1.方法
ペプチド
WT134-51(WAPVLDFAPPGASAYGSL)(配列番号2)、WT137-45(VLDFAPPGA)(配列番号5)またはWT140-48(FAPPGASAY)(配列番号6)を使用した。NetMHC-4.0(http://www.cbs.dtu.dk/services/NetMHC-4.0)での解析によると、WT137-45はHLA-A*02:01、HLA-A*02:06、およびHLA-A*02:07に、WT140-48はHLA-A*26:01およびHLA-A*26:03に対する親和性が高かった。
HLA-A*02:01(5例)、HLA-A*02:06(7例)、HLA-A*02:07(2例)またはHLA-A*26:01(2例)陽性の16名のがん患者(口腔咽頭癌、肺癌、食道癌、胃癌、乳癌、膵癌、卵巣癌、胆嚢癌、胆管癌、前立腺癌、子宮癌)の凍結保存してある末梢血単核細胞(PBMC)を溶解し、Ficoll‐Plaque Premiumを用いた密度勾配遠心により細胞を単離し、無血清AIM-V培地にサスペンドした。10cmのPrimariaセルカルチャーディッシュに細胞を播種し、37℃5%CO2インキュベーター内で30分以上放置することにより、細胞をプラスチックに付着する単球と非付着性(NAD)細胞とに分離し、それぞれを凍結保存した。また、付着単球を顆粒球マクロファージコロニー刺激因子GM-CSF(50ng/ml)およびIL-4(50ng/ml)を含む無血清AIM-V培地で37℃5%CO2インキュベーター内で5日間培養することにより未成熟樹状細胞(imDC)を作製し、凍結保存した。
凍結保存したimDC(1×105細胞)を解凍し、5ng/mlのGM-CSF、50ng/mlのプロスタグランジンE2、および10μg/mlのOK-432の存在下で100μgのWT134-51を添加したのち、37℃5%CO2で24時間インキュベートすることにより、WT1ペプチドパルス成熟樹状細胞(mDC/WT134-51)を作製した。一方、凍結保存したimDC(1×105細胞)を解凍し、5ng/mlのGM-CSF、50ng/mlのプロスタグランジンE2、および10μg/mlのOK-432の存在下で37℃5%CO2で24時間インキュベートすることにより、WT1ペプチド非パルス成熟樹状細胞(mDC)を作製した。また、imDC(1×104細胞)に25μgのWT134-51、WT137-45、またはWT140-48を添加したのち、37℃5%CO2で24時間インキュベートすることにより、WT1ペプチドパルス未成熟樹状細胞(imDC/WT134-51、imDC/WT137-45またはimDC/WT140-48)を作製した。
凍結保存したNAD細胞を解凍し、1%非必須アミノ酸、1mMピルビン酸ナトリウム、50μM2-メルカプトエタノール、10%非働化ウシ胎仔血清、20U/mlのIL-2、および20ng/mlのIL-7を含むRPMI-1640培地にサスペンドし、24穴プレートにて37℃5%CO2インキュベーター内で24時間培養した。その後、mDC/WT134-51(1×105細胞)とNAD細胞(1×106細胞)とを、1%非必須アミノ酸、1mMピルビン酸ナトリウム、50μM2‐メルカプトエタノール、10U/mlのIL-2、10ng/mlのIL-7および10%非働化ウシ胎仔血清を含むRPMI-1640培地にサスペンドし、37℃5%CO2インキュベーター内で培養した。7日後(8日目)、mDC/WT134-51(1×105細胞)を新しく調製し、10U/mlのIL-2および10ng/mlのIL-7を含む新しい細胞培養培地でさらに培養を継続した。この操作をさらに2回(15日目および22日目)行い、計4回刺激した(1、8、15および22日目)。コントロールとして、NAD細胞にIL-2(10U/ml)とIL-7(10ng/ml)とを7日間隔で4回(1、8、15および22日目)添加した。
mDC/WT134-51(1×105細胞)による4回刺激培養により誘導されたT細胞を、再刺激としてimDC/WT134-51、imDC/WT137-45またはimDC/WT140-48(1×105細胞)とともに96穴U底プレートに播種し、ゴルジストップを添加して、37℃5%CO2インキュベーター内で6時間培養した。その後、ヒトFc受容体遮断薬を添加し、4℃で5分間インキュベートした。PE/Cy5標識抗ヒトCD8抗体(クローンRPA-T8;eBioscience)およびAPC/Cy7標識抗ヒトCD4抗体(クローンOKT4;BioLegend)と4℃で20分間反応後、BD Cytofix/Cytoperm Plus Fixation/permeabilized with BD GolgiStop(BD Biosciences)を用いて透過処理を行った。その後、APC標識抗ヒトIFN-γ抗体(クローンB27;BioLegend)とインキュベートし、CD8陽性細胞中のIFN-γ産生性細胞の頻度を分析した。
Paired t-testまたはWilcoxon signed-rank testで検定した。
