기술분야
본 발명은 사스-코로나바이러스-2의 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률이 개선된 제형을 제조하기 위한 조성물, 흡착률을 개선하는 방법, 및 흡착률이 개선된 제형을 제조하는 방법에 관한 것이다.
사스-코로나바이러스-2 (SARS-CoV-2)는 중증 급성 호흡기 증후군 코로나바이러스 2 (Severe Acute Respiratory Syndrome Coronavirus 2) 또는 코비드19 (COVID19)로 불리며, 한국에서는 코로나 19로 명명된다. 사스-코로나바이러스-2는 NA 바이러스이며, 인간대 인간 (Human-to-human) 감염이 확인되었다. 상기 바이러스 감염 후 2~14일간 발열, 호흡곤란, 신장 및 간 손상, 기침, 폐렴 등의 증상이 관찰되며, 아직까지 치료제는 개발되지 못하고 있는 상황이므로 감염을 예방하고, 지역사회에의 확산을 방지하기 위해 백신에 대한 연구가 절실하다.
한편, 재조합 단백질을 함유하는 백신은 면역 반응을 유발하는 보조제(adjuvant)를 포함하는 것이 일반적이다. 특히 알루미늄 염 보조제는 현재 가장 광범위하게 사용되는 보조제이다. 알루미늄 염 보조제는 백신의 면역원성을 높이고, 백신 내 재조합 단백질의 용량 수준을 감소시키고, 체내에서 백신 항원을 방어할 수 있는 항체 생성을 증가시키는 등 백신의 효과를 개선시킨다. 알루미늄 염 보조제가 이러한 효과를 나타내기 위해서는 항원이 반드시 보조제의 표면 상에 흡착되어야 하므로, 알루미늄 염 보조제와의 흡착률을 높이는 기술의 개발이 필요하다.
이에, 사스-코로나바이러스-2에 대한 재조합 단백질 백신에 알루미늄 염 보조제를 사용함에 있어서도, 재조합 항원의 알루미늄 염 보조제에 대한 흡착률을 높인 백신 제형의 개발이 필요한 실정이다.
이에, 사스-코로나바이러스-2에 대한 재조합 단백질 백신에 알루미늄 염 보조제를 사용함에 있어서도, 재조합 항원의 알루미늄 염 보조제에 대한 흡착률을 높인 백신 제형의 개발이 필요한 실정이다.
본 발명자들은 사스-코로나바이러스-2의 재조합 단백질 백신을 제조함에 있어서 알루미늄 염 보조제(Aluminum adjuvant)와의 흡착률을 높여 항원의 안정성 및 면역원성을 향상시키기 위한 최적의 조건을 확인하였다. 보다 구체적으로, NaCl 농도와 염산염(phosphate) 농도 사이에 교호작용(interaction)이 존재하여, NaCl 농도가 높고 인산염 농도가 낮을수록 흡착률이 향상되는 것을 확인하였으며, 이에 따라 NaCl 농도 및 인산염 농도를 조절하여 흡착률을 최대화할 수 있는 조건을 확인하여 발명을 완성하였다.
일 예는, 사스-코로나바이러스-2의 항원성 폴리펩티드; 알루미늄 염 보조제; 및 50 mM 내지 500 mM 의 염화나트륨(NaCl) 및 0.001 mM 내지 50 mM 의 인산염 (phosphate)을 포함하는 완충액을 포함하는, 사스-코로나바이러스-2의 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률이 개선된 제형을 제조하기 위한 조성물을 제공한다.
다른 예는, 50 mM 내지 500 mM 의 염화나트륨(NaCl) 및 0.001 mM 내지 50 mM 의 인산염 (phosphate)을 포함하는 완충액에서 사스-코로나바이러스-2의 항원성 폴리펩티드 및 알루미늄 염 보조제를 흡착시키는 단계를 포함하는, 상기 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률을 개선하는 방법을 제공한다.
다른 예는, 50 mM 내지 500 mM 의 염화나트륨(NaCl) 및 0.001 mM 내지 50 mM 의 인산염 (phosphate)을 포함하는 완충액에서 사스-코로나바이러스-2의 항원성 폴리펩티드 및 알루미늄 염 보조제를 흡착시키는 단계를 포함하는, 상기 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률이 개선된 제형을 제조하는 방법을 제공한다.
본 발명에 따른, 알루미늄 염 보조제와의 흡착률이 높은 사스-코로나바이러스-2 백신 제형은 체내에서 높은 면역반응을 유도하여 효과적으로 백신 항원을 방어할 수 있는 항체 생성이 가능한 효과가 있다.
