 다음 MgSO4로 건조하고, 감압 농축하였다. 혼합물을 헥세인(n-hexane)과 에틸아세테이트 혼합용매(8:1)를 사용하여 silica gel column chromatography하여 6.8 g(수율 91.8%)의 목적물 4d를 얻었다.

(4-4) 소듐(4-멘틸카르보닐옥시)운데카노에이트[Sodium 4-(Mentylcarbonyloxy)undecanoate, 5d]의 합성
4-(멘틸카르보닐옥시)운데칸산 (4-(Mentylcarbonyloxy)undecanoic acid, 4d) 2.5g(7.5 mmol)을 95% 에탄올 20 mL에 녹이고 98 % NaOH 0.29 g(0.95 eq)를 넣고 실온에서 2 시간 동안 교반하였다. Azotrope 현상을 이용하여 물과 에탄올을 날리고 톨루엔을 추가하여 물을 제거한 다음, 헥세인(n-hexane), 에틸 아세테이트를 넣어 여과하여 흰색 고체를 얻었다.
5. 소듐 4-(벤질옥시카르보닐옥시) 운데카노에이트[sodium 4-(Benzyloxycarbonyloxy)undecanoate, 5e]의 합성
(5-1)에틸 4-하이드록시운데카노에이트[Ethyl 4-hydroxyundecanoate, 2d]의 합성
γ-운데카락톤 10 g (54.2 mmol)을 메탄올 50 mL에 녹이고 교반하면서 KOH 3.9 g (56.9 mmol, 1.05 eq.)을 서서히 넣고 실온에서 12시간 동안 반응시켰다. 반응액을 감압농축시킨 다음 DMF 50 mL를 넣고 교반하면서 브로모에탄 5.9 g (54.2 mmol, 1 eq.)을 넣고 12시간 동안 반응시켰다.
반응액에 물 80 mL를 녛고 에틸 아세테이트로 추출한 다음, 물과 소금물로 씻어주었다. 유기층을 MgSO4로 건조한 다음 감압농축하여 10.7 g (85.6%, 2 steps)의 목적물 2d를 얻었다.
1H NMR (CDCl3, 400.13MHz); δ 4.12 (q, 2H, J = 8 Hz, COO-CH2-), 3.59 (m, 1H, CH-O), 2.43 (m, 2H, CO-CH2), 1.81 ~ 0.92 (m, 20H, alkyl)
(5-2)에틸 4-(벤질옥시카르보닐옥시)운데카노에이트[Ethyl 4-(benzyloxycarbonyloxy)undecanoate, 3e]의 합성
에틸 4-하이드록시운데카노에이트 (2d) 8.3 g (36 mmol)을 THF 50 mL에 녹이고, 피리딘 5.5 g (72.3 mmol, 2 eq.)을 넣고 얼음물로 냉각하고 교반하면서, 벤질클로로포메이트 (benzylchloroformate) 6.1 g (35.3 mmol, 1 eq.) THF 20 mL 용액을 천천히 적가 (dropping )시켰다. 1시간 후 반응액을 실온까지 올리고, 밤새 반응시킨 다음, 물을 넣고 에틸 아세테이트로 추출하였다. 유기 층을 각각 묽은 염산, 탄산수소 나트륨 포화용액 및 소금물로 씻어 준 다음 MgSO4로 건조하고, 감압농축하여 9.9 g (수율 75.6%)의 목적물 3e을 노란색 액체로 얻었다. 1H NMR (CDCl3, 400.13MHz); δ 7.37 ~ 7.34 (m, 5H, ph), 5.14 (m, 2H, O-CH2-Ph), 4.12 (brs, 1H, O-CH-), 2.42 (m, 2H, CO-CH2-), 1.90 ~ 0.79 (m, 21H, alkyl)(도 24).
(5-3)4-(벤질옥시카르보닐옥시)운데카노익 엑시드[4-(Benzyloxycarbonyloxy)undecanoic acid, 4e]의 합성
에틸 4-(벤질옥시카르보닐옥시)운데카노에이트 (3e) 10 g (27.5 mmol)을 THF 30 mL 및 증류수 20 mL에 녹이고, 수산화리튬 1수화물 1.7 g (41.4 mmol, 1.5 eq.)을 넣고 실온에서 12 시간 동안 반응시켰다. 증류수 20 mL를 추가하고 에터로 추출하였다. 물 층을 진한 염산을 넣어 pH 3으로 조정한 다음 에틸 아세테이트로 추출하였다. 유기 층을 소금물로 씻어 준 다음 MgSO4로 건조하고, 감압 농축하여 8.2 g (수율 89%)의 목적물 4e를 얻었다.
