嗅覚受容体の化学物質に対する応答活性を向上させる技術を提供することを課題とし、該課題を、昆虫嗅覚受容体タンパク質中の特定の部位において特定の条件を満たすように変異を導入すること、により解決する。

Description

変異型昆虫嗅覚受容体タンパク質
 本開示は、変異型昆虫嗅覚受容体タンパク質等に関する。
 ヒトの特定の疾患や精神状態等を特徴付ける匂い物質群が同定されており、診断マーカーとしての利用価値が高いことから、これらをターゲットとした様々な匂いセンサの開発が盛んになっている。生物の嗅覚受容体は、多様性、感度、選択性等の面で半導体等の従来の匂いセンサ素子にはない優れた特性を有することから、嗅覚受容体をセンサ素子とした新しい匂いセンサの開発が期待されている。
 一般に匂いマーカーは低濃度であるため、高活性な嗅覚受容体を作出することが望ましい。現状、嗅覚受容体において、置換されることにより、透過するイオンの選択性が変化するアミノ酸部位が報告されている。

 本開示は、嗅覚受容体の化学物質に対する応答活性を向上させる技術を提供することを課題とする。
 本発明者は上記課題に鑑みて鋭意研究を進めた結果、昆虫嗅覚受容体タンパク質中の特定の部位において特定の条件を満たすように変異を導入することにより、化学物質に対する応答活性を向上させることができることを見出した。本発明者は、この知見に基づいてさらに研究を進めた結果、本開示の発明を完成させた。即ち、本開示は、下記の態様を包含する。
 項1. 変異型昆虫嗅覚受容体タンパク質であって、
対応する野生型昆虫嗅覚受容体タンパク質中の一般式(1):

下記条件1~3:
(条件1)前記アミノ酸配列Bにおいて、X3及び/又はX4が置換しており、X3が分岐鎖アミノ酸であり且つ/或いはX4が無電荷極性アミノ酸であること、
(条件2)前記アミノ酸配列AにおいてX5及びX6が共に正電荷極性アミノ酸である場合に、前記アミノ酸配列Bにおいて、X6が正電荷極性アミノ酸以外のアミノ酸であること、及び

 1.定義等
 本明細書中において、「含有」及び「含む」なる表現については、「含有」、「含む」、「実質的にからなる」及び「のみからなる」という概念を含む。
 本明細書において、アミノ酸配列の「同一性」とは、2以上の対比可能なアミノ酸配列の、お互いに対するアミノ酸配列の一致の程度をいう。従って、ある2つのアミノ酸配列の一致性が高いほど、それらの配列の同一性又は類似性は高い。アミノ酸配列の同一性のレベルは、例えば、配列分析用ツールであるFASTAを用い、デフォルトパラメータを用いて決定される。若しくは、Karlin及びAltschulによるアルゴリズムBLAST(Karlin S, Altschul SF.“Methods for assessing the statistical significance of molecular sequence features by using general scoringschemes”Proc Natl Acad Sci USA.87:2264-2268(1990)、KarlinS,Altschul SF.“Applications and statistics for multiple high-scoring segments in molecular sequences.”Proc Natl Acad Sci USA.90:5873-7(1993))を用いて決定できる。このようなBLASTのアルゴリズムに基づいたBLASTXと呼ばれるプログラムが開発されている。これらの解析方法の具体的な手法は公知であり、National Center of Biotechnology Information(NCBI)のウェブサイト(http://www.ncbi.nlm.nih.gov/)を参照すればよい。また、塩基配列の「同一性」も上記に準じて定義される。
 本明細書中において、「保存的置換」とは、アミノ酸残基が類似の側鎖を有するアミノ酸残基に置換されることを意味する。例えば、リジン、アルギニン、ヒスチジンといった塩基性側鎖を有するアミノ酸残基同士で置換されることが、保存的な置換にあたる。その他、アスパラギン酸、グルタミン酸といった酸性側鎖を有するアミノ酸残基;グリシン、アスパラギン、グルタミン、セリン、トレオニン、チロシン、システインといった非帯電性極性側鎖を有するアミノ酸残基;アラニン、バリン、ロイシン、イソロイシン、プロリン、フェニルアラニン、メチオニン、トリプトファンといった非極性側鎖を有するアミノ酸残基;トレオニン、バリン、イソロイシンといったβ-分枝側鎖を有するアミノ酸残基;チロシン、フェニルアラニン、トリプトファン、ヒスチジンといった芳香族側鎖を有するアミノ酸残基同士での置換も同様に、保存的な置換にあたる。
 本明細書において、DNA、RNAなどのヌクレオチドには、次に例示するように、公知の化学修飾が施されていてもよい。ヌクレアーゼなどの加水分解酵素による分解を防ぐために、各ヌクレオチドのリン酸残基(ホスフェート)を、例えば、ホスホロチオエート(PS)、メチルホスホネート、ホスホロジチオネート等の化学修飾リン酸残基に置換することができる。また、各リボヌクレオチドの糖(リボース)の2位の水酸基を、-OR(Rは、例えばCH3(2´-O-Me)、CH2CH2OCH3(2´-O-MOE)、CH2CH2NHC(NH)NH2、CH2CONHCH3、CH2CH2CN等を示す)に置換してもよい。さらに、塩基部分(ピリミジン、プリン)に化学修飾を施してもよく、例えば、ピリミジン塩基の5位へのメチル基やカチオン性官能基の導入、あるいは2位のカルボニル基のチオカルボニルへの置換などが挙げられる。さらには、リン酸部分やヒドロキシル部分が、例えば、ビオチン、アミノ基、低級アルキルアミン基、アセチル基等で修飾されたものなどを挙げることができるが、これに限定されない。また、ヌクレオチドの糖部の2´酸素と4´炭素を架橋することにより、糖部のコンフォーメーションをN型に固定したものであるBNA(LNA)等もまた、好ましく用いられ得る。
 嗅覚受容体タンパク質は、7回膜貫通構造を有する膜タンパク質であり、そのアミノ末端(以下、「N末端」という場合もある。)からカルボキシル末端(以下、「C末端」という場合もある。)に向かって順に、N末端領域(NT)、第1膜貫通ドメイン(TM1)、第1細胞外ループ(EC1)、第2膜貫通ドメイン(TM2)、第1細胞内ループ(IC1)、第3膜貫通ドメイン(TM3)、第2細胞外ループ(EC2)、第4膜貫通ドメイン(TM4)、第2細胞内ループ(IC2)、第5膜貫通ドメイン(TM5)、第3細胞外ループ(EC3)、第6膜貫通ドメイン(TM6)、第3細胞内ループ(IC3)、第7膜貫通ドメイン(TM7)、及びC末端領域(CT)が連結されて構成される。本開示において、各領域は、TMpred(K. Hofmann, W. Stoffel, TMbase - a database of membrane spanning proteins segments, Biol. Chem. Hoppe-Seyler, 374 (1993), p. 166、https://embnet.vital-it.ch/software/TMPRED_form.html)を用いた構造予測(条件はデフォルト)により決定される。
 本明細書において、アミノ酸の変異は、具体的には、アミノ酸の欠失、置換、挿入、又は付加である。嗅覚受容体タンパク質は、化学物質応答活性への影響が少ないアミノ酸が欠失していてもよく、例えば膜貫通領域以外のアミノ酸の一部が欠失している嗅覚受容体ペプチドであってもよい。
 本明細書において、ORは嗅覚受容体(Odorant receptor)を示し、Orcoは嗅覚受容体共受容体(Odorant recetor co-receptor)を示し、Dmはキイロショウジョウバエ(Drosophila melanogaster)を示し、Bmはカイコガ(Bombyx mori)を示し、Agはガンビエハマダラカ(Anopheles gambiae)を示し、Aaはネッタイシマカ(Aedes aegypti)を示す。
