Abstract
双载体以及利用synNotch技术提供一种靶抗原依赖性调控细胞因子的表达,从而局部释放细胞因子增强T细胞的功能,同时也降低细胞因子的非特异性的毒副作用。利用一种合成的notch受体,构建质粒载体来实现细胞因子的局部分泌。其中一个质粒载体以特异性抗体为胞外段来识别靶抗原,notch核心包括跨膜区和notch受体特异性的酶切位点,GAL4VP64转录因子作为胞内段。当识别到靶抗原时,notch上酶切位点会被相应的酶识别水解切割胞内转录因子GAL4VP64。另外一个质粒载体带有GAL4转录因子的DNA识别结合区GAL4UAS以及相应启动目的基因的最小启动子,当GAL4VP64被水解切割后会结合到结合区从而启动后续基因的转录,表达IL12并分泌到细胞外。

本申请要求申请日为2018年1月19日的中国专利申请CN201810053219.6的优先权。本申请引用上述中国专利申请的全文。
本发明属于免疫治疗领域,具体涉及一种质粒载体对以及靶抗原依赖性调控IL12的表达的免疫细胞。
IL12是一种具有多种生物学活性的免疫细胞生长刺激因子,也是一种异源二聚体细胞因子,能促进T辅助细胞1(Th1)的增殖;诱导NK细胞和T细胞产生γ干扰素;提高NK细胞的细胞毒性作用;促进细胞毒性T细胞的形成。目前直接加载IL12以增强嵌合抗原受体(chimeric antigen receptor,CAR)抗肿瘤效力,但这种加载方式会使得IL12随着CAR-T全身性分泌,还容易造成IL12的表达量随着T细胞的扩增而不受控制,从而给机体带来严重的毒副作用。
美国专利US9,834,608B2(该专利以其全文在此引作参考)公开了一种结合触发转录开关多肽及其编码核酸和宿主细胞。该多肽为嵌合Notch受体多肽,包括胞外域、Notch调节区以及胞内域,其中胞外域含有特异性结合对的第一成员,该第一成员与特异性结合对的第二成员特异性结合,Notch调节区含有Lin 12-Notch重复单元、S2蛋白水解切割位点和含S3蛋白水解切割位点的跨膜域,以及其中特异性结合对的第一成员与第二成员结合诱导S2和S3蛋白水解切割位点的切割,从而释放胞内域。第一成员选自抗体、基于抗体的识别支架、基于非抗体的识别支架、抗原、受体的配体、受体、基于非抗体的识别支架的靶、胞外基质组分和粘附分子。胞内域含转录激活因子,而胞内域的释放引起转录激活因子诱导内源基因在细胞中的表达。或者,胞内域含转录阻遏因子,胞内域的释放引起转录阻遏因子抑制内源基因在细胞中的表达。
然而,该美国专利没有提及在免疫细胞中控制表达和局部分泌IL-12的必要性及其有益效果。
发明内容
为解决现有技术缺乏控制表达和局部分泌IL-12的免疫细胞,本发明提供一种质粒载体对以及利用synNotch技术提供一种靶抗原依赖性调控IL12的表达的免疫细胞。具体地,本发明在NK92细胞中利用一种合成的Notch(synNotch)受体为核心,构建两个质粒载体来实现IL12的局部分泌。其中一个质粒载体以antiGPC3 scfv(SEQ ID NO:2)为胞外段来识别靶抗原GPC3;notch核心(notch core,SEQ ID NO:3)包括跨膜区,并包含两个酶切位点;GAL4VP64(SEQ ID NO:4)转录因子作为胞内段。当识别到靶抗原GPC3时,notch核心上的酶切位点会被相应的酶识别水解切割并释放胞内转录因子GAL4VP64。另一个质粒载体带有GAL4转录因子的DNA识别结合区GAL4UAS(SEQ ID NO:5)以及相应启动目的基因IL12的minimal CMV启动子(SEQ ID NO:6),当GAL4VP64被水解切割后会结合到结合区GAL4UAS从而启动后续基因的转录,表达IL12并分泌到细胞外。
