マウスなどの他のモデル生物にはないショウジョウバエの利点は、飼育のコストが低く大規模なスクリーニングに適している点や、繁殖が早いために複数世代かかるような遺伝子操作を短期間に行える点にある。また、記憶の分野においては、cAMP-PKAシグナルと呼ばれる分子機構がホニュウ類とキイロショウジョウバエとで共通して重要な役割を担っていることが報告されている。このように、生存に関わる重要な分子メカニズムは進化的によく保存されており、ショウジョウバエをモデルとした研究が、ヒトの記憶の分子メカニズムの解明に資することが期待されている。
ヒト以外の動物の場合、筆記や言語によるコミュニケーションが取れないため、記憶はその動物の「行動の変化」として測定される。動物の行動が変化する理由は、脳を構成する1つ1つの神経細胞(ニューロン)同士の接続の変化によると信じられている。つまり、記憶は、脳の細胞が変化することによって、脳の機能が変化し、動物の行動が変わるという現象と考えられている。そのため、記憶の研究は、動物の行動の変化を観察するアプローチと、脳神経細胞の変化を観察するアプローチとが存在する。
キイロショウジョウバエにおいては、「T-maze」と呼ばれる方法が記憶を測定する方法として最も一般的に用いられている(特許文献1)。この方法においては、2種類の匂い物質(仮に「匂い1」及び「匂い2」とする)を用意し、100匹のハエに「匂い1」を嗅がせながら、電気ショックまたはエサを与える。その後、T字型の迷路にそれらのハエを入れ、分かれ道のうち片側からは「匂い1」が、もう片側からは「匂い2」が漂ってくるようにし、どちらへ行くかをハエに選択させる。ハエは電気ショックと対にされた場合には「匂い1」を避け「匂い2」のほうへ進みがちになる。一方、エサと対にした場合には、「匂い1」の方へ進みがちになる。全く記憶が形成されていない場合には、それぞれの匂いを選ぶ個体数はほぼ50対50になるが、記憶が形成されるとこの数値に差異が生じる。T-mazeは、これを記憶の点数として評価する手法である。
また、ショウジョウバエにおいて脳を観察するためには、解剖によって頭部に穴をつくり、脳を露出させる必要がある。脳を露出された状態でショウジョウバエを生存させるためには、体液(細胞外液)の組成を模した水溶液を灌流するのが常套手段である。例えば、非特許文献1~3には、以下の脳灌流液が記載されている。

古くからある多くの記憶の実験系は、行動の変化を観察して点数化するのみであり、神経細胞の変化はモニターできない。例えば、特許文献1の方法では、記憶の点数化が100匹のキイロショウジョウバエの行動の総計で為されるのに対して、脳の観察は1匹ずつしか行うことはできないため、脳の変化と行動の変化との対応付けが困難である。さらに、キイロショウジョウバエが2種類の匂いのうちどちらかを選択し、そちらへ歩いて向かうことで記憶の点数化が為されるため、顕微鏡下に頭部を固定する必要がある脳の観察を同時に行うことは出来ない。
また、脳の観察に関しても、脳を露出させたショウジョウバエを、充分な時間、行動可能な状態で維持することが困難である。例えば、非特許文献1~3に記載の方法では、観察のために脳を露出させると、30分ほどで行動しなくなる。記憶形成に伴う脳の変化を追跡するためには、脳を露出させてから行動テストを行い、その後にショウジョウバエを訓練し、一定の間隔で行動テストを繰り返して記憶の保持を観察する必要がある。30分という限られた時間の中では、記憶の保持を観察することができない。
本発明の一態様は、ショウジョウバエにおいて、観察のために脳を露出させた状態でも実行可能な学習方法を提供することを一つの目的とする。本発明の他の態様では、脳を露出させた状態でショウジョウバエを長時間生かし続けることが可能な脳灌流液を提供することを目的とする。
上記の課題を解決するために、本発明には、例えば、以下の態様が含まれる。
1)ショウジョウバエに対して条件刺激を付与する方法であって、ショウジョウバエに肢でつかませておいた物体から当該肢を引き離すことを、条件刺激として付与する方法。
2)トレハロース及びグルコースから選択される少なくとも一つと、塩類と、を含んでなる水溶液であり、浸透圧が225mOsm以上261mOsm以下の範囲内である、昆虫用の脳灌流液。
3)紙を介して、餌の溶液をショウジョウバエに対して与える方法であって、上記餌の溶液をショウジョウバエに対して与える前に、上記紙に対して、餌の溶液の送液と吸液とを繰り返す工程を含む、方法。

本発明の一態様によれば、ショウジョウバエにおいて、観察のために脳を露出させた状態でも実行可能な学習方法を提供することが出来る。本発明の他の態様によれば、脳を露出させた状態でショウジョウバエを長時間生かし続けることが可能な脳灌流液を提供することが出来る。
〔実施形態1〕
以下、本発明の一実施形態について、詳細に説明する。

<1.条件付けに用いる刺激を付与する方法>

(ショウジョウバエ)
ショウジョウバエは、ショウジョウバエ科に属する昆虫を指し、より好ましくはショウジョウバエ属に属する昆虫を指す。その種類は特に限定されないが、キイロショウジョウバエが好ましい。