 本明細書において、化学物質応答活性とは、嗅覚受容体が化学物質を認識し、その嗅覚受容体と嗅覚受容体共受容体とが形成した嗅覚受容体複合体が活性化されてイオンチャネル活性を示す性質をいう。嗅覚受容体の化学物質応答活性は、化学物質と接触した嗅覚受容体と嗅覚受容体共受容体とが形成する嗅覚受容体複合体のイオンチャネル活性を指標として測定することができる。例えば、(a)嗅覚受容体、(b)嗅覚受容体共受容体、及び (c)嗅覚受容体が応答した場合に細胞内に流入するイオン(カルシウムイオン等)により発色又は発光するタンパク質を発現する細胞と、化学物質とを接触させ、当該細胞の発光量を測定する。測定された発光量が多い程、嗅覚受容体の化学物質の応答活性が高いと判定する。後述の試験例1-2及び試験例1-3に、化学物質応答活性を測定した具体例が記載されている。
 各嗅覚受容体が応答活性を有する化学物質(匂い物質等)は、公知である、或いは公知のスクリーニング方法によって決定することができる。例えば、DmOR47aが応答活性を有する化学物質にはペンチルアセテートが含まれ、BmOR56が応答活性を有する化学物質にはシスジャスモンが含まれ、AgOR1が応答活性を有する化学物質にはフェノールが含まれ、AaOR9が応答活性を有する化学物質にはスカトールが含まれ、AgOR2が応答活性を有する化学物質にはインドールが含まれる。
 2.嗅覚受容体タンパク質
 本開示は、その一態様において、変異型昆虫嗅覚受容体タンパク質であって、対応する野生型昆虫嗅覚受容体タンパク質中の一般式(1):φ7X1X2Z1X3X4Z2Z3φ65φ4φ3φ2φ1X5X6で示されるアミノ酸配列Aが変異してなるアミノ酸配列Bを含み、条件1~3からなる群より選択される少なくとも1種の条件を満たす、変異型昆虫嗅覚受容体タンパク質(本明細書において、「本開示の嗅覚受容体タンパク質」と示すこともある。)に関する。以下に、これについて説明する。
 本開示の嗅覚受容体タンパク質は、野生型昆虫嗅覚受容体タンパク質中のアミノ酸が変異してなる変異型昆虫嗅覚受容体タンパク質である。
 野生型昆虫嗅覚受容体タンパク質は、昆虫由来の、すなわち昆虫が内在的に発現する(固有に有する)嗅覚受容体であり、且つ一般式(1):φ7X1X2Z1X3X4Z2Z3φ65φ4φ3φ2φ1X5X6で示されるアミノ酸配列Aを含むものである限り、特に制限されない。
 一般式(1)中、X1~X6及びZ1~Z3は、野生型昆虫嗅覚受容体のアミノ酸配列に由来するアミノ酸を示す。例えば、野生型昆虫嗅覚受容体中、アミノ酸配列Aに相当する配列がFSSIVRTAMSYITMLRS(配列番号1で示されるアミノ酸配列中の一部の配列)である場合、下線部のアミノ酸の左から順に、X1、X2、Z1、X3、X4、Z2、Z3、X5、X6である。
 一般式(1)中、φ1は、疎水性アミノ酸を示す。疎水性アミノ酸としては、例えばアラニン、バリン、ロイシン、イソロイシン、メチオニン、プロリン、フェニルアラニン、トリプトファン等が挙げられる。これらの中でも、好ましくはロイシン、メチオニン、イソロイシン、バリン、アラニン、フェニルアラニン等が挙げられ、より好ましくはロイシン、メチオニン、イソロイシン、バリン等が挙げられ、さらに好ましくはロイシン、メチオニン等が挙げられ、よりさらに好ましくはロイシンが挙げられる。
 一般式(1)中、φ2~φ7はそれぞれ独立して無電荷極性アミノ酸又は疎水性アミノ酸を示す。無電荷極性アミノ酸としては、例えばグリシン、トレオニン、セリン、アスパラギン、グルタミン、チロシン、システイン等が挙げられる。疎水性アミノ酸としては、例えばアラニン、バリン、ロイシン、イソロイシン、メチオニン、プロリン、フェニルアラニン、トリプトファン等が挙げられる。
 φ2として、好ましくは疎水性アミノ酸が挙げられ、より好ましくはロイシン、フェニルアラニン、イソロイシン、バリン、メチオニン等が挙げられ、さらに好ましくはロイシンが挙げられる。
 φ3として、好ましくはトレオニン、アラニン、メチオニン、ロイシン等が挙げられ、より好ましくはトレオニンが挙げられる。
 φ4として、好ましくはフェニルアラニン、ロイシン、チロシン、イソロイシン、バリン等が挙げられ、より好ましくはフェニルアラニン、ロイシン、チロシン等が挙げられる。
 φ5として、好ましくはチロシン、フェニルアラニン、イソロイシン、システイン、メチオニン等が挙げられ、より好ましくはチロシンが挙げられる。
 φ6として、好ましくはチロシン、フェニルアラニン、グリシン、トリプトファン、バリン、ロイシン、イソロイシン、アラニン、セリン、メチオニン等が挙げられ、より好ましくはチロシン、フェニルアラニン、グリシン、トリプトファン等が挙げられ、さらに好ましくはチロシン、フェニルアラニン等が挙げられる。

 一般式(1)中、Uは、無電荷極性アミノ酸を示す。無電荷極性アミノ酸としては、例えばグリシン、トレオニン、セリン、アスパラギン、グルタミン、チロシン、システイン等が挙げられる。これらの中でも、好ましくはセリン、トレオニン、グルタミン、アスパラギン、グリシン等が挙げられ、より好ましくはセリン、トレオニン、グルタミン等が挙げられ、さらに好ましくはセリン、トレオニン等が挙げられる。
 野生型昆虫嗅覚受容体タンパク質中のアミノ酸配列Aの位置は、特に制限されるものではない。アミノ酸配列Aは、その配列的特徴から、通常、野生型昆虫嗅覚受容体タンパク質のアミノ酸配列のC末端側に含まれる配列である。C末端側とは、例えば、野生型昆虫嗅覚受容体タンパク質のアミノ酸配列のC末端から、C末端のアミノ酸から数えて例えば70番目、好ましくは60番目、より好ましくは50番目、さらに好ましくは40番目、よりさらに好ましくは35番目のアミノ酸までである。
 野生型昆虫嗅覚受容体タンパク質の由来昆虫としては、好ましくは内翅上目昆虫が挙げられ、より好ましくはカ科、ショウジョウバエ科等の双翅目昆虫;カイコガ科等の鱗翅目昆虫;ミツバチ科等の膜翅目昆虫等が挙げられ、さらに好ましくはカ科、ショウジョウバエ科等の双翅目昆虫が挙げられ、よりさらに好ましくはカ科昆虫が挙げられる。カ科の昆虫としては、例えば、ガンビエハマダラカ(Anopheles gambiae)、ネッタイシマカ(Aedes aegypti)、ネッタイイエカ(Culex quinquefasciatus)等が挙げられる。ショウジョウバエ科の昆虫としては、例えば、キイロショウジョウバエ(Drosophila melanogaster)、ウスグロショウジョウバエ(Drosophila pseudoobscura)、クロショウジョウバエ(Drosophila virillis)等が挙げられる。カイコガ科の昆虫としては、例えば、カイコガ(Bombyx mori)、クワコ(Bombyx mandarina)、イチジクカサン(Trilocha varians)等が挙げられる。ミツバチ科の昆虫としては、例えば、セイヨウミツバチ(Apis mellifera)、ヒメミツバチ(Apis florea)、オオミツバチ(Apis dorsata)、セイヨウオオマルハナバチ(Bombus terrestris)等が挙げられる。
 野生型昆虫嗅覚受容体タンパク質として、具体的には、例えば、DmOR47a(アミノ酸配列:配列番号1)、BmOR56(アミノ酸配列:配列番号2)、AgOR1(アミノ酸配列:配列番号3)、AaOR9(アミノ酸配列:配列番号4)、AgOR28(アミノ酸配列:配列番号65)、AgOR47(アミノ酸配列:配列番号66)、AgOR11(アミノ酸配列:配列番号67)、AgOR27(アミノ酸配列:配列番号68)、AaOR5(アミノ酸配列:配列番号69)、AaOR31(アミノ酸配列:配列番号70)、AaOR72(アミノ酸配列:配列番号71)、AaOR110(アミノ酸配列:配列番号72)、AgOR2(アミノ酸配列:配列番号73)、AgOR10(アミノ酸配列:配列番号74)、AaOR15(アミノ酸配列:配列番号75)等が挙げられる。
