Abstract
자연 살해 세포의 배양용 조성물, 키트 및 이를 이용한 방법에 관한 것이다. 일 양상에 따른 자연 살해 세포 배양용 조성물, 및 이를 이용한 자연 살해 세포 배양 방법에 따르면 말초혈액 단핵구에서 자연 살해 세포를 배양함에 있어서, 자연 살해 세포를 적어도 일면에 활성화 수용체 리간드, 억제성 수용체 리간드, 보조자극 수용체 리간드, 사이토카인, 사이토카인 수용체, 면역관문 리간드, 블로킹 (blocking) 항체, 또는 이들의 조합이 부착된 자성 입자를 포함하는 자연 살해 세포 배양용 조성물을 함유하는 배지에서 배양하여, 대량으로 증식시키고 자연 살해 세포의 활성화 또는 억제, 자연 살해 세포의 확장을 촉진시킬 수 있다. 따라서, 이를 이용하여 배양된 자연 살해 세포를 면역세포 치료제로 유용하게 사용할 수 있다. 또한, 자성 입자는 배지로부터 쉽게 분리할 수 있어 간편하고, 경제적이며 안전한 자성 입자를 사용하기 때문에 임상적으로 안전성이 우수하다.
Description
자연 살해 세포의 배양용 조성물, 키트 및 이를 이용한 방법에 관한 것이다. 본 출원은 2019년 5월 15일 출원된 대한민국 특허출원 제10-2019-0057136호를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다.
현재 암의 일반적인 치료법은 외과적 수술(surgery), 방사선 요법(radiation therapy), 항암화학요법(chemotherapy) 등이 있으며, 암의 종류 및 병기에 따라 단독 혹은 복합적으로 사용되고 있지만, 환자들에게 심한 부작용과 고통을 수반하고, 기존 치료법은 정상 세포에 어느 정도 손상을 미치는 점에서 한계가 있다. 최근 부각되고 있는 암 면역치료법(cancer immunotherapy)은 인체 고유의 면역 시스템을 활용하여 정상 세포의 손상을 최소화하여 암세포를 보다 특이적으로 제거하는 치료법으로, 여러 세부 분야(항체 치료법, 면역세포 치료법, 바이러스 면역치료법, 나노 기술 면역치료법 등)의 연구가 활발히 진행되고 있다. 이 중 면역세포 치료는 환자의 혈액에서 얻어진 림프구(lymphocyte) 중, 자연 살해 세포(natural killer cell: NK 세포), 자연살생 T 세포(natural killer T cell), T 세포, B 세포, 수지상 세포(dendritic cell) 등의 세포 수를 늘리고, 체외(in vitro) 에서 기능을 강화하여, 이를 다시 환자 몸에 되돌림으로써, 암을 치료하는 방법이다. 이러한 면역세포를 이용한 치료법은 면역 반응 조절 치료에 좋은 효과를 보이고, 독성 및 안전성 면에서 우수하다고 평가된다.
이 중 NK 세포는 선천면역(innate immunity)을 담당하는 중요한 세포로, 바이러스에 감염된 세포나 종양세포 등 비정상 세포를 스스로 식별하여 사멸시키는 기능을 가진다. 또한 T 세포가 인식하지 못하는 종양 및 바이러스에 감염된 세포를 인식할 수 있고, T 세포에 비해 안전성이 우수한 특징이 있다. 이에 지난 10 여 년 동안 환자들의 면역 시스템을 이용한 종양 면역치료가 꾸준히 발전되어 왔고, 이를 이용한 세포 치료제(cell therapy product)도 상업화되고 있다.
NK 세포를 이용한 세포치료제 개발을 위해, NK 세포의 기능 강화 및 활성화, 배양, 확장이 필요하다. 종래 NK 세포의 배양 또는 확장을 위해 배양 시 공여자 세포가 필요하다. 종래 공여자 세포로 K562와 같은 세포가 사용되는데, 이는 암 세포로 임상에 사용하기 부적합하다.
따라서, 암과 같은 난치성 질환의 치료를 위한 세포치료제로서 면역 거부 반응을 해소할 수 있으며, 환자 맞춤형 세포 치료를 제공할 수 있는, NK 세포를 대량으로 증식하는 방법 및 활성이 강화 또는 억제된 NK 세포를 배양하는 방법에 대한 연구가 필요하다.

