Abstract
본 발명은 섬유화 튜머로이드(Fibrosis-encapsulated tumoroid, FET)의 제조 방법 및 그의 용도에 관한 것이다. 본 발명은 유도만능줄기세포 유래 세포를 활용함으로써 실제 고형 암 조직에 가까운 유사체를 제조하였으며, 이는 인간 고형암의 특징을 그대로 반영하지 못하여 임상 검증 단계에서 실패할 확률이 매우 높은 기존의 튜머로이드를 획기적으로 개선하였으므로 항암 신약 개발 및 정밀 의료 분야에 널리 이용될 수 있을 것으로 기대된다.
Description
본 발명은 인간 고형암의 약물 전달 과정을 모사하는 섬유화 튜머로이드 모델의 제조 방법 및 이를 이용하여 암의 치료 또는 개선용 물질을 스크리닝하는 방법에 관한 것이다.
암은 인류가 해결해야 할 난치병 중의 하나로, 전 세계적으로 이를 치유하기 위한 개발에 막대한 자본이 투자되고 있는 실정이며, 우리나라의 경우, 질병 사망원인 중 제 1위의 질병으로서 연간 약 10만 명 이상이 진단되고, 약 6만 명 이상이 사망하고 있다. 지난 10년간 암 진단과 치료에 있어 비약적으로 발전하고 있지만, 암 발병으로 인한 치사율은 여전히 높다.
기존의 신약개발 연구를 통해 개발된 항암 치료제들이 임상 단계에서 부작용이 나타나거나 낮은 효용성에 의해 실제 암환자들에게 활발하게 활용되지 못하고 있다. 또한 같은 암 환자라 하더라도 동일한 항암 치료제에 대한 반응성이 상이한 경우가 많아 이에 대한 연구가 절실하다. 현재, 이러한 신약개발 연구 및 개인별 다양한 암 세포 스페로이드로서 암 조직부를 구성하는 튜머로이드들이 다수 보고되어 오고 있으며, 다양한 세포로 구성된 경우에는 헤테로 셀룰러 또는 멀티 셀룰러 튜머로이드라고 칭한다. 그러나 현재 널리 사용되고 있는 2차원 배양 또는 단순한 스페로이드에 기반한 스크리닝 방법을 통해서는 인간 고형암의 특징을 온전히 반영하지 못하여 개발된 약물이 임상 시험 단계에서 실패하는 경우가 매우 높은 문제가 있다.
이러한 수요를 충족할 수 있는 새로운 튜머로이드의 배양 방법이 본 발명의 근간이 되는 삼차원 섬유화 튜머로이드의 배양 방법이다. 일반적인 기존의 오가노이드는 줄기세포 혹은 암 조직 유래 세포를 이용하여 만드는 스페로이드로서, 장기 또는 암 조직과 유사한 분화를 보인다(Sci Rep. 2016 Jan 11;6:19103.). 그러나 이는 실제 암 조직을 그대로 모사하였다고 볼 수 없다. 암 조직을 둘러싸고 있는 섬유화 조직 구조는 모든 고형암이 공유하는 특징으로 오가노이드의 기존 배양 방법으로는 자연스럽게 형성될 수 없으며, 인위적으로 구성되어져야만 한다. 따라서 실제 인간 고형암에서 나타나는 섬유아세포의 구조 및 혈관 구조의 형태를 그대로 반영하여 인체에서와 동일하게 구현되도록 튜머로이드에 섬유화층을 인위적으로 구성하는 방법에 대한 개발이 요구되는 실정이다.
본 발명자들은 인간 고형암의 특징을 구현하는 섬유화 튜머로이드 모델을 발명하였으며, 더 나아가 혈관으로부터 암 세포로 약물이 전달되는 과정 즉, 인체에서와 동일한 약물 전달 과정을 모사함으로써 신약 개발에 널리 활용될 수 있다. 아울러, 기존의 방법에 비하여 최적의 방법이 적용되어 비용과 시간을 단축할 수 있으므로 시간과 체력적 한계가 있는 암 환자의 선제적 항암제 선별이 가능하게 되므로 실제 환자에게 맞춤형 의료를 제공하는 발판이 될 것으로 기대된다.
본 발명의 일 목적은 인간 고형암 조직과 동일한 형태의 섬유화층 및/또는 혈관세포층으로 구성된 3 차원 섬유화 튜머로이드를 단시간에 높은 수율로 배양하는 방법을 제공하고자 한다.
본 발명의 다른 목적은 본 발명의 방법에 따라 제조된 섬유화 튜머로이드를 제공하고자 한다.
본 발명의 또 다른 목적은 본 발명의 방법에 따라 제조된 섬유화 튜머로이드가 이식된 인간을 제외한 동물 모델을 제공하고자 한다.
본 발명의 또 다른 목적은 본 발명에서 제공하는 섬유화 튜머로이드를 이용하여 암의 치료 또는 개선용 물질을 스크리닝하는 방법을 제공하고자 한다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업계에서 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.

본 발명은 섬유화 튜머로이드(Fibrosis-encapsulated tumoroid, FET)를 제조하는 방법과 이에 의해 제조된 섬유화 튜머로이드의 다양한 용도에 관한 것이다.
본 발명의 일 구현 예에 따르면, 섬유화 튜머로이드의 제조 방법에 관한 것이다.
본 발명의 일 구체예에서, 상기 "튜머로이드"란 줄기세포나 장기 기원 세포로부터 분리한 세포를 3D 배양법으로 다시 응집 또는 재조합하여 만들어진 세포집합체를 의미하는 오가노이드의 일종으로 암 세포의 특성을 갖는 세포집합체를 말한다. 서스펜션 세포 배양물로부터 형성된 튜머로이드 또는 세포 클러스터를 포함할 수 있다. 상기 튜머로이드는 이하 명세서 상에서 고형암 유사체, 오가노이드 또는 스페로이드의 용어를 혼용하여 사용할 수 있다.
본 발명의 일 구체예에서, 상기 "오가노이드"는 줄기세포 혹은 암 조직 유래 세포를 이용하여 만드는 스페로이드로서 장기 혹은 암조직과 유사한 분화를 보인다. 상기 오가노이드는 구체적으로 기관 또는 조직을 구성하는 여러 종류의 세포들 중 하나 이상의 세포 종류를 포함하며, 조직 또는 기관의 형태와 기능을 재현할 수 있어야 한다. 특정 질환을 가진 환자의 세포를 활용할 경우 유전적, 생리적 및 환경적 특성을 그대로 반영한 조건을 제공할 수 있어 각 개인별 적합한 약물을 선별할 수 있는 약물 스크리닝에 널리 이용되고 있다.
본 발명의 섬유화 튜머로이드의 제조 방법은 개체로부터 분리된 암 세포를 수득하는 단계를 포함할 수 있다.
