Abstract
本開示は、多血小板血漿から成長因子群を効率的に回収し、活性化させた成長因子混合物を提供する。 本開示は、血液から成長因子混合物を調製する方法であって、血液に対して第一の遠心分離処理を行い、上清を回収する工程と、該上清に対して第二の遠心分離処理を行い、多血小板血漿を得る工程と、該多血小板血漿に対して、血小板を活性化させる処理を行う工程と、血小板を活性化させた該多血小板血漿から成長因子混合物を得る工程と、を含む、方法を提供する。
Description
本開示は、成長因子混合物を調整する方法に関する。
従来、血小板を多く含んだ血漿(多血小板血漿、PRP)を血液から分離し、患部に注射するPRP療法が知られている。PRP療法は、血小板に含まれる血小板由来増殖因子(PDGF)、トランスフォーミング増殖因子β(TGF-β)、線維芽細胞増殖因子(FGF)、血管内皮増殖因子(VEGF)、及び上皮成長因子(EGF)などの成長因子が持つ組織修復能力を利用し、治癒力を高める手法である。
ところで、この多血小板血漿をさらに濃縮して活性化し、濃縮活性化した多血小板血漿から血小板由来成分を回収して得られた成長因子混合物は、無細胞化されているため安全性が高く、またすでに濃縮活性化されていることから利便性が高い。そのため、再生医療分野における有望な材料として期待されている。
したがって、本開示は以下を提供する。

本開示によれば、多血小板血漿から成長因子群を効率的に回収でき、また活性化されているため、医療の現場において有用である。また、無細胞化されていることから、再生医療等安全性確保法などの法令上の制約を回避することができ、利便性が高い。

本明細書において、「約」とは、後に続く数値の±10%を意味する。
本明細書において、「成長因子混合物」とは、任意の成長因子の混合物を言い、代表的に血小板由来のサイトカインやその他成長因子を含むものであり、例えば、PDGF、TGF-β、EGFなどの成長因子が混合したものをいう。「混合物」は特に限定されず、天然の状態で2つ以上の要素が共存しているものであればよく、積極的に混合する操作によって生成されたものでなくてもよく、そうであってもよい。
本明細書において、「多血小板血漿」(またはPRPともいう)とは、通常の血漿よりも血小板を多く含む血漿濃縮物を指し、血液中の血漿を遠心分離処理することによって得ることができる。血小板の濃度は血液(全血)の約1倍以上であり、代表的に、約2~約7倍である。多血小板血漿は例えば、約10×104個/μL以上、約25×104個/μL以上、約50×104個/μL以上、または好ましくは約100×104個/μL以上の濃度の血小板を含む。あるいは、また、通常、多血小板血漿には、PDGF、TGF-β、VEGF、EGF、FGF、IGF、またはHGFなどの成長因子が豊富に含まれる。また本明細書において、「白血球含有多血小板血漿」とは、上記のような多血小板血漿に白血球が含まれるものをいう。
本明細書において血小板の「活性化」とは出血時に血小板に生じる一連の反応、それと同等の反応、または一部の反応をいい、衝撃や刺激物質により、血小板内の細胞骨格系が変化し、多数の長い突起を出し金平糖のような形をとること、または血小板内で生成された成長因子が血小板外へ放出されること、または血小板内に貯蔵されていた成長因子が血小板外へ放出されることをいう。

