본 발명은 위암 개체로부터 분리된 시료에서 측정된 ERH(Enhancer of Rudimentary Homolog) mRNA 또는 단백질 수준이 대조군의 수준보다 높으면 위암 예후가 대조군 대비 좋을 것으로 판단함으로써, 위암 예후를 쉽고 간단한 방법으로 예측할 수 있는 정보 제공 방법에 관한 것이다.

본 발명은 위암의 예후 예측용 ERH 마커 및 이의 용도에 관한 것으로서, 위암 환자에게서의 생존률 예후를 예측할 수 있는 ERH 유전자, 이의 발현수준을 측정하는 제제를 포함하는 위암 예후 예측용 조성물 및 키트, 및 상기 마커를 이용하여 예후를 예측하는 방법에 관한 것이다.
위암은 전세계적으로 암으로 인한 사망 원인의 2위를 차지하며 한국에서도 주요 암 사망의 원인 중 하나이다. 위암의 근본적인 치료는 수술적인 제거이지만, 조기 위암을 제외하고는 수술적 절제 후에도 상당수의 환자에서 재발된다. 2기 위암의 경우는 5년 생존율이 30-70%, 3기 위암의 경우는 5-30%에 불과하다. 같은 병기의 환자들에 대한 치료적 완전절제술 후 예후를 예측하는 것은 여전히 어려운 과제로 남아있다.
임상적, 병리적 분류법을 이용하여 위암의 예후를 예측하는 모델을 만들기 위한 상당한 연구가 있었음에도, 수술 후에 재발할 확률이 높아 수술 후 보조적 항암치료가 필요한 환자군과 재발 확률이 낮아 수술 후 보조적 항암치료의 필요성이 낮은 환자군을 구별하는 방법은 정립되어 있지 않다. 그렇기 때문에 현재는 수술이 가능한 위암환자의 경우 모두 일괄적으로, 치료적 완전절제술을 받고 조기 위암인 경우를 제외하고는 대부분 추가적인 보조 항암치료를 받는다. 그러나, 수술적 병기가 같은 경우에도 수술 후 환자의 예후는 다양하게 나타나기 때문에 각 환자에 맞는 정확한 예후의 예측과 맞춤치료가 절실히 필요한 상황이다.
진행성 위암의 경우에도 마찬가지로 동일한 치료를 함에도 환자에 따라 예후가 매우 다양하게 나타나므로 정확한 예후 예측이 절실히 필요하다. 이러한 위암의 임상적 다양성은 위암의 분자생물학적인 다양성 때문이라고 할 수 있고, 이는 여러 유전자의 변화에 의한다. 그러나, 이에 대한 연구는 아직 미미한 실정이며 위암의 예후를 예측할 수 있는 바이오마커로 승인되어 사용중인 것은 없는 상태이다.
이에 본 발명자들은 위암의 예후를 예측할 수 있는 바이오마커를 규명하기 위해 노력한 결과, 높은 ERH 발현을 보이는 위암 환자군이 낮은 ERH 발현을 보이는 환자군 대비 생존율, 전이 및 침투율이 낮은 것을 확인하고 본 발명을 완성하게 되었다.
본 발명의 목적은 ERH (Enhancer of Rudimentary Homolog) 유전자 또는 상기 유전자가 암호화하는 단백질을 포함하는, 위암 예후 예측용 마커 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 마커 유전자의 mRNA 또는 상기 유전자가 암호화하는 단백질 수준을 측정하는 제제를 포함하는, 위암 예후 예측용 조성물, 및 상기 조성물을 포함하는 위암 예후 예측용 키트, 및 예후 예측을 위한 정보제공방법을 제공하는 것이다.

이하, 본 발명을 상세히 설명한다.
본 발명은 위암 개체로부터 분리된 시료에서 측정된 ERH(Enhancer of Rudimentary Homolog) mRNA 또는 단백질 수준이 대조군의 수준보다 높으면 위암 예후가 대조군 대비 좋을 것으로 판단하는 위암 예후 예측을 위한 정보제공방법에 관한 것이다.
이하 본 발명의 일 구현예에 따른 방법을 상세히 설명한다.
‘위암 개체’ 는 위암 세포 또는 위암 조직을 가지는 개체로서, 이는 인간을 포함한 포유류일 수 있다.