一態様において、ショウジョウバエは、頭部が切開されている。また、頭部が切開されていることにより、ショウジョウバエは、触角が取り除かれているか、脳が露出しているか、触角が取り除かれると共に脳が露出している。頭部が切開されているショウジョウバエは、必要に応じて脳灌流液を用いた脳の灌流が行われている。
また、別の一態様において、ショウジョウバエは、消化器官(例えば、食道)の少なくとも一部が取り除かれていて、栄養の摂取が出来ない状態のものである。
さらに別の一態様において、ショウジョウバエには、口吻の動きを規制する構造物が設けられている。口吻の動きを規制する構造物とは、例えば口吻の折り畳みを阻害する構造物であり得る。構造物は、例えば接着剤を滴下して硬化させたものなどであってもよい。構造物を設ける位置は、例えば、ショウジョウバエの頭部における口吻の下部付近であり、構造物を設けない場合と比較して、口吻の伸展の方向がより上向きに動くようになる。また、構造物を設けない場合と比較して、口吻の折り畳みが阻害されることによって、口吻の根元(フルクラムと呼ばれる構造)が脳のSubesophageal zone(SEZ)を覆い隠すことが防止される。この結果、実施例にも示すように、SEZの観察と、口吻の観察とを一度に行うことができる。
(条件刺激の付与)
条件刺激とは、後述する無条件刺激と対にして呈示することによって、条件刺激が無条件刺激によって引き起こされる行動に連合され、ショウジョウバエの行動に変化を引き起こすようになる刺激である。具体的には、条件刺激と無条件刺激との対呈示によって条件付けされたショウジョウバエは、条件刺激を呈示されるだけで、無条件刺激を呈示されたときと同様の行動をとる(つまり記憶が形成される)ようになる。
条件刺激とは、後述する無条件刺激と対にして呈示することによって、条件刺激が無条件刺激によって引き起こされる行動に連合され、ショウジョウバエの行動に変化を引き起こすようになる刺激である。具体的には、条件刺激と無条件刺激との対呈示によって条件付けされたショウジョウバエは、条件刺激を呈示されるだけで、無条件刺激を呈示されたときと同様の行動をとる(つまり記憶が形成される)ようになる。
本実施の形態では、「ショウジョウバエに肢でつかませておいた物体から肢を引き離すこと」を、条件刺激として用いる。この条件刺激は、ショウジョウバエに肢で物体につかませておいてから、ショウジョウバエと物体との相対的な移動、すなわち、1)ショウジョウバエを固定し、物体を動かすこと、2)物体を固定し、ショウジョウバエを動かすこと、又は、3)ショウジョウバエと物体とを何れも動かすこと、によって実現できる。ショウジョウバエと物体との相対的な移動は、1)ショウジョウバエを固定し、物体を動かすこと、すなわち物体を肢から引き離す方向に動かすこと、が好ましい。物体を動かす方向は、例えば、上方向、下方向、水平方向、回転方向、及び、これらの組合せの方向等から選択される。
物体は、ショウジョウバエが肢でつかむことが出来るものであれば、その形状、材質、及びサイズ等は特に限定されない。好ましい一例では、物体は木製であり、棒状である。棒の直径は好ましくは2mm以上であり、棒の長さ(ハエがつかむ部分の長さ)は好ましくは3mm以上である。
条件刺激として機械的な刺激を用いる利点は、1)まず条件刺激として優れていること、2)頭部が切開され触角が取り除かれたショウジョウバエにも適用できること、及び、3)視覚刺激ではないので、励起光の照射を伴う脳の蛍光観察等を行う上でも支障のないこと、等が挙げられる。
(無条件刺激の付与)
条件刺激と対呈示される無条件刺激は、例えば、「ショウジョウバエに対して餌を与えること」、が好適である。刺激を可能な限り一定にする(再現性あるものにする)ため、餌は溶液として、ショウジョウバエに与えることが好ましい。また、機械抄紙の紙よりも目の少ない紙を介して、餌の溶液を与えることがより好ましい。機械抄紙の紙は、抄紙時の流れ方向に沿って繊維構造が揃う(目がある)ため、ショウジョウバエの口吻が紙のどこに当たるかで付与される刺激が異なってくるし、紙からの水分の蒸発スピードも一様ではない。なお、機械抄紙の紙よりも目の少ない紙とは、例えば、バッチ式抄紙の紙である。バッチ式抄紙の紙とは、例えば、手漉きの紙が挙げられ、目の少なさの観点で、流し漉きよりもため漉きの手漉きの紙が好ましい。なお、実施例では、表面の凹凸が極めて少なく吻表面全体を一様に刺激することが可能との理由で、手漉きの和紙(雁皮紙)を用いて餌の溶液(スクロース溶液)を与えたが、従来の方法(キムワイプをちぎった繊維を用いるShiraiwa and Carlson, 2007)よりも、無条件刺激の付与の再現性が明らかに優れていた。
条件刺激と対呈示される無条件刺激は、例えば、「ショウジョウバエに対して餌を与えること」、が好適である。刺激を可能な限り一定にする(再現性あるものにする)ため、餌は溶液として、ショウジョウバエに与えることが好ましい。また、機械抄紙の紙よりも目の少ない紙を介して、餌の溶液を与えることがより好ましい。機械抄紙の紙は、抄紙時の流れ方向に沿って繊維構造が揃う(目がある)ため、ショウジョウバエの口吻が紙のどこに当たるかで付与される刺激が異なってくるし、紙からの水分の蒸発スピードも一様ではない。なお、機械抄紙の紙よりも目の少ない紙とは、例えば、バッチ式抄紙の紙である。バッチ式抄紙の紙とは、例えば、手漉きの紙が挙げられ、目の少なさの観点で、流し漉きよりもため漉きの手漉きの紙が好ましい。なお、実施例では、表面の凹凸が極めて少なく吻表面全体を一様に刺激することが可能との理由で、手漉きの和紙(雁皮紙)を用いて餌の溶液(スクロース溶液)を与えたが、従来の方法(キムワイプをちぎった繊維を用いるShiraiwa and Carlson, 2007)よりも、無条件刺激の付与の再現性が明らかに優れていた。
一態様において、紙は、餌の溶液を貯蔵した供給手段(例えば、マイクロインジェクタに接続した注射針)に連結されている。ショウジョウバエに無条件刺激を付与する直前まで、供給手段から紙に対して、餌の溶液の送液と吸液とを繰り返すことによって、紙から水分が蒸発した場合でも、紙に含まれる餌の溶液の濃度を略一定に保つことができ、再現性よく無条件刺激の付与を行うことができる。
条件刺激と無条件刺激とを対呈示する方式は特に限定されず、条件刺激の付与の開始後速やかに無条件刺激を付与する方式等が挙げられる。
条件刺激と無条件刺激との対呈示は、少なくとも1回以上であり、通常は、所望する状態になる(記憶が形成される)まで複数回行われる。対呈示の回数は、例えば、2回以上、3回以上、4回以上、又は、5回以上であってよく、例えば、10回以下、9回以下、又は、8回以下であってよい。
条件刺激の時間、無条件刺激の時間、これら刺激の対呈示を複数回行う場合の対呈示同士の間隔等の諸条件も、ショウジョウバエの種類等に応じて適宜設定すればよい。
キイロショウジョウバエの場合は、rod removal(棒を離す)刺激(条件刺激)を与えながら、スクロース水溶液を与える(無条件刺激)と、パブロフの犬がベルの音を聞くだけで唾液の分泌という摂食行動をとるようになったように、ハエはrod removal刺激のみで、吻を伸展させるという摂食行動を示すようになった(実施例参照)。吻の伸展は、キイロショウジョウバエの頭部を固定した状態でも観察し定量可能な行動である。吻を最大に伸ばしたときの長さに対してどのくらい伸ばしたか、その割合(%)を記憶の点数として評価する。最初は、rod removal刺激のみでは、ほとんど吻を伸ばすことはなく、記憶の点数としてはほぼ0%であるが、rod removal刺激とスクロース水溶液による刺激とを対にして呈示する操作を30秒間隔で5回くり返す(訓練)と、ほぼ全ての個体でこの点数が増大する。また、記憶形成に異常があると報告されている、キイロショウジョウバエの突然変異体「dnc1」「rut2」では、訓練後の点数の上昇が小さく、この点数化法が記憶の評価方法として理にかなっていることの裏付けも得られた。
<2.試験の方法>
本発明の一態様は、上記<1.条件付けに用いる刺激を付与する方法>の欄に記載の方法によって、条件刺激が付与されると無条件刺激に関する応答(例えば、口吻を伸ばすこと)を示すようになったショウジョウバエを用いて、記憶に関係する試験を行う方法である。