도 1은 반응(흡착률)에 대한 주효과 인자들(NaCl 농도, 인산염 농도)의 영향이 도표화된 예측 분석도(Prediction Profiler)이다. 주효과 인자들은 통계적 분석을 통해 NaCl 농도와 인산염(phosphate) 농도가 선정되었다. 즉, NaCl 농도와 인산염 농도가 반응(흡착률)에 큰 영향을 나타낸다.
도 2는 주효과로 선정된 인자(NaCl 농도, 인산염 농도)들의 교호작용(interaction)을 나타낸 도표이다. 교호작용이 있을 경우에는 상대인자의 각 수준에 대응되는 반응(흡착률)의 변화가 교차하거나 평행이 아니다.
도 3은 RBDP2ex1의 제형 실험 디자인 설계를 기반으로 진행된 실험 결과를 바탕으로 도출된 반응(흡착률)의 등고선도(Contour profiler)이다. 흰색영역은 통계적으로 분석된 반응(흡착률)이 90% 이상으로 예상되는 구간이다.

알루미늄 염 보조제는 백신 항원에 대한 면역 반응을 자극하는 것으로 당업계에 통상적으로 사용되는 면역보조제이다. 백신 항원이 알루미늄 염 보조제 상에 물리적으로 흡착되면, 주사 부위에서 데포(depot)로 작용하여 항원이 투여 후 서서히 방출되도록 하거나, 항원 제시 세포에 의한 항원의 보다 효율적인 유입을 허용하거나, 또는 Th2 면역 반응을 유도하는 등의 기전을 통하여 면역 반응을 강화시키는 것으로 알려져 있으나, 그 작용기전이 완전히 밝혀지지는 않았으며 이에 대한 연구가 활발하게 진행되고 있다.
본 발명에서 알루미늄 염 보조제는 알루미늄 하이드록사이드 (Al(OH)3), 알루미늄 옥시하이드록사이드(AlOOH), 알루미늄 옥시하이드록시 포스페이트(AlOOHPO4), 알루미늄 포스페이트 (AlPO4), 알루미늄 하이드록시포스페이트(AlOHPO4 또는 Al(OH)x(PO4)y), 무정형 알루미늄 하이드록시포스페이트 설페이트(AlHO9PS-3 또는 AAHS) 또는 소위 "알룸" (AlK(SO4)2)의 형태이거나, 이들 중 2종 이상의 임의의 조합물일 수 있으나, 이에 제한되지 않는다.
알루미늄 옥시하이드록사이드(AlOOH)는 부분적으로 결정체이며, 알루미늄 하이드록사이드 (Al(OH)3)과는 적외선 (IR) 분광 스펙트럼에서 특히 1070 cm-1에서 흡수 밴드가 존재하고 3090 내지 3100 cm-1에서 강한 숄더를 보이는 점에서 구분할 수 있다. 알루미늄 옥시하이드록시 포스페이트(AlOOHPO4)는 알루미늄 옥시하이드록사이드(AlOOH)에 포스페이트 이온을 첨가함으로써 수득할 수 있다. 그러나 당업계에서는 알루미늄 하이드록사이드 (Al(OH)3), 알루미늄 옥시하이드록사이드(AlOOH), 또는 알루미늄 옥시하이드록시 포스페이트(AlOOHPO4) 를 모두 "알루미늄 하이드록사이드" 보조제로 통칭하기도 한다.
알루미늄 하이드록시포스페이트(AlOHPO4 또는 Al(OH)x(PO4)y) 또는 알루미늄 하이드록시포스페이트 설페이트는 보통 침전에 의해 수득할 수 있으며, 침전시키는 동안의 반응 조건 및 농도는 염내의 하이드록실기에 대한 포스페이트 치환 정도에 영향을 준다. 이들은 보통 비결정질이고, 일반적으로 PO4/Al의 몰비가 0.3 내지 1.2 이며 하이드록실기가 존재하는가에 따라 알루미늄 포스페이트 (AlPO4)와 구분할 수 있다. 예를 들면, 200℃에서 가열하였을 때 3164 ㎝-1에서 IR 스펙트럼 밴드가 있는 것은 하이드록실 구조가 존재하는 것을 의미한다. 그러나 당업계에서는 알루미늄 포스페이트 (AlPO4), 알루미늄 하이드록시포스페이트(AlOHPO4 또는 Al(OH)x(PO4)y), 무정형 알루미늄 하이드록시포스페이트 설페이트(AlHO9PS-3 또는 AAHS) 를 모두 "알루미늄 포스페이트" 보조제로 통칭하기도 한다.
일예로, 본원에서 알루미늄 염 보조제는 최종 백신 제형 내 Al3+의 농도가 5 mg/ml 내지 15 mg/ml 이 되도록 사용할 수 있으나, 이에 제한되는 것은 아니다.