 本開示の嗅覚受容体タンパク質は、アミノ酸配列Aが変異してなるアミノ酸配列Bを含む。変異は、好ましくは置換である。
 本開示の嗅覚受容体タンパク質は、アミノ酸配列Bについて、条件1~3からなる群より選択される少なくとも1種の条件を満たす。
 条件1は、アミノ酸配列Bにおいて、X3及び/又はX4が置換しており、X3が分岐鎖アミノ酸であり且つ/或いはX4が無電荷極性アミノ酸であること、である。
 条件1において、「X3及び/又はX4が置換」とは、アミノ酸配列BにおけるX3及び/又はX4が、アミノ酸配列AのX3及び/又はX4から置換していること(すなわち、異なるアミノ酸であること)を示す。
 条件1において、アミノ酸配列B中のX3で示される分岐鎖アミノ酸としては、例えばバリン、ロイシン、イソロイシン等が挙げられる。これらの中でも、好ましくはイソロイシン、ロイシン等が挙げられる。
 条件1において、アミノ酸配列B中のX4で示される無電荷極性アミノ酸としては、例えばグリシン、トレオニン、セリン、アスパラギン、グルタミン、チロシン、システイン等が挙げられる。これらの中でも、好ましくはセリン、アスパラギン等が挙げられる。
 条件1において、アミノ酸配列B中、X3が分岐鎖アミノ酸であり且つX4が無電荷極性アミノ酸であることが好ましい。
 条件2は、アミノ酸配列AにおいてX5及びX6が共に正電荷極性アミノ酸である場合に、アミノ酸配列Bにおいて、X6が正電荷極性アミノ酸以外のアミノ酸であること、である。すなわち、条件2は、アミノ酸配列AにおいてX5及びX6が共に正電荷極性アミノ酸である場合に限定された条件である。
 条件2において、アミノ酸配列A中のX5又はX6で示される正電荷極性アミノ酸としては、例えばリシン、アルギニン、ヒスチジン等が挙げられ、好ましくはリシン、アルギニン等が挙げられる。
 条件2において、アミノ酸配列B中のX6で示されるアミノ酸は、正電荷極性アミノ酸以外のアミノ酸である限り、特に制限されない。該アミノ酸としては、好ましくは無電荷極性アミノ酸が挙げられる。該無電荷極性アミノ酸としては、例えばグリシン、トレオニン、セリン、アスパラギン、グルタミン、チロシン、システイン等が挙げられる。これらの中でも、とりわけ好ましくはセリン、トレオニン等が挙げられる。
 条件3は、アミノ酸配列AにおいてX1及びX2の一方又は両方が正電荷極性アミノ酸である場合に、アミノ酸配列Bにおいて、X1及び/又はX2が置換しており、X1が無電荷極性アミノ酸であり且つ/或いはX2が疎水性アミノ酸であること、である。すなわち、条件3は、アミノ酸配列AにおいてX1及びX2の一方又は両方が正電荷極性アミノ酸である場合に限定された条件である。
 条件3において、アミノ酸配列A中のX1及びX2の内の少なくともX2が正電荷極性アミノ酸であることが好ましく、X2のみが正電荷極性アミノ酸であることがより好ましい。
 条件3において、アミノ酸配列A中のX1又はX2で示される正電荷極性アミノ酸としては、例えばリシン、アルギニン、ヒスチジン等が挙げられ、好ましくはリシン、アルギニン等が挙げられ、より好ましくはアルギニンが挙げられる。
 条件3において、「X1及び/又はX2が置換」とは、アミノ酸配列BにおけるX1及び/又はX2が、アミノ酸配列AのX1及び/又はX2から置換していること(すなわち、異なるアミノ酸であること)を示す。
 条件3において、アミノ酸配列B中のX1で示される無電荷極性アミノ酸としては、例えばグリシン、トレオニン、セリン、アスパラギン、グルタミン、チロシン、システイン等が挙げられる。これらの中でも、好ましくはセリン、グルタミン等が挙げられる。
 条件3において、アミノ酸配列B中のX2で示される疎水性アミノ酸としては、例えばアラニン、バリン、ロイシン、イソロイシン、メチオニン、プロリン、フェニルアラニン、トリプトファン等が挙げられる。これらの中でも、好ましくはアラニン、バリン、ロイシン、イソロイシン等が挙げられ、より好ましくはアラニンが挙げられる。
 条件3において、アミノ酸配列B中、X1が無電荷極性アミノ酸であり且つX2が疎水性アミノ酸であることが好ましい。
 本開示の一態様において、本開示の嗅覚受容体タンパク質は、アミノ酸配列Bについて、条件1A~3Aからなる群より選択される少なくとも1種の条件を満たすことが好ましい。
 条件1Aは、アミノ酸配列Bにおいて、X3が分岐鎖アミノ酸であり且つ/或いはX4が無電荷極性アミノ酸であること、である。条件1Aは、アミノ酸配列BにおいてX3及び/又はX4が置換しているか否かが関係ない以外は、条件1と同様である。つまり、条件1Aは、アミノ酸配列BにおいてX3及び/又はX4がアミノ酸配列Aから置換していない場合も包含する。
 条件2Aは、アミノ酸配列Bにおいて、X6が正電荷極性アミノ酸以外のアミノ酸であること、である。条件2Aは、アミノ酸配列AにおいてX5及びX6が共に正電荷極性アミノ酸であるか否かが関係ない以外は、条件2と同様である。つまり、条件2Aは、アミノ酸配列BにおいてX5及び/又はX6が置換していない場合も包含する。
 条件3Aは、アミノ酸配列Bにおいて、X1が無電荷極性アミノ酸であり且つ/或いはX2が疎水性アミノ酸であること、である。条件3Aは、アミノ酸配列AにおいてX1及びX2の一方又は両方が正電荷極性アミノ酸であるか否かが関係ない以外は、条件3と同様である。つまり、条件3Aは、アミノ酸配列BにおいてX1及び/又はX2がアミノ酸配列Aから置換していない場合も包含する。
 本開示の嗅覚受容体タンパク質は、化学物質応答活性が著しく低減しない限りにおいて、上記した条件1~3の変異以外の他のアミノ酸変異を含むことができる。「著しく低減しない」とは、例えば、他のアミノ酸変異を含む本開示の嗅覚受容体タンパク質の化学物質応答活性が、対応する野生型昆虫嗅覚受容体の化学物質応答活性よりも高く、且つ他のアミノ酸変異を含まない本開示の嗅覚受容体タンパク質の化学物質応答活性100%に対して、例えば50%以上、好ましくは60%以上、より好ましくは70%以上、さらに好ましくは80%以上、よりさらに好ましくは90%以上である、ことを意味する。
 本開示の嗅覚受容体タンパク質のアミノ酸配列は、条件1~3からなる群より選択される少なくとも1種の条件における変異のみを含む変異型昆虫嗅覚受容体のアミノ酸配列に対して、例えば85%以上、より好ましくは90%以上、さらに好ましくは95%以上、よりさらに好ましくは97%以上の同一性を有する。
 本開示の嗅覚受容体タンパク質が他のアミノ酸変異を含む場合、本開示の嗅覚受容体タンパク質のアミノ酸配列は、条件1~3からなる群より選択される少なくとも1種の条件における変異のみを含む変異型昆虫嗅覚受容体のアミノ酸配列に対して、1若しくは複数個のアミノ酸が変異したアミノ酸配列であることが好ましい。「複数個」とは、例えば2~50個、好ましくは2~30個、より好ましくは2~20個、さらに好ましくは2~10、よりさらに好ましくは2~5個である。
 他のアミノ酸変異は、例えば置換、欠失、付加、挿入等が挙げられ、好ましくは置換が挙げられ、中でもより好ましくは保存的置換が挙げられる。また、一般式(1)中のφ2~φ7における変異の場合、無電荷極性アミノ酸と疎水性アミノ酸との置換も好ましい。
 本開示の嗅覚受容体タンパク質の化学物質応答活性は、対応する野生型昆虫嗅覚受容体の化学物質応答活性よりも高い。本開示の嗅覚受容体タンパク質の化学物質応答活性は、対応する野生型昆虫嗅覚受容体の化学物質応答活性に対して、例えば1.2倍以上、1.5倍以上、2倍以上、3倍以上、4倍以上、5倍以上、6倍以上、7倍以上、8倍以上、9倍以上、10倍以上、15倍以上、又は20倍以上である。