一実施形態において、上清は白血球を含むことができる。本開示では、白血球を含む上清は、全血の約1.5倍以上の濃度で白血球を含むことが好ましいが、これに限定されない。
一実施形態において、多血小板血漿は白血球含有多血小板血漿とすることができる。本開示では、白血球含有多血小板血漿は、全血の約5倍以上の濃度で白血球を含むことが好ましいが、これに限定されない。
本開示の一実施形態において、血液に対する第一の遠心分離処理と、この第一の遠心分離処理によって得られる上清に対する第二の遠心分離処理とは同じ条件で行うことが好ましい。一実施形態において、第一または第二の遠心分離処理は、約100~約3000Gの遠心分離とすることができ、好ましくは約500~約1000G、または約940Gとしてもよい。処理時間は約2~約30分とすることができ、好ましくは約5~約20分、または約15分である。
血液や上清に対する遠心処理は、室温で行うことが好ましく、例えば約20℃~約27℃で行うことができ、さらに好ましくは約24℃で行われる。
以上のようにして血液(全血)に対して第一の遠心分離処理を行うことにより、上清を分離することができ、またこの上清に対して第二の遠心分離処理を行うことにより、多血小板血漿を分離することができる。また一実施形態において、第一の遠心分離処理と第二の遠心分離処理は必要に応じて複数回行うこともできる。
本開示の一実施形態において、上記のようにして回収した多血小板血漿に対して、血小板を活性化させる処理を行う工程は、多血小板血漿に、血小板を活性化させる薬剤を添加することで行うこともできる。このようにして血小板が活性することによって血小板由来の成長因子が多数放出され、また活性化される。
あるいは、血小板を活性化させる処理は、薬剤を使用しない形でもよい。例えば低温刺激による方法、超音波処理による方法、血小板を基材に付着させる方法などであってもよい。この場合の低温刺激温度は、約0℃以上、好ましくは約4℃以上などであり得る。
血小板を活性化する薬剤は、特に限られるものではないが、塩化カルシウム、リン酸カルシウム、乳酸カルシウム、炭酸カルシウム等のカルシウムイオン源化合物を用いることができ、カルシウムイオンが存在することによって血小板が活性化される。
本開示の一実施形態において、血小板を活性化する薬剤は溶液として添加することができ、その濃度は特に限られるものではないが、例えば約1質量%以上、約1.5質量%以上、約2質量%以上、約2.5質量%以上とすることができる。また一実施形態において、その添加量は特に限られるものではないが、例えば、多血小板血漿の液量を基準として約0.05体積倍以上、約0.1体積倍以上、約0.15体積倍以上、約0.2体積倍以上、約0.25体積倍以上、約0.3体積倍以上とすることができる。
本開示の一実施形態において、血小板を活性化させた該多血小板血漿から成長因子混合物を得る工程は、血小板を活性化させた前記多血小板血漿から細胞を除去する処理を含むことができる。この場合、細胞の除去は多血小板血漿から細胞を除去することができる手段であれば特に限られるものではなく、フィルタリング処理を好ましく使用することができる。フィルタリング処理は多血小板血漿に含まれる白血球や血小板、赤血球などの種々の細胞を除去するものであり、除去対象となる細胞のサイズに応じて、メンブレン孔径は適宜設定することができる。一実施形態において、必要に応じて複数のメンブレン孔径のフィルターを用いることができ、これらを組み合わせて複数回フィルタリングすることにより、血小板以外の細胞を除去することができる。例えば、フィルターとしては、孔径が約5μm以下、約1μm以下、約0.7μm以下、約0.5μm以下、約0.3μm以下のメンブレンフィルターを使用することができ、除去対象となる細胞をフィルタリングできるものであればよい。
一実施形態において、フィルタリング処理は遠心分離処理によって行うことができ、上記のようなフィルターを用いて多血小板血漿を遠心分離処理することで血小板や白血球などの種々の細胞を除去することができる。この場合、遠心分離処理の条件としては、第一の遠心分離処理及び第二の遠心分離処理と同様の条件を採用することができる。
成長因子混合物やその前段階の無細胞系血漿が細胞(特に血小板)を実質的に含有しない、または全く含有しないことは、例えば、セルカウンターを用いて1μm以上の細胞が確認されないことによって実証できる。あるいは血球計算盤や血球計測機などを用いて血小板が観測されないことによって実証してもよい。いずれかの手段によって血小板を含まないことが確認できればよい。
以上のようにして、多血小板血漿に含まれる血小板や白血球などの種々の細胞を除去することで、成長因子混合物は、細胞を実質的に含有しないものとすることができる。そのため、従来の多血小板血漿では細胞を含むものであることから種々の法令の制限を受けていたところ、本開示の成長因子混合物は、細胞を含有しないことから、制約を受けることなく再生医療の材料として使用することができる。
本開示の一実施形態において、上記のようにして第二の遠心分離処理を行った後に、フィブリノーゲン及び/またはアルブミンが含まれる上清を除去することもできる。この場合には、沈殿した(分離した)血液細胞を少量のPBS(リン酸緩衝生理食塩水)で懸濁して血液細胞の懸濁物(多血小板血漿)を得ることができる。その後、この懸濁物に上記のような血小板を活性化する薬剤が添加される。
一実施形態において、上記のようにしてフィブリノーゲン及び/またはアルブミンを除去することにより、本開示の方法によって最終的に得られる成長因子混合物において、フィブリノーゲンは、約10mg/mL未満、約7mg/mL未満、約5mg/mL未満、約4mg/mL未満、約3mg/mL未満、約2mg/mL未満、約1mg/mL未満のレベルで実質的に除去され、またアルブミンは、約50mg/mL未満、約40mg/mL未満、約37mg/mL未満、約30mg/mL未満、約20mg/mL未満、または約10mg/mL未満のレベルで実質的に除去されることができる。
本開示の一実施形態において、上記のように細胞を除去することで得られた成長因子混合物は、凍結乾燥されることなく使用することができる。
本開示の一実施形態において、本開示の方法によって得られる成長因子混合物は、通常の多血小板血漿が含有する成長因子を含有することができる。このような成長因子としては、例えば、PDGF、TGF-β、VEGF、EGF、FGF、IGF、HGF等が挙げられる。本開示の成長因子混合物は少なくともこれら因子を含有することができる。
一実施形態において、各種成分は、例えば以下の範囲で含まれることができる。

