The microneedle layer 60 can be provided on the opposite side of the conductive layer 64 in the adhesive layer 56, that is, on the surface in contact with the user's skin when the sheet electrode 50 is attached to the user's skin.
When using the beauty device 100, first, the sheet electrode 50 is attached to a desired position on the user's skin S. At that time, the orientation of the second portion 50b is determined so that the apparatus main body 10 can be arranged at an appropriate position on the surface of the user's skin S. Next, the bottom wall 12a of the housing main body 12 is made to face the user's skin S, and as shown in FIG. 11, the first electrode 16 is brought into contact with the second portion 50b of the sheet electrode 50, and the second electrode is used. The device body 10 is attached to the surface of the user's skin S by arranging the 18 so as to be in contact with the user's skin S. Next, the switch 22 is turned on to apply a pulse wave to the user's skin S. As an example of the pulse wave, it may be a pulse-shaped pulse wave as shown in FIG. 120.
Protoc.
<Example 1>
<Collagen production promoting action by electrical stimulation>
METHODS: A cell reactor was prepared by the method described in N. Tandon et al., Nat. Protocol. 4 (2009) 155-173. A resistance of 1 kΩ was applied to human fibroblasts (Normal Human Thermal Fiberblast manufactured by ATCC in the United States) in a cell reactor using a voltage divider, and a pulse wave generating device was used to generate 1 Hz, 500 Hz, 2000 Hz pulse waves (5 V, 5 μA). The stimulus was given for 2 hours. FIG. 120 shows a waveform at 2000 Hz. Even in the case of 1 Hz and 500 Hz, they have the same pulse-like waveform except that only the frequency is different.
The amount of collagen in fibroblasts was measured as follows using a Sircol ™ soluble collagen assay kit (Biocolor Ltd.).
1.0 ml of Sircol ™ dye reagent is added to a tube containing 100 μl of the above fibroblast supernatant. Cover the tube and mix manually at 5 minute intervals for a total of 30 minutes. The tube is then transferred to a microcentrifuge to remove unbound dye by suction. After adding 750 μl of ice-cold acid-salt wash reagent, transfer the tube to a microcentrifuge and centrifuge at 12,000 rpm for 10 minutes. The acid-salt cleaning reagent is removed by suction and 250 μl of alkaline reagent is added to each tube. The collagen-binding pigment is released into the solution using a vortex mixture. Then, 200 μl of the solution in each tube is transferred to the wells of the 96-well plate. Read at 555 nm using a plate reader (Thermo Electron Corporation). The quantification is performed by triplicate, and the case where no electrical stimulation is applied to the cells is used as a control and contrasted. The concentration of collagen produced is determined by contrasting with the calibration curve and standardized by the number of cells lysed by the assay.
The results are shown in FIG. When the frequencies were 500 Hz and 2000 Hz, high collagen production promoting effects were observed, about 2.0 times and about 6.0 times, respectively, as compared with the subject. In particular, remarkable collagen production is observed when the frequency is 2000 Hz, and it can be seen that the higher the frequency, the more the collagen production is promoted. The apparatus of the present invention has the effect of promoting collagen production in skin cells simply by applying a skin electrode to the skin and applying a pulse wave at a high frequency.

<Moisturizing effect by electrical stimulation>
Subject: 10 healthy men in their 40s and 60s who have no skin problems Equipment used:
-Device of the present invention (self-made)
・ Omron skin gel pad (conductive electrode (carbon black) adhesive gel, model number: HVLLPAD) 2 cm x 2 cm
-Courage + Khazaka Corneometer (model number: CM825)
Method: The experiment was performed at a temperature of 24 ° C. and a humidity of 45%. After washing the skin with soap and water to remove water, the skin water content was measured with a corneometer. The gel pad was attached to the skin. The gel pad was connected to the device of the present invention, and the skin was subjected to electrical stimulation of a pulse wave of 5 V, 250 μA, 500 Hz for 20 minutes. The pad was removed from the skin, and at 15, 30, 45, and 60 minutes, the water content of the skin was measured over time with a corneometer. As a control, the water content was also measured when the cosmetic was applied for the same time without the pulse wave and when neither the pulse wave nor the cosmetic was given.