 本開示の嗅覚受容体タンパク質は、化学物質応答活性が著しく損なわれない限りにおいて、他のアミノ酸配列、例えばタンパク質タグ、蛍光タンパク質、発光タンパク質、シグナル配列等のタンパク質又はペプチドが付加されたものであってもよい。タンパク質タグとしては、例えばビオチン、Hisタグ、FLAGタグ、Haloタグ、MBPタグ、HAタグ、Mycタグ、V5タグ、PAタグ等が挙げられる。
 本開示の嗅覚受容体タンパク質は、化学物質応答活性が著しく損なわれない限りにおいて、化学修飾されたものであってもよい。
 本開示の嗅覚受容体タンパク質は、C末端がカルボキシル基(-COOH)、カルボキシレート(-COO)、アミド(-CONH2)またはエステル(-COOR)の何れであってもよい。
 ここでエステルにおけるRとしては、例えば、メチル、エチル、n-プロピル、イソプロピル、n-ブチルなどのC1-6アルキル基;例えば、シクロペンチル、シクロヘキシルなどのC3-8シクロアルキル基;例えば、フェニル、α-ナフチルなどのC6-12アリール基;例えば、ベンジル、フェネチルなどのフェニル-C1-2アルキル基;α-ナフチルメチルなどのα-ナフチル-C1-2アルキル基などのC7-14アラルキル基;ピバロイルオキシメチル基などが用いられる。
 本開示の嗅覚受容体タンパク質は、C末端以外のカルボキシル基(またはカルボキシレート)が、アミド化またはエステル化されていてもよい。この場合のエステルとしては、例えば上記したC末端のエステルなどが用いられる。
 さらに、本開示の嗅覚受容体タンパク質には、N末端のアミノ酸残基のアミノ基が保護基(例えば、ホルミル基、アセチル基などのC1-6アルカノイルなどのC1-6アシル基など)で保護されているもの、生体内で切断されて生成し得るN末端のグルタミン残基がピログルタミン酸化したもの、分子内のアミノ酸の側鎖上の置換基(例えば-OH、-SH、アミノ基、イミダゾール基、インドール基、グアニジノ基など)が適当な保護基(例えば、ホルミル基、アセチル基などのC1-6アルカノイル基などのC1-6アシル基など)で保護されているもの、あるいは糖鎖が結合したいわゆる糖蛋白質などの複合蛋白質なども包含される。
 本開示の嗅覚受容体タンパク質は、酸または塩基との塩の形態であってもよい。塩は、特に限定されず、酸性塩、塩基性塩のいずれも採用することができる。例えば酸性塩の例としては、塩酸塩、臭化水素酸塩、硫酸塩、硝酸塩、リン酸塩等の無機酸塩; 酢酸塩、プロピオン酸塩、酒石酸塩、フマル酸塩、マレイン酸塩、リンゴ酸塩、クエン酸塩、メタンスルホン酸塩、パラトルエンスルホン酸塩等の有機酸塩; アスパラギン酸塩、グルタミン酸塩等のアミノ酸塩等が挙げられる。また、塩基性塩の例として、ナトリウム塩、カリウム塩等のアルカリ金属塩; カルシウム塩、マグネシウム塩等のアルカリ土類金属塩等が挙げられる。
 本開示の嗅覚受容体タンパク質は、溶媒和物の形態であってもよい。溶媒は、特に限定されず、例えば水、エタノール、グリセロール、酢酸等が挙げられる。
 本開示の嗅覚受容体タンパク質は、公知の遺伝子工学的手法に従って容易に製造することができる。例えば、PCR、制限酵素切断、DNA連結技術、in vitro転写・翻訳技術、リコンビナントタンパク質作製技術等を利用して製造することができる。
 3.ポリヌクレオチド、細胞、非ヒト動物
 本開示は、その一態様において、本開示の嗅覚受容体タンパク質のコード配列を含む、ポリヌクレオチド(本明細書において、「本開示のポリヌクレオチド」と示すこともある。)、本開示のポリヌクレオチドを含む、細胞(本明細書において、「本開示の細胞」と示すこともある。)、本開示の細胞を含む、非ヒト動物(本明細書において、「本開示の非ヒト動物」と示すこともある。)に関する。以下に、これらについて説明する。
 本開示の嗅覚受容体タンパク質のコード配列は、本開示の嗅覚受容体タンパク質をコードする塩基配列からなるポリヌクレオチドである限り、特に制限されない。
 本開示のポリヌクレオチドは、その一態様において、本開示の嗅覚受容体タンパク質の発現カセットを含む。
 本開示の嗅覚受容体タンパク質の発現カセットは、細胞内で本開示の嗅覚受容体タンパク質を発現可能なポリヌクレオチドである限り特に制限されない。本開示の嗅覚受容体タンパク質の発現カセットの典型例としては、プロモーター、及びそのプロモーターの制御下に配置された本開示の嗅覚受容体タンパク質のコード配列を含むポリヌクレオチドが挙げられる。
 本開示の嗅覚受容体タンパク質の発現カセットに含まれるプロモーターとしては、特に制限されず、対象細胞に応じて適宜選択することができる。プロモーターとしては、例えばpol II系プロモーターを各種使用することができる。pol II系プロモーターとしては、特に制限されないが、例えばCMVプロモーター、EF1プロモーター、SV40プロモーター、MSCVプロモーター等が挙げられる。その他にも、プロモーターとして、例えばtrcやtac等のトリプトファンプロモーター、lacプロモーター、T7プロモーター、T5プロモーター、T3プロモーター、SP6プロモーター、アラビノース誘導プロモーター、コールドショックプロモーター、テトラサイクリン誘導性プロモーター等が挙げられる。
 本開示の嗅覚受容体タンパク質の発現カセットは、必要に応じて、他のエレメント(例えば、マルチクローニングサイト(MCS)、薬剤耐性遺伝子、複製起点、エンハンサー配列、リプレッサー配列、インスレーター配列、レポータータンパク質(例えば、蛍光タンパク質等)コード配列、薬剤耐性遺伝子コード配列などが挙げられる。)を含んでいてもよい。
 本開示のポリヌクレオチドは、ベクターの形態であることができる。使用目的(クローニング、タンパク質の発現)に応じて、また宿主細胞の種類を考慮して適当なベクターが選択される。大腸菌を宿主とするベクターとしてはM13ファージ又はその改変体、λファージ又はその改変体、pBR322又はその改変体(pB325、pAT153、pUC8など)等、酵母を宿主とするベクターとしてはpYepSec1、pMFa、pYES2、pPIC3.5K等、昆虫細胞を宿主とするベクターとしてはpAc、pVL等、哺乳類細胞を宿主とするベクターとしてはpcDNA、pCDM8、pMT2PC等を例示することができる。
 本開示の細胞は、本開示のポリヌクレオチドを含む限り特に制限されない。細胞としては、例えば、Escherichia coli K12等の大腸菌、Bacillus subtilis MI114等のバチルス属細菌、Saccharomyces cerevisiae AH22等の酵母、Spodoptera frugiperda 由来の Sf細胞系もしくはTrichoplusia ni由来のHighFive細胞系、嗅神経細胞等の昆虫細胞、COS7細胞等の動物細胞等を挙げることができる。動物細胞としては、好ましくは、哺乳動物由来の培養細胞、具体的には、COS7細胞、CHO細胞、HEK293細胞、HEK293FT細胞、Hela細胞、PC12細胞、N1E-115細胞、SH-SY5Y細胞等が挙げられる。
 本開示の細胞は、後述の化学物質検出用途にそのまま使用できるという観点から、昆虫の嗅覚受容体共受容体のコード配列を含むことが好ましい。昆虫の嗅覚受容体共受容体は、嗅覚受容体と同様に7回膜貫通構造を有する膜タンパク質であるものの、それ自身は匂い物質を認識せず、嗅覚受容体とヘテロ複合体を形成して機能する。嗅覚受容体と嗅覚受容体共受容体とから構成されるヘテロ複合体である嗅覚受容体複合体は、匂い物質で活性化されるイオンチャンネル活性が備わっており、活性化されるとナトリウムイオン(Na+)、カルシウムイオン(Ca2+)等の陽イオンを細胞内に流入させる。