본 명세서에서 용어 "자연 살해 세포(natural killer cell: NK cell)"는 림프구의 일종인 거대 과립형 림프구(Large granular lymphocytes: LGL)로서, 감염된 바이러스 및 종양세포를 죽이는 능력이 뛰어나고, 대부분의 정상 세포는 죽이지 않는 특성을 가진다. 따라서, 자연 살해 세포는 활성 세포 독성 T 림프구가 다량으로 생성되기 전에 바이러스 감염 혹은 종양 형성 초기 단계에서 중요한 역할을 한다. 예를 들면, 자연 살해 세포가 표적 세포와 접촉하는 경우, 몇몇 분자는 표적 세포의 막에 구멍을 형성하여 세포를 용해시키며, 또 다른 분자는 표적 세포에 들어가서 핵 DNA의 분절을 증가시켜, 괴사(necrosis), 세포사멸(Apotosis) 또는 세포 예정사(Programmed cell death)를 야기한다.
상기 자연 살해 세포는, 예를 들면, 포유동물, 인간, 원숭이, 돼지, 말, 소, 양, 개, 고양이, 마우스 또는 토끼 등으로부터 유래한 것일 수 있다. 상기 자연 살해 세포는 정상인 또는 암환자로부터 수득한 것일 수 있다. 상기 자연 살해 세포는 혈액, 말초혈액 단핵구(Peripheral blood mononuclear cell: PBMC)로부터 분리된 것일 수 있다. 혈액을 분리하는 방법, 이로부터 PBMC를 분리하는 방법, 이로부터 자연 살해 세포를 분리하는 방법은 공지된 방법으로 수행되는 것일 수 있다.
상기 조성물은 적어도 일면에 활성화 수용체 리간드, 억제성 수용체 리간드, 보조자극 수용체 리간드, 사이토카인, 사이토카인 수용체, 면역관문 리간드, 블로킹 (blocking) 항체, 또는 이들의 조합이 부착된 자성 입자를 포함함으로써, 용해성 활성화 수용체 리간드, 억제성 수용체 리간드, 보조자극 수용체 리간드, 사이토카인, 사이토카인 수용체, 면역관문 리간드, 블로킹(blocking) 항체, 또는 이들의 조합을 포함하는 자연 살해 세포 배양용 조성물에 비해 자연 살해 세포를 활성화, 증식, 확장 또는 억제하는 효과가 증가한다.
일 양상에 따른 조성물에서, 상기 활성화 수용체 리간드는 고형암, 또는 혈액 암세포를 포함하는 변형된 세포(transformed cells), 바이러스 감염세포(virus infected cells), 및 스트레스성 세포(stressed cells)에서 발현하는 단백질로서, 상기 언급된 NK세포 수용체와의 결합을 통해 NK세포의 활성화 및 작동 기능을 유발 가능한 물질을 지칭할 수 있다. 상기 활성화 수용체 리간드는 NK세포 활성화 수용체 구조에 따른 3가지 분류인 natural cytotoxicity receptor(NCR) family, NKG2 family, killer cell immunoglobulin like receptor(KIR) family에 대한 리간드를 포함할 수 있다. 또한, 상기 활성화 수용체 리간드는 예를 들어, BAG6, AICL, MICA, MICB, CADM1, IgG, CD48, NTB-A/SLAMF6, CD70, CD155, CD319, C8, C9, 및 CS1로 구성된 군에서 선택된 1종 이상일 수 있다.
용어 "활성화 수용체 리간드"는 수용체의 특정 부위게 결합하여 수용체를 활성화할 수 있는 물질을 지칭할 수 있다.
일 양상에 따른 조성물에서, 상기 억제성 수용체 리간드는 예를 들어, HLA-A, HLA-B, HLA-BW4, HLA-C1, HLA-C2, HLA-E, HLA-G, CD112/Nectin-2, CD112/Nectin-3, 카드헤린(cadherin), 콜라겐, OCIL, 및 CLEC2D로 구성된 군에서 선택된 1종 이상일 수 있다.

용어 "블로킹 항체"는 항원과 결합할 때 반응이 없지만, 다른 기존의 항체가 항원과 결합하는 것을 방지하는 immunoglobulin 단백질을 지칭할 수 있다. 구체적으로, 상기 블로킹 항체는 Y자형의 단백질로서 2개의 경사슬과 2개의 중사슬 2개가 이황화 결합을 하여 이루어져 있으며, 불변영역과 가변영역을 지닌다. 중쇄의 불변영역의 차이에 따라 IgA, IgD, IgM, IgE, IgG isotype을 지닐 수 있고, 가변영역 내 아미노산 서열의 변형을 통하여 다수 단백질간의 결합을 억제할 수 있다. 상기 언급된 블로킹 항체는 NK세포의 활성화, 억제성, 보조자극, 면역관문 역할을 수행하는 수용체/리간드에 대한 것일 수 있다.