この試験では、上記条件刺激と同種の刺激をショウジョウバエに付与して、ショウジョウバエの応答(無条件刺激に関する応答の程度)を観察することや、ショウジョウバエの脳を観察すること、等が行われる。
ショウジョウバエの脳を観察することには、ショウジョウバエの脳を露出し、特に脳のSEZを露出して観察することが含まれる。SEZの観察においては、上記のとおり口吻の下部に滴下された接着剤等によって、ショウジョウバエの頭部に構造物が設けられていることが望ましい。これにより、フルクラムと呼ばれる口吻の根元構造がSEZの観察の妨げとなることを防ぎ、SEZの変化と口吻の動きの変化とを一度に観察することができる。
ショウジョウバエの口吻の動きの観察においては、ショウジョウバエに視覚的刺激を与えない条件下で観察を行うことが好ましい。具体的には観察時の光源としてショウジョウバエが視認できない波長の光を用いることが好ましく、特に波長700nm以上780nm以下の範囲内の赤色光を用いることがより好ましい。観察時の光源は、さらに好ましくは710nm以上770nm以下の範囲内、又は、720nm以上760nm以下の範囲内、又は、730nm以上750nm以下の範囲内、又は、735nm以上745nm以下の範囲内の赤色光である。
記憶現象においては、脳の神経細胞の変化によって動物の行動が変化するが、本発明の一実施形態では、上記のとおりこれらを同時に観察することができる(実施例も参照)。カルシウムイオンの濃度によって蛍光強度が変わるタンパク質(Ca2+インジケータタンパク質)GCaMPを、遺伝子組換え技術を用いて脳の特定の神経細胞にのみ発現させておくと、神経細胞が活動する際には細胞外からカルシウムイオンが流入するため、蛍光強度の変化が生じる。この蛍光強度の変化を定量することによって、動物の神経活動をモニターすることができる。この手法を用いて、吻を伸ばすなどの摂食行動を引き起こすコマンドニューロンであるフィーディング・ニューロンの活動をモニターすると、最初はrod removal刺激のみでは、GCaMPの蛍光増大はほとんど見られない(フィーディング・ニューロンはほとんど活動しない)が、訓練後には蛍光増大が観察される(フィーディング・ニューロンが活動する)ようになる。この神経活動の変化は、上述の吻を伸ばすようになるという行動の変化とよく対応しており、これも記憶を評価するための指標となる。なお、上記蛍光強度の変化の定量は、例えば二光子蛍光顕微鏡を使用することで行われ得る。二光子蛍光顕微鏡を使用することは、フィーディング・ニューロンに焦点をしぼった観察を行う点で好ましい。また、ショウジョウバエの脳への負担が少ない方法であることが好ましく、例えば、1)ガルバノスキャンモードを適用して、観察の対象領域(ここではフィーディング・ニューロン)を、遠赤外光を照射して探索し、2)観察の対象領域(ここではフィーディング・ニューロン)を発見次第、レゾナントスキャンモードを適用してその神経活動を観察する、方法などが採用され得る。レゾナントスキャンモード適用時にショウジョウバエの脳に照射される遠赤外線の波長は、二光子励起を発生させるものであれば特に限定されないが、900nmであることが好ましい。同遠赤外線の照射時間は、観察に必要な長さであり得、例えば10秒以上であり得る。
<3.装置>
本発明の一態様に係る装置は、上記<1.条件付けに用いる刺激を付与する方法>欄に記載の方法や、上記<2.試験の方法>欄に記載の方法を実施するのに適した装置である。この装置は、(1)ショウジョウバエが肢でつかむ対象となる「物体」と、(2)ショウジョウバエを生きた状態で物理的に固定するための「固定部材」と、(3)固定部材と上記物体とを相対的に移動させる「駆動手段」(条件刺激の付与手段)と、を備えている。さらに、(4)無条件刺激の付与手段、(5)ショウジョウバエの脳を観察する観察手段、及び、(6)ショウジョウバエの脳に対して脳灌流液を用いた灌流を行う灌流手段、等を備えてもよい。
本発明の一態様に係る装置は、上記<1.条件付けに用いる刺激を付与する方法>欄に記載の方法や、上記<2.試験の方法>欄に記載の方法を実施するのに適した装置である。この装置は、(1)ショウジョウバエが肢でつかむ対象となる「物体」と、(2)ショウジョウバエを生きた状態で物理的に固定するための「固定部材」と、(3)固定部材と上記物体とを相対的に移動させる「駆動手段」(条件刺激の付与手段)と、を備えている。さらに、(4)無条件刺激の付与手段、(5)ショウジョウバエの脳を観察する観察手段、及び、(6)ショウジョウバエの脳に対して脳灌流液を用いた灌流を行う灌流手段、等を備えてもよい。
装置の一具体例について、図3に基づき説明する。この装置では、ショウジョウバエが肢でつかむ対象となる物体として棒(rod)を備え、この棒はステッピングモータ(駆動手段)によって上下方向に動かすことが出来る。また、装置は、ショウジョウバエを生きた状態で物理的に固定し、観察するための観察チャンバを備える。頭部が切開され脳が露出したショウジョウバエは、その頭部において、観察チャンバに固定されている。
この装置はさらに、スクロース溶液(餌の溶液)を、和紙を介してショウジョウバエに与えるためのマイクロインジェクタ(無条件刺激の付与手段);ショウジョウバエの脳内を蛍光観察する二光子蛍光顕微鏡(観察手段);及び、ショウジョウバエの脳に対して脳灌流液を用いた灌流を行う灌流装置;を備えている。
さらに、この装置は、ショウジョウバエの行動を記録するカメラ;操作時の観察用の実体顕微鏡;及び、カメラ及び実体顕微鏡用の光源(波長740nmの赤色光);等を備えている。
ステッピングモータの動作は、図示しない制御回路によって制御されている。この制御によって、ショウジョウバエの訓練時には、条件刺激をプログラムされた条件にて、ショウジョウバエに呈示する。訓練を受けたショウジョウバエを実験に用いる際には、この制御によって、条件刺激と同様の刺激をプログラムされた条件にて、ショウジョウバエに呈示する。
<4.脳灌流液>
本発明の一態様に係るショウジョウバエ用の脳灌流液は、1)トレハロース及びグルコースから選択される少なくとも一つと、2)塩類と、を含んでいる水溶液であり、3)浸透圧が225mOsm以上261mOsm以下の範囲内である。また、この脳灌流液は、ショウジョウバエ用に限定されず、昆虫用の脳潅流液として用いることも出来る。