'시료'는 위암 개체로부터 분리된 것으로서, 그 예로는 조직, 세포, 혈액, 혈청, 혈장, 타액 또는 뇨일수 있고, 구체적으로는 진단 대상으로부터 분리된 암 조직일 수 있으나, 이에 제한되는 것은 아니다.
'ERH'(NM_004450) 는 기초 상동체의 인핸서로 성인과 태아조직 모두에서 편재적으로 발현되며, 주로 핵에 위치하는 것으로 ERH와 결합하는 단백질들은 전사 조절, 세포 성장 또는 세포 주기 진행에 관여하는 것으로 보고된 유전자 또는 단백질이다.
mRNA 수준을 측정하는 것은 당 분야에 공지된 방법으로 수행될 수 있으며, 예를 들면 노던 블로팅, RT-PCR, real-time PCR 등을 이용하여 측정할 수 있으나, 이에 제한되는 것은 아니다.
단백질 수준을 측정하는 것은 당 분야에 공지된 방법으로 수행될 수 있으며, 예를 들면 웨스턴 블로팅, 면역염색, 면역 전기영동, 단백질 면역 침전, 효소 결합 면역 침강 분석법, 액체 크로마토그래피 등을 이용하여 측정할 수 있으나, 이에 제한되는 것은 아니다. 예를 들면 위암 조직의 면역 염색으로 ERH 단백질이 구분되도록 염색하여 확인될 수 있다.
대조군은 비교 대상으로서, 정상 개체의 시료로부터 얻어진 값, 다른 위암 개체의 시료로부터 얻어진 값, 복수의 위암 개체의 시료에서 얻어진 값의 평균값 등일 수 있다.
'예후'는 환자의 상태나 질병의 치료 결과에 대한 예측을 의미하는 것으로, “예후가 좋다”는 피검자가 저위험군으로 예측되는 경우에 사용되고, 재발 가능성이 낮은 경우, 전이성 암이 아니거나 전이될 가능성이 낮은 경우, 1기 또는 2기 위암이거나 3기 또는 4기 위암으로 발전될 가능성이 낮은 경우, 위암으로 사망할 가능성이 낮은 경우에 사용한다. “예후가 좋지 않다”는 피검자가 고위험군으로 예측되는 경우에 사용되고, 재발 가능성이 높은 경우, 전이성 암이거나 전이될 가능성이 있는 경우, 3기 또는 4기 위암이거나 3기 또는 4기 위암으로 발전될 가능성이 있는 경우, 위암으로 사망할 가능성이 높은 경우를 의미한다. 이외에도 2개 이상의 개체에서 상대적으로 예후가 좋고 나쁨을 나타낼 수도 있다. 본 발명의 일 실시예에서, 예후가 좋지 않은 개체는 시간이 지남에 따라 해당 개체의 위암이 더 높은 TNM (Tumor Node Metastasis) 단계로 진행될 것으로 예측되는 환자를 의미한다.
“예후 예측”은 “예후 진단”과 상호 교환적으로 사용 가능하고, 특정 질병 또는 질환에 걸린 한 객체의 예후 (예를 들어, 전-전이성 또는 전이성 암 상태의 동정, 암의 단계 결정, 치료에 대한 암의 반응성 결정, 또는 고위험군 및 저위험군의 결정)를 판정하는 것, 또는 테라메트릭스(therametrics)(예를 들어, 치료 효능에 대한 정보를 제공하기 위하여 객체의 상태를 모니터링 하는 것)를 포함한다.
또한 본 발명은 ERH (Enhancer of Rudimentary Homolog) 활성 또는 발현을 증가시키는 물질을 포함하는 위암의 전이 억제용 약학 조성물에 관한 것이다.
ERH 활성 또는 발현을 증가시키는 물질은 ERH 단백질의 활성을 증가시키는 물질, ERH 유전자, mRNA, 또는 단백질의 발현을 증가시키는 물질이라면 제한없이 적용될 수 있다. 그 종류는 예를 들면 저분자량 약물, 유전자 약물, 단백질 약물, 추출물, 핵산, 올리고뉴클레오티드, 단백질, 펩타이드, 항체, RNA, DNA, PNA, 압타머, 화학약품, 효소, 아미노산, 당, 지질, 화합물(천연화합물 및/또는 합성화합물) 및 이들을 구성하는 요소로 이루어진 군에서 선택된 적어도 하나일 수 있으나, ERH를 코딩하는 유전자의 mRNA 또는 상기 유전자가 코딩하는 단백질의 발현을 증가시키는 효능을 가진 물질이라면 특별히 제한되지 아니한다. ERH를 코딩하는 유전자(예를 들면 cDNA)가 도입된 벡터를 사용할 수도 있고, 그 유전자로는 적용하고자 하는 개체와 동일종 또는 다른 종 유래 유전자를 사용할 수 있다. 예를 들면 인간의 경우 인간 유래의 ERH를 코딩하는 유전자(NM_004450)를 사용할 수 있다.