본 명세서 내 "암 조직" 및 "암 세포"는 "종양 조직" 및 "종양 세포"와 동일한 개념으로 사용된다.
본 발명에서 상기 암 세포는 암 환자로부터 채취한 세포로, 상기 "암"은 포유류에서 전형적으로 조절되지 않는 세포 성장으로 특징 지어진 생리적 상태를 나타내거나 가리킨다. 본 발명에서 상기 암은 그 발생 부위에 따라 유방암, 자궁암, 식도암, 위암, 뇌암, 직장암, 대장암, 폐암, 피부암, 난소암, 자궁경부암, 신장암, 혈액암, 췌장암, 전립선암, 고환암, 후두암, 구강암, 두경부암, 갑상선암, 간암, 방광암, 골육종, 림프종, 백혈병 또는 이들의 조합으로 구성된 군으로부터 선택된 암일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 상기 암 세포는 일반적으로 스페로이드가 형성되는 세포에 해당하나, 상기와 같은 스페로이드 형성능은 모든 암 세포에 적용이 되는 것은 아니며, 일부 암 세포에서는 비 스페로이드 형성세포(non-spheroid forming cell, NSF)의 특성을 갖는 것을 포함하고 있다.
본 발명의 섬유화 튜머로이드의 제조 방법은 상기 암 세포와 함께 유도만능줄기세포(Induced pluripotent stem cell, iPSC) 또는 유도만능줄기세포 유래 세포를 공배양(co-culture)하여 스페로이드(spheroid) 형태의 배양체를 형성하는 단계를 포함할 수 있다.
본 발명의 일 구체예에서, 상기 "유도만능줄기세포(induced pluripotent stem cell, iPSC)"는 자기 복제 능력을 가지면서 두 개 이상의 세포로 분화하는 능력을 갖는 세포인 줄기세포의 일종을 말한다. 다능성이 없는 성인의 피부 세포와 같은 체세포에 역분화를 일으키는 4가지 특정 유전자를 도입한 후 발현시키거나 역분화를 일으키는 4가지 유전자가 도입된 세포에서 만들어진 역분화 유도 단백질을 추출하여 이를 다시 체세포에 주입함으로써 배아 줄기세포와 같은 다능성을 갖는 줄기세포를 만들 수 있는데 이를 유도 다능성 줄기세포 또는 역분화 줄기세포라고 한다. 2006년 생쥐의 피부 섬유아세포에 몇 가지 유전자를 도입하여 배아줄기세포처럼 만능성을 가진 줄기세포를 만드는데 성공하였으며, 2007년 성인의 피부세포에 유전자를 도입하여 유도만능줄기세포를 만드는데 성공하였다. 이미 성숙하고 분화된 세포를 미성숙한 세포로 역분화시켜서 다시 모든 조직으로 발전시킬 수 있는 세포로서, 현재 다양한 분야에서 이를 이용한 연구가 활발하게 진행되고 있다.
본 발명에서 상기 암 세포는 비 스페로이드 형성세포(non-spheroid forming cell; NSF)를 포함할 수 있고, 링커 세포(linker cell)인 유도만능줄기세포 또는 그 유래의 세포로 바람직하게는 유도만능줄기세포로부터 유래된 중간엽 줄기세포(iPSC derived MSC, iMSC)를 공배양 함으로써 NSF를 스페로이드화 시킬 수 있다.
본 발명에서 상기 암 세포와 함께 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포의 공배양 시 이들 세포를 0.01:1 내지 100:1, 바람직하게는 0.1:1 내지 10:1, 1:1 내지 5:1, 또는 1.5:1 내지 2.5:1의 세포 수의 비율로 혼합하여 배양할 수 있다.
본 발명에서 상기 암 세포와 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포의 공배양 시 사용되는 배지 조성물 내에 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포는 1 x 104/cm2 내지 10 x 104/cm2, 2 x 104/cm2 내지 8 x 104/cm2, 3 x 104/cm2 내지 7 x 104/cm2 또는 4 x 104/cm2 내지 6 x 104/cm2의 양으로 포함될 수 있으나, 이에 제한되는 것은 아니다.
또한, 본 발명에서 상기 공배양 시 사용되는 배양 배지는 특별히 제한하지 않으나, 예를 들면 DMEM(Dulbeco's Modified Eagle's Medium), IMDM(Iscove's Modified Dulbecco's Medium), a-MEM(Alpha Modification of Eagle's Medium), TI-free OGM 배지(TI-free Osteoblast Growth Medium), F12(Nutrient Mixture F-12), RPMI 1640 배지(RPMI 1640 Medium), 윌리암 배지 E(Williams' s medium E), 맥코이 5A(McCoy' s 5A), E8 배지(Essential 8 medium), SFM 배지(StemPro-34 SFM medium), N2B2 배지(N2B2 medium), OBM 배지(OGM Osteoblast Growth Medium), 성장 배지-5(Growth medium-5), 성장 배지-10(Growth medium-10) 및 DMEM/F12(Dulbecco's Modified Eagle Medium: Nutrient Mixture F-12)로 이루어진 군에서 선택된 1종 이상의 배양 배지를 사용하여 수행될 수 있으나, 이에 제한되는 것은 아니다. 또한, 상기 배지에 B27 보충제(supplement), N2 보충제, G5 보충제 또는 포스콜린(Forskolin) 중 적어도 하나 이상을 추가로 더 포함할 수 있다.
또한, 본 발명에서 상기 공배양 시 사용되는 배양 배지에 암유래 섬유아세포(cancer-associated fibroblast); 내피세포(endothelial cell); iPSC(Induced pluripotent stem cell)로부터 분화된 중간엽줄기세포(iMSC); 및 성장 인자(growth factor) 중 적어도 하나 이상을 추가로 더 포함할 수 있다.
본 발명에서 상기 성장 인자는 혈관내피세포 성장인자(Vascular Endothelial Growth Factor, VEGF), 혈관내피세포 성장인자 A(Vascular Endothelial Growth Factor A, VEGFA), 형질전환성장인자 베타(Transforming growth factor beta, TGF-β), 염기성 섬유아세포 성장 인자(basic fibroblast growth factor; bFGF), 상피세포 성장 인자(epidermal growth factor; EGF) 및 인슐린-유사 성장 인자-1(Insulin-like growth factor-1; IGF-1)로 이루어진 군에서 선택된 1종 이상일 수 있으며, 바람직하게는 형질전환성장인자 베타일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 섬유화 튜머로이드의 제조 방법은 상기와 같이 형성된 배양체에 섬유화층을 유도하는 단계를 포함할 수 있다.