本開示の一実施形態において、本開示の方法によって得られる成長因子混合物は、その投与される組織は特に限られるものではなく、関節や筋腱にも投与することができる。また一実施形態において、本開示の成長因子混合物は、-約80℃~約25℃で保存することができ、使用の際には約20~約25℃の室温で保存することもできる。
本開示の一実施形態において、本開示の方法の一連の流れは以下のようにすることができる。すなわち、採血後、約2000rpm(980g)、15分、20~25℃で第1遠心を行い、血清上部1/2~2/3は廃棄し、中間層を採取する。このとき、リンパ球分離液やBDバキュテイナCPT採血管などを用いて赤血球を除外することもできる。その後、約10ml程度を遠沈管に回収し、フィブリンが低減するようにPBSを加えて希釈し、約2000rpm(980g)、15分、20~25℃で第2遠心を行う。上清を廃棄し、必要な場合にはPBSや2% CaCl2を加えて、よく攪拌し、20~25℃で血小板を活性化させ成長因子を放出させる。
その後、この溶液を凍結融解しない場合には、約2000rpm(980g)、2分、20~25℃でさらに遠心することができる。凍結融解する場合には、凍結する前に、血小板以外の細胞(白血球や赤血球)を除去するためのフィルター処理等を実施してもよく、
-80℃で、30分以上の凍結を行う。融解後、14000rpm、5分、20~25℃で遠心することができる。