1H NMR (CDCl3, 400.13MHz); δ 7.37 ~ 7.35 (m, 5H, ph), 5.14 (m, 2H, O-CH2-Ph), 4.48 (m, 1H, O-CH-), 2.47 (m, 2H, CO-CH2-), 1.90 ~ 0.79 (m, 21H, alkyl)

4-(벤질옥시카르보닐옥시)운데카노익 엑시드[4-(Benzyloxycarbonyloxy)undecanoic acid, 4e, 2.5g(7.5 mmol)을 95% 에탄올 20 mL에 녹이고 98 % NaOH 0.29 g(0.95 eq)를 넣고 실온에서 2 시간 동안 교반하였다. Azotrope 현상을 이용하여 물과 에탄올을 날리고 톨루엔을 추가하여 물을 제거한 다음, 헥세인(n-hexane), 에틸 아세테이트를 넣어 여과하여 흰색 고체를 얻었다.
실험예
5d 화합물 (2B)이 열에 노출 시 열적특성(pyrolytic behavier)을 확인하기 위해서 열분해 시험을 진행하였으며, 이는 통상적인 알려진 열분해-가스크로마토그래피/질량분석 (Pyrolysis-Gas Chromatography/Mass Spectrometry [Py-GC/MS]) 방법에 의해서 관찰하였다. 열분해 장비(Pyrolyzer)는 『Double-Shot Pyrolyzer 2020iD』(Frontier Lab, Japan)를 GC/MS(Agilent 6890 GC, USA/Aginelt 7890 MSD, USA) 장비에 연결되어 있는 시스템에서 수행하였다. 2B를 에틸알콜(Ethyl alcohol) 용액에 2.5% 농도로 희석한 후 pyrolyzer sample cup에 10ul loading한 후 열분해 시켰다. 열분해 온도는 Double-Shot Pyrolyzer의 고로(Furnace)의 온도를 지정하여 샘플이 겪게 되는 온도를 조절하였는데 최초 열분해 온도는 80 oC에서 30초 동안 샘플이 놓여진 샘플컵을 고로(Furnace에 노출시켜 샘플컵내 타켓 화합물(2B)이 열분해를 겪도록 하였다. 열에 의해 생성된 혹은 열에 의해 휘발된 성분은 곧 바로 GC/MS의 주입구(Injector)로 주입되어 분리(separation)되었다. 열분해 후 GC/MS 분석되는 동안 샘플컵을 고로(Furnace)로부터 벋어나게 해서 열분해 온도의 영향을 받지 않도록 하였고, 최초 열분해에 의한 GC/MS 분석이 끝난 후에는 최초 사용된 샘플컵을 새로이 화합물을 주입하지 않고 다시 열분해를 겪게 하였는데 이 때 열분해 온도는 10 oC 높은 90 oC에서 30초 동안 열분해를 겪게 하였다. 역시 열분해가 끝난 후 샘플컵은 furnace에서 벋어나게 하여 열분해 온도의 영향을 받지 않도록 하였다. 이러한 방식으로 최초 시료를 샘플컵에 loading한 후 열분해시킬 때 온도는 80oC, 90oC, 100oC에서 최종적으로는 320 oC 까지 승온하면서 열분해 실험을 수행하였다. 그 결과 열분해 온도가 높아지면서 겪게 되는 화합물의 열분해 특성을 온도대별로 분할해서 고찰할 수 있었다. 그 결과는 도 15 및 도 16에 나타내었다.
[분해 매커니즘]
도 15 및 도 16에서 [2B] 화합물은 열분해 시험결과 약 120 oC에서 온도에서 멘톨 및 감마-운데카락톤이 분해되는 것을 확인할 수 있다.
즉, 상기 분해 메커니즘에서 락톤 [1B, gamma-운데카락톤]을 개환 (ring-opening)하고, 하이드록실기를 카보네이트 연결기 (carbonate linkage)로 L-멘톨과 공유결합시켜 [2B] 화합물을 제조하였다. [2B] 화합물이 제품 매트릭스에 적용한 후 열에 의해 L-멘톨 ([3B])과 CO2가 생성되면서 하이드록실기가 노출된 [4B] 화합물이 행성된다. [4B] 화합물은 역시 열 (heat)에 의해 고리 닫힘 (ring-closing, intramolecular esterification)되어 gamma-운데카락톤 [5B]가 생성된다. [2B] 상태에서 하이드록실기가 멘틸 카보네이트기 (Menthyl carbonate group)로 보호되어 상온에서는 고리 닫힘 (ring-closing, intramolecular esterification)이 발생하는 것을 억제할 수 있다. [2B] 화합물의 열분해 패턴을 보면 120 oC에서 260 oC에 온도에 이르는 동안 멘톨이 열분해 되어 발현되고 gamma-락톤은 120oC에서 200oC에 이르는 동안 일차로 발현되고 이어서 200 oC에서 300 oC에 이르는 동안 이차 발현이 풍부하게 된다. 아마도 보호기(protecting group)으로 활용한 멘톨이 열에 의해 deprotecting되어 발현되더라도 [4B] 형태의 화합물 상태로 즉 중간 상태 (intermediate)로 잠시 존재하는 듯 하다. 결국에는 분자간 에스터화반응 (intramoleculat esterification)에 의해 락톤이 생성되지만, 염 (Salt form)상태에서는 이러한 고리 닫힘(ring-closing)이 지연(retardation)될 수 있다. 또한, 열분해 실험 결과 온도가 높아짐에 따라 멘솔이 열분해 되어 발현되고, salt 형태의 [4B] 상태일 때 좀 더 높은 온도에서 분자간 에스터화반응 (intramoleculat esterification)이 일어나서 락톤 [5B]가 생성되었다. 즉 멘솔이 열분해 되는 온도대와 시간차이를 두고 높은 온도대에서 나머지 열분해(Ring-Closing)이 일어나는 것을 알 수 있었다.