일 양상에 따른 조성물에서, 상기 자성 입자 표면의 적어도 일면은 단백질 G(protein G) 또는 단백질 A(protein A)로 코팅된 것일 수 있다. 상기 단백질 G 또는 단백질 A는 면역글로불린과 결합 친화력이 우수한 것으로서 자성 입자에 코팅될 수 있는 것이라면 그 종류가 제한되는 것은 아니다.
일 양상에 따른 조성물에서, 상기 활성화 수용체 리간드, 억제성 수용체 리간드, 보조자극 수용체 리간드, 사이토카인, 사이토카인 수용체, 면역관문 리간드, 및 블로킹 (blocking) 항체는 인간 면역글로불린과 융합된 형태일 수 있다.
일 양상에 따른 조성물에서, 상기 IL-15R은 인터루킨-15 수용체 α(interleukin-15 receptor α: IL-15Rα)일 수 있다.
상기 IL-15R은 NK 세포에서 발현되는 IL-15 수용체로서, NK 세포의 성장과 분화를 증진시키는 기능에 관여할 수 있다. 이러한 수용체들 중에서, 예를 들어, IL-15Rα는 전형적 신호전달(classic signaling) 뿐만 아니라, 트랜스-신호전달 (trans-signaling) 역시 수행할 수 있다. 상기 트렌스-신호전달은 세포가 IL-15 Rα를 발현하지 않을지라도, 이웃 세포 표면에 IL-15 Rα가 발현되고 IL-15가 상기 수용체가 결합할 경우, 세포간 트랜스로 cross-linking되어 활성화 신호가 전달되는 것을 지칭할 수 있다. 상기와 같이 트랜스-신호전달을 수행할 수 있는 수용체들의 생물학적 메커니즘은 일 양상에 따른 배양용 조성물에 접목할 수 있다.
상기 4-1BB 리간드는 자연 살해 세포의 확장에 관여하는 분자로 알려져 있다.

상기 자성 입자는 표면에 단백질 G 또는 단백질 A가 부착될 수 있는 모든 자성 입자들 중에서 선택될 수 있다. 예컨대, 상기 자성 입자는 평균 입경이 약 500nm 내지 약 10μm, 약 550nm 내지 약 9μm, 약 600nm 내지 약 8μm, 약 650nm 내지 약 7μm, 약 600nm 내지 약 6μm인 자성 입자일 수 있으나, 이에 제한되는 것은 아니다.
상기 자성 입자는 작은 입자 크기를 가짐으로써 개별 입자가 단일자기구역을 갖게 되고, 이로 인해 외부 자기장이 존재 시에만 자성의 특성을 갖는 초상자성(superparamagnetism)을 나타낸다. 초상자성을 나타내는 자성 입자는 외부 자기장 인가에 의해 간단하고 빠르게 분리될 수 있다. 자기장 인가에 의한 자성 입자의 분리는 pH, 온도, 이온 등과 같은 주변 환경에 영향을 받지 않으므로 안정성 및 민감성이 우수하다.

이하 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 하나 이상의 구체예를 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1: NK 세포 배양용 자성 입자를 이용한 NK 세포 배양
1.1
NK 세포 배양용 자성 입자의 제조
NK 세포 배양시 지지 세포(feeder cell)의 역할을 할 수 있는 자성 입자로, 4-1BB 리간드 또는 IL-15Rα가 부착된 자성 입자를 제조하였다. 자성 입자에 특정 단백질(4-1BB 리간드 또는 IL-15Rα)을 부착시키기 위해, 단백질 G(Protein G)로 코팅된 자성 입자를 사용하였다. 단백질 G는 인간 면역글로불린과 매우 높은 결합 친화성을 갖기 때문에, 인간 면역글로불린과 융합된 특정 단백질을 사용하면 단백질 G와 면역글로불린 사이의 결합을 통해 특정 단백질을 자성 입자에 부착시킬 수 있다.
구체적으로, 단백질 G로 코팅된 자성 입자인 Dynabeads (Thermo Fisher Scientific, Novex 쪠)를 구입하였다. 특정 단백질로는 인터루킨-15 수용체 알파 (interleukin-15 receptor alpha: IL-15Rα)와 4-1BB 리간드(4-1BB ligand: 4-1BBL)를 사용하였고, IL-15Rα가 융합된 면역글로불린-태그(tagged) 단백질(이하 'IL-15Ra_IgG1Fc', R&D systems, Minneapolis, MN, USA) 및 4-1BBL이 융합된 면역글로불린-태그 단백질(이하 '4-1BBL_IgG1Fc', ACRObiosystems, Newark, DE, USA)을 구입하였다.