 同様の観点から、本開示の細胞は、嗅覚受容体が応答した場合に細胞内に流入するイオン(カルシウムイオン等)により発色又は発光するタンパク質のコード配列を含むことが好ましい。このようなタンパク質としては、イクオリン、Yellow Cameleon(YC)、GCaMP等が挙げられる。或いは、本開示の細胞は、カルシウムイオン依存性蛍光色素(例えばFura-2、Fluo-3、Fluo-4等)を含むことが好ましい。
 同様の観点から、本開示の細胞は、本開示の嗅覚受容体タンパク質を含む、すなわち、本開示の細胞においては本開示の嗅覚受容体タンパク質が発現している。この場合、本開示の嗅覚受容体タンパク質は、7回膜貫通構造を有するので、膜タンパク質として、細胞膜上に配置される。
 本開示の非ヒト動物は、本開示の細胞を含む限り特に制限されない。非ヒト動物としては、特に制限されないが、後述の化学物質検出用途に好適であるという観点から、昆虫が好ましい。昆虫については、上記「2.嗅覚受容体タンパク質」における説明と同様である。また、同様の観点から、本開示の非ヒト動物における本開示の細胞は、嗅神経細胞を含むことが好ましい。
 4.用途
 本開示の嗅覚受容体タンパク質は、化学物質に応答して、細胞内に陽イオン(ナトリウムイオン(Na+)、カルシウムイオン(Ca2+)等)を流入させる活性を有する。この陽イオンを検出することにより、或いはこの陽イオンの流入により惹起される嗅神経細胞活性化に基づく行動変化を検知することにより、化学物質を検出することができる。このため、本開示の嗅覚受容体タンパク質は、化学物質検出素子として利用することができる。
 本開示は、その一態様において、本開示の嗅覚受容体タンパク質からなる化学物質検出素子(本開示の検出素子)、さらには本開示の検出素子を含む細胞、脂質二重膜(人工細胞膜)、又は該細胞を含む非ヒト動物を備える、化学物質検出センサ(本開示の検出センサ)、に関する。
 細胞、脂質二重膜、又は非ヒト動物を利用した化学物質検出センサは、既に公知であり、その具体的な構成は、例えば特許文献1、特許文献2、特許文献3等に記載の構成を採用することができる。本開示の細胞又は脂質二重膜を備える化学物質検出センサは、例えば、本開示の細胞又は脂質二重膜を保持する容器、本開示の細胞における化学物質応答(例えば光)を検出すると信号を出力するセンサ、及び前記信号に基づいて化学物質を検出する判定器を備える。本開示の非ヒト動物を備える化学物質検出センサは、例えば、本開示の非ヒト動物の動作を検知する検知部(例えば動体センサ、振動センサ、音センサ等)を備える。
 以下に、実施例に基づいて本発明の一態様を詳細に説明するが、本発明はこれらの実施例によって限定されるものではない。

 試験例1.DmOR47aの化学物質応答活性の測定
 嗅覚受容体(野生型DmOR47a及び変異型DmOR47a)の化学物質に対する応答活性を測定した。
 試験例1-1.発現プラスミドの作製
 <比較例1-1.野生型DmOR47a発現プラスミドの作製>
 ショウジョウバエ(Drosophila melanogaster)成虫RNA(TAKARA社製)を鋳型に用いて、Super Script III逆転写酵素(インビトロジェン社製)を添加し、55℃で50分間、次いで75℃で15分間保温することにより逆転写反応を行い、cDNAを得た。得られたcDNA1μLを鋳型に用い、10μLのフォワードプライマーDmOR47a-5’(5’-caccatggacagttttctgcaagtacagaa;配列番号9)1μL、10μMのリバースプライマーDmOR47a-3’(5’-ttaggagaatgatctcagcattgtgatgta;配列番号10)1μL及びKOD Plus neo DNAポリメラーゼ(東洋紡製)1μLを用いてPCRを行った。PCR反応は、(1)94℃、2分間、(2)98℃、10秒間、(3)68℃、1.5分間で行い、(2)~(3)の工程は35サイクル繰り返し行った。得られたPCR産物をアガロースゲル電気泳動した後、当該ゲル上で検出された約1.2kbのDNAを回収した。回収されたDNAをpENTR/D-TOPOベクター(インビトロジェン社製)へ導入し、pENTR-DmOR47aと名付けたプラスミドを得た。4μLのpENTR-DmOR47a、1μLのpcDNA6.2V5-DEST及び2μLのLRクロナーゼII(インビトロジェン社製)を混合し室温で1時間保持した。得られた混合物を大腸菌へ導入して培養することにより、pcDNA6.2-DmOR47aと名付けた発現プラスミドを得た。発現プラスミドpcDNA6.2-DmOR47aの塩基配列をDNAシーケンサーにより解析したところ、配列番号5で示される塩基配列を含有することが判った。配列番号5で示される塩基配列は配列番号1で示されるアミノ酸配列をコードする。図1に、一般式(1):φ7X1X2Z1X3X4Z2Z3φ65φ4φ3φ2φ1X5X6で示されるアミノ酸配列と、該アミノ酸配列に相当する、配列番号1で示されるアミノ酸配列中の一部のアミノ酸配列との対応関係を示す。
 <実施例1-1.変異型DmOR47a(RX4S)発現プラスミドの作製>
 野生型DmOR47aのアミノ酸配列において372番目のアミノ酸残基(一般式(1)のX4)であるアルギニンがセリンに変異した変異型DmOR47a(RX4S)(アミノ酸配列:配列番号11)のコード配列(塩基配列:配列番号12)を含むプラスミド(pcDNA6.2)を、以下のように作製した。
 野生型DmOR47aのアミノ酸配列において372番目のアミノ酸残基(一般式(1)のX4)であるアルギニンがセリンに変換するように設計したPCRプライマー(プライマーF:CCTTCTCATCGATTGTTAGCACGGCGATGTCCTAC(配列番号13)及びプライマーR:GTAGGACATCGCCGTGCTAACAATCGATGAGAAGG(配列番号14))を合成した。これらのプライマーを用いて、pcDNA6.2-DmOR47aを鋳型とするPCR増幅を行った(反応手順、条件は定法による)。増幅後のPCR反応液をDpnIで消化した後に、大腸菌(DH5α)にトランスフォームした。トランスフォーム後の大腸菌を、25cm四方の(アンピシリンを添加済み)プレートに播種して、37℃で培養してコロニーを形成させた。コロニーを2つ選択し、再度LB液体培地で培養した。その後、それらが保持するプラスミドの塩基配列を決定し、変異型DmOR47a(RX4S)発現プラスミドを得た。
 <実施例1-2.変異型DmOR47a(VX3L-RX4S)発現プラスミドの作製>
 野生型DmOR47aのアミノ酸配列において371番目のアミノ酸残基(一般式(1)のX3)であるバリンがロイシンに変異し、372番目のアミノ酸残基(一般式(1)のX4)であるアルギニンがセリンに変異した変異型DmOR47a(VX3L-RX4S)(アミノ酸配列:配列番号15)のコード配列(塩基配列:配列番号16)を含むプラスミド(pcDNA6.2)を、作製した。具体的には、PCRプライマーとして、プライマーF:CCTTCTCATCGATTCTTAGCACGGCGATGTCC(配列番号17)及びプライマーR:GGACATCGCCGTGCTAAGAATCGATGAGAAGG(配列番号18)を使用し、PCRの鋳型として変異型DmOR47a(RX4S)発現プラスミド(実施例1-1)を使用する以外は、実施例1-1と同様にして作製した。
 <実施例1-3.変異型DmOR47a(VX3L-RX4S+alpha)発現プラスミドの作製>
 変異型DmOR47a(VX3L-RX4S)において、さらに複数の変異が追加された変異型DmOR47a(VX3L-RX4S+alpha)(アミノ酸配列:配列番号19)のコード配列(塩基配列:配列番号20)を含むプラスミド(pcDNA6.2)を、作製した。変異箇所については、図1の通りである。具体的には、PCRの鋳型として変異型DmOR47a(VX3L-RX4S)発現プラスミド(実施例1-2)を使用して、各変異導入用プライマーセットを使用して順次変異を導入して作製した。