'전이'는 장기 또는 조직에 발생한 암 병소로부터 암 세포가 이동하여 다른 장기 또는 조직에 결합하여 증식하는 상태를 의미한다.
약학 조성물은 약학적으로 허용가능한 담체를 추가로 포함할 수 있으며, 담체와 함께 제제화될 수 있다. "약학적으로 허용가능한 담체"란 생물체를 자극하지 않고 투여 화합물의 생물학적 활성 및 특성을 저해하지 않는 담체 또는 희석제를 말한다. 액상 용액으로 제제화되는 조성물에 있어서 허용되는 약제학적 담체로는, 멸균 및 생체에 적합한 것으로서, 식염수, 멸균수, 링거액, 완충 식염수, 알부민 주사용액, 덱스트로오스 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다.
본 발명의 조성물은 상기 ERH mRNA 또는 그 단백질의 발현을 증가시키는 물질을 유효성분으로 포함하는 어떠한 제형으로도 적용가능하며, 경구용 또는 비경구용 제형으로 제조할 수 있다. 본 발명의 약학적 제형은 구강(oral), 직장(rectal), 비강(nasal), 국소(topical; 볼 및 혀 밑을 포함), 피하, 질(vaginal) 또는 비경구(parenteral; 근육 내, 피하 및 정맥내를 포함) 투여에 적당한 것 또는 흡입(inhalation) 또는 주입(insufflation)에 의한 투여에 적당한 형태를 포함한다.
또한 본 발명은 위암 개체로부터 분리된 샘플 내 ERH mRNA 또는 그 단백질의 발현을 증가시키는 물질을 선별하는 단계를 포함하는 위암의 전이 억제제 후보물질의 스크리닝 방법에 관한 것이다.
스크리닝 방법은 피검물질을 개체에 처리하여 ERH mRNA 또는 그 단백질의 발현 수준이 증가한다면 위암세포의 이동 및 침입이 억제되는 것이므로, 그러한 피검물질을 위암의 전이 억제제 후보물질로 선별할 수 있다.
또한, 상기 방법은 위암을 지닌 개체로부터 분리된 시료에 피검물질을 처리하여, 처리 전후 ERH mRNA 또는 단백질의 발현 정도를 비교할 수도 있고, 여러 개체 중 일부 개체에 피검물질을 처리하여 대조군과 상기 발현 수준을 비교할 수도 있다.
'후보물질'은 새롭게 합성된 또는 공지된 물질로 위암의 전이 억제 효과를 나타낼 것으로 기대되는 물질을 제한 없이 포함할 수 있고, ERH mRNA 또는 단백질의 발현량에 영향을 미치는지 여부를 검사하기 위하여 스크리닝에서 이용되는 미지의 물질을 의미한다. 상기 후보물질은 화학물질, 뉴클레오타이드, 안티센스-RNA, siRNA(small interference RNA) 및 천연물 추출물을 포함하나, 이에 한정되는 것은 아니다.
이외, 본 발명의 스크리닝 방법에 있어서, 시료, 발현 수준의 측정 등에 관련한 사항은 전술한 바와 같다.
실시예
1. 실험 방법
(1) 조직 마이크로어레이 (TMA)용 위 조직 샘플
경상대학교병원에서 위 절제술을 받은 319 명의 GC 조직 표본 (2004.1.1~2007.12.31) 및 환자에 대한 의료 정보와 임상병리학적 데이터를 수집했다. 의료 차트를 통해 사망률을 평가하였다. 이 연구는 헬싱키 선언 (2013년 개정)에 따라 수행되었으며, 경상대학교병원 기관심의위원회의 승인(GNUHIRB 2009-54) 및 모든 환자의 동의를 얻어 수행되었다.