凍結融解する場合、およびしない場合のいずれにおいても、遠心後の上清を約0.45μmのフィルターでろ過し、この溶液についてVEGF、フィブリノーゲン、アルブミンなどを測定することができる。また細胞がないことを確認し、本開示の成長因子混合物とすることができる。
本開示の成長因子混合物または多血小板血漿は、対象の疾患、障害または状態を治療するために用いることができる。例えば本開示の成長因子混合物または多血小板血漿は、対象から得た血液を用いて調整し、治療有効量を対象に投与することで、その対象の疾患、障害または状態を治療することができる。
一実施形態において、本開示の成長因子混合物または多血小板血漿を用いた治療は、疾患の状態、種類、患者の状態、患者の希望などにより、投与量、投与期間、投与回数などを調整することができる。例えば、膝治療の場合、医師の判断で、片足に2本(1本は任意の一定量)投与したり、両足にそれぞれ1本ずつ投与することもできる。また他の実施形態において、1本投与した後に、さらに追加で1本または複数を投与することもできる。
他の実施形態において、本開示の成長因子混合物または多血小板血漿を用いた治療は、膝を含む関節の治療にも用いることができ、例えば、変形性膝関節症、変形性股関節症、半月板損傷、靭帯損傷、膝関節痛などの治療に用いることができる。また一実施形態において、本開示の成長因子混合物または多血小板血漿は、皮膚、骨、靭帯、アキレス腱、腱鞘炎、歯槽骨、半月板、卵巣、子宮内膜、角膜、その他軟部組織などの身体組織の再生治療に用いることもできる。また一実施形態において、本開示の成長因子混合物または多血小板血漿は、損傷治癒の促進(早期治癒)や鎮痛軽減、手術創の治癒補助などのために用いることができる。
他の実施形態において、本開示の成長因子混合物または多血小板血漿は、薄毛治療などの美容のために用いることもでき、例えば、目的の美容の種類に応じて、投与方法、投与量、投与期間を調整することができる。一実施形態において、本開示の成長因子混合物または多血小板血漿は、美容目的のために、例えば1週間おきに1本を投与したり、1ヶ月おきに2本の投与を2~3回繰り返すこともできる。

採血から成長因子混合物の生成までの一連の流れを図1に示した。まず診療所にて採血(採血管4本分/約40mL)した。この際、ヘパリン採血管4本(ベノジェクトII真空採血管(滅菌品)、ヘパリンナトリウム入)を用いた。またヘパリン採血管に代えて、BDバキュテイナ CPTを使用することもできる。採血後、静かに約10回転倒混和し、冷蔵で輸送した。この際、0~10℃で0~72時間に亘って血液を予冷蔵しておくこともできる。

上清(血漿+白血球層)を50mL遠沈管(Corning社製)に回収し、W-PRP(WBC-containing platelet-rich plasma;白血球含有多血小板血漿)を得た。この白血球含有多血小板血漿に対して、卓上多本架遠心機H-40F(スイングローター:RF-121-S/コクサン社製)を用いて第2遠心(940G,15分,20~25℃)を行った。BDバキュテイナ CPTを使用した場合、第2遠心:1500G,15分。
上清を除去し(沈殿物+上清0.6mL以下)、Total volumeが3.6mLとなるようPBS(-)を添加した。さらに2% CaCl2 0.9mLを添加した(ニプロ 塩化カルシウム注2%「NP」)。よく攪拌し、血小板を活性化させ成長因子を放出させた(20min,20~25℃)。
活性化させた白血球含有多血小板血漿溶液を5mLまたは10mLシリンジに充填し、無細胞化フィルタリングを行った。無細胞化フィルターとしてはミニザルト孔径0.45μm(ザルトリウス社製)を用いた。
無細胞化加工済溶液中(成長因子混合物)に細胞が存在しないことを目視(血球計算盤:10μL)で確認し、3mLクライオジェニックバイアル(Corning社製)2本に1.5mLずつ分注した(製品容量3mL)。ガラスバイアル瓶(ゴム栓、アルミキャップ付き;Weatonまたはマムエル社製)5mLを1本または3mLを2本使用することもできる。残液は無菌検査等で使用した。
診療所へ発送するまで-20℃で凍結保存し(無菌検査結果が出るまで約3週間)、診療所へ発送した(冷凍便。ドライアイスを同封)。製品の使用期限は、冷凍保存の場合は検査結果判明日から3ヶ月、冷蔵保存の場合は解凍後2日間とした(解凍後の再凍結は控える)。
(実施例2:成長因子混合物の作成(2))

細胞加工施設に到着次第、血液を室温に戻した。ヘパリン採血管3本の血液30mLを、リンパ球分離溶液を含むリンパ球分離チューブ1本(Leucosep(商標) 血液量30mL、グライナージャパン社製)に移し、卓上多本架遠心機LCX-100(トミ
ー精工社製)を用いてリンパ球分離チューブ1本を遠心した(第1遠心:1200G,15min,15~25℃)。BDバキュテイナ CPTを使用した場合、リンパ球分離チューブに移さず、遠心(第1遠心:1500G,15分)を行ってよい。

