Description
본 발명은 회전 반응기 내에서 비혼화성인 액상 동심층들의 적층 구조를 형성하는 반응 방법에 관한 것이다.
또한, 회전 반응기 내에서 형성되는 비혼화성인 액상 동심층들을
포함하는
반응 시스템에 관한 것이다.
선행 비특허문헌 1, 2 및 3은 비혼화성인 액상층들의 적층 구조에 대하여 개시하였으나, 상기 종래의 적층 구조에서는 인접하지 않은 액상층들끼리 합쳐질 수 있는 등 안정성이 충분하지 못한 문제가 있다.
한편, 선행 비특허문헌 4, 5 및 6은 다단계 유기합성 반응 같은 다단계 반응을 위한 종래의 반응 시스템에 관한 것이나, 시간 및 공간적 측면에서 모두 효율적으로 진행함에는 어려움이 있으며 또한 상당히 정교한 조작도 필요한 문제가 있다.

본 발명은 회전 반응기 내에서 비혼화성인 액상층들의 안정된 적층 구조를 형성하는 것을 통하여, 시간 및 공간적 측면에서 다단계 반응을 효율적으로 진행할 수 있는 기반을 제공하고자 한다.
상기 목적 달성을 위하여, 본 발명은 원통형 반응기; 및 상기 원통형 반응기를 회전시키는 구동부;를 포함하는 회전 반응기를 이용한 반응 방법으로서,
회전 중인 상기 원통형 반응기의 내부에 제1액상 물질부터 제n액상 물질(단, n은 2이상의 정수)까지 순차적으로 주입함에 따라, 상기 원통형 반응기가 회전하는 원심력에 의하여 상기 원통형 반응기의 내측면에 인접하거나 가까운 순으로 제1액상 동심층(concentric layer)부터 제m액상 동심층(단, m은 2이상의 정수)까지 순차적으로 인접되게 형성하는 단계를 포함하고,
상기 액상 물질은 n이 커질수록 밀도가 작아지고,
상기 액상 동심층은 m이 커질수록 밀도가 작아지고,
n-m ≥ 0의 관계가 성립되고,
순차적으로 주입되는 상기 제1액상 물질과 제2액상 물질은 비혼화성(immiscible)이며, n이 3이상인 경우 제3액상 물질부터는 이전 순서로 주입된 액상 물질과 혼화성(miscible) 또는 비혼화성(immiscible)이고,
상기 형성된 액상 동심층들에 있어서, 서로 인접하는 액상 동심층들은 비혼화성인 반응 방법을 제공한다.
상기 각각의 액상 물질은 독립적으로 하나 이상의 용매를 포함할 수 있으며, 또는 하나 이상의 용매와 하나 이상의 반응물질을 포함할 수 있다.
상기 제1액상 물질부터 제n액상 물질에 있어 적어도 하나 이상의 수용액이 사용될 수 있으며, 상기 제1액상 동심층부터 제m액상 동심층에 있어 적어도 하나 이상의 수용액 동심층이 구비될 수 있다.
상기 원통형 반응기는 60 내지 200 mm의 직경(diameter) 및 5 내지 55 mm의 높이(height)를 갖을 수 있다.
상기 원통형 반응기의 회전 속도는 800 rpm 이상일 수 있다.
상기 각각의 액상 동심층의 두께는 150 μm 이상일 수 있다.
상기 원통형 반응기의 내부의 바닥면의 중심부 상에 맨드릴(mandrel)이 구비될 수 있다.
상기 맨드릴의 주입구에 연결되고 상기 원통형 반응기의 바닥면의 하부에 임베드된(embedded) 주입 채널들을 통하여, 상기 액상 물질들이 상기 원통형 반응기의 내부에 주입될 수 있다.
상기 맨드릴의 주입구에 연결되고 상기 원통형 반응기의 바닥면의 하부에 임베드된 주요 주입 채널과 상기 주요 주입 채널에서 유래하는 2 이상의 가지 주입 채널들을 통하여, 상기 액상 물질들이 상기 원통형 반응기의 내부에 주입될 수 있다.
상기 반응 방법은 상기 서로 인접하는 액상 동심층들 사이에 상 전이(phase transfer)가 일어나는 단계를 추가로 포함할 수 있다.