(2) TMA 분석 및 조직 면역염색 (IHC)
종양 파라핀 블록 (기증자 블록)에서 직경 2mm 핵심조직 생검 샘플을 얻었다. 이 표본을 새로운 파라핀 블록에 삽입하고, IHC 염색은 4μm 절편에 수행되었다. 간단히 말해서, 조직 절편을 탈파라핀화 및 재수화하고, 슬라이드를 ERH 특이 항체 (토끼 다클론 항체, 1:25; Atlas Antibodies AB)로 실온에서 60 분간 배양하였다. 발현을 검출하기 위해 UltraVision LP 검출 시스템 (Lab Vision Corporation)을 사용하였다. ERH 발현에 대한 스코어링은 임상병리학적 데이터를 모르는 상태에서 병리학자(K.H. Ko)에 의해 수행되었다. 핵에서 조직 화학적 반응 정도는 ERH 양성 세포의 비율에 따라 점수를 매겼다 (도 1): 2+ (25-49 %), 3+ (50-74 %).
(3) 통계학적 분석
모든 통계 분석은 SPSS Statistics 버전 24 (IBM SPSS, Inc.)를 사용하여 수행되었고, 데이터는 평균±SD를 나타낸다. 평균값에 대한 통계적 분석은 t-test를 통해 수행되었으며, 빈도에 대한 통계적 분석은 χ2 테스트로 수행되었다. 카플란-메이어 방법은 생존 분석에 적용되었다. 모든 분석에서 0.05 미만의 P 값은 통계적으로 유의한 것으로 간주된다.
(4) 세포 배양
인간 위 선암 세포주 MKN74와 SNU601은 한국 세포주 은행에서 구입하였다. 세포는 열로 비활성화시킨 FBS (Gibco) 및 페니실린 (100U/mL, Thermo Fisher Scientific Inc.) 가 포함된 RPMI 1640 배지 (Gibco)에서 37℃, 5% CO
2 대기에서 배양되었다.
(5) 웨스턴 블롯 분석
세포를 1% 프로테아제 억제제 칵테일 (Sigma)이 포함된 용해 (RIPA) 버퍼에 현탁하고 라이세이트를 4 ℃에서 15분간 13,000 rpm에서 원심 분리하였다. 단백질 농도는 Bio-Rad 단백질 분석 키트를 사용하여 정량되었다. 그런 다음 추출된 단백질의 30μg 샘플을 15% SDS-폴리 아크릴아미드 겔 전기 영동을 거쳐 PVDF 막 (iBlot 2PVDF Regular Stacks, Invitrogen)으로 트랜스퍼 하였다. 막은 TBST 용액 [10mM Tris-HCl (pH 8.0), 150mM NaCl 및 0.05 % Tween 20] 내 5% 무지방 분유와 함께 실온에서 1시간 동안 블로킹하였다. 단백질 블롯은 1차 항체 (ERH 1:1000, Sigma)로 4 ℃에서 밤새 배양되었다. 검출은 향상된 화학 발광 시약 (ECL, Thermo)을 사용하여 수행하였고, β-액틴 (1:1000, Santa Cruz Biotech) 항체를 사용하여 로딩된 샘플의 양을 표준화하였다.
(6) RNA 간섭 실험
ERH 유전자를 표적으로 하는 두 개의 서로 다른 siRNA 올리고 뉴클레오티드 듀플렉스 (siERH-3 및 siERH-4로 지정됨)는 Dharmacon에서 구입하였다. siERH-3의 서열은 5'-UCAGUCAGUUFUUUFAUUU-3'이고, siERH-4의 서열은 5'-GAACUUAUGCUGACUACGA-3'이다. 각 siRNA 듀플렉스의 일시적 형질 도입은 제조업체의 지침에 따라 Lipofectamine
TM 3000 (Invitrogen) 형질도입 시약을 사용하여 수행되었다.
(7) ERH 발현 플라스미드의 구축 및 형질 도입
인간 ERH cDNA는 OriGene (RC200367)에서 구입했다. 제조업체의 지침에 따라 Lipofectamine
TM 3000 (Invitrogen)을 사용하여 ERH 발현 플라스미드를 세포에 형질 도입하였다. 48시간 후, 세포 선별을 위해 네오마이신으로 처리하였다. 네오마이신 내성 클론에서 ERH 발현은 면역 블롯팅에 의해 조사되었다. pCMV6 공벡터는 대조군으로 사용되었다.