실시예 2
제조예의 목적물 (합성된 소듐 (4-멘틸카르보닐옥시)헵타노에이트, 5a, 1 중량%), 물 (40 중량%), 우유 (10 중량%) 및 밀가루 (49 중량%)를 혼합하고 반죽하여 도우를 제조하고, 전기오븐에서 200 온도에서 1 시간 동안 가열하여 베이킹하였다. 오븐에서 꺼낸 이후에 냄새를 맡아서 향미 (예 : 목적물의 합성에 사용된 락톤향 및 멘톨향)가 발현되는 것을 확인하였다.
실시예 3
제조예의 목적물(합성된 소듐 5-(메틸카보닐옥시)데카노에이트, 5c, 0.01 내지 5 중량%), 베이스 기질(펄프, 95 내지 99 중량%) 및 기타 첨가제(잔량)를 혼합 이후에 롤루롤을 이용하여 시트(2 mm 두께)로 제조하고, 상온에서 건조하였다. 상기 시트는 실온에서 냄새를 맡아 보았으나 목적물의 합성에 사용된 향료 화합물의 냄새는 나지 않았다. 다음으로, 상기 시트는 궐련 담배의 궐련지로 적용하여 통상적인 궐련 담배로 제작하고 담배를 흡연하였으며, 흡연 중 향미(예 : 목적물의 합성에 사용된 락톤향 및 멘톨향)가 발현되는 것을 확인하였다.
실시예 4
제조예의 목적물(합성된 소듐 (4-멘틸카르보닐옥시)운데카노에이트, 5d, 0.003 내지 0.02 중량%), 담배 분말(tobacco powder, 90 내지 99 중량%, 0.03 mm 내지 약 0.12 mm의 평균 입자 크기) 및 기타 첨가제(잔량)를 혼합한 이후에 통상적인 방법으로 담배 조성물을 제조하였다. 상기 담배 조성물을 흡연 매질로 적용하고 궐련지를 싸서 래핑한 이후에 필터 및 권지를 구성하여 통상적인 궐련 담배를 제조하였다. 궐련 담배를 흡연하였으며, 주류연과 부류연에서 흡연 중 향미가 발현되는 것을 확인하였다.
이상과 같이 실시예들이 비록 한정된 실시예와 도면에 의해 설명되었으나, 해당 기술분야에서 통상의 지식을 가진 자라면 상기의 기재로부터 다양한 수정 및 변형이 가능하다. 예를 들어, 설명된 기술들이 설명된 방법과 다른 순서로 수행되거나, 및/또는 설명된 구성요소들이 설명된 방법과 다른 형태로 결합 또는 조합되거나, 다른 구성요소 또는 균등물에 의하여 대치되거나 치환되더라도 적절한 결과가 달성될 수 있다. 그러므로, 다른 구현들, 다른 실시예들 및 특허청구범위와 균등한 것들도 후술하는 특허청구범위의 범위에 속한다.
Claims (20)
- 제1항에 있어서,상기 향료 화합물은,하이드록실기 갖는 고리형 모노페르펜계 화합물, 하이드록실기 갖는 모노페르텐계 비고리 화합물, 하이드록실기 갖는 탄소수 6 내지 10의 방향족 화합물 및 하이드록실기 갖는 탄소수 5 내지 6의 비방향족 고리에서 선택되는 것인,향미제.
- 제1항에 있어서,상기 전이금속은, Zr, Mg, Ca, Co, Rh, Ir, Nb, Pd, Pt, Fe, Ru, Os, Cr, Mo, W, Mn, Tc, Re, Cu, Ag 및 Au에서 선택되고,상기 알칼리 금속은, Li, Na, K, Rb 및 Cs에서 선택되는 것인,향미제.
- 제1항에 있어서,상기 향미제는,열분해 시 향미를 발현하는 향미제 화합물인 것인,향미제.
- 제1항에 있어서,상기 향미제는,열분해 시 상기 향료 화합물, 락톤 화합물 및 이산화탄소로 분해되는 것인,향미제.
- 제1항에 있어서,상기 향미제는, 식품용 또는 흡연 물품용 향미제인 것인,화합물.
- 제1항의 향미제; 를 포함하는, 조성물.
