Definitions
- the present disclosure relates to a method and an apparatus for designing a simulated sample used in evaluating the performance of a near-infrared spectroscope.
- NIRS Near-infrared spectroscopy
- TRS Time Resolved Spectroscopy
- a simulated sample (phantom) is used to evaluate the performance (measurement accuracy, reproducibility, stability, etc.) of the near-infrared spectroscope.
- the simulated sample is required to have optical characteristics close to the optical characteristics of the object of spectroscopic measurement. Further, in order to evaluate the reproducibility and long-term stability of the near-infrared spectroscope, it is desired that the change over time in the optical characteristics of the simulated sample is small.
- resins, water, gelatin, agar (water base), oil (lard, butter) and the like have been used as base materials for simulated samples.
- Patent Document 1 describes the invention of a simulated sample using a resin as a base material. Since the simulated sample is often used to evaluate basic characteristics such as measurement accuracy at each wavelength of the near-infrared spectroscope, a dye (for example, carbon ink) whose absorption spectrum does not change significantly in the near-infrared region is used as a base material. It is contained in. As the wavelength becomes longer in the near infrared region, the absorption of carbon ink decreases monotonically. Therefore, it is difficult for this simulated sample to reproduce the absorption spectrum of living tissue.

- FIG. 1 is a flowchart of a simulated sample design method and a simulated sample preparation method.
- the simulated sample design method includes a modification step S11 and a calculation step S12.
- the simulated sample preparation method includes a design step S10 and a preparation step S13 including a modification step S11 and a calculation step S12.
- the simulated sample to be designed and manufactured is one in which a resin is used as a base material and two or more N kinds of dyes are contained in the base material, and it is also preferable that the simulated sample contains scattered particles.
- the correction step S11 first, the target of spectroscopic measurement by the near-infrared spectroscopic apparatus, the resin used as the base material of the simulated sample, and the N kinds of dyes contained in the base material are determined. Then, the absorption spectrum of the target is modified based on the refractive index of the target of spectroscopic measurement and the refractive index of the resin to create a corrected absorption spectrum. Details of this modification will be described later.
- the target of spectroscopic measurement is arbitrary, but it is a living tissue such as a breast, a head, a leg / arm muscle, and the like.
- the amount of hemoglobin, the amount of water, and the amount of fat differ depending on the target site. Therefore, the absorption spectrum of the target differs depending on the target and may differ depending on the state of the target.
- the resin and dye are determined according to the subject.
- the resin used as the base material of the simulated sample may be in a cured state or may be used in a liquid state.
- the cured resin is used as a base material, the dye and scattered particles are mixed and then cured.
- various resins such as epoxy resin, polyurethane resin, and silicone rubber can be used.
- the resin may be cured by any method such as 1-component curing type, 2-component mixed type, ultraviolet curing type, and thermosetting type. It is desirable to use a resin having a transmittance of 95% or more in the wavelength range of approximately 700 nm to 1100 nm.
- the N types of dyes those in which the absorption spectrum of the simulated sample can accurately approximate the target absorption spectrum by appropriately setting the content concentration of each dye in the base material (resin) are used.
- the pigment A and the pigment B among the N types of pigments can be as follows.
- Dye A is selected so that the simulated sample can reproduce the light absorption characteristics of deoxygenated hemoglobin (HHb) by itself or by mixing with other dyes.
- HHb deoxygenated hemoglobin
- Various things such as 761 can be used.
- Dye B is selected so that the simulated sample can reproduce the light absorption characteristics of oxygenated hemoglobin (O 2 Hb) by itself or by mixing with other dyes.
- a dye B it is desirable that the absorption gradually increases from the wavelength of 700 nm toward the long wavelength side and has an absorption peak near the wavelength of 900 nm in the near infrared region of a wavelength of 700 nm to 1100 nm, similar to oxygenated hemoglobin.
- Fujifilm's Pro-Jet 900NP, Exciton's IRA 912, IRA 931, etc. can be used.
- the scattered particles are added to the base material in order to reproduce the light scattering characteristics close to the target in the simulated sample.
- various particles such as titanium oxide (TiO 2 ), aluminum oxide (Al 2 O 3 ), silica particles (SiO 2 ), silicon powder (Si), and acrylic powder can be used.
- the diameter of the scattered particles is preferably about 0.01 ⁇ m to 10 ⁇ m.
- the simulated sample may have a plurality of regions having different absorption spectra.
- the absorption spectra of the internal region of the simulated sample and the peripheral region surrounding the internal region may be different from each other.
- the processing of the correction step S11 is performed for each area, and the processing of the calculation step S12 and subsequent steps is performed.
- the cancer tissue in the normal tissue, the pathological condition such as the change in the hemoglobin concentration or the tissue oxygen saturation in the living body, and the change in the living tissue, etc., can be reproduced according to the actual measurement. It is possible to realize a simulated sample that is also possible.
- the absorption spectrum of the simulated sample composed of the base material containing the N types of dyes is in the near infrared region with respect to the corrected absorption spectrum.
- the content concentration of each of the N types of dyes to be contained in the base material is determined so as to approximate in the predetermined wavelength range of.
- the predetermined wavelength range is a wavelength range used for spectroscopic measurement of an object, and includes, for example, 750 nm to 850 nm when the object is a biological tissue.
- the least squares method can be used to calculate the content concentration of each dye. That is, the absorption coefficient of the resin at the wavelength ⁇ is expressed as ⁇ ⁇ a, resin .
- the absorption coefficient of the nth dye among the N kinds of dyes at the wavelength ⁇ per unit concentration is expressed as ⁇ ⁇ a, n, and the content concentration of the nth dye is expressed as C n .
- the value of the corrected absorption spectrum at the wavelength ⁇ is expressed as ⁇ ⁇ a, tissue .
- the absorption coefficient ⁇ ⁇ a, phantom (C) at the wavelength ⁇ of the simulated sample is expressed by the following equation (1). Then, the following (2) F of the formula (C) is minimized (or smaller than a predetermined threshold value) such Request containing a concentration C 1 ⁇ C N. The summation calculation on the right side of equation (2) is obtained for each wavelength within a predetermined wavelength range in the near infrared region.
- the degree of agreement between the absorption spectrum of the simulated sample composed of the base material containing N kinds of dyes at the obtained content concentration and the modified absorption spectrum may be obtained.
- the degree of agreement is expressed by, for example, the above equation (2).
- the concentration of each of the N types of dyes to be contained in the base material is obtained for each group consisting of any N types of dyes among the three or more M types of dyes, and the degree of agreement is obtained. It is preferable to select the N dyes and content concentrations of any group based on the number and degree of agreement of the dye types in each group. As the number of types of dyes contained in the base material increases, the preparation of a simulated sample becomes more complicated. Therefore, it is preferable that the number of types of dyes is small if a desired degree of agreement can be obtained. Description
a,resinと表す。
a,tissueと表す。
この(8)式は、樹脂の吸収係数が見かけ上ではcresin/ctissue(=ntissue/nresin)倍されていることを表している。
