Abstract
L'invention concerne un kit permettant de construire une bibliothèque de séquençage de TCR monocellulaire humain et son utilisation. L'invention concerne également une méthode basée sur un kit destinée à construire une bibliothèque de séquençage de TCR monocellulaire humain, comprenant : l'utilisation d'une puce microfluidique pour effectuer une étape de capture et d'encapsulation de cellules isolées, puis la réalisation d'étapes telles qu'une transcription inverse d'ARN, une pré-amplification d'ADNc, une première amplification, une seconde amplification, une fragmentation, une amplification et une purification. Lorsque le kit et la bibliothèque de séquençage de TCR monocellulaire humain sont appliqués au séquençage du répertoire immunitaire de cellules immunitaires, de 500 à 30000 cellules peuvent être séparées en une seule expérience.