본 발명의 일 구체예에서, 상기 "섬유아세포(fibroblast)"란 선유아세포라고도 명명되며, 결합조직의 고유세포로서 조면소포체와 골지체의 양호한 발육을 특징으로 하는 타원성인 핵과 방추상인 원형질을 갖춘 세포를 말한다. 근아세포(myoplast)와는 밀접한 관계가 있으며 암세포 또는 근육 등의 초배배양에서는 실질세포이외에 존재하는 세포를 총칭하기도 한다. 혈청 또는 섬유아세포 증식인자(fibroblast growth factor: FGF) 등에 의해 증식한다. 암 세포의 특징인 접촉저지기능상실(loss of function for contact inhibition)을 이용하여 발암의 선별(screening)에 쓰이며, 더 나아가 세포의 증식연구, 특히 증식인자(growth factor)의 신호전달기구를 연구하는 데 많이 이용되고 있다.
본 발명의 일 구체예에서, 상기 "내피세포"는 혈관 및 림프관의 내벽을 덮고 있는 한 개 층의 편평세포를 말한다. 원래 상피성 기관의 대부분은 외배엽성 또는 내배엽성이고 간엽에서 생성된 장막강의 벽이나 혈관, 림프관 내벽을 이전에는 가성상피라고 하였으나 현재는 내피라는 용어로 사용되고 있다. 단층 편평상피에서 얇은 판상의 세포가 1층으로 배열되어 세포의 외측면에서 서로 견고하게 결합하는 것으로 알려져 있다. 주로 내피 세포는 모든 혈관의 내부 표면의 내층을 형성하고, 혈액과 조직 간의 비-혈전형성 계면을 구성한다. 내피 세포는 새로운 모세혈관과 혈관이 발생하는데 중요한 성분이기도 하며 내피 세포는 종양 성장 및 전이뿐만 아니라 각종 비-신생물성 질병 또는 장애와 연관된 혈관신생 또는 신혈관신생 과정 동안에 증식된다.
본 발명의 일 구체예에서, 상기 "혈관내피세포"는 혈관 내강이 배접하여 내막을 구성하는 한 층의 세포로서 중배엽 기원이며, 내막 박리에 의해 아세틸 콜린에 의한 혈관 이완 현상이 소실되는 것이 제시된 이후 이것의 기능적 중요성이 주목을 받게 되었다. 이러한 이완을 매개하는 내피세포 유래 평활근 이완 인자나 혈관 수축 작용을 하는 엔도텔린 등의 생산에 의한 혈관 긴장성 조절 외에 혈관내 혈액 응고의 조절과 혈관 투과성 조절, 물질교환, 백혈구 세포와의 상호작용에 의한 생체 방어 등 다양한 기능을 갖고 있는 것으로 알려져 있다.
본 발명에서 상기 "혈관내피세포 성장인자"란 세포에서 만들어진 새로운 혈관형성을 자극하는 물질로 배양하수체 세포의 상층액에서 혈관내피세포에 대한 생장활성을 지표로 하여 분리되는 폴리펩티드를 말한다. 단일유전자에서 121, 165, 189, 206 아미노산 잔기의 4종의 폴리펩티드를 번역하여 SS결합에 의한 효모 2합체로서 생산되며, 대식세포, 평활근세포, 종양세포 등 여러 가지 세포가 생산하는 것으로 알려져 있다.
본 발명에서 상기 "형질전환 성장인자 베타"란 25kDa의 2합체 구조를 한 단백질로 세포내에 신호를 전달하는 TGF-β 수용체(Transforming growth factor beta receptor, TGF-β receptor)와 결합하여 세린/트레오닌 인산화효소를 활성화시키고 세포 내에 신호를 전달하게 된다. 포유류에는 구조가 유사한 3종류의 동형(TGF-β1, β2, β3)이 있다. 원래 섬유아세포의 연한천 중에서 증식을 촉진시키는 물질 중의 하나로 발견되었으나, 현재는 대부분 세포의 생장억제인자로 알려져 있다. TGF-β는 혈소판이나 태반, 골격 등 여러 조직에서 생산되고 세포의 생장억제, 세포외기질의 생산, 면역능 억제 같은 작용을 하는 것으로 알려져 있다.
본 발명에서 상기의 섬유화층은 섬유아세포(fibroblast), 섬유아세포의 전구체 세포, 암유래 섬유아세포(cancer-associated fibroblast), 유도만능줄기세포로 유도된 중간엽 줄기세포(iMSC), 및 유도만능줄기세포로 유도된 암유래 섬유아세포(iPSC derived cancer-associated fibroblast)로 구성된 군으로부터 선택된 적어도 하나에 의하여 유도되는 것일 수 있으며, 섬유아세포와 유사한 특성을 갖는 세포에 해당한다면 이에 제한되지 않고 모두 포함될 수 있다.
본 발명의 상기의 섬유아세포층을 유도하기 위하여 상기 배양체와 유도만능줄기세포 또는 유도만능줄기세포 유래 세포를 혼합 배양하여, 상기 배양체의 외곽에 섬유아세포층을 유도할 수 있다.
본 발명에서 상기 유도만능줄기세포 유래 세포는 유도만능줄기세포로부터 유도된 중간엽 줄기세포(iMSC), 및 유도만능줄기세포로부터 유도된 암유래 섬유아세포(iPSC derived cancer-associated fibroblast) 중 적어도 하나일 수 있다.
본 발명에서 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포는 상기 배양체 내 암세포와 상호작용을 하여 상기 암세포가 분비하는 TGF-β에 의해 암유래 섬유아세포(cancer-associated fibroblast)로 분화 유도될 수 있다.
본 발명에서 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포;와 상기 배양체는 5:1 내지 2:1, 바람직하게는 3:1의 세포수의 비율로 혼합하여 배양될 수 있다.
본 발명에서 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포;와 상기 배양체의 혼합 배양 시 사용되는 배지 조성물 내에 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포는 1 x 104/cm2 내지 10 x 104/cm2, 2 x 104/cm2 내지 8 x 104/cm2, 3 x 104/cm2 내지 7 x 104/cm2 또는 4 x 104/cm2 내지 6 x 104/cm2의 양으로 포함될 수 있다.
본 발명에서 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포;와 상기 배양체의 혼합 배양 시 사용되는 배지 조성물에는 성장 인자가 추가로 포함될 수 있다. 여기서, 상기 성장 인자는 혈관내피세포 성장인자(Vascular Endothelial Growth Factor, VEGF), 혈관내피세포 성장인자 A(Vascular Endothelial Growth Factor A, VEGFA), 형질전환성장인자 베타(Transforming growth factor beta, TGF-β), 염기성 섬유아세포 성장 인자(basic fibroblast growth factor; bFGF), 상피세포 성장 인자(epidermal growth factor; EGF) 및 인슐린-유사 성장 인자-1(Insulin-like growth factor-1; IGF-1)로 이루어진 군에서 선택된 1종 이상일 수 있으며, 바람직하게는 형질전환성장인자 베타일 수 있으나, 이에 제한되는 것은 아니다.