上清を除去し(沈殿物+上清1.0mL以下)、Total volumeが6mLとなるようPBS(-)を添加した。さらに2% CaCl2 1.5mLを添加した(大塚塩化カルシウム注2%)。よく攪拌し、血小板を活性化させ成長因子を放出させた(60min,20~25℃)。
活性化させた白血球含有多血小板血漿溶液を10mLシリンジに充填し、無細胞化フィルタリングを行った。無細胞化フィルターとしてはミニザルト孔径0.2μm(ザルトリウス社製)を用いた。
無細胞化加工済溶液中(成長因子混合物)に細胞が存在しないことを自動セルカウンターで確認し、5mLクライオジェニックバイアル(Corning社製)2本に3mLずつ分注した(製品容量約6mL)。ガラスバイアル瓶(ゴム栓、アルミキャップ付き;Weatonまたはマムエル社製)10mLを1本または5mLを2本使用することもできる。残液は無菌検査等で使用した。

無細胞化加工済溶液中(成長因子混合物)に細胞が存在しないことを目視(血球計算盤:10μL)で確認し、3mLクライオジェニックバイアル(Corning社製)2本に1.5mLずつ分注した(製品容量3mL)。ガラスバイアル瓶(ゴム栓、アルミキャップ付き;Weatonまたはマムエル社製)5mLを1本または3mLを2本使用することもできる。残液は無菌検査等で使用した。
診療所へ発送するまで-20℃で凍結保存し(無菌検査結果が出るまで約3週間)、診療所へ発送した(冷凍便。ドライアイスを同封)。製品の使用期限は、冷凍保存の場合は検査結果判明日から3ヶ月、冷蔵保存の場合は解凍後2日間とした(解凍後の再凍結は控える)。
(実施例5:成長因子混合物の作成(4))
採血から成長因子混合物の生成までの一連の流れは図2と同様である。まず診療所にて採血(採血管3本分/約30mL)した。この際、ヘパリン採血管3本(BDバキュテイナ採血管、ヘパリンナトリウム入)を用いた。またヘパリン採血管に代えて、BDバキュテイナ CPTを使用することもできる。採血後、静かに約20回転倒混和し、冷蔵で輸送した。この際、0~15℃で0~72時間に亘って血液を予冷蔵しておくこともできる。

細胞加工施設に到着次第、血液を室温に戻した。ヘパリン採血管3本の血液30mLを、リンパ球分離溶液を含むリンパ球分離チューブ1本(Leucosep(商標) 血液量30mL、グライナージャパン社製)に移し、卓上多本架遠心機LCX-100(トミ
ー精工社製)を用いてリンパ球分離チューブ1本を遠心した(第1遠心:1200G,15min,15~25℃)。BDバキュテイナ CPTを使用した場合、リンパ球分離チューブに移さず、遠心(第1遠心:1500G,15分)を行ってよい。

白血球層よりも上方の5~10mm程度残して血漿を回収(廃棄)し、残った血漿を15mL遠沈管(IWAKI社製)1本に回収し、多血小板血漿を得た。この多血小板血漿に対して、卓上多本架遠心機LCX-100(トミー精工製)を用いて第2遠心(1200G,15min,15~25℃)を行った(BDバキュテイナ CPTを使用した場合、第2遠心:1500G,15分)。
上清を除去し(沈殿物+上清1.0mL以下)、Total volumeが6mLとなるようPBS(-)を添加した。さらに2% CaCl2 1.5mLを添加した(大塚塩化カルシウム注2%)。よく攪拌し、血小板を活性化させ成長因子を放出させた(60min,20~25℃)。
活性化させた多血小板血漿溶液を10mLシリンジに充填し、無細胞化フィルタリングを行った。無細胞化フィルターとしてはミニザルト孔径0.2μm(ザルトリウス社製)を用いた。
無細胞化加工済溶液中(成長因子混合物)に細胞が存在しないことを自動セルカウンターで確認し、5mLクライオジェニックバイアル(Corning社製)2本に3mLずつ分注した(製品容量約6mL)。ガラスバイアル瓶(ゴム栓、アルミキャップ付き;Weatonまたはマムエル社製)10mLを1本または5mLを2本使用することもできる。残液は無菌検査等で使用した。
診療所へ発送するまで-80℃で凍結保存し(無菌検査結果が出るまで約3週間)、診療所へ発送した(冷凍便。ドライアイスを同封)。製品の使用期限は、冷凍保存の場合は検査結果判明日から2ヶ月、冷蔵保存の場合は解凍後3日間とした(解凍後の再凍結は控える)。
(実施例6:成長因子混合物の作成(5))