본 발명은 또한 회전 반응기 내에서 형성되는 액상 동심층들을
포함하는
반응 시스템으로서,
회전하는 원통형 반응기;
상기 원통형 반응기를 회전시키는 구동부; 및
상기 원통형 반응기의 내측면에 인접하거나 가까운 순으로 순차적으로 인접되게 형성된 제1액상 동심층(concentric layer)부터 제m액상 동심층을 포함하고;
회전 중인 상기 원통형 반응기의 내부에 제1액상 물질부터 제n액상 물질(단, n은 2이상의 정수)까지 순차적으로 주입함에 따라, 상기 원통형 반응기가 회전하는 원심력에 의하여 상기 액상 동심층들이 순차적으로 인접되게 형성되고;
상기 액상 물질은 n이 커질수록 밀도가 작아지고;
상기 액상 동심층은 m이 커질수록 밀도가 작아지고;
n-m ≥ 0의 관계가 성립되고;
순차적으로 주입되는 상기 제1액상 물질과 제2액상 물질은 비혼화성(immiscible)이며, n이 3이상인 경우 제3액상 물질부터는 이전 순서로 주입된 액상 물질과 혼화성(miscible) 또는 비혼화성(immiscible)이고;
상기 형성된 액상 동심층들에 있어서, 서로 인접하는 액상 동심층들은 비혼화성인 반응 시스템을 제공한다.
회전 반응기를 이용하는 본 발명의 반응 방법 및 반응 시스템은 비혼화성인 액상 동심층들의 적층 구조를 형성하며, 액상 동심층들의 전체적 적층 구조를 손상시키지 않으면서 특정 동심층에 추출 및 혼화성 물질 추가 등의 변형을 가할 수 있다.
또한, 본 발명의 반응 방법 및 반응 시스템은 서로 인접하는 액상 동심층들 사이에서 상 전이가 일어나는 것도 가능하게 하므로, 다단계 유기합성 반응 같은 다단계 반응을 시간 및 공간적 측면에서 효율적으로 진행할 수 있는 장점도 있다.

본 발명의 비제한적인 실시예들에 따른 도 1-(a), 도 2-(a) 및 도 3에서와 같이, 본 발명의 반응 방법에 이용되는 회전 반응기(10)는 원통형 반응기(12); 및 상기 원통형 반응기를 회전시키는 구동부(14);를 포함한다.
상기 원통형 반응기는 원통 형상의 재질로 이루어질 수 있으며, 상기 재질은 비제한적으로 SiN
x, SiO
2, TiO
2, 쿼츠(Quartz) 및 테플론(Teflon, polytetrafluoroethylene)으로 이루어진 군에서 선택된 하나 또는 둘 이상일 수 있다.
회전 중인 상기 원통형 반응기의 내부에 제1액상 물질부터 제n액상 물질(단, n은 2이상의 정수)까지 순차적으로 주입함에 있어서, 상기 액상 물질은 n이 커질수록 밀도가 작아지고, n은 바람직하게 2 내지 20의 정수일 수 있다. 도 1-(a), 도 2-(a) 및 도 3의 도시에서와 같이, 원통형 반응기 마개(11)의 중심에 위치한 주입부를 통하여 상기 각각의 액상물질을 원통형 반응기의 내부에 주입할 수 있으며, 비제한적으로 주사기(syringe)를 사용하여 상기 각각의 액상물질을 원통형 반응기 내부의 바닥면 상에 떨어뜨릴 수 있다(도 3).
상기 원통형 반응기 마개의 재질은 비제한적으로 SiN
x, SiO
2, TiO
2 및 쿼츠(Quartz)로 이루어진 군에서 선택된 하나 또는 둘 이상일 수 있다.
회전 중인 원통형 반응기의 내부에 순차적으로 주입되는 상기 제1액상 물질과 제2액상 물질은 비혼화성이며, n이 3이상인 경우 제3액상 물질부터는 이전 순서로 주입된 액상 물질과 혼화성 또는 비혼화성이다. 비제한적인 일례로, 제1액상 물질부터 제3액상 물질까지 순차적으로 원통형 반응기의 내부에 주입하는 경우(밀도 크기: 제1액상 물질 > 제2액상 물질 > 제3액상 물질), 제1액상 물질과 제2액상 물질은 비혼화성이며, 제2액상 물질과 제3액상 물질은 혼화성 또는 비혼화성이다. 비제한적인 다른 일례로, 제1액상 물질부터 제4액상 물질까지 순차적으로 원통형 반응기의 내부에 주입하는 경우(밀도 크기: 제1액상 물질 > 제2액상 물질 > 제3액상 물질 > 제4액상 물질), 제1액상 물질과 제2액상 물질은 비혼화성이며, 제2액상 물질과 제3액상 물질은 혼화성 또는 비혼화성이며, 제3액상 물질과 제4액상 물질은 혼화성 또는 비혼화성이다.