본원에서 알루미늄 염 보조제는 염화나트륨(NaCl) 및 인산염(phosphate)을 포함하는 완충액 내에 현탁될 수 있다.
특히, 본 발명에서는 NaCl 농도와 염산염(phosphate) 농도 사이에 교호작용(interaction)이 존재하여, NaCl 농도가 높고 인산염 농도가 낮을수록 흡착률이 향상되는 것을 확인하였으며, 이에 따라 NaCl 농도 및 인산염 농도를 조절하여 흡착률을 최대화할 수 있는 조건을 확인하였다.
구체적으로, 완충액은 50 - 500 mM, 예를 들어, 75 - 400 mM, 100 - 300 mM, 250 - 350 mM, 280 - 320 mM, 290 - 310 mM, 또는 300 mM 의 염화나트륨(NaCl)을 포함할 수 있다.
또한, 완충액은 0.001 - 50 mM, 예를 들어, 0.01 - 35 mM, 0.1 - 20 mM, 0.001 - 10 mM, 0.001 - 0.2 mM, 0.03 - 0.07 mM, 또는 0.05mM 의 인산염 (phosphate)을 포함할 수 있다.
염화나트륨(NaCl) 의 상기 정의된 농도 범위에 미달하는 경우 흡착률이 기준치에 미달하는 문제가 발생할 수 있고, 상기 농도 범위를 초과하는 경우 제형 안정성에 문제가 발생할 수 있다. 또한, 인산염(phosphate) 의 상기 정의된 농도 범위에 미달하는 경우 pH 조절 능력 저하로 인한 pH 조절 실패의 문제가 발생할 수 있고, 상기 농도 범위를 초과하는 경우 흡착률이 기준치에 미달하는 문제가 발생할 수 있다.
본원에서 사스-코로나바이러스-2의 항원성 폴리펩티드는 사스-코로나바이러스-2의 스파이크 단백질 (spike protein)의 리셉터 결합 도메인(RBD, receptor-binding domain)을 포함하는 폴리펩티드 및 사스-코로나바이러스-2의 N 단백질을 포함하는 폴리펩티드 중에서 선택되는 1종 이상일 수 있다.
사스-코로나바이러스-2는 ACE2 (Angiotensin Converting Enzyme2) 수용체를 통해 숙주세포의 표면에 강하게 부착하는 것으로 알려져 있으며, SARS-CoV-2의 스파이크 단백질의 RBD 는 ACE2 수용체와 결합하는데 이용되는 것으로 알려져 있다. RBD 는 스파이크 단백질의 전장 폴리펩타이드 서열의 331-524번째 아미노산으로 구성된다. 본원에서는 "사스-코로나바이러스-2의 스파이크 단백질 RBD를 포함하는 폴리펩티드"란, RBD (스파이크 단백질의 331-524번째 아미노산)로 구성되거나, RBD 를 포함하면서 C-말단과 N-말단 방향으로 적어도 1, 2, 3, 4, 또는 5개 이상의 아미노산을 더 포함하는 폴리펩티드를 말한다. 예를 들어, 스파이크 단백질의 328-531번째, 321-545번째, 321-591번째, 또는 321-537번째 아미노산으로 구성된 폴리펩티드가 본원의 항원성 폴리펩티드 범주에 포함될 수 있다. 또한, 전술한 서열들과 적어도 75%, 80%, 85%, 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99% 이상, 또는 100%의 동일한 아미노산 서열을 포함하거나 이로 이루어진 폴리펩티드가 본원의 항원성 폴리펩티드 범주에 포함될 수 있다.
사스-코로나바이러스-2의 N 단백질은 사스-코로나바이러스-2 의 뉴클레오캡시드 단백질(nucleocapsid) 을 말하며, 이와 기능적 동일성을 갖는 단편, 및/또는 유사체를 포함할 수 있다. 상기 단편 및/또는 유사체는 N 단백질의 서열과 적어도 75%, 80%, 85%, 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99%, 또는 100% 서열 동일성을 가질 수 있으며, 기능적 동일성을 가질 수 있다. 기능적 동일성을 갖는다는 의미는 상기 N 단백질이 본 발명에서 목적하는 바와 유사한 목적, 효과를 달성할 수 있다는 것을 의미한다. N 단백질은 세포성 면역을 유도할 수 있으며, RBD 함유 폴리펩티드과 함께 사용하여 증가된 보호면역원성을 유도할 수 있다. N 단백질은 안정성이 높고 상당한 면역원성 유도능을 보이며, 세포독성 림프구(Cytotoxic T lymphocytes)의 유도와 관련이 있으므로 이를 이용한 세포성 면역은 바이러스를 감염 초기에 효과적으로 방어할 수 있다. 또한, N 단백질의 투여로 RBD-특이적 IgG 역가의 높은 증가를 보일 수 있다. 따라서, RBD 함유 폴리펩티드와 N 단백질의 동시 투여로, 향상된 세포성 면역원성 증가를 기대할 수 있다
RBD 함유 폴리펩티드와 N 단백질을 포함하는 폴리펩티드의 혼합 비율은 1: 1 내지 500의 중량비, 바람직하게 1: 1 내지 400, 바람직하게 1: 1 내지 300, 바람직하게 1: 1 내지 200, 바람직하게 1: 1 내지 100, 바람직하게 1: 1 내지 80의 중량비, 바람직하게 1:30 내지 50일 수 있다. 상기 비율로 포함될 때 항체와의 결합력이 우수하거나, 높은 중화 항체가가 확인될 수 있다.