本发明减少毒性产生,只有当接触到特定的靶抗原时才会诱导IL12的分泌表达,从而在肿瘤附近局部释放IL12并增强CAR-T效果。并且,所使用的NK92细胞系在体内基本不扩增,从而避免了T细胞在体内扩增导致被诱导表达的IL12量不受控制所带来的严重毒副作用。另外,相比NK细胞,NK92可以批量生产,更适合于产业化应用。
一方面,本发明涉及质粒载体对,其包括第一载体和第二载体,所述第一载体包含Notch核心,还包括其他胞外段和胞内段的编码核酸分子,所述胞内段优选地含有转录激活因子;所述第二载体含有与所述第一载体中的胞内段特异性识别和结合的核酸分子以及编码IL12的核酸分子。
在一较佳的具体实施例中,所述第一载体的Notch核心具有SEQ ID NO:3所示的核酸序列。
根据本发明,第一载体的胞内段含有转录激动因子(transcriptional activator),例如四环素控制的转录激动因子(tTa)、GAL4VP64和ZFHD1-VP64,第二载体含有与上述转录激动因子相对应的DNA结合区,例如,Tet、UAS和ZFRE。优选地,第一载体的胞内段含有GAL4VP64,而第二载体含有GAL4UAS。第一载体的胞内段为DNA结合多肽,例如Zip(-)Gal4DNA结合多肽,或者NLS VP64Zip(+)。
本发明第一载体中的胞外段没有特别限定,只要能与目标对象特异性结合配对即可,例如可以含有抗原或该抗原的抗体或其片段、受体或该受体的配体、基于非抗体的识别支架或其靶、粘附分子或其胞外基质、Fc或其受体、受体或其共受体等。
本发明的抗体与抗原特异性结合,包括纳米抗体,单链抗体,二价抗体,三价抗体或 微型抗体。抗体片段是指完整抗体的一部分,例如完整抗体的抗原结合区或可变区。抗体片段包括Fab,Fab’,F(ab’)2以及Fv片段。Fv为最小的抗体片段,其含有完整的抗原识别和结合位点。单链Fv(scFv)是指抗体的VH和VL结构域,这些结构域呈现单链多肽形式。在有些实施方式中,Fv多肽还包括VH和VL之间的接头。胞外段优选含有抗体,更优选含有scFv。
本发明的抗体或其片段特异性针对抗原上的表位,所述抗原尤其是癌细胞相关抗原,包括乳腺癌,B细胞淋巴癌,前列腺癌,霍奇金淋巴瘤,卵巢癌,肺癌(例如小细胞肺癌),间皮瘤,黑素瘤,急慢性淋巴性白血病,成神经细胞瘤,胶质瘤,成胶质细胞瘤,成神经管细胞瘤,结肠癌,胃癌,肝癌等。
根据本发明,所述肿瘤抗原包括前列腺特异性膜抗原(PSMA)、癌胚抗原(CEA)、IL13Ralpha、HER-2、CD19、NY-ESO-1、HIV-1Gag、Lewis Y、MART-1、gp100、酪氨酸酶、WT-I、hTERT、间皮素、EGFR、EGFRvIII、GPC3、EphA2、HER3、EpCAM、MUC1、MUC16、CLDN18.2、叶酸受体、CLDN6、CD30、CD138、ASGPR1、CDH16、GD2、5T4、8H9、αvβ6整合素、B细胞成熟抗原(BCMA)、B7-H3、B7-H6、CAIX、CA9、CD20、CD22、κ轻链(kappa light chain)、CD33、CD38、CD44、CD44v6、CD44v7/8、CD70、CD123、CD171、CSPG4、EGP2、EGP40、ERBB3、ERBB4、ErbB3/4、FAP、FAR、FBP、胚胎型AchR、GD2、GD3、HLA-AI MAGE A1、MAGE3、HLA-A2、IL11Ra、KDR、Lambda、MCSP、NCAM、NKG2D配体、PRAME、PSCA、PSC1、ROR1、Sp17、SURVIVIN、TAG72、TEM1、TEM8、VEGRR2、HMW-MAA、VEGF受体、和/或纤连蛋白、腱生蛋白或肿瘤坏死区的癌胚变体。