細胞加工施設に到着次第、血液を室温に戻した。卓上多本架遠心機H-40F(スイングローター:RF-121-S/コクサン社製)を用いてヘパリン採血管4本を遠心した(第1遠心:940G,15分,20~25℃)。ヘパリン採血管4本の血液40mLを、リンパ球分離溶液を含むリンパ球分離チューブに移して遠心する場合は第1遠心:940~1800G,15分、20~25℃で遠心。BDバキュテイナ CPTを使用した場合、第1遠心:1500G,15分(ヘパリン)もしくは20分(クエン酸)、20~25℃で遠心。なお、遠心機は他の機種でもよく、ローターはスイング式を使用してもよく、アングル式を使用してもよい。
上清を50mL遠沈管(Corning社製)に回収し、多血小板血漿を得た。この上清を採取する際、血漿上部の一部~ほぼ全てを廃棄してもよく、白血球層(中間層)付近に存在する血小板をより多く採取できるよう、白血球層より下層も含めて約10mL以上採取してもよい。この多血小板血漿に対して、Total volumeが30mL以上となるようPBS(-)を添加した後、卓上多本架遠心機H-40F(スイングローター:RF-121-S/コクサン社製)を用いて第2遠心(940G,15min,20~25℃)を行った。第1遠心でリンパ球分離溶液を含むリンパ球分離チューブに移して遠心した場合、第2遠心:940~1800G,15分、20~25℃。第1遠心でBDバキュテイナCPTを使用した場合、第2遠心:1500G~1800G,15分~20分。なお、上記遠心の前にフィルター等により血小板以外の細胞を除去してもよい。
上清を除去し(沈殿物+上清1mL以下)、Total volumeが4.8mLとなるようPBS(-)を添加した。さらに2% CaCl2 1.2mLを添加した(ニプロ 塩化カルシウム注2%「NP」)。上清を除去した後の液量に応じてTotal volumeおよびCaCl2の濃度や液量を変更してもよく、例えば上清を除去したときの液量が1.2mL以下の場合はTotal volumeが4mLとなるようPBS(-)を添加し、2% CaCl2 1mLを添加してもよい。その後、よく攪拌し、血小板を活性化させ成長因子を放出させた(30分,20~25℃)。活性化の時間は適宜調整することが可能で、30分以内でも、120分までのばしてもよい。
活性化させた多血小板血漿溶液に対して凍結(30分以上、-80℃)と融解を行った。凍結を行う前にフィルター等により血小板以外の細胞(白血球や赤血球)を除去してもよい。解凍は翌日以降でもよく、解凍の温度と時間は、室温30分以上でも4℃で2時間以上でもよい。その後、卓上型遠心機5418(Eppendorf社製)を用いて遠心(14000rpm,5分,20~25℃)を行い、上清を5mLまたは10mLシリンジに充填し、無細胞化フィルタリングを行った。無細胞化フィルターとしてはミニザルト孔径0.45μm(ザルトリウス社製)を用いた。
無細胞化加工済溶液中(成長因子混合物)に細胞が存在しないことを目視(血球計算盤:10μL)で確認し、3mLクライオジェニックバイアル(Corning社製)2本に約1.5mLずつ分注した(製品容量約3mL)。必要に応じて他の容器・本数・容量を選択してもよく、例えば、ガラスバイアル瓶(ゴム栓、アルミキャップ付き;Weatonまたはマムエル社製)5mLを1本または3mLを2本使用することもできる。残液は無菌検査等で使用した。