The result of skin water content is shown in FIG. The device of the present invention has an effect of improving wrinkles on the skin simply by applying a skin electrode to the skin and applying a pulse wave at a high frequency.
<実施例4>
<Example 4>
<Wrinkle improving effect by multiple electrical stimuli>
The same subjects as in Examples 2 and 3 were given the same 20-minute electrical stimulation except that the electrical stimulation was applied three times after 0 hour, 24 hours, and 48 hours. Skin wrinkles were imaged using an ANTERA 3D camera and the wrinkle depth was quantified by determining the Indentation Index (μm) using Miravex 1 software.
The result of wrinkle measurement is shown in FIG. It decreased by about 6.0 by the first stimulus, but then returned to the original level, but decreased by about 2.0 by the second stimulus and further decreased by the third stimulus. It was found that the wrinkle improving effect was further enhanced by applying a plurality of stimuli in this way as compared with a single stimulus.
From the above results, it was found that the beauty device of the present invention has the effects of promoting collagen production in the skin, moisturizing, and improving wrinkles. When the collagen production of the skin is promoted, moisturized, and wrinkles are improved, it is effective for the elasticity and elasticity of the skin.
<実施例5>
<Example 5>
For the purpose of demonstrating that the device of the present invention exerts an excellent cosmetic effect, the effect was compared with a commercially available facial device. As a commercially available facial device, another company's product was used. The device activates the skin by passing a weak electric current through the skin. The wrinkle reducing effect was investigated by determining the indentation index (μm) and quantifying the wrinkle depth in the same manner as in Example 4.
The result is shown in FIG.
“Before application” in measurement 1 on the left side of the figure indicates the dent index of the application site immediately before application by another company's product.
“Gel application” in measurement 2 indicates the dent index of the skin area to which only gel application is applied. When using other companies' products, it is necessary to apply gel to the application area before applying current stimulation. Therefore, in order to eliminate the beauty effect of the gel application itself, the dent index in which only the gel application was applied was measured.
“After application” in measurement 3 indicates the dent index at the location where the gel was applied and the current stimulus was applied. The current stimulation was performed by giving the current stimulation to the skin for 5 minutes according to the instruction manual of the product of another company.
Since there is no difference in the dent index between measurements 2 and 3, it is considered that the effect seen by applying the products of other companies is not the effect of electrical stimulation but the effect of gel application. Therefore, it can be seen that the wrinkle reduction effect of the current stimulation of other companies' products cannot be expected so much if it is limited to the effect using the dent index as an index.
The right side of FIG. 18 shows the amount of change in the depression index with respect to “before application” of measurement 1. The amount of change "after application" is the value obtained by subtracting the amount of change in <gel application only> from the amount of change in <gel application + current stimulation>, and the effect of using the dent index as an index is the current stimulation by other companies' products. It is clear from this figure that we cannot expect it.
A direct comparison test was conducted between the apparatus of the present invention and a product of another company under the same environment. The result is shown in FIG. 197.
"Before application" on the left side of the figure shows the dent index of the part where only the gel is applied in the case of another company's product, and in the case of the present invention, shows the dent index of the skin in the state where no treatment is applied.
“After application” indicates the dent index of the portion where the gel is applied and the current stimulus is applied in the case of another company's product, and in the case of the present invention, the dent index measured after applying a 0.5 kHz pulse for 20 minutes is indicated. In the case of the device of the present invention, no cosmetics such as gel is used. The right side of FIG. 19 shows the amount of change in the depression index with respect to “before application”.
From both figures, the wrinkle improvement effect using the dent index as an index cannot be expected with the current stimulation by the products of other companies, but the clear effect was confirmed by the apparatus of the present invention.