상기 면역글로불린-태그 단백질 및 단백질 G가 코팅된 자성 입자를 약 1시간 동안 차가운 방에서 반응시키고, 상층액은 완충액 흡입을 사용하여 MagneSphere 자기 스탠드(Promega, Madison, USA)로부터 제거함으로써 'NK 세포 배양용 자성 입자'를 제조하였다.

NK 세포의 배양
NK 세포는 말초 혈액 단핵구 (Peripheral blood mononuclear cells: PBMCs)로부터 배양하여 얻었다. CHA 의과학 대학교에서 기관 생명윤리위원회(IRB)의 승인 (1044308-201701-BR-005)하에 건강한 5명의 혈액 기증자로부터 PBMC를 수득하였다. 채혈된 전혈을 인산염 완충 식염수(phosphate buffered saline: PBS)로 희석하였다. PBS로 희석한 혈액 시료로부터 SepMate 쪠 튜브 (STEMCELL Technologies, Inc., Vancouver, Canada)로 PBMC를 분리하였다. PBMC 분리는 Histopaque-1077 (Sigma-Aldrich, St. Louis, USA)을 사용하여 피콜-하이팩(Ficoll-Hypaque) 밀도 구배 원심 분리에 의해 수행하였다.
수득한 PBMC의 배양은 24 웰에서 1Х106 PBMC/웰의 밀도로 수행하였다. 배지의 일부는 PBMC 형태에 따라 매 2-4 일마다 변경되거나 추가하였으며, 세포 계수는 매 6 일 (6 일 및 12 일)마다 수행하였다. PBMC는 10%의 열-불활성화 FBS (Gibco, Thermo Fisher, USA), 베타-메르캅토에탄올 50μM 및 1% 페니실린 및 스트렙토마이신(P/S)으로 보충된 basic RPMI-1640 (Hyclone, Logan, USA) 에스)에서 유지하였다.
이하에서, NK 세포는 실시예 1.2에서 수득한 전체 PBMC를 사용하여 PBMC 중의 NK 세포를 배양하는 방식을 통해 배양하였고, 실험에 사용된 실험군은 다음과 같다: ① 용해성 형태인 IL-15Rα 또는 4-1BBL을 포함한 배지에 배양 ② 실시예 1.2의 자성 입자를 포함한 배지에 배양 ③ 특정 분자가 부착되지 않은 자성 입자를 포함한 배지에 배양.
실험예 1: NK 세포 배양용 자성 입자를 이용한 NK 세포 증식 평가
상기 실시예 1.1에서 제조한 NK 세포 배양용 자성 입자를 이용하는 경우의 NK 세포 증식 효율을 평가하였다. NK 세포 증식을 측정하기 위해, 상기 실험군 3종에 대하여 매 6 일마다 (0 일, 6 일 및 12 일째) 유세포 분석을 수행하였다. PBMC의 증식은 혈구 계수기로 측정하였다. PBMC 계수 후, 일부 시료를 유세포 분석에 사용하였다. CD3 PE 항체 (Thermo Fisher Scientific) 및 CD56 FITC 항체 (Thermo Fisher Scientific)를 PBMC 시료에 처리하여 염색하였다. 이 두 항체의 염색을 통해 수득된 PBMC에서 NK 세포, NKT 세포,T 세포, B 세포 및 단핵구의 개체군을 구별할 수 있다.

도 1에 나타낸 바와 같이, 용해성 IL-15를 이용하여 배양한 경우에 비해, 4-1BBL 또는 IL-15Rα이 부착된 일 양상에 따른 NK 세포 배양용 자성 입자를 이용하여 배양한 PBMC 군에서 세포들이 더 잘 응집된 것을 확인하였다.
배양 시 NK 세포가 응집되어 클러스터를 형성하는 현상은 NK 세포의 활성화가 강화되는 것임이 공지되어 있다. 따라서, 일 양상에 따른 4-1BBL 또는 IL-15Rα이 부착된 자성 입자를 이용하여 PBMC를 배양하는 경우 PBMC가 잘 응집하므로 NK 세포가 활성화되는 것을 알 수 있다.
도 2는 용해성 IL-15(a), 용해성 IL-15 및 자성 입자(b), 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자(c), 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자(d)를 이용한 배양 6일, 12일차에 혈구계(hemocytometer)를 이용하여 세포를 계수한 결과이다.