또한, 본 발명에서 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포;와 상기 배양체의 혼합 배양 시 사용되는 배지 조성물에 추가로 포함되는 성장 인자의 양은 10 내지 1000 ng/ml, 50 내지 500 ng/ml, 60 내지 200 ng/ml 또는 80 내지 120 ng/ml일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 섬유화 튜머로이드의 제조 방법은 필요에 따라서는 상기와 같이 유도된 섬유화층을 치밀화(compaction)하는 단계를 더 포함할 수 있다.
본 발명에서 상기 섬유화층의 치밀화는 35 내지 40 ℃의 온도 및 5 부피% CO2 조건 하에서 1 내지 10 일, 바람직하게는 1 내지 5 일 동안 추가로 배양하여 수행될 수 있다.
본 발명의 섬유화 튜머로이드의 제조 방법은 이후 섬유화층이 유도된 스페로이드(Fibrosis-encapsulated tumoroid; FET)를 선별하는 단계를 포함할 수 있다.
또한, 본 발명의 섬유화 튜머로이드의 제조 방법은 선별된 스페로이드(FET)에 혈관세포층을 유도하는 단계를 포함할 수 있다.
본 발명의 상기의 혈관세포층을 유도하기 위하여 상기 FET와 내피세포(endothelial cell) 또는 혈관내피세포(Vascular endothelial cell)를 혼합 배양하여, 상기 FET의 외곽에 혈관세포층을 유도할 수 있다.
본 발명에서 상기 내피세포 또는 혈관내피세포는 유도만능줄기세포로부터 분화 유도된 것일 수 있고, 바람직하게는 유도만능줄기세포로부터 유도된 중배엽 세포로부터 유도된 내피세포 또는 혈관내피세포로부터 형성되는 것일 수 있으나, 상기 세포와 유사한 특성을 가진 세포에 해당한다면 이에 제한되지 않고 모두 포함될 수 있다.
본 발명에서 상기 내피세포 또는 혈관내피세포;와 상기 FET는 5:1 내지 2:1, 바람직하게는 3:1의 세포수의 비율로 혼합하여 배양될 수 있다.
본 발명에서 상기 FET와, 내피세포 또는 혈관내피세포의 혼합 배양 시 사용되는 배지 조성물 내에 상기 유도만능줄기세포 또는 유도만능줄기세포 유래 세포는 1 x 104/cm2 내지 10 x 104/cm2, 2 x 104/cm2 내지 8 x 104/cm2, 3 x 104/cm2 내지 7 x 104/cm2 또는 4 x 104/cm2 내지 6 x 104/cm2의 양으로 포함될 수 있다.
또한, 본 발명에서 상기 FET와, 내피세포 또는 혈관내피세포의 혼합 배양 시 사용되는 배지 조성물에는 성장 인자가 추가로 포함될 수 있다. 여기서, 상기 성장 인자는 혈관내피세포 성장인자(Vascular Endothelial Growth Factor, VEGF), 혈관내피세포 성장인자 A(Vascular Endothelial Growth Factor A, VEGFA), 형질전환성장인자 베타(Transforming growth factor beta, TGF-β), 염기성 섬유아세포 성장 인자(basic fibroblast growth factor; bFGF), 상피세포 성장 인자(epidermal growth factor; EGF) 및 인슐린-유사 성장 인자-1(Insulin-like growth factor-1; IGF-1)로 이루어진 군에서 선택된 1종 이상일 수 있으며, 바람직하게는 형질전환성장인자 베타일 수 있으나, 이에 제한되는 것은 아니다.
또한, 본 발명에서 상기 FET와, 내피세포 또는 혈관내피세포의 혼합 배양 시 사용되는 배지 조성물에 추가로 포함되는 성장 인자의 양은 10 내지 1000 ng/ml, 50 내지 500 ng/ml, 60 내지 200 ng/ml 또는 80 내지 120 ng/ml일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 상기 FET와, 내피세포 또는 혈관내피세포의 혼합 배양은 2일 내지 14일 동안 또는 3일 내지 10일, 바람직하게는 4일 내지 7일 동안 수행될 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 상기 FET와, 내피세포 또는 혈관내피세포의 혼합 배양은 5% CO2 및 37℃의 온도 조건 하에서 수행될 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 섬유화 튜머로이드의 제조 방법은 필요에 따라서는 상기와 같이 유도된 혈관세포층을 치밀화(compaction)하는 단계를 더 포함할 수 있다.
본 발명의 섬유화 튜머로이드의 제조 방법은 이후 혈관세포층이 유도된 스페로이드(Endothelial cell layered FET; eFET)를 선별하는 단계를 포함할 수 있다.
본 발명의 다른 구현 예에 따르면, 본 발명의 방법으로 제작된 섬유화 튜머로이드에 관한 것이다.
본 발명에 따른 섬유화 튜머로이드는 암 환자로부터 채취한 암 세포를 3 차원 배양한 것으로서, 인체 내 고형암의 구조적 특징인 튜머로이드 외층에 캡슐화 과정을 통해 섬유화층을 치밀화하여 구현한 것에 특징이 있다. 더 나아가, 본 발명에서는 상기와 같이 섬유화층이 캡슐화된 튜머로이드에 추가로 혈관세포층의 형성을 유도함으로써, 최종 제조되는 튜머로이드에 약물을 처리하여 스크리닝하는 경우 인간 고형암 조직에서 나타나는 약물 전달 과정으로 항암제가 혈관벽과 섬유화 조직을 차례로 지나 암조직에 도달하는 일련의 과정을 인체에서와 동일하게 모사할 수 있는 것에 기술적 특징이 존재한다고 할 것이다.
통상적으로 암 세포에는 스페로이드 형성능이 있는 세포와 비 스페로이드 형성능이 있는 세포로 이루어져 있는데, 본 발명에서는 만능유래줄기세포를 활용하여 비 스페로이드 형성능이 있는 암세포도 스페로이드화 시킬 수 있다는 점에서 튜머로이드 제조가 가능한 암종을 넓힐 수 있어 기존의 발명들과 차별점이 존재한다고 할 것이다.
또한, 본 발명에서는 환자 유래 암세포를 이용하여 섬유화 튜머로이드를 보관하거나, 지속적으로 배양하여 대량 제작에 이용할 수 있다.
본 발명의 또 다른 구현 예에 따르면, 본 발명의 방법으로 제작된 섬유화 튜머로이드가 이식된 인간을 제외한 동물 모델에 관한 것이다.