HLA-A*02:01拘束性WT137-45特異的CD8陽性T細胞の誘導
HLA-A*02:01陽性者(5例)のNAD細胞を7日毎にmDC/WT134-51で4回刺激したのち、imDC/WT137-45またはimDCで再刺激し、IFN-γ産生CD8陽性T細胞の頻度を解析した。その結果、5例中3例(60%)において、imDC再刺激と比較してimDC/WT137-45再刺激によりCD8陽性T細胞中のIFN-γ産生CD8陽性T細胞の頻度が10%以上増加した。
HLA-A*02:06陽性者(7例)のNAD細胞を7日毎にmDC/WT134-51で4回刺激したのち、imDC/WT137-45またはimDCで再刺激し、IFN-γ産生CD8陽性T細胞の頻度を解析した。その結果、7例中2例(28.6%)において、imDC再刺激と比較してimDC/WT137-45再刺激によりCD8陽性T細胞中のIFN-γ産生CD8陽性T細胞の頻度が10%以上増加した。
HLA-A*02:07陽性者(2例)のNAD細胞を7日毎にmDC/WT134-51で4回刺激したのち、imDC/WT137-45またはimDCで再刺激し、IFN-γ産生CD8陽性T細胞の頻度を解析した。その結果、2例中1例(50%)において、imDC再刺激と比較してimDC/WT137-45再刺激によりCD8陽性T細胞中のIFN-γ産生CD8陽性T細胞の頻度が10%以上増加した。
HLA-A*26:01陽性者(2例)のNAD細胞を7日毎にmDC/WT134-51で4回刺激したのち、imDC/WT140-48で再刺激し、IFN-γ産生CD8陽性T細胞の頻度を解析した。その結果、2例中2例(100%)において、IFN-γ産生CD8陽性T細胞が認められた。
1.方法
PBMCの刺激
HLA-A*02:01/24:02陽性者(3例)、HLA-A*02:06/24:02陽性者(1例)、HLA-A*02:07/24:02陽性者(2例)、およびHLA-A*24:02/24:02陽性者(1例)(がん患者または健常人)の末梢血を採取し、リンパ球分離溶液(ナカライテスク)を用いた密度勾配遠心により単核球分画を単離し、PBMCを得た。
各刺激の7~12日後に、24穴プレートの各ウェルの細胞と培地とをピペティングでよく混和し、細胞含有培地を500μlずつ回収した。遠心して上澄みを除去後、細胞に10μg/mlブレフェルジンA(BFA、SIGMA)含有hCMを800μl添加し、細胞浮遊液を調製した。96穴プレートのウェルに、WT137-45およびWT140-48をそれぞれ最終濃度20μg/mlとなるよう添加した。コントロールとしてペプチド添加なしのウェルも用意した。これらのウェルに、調製した細胞浮遊液を1ウェルにつき200μl添加し、37℃で4時間反応させた。
各刺激の7~12日後に、24穴プレートの各ウェルの細胞と培地とをピペティングでよく混和し、細胞含有培地を500μlずつ回収し、2等分して2本のFACSチューブに入れた。細胞をFACS Bufferで2回洗浄後、Clear Back (Human FcR blocking reagent,MBL)を加え、室温5分反応させた。2本のチューブの1本に、PE標識 HLA-A*02:01 WT137-45 Tetramer(MBL)を加え、氷上で60分間反応させた。もう1本のチューブはテトラマー非添加チューブとした。これらのチューブに、Pacific Blue標識抗ヒトCD3抗体(クローンUCHT1、BD Biosciences)およびFITC標識抗ヒトCD8抗体(クローンT8、サイトスタット/コールタークローン、BECKMAN COULTER)を加え、氷上で30分間反応させた。細胞をFACS Bufferで2回洗浄後、7-AAD Viability Staining Solution(BioLegend)を添加したFACS Bufferに浮遊させ、BD FACSCanto II フロサイトメーター(BD Biosciences)で測定し、FLOWJOソフトウエア(BD Biosciences)で解析した。
1.方法
PBMCの刺激
HLA-A*01:01陽性者、HLA-A*26:01陽性者、HLA-B*35:01陽性者、HLA-C*03:03陽性者、HLA-C*05:01陽性者、およびHLA-C*07:01陽性者(各1例)(がん患者または健常人)の末梢血を採取し、リンパ球分離溶液(ナカライテスク)を用いた密度勾配遠心により単核球分画を単離し、PBMCを得た。
前3回の刺激に使用したPBMCとはHLAクラスIの1つのアレルのみが異なるPBMC(表4参照)にWT137-45またはWT140-48をパルスした細胞またはペプチドをパルスしていない細胞をAPCとして使用して、細胞内サイトカインアッセイおよびテトラマーアッセイを行った。APC作製のため、3×105細胞のPBMCを300μlのRPMI1640(血清不含)に浮遊させ、WT137-45(20μg/ml)またはWT140-48(20μg/ml)を添加し、またはペプチドを添加せず、37℃で1時間反応させた。この細胞をPBSで2回洗浄し、1×105細胞/100μlとなるようブレフェルジンA(BFA)(SIGMA)添加hCMを添加し、APC浮遊液を調製した。第1回刺激の21日以降に、刺激後の細胞と培地とをピペティングでよく混和し、各ウェルより細胞含有培地を900μlずつ回収し、遠心して上澄みを除去後、BFA添加hCMを添加し細胞浮遊液を調製した。丸底96ウエルプレートに、上記の調製したAPCと細胞浮遊液、各100μlずつ添加し、37℃5%CO2で4時間共培養した。反応はトリプリケートで行った。