도 2에 나타낸 바와 같이, 모든 군에서 PBMC의 수는 12일차에 전반적으로 증가한 것을 확인하였다. 그러나, 특히 일 양상에 따른 4-1BBL 및/또는 IL-15Rα가 부착된 NK 세포 배양용 자성 입자를 이용하여 PBMC를 배양한 경우 세포 수가 크게 증가한 것을 확인하였다.
도 3은 용해성 IL-15(a), 용해성 IL-15 및 자성 입자(b), 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자(c), 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자(d)를 이용한 배양 6일, 12일차에 혈구계(hemocytometer)를 이용하여 세포를 계수한 결과를 대응 표본(paired t-test)으로 확인한 그래프이다.
도 3에 나타낸 바와 같이, 용해성 IL-15 형태로 처리된 PBMC 수와 실험군을 비교한 결과 용해성 IL-15와 자성 입자를 처리한 PBMC 수는 유의한 차이가 없었다. 반면, 4-1BBL_IgG1Fc 또는 IL-15Rα_IgG1Fc이 부착된 자성 입자를 이용하여 PBMC를 배양한 경우의 세포 수가 유의적으로 증가한 것을 확인하였고, 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 모두 부착된 자성 입자를 이용한 경우에 NK 세포 수가 가장 많이 증가하였다.
실험예 2: NK 세포 배양용 자성 입자의 이용에 따른 PBMC 내 NK 세포 구성 비율 평가
상기 실시예 1.1에서 제조한 NK 세포 배양용 자성 입자를 이용하는 경우 PBMC 내의 NK 세포 구성 비율이 변화하는지 평가하였다. PBMC를 각 CD 마커(CD3, CD56)로 염색한 후 세포 집단을 유세포 분석법으로 확인하였다.
구체적으로는, NK 세포 마커 CD3 PE 항체 (Thermo Fisher Scientific) 및 CD56 FITC 항체 (BD PharmingenTM)를 사용하여 배양 0 일, 6 일 및 12 일차에 PBMC에서의 NK 세포의 비율을 유세포 계측 분석기 CytoFLEX (Beckman Coulter, Inc., Brea, CA, USA)를 통해 측정하였다.
도 4는 배양 0일차(a), 배양 12일차(b), 용해성 IL-15를 이용한 배양 12일차(c), 용해성 IL-15 및 특정 분자가 부착되지 않은 자성 입자를 이용한 배양 12일차(d), 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자를 이용한 배양 12일차(e), 및 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자를 이용한 배양 12일차(f)에 CD3, CD56 마커로 염색한 FACS 결과를 나타낸 도이다. FACS 데이터에서, x축은 CD56을 나타내고, y축은 CD3을 나타낸다.
도 4에 나타낸 바와 같이, 배양 전(a) 평균 NK 세포의 비율은 15.58±4.40% (범위, 7.29-32.10)이었다. 각 조건에 따라 배양한 결과, 도 4c 및 도 4d에서 NK 세포의 비율은 각각 11.73±0.93% (범위, 5.05-18.4) 또는 19.19±2.35% (범위, 6.70-35.70)이었다. 반면, 4-1BBL_IgG1Fc 또는 4-1BBL_IgG1Fc가 부착된 자성 입자와 배양된 PBMC(도 4e 및 도 4f)에서 NK 세포의 비율은 각각 46.74±6.45% (범위, 4.53-74.70) 및 49.59±6.43% (범위, 9.00-79.20)로 NK 세포 마커 발현이 크게 증가한 것을 확인하였다(p <0.001).
도 5는 용해성 IL-15(a), 용해성 IL-15 및 자성 입자(b), 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자(c), 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자(d)를 이용한 배양 12일차에 CD3, CD56 마커로 염색한 FACS 결과를 5명의 공여자 사이에서 비교한 결과이다. 도 4 및 도 5에 나타낸 바와 같이, 일 양상의 특정 분자가 부착된 자성 입자와 함께 배양된 군에서 PBMC 내 NK 세포의 비율은 통계적으로 유의하게 증가된 것을 확인하였다.
또한, 배양 후 NK 세포 수의 변화를 확인하기 위해, 항체 염색에 의해 얻어진 NK 세포의 백분율에 도 2서 얻은 각각의 PBMC 수를 곱하였다. 이후 각 공여자에서의 NK 세포의 수를 실험 조건에 따라 플롯팅하였다.
도 6은 용해성 IL-15(a), 용해성 IL-15 및 자성 입자(b), 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자(c), 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자(d)를 이용한 배양 6일차 및 12일차에 각 공여자에서 NK 세포의 수를 계산할 결과를 나타낸 그래프이다.