본 발명에서 상기 "동물 모델"은 질환 동물 모델을 의미한다. 구체적으로, 동물 모델은 인간의 질병과 유사한 상태의 질병에 걸리거나 선천적으로 그 질병에 걸리도록 만들어낸 동물 모델일 수 있다. 본 명세서에서 동물 모델은 알레르기성 호흡기 질환의 동물 모델일 수 있다. 또한, 본 발명의 알레르기성 호흡기 질환의 동물 모델로 이용될 수 있는 동물은 인간을 제외한 포유 동물로, 예를 들면, 래트, 마우스, 모르모트, 햄스터, 토끼, 원숭이, 개, 고양이, 소, 말, 돼지, 양 및 염소로 구성된 군으로부터 선택될 수 있고, 보다 바람직하게는 마우스일 수 있다.
본 발명의 또 다른 구현 예에 따르면, 본 발명의 방법으로 제작된 섬유화 튜머로이드를 이용한 항암제의 스크리닝 방법에 관한 것이다.
본 발명에서, "항암제"는 본 발명의 섬유화 튜머로이드를 이용하여 스크리닝되는 후보 물질로 암 세포 또는 암 줄기세포의 증식을 억제하거나 증식을 지연시키는 모든 작용제라면 제한없이 포함할 수 있다. 화학항암제, 표적항암제, 면역항암제, 또는 대사항암제 등이 포함될 수 있으며, 체내에 투여되어 빠르게 분열하는 암 세포 또는 암 줄기세포를 공격하는 약물의 종류에 해당한다면, 이에 제한되는 것은 아니다.
본 발명의 일 구체예에서, 상기 "스크리닝"이란 신약 개발 과정 중에 거치게 되는 수많은 절차들 중의 하나로, 보다 구체적으로는 기초 연구로부터 선정된 화합물 스크리닝을 통해 유효 물질 및 선도 물질을 발굴하여 후보물질을 확정하는 절차를 말한다. 이는 신약 개발에 소요되는 시간과 비용을 줄여줄 수 있으며, 대상 질병이 선정된 후 이를 표적으로 하는 신약 유효 물질을 발굴할 수 있다. 예를 들어, 신약뿐만 아니라 예방제 또는 화장료 조성물 등을 조사하였을 때 피부 질환, 보다 구체적으로는 아토피 피부염 질환으로 나타나는 증상이 호전되거나 이롭게 변경되는 물질이라면, 이에 제한없이 포함될 수 있다.
본 발명의 항암제의 스크리닝 방법은 본 발명의 섬유화 튜머로이드를 준비하는 단계를 포함할 수 있다.
본 발명에서 상기 섬유화 튜머로이드는 인간 고형암의 특징을 구현함과 동시에 인간 고형암에서 나타나는 약물 전달 과정을 그대로 모사하였으므로 환자 개개인별 맞춤형 치료제 스크리닝에 이용할 수 있다.
본 발명에서 상기 항암제의 스크리닝 방법은 상기 섬유화 튜머로이드에 후보 물질을 처리하는 단계를 포함할 수 있다.
본 발명에서 상기 후보 물질은 천연 화합물, 합성 화합물, RNA, DNA, 폴리펩티드, 효소, 단백질, 리간드, 항체, 항원, 박테리아 또는 진균의 대사 산물 및 생활성 분자로 이루어진 군으로부터 선택되는 1종 이상일 수 있으나, 이에 제한되는 것은 아니다.

본 발명의 항암제의 스크리닝 방법은 상기 후보 물질이 처리된 섬유화 튜머로이드에 항암 효과가 나타나는 경우에 상기 후보 물질을 암 치료 또는 개선용 물질로 판단하는 단계를 포함할 수 있다.
본 발명에의 항암제의 스크리닝 방법은 본 발명에서 제공하는 인체 내 고형암의 특징을 그대로 반영하여 제작된 섬유화 튜머로이드를 이용함으로써 각 개개인별 최적의 항암제의 선제적 선별이 가능하므로 맞춤형 항암 치료제 개발에 널리 이용될 수 있을 것으로 기대된다.
본 발명은 인간 고형암의 특징을 구현하는 섬유화 튜머로이드의 제조방법 및 그의 용도에 관한 것이다. 본 발명에서 제공하는 섬유화 튜머로이드는 튜머로이드 외층에 섬유화층과 혈관세포층이 캡슐화 과정을 통해 형성되어 인간 고형암의 구조적 특징을 그대로 반영하고 있으며, 이에 약물을 처리하는 경우 체내 약물 전달 과정으로 약물이 혈관벽과 섬유화 조직을 차례로 지나 암조직에 도달하는 일련의 과정이 그대로 구현될 수 있다. 따라서, 본 발명에 따른 섬유화 튜머로이드를 활용하는 경우 정확한 약물 스크리닝을 통해 임상 검증 단계에서의 성공률을 높일 수 있을 뿐만 아니라 환자 유래 암 세포를 이용하여 보다 효과적으로 각 개개인별 최적의 항암제의 선제적 선별이 가능하므로 맞춤형 항암 치료제 개발에 널리 이용될 수 있을 것으로 기대된다.

본 발명의 일 구현 예에서는 (a) 개체로부터 분리된 암 세포를 수득하는 단계; 및 (b) 상기 암 세포와 함께 유도만능줄기세포(Induced pluripotent stem cell, iPSC) 또는 유도만능줄기세포 유래 세포를 공배양(co-culture)하여 스페로이드(spheroid) 형태의 배양체를 형성하는 단계;를 포함하는 섬유화 튜머로이드의 제조 방법에 관한 것이다.
본 발명의 다른 구현 예에서는 본 발명의 방법으로 제작된 섬유화 튜머로이드에 관한 것이다.
본 발명의 또 다른 구현 예에서는 본 발명의 방법으로 제작된 섬유화 튜머로이드가 이식된 인간을 제외한 동물 모델에 관한 것이다.
본 발명의 또 다른 구현 예에서는 본 발명의 방법으로 제작된 섬유화 튜머로이드를 이용한 항암제의 스크리닝 방법에 관한 것이다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로서, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1: 암 세포주의 배양 및 링커 세포(Linker cell)의 준비
1.1 암 세포주의 배양
본 발명의 발명자들은 연세대학교 의과대학 평가위원회 (Institutional Review Board, IRB)의 승인을 얻어 모든 실험을 수행하였으며, 기 구축된 유도만능줄기세포 풀을 활용하여 실험에 이용하였다. 또한, 미국 세포주 은행(American Type Culture Collection; ATCC)으로부터 인간 유방암 세포주인 BT-474, MCF-7, SK-BR-3, BT-20, MDA-MB-231 및 Hs578T, 췌장암 세포주인 PANC-1, MIA Paca-2, AsPC-1 및 BxPC-3를 수득하여 10% FBS가 포함된 배지에서 배양하여 실험에 이용하였으며, 암 세포의 배양 외에 다른 세포 등의 배양을 위한 하기 실시예에서 사용되는 모든 배지는 조성이 알려져 있거나 판매되는 상품이나, 본 연구에서는 배양의 최적화를 위하여 2가 이상의 배지를 혼합하여 사용하거나 VEGFA 등을 첨가하여 사용하였다.