トレハロースは、昆虫の血糖の成分であり、グルコースが2つ結合した構造の糖である。グルコースはトレハロースの代替として利用が可能である。脳灌流液における、トレハロース及びグルコースの濃度(両方を用いる場合は合計の濃度)の下限は、例えば、1mM以上、3mM以上、4mM以上、又は、5mM以上である。トレハロース及びグルコースの濃度(両方を用いる場合は合計の濃度)の上限は、例えば、75mM以下、70mM以下、65mM以下、又は、60mM以下である。
塩類は、一般的な生理的食塩水を構成する塩類から適宜選択すればよい。塩類は、例えば、NaCl、KCl、MgCl2、及び、CaCl2からなる群より選択される少なくとも一種であり、複数種を用いることが好ましい。脳灌流液における、塩類の濃度の下限は、例えば、50mM以上、60mM以上、70mM以上、又は、75mM以上である。塩類の濃度の上限は、例えば、160mM以下、150mM以下、140mM以下、又は、130mM以下である。塩類としてNaClを用いる場合、その濃度の下限は、例えば、40mM以上、50mM以上、60mM以上、又は、70mM以上であり、濃度の上限は、例えば、150mM以下、140mM以下、130mM以下、又は、120mM以下である。塩類としてKCl、MgCl2、及び、CaCl2を用いる場合、それぞれの濃度の下限は、例えば、0.5mM以上、0.6mM以上、又は、1mM以上であり、濃度の上限は、例えば、10mM以下、7mM以下、又は、5mM以下である。
脳灌流液の浸透圧の下限は、230mOsm以上、235mOsm以上、又は、240mOsm以上が好ましい場合があり、上限は、255mOsm以下、又は、250mOsm以下が好ましい場合がある。なお、脳灌流液の浸透圧の調整は、スクロース等の体液中においては代謝されない浸透圧調節剤を用いて行えばよい。
脳灌流液のpHは、例えば、6.5以上であり、7.0以上であることが好ましい。脳灌流液のpHは、例えば、8.0以下であり、7.5以下であることが好ましい。pHの変動を防止するため、脳灌流液はpH緩衝液であることが好ましい。
従来のキイロショウジョウバエ用の脳灌流液では、脳を露出させた状態でのキイロショウジョウバエの観察は、30分程度が限度であった。本発明の一態様に係るショウジョウバエ用の脳灌流液では、驚くべきことに、脳を露出させた状態でのキイロショウジョウバエの観察が、少なくとも2時間以上、3時間以上、4時間以上、5時間以上、10時間以上、又は、24時間以上にわたり可能となった。
脳灌流液は、本来は、生物の体液組成を模した組成物を用いるのが常套手段である。しかし、ショウジョウバエの場合は体が小さい(キイロショウジョウバエは体長が2~3mm)ため、細胞を壊さずに体液を回収することが困難である。また、細胞内と細胞外では液体成分の組成が異なり、すりつぶしてしまうと両者が混ざり、正確な体液組成の測定は不可能となる。そのため従来は、他の体の大きな昆虫の体液組成を参考にするなどして、場当たり的に組成を決めて実験を行っており、ここに改善の余地が存在した。
<5.応用など>
1つ1つの神経細胞の変化と、記憶形成による行動の変化の因果関係を追及することが、記憶研究の最も大きなテーマの1つである。記憶研究においても、ある薬を投与したら行動の点数がアップしたりダウンしたりするというだけではなく、その原因となる細胞の変化がどうなっているのか(神経活動に影響しているのか、あるいは神経細胞の形態に影響しているのかなど)をモニターする意義がある。
1つ1つの神経細胞の変化と、記憶形成による行動の変化の因果関係を追及することが、記憶研究の最も大きなテーマの1つである。記憶研究においても、ある薬を投与したら行動の点数がアップしたりダウンしたりするというだけではなく、その原因となる細胞の変化がどうなっているのか(神経活動に影響しているのか、あるいは神経細胞の形態に影響しているのかなど)をモニターする意義がある。
本発明の一態様では、キイロショウジョウバエの脳を露出させた状態で記憶形成をリアルタイムで観察することが可能となった。そこで発明者たちは、オプトジェネティクスと呼ばれる、神経細胞へ光を照射してその活動を操作する技術を用いて、フィーディング・ニューロンで起こる分子や細胞の変化によって、記憶が形成されることを明らかにした。これによって、キイロショウジョウバエの脳を構成する約20~25万の神経細胞の中から、フィーディング・ニューロンに焦点をしぼった解析を行うことができ、記憶形成の過程でどのような細胞変化がみられるか、すなわち記憶の脳内変化を、初めて観察することが可能になった。この技術的進歩により、記憶形成の過程で働く分子群の機能をしらべることが可能になった。ひいては、記憶形成の過程に作用する薬剤の探索も可能にした。より具体的には、以下に示す応用が考えられる。
1)ショウジョウバエ遺伝学を使った記憶に関与する分子の探索
本発明は、従来法のように記憶の結果生ずる行動変化を査定するだけでなく、フィーディング・ニューロン上に生じるシナプスの変化を細胞レベルで調べることができる。ショウジョウバエ遺伝学を駆使すると、記憶の形成に関与する、シナプスの前後の細胞(シナプスの後の細胞は(入力をうけるのは)フィーディング・ニューロンである。)での各分子の機能を調べることができる。この方法によって、記憶過程で働いている分子群を検索することが可能になる。
2)薬剤投与
無作為に記憶機構に影響を与える薬剤をスクリーニングすることも可能であるが、さらに、1)で記憶形成のために機能する分子群を同定し、それらの分子群を創薬のターゲットとして、薬剤をデザインすることも可能になる。本発明の一態様では、薬剤を還流することによって、記憶形成の現場に直接与えることができる。さらに、必要に応じては小ピペットを用いることによって、薬剤を局所的にフィーディング・ニューロンのみに投与することも可能である。その利点は、記憶形成とは直接関係のない神経細胞への薬剤の作用によって、副作用として動物の行動が変化し、一見すると記憶に効果があるように見えるというような偽陽性をひろう可能性(例えば、ハエの行動観察のみを行う従来技術では、記憶とは無関係に、ハエの一般的な活動が亢進した結果、歩行量が上昇した場合でも、一見すると記憶の点数が良くなったように見えることもある)を低くすることができる。そのため、薬剤投与したうえで記憶を担う細胞変化を査定することによって、真に記憶形成過程への薬剤の影響について知ることができる。