(8) 증식 분석
3-(4,5-디메틸티아졸-2-일)-2,5-디페닐테트라졸륨 브로마이드 (MTT) 분석 (Sigma)을 사용하여 세포 생존력을 분석하였다. MKN74 및 SNU601 세포를 웰당 2x10
4 세포로 24 웰 플레이트에 접종하였다. 그 다음 이 플레이트는 37℃에서 5% CO
2-와 함께 가습 인큐베이터에서 배양되었다. 24, 48, 72 또는 96시간 동안 배양한 후, MTT를 포르마잔으로 전환하기 위해 3시간 동안 200μL의 MTT 용액 (0.5mg/mL)을 첨가하여 MTT 분석을 수행하였다. 상청액을 제거하고 500μL의 DMSO를 각 웰에 첨가하였다. 570nm에서 용액의 광학 밀도 (OD)는 VersaMax ELISA 마이크로 플레이트 리더 (Molecular Devices/Versamax)를 사용하여 측정되었다. 각 실험은 독립적으로 세 번 반복되었다.
(9) 침입 및 이동 분석
암 세포의 침입 능력은 Matrigel-기반 Transwell 시스템을 사용하여 평가되었다. Transwell 시스템의 상부 챔버를 Matrigel (BD Bioscience # 356234)으로 코팅하고 37℃에서 4시간 또는 밤새 배양하였다. MKN74 및 SNU601 세포를 1x10
5로 조정하고 FBS가 없는 배지에서 Transwell의 상부 챔버에 접종하였다. 37℃의 5% 대기 CO2 인큐베이터에서 48 또는 72시간 동안 배양한 후 면봉으로 닦아 막 윗면의 세포를 제거하고, 밑면의 세포는 4% 포름알데히드 고정액(PBS에 0.1% Triton X-100 포함)으로 고정시키고, 4,6-디아미디노-2-2-페닐인돌 (DAPI) 용액으로 염색했다. 무작위로 선택된 5개의 10배 확대 필드를 카운팅하여 정량화를 수행했다. 세포 침입에 대한 값은 3중 실험에 대해 필터당 5개 이상의 필드에서 현미경 필드당 평균 세포 수로 표현된다. 실험은 3회 이상 독립적으로 수행되었다. 상처 치유 분석을 위해, MKN74 세포 및 SNU601 세포 (70μL 배지에 웰당 1.4 x 105)를 6-웰 플레이트의 배양 인서트-2-웰 (ibidi)에 접종하였다. 인서트 웰을 제거한 후, 세포를 새로운 배지에서 배양하였다. 이동 분석의 현미경 사진은 현미경으로 0, 24 및 48시간에 촬영되었다. NIH ImageJ 소프트웨어를 사용하여 세포 이동을 정량화했다. 데이터는 3중 독립 실험에 대한 평균± SD로 표시된다.
2. 실험 결과
(1) TMA 샘플에서 ERH-과발현된 환자는 종양 침입, TNM 분류 및 암 재발률이 감소하였다.
TMA 블록으로 319 개의 GC 조직 샘플에서 ERH 발현을 분석하기 위해 면역 조직 화학 염색을 수행했다. 피험자는 평균 연령이 62.1±11.1 세인 남자 208 명과 여자 111 명으로 구성되었다. 평균 종양 크기는 4.2±2.6 cm이었고 전이된 림프절 (LN)의 평균 수는 2.2였다.
종양 TNM 단계 분류는 다음과 같다: 1기 (60.2%, n=192); 2기 (17.9%, n=57); 및 3기 (21.9%, n=70). 평균 추적 기간은 55.5 개월이었고 GC는 18.5% (n=59)에서 재발하였다. 암 세포의 세포질에서 ERH 발현 수준을 결정하기 위해 사용되는 염색 강도는 각 조직마다 달랐다. 319 개의 GC 조직 샘플 중 11.9%은 2+, 88.1%은 3+을 기록했다. 점수가 2+ 및 3+ 인 샘플은 각각 ERH 단백질 저발현 (ERH-UE) 및 과발현을 갖는 것으로 간주되었다 (도 1).