 <参考例1-1.キメラOrco発現プラスミドの作製>
 既報(特開2018-50556号公報)に従って、ショウジョウバエ共受容体の一部とミツバチ共受容体の一部との融合タンパク質Dm(NT-TM4)AmORCO(アミノ酸配列:配列番号21)のコード配列(塩基配列:配列番号22)を含むプラスミド(pcDNA3.1)を、作製した。Dm(NT-TM4)AmORCOでは、N末端領域(NT)から第4膜貫通ドメイン(TM4)までのアミノ酸配列(そのN末端アミノ酸残基から234番目のアミノ酸残基までのアミノ酸配列)がショウジョウバエ共受容体由来(Dm(NT-TM4)であり、第2細胞内ループからC末端領域までのアミノ酸配列がミツバチ共受容体由来(Am(IC2-CT))である。
 試験例1-2.発現プラスミドの細胞への導入
 HEK293FT細胞(Invitrogen社から購入)を10cmシャーレに3×10^6cells/シャーレで播種し、10% FBSを含むDMEM培地(ナカライテスク社製)中で、37℃、5%CO2条件下にて約24時間培養した。作製したいずれかの嗅覚受容体発現プラスミド1.5μgと、嗅覚受容体共受容体発現プラスミド3.0μgと、GFP-イクオリン(GAP)発現プラスミド8μgとを、12.5μLのPlus試薬及び31.25μLのリポフェクトアミンLTX(Invitrogen社製)と混合し、10分間保持した。その後、この混合液を上記細胞にトランスフェクションした。トランスフェクション開始4時間後に96wellプレートに9×10^4cells/wellで播種し、10%FBSを含むDMEM培地(ナカライテスク社製)中で、37℃、5%CO2条件下にて約24時間培養した。これにより、嗅覚受容体発現プラスミド、嗅覚受容体共受容体発現プラスミド、及びGAP発現プラスミドがトランジェントに導入された形質転換細胞を得た。
 試験例1-3.活性測定
 GFP-イクオリン(GAP)はカルシウムイオンと結合すると、セレンテラジン等の基質の存在下で活性化されて緑色蛍光を発する。したがって、GAPを発現する細胞の細胞内カルシウムイオン濃度の上昇は直接、蛍光量の増大として現れることになるため、被検物質の添加により嗅覚受容体複合体がイオンチャネルとして機能しているかどうかを蛍光量の変化から測定することができる。
 上記形質転換細胞の培養液を除去してAssay buffer(0.5μMセレンテラジンh(プロメガ社製)及び0.3%のBSAを含むHanks-HEPES(20 mM pH7.4))に置換し、更に4時間、室温で静置した。次いで、Flexstation 3(Molecular devices社製)を用いて、嗅覚受容体に対応する被検物質を細胞の培養液中に添加するとともに細胞の蛍光量を測定した。
 本試験例では、被検物質として、DmOR47aが認識する化学物質であるペンチルアセテートを使用した。
 結果を図1に示す。実施例1-1~1-3の変異型DmOR47aは野生型DmOR47aに比べて化学物質応答活性が高いことが分かった。
 試験例2.BmOR56の化学物質応答活性の測定
 嗅覚受容体(野生型BmOR56及び変異型BmOR56)の化学物質に対する応答活性を測定した。
 試験例2-1.発現プラスミドの作製
 <比較例2-1.野生型BmOR56発現プラスミドの作製>
 いずれか一方のDNA鎖に配列番号6で示される塩基配列を有する二本鎖DNAを合成した。この二本鎖DNA100ngを鋳型に用いて、10μMのフォワードプライマーBmOR56-5’(5’-tggaattctgcagatcaccatgaagctcctggagaagctag;配列番号23)1μL、10μMのリバースプライマーBmOR56-3’(5’-gccactgtgctggattcatgttttattcatttgcgactgac;配列番号24)1μL及びKOD Plus neoDNAポリメラーゼ(東洋紡製)1μLを用いてPCRを行った。PCR反応は、(1)94℃、2分間、(2)98℃、10秒間、(3)63℃、30秒間、(4)68℃、1.5分間で行い、(2)~(4)の工程は35サイクル繰り返し行った。得られたPCR産物を、EcoRVで消化されたpcDNA6.2(インビトロジェン社製)に、In-Fusion HD Cloning Kit(TAKARA社製)を使用して連結した。連結されたDNAを大腸菌へ導入して培養することにより、pcDNA6.2-BmOR56と名付けた発現プラスミドを得た。発現プラスミドpcDNA6.2-BmOR56の塩基配列をDNAシーケンサーにより解析したところ、配列番号6で示される塩基配列を含有することが判った。配列番号6で示される塩基配列は配列番号2で示されるアミノ酸配列をコードする。図2に、一般式(1):φ7X1X2Z1X3X4Z2Z3φ65φ4φ3φ2φ1X5X6で示されるアミノ酸配列と、該アミノ酸配列に相当する、配列番号2で示されるアミノ酸配列中の一部のアミノ酸配列との対応関係を示す。
 <実施例2-1.変異型BmOR56(IX1Q)発現プラスミドの作製>
 野生型BmOR56のアミノ酸配列において380番目のアミノ酸残基(一般式(1)のX1)であるイソロイシンがグルタミンに変異した変異型BmOR56(IX1Q)(アミノ酸配列:配列番号25)のコード配列(塩基配列:配列番号26)を含むプラスミド(pcDNA6.2)を、以下のように作製した。
 野生型BmOR56のアミノ酸配列において380番目のアミノ酸残基(一般式(1)のX1)であるイソロイシンがグルタミンに変換するように設計したPCRプライマー(プライマーF:CGGTCGCCACTTTTCAGAGGATCCTCAAAGGAG(配列番号27)及びプライマーR:CTCCTTTGAGGATCCTCTGAAAAGTGGCGACCG(配列番号28))を合成した。pcDNA6.2-BmOR56を鋳型として、これらのプライマーを用いたPCRにより上記実施例と同様にして変異導入を行い、変異型BmOR56(IX1Q)発現プラスミドを得た。
 <実施例2-2.変異型BmOR56(RX2A)発現プラスミドの作製>
 野生型BmOR56のアミノ酸配列において381番目のアミノ酸残基(一般式(1)のX2)であるアルギニンがアラニンに変異した変異型BmOR56(RX2A)(アミノ酸配列:配列番号29)のコード配列(塩基配列:配列番号30)を含むプラスミド(pcDNA6.2)を、作製した。具体的には、PCRプライマーとして、プライマーF:GTCGCCACTTTTATCGCAATCCTCAAAGGAGCC(配列番号31)及びプライマーR:GGCTCCTTTGAGGATTGCGATAAAAGTGGCGAC(配列番号32)を使用する以外は、実施例2-1と同様にして作製した。
 <実施例2-3.変異型BmOR56(KX4S)発現プラスミドの作製>
 野生型BmOR56のアミノ酸配列において384番目のアミノ酸残基(一般式(1)のX4)であるリシンがセリンに変異した変異型BmOR56(KX4S)(アミノ酸配列:配列番号33)のコード配列(塩基配列:配列番号34)を含むプラスミド(pcDNA6.2)を、作製した。具体的には、PCRプライマーとして、プライマーF:CACTTTTATCAGGATCCTCAGCGGAGCCTATAGTTACTAC(配列番号35)及びプライマーR:GTAGTAACTATAGGCTCCGCTGAGGATCCTGATAAAAGTG(配列番号36)を使用する以外は、実施例2-1と同様にして作製した。
 <実施例2-4.変異型BmOR56(KX4N)発現プラスミドの作製>