1.2 링커 세포의 준비
배아체 형성 배지(Embryonic Body(EB) formation medium(EBFM, AggreWell, STEMCELL Technologies))를 이용하여, 1x104 개의 유도만능줄기세포(Induced pluripotent stem cell, iPSC)로 배아체(EB)를 구성하였다. 상기 배아체를 하기 표 1의 구성 성분을 포함한 Essential 8(E8) 배지에서 5일 동안 배양하였으며, 5일째 되는 날 상기 배아체를 0.1%의 젤라틴 코팅 디쉬(gelatin coated dish)에 붙여 하기 표 2의 성분으로 구성된 중간엽 줄기세포(Mesenchymal stem cell, MSC) 배양 배지(culture medium)에 2주 동안 추가로 배양하여 iPSC 유래 중간엽 세포(iMSC)로 분화시켜 링커 세포(linker cell)를 제조하였다.

본 발명은 기존의 발명들과 차별화하여 유도만능세포를 활용하였으며, 이를 이용하여 고형암 유사체를 구축하는 실험 설계도로서의 모식도를 도 1에 나타내었다. 이하에서 상기 설계에 따른 섬유화 튜머로이드의 제조 과정을 상세히 설명한다.
실시예 2: 비 스테로이드 형성 암 세포의 튜머로이드 형성 유도
암 세포와 iMSC를 배양 플레이트로부터 떼어내어 PBS를 이용하여 워싱한 후 펠렛 형태를 준비하였다. 오가노이드 기본배지(Organoid Basal Medium, OBM)와 배아체 형성 배지(EB formation medium, EBFM)의 혼합배지를 이용하여 암 세포(8 x 104/ml)와 iMSC (4 x 104/ml)를 준비하였다. 이 때 혼합배지는 OBM : EBFM의 비율을 1:1로 유지하였다. OBM 배지의 조성은 하기 표 3과 같다.

플레이트(ultra-low attachment, U-shape plate) 상에서, 상기에서 준비한 암 세포 12.5㎕ 및 iMSC 12.5㎕를 혼합하였다. 세포배양 인큐베이터에 37℃, 5%의 CO2 조건에서 1일 동안 스페로이드 형성을 유도하였다. 스페로이드 형성이 유도된 이후 1일 동안 추가적으로 치밀화(compaction)를 유도하여 사용하였다. 이때 암 세포와 iMSC의 비율을 2 : 1로 유지(예를 들어, 1x103 개 : 5x102 개)한 채 각 세포의 수를 늘리게 되면 튜머로이드의 크기를 증가시킬 수 있다. 그러나, 섬유화층과 혈관세포층의 추가적 구축을 최적화하기 위하여는 상기에서 언급한 세포의 개수가 가장 적절한 것을 확인하였다. 추가적으로 동일한 볼륨(25㎕)의 성장배지-10(Growth medium - 10)을 첨가하고 스페로이드 치밀화(spheroid compaction)가 유도되었으며, 이 때, 24시간 이상의 세럼-프리 상태 유지는 세포의 전사체 발현 변화를 유도할 수 있어, 세럼이 있는 배지를 추가로 첨가하였다. 전사체 발현 변화는 세포의 성질 변화를 유도할 수 있으며, 최종적 세럼 농도는 5%가 되도록 하였다. 상기 성장배지-10(Growth medium - 10)의 조성은 하기 표 4와 같다.

상기 실시예 2에 따라 제조된 비 스페로이드 형성세포의 스페로이드 형성 유도 결과를 서로 다른 종류의 암 세포에서 관찰한 사진을 도 3에 나타내었다. NSF#1은 유방암 세포주인 MDA-MB-231이며, NSF#2는 유방암 세포주인 SK-BR-3이며, NSF#3은 췌장암 세포주인 AsPC-1를 이용하였으며, 도 3을 참조하면 링커 세포(iMSC)에 의하여 다양한 암종의 비 스페로이드 형성세포가 1일 이내에 스페로이드를 형성한 것을 확인할 수 있었다. 링커 세포는 iPSC로부터 분화된 iMSC로 초록색 형광물질로 염색하여 확인한 결과 스페로이드 형성에 사용된 후 5일 이내에 90% 이상이 사멸된 것을 확인하였다(도 4 참조). 이러한 링커 세포의 사멸로 인하여, 튜머로이드의 특징이 유지될 수 있다. 또 하나의 기존 발명과의 차별점은 상기 링커 세포를 이용하여 비 스페로이드 형성 세포를 스페로이드화로 유도한 것에 있다.
실시예 3: 유도만능줄기세포를 이용하여 섬유화층 형성(Fibrosis-encapsulated Tumoroid, FET) 유도
유도만능줄기세포(Induced pluripotent stem cell, iPSC)로부터 활성화 섬유아세포(섬유화조직의 주 구성세포)의 전구체(iMSC)를 유도하고, 튜머로이드 외곽에 섬유화층 형성을 유도하여 FET를 제조하고, 혈관세포층의 형성을 유도하여 eFET(Endothelial cell layered FET)를 제조하는 과정의 모식도를 도 5에 나타내었다. 유도만능줄기세포로부터 유도 또는 기원한 섬유아세포의 전구체를 iMSC라고 하며, 이는 암 세포가 분비하는 TGF-β(Transforming growth factor-beta)에 의해 활성화 섬유아세포로 분화된다. 본 실험에서는 섬유화층 또는 혈관세포층의 형성을 확인하기 위하여 형광 단백체를 과발현하는 iMSC를 사용하였다. 먼저, 캡슐화 배지 1(Encapsulation medium 1)을 이용하여, 플레이트(ultra-low attachment, U-shape plate) 상에서, 튜머로이드 형성에 사용된 세포수의 3배인 3x103 개의 iMSC를 이용하여 튜머로이드 외곽에 섬유화층 형성을 유도하였다. 이때 iMSC는 1.2x105/ml로 준비하여, 25ul를 사용하였으며, 사용한 캡슐화 배지 1은 동량의 TGF-b inhibitor(TI)-free Organoid Growth Medium(OGM)와 Embryonic Body(EB) formation medium(EBFM / AggreWell - STEMCELL Technologies)의 혼합배지로서 사용하였으며, TI-free OGM 배지의 조성은 하기 표 5와 같다.