〔実施例1〕
以下、本発明のショウジョウバエの脳と行動を同時的に観察する方法に係る一実施例について説明するが、本発明は以下の記載によっては限定されない。表1に重要材料一覧を示す。
<実験モデルと条件の詳細>
標準プロトコルに従い、キイロショウジョウバエは、明暗12時間サイクル、25℃下におかれた。すべての実験において雌個体が使われた。
以下、本発明のショウジョウバエの脳と行動を同時的に観察する方法に係る一実施例について説明するが、本発明は以下の記載によっては限定されない。表1に重要材料一覧を示す。

<ハエ個体の系統>
野生型としてCanton-Sが使われた。dnc1突然変異体(参考文献1)、及びrut2突然変異体(参考文献2)は、パブロフ条件付けアッセイにおいて記憶形成に異常を示すことが確認されている。

GMR81E10-GAL4はJanelia Farm(参考文献3)によって単離され、フィーディング・ニューロンにおける遺伝子の発現に使われた。GMR81E10-GAL4とUAS-mCD8-gfp(参考文献4)をヘテロ接合体として発現する個体を、フィーディング・ニューロンの解剖学的構造を可視化するために使用した。
=方法詳細=
<準備・設計と生理食塩水>
今回の実験においては、FLIES(Fly brain Live Imaging and Electrophysiology
Stage)(参考文献5)と呼ばれる記録チャンバを改良して、ハエの行動観察と脳のイメージングを行った。頭部の切開には、ピンセットを加工して作成した10ミクロンのものも切断できる超微細はさみを用いた。長方形の端部を持つピンセット(約100μm幅)を、内部器官と外骨格を取り除くために使った。ハエの空腹が満たされること、そして給餌による報酬効果を避けるために、食道は取り除いた。成虫の頭部の気嚢は、解剖中、可能な限りそのまま維持した。図3及び4に示すようなイメージング及び光学的操作のために、口吻の根元(フルクラムと呼ばれる構造)がSEZを覆い隠すのを防ぐため、口吻の下部に、光硬化型の接着剤を一滴滴下した。その結果、しばしば口吻が伸展時に前方よりも上向きに動くようになった。実験はSY2020と呼称される新しい生理食塩水(組成:NaCl85mM、KCl1mM、MgCl22.5mM、CaCl23mM、NaHCO310mM、HEPES-NaOH5mM、トレハロース5mM、スクロース35mM、pH7.2)を用いて行われた。SY2020は既存の体液組成に似た溶液を改良して作られ(参考文献6~10)、24時間以上の実験操作にあたりプレップを良好に維持するのに最適であった。

<条件付け>
0.65±0.05mgの木片を3-5日齢の成虫雌個体の胸部背側に糊付けし、その状態でハエ個体が動けるようにしておき、そのハエを麻酔を行うことなく記録チャンバに取りつけられるようにした。野生型個体については、条件付け中にスクロース刺激に対して強く反応するように、実験前に4mlのMilli-Q水を浸したペーパータオルを入れたプラスチックバイアル内で27-28時間かけて飢餓状態にさせた。一方、GCaMP6mを発現する個体(参考文献11)は47時間、ハロロドプシンを発現させた個体(参考文献12)は40-44時間を飢餓状態にするために必要とした。解剖後の個体の頭部気嚢が勢い良く拍動し、気管を通して酸素の供給が良好に行われている場合に限り条件付けアッセイを行った。