선택적으로, 항원성 폴리펩티드는 N-말단 또는 C-말단에 시그널 펩티드 및 Tetanus 독소의 P2 도메인 중 1종 이상을 더욱 포함할 수 있다. 시그널 펩티드는 숙주 세포에서 단백질을 분비 경로로 지시하는, 새로 합성된 폴리펩타이드 사슬의 N-말단에 존재하는 짧은 펩티드 (일반적으로 5-30개 폴리펩타이드 길이를 갖지만, 이에 제한되지 않는다)를 의미한다. 본원에서 언급된 시그널 펩티드는 단백질 분비 과정에서 제거된다. 시그널 펩티드는 이종 유래일 수 있으며, 예를 들어 murine phosphatase 시그널 펩타이드 서열, honeybee melittin 시그널 펩타이드 서열, 인간 알부민 시그널 펩타이드 서열 등을 예시할 수 있다. Tetanus 독소의 P2 도메인은 더 향상된 면역 증강 효과를 나타내기 위하여 본원의 항원성 폴리펩티드에 연결될 수 있다.
시그널 펩티드 및/또는 Tetanus 독소의 P2 도메인은 항원성 폴리펩티드에 직접 연결되거나 링커를 통해 연결될 수 있다. 링커는 G(Gly, 글라이신), S(Ser, 세린), 및 A(Ala, 알라닌) 중 하나 이상을 포함하는 펩티드 링커일 수 있고, 바람직하게는 Gly-Ser-Gly-Ser-Gly (GSGSG), Gly-Ser-Ser-Gly (GSSG), Gly-Ser-Gly-Gly-Ser (GSGGS), Gly-Ser-Gly-Ser (GSGS), 및 Gly-Ser-Gly-Ser-Ser-Gly (GSGSSG)로 이루어진 군에서 선택된 어느 하나 이상의 펩티드 링커일 수 있으나, 이에 제한되지 않는다.
항원성 폴리펩티드를 코딩하는 폴리뉴클레오티드는 숙주에 대하여 코돈 최적화되는 것이 바람직하다.
일 구현예로, 본원의 항원성 폴리펩티드는 적합한 발현 벡터를 사용하여, 원핵 또는 진핵 발현 시스템에서 클로닝 및 발현에 의해 제조할 수 있다. 예를 들어, 당업계 공지된 배큘로바이러스 발현 시스템(BEVS), CHO 세포 발현 시스템, 또는 E. coli 발현 시스템을 사용할 수 있으나, 이에 제한되지 않는다. 벡터는 임의의 적절한 유형일 수 있고, 비제한적으로 파아지, 바이러스, 플라스미드, 파지미드(phagemid), 코스미드(cosmid), 백미드(bacmid) 등을 포함할 수 있다. '숙주세포'라는 용어는 배양액에서 성장할 수 있고 목적하는 단백질 재조합 산물 단백질을 발현할 수 있는 세포를 지칭한다. 바람직한 숙주 세포는 진핵 숙주 세포일 수 있고, 예를 들어, 곤충 세포로 배큘로바이러스 발현시스템을 이용하는 Sf9, Sf21과 같은 Spodopterafrugiperda (Sf) 세포, 하이 파이브 (Hi-5) 세포와 같은 Trichoplusiani 세포 및 Drosophila S2 세포들을 포함할 수 있고, 포유류 세포로 중국 햄스터 난소(CHO) 세포, 예를 들어 CHO K1, CHO pro3-, CHO DG44, CHO P12 를 포함할 수 있다. 그 외에도, 진핵 숙주 세포의 예로 효모, 조류, 식물, 꼬마선충(또는 선충) 등을 포함할 수 있다. 원핵 숙주 세포들은, 예를 들어, 대장균(E. coli, B. subtilis), 살모넬라티피균 (Salmonella typhi) 및 마이코박테리아와 같은 박테리아 세포를 포함할 수 있다. 벡터의 도입 후, 상기 숙주 세포를 일반배지 또는 선택성 배지(벡터 함유 세포의 성장을 위해 선택한다)에서 증식시키면, 상기 클로닝된 유전자 서열(들)의 발현 결과 목적하는 단백질이 생산된다. 상기 재조합 단백질의 정제를 상기 목적으로 공지된 방법들 중 어느 하나, 즉 추출, 침전, 크로마토그래피, 한외여과, 전기영동 등을 수반하는 임의의 통상적인 과정에 의해 수행할 수 있다.