根据本发明,胞外段包括但不限于,例如anti-CD19scFv、anti-间皮素scFv、anti-GFP nanobody、anti-GPC3scFv等,优选anti-GPC3scFv(即抗GPC3scFv)。
根据本发明,Notch核心包括Notch跨膜区以及一个或多个切割位点。
根据本发明,Notch核心的胞外末端任选可添加一个或多个表皮细胞生长因子(Epidermal Growth Factor,EGF)重复单元。EGF重复单元可以减少基础本底的非特异激活。
根据本发明,第一载体和第二载体还任选包括信号序列(例如,SEQ ID NO:1)、接头、启动子、标签(tag)和报道子(reporter)等元件,其中标签包括血凝素(HA)、C-myc和Flag等中的一种或多种,报道子包括绿色荧光蛋白(GFP)、蓝色荧光蛋白(BFP)和mcherry等中的一种或多种。
根据本发明,所述第一载体依序包括PGK启动子-信号序列-myc标签–anti-GPC3scFv-notch core-GAL4-接头-VP64,所述第二载体依序包括GAL4UAS-CMV minimal启动子-IL12-PGK启动子-mcherry。
根据本发明,所述第一载体的开放阅读框含有如SEQ ID NO:21所示的核苷酸序列,所述第二载体的开放阅读框含有如SEQ ID NO:22所示的核苷酸序列。由上述核苷酸序列编码的氨基酸序列即本发明所述载体的元件和/或载体所编码的多肽。
根据本发明,一个或多个氨基酸的替换、缺失和/或添加,通常不会从实质上影响多肽变体的功能,尤其是在多肽的保守区已知的情况下。保守性替换(例如,一种碱性氨基酸替换另一种碱性氨基酸,一种酸性氨基酸替换另一种酸性氨基酸)对本领域技术人员而言也是容易想到的。因此,除本发明载体元件和/或载体所编码的多肽的氨基酸序列外,与本发明载体元件和/或载体所编码的多肽的氨基酸序列具有至少50%,优选70%、75%、80%、85%或90%,更优选95%、96%、97%、98%、99%的同一性的功能性变体在保护范围之内。另外,编码多肽的核苷酸密码子可以是简并的,因此与本发明载体元件和/或载体的核苷酸序列具有至少50%,优选70%、75%、80%、85%或90%,更优选95%、96%、97%、98%或99%的同一性的功能性变体同样在保护范围之内。进一步更优选地,anti-GPC3scFv具有SEQ ID NO:2所示的核苷酸序列具有至少90%的同一性。
另一方面,本发明还提供以本发明的质粒载体对修饰的免疫细胞。所述免疫细胞为T细胞或NK细胞。优选地,所述免疫细胞为NK细胞。所述细胞转染有上述任一项的质粒载体对。所述NK细胞为天然NK细胞或永生化的NK细胞。优选地,为永生化的NK细胞。更优选地,为NK92细胞。
又一方面,本发明提供以上任一项的质粒载体对或以上任一项的免疫细胞在制备用于局部分泌IL12或者调控表达IL12的药物中的应用。