 野生型BmOR56のアミノ酸配列において384番目のアミノ酸残基(一般式(1)のX4)であるリシンがアスパラギンに変異した変異型BmOR56(KX4N)(アミノ酸配列:配列番号37)のコード配列(塩基配列:配列番号38)を含むプラスミド(pcDNA6.2)を、作製した。具体的には、PCRプライマーとして、プライマーF:CACTTTTATCAGGATCCTCAACGGAGCCTATAGTTACTAC(配列番号39)及びプライマーR:GTAGTAACTATAGGCTCCGTTGAGGATCCTGATAAAAGTG(配列番号40)を使用する以外は、実施例2-1と同様にして作製した。
 <実施例2-5.変異型BmOR56(IX1Q-RX2A-KX4S)発現プラスミドの作製>
 野生型BmOR56のアミノ酸配列において、380番目のアミノ酸残基(一般式(1)のX1)であるイソロイシンがグルタミンに変異し、381番目のアミノ酸残基(一般式(1)のX2)であるアルギニンがアラニンに変異し、384番目のアミノ酸残基(一般式(1)のX4)であるリシンがセリンに変異した変異型BmOR56(IX1Q-RX2A-KX4S)(アミノ酸配列:配列番号41)のコード配列(塩基配列:配列番号42)を含むプラスミド(pcDNA6.2)を、作製した。具体的には、上記実施例と同様にして、各変異導入用プライマーセットを使用して順次変異を導入して作製した。
 試験例2-2.発現プラスミドの細胞への導入、及び活性測定
 試験例1-2と同様にして発現プラスミドを細胞へ導入し、試験例1-3と同様にして活性測定した。本試験例では、被検物質として、シスジャスモンを使用した。
 結果を図2に示す。実施例2-1~2-5の変異型BmOR56は野生型BmOR56に比べて化学物質応答活性が高いことが分かった。
 試験例3.AgOR1の化学物質応答活性の測定
 嗅覚受容体(野生型AgOR1及び変異型AgOR1)の化学物質に対する応答活性を測定した。
 試験例3-1.発現プラスミドの作製
 <比較例3-1.野生型AgOR1発現プラスミドの作製>
 いずれか一方のDNA鎖に配列番号7で示される塩基配列を有する二本鎖DNAを合成した。この二本鎖DNA100ngを鋳型に用いて、10μMのフォワードプライマーAgOR1-5’(5’- tggaattctgcagatcaccatgaagctgaacaaactgaac;配列番号43)1μL、10μMのリバースプライマーAgOR1-3’(5’- gccactgtgctggatttactctgattccatgctctg;配列番号44)1μL及びKOD Plus neoDNAポリメラーゼ(東洋紡製)1μLを用いてPCRを行った。PCR反応は、(1)94℃、2分間、(2)98℃、10秒間、(3)63℃、30秒間、(4)68℃、1.5分間で行い、(2)~(4)の工程は35サイクル繰り返し行った。得られたPCR産物を、EcoRVで消化されたpcDNA6.2(インビトロジェン社製)に、In-Fusion HD Cloning Kit(TAKARA社製)を使用して連結した。連結されたDNAを大腸菌へ導入して培養することにより、pcDNA6.2-AgOR1と名付けた発現プラスミドを得た。発現プラスミドpcDNA6.2-AgOR1の塩基配列をDNAシーケンサーにより解析したところ、配列番号7で示される塩基配列を含有することが判った。配列番号7で示される塩基配列は配列番号3で示されるアミノ酸配列をコードする。図3に、一般式(1):φ7X1X2Z1X3X4Z2Z3φ65φ4φ3φ2φ1X5X6で示されるアミノ酸配列と、該アミノ酸配列に相当する、配列番号3で示されるアミノ酸配列中の一部のアミノ酸配列との対応関係を示す。
 <実施例3-1.変異型AgOR1(MX3L-KX4S)発現プラスミドの作製>
 野生型AgOR1のアミノ酸配列において401番目のアミノ酸残基(一般式(1)のX3)であるメチオニンがロイシンに変異し、402番目のアミノ酸残基(一般式(1)のX4)であるリシンがセリンに変異した変異型AgOR1(MX3L-KX4S)(アミノ酸配列:配列番号45)のコード配列(塩基配列:配列番号46)を含むプラスミド(pcDNA6.2)を、以下のように作製した。
 野生型AgOR1のアミノ酸配列において401番目のアミノ酸残基(一般式(1)のX3)であるメチオニンがロイシンに変換し、402番目のアミノ酸残基(一般式(1)のX4)であるリシンがセリンに変換するように設計したPCRプライマーF:CACATTTTTGCAGATTTTGAGCCTATCGTACTCCTATC(配列番号47及び49)及びプライマーR:GATAGGAGTACGATAGGCTCAAAATCTGCAAAAATGTG(配列番号48及び50))を合成した。pcDNA6.2-AgOR1を鋳型として、これらのプライマーを用いたPCRにより上記実施例と同様にして変異導入を順次行い、変異型AgOR1(MX3L-KX4S)発現プラスミドを得た。
 <実施例3-2.変異型AgOR1(MX3L-KX4N)発現プラスミドの作製>
 野生型AgOR1のアミノ酸配列において401番目のアミノ酸残基(一般式(1)のX3)であるメチオニンがロイシンに変異し、402番目のアミノ酸残基(一般式(1)のX4)であるリシンがアスパラギンに変異した変異型AgOR1(MX3L-KX4 N)(アミノ酸配列:配列番号51)のコード配列(塩基配列:配列番号52)を含むプラスミド(pcDNA6.2)を、作製した。具体的には、401番目のアミノ酸残基(一般式(1)のX4)であるリシンがアスパラギンに変換し、402番目のアミノ酸残基(一般式(1)のX4)であるリシンがアスパラギンに変換するように設計したPCRプライマーF:CACATTTTTGCAGATTTTGAACCTATCGTACTCCTATC(配列番号53)及びプライマーR:GATAGGAGTACGATAGGTTCAAAATCTGCAAAAATGTG(配列番号54))を合成し、実施例3-1と同様にして作製した。
 <実施例3-3.変異型AgOR1(MX3L-KX4S+alpha)発現プラスミドの作製>
 変異型AgOR1(MX3L-KX4S)において、さらに複数の変異が追加された変異型AgOR1(MX3L-KX4S+alpha)(アミノ酸配列:配列番号55)のコード配列(塩基配列:配列番号56)を含むプラスミド(pcDNA6.2)を、作製した。変異箇所については、図3の通りである。具体的には、PCRの鋳型として変異型AgOR1(MX3L-KX4S)発現プラスミド(実施例3-1)を使用して、各変異導入用プライマーセットを使用して順次変異を導入して作製した。
 試験例3-2.発現プラスミドの細胞への導入、及び活性測定
 試験例1-2と同様にして発現プラスミドを細胞へ導入し、試験例1-3と同様にして活性測定した。本試験例では、被検物質として、フェノールを使用した。
 結果を図3に示す。実施例3-1~3-3の変異型AgOR1は野生型AgOR1に比べて化学物質応答活性が高いことが分かった。
 試験例4.AaOR9の化学物質応答活性の測定
 嗅覚受容体(野生型AaOR9及び変異型AaOR9)の化学物質に対する応答活性を測定した。
 試験例4-1.発現プラスミドの作製
 <比較例4-1.野生型AaOR9発現プラスミドの作製>