飢餓状態にしたのち、図3のAに示す通りハエ個体を記録チャンバにセットした。ハエ個体には740nm赤色光(fiber coupled LED M740F2,Thorlabs)を光学ファイバー(中心直径200mm,NA 2.2,Thorlabs)から照射し、視覚的刺激を避けながらスクロースを与えられるようにした。個体の行動は、CMOSカメラ(DMK23U445,Imaging Source)を取り付けたVideo-Zoom顕微鏡XV-440を用いて、その側方から記録した。映像の取得にはIC capture 2.4 software (Imaging Source)を使用した。rod removalについてはLabView 2017で駆動するステッピングモータ(TAMM40-10C and GSC-01,SigmaKoki)を使用して行った。ステッピングモータには小型マニピュレータ(YOU-2,Narishige)を搭載した。ロッド(直径2mm、長さ10mm)は、操作の融通のため、より長い棒に接続し、マニピュレータに適切な角度で固定した。棒をマニピュレータとともに個体の近くに配置し、ハエ個体がその肢でつかめるようにした。ステッピングモータの動作により、個体の肢からロッドを離し、またロッドを再びつかむことができるように、もとの位置にロッドを戻した。
スクロースによる刺激は、スクロース溶液を含む紙片の接触により行った。長方形(幅400μm、長さ数mm)の和紙(雁皮紙(Haibara))の紙片を、加工して曲げたピンを用いて、注射針の中に保持されるようにした。注射針はスクロース溶液を供給するための、シリコンチューブ及びマイクロインジェクタ(IM-11-2, Narishige)に接続した(図3のA)。アームスタンド(Nikon)に搭載した実体顕微鏡SMZ-800、またはアームスタンドF10(Wraymer)に搭載した実体顕微鏡LW-820(Wraymer)を通して実験ハエ個体を見ながら、ジョイスティックマニピュレータ(MN-151, Narishige)を用いてこの注射針を操作した。この方法においては、ハエ個体は報酬としてスクロースを飲み込むことはできず、報酬としては吻表面の感覚毛による甘味感覚受容に限られる。顕微鏡のステージとコンデンサーは、マニピュレータM-152(Narishige)及び高さ調整台(P-1A, Narishige)とともに可動観察台に取り付けられた記録チャンバへ交換した。生理食塩水は酸素供給のために、ポンプ(TP10-SA, As-One)または重力によって灌流させた。灌流は、解剖後ハエ個体の状態が安定するまで(自発的な口吻の伸展が落ち着きスクロース溶液への反応が強くなるまで)2時間以上続けた。2光子イメージングと光学的活性化を除いた条件付け実験は25℃で行った。
<カルシウムイメージングとレーザー不活性化>
カルシウムイメージング実験にあたり, 可視化のためにFVMPE-RS2光子顕微鏡(Olympus)が、励起のためにInSight DS Dual-OL(Spectra-Physics)が使用された。GMR81E10-GAL4の発現がおおむねSEZ内の1対のフィーディング・ニューロンに制限されている一方(参考文献13)、GMR81E10-GAL4の発現は、口吻から脳のSEZ腹側後部へと投射する感覚神経線維束においても確認される。感覚神経線維束が投射する部位は、フィーディング・ニューロンの可視化と操作を行った領域の外にある。第3染色体上にUAS-GCaMP6mとGMR81E10-GAL4をもつヘテロ接合体において、フィーディング・ニューロンのmain dendritic trunkのGCaMP蛍光を定量した。条件付け前に、フィーディング・ニューロンのmain dendritic trunkをガルバノスキャンモードで特定し、定量には15ピクセル(4.97mm)×30ピクセル(9.93mm)のROI(Region of interest)を設定した。同ROI設定で、高速スキャニングを行うためにレゾナントスキャニングモードに切り替えた。レゾナントスキャニングモードにおいては、15ピクセル×30ピクセルのROIを含む142ピクセル(47.07mm)×512ピクセル(169.71mm)の範囲を、1スキャン当たり50ミリ秒で200サイクル、合計10秒間スキャンした。7.6秒持続する強い条件刺激の際には、条件刺激開始1秒前から10秒間、900nm遠赤外線(5%power)でスキャンした。ROI内における蛍光強度の平均値は、FVMPE-RS2光子顕微鏡に付属するFV31S-SW software(Olympus)によって定量した。

第2染色体に20XUAS-eNpHR3.0-EYFP(参考文献14)というハロロドプシンを発現させるトランスジーンを、第3染色体にGMR81E10-GAL4を持つホモ接合体を、レーザーを用いたフィーディング・ニューロンの活動抑制実験に使用した。レーザー光の照射はフィーディング・ニューロンの樹状突起がmain dendritic trunkの中心から背側にかけて広がる領域に限定して行った(参考文献15)。条件付け前に、ハロロドプシンと連結されたYFPの蛍光によってフィーディング・ニューロンを特定するため、ハエ個体の脳SEZはガルバノスキャンモード(925nm)によってスキャンされた。不活性化のため、脳の両側のフィーディング・ニューロンの背側樹状突起に588nmレーザーを照射した。レーザーシステムの維持のため、カルシウムイメージングとレーザーによる不活性化実験は、21℃にて行われた。
<画像加工>
すべての画像は比較のために全く同じ方法でAdobe PHOTOSHOP(登録商標) 2020 21.2. 2. 又は Final cut Pro X10.5.1を用いて加工した。
すべての画像は比較のために全く同じ方法でAdobe PHOTOSHOP(登録商標) 2020 21.2. 2. 又は Final cut Pro X10.5.1を用いて加工した。
<定量と統計分析>
統計分析はGraphPad Prism 8.4.3(GraphPad Software)を用いた標準的な方法(参考文献16)に従って行われた。行動解析のデータと、Ca2+イメージングのデータのうちKolmogorov-Smirnovの正規性検定を通過したものは,スチューデントのt検定のようなパラメトリック検定を行った。Kolmogorov-Smirnovの正規性検定を通過しなかったものについては、Mann-Whitney U 検定又はKruskal-Wallis 検定のようなノンパラメトリック検定を行った。F検定は図3において条件付け後の変化を解析するために行った。