본원에서 항원성 폴리펩티드의 농도는 목적에 따라 당업자에 의해 적절히 선택될 수 있다. 예를 들어, 최종 백신 제형 내 항원성 폴리펩티드의 농도가 10 내지 200 ㎍/ml, 20 내지 150 ㎍/ml, 25 내지 100 ㎍/ml, 25 내지 55 ㎍/ml, 또는 30 내지 50 ㎍/ml이 되도록 사용할 수 있으나, 이에 제한되는 것은 아니다.
본원의 조성물은, 보조제, 완충액, 및 항원성 폴리펩티드 외에도, 선택적으로 탄산나트륨(Sodium Carbonate)을 더욱 포함할 수 있다. 탄산나트륨은 안정성 향상 및 면역원성 증가의 목적으로 포함할 수 있으며, 예를 들어, 0.001 내지 50 mM, 0.01 내지 25mM, 0.1 내지 20mM 또는 20mM의 농도로 포함할 수 있다. 상기 정의된 농도 범위에 미달하는 경우 pH 조절에 실패하는 문제가 발생할 수 있고, 상기 농도 범위를 초과하는 경우 흡착률 문제 또는 항원의 안정성을 보증할 수 없는 문제가 발생할 수 있다.
본원의 조성물은 pH 가 6.0 내지 8.0, 예를 들어, pH 6.0 내지 7.8, 또는 pH 6.5 내지 7.6, 구체적으로 pH 7.0 일 수 있다. 상기 정의된 농도 범위에 미달하는 경우 항원 단백질의 변성 및 안정성의 문제가 발생할 수 있고, 상기 농도 범위를 초과하는 경우에도 역시 항원 단백질의 변성, 안정성 문제 및 높은 pH로 인한 주사 통증이 발생할 수 있다.
본 발명은 또한 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률을 개선하는 방법, 및 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률이 개선된 제형을 제조하는 방법을 제공하는데, 이 방법들은, 50 mM 내지 500 mM 의 염화나트륨(NaCl) 및 0.001 mM 내지 20 mM 의 인산염 (phosphate)을 포함하는 완충액에서 사스-코로나바이러스-2의 항원성 폴리펩티드 및 알루미늄 염 보조제를 흡착시키는 단계를 포함한다.
본원에서 흡착률의 개선이란, 50 mM 내지 500 mM 의 염화나트륨(NaCl) 및 0.001 mM 내지 20 mM 의 인산염 (phosphate)을 포함하는 완충액을 사용하지 않은 경우의 흡착률과 비교하여, 또는 통상적으로 알려진 흡착률과 비교하여 흡착률이 증가된 것을 말한다. 흡착률은 90% 이상인 것이 바람직하며, 예를 들어 90% 이상, 91% 이상, 92% 이상, 93% 이상, 94% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 또는 99% 이상일 수 있다.
본원의 방법들에서, 완충액, 항원성 폴리펩티드, 및 알루미늄 염 보조제에 관한 내용은 전술한 바와 동일하게 적용된다.
본원에서 흡착 단계는, 상기 완충액, 항원성 폴리펩티드, 및 알루미늄 염 보조제를 혼합함으로써 수행될 수 있으며, 상기 3가지 성분은 임의의 순서로 혼합될 수 있다. 흡착 단계의 pH 는 pH 6.0 내지 8.0일 수 있고, 온도는 2℃ 내지 8℃일 수 있다. 혼합을 통해 항원을 보조제와 접촉하게 한 후, 1시간 이상, 2시간 이상, 4시간 이상, 12시간 이상, 또는 24시간 또는 그 이상 방치하여 흡착을 생성할 수 있으며, 유리하게는 교반과 함께 수행할 수 있다.
이와 같은 방법을 통해, 본원의 일예는 사스-코로나바이러스-2의 항원성 폴리펩티드 및 알루미늄 염 보조제를 포함하며, 사스-코로나바이러스-2의 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률이 90% 이상, 91% 이상, 92% 이상, 93% 이상, 94% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 또는 99% 이상인 백신 제형을 제공한다.