그 후 세포배양 인큐베이터 37℃ 및 5%의 CO2 조건에서 2일 동안 추가로 스페로이드 형성 및 치밀화 과정을 거쳤다. 상기 과정을 거친 다양한 인간 유방암 세포주(BT-474, MCF-7, SK-BR-3, BT-20, MDA-MB-231, 및 Hs578T)의 배양액에 의해 활성화된 섬유아세포의 형태 변화를 형광 현미경으로 관찰하여 도 7에 나타내었다. 상기 결과에 비추어 암 세포주에 따라 다양한 섬유아세포의 형태가 관찰된 것을 확인할 수 있다. 또한 중심부 죽은 세포, 중간층 정지 세포 및 외곽층 증식 세포로 구성된 단순 튜머로이드(Sci Rep. 2016 Jan 11;6:19103.)와 달리 기존의 튜머로이드 외층에 추가적으로 섬유화 구성하였고 이를 형광 염색하여 형광 이미지를 확인하여 도 8a에 나타낸 결과 암 조직 외피에 섬유층이 생성된 것을 확인할 수 있었다.
실시예 4: 유도만능줄기세포를 이용하여 혈관세포층(Endothelial cell layered FET, eFET) 형성 유도
치밀화 과정을 거친 FET에 캡슐화 배지 2(Encapsulation medium 2)를 이용하여 플레이트(ultra-low attachment, U-shape plate) 상에서, 섬유화층 형성에 사용된 세포 수의 3배인 9 x 103의 내피세포(endothelial cell, EC)를 이용하여 FET 외곽에 EC layer의 형성을 유도하였다. EC는 3.6 x 105/ ml로 준비하여, 25 ul를 사용하였으며, 캡슐화 배지 2는 캡슐화 배지 1에 VEGFA 50 ng/ml를 추가하여 조성하였다. 이 후 세포배양 인큐베이터에서 37℃, 5%의 CO2 조건 하에 1일 동안 스페로이드를 형성시켰으며, 유지 배지(Maintenance medium)는 하기 표 6의 성장배지-5(Growth medium-5)에 VEGFA 20 ng/ml를 추가하여 조성하였고 상기 배지로 교체하여 최종 형성을 유도하였다.

상기 방법으로 제조된 eFET의 이미지를 도 8b에 나타내었다. 혈관 내피세포의 경우 형광 염색이 되지 않아, 형광 이미지상의 혈관 내피세포층은 백색 점선으로 표시하였다. 실시예 3 및 실시예 4의 방법으로 제조된 섬유화 튜머로이드(FET 또는 eFET)는 암종 혹은 암의 진행 정도에 따라 섬유화 조직의 두께 정도가 다양하게 나타나는 것을 확인하였으며(도 9 참조), 이러한 섬유화 층의 두께는 섬유아세포의 수를 조절함으로써, 각 두께별 섬유화 튜머로이드를 제작할 수 있다.
실시예 5: 효율적인 EC 분화를 위한 최적의 배지 조성 비율 적용
마트리겔 코팅 접시 상에 iPSC를 4 x 104/cm2를 분주하고, 중배엽(Lateral mesoderm)을 유도하기 위하여 pre-warmed N2B27 Medium supplemented with CHIR-99021(8 uM) and hBMP4(25 ng/ml)로 배지를 교체하였으며, 추가적인 배지의 교체 없이 3일간 유지하였다. 상기 배지의 조성은 하기 표 7과 같고, DMEM/F12 : Meurobasal medium의 비율을 1 : 1로 구성하였다.

4, 5일째 내피세포를 유도하기 위하여 StemPro-34 SFM medium supplemented with 혈관내피세포 성장인자(VEGF, 200 ng/ml) and 포스콜린(Forskolin, 2 uM)로 배지를 교체하였고, 이 때에는 상기 3일 동안과 달리 매일 배지를 교체해야 한다. 6일째 되는 날 내피세포를 분류하여 CD144 positive endothelial cells만을 선별하여 재플레이팅하였으며, 이 때 사용한 배지는 StemPRo-34 supplemented with VEGF(50 ng/ml)를 이용하여 피브로넥틴이 코팅된 플레이트(fibronectin coated plates) 상에 분주하였다. 상기 ThermoFisher Scinentific로부터 구입한 StemPro-34 SFM 배지의 조성은 하기 표 8과 같다.

상기 StemPro-34 medium에는 StemPro-34 SFM medium와 StemPro-34 supplement가 포함되어 있다. 7일째되는 날부터는 Vasculife complete medium에서 세포를 배양하고 유지시켰다.
실시예 6: 섬유화 튜머로이드 항암제 저항성의 확인
본 발명자들은 상기 실시예 1 내지 실시예 4의 방법으로 제조한 고형암 조직 유사체인 섬유화 튜머로이드를 이용하여 임상 단계에서 실효를 거둘 수 있는 약물의 선별이 가능한지를 평가하기 위하여 약물 저항성 테스트를 수행하였다(도 10a 및 도 10b 참조). 또한, 이상에서 개시한 튜머로이드의 형성 과정부터 섬유화 튜머로이드를 제작하고 약물 테스트를 수행하기까지의 시간적 흐름을 도 6에 나타낸 바 있다. 약물 테스트는 세포생존력(Cell viability) 측정을 통해 진행되었다. 항암제가 포함된 군과 포함되지 아니한 대조군의 세포생존력을 유지 배지(Maintenance medium)로 교체한 후 측정하였다. 이때, 공초점현미경(Confocal microscope)을 이용하여 형광 시그널(fluorescent signal)을 측정하여 형광을 발현하지 않는 튜머로이드 상의 살아있는 암세포는 Calsein-AM-blue로, 죽은 암세포는 propidium iodide(PI)로 염색하여 확인하였다. 또 다른 측정방법으로 Promega 사의 CellTiter-Glo Luminescent cell viability assay를 이용하여, 고형암조직 유사체의 cell viability를 측정하였고, 대조군과의 비교를 통해 간접적으로 항암제에 의한 암 세포의 사멸을 확인할 수 있었다. 실험 결과를 도 10a 및 도 10b에 나타내었다. 섬유화 튜머로이드(FET)는 단순 튜머로이드(tumor only)에 비해, 높은 항암제 저항성 즉, 세포 생존성이 높게 나타났으며, 더 강한 Calsein-AM 시그널을 보이는 것을 확인하였다(도 10a 참조). 도 10b를 참조하면, 섬유화 튜머로이드(FET)는 단순 튜머로이드(Simple tumoroid)에 비해, 항암제(cisplatin, 100 uM)에 대한 통계적으로 유의한 저항성을 보이는 것을 확인할 수 있다. 상기에서 제조된 섬유화 튜머로이드는 항암제가 혈관벽과 섬유화 조직을 차례로 지나 암조직에 도달하는 일련의 과정을 인체에서와 동일하게 모사하였으므로 최종적으로 eFET를 제작하여 항암제 테스트를 진행하는 것이 바람직하지만, 비용 또는 시간을 줄이기 위하여 선택적으로 FET만을 사용할 수도 있다. FET를 사용하는 경우 실험 기간을 2일 단축시킬 수 있어 시간 및 체력적 한계가 있는 암 환자를 위한 빠른 항암제의 선제적 선별이 가능하게 하여 정밀 의료 분야에서 크게 활용될 수 있을 것으로 기대된다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
본 발명은 섬유화 튜머로이드(Fibrosis-encapsulated tumoroid, FET)의 제조 방법 및 그의 용도에 관한 것으로, 본 발명의 방법을 이용할 경우 인간 고형암의 특징을 그대로 반영함으로써 후보 물질의 임상 검증 단계에서의 성공 확률을 획기적으로 높이므로 항암 신약 개발 및 정밀 의료 분야에 널리 이용될 수 있다.