회전 중인 상기 원통형 반응기의 내부에 제1액상 물질부터 제n액상 물질까지 순차적으로 주입하면, 하나의 액상 물질 또는 순차적으로 주입되는 둘 이상의 혼화성 액상 물질들이 하나의 액상 동심층을 형성하게 된다. 본 발명의 비제한적인 일 실시예에 관한 도 1-(a) 및 1-(b)의 도시에서와 같이, 제1액상 물질부터 제3액상 물질까지 순차적으로 원통형 반응기의 내부에 주입하는 경우(밀도 크기: 제1액상 물질 > 제2액상 물질 > 제3액상 물질, 제1액상 물질과 제2액상 물질은 비혼화성, 제2액상 물질과 제3액상 물질은 비혼화성), 첫 번째로 주입되는 제1액상 물질은 원통형 반응기의 내측면(12)에 인접한 제1액상 동심층(1)을 형성하며, 두 번째로 주입되는 제2액상 물질은 제1액상 동심층에 인접한 제2액상 동심층(2)을 형성하며, 마지막으로 주입되는 제3액상 물질은 제2액상 동심층에 인접하면서 반응기의 중심부에 가장 가까운 제3액상 동심층(3)을 형성하게 된다. 본 발명의 비제한적인 또 다른 실시예에 따른 도 2-(a) 및 도 2-(b)의 도시에서와 같이, 제1액상 물질부터 제5액상 물질까지 순차적으로 원통형 반응기의 내부에 주입하는 경우(밀도 크기: 제1액상 물질 > 제2액상 물질 > 제3액상 물질 > 제4액상 물질 > 제5액상 물질, 제1액상 물질과 제2액상 물질은 비혼화성, 제2액상 물질과 제3액상 물질은 비혼화성, 제3액상 물질과 제4액상 물질은 비혼화성, 제4액상 물질과 제5액상 물질은 비혼화성), 첫 번째로 주입되는 제1액상 물질은 원통형 반응기의 내측면(12)에 인접한 제1액상 동심층(1)을 형성하며, 두 번째로 주입되는 제2액상 물질은 제1액상 동심층에 인접한 제2액상 동심층(2)을 형성하며, 세 번째로 주입되는 제3액상 물질은 제2액상 동심층에 인접한 제3액상 동심층(3)을 형성하며, 네 번째로 주입되는 제4액상 물질은 제3액상 동심층에 인접한 제4액상 동심층(4)을 형성하며, 마지막으로 주입되는 제5액상 물질은 제4액상 동심층에 인접하면서 반응기의 중심부에 가장 가까운 제5액상 동심층(5)을 형성하게 된다.
회전 중인 상기 원통형 반응기의 내부에 제1액상 물질부터 제n액상 물질까지 순차적으로 주입하는 과정에 의하여, 2개의 혼화성 액상 동심층들 사이에 비혼화성 액상 동심층을 형성함으로서, 상기 2개의 혼화성 액상 동심층들이 섞이는 것을 방지할 수 있다. 비제한적인 일례로, 밀도가 서로 다른 2개의 수용액 동심층들 사이에 유기액상 동심층을 형성함으로서, 상기 2개의 수용액 동심층들을 서로 격리시킬 수 있다.
상기 제1액상 물질부터 제n액상 물질에 있어 바람직하게 적어도 하나 이상의 수용액이 사용될 수 있으며, 상기 제1액상 동심층부터 제m액상 동심층에 있어 바람직하게 적어도 하나 이상의 수용액 동심층이 구비될 수 있다. 비제한적인 일례로, 제1유기액상 물질, 수용액 및 제2유기액상 물질을 순차적으로 회전 중인 원통형 반응기의 내부에 주입하는 경우(밀도 크기: 제1유기액상 물질 > 수용액 >제2유기액상 물질), 원통형 반응기의 내측면에 인접한 순으로 제1유기액상 동심층, 수용액 동심층 및 제2유기액상 동심층이 순차적으로 인접되게 형성된다.
회전 중인 상기 원통형 반응기의 내부에 주입되는 액상 물질은 원통형 반응기의 바닥면을 향하여 수직 방향으로 층을 형성함으로서, 액상 동심층을 형성할 수 있다. 또한, 회전 중인 상기 원통형 반응기의 내부에 주입된 액상 물질에 있어서, 회전에 의한 구심력이 액상 물질의 표면 장력보다 우세하면, 동심원 모양의 액상층의 형성에 보다 유리하다. 따라서, 원통형 반응기의 회전 속도 및 액상 물질의 표면 장력에 대한 적절한 조절이 바람직하다.
회전 중인 상기 원통형 반응기의 내부에 액상 물질들을 순차적으로 주입하는 과정에서, 상기 원통형 반응기 내부의 중심부에 존재하는 기포(air bubble)에 의하여, 반응기의 중심부의 바닥면을 향하여 에어 칼럼(air column)이 서서히 형성될 수 있다. 원통형 반응기 내부의 중심부에 위치하는 상기 에어 칼럼에 의하여, 외부의 공기가 반응기 내부의 아래 방향으로 흐를 수 있는 칼럼이 확보될 수 있으며, 또한 회전하는 반응기 내부에 추가의 액상물질들을 순차적으로 주입할 수 있는 경로도 원활히 확보될 수 있다. 상기 에어 칼럼은 원통형 반응기의 중심부의 바닥면을 향하여 거의 수직 방향으로 형성되며, 반응기 부피에 대한 전체 액상 물질들의 부피 비율에 따라 상기 에어 칼럼의 부피 및 두께가 조절될 수 있다. 상기 에어 칼럼은 반응기 내부에 주입되는 액상 물질들 보다 작은 밀도를 갖으므로, 상기 에어 칼럼의 주위로 액상 물질들에 의한 액상 동심층들이 형성될 수 있다.