도 6에 나타낸 바와 같이, NK 세포의 수가 다른 두 대조군에 비해 특정 분자가 부착된 자성 입자와 함께 배양되었을 때 유의적으로 증가한 것을 확인하였다.
또한, NK 세포(CD3-, CD56+)의 백분율 데이터 이외에 T 세포(CD3+, CD56-) 및 NKT 세포(CD3+, CD56+), B 세포 및 단핵구(CD3-, CD56-)에 관한 데이터 또한 CD3, CD56 항체를 이용하여 동일한 방법으로 얻었다. NK 세포(CD3-, CD56-)를 포함하는 전체 PBMC의 비율에 있어 배율 변화(fold change)를 측정한 결과를 하기 표 1에 나타내었다. 표 1은 NK 세포, T 세포 및 NKT 세포를 포함하는 전체 PBMC의 증가율의 배율 변화를 도표화한 결과이다. 대응 표본(paired t-test)에 의해 통계적 분석을 수행하였다.

(*P<0.05)따라서, NK 세포의 백분율과 수 모두 특정 분자가 부착된 자성 입자와 함께 배양된 경우에 유의적으로 증가하였기 때문에, 특정 분자가 부착된 자성 입자와 함께 PBMC를 배양하는 경우 PBMC 내 환경이 NK 세포 우세 환경으로 유도되는 것을 알 수 있다.
실험예 3: NK 세포 배양용 자성 입자의 이용에 따른 PBMC 매개된 세포사멸능평가
상기 실시예 1.1에서 제조한 NK 세포 배양용 자성 입자를 이용하는 경우 PBMC 내에서 NK 세포의 수가 증가되는 효과 이외에, NK 세포의 면역 세포 기능 향상을 평가하기 위해 K562에 대한 PBMC 매개된 세포사멸능 평가를 수행하였다.
구체적으로, 백혈병 세포주인 K562에 대해 상기한 바와 같이 12 일 동안 배양된 PBMC를 처리하였다. 먼저, 유세포 분석에서 표적 세포인 K562와 작용 세포(effector cell)인 PBMC를 구별하기 위해 CFSE 염색을 수행하였다. 이처럼 작동 세포(NK 세포)만을 게이팅하여 K562와 함께 4 시간 동안 배양한 후, 사멸된 K562 세포를 검출하기 위해 7-AAD 염색을 수행하였다. 여기서 공배양은 E:T = 10:1 비율로 수행하였다. 염색된 7-AAD 값을 검출하여 표적 세포 중 몇%가 사멸되었는지 확인하였다. 사멸된 세포는 K562 도트 플롯(dot plot)에 기초하여 선택하였다.
도 7은 용해성 IL-15(a), 용해성 IL-15 및 자성 입자(b), 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자(c), 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자(d)를 이용한 배양 12일차에 PBMC의 세포사멸능을 평가하기 위해 CFSE 및 7-AAD 염색에 의한 FACS 결과를 나타낸 도이다. FACS 데이터에서 y축은 7-AAD이고, 선상에서 염색된 세포는 실제로 사멸된 세포였다. CSFE는 NK 세포, K562 세포를 구분하기 위한 목적으로 사용되며, 세포질에 광범위하게 염색된다. 7-AAD는 사멸된 세포를 염색하기 위한 목적으로 사용되며, DNA break가 일어나면 염기에 결합하여 염색된다.
도 8은 용해성 IL-15(a), 용해성 IL-15 및 자성 입자(b), 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자(c), 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자(d)를 이용한 배양 12일차에 PBMC의 세포사멸능 평가를 4명의 공여자 사이에서 비교한 결과를 나타낸 그래프이다. 상기 유세포 분석 결과에 대하여 대응 표본(paired t-test)에 의한 통계적 분석의 결과로, 데이터는 평균 ± 표준편차로 표현하였다(* P <0.05).
도 7 및 도 8에 나타낸 바와 같이, 면역 세포의 K562에 대한 세포사멸 기능은 특정 분자가 부착된 자성 입자로 처리한 경우에 증가한 것을 확인하였다.
따라서, 일 양상에 따른 자성 비드와 함께 배양한 PBMC군에서 K562에 대한 세포 사멸능이 향상되기 때문에, 세포의 수뿐만 아니라 세포 독성 기능 또한 향상된 것을 알 수 있다.