再一方面,本发明提供以上任一项的质粒载体对或以上任一项的免疫细胞在制备用于增强CAR-T细胞杀伤作用的药物中的应用。
在一优选的具体实施例中,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与所述CAR-T细胞识别的肿瘤抗原相同。
在一优选的具体实施例中,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与所述CAR-T细胞识别的肿瘤抗原均表达在同一种肿瘤上。

本发明通过实验证明,分别与表达GPC3的细胞系共孵育,未感染的NK92细胞对高表达GPC3的细胞系IL12的分泌量均非常低,而双感染pHRLSIN-antiGPC3-synNotch-GAL4VP64和pHRLSIN-GAL4UAS-IL12-PGK-mcherry的NK92细胞的IL12分泌量显著性升高。
因此,选择针对GPC3的单链抗体能够在NK92细胞中调控表达细胞因子IL12,并能够局部释放IL12。
为了更清楚地描述本发明的技术方案,下面将结合附图作简要介绍。显而易见,这些附图仅是本申请记载的一些具体实施方式。本发明包括但不限于这些附图。

术语“受体”是一类存在于胞膜或胞内的,能与细胞外专一信号分子结合进而激活细胞内一系列生物化学反应,使细胞对外界刺激产生相应的效应的特殊蛋白质。与受体结合的生物活性物质统称为配体(ligand)。
术语“嵌合抗原受体”或“CAR”是指可以由包括但不限于T细胞的免疫细胞表达的工程化分子。CAR在T细胞中表达并且可以重定向T细胞以诱导以由人造受体决定的特异性杀死靶细胞。CAR的细胞外结合结构域可以衍生自鼠、人源化或完全人单克隆抗体。
嵌合抗原受体通常包含(细)胞外抗原结合区。在一些实施方案中,胞外抗原结合区可以是完全人的。在其他情况下,胞外抗原结合区域可以被人源化。在其他情况下,胞外抗原结合区可以是鼠源的,或者所述胞外抗原结合区中的嵌合体由来自至少两种不同动物的氨基酸序列组成。在一些实施方案中,所述胞外抗原结合区可以是非人的,也可以设计多种抗原结合区域。非限制性实例包括衍生自抗体的单链可变片段(scFv)、选自文库的片段抗原结合区(Fab)、单结构域片段或与接合其同源受体的自然配体。在一些实施方案中,胞外抗原结合区域可以包含scFv、Fab或天然配体,以及它们的任何衍生物。细胞外抗原结合区可以指除完整抗体之外的分子,其可以包含完整抗体的一部分并且可以与完整抗体所结合的抗原结合。抗体片段的实例可以包括但不限于Fv、Fab、Fab'、Fab'-SH、F(ab')2;双功能抗体、线性抗体;单链抗体分子(例如scFv);和由抗体片段形成的多特异性抗体。
术语“工程化”及其语法上的其他形式可以指核酸的一个或多个改变,例如生物体基因组内的核酸。术语“工程化”可以指基因的改变、添加和/或缺失。工程细胞还可以指含有加入、缺失和/或改变的基因的细胞。
术语“细胞”或“工程细胞”及其语法上的其他形式可以指人或非人动物来源的细胞。工程细胞也可以指表达CAR的细胞。
术语“转染”是指将外源核酸引入真核细胞。转染可以通过本领域已知的各种手段来实现,包括磷酸钙-DNA共沉淀、DEAE-葡聚糖介导的转染、聚凝胺介导的转染、电穿孔、显微注射、脂质体融合、脂质转染、原生质体融合、逆转录病毒感染和生物弹道技术(biolistics)。
如本文所用,术语“编码核酸分子”、“编码核酸序列”是指沿着脱氧核糖核酸链的脱氧核糖核苷酸的顺序或顺序,这些脱氧核糖核苷酸的顺序决定了沿着多肽(蛋白质)链的氨基酸的顺序。因此,核酸序列编码氨基酸序列。