상기 원통형 반응기는 본 발명의 목적 달성 범위 내라면 크기에 있어 특별한 제한은 없으며, 예를 들면 60 내지 200 mm의 직경(diameter) 및 5 내지 55 mm의 높이(height)를 갖을 수 있다.
상기 원통형 반응기의 회전 속도는 바람직하게 800 rpm 이상이고, 보다 바람직하게 900 내지 10000 rpm이다. 상기 원통형 반응기의 회전 속도가 800 rpm 보다 낮으면, 주입되는 액상 물질이 원통형 반응기의 바닥면을 향하여 수직 방향으로 층을 형성하기 어려울 수 있으므로, 이에 따라 원활한 형태의 액상 동심층을 형성하기 어려울 수 있다.
회전 중인 상기 원통형 반응기의 내부의 각각의 액상 동심층의 두께는 바람직하게 150 μm 이상이고, 보다 바람직하게 1 내지 10 mm 이다. 액상 동심층의 두께가 150 μm 보다 작으면, 액상 동심층의 안정성이 준안정(metastable) 상태에 불과하게 되므로, 액상 동심층의 일 부분이 붕괴되거나 깨질 수 있다. 따라서, 액상 동심층이 150 μm 이상의 두께를 갖게 하기 위하여는, 주입되는 액상 물질의 밀도, 인접하는 액상 동심층과의 밀도 차 및/또는 액상 동심층의 표면 장력 뿐만 아니라 반응기의 크기 및 회전 속도에 대한 적절한 조절이 바람직하다. 예를 들면, 60 내지 200 mm의 직경 및 5 내지 55 mm의 높이를 갖는 원통형 반응기 내에서, 인접하는 액상 동심층과의 밀도 차를 0.1 g/mL 이상으로 하고 회전 속도를 800 rpm 이상으로 조절함으로서, 액상 동심층이 150 μm 이상의 두께를 갖게 할 수도 있다.
상기 구동부는 상기 원통형 반응기의 내부에 액상 동심층들을 형성하도록, 상기 원통형 반응기를 동축을 중심으로 회전시킨다. 상기 구동부는 일례로 전기 모터(electric motor)일 수 있다.
상기 원통형 반응기의 내부의 바닥면의 중심부 상에 맨드릴(mandrel)이 구비될 수 있다. 본 발명의 비제한적인 실시예들에 따른 도 1, 도 2 및 도 3에서와 같이, 본 발명의 반응 방법에 이용되는 회전 반응기(10)는 원통형 반응기(12); 및 상기 원통형 반응기를 회전시키는 구동부(14);를 포함하며, 또한 바닥면의 중심부 상에 맨드릴(13)도 포함할 수 있다.
상기 맨드릴의 재질은 비제한적으로 스테인리스강(stainless steel)일 수 있다.
상기 원통형 반응기가 맨드릴을 구비할 시, 상기 맨드릴의 주입구에 연결되고 상기 원통형 반응기의 바닥면의 하부에 임베드된(embedded) 주입 채널들을 통하여, 상기 액상 물질들이 상기 원통형 반응기의 내부에 주입될 수 있다. 본 발명의 비제한적인 실시예에 따르면, 원통형 반응기의 바닥면의 하부에 임베드된 각각의 주입 채널은 맨드릴의 주입구에 독립적으로 연결되고 원통형 반응기의 내부와도 독립적으로 연결된다. 이에 따라, 특정 액상 물질을 특정 주입 채널을 통하여, 반응기 내부의 특정 위치에 직접적으로 로딩(loading)할 수 있다. 도 4(제1주입 채널 및 제2주입 채널은 미도시)에 따른 비제한적인 일례에 따라, 제1액상 물질부터 제3액상 물질까지 순차적으로 원통형 반응기의 내부에 주입하는 경우(밀도 크기: 제1액상 물질 > 제2액상 물질 > 제3액상 물질, 제1액상 물질과 제2액상 물질은 비혼화성, 제2액상 물질과 제3액상 물질은 비혼화성), 제1액상 물질, 제2액상 물질 및 제3액상 물질을 각각 제1주입 채널, 제2주입 채널 및 제3주입 채널을 통하여 반응기 내부에 순차적으로 주입함에 따라, 제1액상 동심층(1), 제2액상 동심층(2) 및 제3액상 동심층(3)을 순차적으로 형성하게 된다. 구체적으로, 제3액상 물질은 제3주입 채널을 통하여 반응기 내부에 주입되고, 제2액상 동심층(2)에 인접한 위치에 로딩됨으로서 제3액상 동심층(3)을 형성한다.