 いずれか一方のDNA鎖に配列番号8で示される塩基配列を有する二本鎖DNAを合成した。この二本鎖DNA100ngを鋳型に用いて、10μMのフォワードプライマーAaOR9-5’(5’- tggaattctgcagatcaccatgtccgtcgagaagatcctggc;配列番号57)1μL、10μMのリバースプライマーAaOR9-3’(5’- gccactgtgctggatttagctataaacacgtttcaaaagag;配列番号58)1μL及びKOD Plus neoDNAポリメラーゼ(東洋紡製)1μLを用いてPCRを行った。PCR反応は、(1)94℃、2分間、(2)98℃、10秒間、(3)63℃、30秒間、(4)68℃、1.5分間で行い、(2)~(4)の工程は35サイクル繰り返し行った。得られたPCR産物を、EcoRVで消化されたpcDNA6.2(インビトロジェン社製)に、In-Fusion HD Cloning Kit(TAKARA社製)を使用して連結した。連結されたDNAを大腸菌へ導入して培養することにより、pcDNA6.2-AaOR9と名付けた発現プラスミドを得た。発現プラスミドpcDNA6.2-AaOR9の塩基配列をDNAシーケンサーにより解析したところ、配列番号8で示される塩基配列を含有することが判った。配列番号8で示される塩基配列は配列番号4で示されるアミノ酸配列をコードする。図4に、一般式(1):φ7X1X2Z1X3X4Z2Z3φ65φ4φ3φ2φ1X5X6で示されるアミノ酸配列と、該アミノ酸配列に相当する、配列番号4で示されるアミノ酸配列中の一部のアミノ酸配列との対応関係を示す。
 <実施例4-1.変異型AaOR9(RX6T)発現プラスミドの作製>
 野生型AaOR9のアミノ酸配列において374番目のアミノ酸残基(一般式(1)のX6)であるアルギニンがトレオニンに変異した変異型AaOR9(RX6T)(アミノ酸配列:配列番号59)のコード配列(塩基配列:配列番号60)を含むプラスミド(pcDNA6.2)を、以下のように作製した。
 野生型AaOR9のアミノ酸配列において374番目のアミノ酸残基(一般式(1)のX6)であるアルギニンがトレオニンに変換するように設計したPCRプライマー(プライマーF:CTCATATTTCACTCTTTTGAAAACGGTTTATAGCTAA(配列番号61)及びプライマーR:TTAGCTATAAACCGTTTTCAAAAGAGTGAAATATGAG(配列番号62))を合成した。pcDNA6.2-AaOR9を鋳型として、これらのプライマーを用いたPCRにより上記実施例と同様にして変異導入を行い、変異型AaOR9(RX6T)発現プラスミドを得た。
 <実施例4-2.変異型AaOR9(RX6T+alpha)発現プラスミドの作製>
 変異型AaOR9(RX6T)において、さらに複数の変異が追加された変異型AaOR9(RX6T+alpha)(アミノ酸配列:配列番号63)のコード配列(塩基配列:配列番号64)を含むプラスミド(pcDNA6.2)を、作製した。変異箇所については、図4の通りである。具体的には、PCRの鋳型として変異型AaOR9(RX6T)発現プラスミド(実施例4-1)を使用して、各変異導入用プライマーセットを使用して順次変異を導入して作製した。
 試験例4-2.発現プラスミドの細胞への導入、及び活性測定
 試験例1-2と同様にして発現プラスミドを細胞へ導入し、試験例1-3と同様にして活性測定した。本試験例では、被検物質として、スカトールを使用した。
 結果を図4に示す。実施例4-1~4-2の変異型AaOR9は野生型AaOR9に比べて化学物質応答活性が高いことが分かった。
 試験例5.他の昆虫嗅覚受容体の化学物質応答活性の測定
 試験例1~4の結果より、
(A)X3が分岐鎖アミノ酸であり且つ/或いはX4が無電荷極性アミノ酸となるように変異させること、
(B)野生型においてX5及びX6が共に正電荷極性アミノ酸である場合に、X6が正電荷極性アミノ酸以外のアミノ酸になるように変異させること、並びに
(C)野生型においてX1及びX2の一方又は両方が正電荷極性アミノ酸である場合に、X1が無電荷極性アミノ酸であり且つ/或いはX2が疎水性アミノ酸になるように変異させること、
からなる群より選択される少なくとも1つにより、昆虫嗅覚受容体の化学物質応答活性を向上させることができることが分かった。
 また、多数の昆虫嗅覚受容体(一部の例として、AgOR28、AgOR47、AgOR11c、AgOR27a、AaOR5、AaOR31、AaOR72a、AaOR110等)について、野生型と変異型とで化学物質応答活性を比較する試験を試験例1~4と同様にして行ったところ、上記規則を支持する結果が得られた。

Claims (14)

  1. 変異型昆虫嗅覚受容体タンパク質であって、
    対応する野生型昆虫嗅覚受容体タンパク質中の一般式(1):
    φ7X1X2Z1X3X4Z2Z3φ65φ4φ3φ2φ1X5X6
    [式中、X1~X6及びZ1~Z3は野生型昆虫嗅覚受容体のアミノ酸配列に由来するアミノ酸を示し、φ1は疎水性アミノ酸を示し、φ2~φ7はそれぞれ独立して無電荷極性アミノ酸又は疎水性アミノ酸を示し、Uは無電荷極性アミノ酸を示す。]
    で示されるアミノ酸配列Aが変異してなるアミノ酸配列Bを含み、
    下記条件1~3:
    (条件1)前記アミノ酸配列Bにおいて、X3及び/又はX4が置換しており、X3が分岐鎖アミノ酸であり且つ/或いはX4が無電荷極性アミノ酸であること、
    (条件2)前記アミノ酸配列AにおいてX5及びX6が共に正電荷極性アミノ酸である場合に、前記アミノ酸配列Bにおいて、X6が正電荷極性アミノ酸以外のアミノ酸であること、及び
    (条件3)前記アミノ酸配列AにおいてX1及びX2の一方又は両方が正電荷極性アミノ酸である場合に、前記アミノ酸配列Bにおいて、X1及び/又はX2が置換しており、X1が無電荷極性アミノ酸であり且つ/或いはX2が疎水性アミノ酸であること、
    からなる群より選択される少なくとも1種の条件を満たす、
    変異型昆虫嗅覚受容体タンパク質。
  2. 前記条件1を満たし、X3がイソロイシン又はロイシンであり且つ/或いはX4がセリン又はアスパラギンである、請求項1に記載の変異型昆虫嗅覚受容体タンパク質。
  3. 前記条件2を満たし、X6が無電荷極性アミノ酸である、請求項1又は2に記載の変異型昆虫嗅覚受容体タンパク質。
  4. 前記条件2を満たし、X6がセリン又はトレオニンである、請求項1~3のいずれかに記載の変異型昆虫嗅覚受容体タンパク質。
  5. 前記条件3を満たし、X1がセリン又はグルタミンであり且つ/或いはX2がアラニンである、請求項1~4のいずれかに記載の変異型昆虫嗅覚受容体タンパク質。
  6. 条件1において、X3がイソロイシン又はロイシンであり且つ/或いはX4がセリン又はアスパラギンであり、
    条件2において、X6がセリン又はトレオニンであり、
    条件3において、X1がセリン又はグルタミンであり且つ/或いはX2がアラニンである、請求項1~5のいずれかに記載の変異型昆虫嗅覚受容体タンパク質。
  7. 下記条件1A~3A:
    (条件1A)前記アミノ酸配列Bにおいて、X3が分岐鎖アミノ酸であり且つ/或いはX4が無電荷極性アミノ酸であること、
    (条件2A)前記アミノ酸配列Bにおいて、X6が正電荷極性アミノ酸以外のアミノ酸であること、及び
    (条件3A)前記アミノ酸配列Bにおいて、X1が無電荷極性アミノ酸であり且つ/或いはX2が疎水性アミノ酸であること、
    からなる群より選択される少なくとも1種の条件を満たす、
    請求項1~6のいずれかに記載の変異型昆虫嗅覚受容体タンパク質。
  8. 前記アミノ酸配列Aが、前記野生型昆虫嗅覚受容体タンパク質のアミノ酸配列のC末端側に含まれる配列である、請求項1~7のいずれかに記載の変異型昆虫嗅覚受容体タンパク質。
  9. 前記野生型昆虫嗅覚受容体タンパク質が内翅上目昆虫の野生型嗅覚受容体タンパク質である、請求項1~8のいずれかに記載の変異型昆虫嗅覚受容体タンパク質。
  10. 請求項1~9のいずれかに記載の変異型昆虫嗅覚受容体タンパク質のコード配列を含む、ポリヌクレオチド。
  11. 請求項10に記載のポリヌクレオチドを含む、細胞。
  12. 請求項11に記載の細胞を含む、非ヒト動物。
  13. 請求項1~9のいずれかに記載の変異型昆虫嗅覚受容体タンパク質からなる、化学物質検出素子。
  14. 請求項13に記載の化学物質検出素子を含む脂質二重膜、細胞、又は前記細胞を含む非ヒト動物を備える、化学物質検出センサ。