백신 조성물은 약학적으로 허용가능한 담체, 희석제 및/또는 부형제를 더욱 포함할 수 있다. 이에 제한되는 것은 아니지만 예를 들면, 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 백신 조성물은 상기 성분들 이외에 TWEEN쪠, 폴리에틸렌 글리콜 (PEG) 등과 같은 비-이온성 계면 활성제, 아스코르브 산을 포함하는 항산화제, 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함하여 사용될 수 있다. 본 발명에서 상기 백신은, 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화 함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다. 본 발명에서 상기 백신의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다. 한편, 본 발명에 따른 백신의 투여량은 바람직하게는 도즈 당 1 ~ 500 ug 일 수 있다. 본 발명의 일 구체 예에서는 상기 재조합 단백질을 유효성분으로 포함하는 백신은 정맥내주사, 근육 내주사, 피하내주사, 경피전달 또는 기도흡입으로 체내에 투여될 수 있으나, 이에 제한되는 것은 아니다.
이하 본 발명을 다음의 실시예에 의하여 보다 구체적으로 설명하고자 한다. 그러나 이들은 본 발명을 예시하기 위한 것일 뿐이며, 본 발명의 범위가 이들 실시예에 의하여 제한되는 것은 아니다. 아래 기재된 실시예들은 발명의 본질적인 요지를 벗어나지 않는 범위에서 변형될 수 있음은 당업자들에게 있어 자명하다.
실시예 1. 통계분석과 모델링
pH, NaCl 농도, NaPi 농도, 온도, 항원비율의 5가지 주효과가 항원의 흡착률에 미치는 영향을 알아보기 위해 DoE 실험을 계획하였다. 실험의 설계와 통계적 분석은 통계프로그램 (JMP 13.0.0, SAS Institute Inc.)을 사용하였다. 실험의 디자인은 주효과, 주효과의 곡률, 주효과 간의 교호작용(interaction)을 확인할 수 있도록 표면반응분석법으로 설계하였다. 실험결과에 대한 분석은 p-value가 0.05 미만인 인자만을 남기도록 하는 backward elimination을 선행하였고 유의미한 인자를 이용하여 모델을 구성하였다. 총 48회의 실험으로 이루어진 데이터를 모아 통계 처리하여, 도 1 내지 도 3에 나타내었다.
실시예 2. RBDP2ex1의 제조
RBDP2ex1는 사스-코로나바이러스-2 (코로나 19 라고도 함) 의 스파이크 단백질 (spike protein)의 리셉터 결합 도메인(RBD, receptor-binding domain)을 포함하면서, 스파이크 단백질의 전장 서열의 321-545 번째 아미노산으로 구성된 폴리펩티드; 상기 폴리펩티드 영역의 N-말단에 직접 연결된, 인간 알부민 시그널 펩티드; 및 상기 폴리펩티드 영역의 C-말단에 링커(GSGSG)를 통해 연결된, Tetanus Toxoid Epitope P2 도메인 영역으로 구성된, 사스-코로나바이러스-2의 재조합 항원성 폴리펩티드이다 (국내특허출원 제10-2021-0055290호 참조; 상기 문헌은 전체가 본원에 참조로서 통합됨).
상기 재조합 항원성 폴리펩티드의 아미노산 서열을 서열번호 1에 나타내었고, 이를 배큘로바이러스 발현시스템에서 발현하기 위하여 코돈 최적화한, 상기 재조합 항원성 폴리펩티드를 코딩하는 폴리뉴클레오티드의 서열을 서열번호 2에 나타내었고, CHO 세포 발현시스템에서 발현하기 위하여 코돈 최적화한, 상기 재조합 항원성 폴리펩티드를 코딩하는 폴리뉴클레오티드의 서열을 서열번호 3에 나타내었다.
본 실시예에서는, RBDP2ex1 를 하이 파이브 (Hi-5) 세포 발현시스템을 이용하여 재조합적으로 생산하였다. 구체적으로, 서열번호 2의 유전자 컨스트럭트를 준비하고 전이 벡터 pFastBac vector에 상기 준비된 컨스트럭트 유전자를 삽입하여 클로닝하고, 유전자 서열을 분석하였다. 제조된 플라스미드를 bacmid 제조용 E. coli에 형질전환 (Transformation)하여 재조합백미드 (Recombinant bacmid)를 제조하고 유전자 서열을 분석하였다. 재조합백미드를 단층으로 배양된 Sf9 세포에 접종하여 형질감염(Transfection)하고 재조합배큘로바이러스 (P0)를 제조하여 플라그시험법으로 정량하였다. 배양된 Hi-5 세포에 재조합배큘로바이러스를 감염시켜 P1 바이러스를 확보하고, 상등액에서 생산된 재조합 단백질을 확인하였다. 상기 P1 바이러스를 Hi-5 세포에 감염시켜 생산된 재조합 단백질을 회수하였다. 회수한 재조합 단백질을 필터로 여과하고, 적절한 크로마토그래피법을 이용하여 정제하였다.