상기 원통형 반응기가 맨드릴을 구비할 시, 상기 맨드릴의 주입구에 연결되고 상기 원통형 반응기의 바닥면의 하부에 임베드된 주요 주입 채널과 상기 주요 주입 채널에서 유래하는 2 이상의 가지 주입 채널들을 통하여, 상기 액상 물질들이 상기 원통형 반응기의 내부에 주입될 수도 있다. 본 발명의 비제한적인 실시예에 따른 도 5에서와 같이, 원통형 반응기의 바닥면의 하부에 임베드된 주요 주입 채널(15)로부터 2이상의 가지 주입 채널들(16)이 유래한다. 특정 액상 물질(이전 순서로 주입된 액상 물질들보다 밀도가 작음)이 맨드릴의 주입구를 통하여 주입되면, 주요 주입 채널의 채워지지 않은 부분을 통과해 흐르면서, 이전 순서로 주입된 액상 물질들에 의하여 사용되지 않은 첫 번째 가지 주입 채널을 통과하여 반응기 내부의 특정 위치에 주입된다. 도 5에 따른 비제한적인 일례에서는, 제1액상 물질부터 제4액상 물질까지 순차적으로 원통형 반응기의 내부에 주입된다(밀도 크기: 제1액상 물질 > 제2액상 물질 > 제3액상 물질 > 제4액상 물질, 제1액상 물질과 제2액상 물질은 비혼화성, 제2액상 물질과 제3액상 물질은 비혼화성, 제3액상 물질과 제4액상 물질은 비혼화성). 구체적으로, 주요 주입 채널에서 제4액상 물질은 이전의 3개의 액상 물질들에 의하여 채워지지 않은 부분을 통과해 흐르면서, 이전의 3개의 액상 물질들에 의하여 사용되지 않은 첫 번째 가지 주입 채널을 통과하여, 반응기 내부에서 제3액상 동심층(3)에 인접한 위치에 주입됨으로서 제4액상 동심층(4)을 형성한다.
본 발명의 도 4에서와 같이, 원통형 반응기의 바닥면의 하부에 임베드된 각각의 주입 채널이 맨드릴(13)의 주입구에 독립적으로 연결되고 원통형 반응기의 내부와도 독립적으로 연결되어 있으면, 특정 주입 채널을 통하여 특정 액상 동심층에 포함된 일부 물질을 추출하는 것도 가능하다. 예를 들면, 본 발명의 비제한적인 실시예에 따른 도 6에서와 같이 이미 형성된 5개의 액상 동심층들에 있어서, 특정한 제3주입 채널을 이용하여 제3액상 동심층(3)에 포함된 일부 물질을 추출할 수 있다. 또한, 상기와 같이 특정 동심층에 추출의 변형을 가하여도, 액상 동심층들의 전체적 적층 구조가 손상되지는 않는다.
본 발명의 도 4에서와 같이, 원통형 반응기의 바닥면의 하부에 임베드된 각각의 주입 채널이 맨드릴(13)의 주입구에 독립적으로 연결되고 원통형 반응기의 내부와도 독립적으로 연결되어 있으면, 특정 주입 채널을 통하여 특정 액상 동심층에 혼화성인 특정 물질을 첨가하는 것도 가능하다. 예를 들면, 본 발명의 비제한적인 실시예에 따른 도 7에서와 같이 이미 형성된 5개의 액상 동심층들에 있어서, 특정한 제3주입 채널을 이용하여 제3액상 동심층(3)에 혼화성인 추가의 제6액상 물질을 첨가할 수 있다. 또한, 상기와 같이 특정 동심층에 혼화성 물질 추가의 변형을 가하여도, 액상 동심층들의 전체적 적층 구조가 손상되지는 않는다.
본 발명의 반응 방법은 상기 서로 인접하는 액상 동심층들 사이에 상 전이(phase transfer)가 일어나는 단계를 추가로 포함할 수 있다. 상기 액상 물질들 중 적어도 하나 이상의 액상 물질이 하나 이상의 용매와 하나 이상의 반응물질을 포함하는 경우, 상기 상 전이가 일어나는 데 보다 유리할 수 있다. 구체적으로, 특정 액상 동심층에서의 반응 생성물이 확산(diffusion)에 의하여 인접한 액상 동심층 내로 전이될 수 있으며, 이러한 확산 전이에 의하여도 액상 동심층들의 전체적 적층 구조가 파괴되지는 않는다. 비제한적인 일례로, 특정 유기액상 동심층에서의 반응 생성물이 수용성을 갖는 경우, 상기 수용성 반응 생성물은 확산에 의하여 인접한 수용액 동심층으로 전이될 수 있다. 또한, 순차적으로 인접되게 형성된 액상 동심층들 사이에서 상 전이가 순차적으로 진행될 수도 있으며, 적절한 다단계 유기 반응 및 무기 반응을 본 발명의 반응 방법에 적용함으로서 상기 순차적 상 전이를 구체적으로 확인할 수 있다.