실험예 4: NK 세포 배양용 자성 입자의 이용에 따른 PBMC의 인터페론 분비 평가
상기 실시예 1.1에서 제조한 NK 세포 배양용 자성 입자를 이용하는 경우 PBMC의 인터페론 분비가 향상되는 효과가 있는지 평가하기 위해 세포 독성 기능과 밀접하게 관련되어 있고, 활성화된 NK 세포에서 높게 분비되는 IFN-γ의 양을 정량화하는 실험을 수행하였다.
상기 기재된 것과 같은 각 실험 조건 하에서 12 일 동안 배양한 상층액을 이용하였다. IFN-γ는 항체가 코팅 된 IFN-γ를 포획한 플레이트를 사용하여 ELISA (enzyme-linked immune-specific assay) 방법으로 검출하였다.
도 9는 용해성 IL-15(a), 용해성 IL-15 및 자성 입자(b), 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자(c), 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자(d)를 이용한 배양 12일차에 PBMC의 배양 상층액에서 IFN-γ를 ELISA에 의해 검출한 결과를 나타낸 도이다. 데이터는 평균 ± 표준편차로 표시하였다 (*** P <0.001).
도 9에 나타낸 바와 같이, 용해성 IL-15_IgGFc과 4-1BB_IgGFc 및 IL-15Rα가 부착된 자성입자와 함께 배양한 PBMC 군을 배양한 배양 상층액에서 IFN-γ가 유의적으로 높게 검출되었다.
실험예 5: NK 세포 배양용 자성 입자의 이용에 따른 NK 세포 수용체의 발현 변화 평가
상기 실시예 1.1에서 제조한 NK 세포 배양용 자성 입자를 이용하는 경우 NK 세포 및 PBMC 매개 세포 독성의 증식 및 백분율이 증가하였기 때문에, NK 세포 수용체에는 어떠한 발현 변화가 나타나는지 분석하였다.
구체적으로, 상기 기재된 각 군별로 3 명의 공여자로부터 얻은 시료를 12일 동안 배양한 다음, 6 가지 종류의 수용체 분자에 대한 항체를 사용하여 동정하였다. 사용된 수용체 분자는 DNAM1, CD27, NKG2A, NKG2D, CD69 및 CD16이다. 사용된 실험군은 다음과 같다. (a) 용해성 IL-15; (b) 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자를 이용한 배양; (c) 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자를 이용한 배양.
도 10은 (a) 용해성 IL-15; (b) 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자를 이용한 배양; (c) 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자를 이용한 배양에서 PBMC 내 NK 세포 표면 상 수용체 발현을 분석한 overlay histogram이다.
또한, 상기 NK 세포 수용체 발현에 대한 데이터를 표로 작성하고 통계 처리하였다. 대응 표본(paired t-test)은 각 공여자의 NK 세포 수용체 발현율 데이터를 사용하여 수행하였다.
도 11은 (a) 용해성 IL-15; (b) 용해성 IL-15 및 4-1BBL_IgG1Fc가 부착된 자성 입자를 이용한 배양; (c) 용해성 IL-15 및 4-1BBL_IgG1Fc 및 IL-15Rα_IgG1Fc가 부착된 자성 입자를 이용한 배양에서 PBMC 내 NK 세포의 수용체 발현을 대응 표본(paired t-test)에 의한 통계적으로 분석한 결과이다. 데이터는 평균 ± 표준편차로 표시하였다. (*P<0.05, **P<0.005, ***P<0.001)
도 10 및 도 11에 나타낸 바와 같이, NK 세포 억제 수용체인 NKG2A는 용해성 IL-15 군(a)에서 약간 발현하였지만, 특정 분자가 부착된 일 양상에 따른 자성 입자를 이용한 군에서는 감소하였다. 반면, 활성화 수용체인 NKG2D, CD69 및 CD16은 일 양상에 따른 자성 입자를 이용한 군에서 발현 백분율이 증가한 것을 확인하였다.
따라서, 일 양상에 따른 자성 입자를 이용한 NK 세포 배양 시 상기 자성 입자를 이용한 배양에 의해 NK 세포에서 억제 수용체는 감소하고 수용체 활성화는 증가한 것을 확인하였다.
Claims (18)
- 적어도 일면에 활성화 수용체 리간드, 억제성 수용체 리간드, 보조자극 수용체 리간드, 사이토카인, 사이토카인 수용체, 면역관문 리간드, 블로킹 (blocking) 항체, 또는 이들의 조합이 부착된 자성 입자를 포함하는 자연 살해 세포(natural killer cell) 배양용 조성물.
- 청구항 1에 있어서, 상기 활성화 수용체 리간드가 NCR family 리간드, NKG2 family 리간드, KIR family 리간드, BAG6, AICL, MICA, MICB, CADM1, IgG, CD48, NTB-A/SLAMF6, CD70, CD155, CD319, C8, C9, 및 CS1로 구성된 군에서 선택된 1종 이상인 것인 조성물.