실시예 3. N 단백질의 제조
N 단백질은 사스-코로나바이러스-2 의 뉴클레오캡시드 단백질(nucleocapsid) 을 말한다. 상기 N 단백질의 아미노산 서열을 서열번호 4에 나타내었고, 상기 N 단백질의 N-말단에 인간 알부민 시그널 펩타이드 서열을 연결한 것을 서열번호 5에 나타내었고, 이를 배큘로바이러스 발현시스템에서 발현하기 위하여 코돈 최적화한 폴리뉴클레오티드의 서열을 서열번호 6에 나타내었고, CHO 세포 발현시스템에서 발현하기 위하여 코돈 최적화한 폴리뉴클레오티드의 서열을 서열번호 7에 나타내었다.
본 실시예에서는, N 단백질을 하이 파이브 (Hi-5) 발현시스템을 이용하여 재조합적으로 산하였다. 구체적으로, 서열번호 6의 유전자 컨스트럭트를 준비하고 전이 벡터 pFastBac vector에 상기 준비된 컨스트럭트 유전자를 삽입하여 클로닝하고, 유전자 서열을 분석하였다. 제조된 플라스미드를 bacmid 제조용 E. coli에 형질전환 (Transformation)하여 재조합백미드 (Recombinant bacmid)를 제조하고 유전자 서열을 분석하였다. 재조합백미드를 단층으로 배양된 Sf9 세포에 접종하여 형질감염(Transfection)하고 재조합배큘로바이러스 (P0)를 제조하여 플라그시험법으로 정량하였다. 배양된 Hi-5 세포에 재조합배큘로바이러스를 감염시켜 P1 바이러스를 확보하고, 상등액에서 생산된 재조합 단백질을 확인하였다. 상기 P1 바이러스를 Hi-5 세포에 감염시켜 생산된 재조합 단백질을 회수하였다. 회수한 재조합 단백질을 필터로 여과하고, 적절한 크로마토그래피법을 이용하여 정제하였다.
실시예 4. 제형 최적화 실험

실시예 2 에서 제조한 항원성 폴리펩티드 RBDP2ex1 및 알루미늄 면역 보조제를, 아래 표 1에 기재된 pH, 인산염(phosphate) 농도(mM), 및 NaCl 농도(mM) 조건으로 혼합하여 제형을 완성하였다. 이때, 2.0 % Alhydrogel 제품을 사용하여 항원 원액과 보조제의 비율이 1:10이 되도록 하였다. 그 후, 흡착도를 BCA 측정법을 이용하여 측정하였다. 이를 위하여, 알루미늄 염 보조제와 흡착된 원액을 12,000 RPM, 5분간 원심분리를 하였다. 원심분리를 통해 흡착원액으로부터 알루미늄 염 보조제와 흡착된 단백질 항원이 분리가 되며, 층분리가 일어난 용액의 상층액을 취하여 단백함량을 측정해 흡착률을 분석할 수 있다. 실시예 1의 DoE 결과, 그리고 표 1 및 도 4 에 나타난 결과를 종합하여 알 수 있는 바와 같이, 흡착률은 NaCl농도가 높고, 인산염 농도가 낮을수록 높은 결과를 나타냈다. 결과적으로 NaCl과 인산염 농도 사이에 교호작용(interaction)이 확인되어, NaCl을 높이고 인산염 농도를 낮추는 방향으로 최적 조건을 확립하였다.

4-2. N protein 제형의 제형 최적화
실시예 2 에서 제조한 N 단백질 및 알루미늄 면역 보조제를, 아래 표 2에 기재된 pH, 인산염(phosphate) 농도(mM), NaCl 농도(mM) 및 조건으로 혼합하여 제형을 완성하였다. 이때, 2.0 % Alhydrogel 제품을 사용하여 항원 원액과 보조제의 비율이 1:10이 되도록 하였다. 그 후, 흡착도를 BCA 측정법을 이용하여 측정하였다. 알루미늄 염 보조제와 흡착된 원액을 12,000 RPM, 5분간 원심분리를 하였다. 원심분리를 통해 흡착원액으로부터 알루미늄 염 보조제와 흡착된 단백질 항원이 분리가 되며, 층분리가 일어난 용액의 상층액을 취하여 단백함량을 측정해 흡착률을 분석할 수 있다. 실시예 1의 DoE 결과, 그리고 표 2 및 도 5에 나타난 결과를 종합하여 알 수 있는 바와 같이, 흡착률은 NaCl농도가 높고, 인산염 농도가 낮을수록 높은 결과를 나타냈다. 결과적으로 NaCl과 인산염 농도 사이에 교호작용(interaction)이 확인되어, NaCl을 높이고 인산염 농도를 낮추는 방향으로 최적 조건을 확립하였다.