WT134-51(WAPVLDFAPPGASAYGSL)(配列番号2)の配列中に含まれるMHCクラスI分子に結合可能な9merペプチドを、NetMHC-4.0(http://www.cbs.dtu.dk/services/NetMHC-4.0)にて解析した。解析したペプチドを表2に、結合予測結果を表3-1~表3-32に示す。表3-1~表3-32中、「Rank」は400,000のランダムな天然ペプチドと比較した%Rank、「H_Avg_Rank」は指定したすべてのアレルに対して計算された%Rankの調和平均、「N_binders」は、そのペプチドでカバーされるアレルの数を示す。「WB」はweak binder、「SB」はstrong binderを意味する。
HLA-A*11:01陽性者、HLA-A*26:03陽性者、HLA-A*15:01陽性者、またはHLA-C*07:02陽性者の末梢血からPBMCを得て、前記I、II、またはIIIに記載のとおり、細胞内サイトカインアッセイおよび/またはテトラマーアッセイを行う。WT134-51および式(5)の化合物、またはWT134-51のみで刺激したPBMCは、HLA-A*11:01、HLA-A*26:03、HLA-A*15:01、またはHLA-C*07:02拘束性のWT137-45特異的CD8陽性T細胞および/またはWT140-48特異的CD8陽性T細胞を誘導することが確認される。
Claims (21)
- HLA-A*02:01、HLA-A*02:06、HLA-A*02:07、HLA-A*26:01、HLA-A*01:01、HLA-B*35:01、HLA-C*03:03、HLA-C*05:01、またはHLA-C*07:01陽性対象における癌を処置または予防するための、WAPVLDFAPPGASAYGSL(配列番号2)のアミノ酸配列からなるペプチドもしくはCTL誘導活性を有するその改変ペプチドまたはその薬学上許容される塩を含む医薬組成物。
- 前記対象が、HLA-A*02:01、HLA-A*02:06、HLA-A*02:07、またはHLA-A*26:01陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-A*02:01、HLA-A*02:06、またはHLA-A*02:07陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-A*02:01陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-A*02:06陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-A*02:07陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-A*26:01陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-A*01:01、HLA-B*35:01、HLA-C*03:03、HLA-C*05:01、またはHLA-C*07:01陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-A*01:01陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-B*35:01陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-C*03:03陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-C*05:01陽性対象である、請求項1に記載の医薬組成物。
- 前記対象が、HLA-C*07:01陽性対象である、請求項1に記載の医薬組成物。
- 配列番号2のアミノ酸配列からなるペプチドまたはその薬学上許容される塩を含む、請求項1~13のいずれかに記載の医薬組成物。
- 癌が、WT1が発現している癌、またはWT1遺伝子の発現レベルの上昇を伴う癌である、請求項1~19のいずれかに記載の医薬組成物。
- 癌が、急性骨髄性白血病、慢性骨髄性白血病、急性リンパ芽球性白血病、慢性リンパ球性白血病、骨髄異形成症候群、多発性骨髄腫、悪性リンパ腫、胃癌、大腸癌、肺癌、乳癌、胚細胞癌、肝癌、皮膚癌、膀胱癌、前立腺癌、子宮癌、子宮頸癌、卵巣癌、脳腫瘍、または神経膠腫である、請求項1~20のいずれかに記載の医薬組成物。