또한, 본 발명의 반응 방법에 의하여 생성된 액상 동심층들에 있어서, 반응기의 내측면에 인접하거나 가장 가까운 액상 동심층 자체가 추출되어도 나머지 액상 동심층들에 의한 적층 구조가 손상되거나 파괴되지 않는다.
나아가, 본 발명의 반응 방법에 의하여, 회전 반응기 내에서 형성되는 액상 동심층들을
포함하는
반응 시스템이 형성된다.
상기 반응 시스템은,
회전하는 원통형 반응기;
상기 원통형 반응기를 회전시키는 구동부; 및
상기 원통형 반응기의 내측면에 인접하거나 가까운 순으로 순차적으로 인접되게 형성된 제1액상 동심층(concentric layer)부터 제m액상 동심층을 포함하고;
회전 중인 상기 원통형 반응기의 내부에 제1액상 물질부터 제n액상 물질(단, n은 2이상의 정수)까지 순차적으로 주입함에 따라, 상기 원통형 반응기가 회전하는 원심력에 의하여 상기 액상 동심층들이 순차적으로 인접되게 형성되고;
상기 액상 물질은 n이 커질수록 밀도가 작아지고;
상기 액상 동심층은 m이 커질수록 밀도가 작아지고;
n-m ≥ 0의 관계가 성립되고;
순차적으로 주입되는 상기 제1액상 물질과 제2액상 물질은 비혼화성(immiscible)이며, n이 3이상인 경우 제3액상 물질부터는 이전 순서로 주입된 액상 물질과 혼화성(miscible) 또는 비혼화성(immiscible)이고;
상기 형성된 액상 동심층들에 있어서, 서로 인접하는 액상 동심층들은 비혼화성인 반응 시스템이다.
실시예
이하, 본 발명을 하기의 실시예에 의하여 더욱 상세히 설명한다.
단, 하기의 실시예는 본 발명을 예시할 뿐, 본 발명의 내용이 하기의 실시예에 의해 한정되는 것은 아니다.
<실시예 1>
도 1의 형태의 반응기를 사용하였으며, 원통형 반응기(직경: 73.5 mm, 높이: 48 mm)를 회전시키기 위한 구동부로 전기 모터를 사용하였다. 상기 원통형 반응기의 내부의 측면 및 바닥면의 재질로 테플론(Teflon, polytetrafluoroethylene)이 사용되었으며, 쿼츠(Quartz) 재질의 원통형 반응기 마개는 증기 증착된 1H,1H,2H,2H-퍼플루오로옥틸트리클로로실란(vapor-deposited 1H,1H,2H,2H-perfluorooctyltrichlorosilane)으로 실란화(silanized) 처리되었다. 또한, 스테인리스강(stainless steel) 재질의 맨드릴이 사용되었다.
표 1에 기재된 하기 3개의 액상 물질들이 사용되었다.

회전 중인 원통형 반응기의 내부에 제1액상 물질부터 제3액상 물질까지 순차적으로 주입하였으며, 회전 속도의 변화에 따라 액상 동심층들의 형성 여부를 관찰하였다. 이때, 형성될 액상 동심층들의 식별을 위하여, 제1액상 물질은 투명하게 유지되었으며, 제2액상 물질은 fluorescein sodium salt에 의하여 노란색으로 염색된 상태이었으며, 제3액상 물질은 Oil Red EGN에 의하여 붉은색으로 염색된 상태이었다. 회전 속도가 100 rpm 및 300 rpm의 경우, 주입되는 액상 물질들이 원통형 반응기의 바닥면을 향하여 수직 방향으로 층을 형성하지 못하여, 원활한 형태의 액상 동심층들이 형성될 수 없었다.
그러나, 회전 속도가 2600 rpm의 경우, 원통형 반응기의 내측면에 인접한 순으로 제1액상 동심층(제1액상 물질로 형성), 제2액상 동심층(제2액상 물질로 형성) 및 제3액상 동심층(제3액상 물질로 형성)이 원활하게 형성된 것을 확인하였다. 이때, 원통형 반응기 내부의 중심부에 존재하는 기포의 부피는 약 0.66 mL로 관찰되었다.