4-3. 결과 종합
RBDP2ex1과 N protein은 백신 제형을 제조할 때 함께 사용되므로, 실시예 1의 DoE 결과 및 실시예 3-1 및 3-2의 실험 결과를 종합하여, 0.02 - 0.05mM 인산염, 300mM NaCl, 및 pH 6.8 - 7.0이라는 최적의 조건을 설정하였다.
실시예 5. 제형 안정성 실험 결과
재조합 코로나 바이러스 백신 완제의약품은 6 개월간 장기 안정성 시험을 실시하였으며 2~8 ℃의 보관조건에서 6개월 시점까지 성상, pH, 실용량, 불용성 이물, 알루미늄 함량, 항원함량, 확인, 단백함량, 흡착도, 무균, 엔도톡신, 이상독성부정시험을 통해 시험항목이 기준에 적합함을 확인하였다. 이를 통해 6 개월 보관 기간 동안 완제의약품의 함량, 안정성에 대한 품질을 보증하였다.

이상의 설명으로부터, 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로서 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (16)
- 사스-코로나바이러스-2의 항원성 폴리펩티드;알루미늄 염 보조제; 및50 mM 내지 500 mM 의 염화나트륨(NaCl) 및 0.001 mM 내지 50 mM 의 인산염 (phosphate)을 포함하는 완충액을 포함하는,사스-코로나바이러스-2의 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률이 개선된 제형을 제조하기 위한 조성물.
- 제1항에 있어서,사스-코로나바이러스-2의 항원성 폴리펩티드가, 사스-코로나바이러스-2의 스파이크 단백질 (spike protein)의 리셉터 결합 도메인(RBD, receptor-binding domain)을 포함하는 폴리펩티드, 및 사스-코로나바이러스-2의 N 단백질을 포함하는 폴리펩티드 중에서 선택되는 1종 이상인, 조성물.
- 제2항에 있어서,상기 항원성 폴리펩티드는 N-말단 또는 C-말단에 시그널 펩티드 및 Tetanus 독소의 P2 도메인 중 1종 이상을 더욱 포함하는, 조성물.
- 제2항에 있어서,상기 항원성 폴리펩티드를 코딩하는 폴리뉴클레오티드는 숙주에 대하여 코돈 최적화된 것인, 조성물.
- 제1항에 있어서,알루미늄 염 보조제는 알루미늄 하이드록사이드 (Al(OH)3), 알루미늄 옥시하이드록사이드(AlOOH), 알루미늄 옥시하이드록시 포스페이트(AlOOHPO4), 알루미늄 포스페이트 (AlPO4), 알루미늄 하이드록시포스페이트(AlOHPO4), 무정형 알루미늄 하이드록시포스페이트 설페이트(AAHS) 또는 알룸 (AlK(SO4)2) 중에서 선택되는 1종 이상인, 조성물.
- 제1항에 있어서,상기 조성물의 pH 가 6.0 내지 8.0인, 조성물.
- 제1항에 있어서,탄산나트륨(Sodium Carbonate)을 더욱 포함하는, 조성물.
- 50 mM 내지 500 mM 의 염화나트륨(NaCl) 및 0.001 mM 내지 50 mM 의 인산염 (phosphate)을 포함하는 완충액에서 사스-코로나바이러스-2의 항원성 폴리펩티드 및 알루미늄 염 보조제를 흡착시키는 단계를 포함하는,상기 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률을 개선하는 방법.
- 50 mM 내지 500 mM 의 염화나트륨(NaCl) 및 0.001 mM 내지 50 mM 의 인산염 (phosphate)을 포함하는 완충액에서 사스-코로나바이러스-2의 항원성 폴리펩티드 및 알루미늄 염 보조제를 흡착시키는 단계를 포함하는,상기 항원성 폴리펩티드와 알루미늄 염 보조제와의 흡착률이 개선된 제형을 제조하는 방법.
- 제8항 또는 제9항에 있어서,사스-코로나바이러스-2의 항원성 폴리펩티드가, 사스-코로나바이러스-2의 스파이크 단백질 (spike protein)의 리셉터 결합 도메인(RBD, receptor-binding domain)을 포함하는 폴리펩티드, 및 사스-코로나바이러스-2의 N 단백질을 포함하는 폴리펩티드 중에서 선택되는 1종 이상인, 방법.