术语“外周血淋巴细胞”及其语法上的其他形式可以指在血液(例如外周血)中循环的淋巴细胞。外周血淋巴细胞可以指不局限于器官的淋巴细胞。外周血淋巴细胞可以包含T细胞、NK细胞、B细胞或其任何组合。
术语“免疫细胞”指可以引发免疫应答的细胞,包括但不限于T细胞、B细胞、NK细胞、NKT细胞、DNT细胞等,它们各自的前体细胞及其后代。免疫应答细胞还可以指淋巴或骨髓谱系的细胞。
术语“免疫细胞”及其语法上的其他形式可以指任何来源的免疫细胞。例如,免疫细胞可以是来自血液的,如自体的T细胞、异体T细胞、自体NK细胞、异体NK细胞,也可以来源自细胞系,如利用EBV病毒感染来制备NK细胞系,从胚胎干细胞和iPSC诱导分化来的NK细胞以及NK92细胞系等。
术语“免疫细胞”也可以是人或非人的。
当用于指核苷酸序列时,术语“序列”及其语法上的其他形式可以包括DNA或RNA,并且可以是单链或双链。核酸序列可以突变。核酸序列可以具有任何长度。
术语“有效量”是指提供治疗或预防益处的量。
本文使用的术语“启动子”定义为由启动多核苷酸序列的特异性转录所需的细胞的合成机制或引入的合成机制识别的DNA序列。
本文使用的术语“载体”是包含分离的核酸并可用于将分离的核酸递送至细胞内部的组合物。在本领域中已知许多载体,包括但不限于线性多核苷酸、与离子或两亲化合 物相关的多核苷酸、质粒和病毒。因此,术语“载体”包括自主复制的质粒或病毒。该术语还应被解释为包括促进核酸转移到细胞中的非质粒和非病毒化合物,例如聚赖氨酸化合物、脂质体等。病毒载体的实例包括但不限于腺病毒载体、腺相关病毒载体、逆转录病毒载体等。
本文使用的术语序列“同一性”通过在比较窗口上比较两个经最佳匹配的序列来确定同一性百分比,其中比较窗口中多核苷酸或多肽序列的部分可以包含添加或缺失(即间隙),例如对于最佳匹配的两个序列而言与参考序列(其不包含添加或缺失)相比20%或更少的间隙(例如5至15%、或10至12%)。通常通过确定在两个序列中发生相同的核酸碱基或氨基酸残基的位置的数目来计算百分比,以产生正确匹配的位置的数目,将正确匹配位置的数目除以参考序列中的位置总数(即窗口大小),并将结果乘以100,以产生序列同一性的百分比。
术语“antiGPC3”、“anti-GPC3”和“抗GPC3”可以互换使用,具有相同意思。
术语“IL12”、“白介素12”及“interleukin-12”可以互换使用,具有相同意思。术语“IL12”是一种具有多种生物学活性的免疫调节因子,如,术语“IL12”可以是指如SEQ ID NO:27中定义的人IL12或者其活性片段,也可以是其他种属。
在一些实施方案中,应用于构建所述的特异结合GPC3的受体的元件或所述的IL12可以是天然存在的,比如其可被分离或纯化自哺乳动物;也可以是人工制备的,比如可以根据常规的基因工程重组技术来生产重组各元件或IL12。优选的,本发明可采用重组的各元件或IL12。
在所述各元件或IL12多肽序列的基础上,经过一个或多个氨基酸残基的取代、缺失或添加而形成的氨基酸序列也包括在本发明中。适当替换氨基酸是本领域公知的技术,所述技术可以很容易地被实施并且确保不改变所得分子的生物活性。这些技术使本领域人员认识到,一般来说,在一种多肽的非必要区域改变单个氨基酸基本上不会改变生物活性。