(実施例7:PDGF-BBおよびVEGFの測定)
採血から成長因子混合物の生成までの一連の流れは図1と同様である。まず診療所にて採血(採血管4本分/約40mL)した。この際、ヘパリン採血管4本(ベノジェクトII真空採血管(滅菌品)、ヘパリンナトリウム入)を用いた。採血後、静かに約10回転倒混和し、冷蔵で輸送した。この際、0~10℃で0~72時間に亘って血液を予冷蔵しておくこともできる。

採血管4本中1本の上清(血漿+白血球層)をPRPとした。成長因子の測定用サンプルとして、細胞がフィルターに詰まりそうな場合は再度遠心した上清を用いて、無細胞化フィルタリングを行った後、測定まで凍結して保存した。残り3本の上清(血漿+白血球層)を15mL遠沈管(Corning社製)に回収し、W-PRP(WBC-containing platelet-rich plasma;白血球含有多血小板血漿)を得た。この白血球含有多血小板血漿に対して、卓上多本架遠心機H-40F(スイングローター:RF-121-S/コクサン社製)を用いて第2遠心(940G,15分,20~25℃)を行った。
上清を除去し(沈殿物+上清0.6mL以下)、Total volumeが3.6mLとなるようPBS(-)を添加した。PBS(-)を添加した溶液にさらに2% CaCl2 0.9mLを添加した(ニプロ 塩化カルシウム注2%「NP」)。よく攪拌し、血小板を活性化させ成長因子を放出させた(20min,20~25℃)。
活性化させた白血球含有多血小板血漿溶液を5mLまたは10mLシリンジに充填し、無細胞化フィルタリングを行った。無細胞化フィルターとしてはミニザルト孔径0.45μm(ザルトリウス社製)を用いた。

Claims (12)
- 血液から成長因子混合物を調製する方法であって、
血液に対して第一の遠心分離処理を行い、上清を回収する工程と、
該上清に対して第二の遠心分離処理を行い、多血小板血漿を得る工程と、
該多血小板血漿に対して、血小板を活性化させる処理を行う工程と、
血小板を活性化させた該多血小板血漿から成長因子混合物を得る工程と、
を含む、方法。 - 前記第一の遠心分離処理と前記第二の遠心分離処理とが同じ条件で行われる、請求項1に記載の方法。
- 前記第一の遠心分離処理と前記第二の遠心分離処理とが、約100G~約3000Gで約2~約30分、室温の条件で行われる、請求項1または2に記載の方法。
- 前記第一の遠心分離処理と前記第二の遠心分離処理とが、約940Gで約15分、約24℃の条件で行われる、請求項1~3のいずれか一項に記載の方法。
- 前記活性化させる処理が、前記多血小板血漿に、血小板を活性化させる薬剤を添加することを含む、請求項1~4のいずれか一項に記載の方法。
- 前記血小板を活性化させる薬剤が塩化カルシウムを含む、請求項5に記載の方法。
- 前記成長因子混合物が凍結乾燥されない、請求項1~6のいずれか一項に記載の方法。
- 前記成長因子混合物を得る工程が、血小板を活性化させた前記多血小板血漿から細胞を除去する処理を含む、請求項1~7のいずれか一項に記載の方法。
- 前記多血小板血漿からの細胞の除去が、フィルタリング処理を含む、請求項8に記載の方法。
- さらに、前記多血小板血漿に対して、フィブリノーゲンを除去する工程を含む、請求項1~9のいずれか一項に記載の方法。
- さらに、前記多血小板血漿に対して、アルブミンを除去する工程を含む、請求項1~10のいずれか一項に記載の方法。
- 前記フィブリノーゲンは約5mg/mL未満となるように除去され、及び/または前記アルブミンは約37mg/mL未満となるように除去される、請求項10または11に記載の方法。