Claims (21)
- (a) 개체로부터 분리된 암 세포를 수득하는 단계; 및(b) 상기 암 세포와 함께 유도만능줄기세포(Induced pluripotent stem cell; iPSC) 또는 유도만능줄기세포 유래 세포를 공배양(co-culture)하여 스페로이드(spheroid) 형태의 배양체를 형성하는 단계;를 포함하는 섬유화 튜머로이드의 제조 방법.
- 제 1항에 있어서,상기 암 세포는 비 스페로이드 형성세포(non-spheroid forming cell; NSF)를 포함하는 것인, 제조 방법.
- 제 2항에 있어서,상기 비 스페로이드 형성세포(NSF)는 유도만능줄기세포 또는 유도만능줄기세포로부터 유래된 중간엽 줄기세포(iPSC derived mesenchymal stem cell; iMSC)에 의하여 스페로이드 형태의 배양체로 형성되는 것인, 제조 방법.
- 제 1항에 있어서,상기 (b)단계의 공배양은 암 세포:유도만능줄기세포 또는 유도만능줄기세포 유래 세포를 0.01:1 내지 100:1의 세포 수 비율로 혼합하여 배양하는 것인, 제조 방법.
- 제 4항에 있어서,상기 (b)단계의 공배양은 암 세포:유도만능줄기세포 또는 유도만능줄기세포 유래 세포를 1.5:1 내지 2.5:1의 세포 수 비율로 혼합하여 배양하는 것인, 제조 방법.
- 제 1항에 있어서,(c) 상기 배양체에 섬유화층을 유도하는 단계를 추가로 포함하는, 제조 방법.
- 제 6항에 있어서,유도된 섬유화층을 치밀화(compaction)하는 단계를 추가로 포함하는, 제조 방법.
- 제 6항에 있어서,(d) 섬유화층이 유도된 스페로이드(Fibrosis-encapsulated tumoroid; FET)를 선별하는 단계;를 추가로 포함하는, 제조 방법.
- 제 6항에 있어서,상기 (c)단계는 섬유아세포(fibroblast), 섬유아세포의 전구체 세포, 암유래 섬유아세포(cancer-associated fibroblast), 유도만능줄기세포로 유도된 중간엽 줄기세포(iMSC), 및 유도만능줄기세포로 유도된 암유래 섬유아세포(iPSC derived cancer-associated fibroblast)로 구성된 군으로부터 선택된 적어도 하나에 의하여 섬유화층이 피막형성(encapsulation)되는 것인, 제조 방법.
- 제 8항에 있어서,(e) 상기 섬유화층 상에 혈관세포층을 유도하는 단계;를 추가로 포함하는, 제조 방법.
- 제 10항에 있어서,상기 혈관세포층은 내피세포(endothelial cell) 또는 혈관내피세포(Vascular endothelial cell)에 의해 유도되는 것인, 제조 방법.
- 제 11항에 있어서,상기 내피세포 또는 혈관내피세포는 유도만능줄기세포(iPSC)로부터 유도된 것인, 제조 방법.
- 제 1항에 있어서,상기 암은 유방암, 자궁암, 식도암, 위암, 뇌암, 직장암, 대장암, 폐암, 피부암, 난소암, 자궁경부암, 신장암, 혈액암, 췌장암, 전립선암, 고환암, 후두암, 구강암, 두경부암, 갑상선암, 간암, 방광암, 골육종, 림프종, 백혈병 또는 이들의 조합으로 구성된 군으로부터 선택된 암인, 제조 방법.
- 제 1항에 있어서,상기 공배양은 DMEM(Dulbeco's Modified Eagle's Medium), IMDM(Iscove's Modified Dulbecco's Medium), a-MEM(Alpha Modification of Eagle's Medium), TI-free OGM 배지(TI-free Osteoblast Growth Medium), F12(Nutrient Mixture F-12), RPMI 1640 배지(RPMI 1640 Medium), 윌리암 배지 E(Williams' s medium E), 맥코이 5A(McCoy' s 5A), E8 배지(Essential 8 medium), SFM 배지(StemPro-34 SFM medium), N2B2 배지(N2B2 medium), OBM 배지(OGM Osteoblast Growth Medium), 성장 배지-5(Growth medium-5), 성장 배지-10(Growth medium-10) 및 DMEM/F12(Dulbecco's Modified Eagle Medium: Nutrient Mixture F-12)로 이루어진 군에서 선택된 1종 이상의 배양 배지에서 수행되는, 제조 방법.
- 제 14항에 있어서,상기 배양 배지는 B27, N2, G5, 또는 포스콜린(Forskolin)을 추가로 포함하는 것인, 제조 방법.
- 제 14항에 있어서,상기 배양 배지에 암유래 섬유아세포(cancer-associated fibroblast); 내피세포(endothelial cell); iPSC(Induced pluripotent stem cell)로부터 분화된 중간엽줄기세포(iMSC); 또는 성장 인자(growth factor)를 추가로 포함하는, 제조 방법.
- 제 16항에 있어서,상기 성장 인자는 혈관내피세포 성장인자(Vascular Endothelial Growth Factor, VEGF); 혈관내피세포 성장인자 A(Vascular Endothelial Growth Factor A, VEGFA); 형질전환 성장인자 베타(Transforming growth factor beta, TGF-β), 염기성 섬유아세포 성장 인자(basic fibroblast growth factor; bFGF), 상피세포 성장 인자(epidermal growth factor; EGF) 및 인슐린-유사 성장 인자-1(Insulin-like growth factor-1; IGF-1)로 이루어진 군에서 선택된 1종 이상인, 제조 방법.
- 제 1항에 있어서,상기 공배양은 2일 내지 14일 동안 배양하는 것인, 제조 방법.
- 제 1항 내지 제 18항 중 어느 한 항의 제조 방법으로 제조된 섬유화 튜머로이드.
- 제 19항의 튜머로이드가 이식된 인간을 제외한 동물 모델.
- (a) 제 19항의 튜머로이드를 준비하는 단계;(b) 상기 튜머로이드에 항암제 후보 물질을 처리하는 단계; 및(c) 상기 튜머로이드에 항암 효과가 나타나는 경우에, 상기 후보 물질을 암 치료 또는 개선용 물질로 판단하는 단계;를 포함하는, 암의 치료 또는 개선용 물질의 스크리닝 방법.