=結果=
<新規パブロフ条件付け実験系>
記憶形成に有効な条件刺激(Conditioned stimulus;CS)として、多くの昆虫が触覚刺激によるインプットには強い反応を見せることから、“rod removal”を採用した。多くの昆虫はrod removalによって、おそらく引き離されたロッドを再度掴もうとして、振り回すように大きく肢を動かした。ハエも、ロッドが取り除かれると他の昆虫と同様に肢を動かした。5秒間のrod removalを条件刺激として開始した直後に、無条件刺激(Unconditioned stimulus;US)として1Mスクロースをハエの口吻に与えると、ハエ個体は、CSによって肢を動かすと同時に、USによって口吻をさかんに伸展した(図1A)。条件付け前のハエはrod removalのみではほとんど口吻を伸展しないのに対し、30秒ごとにスクロースによるUSと、rod removalによるCSを対にして呈示することを5回くりかえすと、条件反射としてrod removalに対して口吻を伸展するようになった(図1のA-D)。条件付け後、CSによって引き起こされた口吻伸展は、スクロースによるUSによって引き起こされた口吻伸展と区別がつかないほどによく一致していた(図5のG)。一方、CSとUSのタイミングをずらした対照実験プロトコルでは、CSによる口吻の伸展はほとんど見られなかった(図1のD)。
この条件反射はCSとUSの対呈示ののち20分間は観察されたが、時間がたつに従い反応が鈍くなった。より長時間強い条件反射を維持するため、従前のような継続的なrod removalの代わりに、CSを更新するための0.3秒の小休止を4回挟んだ7.6秒のrod removalを与えた。このより強いプロトコルによって条件反射はより長く継続され、CSに誘発された口吻の伸展も条件付け1時間後において統計的に有意であり(図1のE)、いくつかのケースでは3時間以上条件反射が観察された。このより強い方法においても、CS-USを対にして与えたグループの条件反射は対照グループ(CS-US非対付与、CSのみ付与、USのみ付与)と統計的に有意な差があった。
「CS-US非対付与」「USのみ付与」の結果については、どちらもCSとUSを対呈示した場合と同じUSによって同じだけ感作の機会があったが、CSとUSを対呈示した場合に観られたような条件反射は形成されなかった。また、スクロースに対する感度は、条件付けの前後で比較しても統計的に有意な差はみられなかった(図1のF)。つまり、スクロース刺激に比べて、CS刺激による入力が特異的に強められていたことから、この実験において、条件反射はフィーディング・ニューロンの非特異的な感作によって引き起こされたとは言えない。これらの結果から、この条件付けプロトコルを用いれば、ライブイメージング(図3)及び光遺伝学的な神経活動の操作(図4)のために脳を露出した動物個体において、連合学習が可能であることが示された。
<新規パブロフ条件付け実験系>
記憶形成に有効な条件刺激(Conditioned stimulus;CS)として、多くの昆虫が触覚刺激によるインプットには強い反応を見せることから、“rod removal”を採用した。多くの昆虫はrod removalによって、おそらく引き離されたロッドを再度掴もうとして、振り回すように大きく肢を動かした。ハエも、ロッドが取り除かれると他の昆虫と同様に肢を動かした。5秒間のrod removalを条件刺激として開始した直後に、無条件刺激(Unconditioned stimulus;US)として1Mスクロースをハエの口吻に与えると、ハエ個体は、CSによって肢を動かすと同時に、USによって口吻をさかんに伸展した(図1A)。条件付け前のハエはrod removalのみではほとんど口吻を伸展しないのに対し、30秒ごとにスクロースによるUSと、rod removalによるCSを対にして呈示することを5回くりかえすと、条件反射としてrod removalに対して口吻を伸展するようになった(図1のA-D)。条件付け後、CSによって引き起こされた口吻伸展は、スクロースによるUSによって引き起こされた口吻伸展と区別がつかないほどによく一致していた(図5のG)。一方、CSとUSのタイミングをずらした対照実験プロトコルでは、CSによる口吻の伸展はほとんど見られなかった(図1のD)。
この条件反射はCSとUSの対呈示ののち20分間は観察されたが、時間がたつに従い反応が鈍くなった。より長時間強い条件反射を維持するため、従前のような継続的なrod removalの代わりに、CSを更新するための0.3秒の小休止を4回挟んだ7.6秒のrod removalを与えた。このより強いプロトコルによって条件反射はより長く継続され、CSに誘発された口吻の伸展も条件付け1時間後において統計的に有意であり(図1のE)、いくつかのケースでは3時間以上条件反射が観察された。このより強い方法においても、CS-USを対にして与えたグループの条件反射は対照グループ(CS-US非対付与、CSのみ付与、USのみ付与)と統計的に有意な差があった。
「CS-US非対付与」「USのみ付与」の結果については、どちらもCSとUSを対呈示した場合と同じUSによって同じだけ感作の機会があったが、CSとUSを対呈示した場合に観られたような条件反射は形成されなかった。また、スクロースに対する感度は、条件付けの前後で比較しても統計的に有意な差はみられなかった(図1のF)。つまり、スクロース刺激に比べて、CS刺激による入力が特異的に強められていたことから、この実験において、条件反射はフィーディング・ニューロンの非特異的な感作によって引き起こされたとは言えない。これらの結果から、この条件付けプロトコルを用いれば、ライブイメージング(図3)及び光遺伝学的な神経活動の操作(図4)のために脳を露出した動物個体において、連合学習が可能であることが示された。
<記憶変異体における条件反射形成の異常>
この条件付けパラダイムと他の学習アッセイとを比較するため、cAMPシグナルに異常があり、嗅覚valenceアッセイにおいて記憶形成効率の著しい低下が報告されているキイロショウジョウバエの突然変異体dunce(dnc)とrutabaga(rut)を用いて実験した。dnc個体とrut個体両方とも同条件付けののち条件反射の成立効率が低下していた(図2のA)。一方で、条件付け中のCSに誘発される反応とUSに誘発される反応は野生型と同レベルであった(図2のB・C)。