- 청구항 1에 있어서, 상기 억제성 수용체 리간드가 HLA-A, HLA-B, HLA-BW4, HLA-C1, HLA-C2, HLA-E, HLA-G, CD112/Nectin-2, CD112/Nectin-3, 카드헤린(cadherin), 콜라겐, OCIL, 및 CLEC2D로 구성된 군에서 선택된 1종 이상인 것인 조성물.
- 청구항 1에 있어서, 상기 보조자극 수용체 리간드가 TNF family 리간드, TLR family 리간드, 4-1BB 리간드, CD28 리간드, NTBA, TLRL, PVR/Nectin-2, 및 PVR로 구성된 군에서 선택된 1종 이상인 것인 조성물.
- 청구항 1에 있어서, 상기 사이토카인이 IFN-α, IFN-β, IFN-γ, IL-1, IL-2, IL-3, IL-4, IL-6, IL-10, IL-12, IL-15, IL-17, IL-18, IL-21, 및 IL-27로 구성된 군으로부터 선택된 1종 이상인 것인 조성물.
- 청구항 1에 있어서, 상기 사이토카인 수용체가 IL-2Rα, IL-15Rα, 또는 이들의 조합인 것인 조성물.
- 청구항 1에 있어서, 상기 면역관문 리간드가 B7 family, galectin family, PVR family, PD-L1, BTLA-4 리간드, CTLA-4 리간드 (CD80), Tim-3 리간드, 및 TIGIT 리간드로 구성된 군으로부터 선택된 1종 이상인 것인 조성물.
- 청구항 1에 있어서, 상기 블로킹 항체가 항-KIR2DL1 모노클로날 항체(mAb), 항-KIR2DL2 mAb, 항-KIR2DL3 mAb, 항-KIR2DL5A mAb, 항-KIR2DL5B mAb, 항-KIR3DL1 mAb, 항-KIR3DL2 mAb, 항-KIR2DL4 mAb, 항-CD94/NKG2A mAb, 항-CD94/NKG2B mAb, 항-CD96 mAb, 항-CEACAM-1 mAb, 항-ILT2/LILRB mAb, 항-KLRG1 mAb, 항-LAIR1 mAb, 항-NKRP1A mAb, 항-Siglec3 mAb, 항-Siglec7 mAb, 및 항-Siglec9 mAb로 구성된 군으로부터 선택된 1종 이상인 것인 조성물.
- 청구항 1에 있어서, 상기 자성 입자 표면의 적어도 일면은 단백질 G 또는 단백질 A로 코팅된 것인 조성물.
- 청구항 1에 있어서, 상기 활성화 수용체 리간드, 억제성 수용체 리간드, 보조자극 수용체 리간드, 사이토카인, 사이토카인 수용체, 면역관문 리간드, 및 블로킹 (blocking) 항체는 인간 면역글로불린과 융합된 형태인 것인 조성물.
- 청구항 10에 있어서, 상기 인간 면역글로불린은 인간 면역글로불린 G인 것인 조성물.
- 청구항 1에 있어서, 상기 자성 입자의 크기는 500nm 내지 10μm인 것인 조성물.
- 청구항 1에 있어서, 상기 배양은 자연 살해 세포의 증식 또는 활성화를 위한 것인 조성물.
- 청구항 1에 있어서, 상기 자연 살해 세포는 말초 혈액 단핵구(Peripheral blood mononuclear cell: PBMC)에 포함된 형태인 것인 조성물.
- 자연 살해 세포를 적어도 일면에 활성화 수용체 리간드, 억제성 수용체 리간드, 보조자극 수용체 리간드, 사이토카인, 사이토카인 수용체, 면역관문 리간드, 블로킹 (blocking) 항체, 또는 이들의 조합이 부착된 자성 입자를 포함하는 자연 살해 세포 배양용 조성물을 함유하는 배지에서 배양하는 단계;를 포함하는 자연 살해 세포를 배양하는 방법.
- 청구항 15에 있어서, 상기 배양하는 단계 전에, 말초 혈액 단핵구를 수득하는 단계;를 더 포함하는 것인 방법.
- 청구항 15에 있어서, 상기 배양하는 단계 후에, 상기 자성 입자를 배지로부터 제거하는 단계;를 더 포함하는 것인 방법.
- 청구항 15에 있어서, 상기 배양은 6일 내지 21일 동안 수행되는 것인 방법.