면역 조직 화학적 분석 결과는 ERH-OE (3점 이상) 환자와 ERH-UE (2점 이상) 환자를 비교하였다. WHO 분류, LN 전이, 암 관련 사망 측면에서 두 그룹간에 차이는 없었다 (P> 0.05). 그러나 ERH-UE 그룹에서 진행된 GC (AGC) 비율은 ERH-OE 그룹에서보다 유의하게 높았다 (73.7% vs. 43.1%; P<0.01). TNM 단계 측면에서 ERH-UE 그룹에서의 높은 단계 비율은 ERH-OE 그룹에서보다 유의하게 높았다 (1기, 2기, 3-4기; 각각 28.9% vs. 64.4%, 42.1% vs. 14.6% 및 28.9% vs. 21%, P<0.01).
또한 암 재발률은 ERH-OE 그룹에서보다 ERH-UE 그룹에서 더 높았다 (31.6% vs. 16.7%, P=0.04) (표 1). 두 그룹 간의 누적 생존율 분석에 따르면 ERH-OE 그룹의 환자 (85.8 개월, 95% CI: 82.7 ~ 88.9 개월)는 ERH-UE 그룹 (67.7 개월, 95% CI: 58.7 ~ 76.7 개월)보다 누적 생존 시간이 더 길었고, 그 차이는 유의미했다 (로그-순위 테스트, P=0.04) (도 2).

(2) ERH-OE는 GC 세포 이동 및 침입을 억제하지만, 시험관 내 세포 증식에는 영향을 미치지 않는다.
ERH-OE가 암 세포의 증식, 이동 및 침입에 미치는 영향을 SNU601 및 MKN74 세포에서 평가했다. 분석 전에, 두 SNU601 세포 그룹 (oeERH-1 및 2) 및 MKN74 세포 그룹 (oeERH-1 및 2)의 세포에서 ERH-OE를 ERH에 대한 항체로 면역 블롯팅하여 확인하였다 (도 3A). ERH-과발현 SNU601 및 MKN74 세포와 각 대조군 세포 사이의 증식 속도에는 차이가 없었다 (도 3B). 그러나, ERH-과발현 세포주의 두 그룹의 이동 및 침입 비율은 대조군 세포보다 낮았다 (도 4A, B).
(3) ERH 발현의 감소는 GC 세포 이동 및 침입을 유발하지만 시험관 내 증식에는 영향을 미치지 않았다.
GC 세포의 증식, 이동 및 침입에서 ERH의 기능은 SNU601 및 MKN74 세포에서 각각 ERH 발현을 억제하기 위한 ERH-특이적 siRNA (siERH-1 & 2)를 사용하여 평가되었다. 두 가지 다른 ERH siRNA의 특이성이 처음 확인되었다 (도 5A).
ERH가 감소된 SNU601 및 MKN74 세포의 이동률은 각 대조군 세포에 비해 증가하였음에도 불구하고 (도 5B), ERH가 감소된 SNU601 및 MKN74 세포의 증식률은 대조군 세포에 비해 크게 다르지 않았다 (도 6A). ERH 감소는 세포 침입률에 유사한 영향을 미쳤으며, 이에 따라 ERH 감소된 SNU601 및 MKN74 세포의 침입률은 대조군 세포에 비해 현저하게 증가했다 (도 6B). 이러한 결과는 ERH 발현이 GC 세포의 침입을 감소시킨다는 것을 의미한다.
Claims (6)
- 위암 개체로부터 분리된 시료에서 측정된 ERH(Enhancer of Rudimentary Homolog) mRNA 또는 단백질 수준이 대조군의 수준보다 높으면 위암 예후가 대조군 대비 좋을 것으로 판단하는 위암 예후 예측을 위한 정보제공방법.
- 청구항 1에 있어서, 상기 시료는 위암 조직인 위암 예후 예측을 위한 정보제공방법.
- 청구항 1에 있어서, 상기 ERH 단백질 수준은 위암 조직의 면역 염색으로 확인되는 위암 예후 예측을 위한 정보제공방법.
- ERH (Enhancer of Rudimentary Homolog) 활성 또는 발현을 증가시키는 물질을 포함하는 위암의 전이 억제용 약학 조성물.
- 청구항 4에 있어서, 상기 물질은 ERH를 코딩하는 유전자가 도입된 벡터인, 위암의 전이 억제용 약학 조성물.
- 위암 개체로부터 분리된 샘플 내 ERH(Enhancer of Rudimentary Homolog) mRNA 또는 그 단백질의 발현을 증가시키는 물질을 선별하는 단계를 포함하는 위암의 전이 억제제 후보물질의 스크리닝 방법.