<条件付けによるフィーディング・ニューロンのCSに対する応答性の獲得>
条件付け実験の間を通してフィーディング・ニューロンの活動を観察するため、カルシウムインジケータたんぱく質GCaMP6mの蛍光強度を測定した。フィーディング・ニューロンにおいて、カルシウムインジケータをGMR81E10-GAL4driverによって発現させた。GCaMP6mは口吻にスクロース溶液を与えることで強い反応を示す(図6のE)。条件付け前は、CSによるフィーディング・ニューロンの活動は検出されなかった。条件付け後は、GCaMP6mの蛍光増大として、フィーディング・ニューロンのCSに対する(甘味刺激なし)活動が検出された。条件反射の強さを評価するために、筆者らは次にフィーディング・ニューロンの条件刺激に対する反応と、スクロースを与えた際の反応とを比較した。ハエ個体の口吻を1mM、10mM、100mMのスクロース溶液で刺激し、GCaMPの蛍光を観測した。条件付け20分後の個体で観察される条件刺激に対するフィーディング・ニューロンの活動及び口吻伸展は、ちょうど10mMスクロースによるものと同程度の強さであった。以上の結果から、条件付けによってフィーディング・ニューロンのCSに対する反応性が強化され、その反応性の変化によって摂食行動(口吻伸展)を起こすことができるようになったことがわかる。

<条件付け中にフィーディング・ニューロンの活動を不活性化すると、可塑性的な変化が抑制される>
条件付けの間フィーディング・ニューロンを不活性化し、条件付け後10分ごとにCS単独刺激に対する摂食行動を定量した。条件付け中にフィーディング・ニューロンでハロロドプシンをはたらかせると、USに対する反応は抑制された一方で(図4のB)、CSによる反応は変化がなかった(図7のD)。すべてのCS-US対呈示においてハロロドプシンを介した不活性化を行うと、CSによって口吻伸展が起こるようになるという可塑的な行動変化はほとんど起こらなくなった一方で、不活性化のタイミングをずらした場合にはこれが起きた(図4のA・C)。以上の結果から、フィーディング・ニューロンが条件付け中に活動することが、条件反射の成立には必要であることが示唆された。このことから、フィーディング・ニューロン上のシナプスの変化によって、行動の変化がもたらされていると考えられる。
条件付けの間フィーディング・ニューロンを不活性化し、条件付け後10分ごとにCS単独刺激に対する摂食行動を定量した。条件付け中にフィーディング・ニューロンでハロロドプシンをはたらかせると、USに対する反応は抑制された一方で(図4のB)、CSによる反応は変化がなかった(図7のD)。すべてのCS-US対呈示においてハロロドプシンを介した不活性化を行うと、CSによって口吻伸展が起こるようになるという可塑的な行動変化はほとんど起こらなくなった一方で、不活性化のタイミングをずらした場合にはこれが起きた(図4のA・C)。以上の結果から、フィーディング・ニューロンが条件付け中に活動することが、条件反射の成立には必要であることが示唆された。このことから、フィーディング・ニューロン上のシナプスの変化によって、行動の変化がもたらされていると考えられる。

本発明は、記憶に関与する分子の探索及びそれらに作用する薬剤の探索に利用することができる。
Claims (14)
- ショウジョウバエに対して条件刺激を付与する方法であって、
ショウジョウバエに肢でつかませておいた物体から当該肢を引き離すことを、条件刺激として付与する方法。 - ショウジョウバエに対して餌を与えることを、上記条件刺激と共に条件付けの際に呈示する無条件刺激として付与する、請求項1に記載の方法。
- 機械抄紙の紙よりも目の少ない紙を介して、上記餌の溶液をショウジョウバエに対して与える、請求項2に記載の方法。
- 上記餌の溶液をショウジョウバエに対して与える前に、上記紙に対して、餌の溶液の送液と吸液とを繰り返す、請求項3に記載の方法。
- ショウジョウバエは、消化器官の少なくとも一部が取り除かれている、請求項2に記載の方法。
- 上記のショウジョウバエは、口吻の折り畳みを阻害する構造物が設けられている、請求項2に記載の方法。
- 請求項1に記載の方法によって、上記条件刺激が付与されると無条件刺激に関する応答を示すようになったショウジョウバエを用いて、記憶に関係する試験を行う、試験の方法。
- 観察光源として波長700nm以上780nm以下の範囲内の赤色光を用いる、請求項7に記載の方法。
- 上記ショウジョウバエは脳を露出されたものであり、当該脳における観察の対象領域をガルバノスキャンモードで遠赤外光を照射して探索する、請求項7に記載の方法。
- 請求項1~9の何れか一項に記載の方法を実施する装置であって、
ショウジョウバエが肢でつかむ対象となる物体と、
ショウジョウバエを生きた状態で物理的に固定するための固定部材と、
固定部材と上記物体とを相対的に移動させる駆動手段と、
を備える装置。 - トレハロース及びグルコースから選択される少なくとも一つと、
塩類と、を含んでなる水溶液であり、
浸透圧が225mOsm以上261mOsm以下の範囲内である、昆虫用の脳灌流液。 - ショウジョウバエ用である、請求項11に記載の脳灌流液。
- pHが6.5以上8.0以下の範囲内にあるpH緩衝液である、請求項11に記載の脳灌流液。
- 紙を介して、餌の溶液をショウジョウバエに対して与える方法であって、
上記餌の溶液をショウジョウバエに対して与える前に、上記紙に対して、餌の溶液の送液と吸液とを繰り返す工程を含む、方法。