<실시예 2>
도 1의 형태의 반응기를 사용하였으며, 원통형 반응기(직경: 120 mm, 높이: 8 mm)를 회전시키기 위한 구동부로 전기 모터를 사용하였다. 상기 원통형 반응기의 내부의 측면 및 바닥면의 재질로 테플론(Teflon, polytetrafluoroethylene)이 사용되었으며, 쿼츠(Quartz) 재질의 원통형 반응기 마개는 증기 증착된 1H,1H,2H,2H-퍼플루오로옥틸트리클로로실란(vapor-deposited 1H,1H,2H,2H-perfluorooctyltrichlorosilane)으로 실란화(silanized) 처리되었다. 또한, 스테인리스강(stainless steel) 재질의 맨드릴이 사용되었다.
표 2에 기재된 하기 4개의 액상 물질들이 사용되었다(밀도 크기: 제1액상 물질 > 제2액상 물질 > 제3'액상 물질 > 제3''액상 물질).

1000 rpm의 회전 속도로 회전하는 원통형 반응기에, 제1액상 물질, 제2액상 물질, 제3'액상 물질 및 제3''액상 물질을 순차적으로 주입하였다. 제3'액상 물질 및 제3''액상 물질을 주입한 즉시, 제3'액상 물질 및 제3''액상 물질에 의한 액상 동심층들(제3'액상 동심층, 제3''액상 동심층)이 혼합되어 하나의 동심층(제3액상 동심층)으로 합쳐졌다. 이에 따라, 반응기 내부에 3개의 액상 동심층들(제1액상 동심층: DCM 층; 제2액상 동심층: 물 층; 제3액상 동심층: 톨루엔 층)이 최종적으로 형성되었으며(도 8), 각각의 액상 동심층의 두께는 1.5 mm이었다. 회전하는 반응기의 중심부에서 가장 가까운 제3액상 동심층에서, 트리페닐포스핀과 메틸 브로모아세테이트의 반응에 의하여 흰색 침전이 형성되었으며, 이는 수용성 물질인 포스포늄 염(phosphonium salt)의 형성을 의미하였다. 상기 수용성 포스포늄 염은 제2액상 동심층(물 층)으로 서서히 전이되었으며, 상기 포스포늄 염과 탄산칼륨과의 반응에 의하여 비수용성인 일라이드(ylide)가 형성되었다. 상기 일라이드는 제1액상 동심층으로 전이되었으며, 상기 일라이드와 테레프타랄데하이드의 위티그 반응(wittig reaction)이 72 시간 동안 상온에서 진행되었다. 이후 반응을 종료하고, 반응기의 반응 생성물들을 분리기(separator)로 옮겨 유기상을 분리하고, 분리된 유기상을 건조 및 정제하였다. 최종적으로, 목표 화합물인 디에스터 화합물(diester product, P
2)이 85%의 수율로 얻어진 것을 확인하였으며, 부산물인 모노에스터 화합물(monoester product, P
1)의 수율은 6%에 불과함을 확인하였다(도 8).
따라서, 공지의 다단계 유기 반응을 본 발명의 반응 방법 및 반응 시스템에 적용함으로서, 하나의 반응기 내에서의 상 전이 과정에 의하여 다단계 반응을 시간 및 공간적 측면에서 효율적으로 진행할 수 있었다.
<실시예 3>
도 1의 형태의 반응기를 사용하였으며, 원통형 반응기(직경: 120 mm, 높이: 8 mm)를 회전시키기 위한 구동부로 전기 모터를 사용하였다. 상기 원통형 반응기의 내부의 측면 및 바닥면의 재질로 테플론(Teflon, polytetrafluoroethylene)이 사용되었으며, 쿼츠(Quartz) 재질의 원통형 반응기 마개는 증기 증착된 1H,1H,2H,2H-퍼플루오로옥틸트리클로로실란(vapor-deposited 1H,1H,2H,2H-perfluorooctyltrichlorosilane)으로 실란화(silanized) 처리되었다. 또한, 스테인리스강(stainless steel) 재질의 맨드릴이 사용되었다.
표 3에 기재된 하기 3개의 액상 물질들이 사용되었다(밀도 크기: 제1액상 물질 > 제2액상 물질 > 제3액상 물질).

1000 rpm의 회전 속도로 회전하는 원통형 반응기에, 제1액상 물질, 제2액상 물질 및 제3액상 물질을 순차적으로 주입하였다. 이에 따라, 반응기 내부에 3개의 액상 동심층들(제1액상 동심층: TCE 층; 제2액상 동심층: 물 층; 제3액상 동심층: AcOEt 층)이 형성되었으며(도 9), 각각의 액상 동심층의 두께는 2 mm이었다. 제3액상 동심층에서 아미노페놀과 무수초산(Ac
2O)의 N-아실레이션(N-acylation) 반응을 적절히 진행시키기 위하여, 회전하는 반응기의 온도가 일단 2시간 동안 상온으로 유지되었다. 이후, 반응기의 온도가 40
oC로 승온되어 22시간 동안 유지된 후, 상기 N-아실레이션 반응 생성물은 제2액상 동심층(물 층)으로 전이되었다. 상기 N-아실레이션 반응 생성물과 탄산칼륨의 반응에 의하여, 상기 N-아실레이션 반응 생성물의 페놀 수산기(phenolic hydroxyl)가 탈양성자화(deprotonated) 되었다. 상기 탈양성자화된 반응 생성물은 테트라부틸암모늄 브로마이드(TBAB)의 매개하에 제1액상 동심층으로 전이되었으며, 상기 탈양성자화된 반응 생성물과 요오드화에틸의 반응에 의하여 페나세틴(phenacetin, N-(4-ethoxyphenyl)acetamide)이 제1액상 동심층에서 형성되었다(도 9). 이후 반응을 종료하고, 반응기의 반응 생성물들을 분리기(separator)로 옮겨 유기상을 분리하고, 분리된 유기상을 건조 및 정제하였다. 최종적으로, 목표 화합물인 페나세틴이 37%의 수율로 얻어진 것을 확인하였다.
따라서, 통증 완화 및 발열 완화 약물로 알려진 페나세틴 역시 본 발명의 반응 방법 및 반응 시스템에 의하여 시간 및 공간적 측면에서 효율적으로 합성될 수 있었다.

- 원통형 반응기; 및 상기 원통형 반응기를 회전시키는 구동부;를 포함하는 회전 반응기를 이용한 반응 방법으로서,회전 중인 상기 원통형 반응기의 내부에 제1액상 물질부터 제n액상 물질(단, n은 2이상의 정수)까지 순차적으로 주입함에 따라, 상기 원통형 반응기가 회전하는 원심력에 의하여 상기 원통형 반응기의 내측면에 인접하거나 가까운 순으로 제1액상 동심층(concentric layer)부터 제m액상 동심층(단, m은 2이상의 정수)까지 순차적으로 인접되게 형성하는 단계를 포함하고,상기 액상 물질은 n이 커질수록 밀도가 작아지고,상기 액상 동심층은 m이 커질수록 밀도가 작아지고,n-m ≥ 0의 관계가 성립되고,순차적으로 주입되는 상기 제1액상 물질과 제2액상 물질은 비혼화성(immiscible)이며, n이 3이상인 경우 제3액상 물질부터는 이전 순서로 주입된 액상 물질과 혼화성(miscible) 또는 비혼화성(immiscible)이고,상기 형성된 액상 동심층들에 있어서, 서로 인접하는 액상 동심층들은 비혼화성인 반응 방법.

- 제7항에 있어서,상기 맨드릴의 주입구에 연결되고 상기 원통형 반응기의 바닥면의 하부에 임베드된(embedded) 주입 채널들을 통하여, 상기 액상 물질들이 상기 원통형 반응기의 내부에 주입되는 반응 방법.
- 제7항에 있어서,상기 맨드릴의 주입구에 연결되고 상기 원통형 반응기의 바닥면의 하부에 임베드된 주요 주입 채널과 상기 주요 주입 채널에서 유래하는 2 이상의 가지 주입 채널들을 통하여, 상기 액상 물질들이 상기 원통형 반응기의 내부에 주입되는 반응 방법.
- 제1항에 있어서,상기 서로 인접하는 액상 동심층들 사이에 상 전이(phase transfer)가 일어나는 단계를 추가로 포함하는 반응 방법.
- 회전 반응기 내에서 형성되는 액상 동심층들을 포함하는 반응 시스템으로서,회전하는 원통형 반응기;상기 원통형 반응기를 회전시키는 구동부; 및상기 원통형 반응기의 내측면에 인접하거나 가까운 순으로 순차적으로 인접되게 형성된 제1액상 동심층(concentric layer)부터 제m액상 동심층을 포함하고;회전 중인 상기 원통형 반응기의 내부에 제1액상 물질부터 제n액상 물질(단, n은 2이상의 정수)까지 순차적으로 주입함에 따라, 상기 원통형 반응기가 회전하는 원심력에 의하여 상기 액상 동심층들이 순차적으로 인접되게 형성되고;상기 액상 물질은 n이 커질수록 밀도가 작아지고;상기 액상 동심층은 m이 커질수록 밀도가 작아지고;n-m ≥ 0의 관계가 성립되고;순차적으로 주입되는 상기 제1액상 물질과 제2액상 물질은 비혼화성(immiscible)이며, n이 3이상인 경우 제3액상 물질부터는 이전 순서로 주입된 액상 물질과 혼화성(miscible) 또는 비혼화성(immiscible)이고;상기 형성된 액상 동심층들에 있어서, 서로 인접하는 액상 동심층들은 비혼화성인 반응 시스템.