所述各元件或IL12的多肽的生物活性片段都可以应用到本发明中。在这里,所述的生物活性片段的含义是指作为一种多肽,其作为全长多肽的一部分,仍然能保持全长的多肽的全部或部分功能。通常情况下,所述的生物活性片段至少保持50%的全长多肽的活性。在更优选的条件下,所述活性片段能够保持全长多肽的60%、70%、80%、90%、95%、99%、或100%的活性。
在所述各元件或IL12多肽序列的基础上,经修饰或改良的多肽也可以应用到本发明 中,比如,可采用为了促进其半衰期、有效性、代谢、和/或多肽的效力而加以修饰或改良的多肽。也就是说,任何不影响多肽的生物活性的变化形式都可用于本发明中。
术语“肿瘤”指的是一种以细胞或组织的病理性增生为特征的疾病,及其随后的迁移或侵袭其他组织或器官。肿瘤生长通常是不受控制的和进行性的,不诱导或抑制正常细胞增殖。肿瘤可影响多种细胞、组织或器官,包括但不限于选自膀胱、乳腺、食管、肠、肾、肝、肺、淋巴结、神经组织、卵巢、胰腺、前列腺、骨骼肌、皮肤、脊髓、脾、胃、子宫器官,或组织或相应的细胞。本发明所述的肿瘤,可包括,但不限于肝癌、胃癌、肺癌、乳腺癌、头颈癌、膀胱癌、卵巢癌、宫颈癌、肾癌、胰腺癌、宫颈癌、脂肪肉瘤、黑色素瘤、肾上腺癌、神经鞘瘤、恶性纤维组织细胞瘤、食道癌。较佳地,所述的“肿瘤”包括但不限于:肝癌、胃癌、肺癌、乳腺癌。
细胞外抗原结合区可以结合任何互补靶。细胞外抗原结合区可以衍生自已知可变区序列的抗体。细胞外抗原结合区可以从获自可获得的小鼠杂交瘤的抗体序列中得到。或者,可以从肿瘤细胞或原代细胞例如肿瘤浸润淋巴细胞(TIL)的全外切割测序获得细胞外抗原结合区。
在一些情况下,细胞外抗原结合区的结合特异性可以通过互补决定区或CDR,如轻链CDR或重链CDR来确定。在许多情况下,结合特异性可以通过轻链CDR和重链CDR来确定。与其他参考抗原相比,给定的重链CDR和轻链CDR的组合可以提供给定的结合袋,其可以赋予抗原(例如GPC3)更大的亲和力和/或特异性。例如,特异于磷脂酰肌醇蛋白聚糖-3的CDR可以在CAR的细胞外结合区域中表达,使得靶向GPC3的CAR可以将免疫应答细胞靶向表达GPC3的肿瘤细胞。

通过使用基因工程,可以以各种方式修饰细胞外抗原结合区。在一些情况下,可以突变细胞外抗原结合区域,从而可以选择细胞外抗原结合区域以对其靶标具有更高的亲和力。在一些情况下,细胞外抗原结合区域对其靶标的亲和力可针对可在正常组织上以低水平表达的靶标进行优化。可以进行此优化,以尽量减少潜在的毒性。在其他情况下,对靶标的膜结合形式具有更高亲和力的细胞外抗原结合区域的克隆可以优于其可溶形式的对应物。可以进行这种修饰,因为也可以检测到不同水平的可溶形式的靶标,并且它们的靶向可引起不期望的毒性。
在一些情况下,细胞外抗原结合区域包括铰链或间隔区。术语铰链和间隔区可以互换使用。铰链可以被认为是用于向细胞外抗原结合区提供柔性的一部分。
术语“治疗”是指在试图改变个人或处理细胞引起的的疾病过程中的临床干预,既可以进行预防也可以在临床病理过程干预。治疗效果包括但不限于,防止疾病的发生或复发、减轻症状、减少任何疾病直接或间接的病理后果、防止转移、减慢疾病的进展速度、改善或缓解病情、缓解或改善预后等。
术语“组成性表达”是指在所有的生理条件下的表达。
术语“诱导表达”是指在一定条件下的表达,所述的条件例如T细胞与抗原发生结合的时候。本领域技术人员如何进行常规的“诱导表达”。
术语“Notch受体”具有三个关键组分:1)配体结合表皮生长因子(EGF)重复序列,2)在激活期间控制受体的裂解的notch core(notch核心),以及3)被释放并调控转录的细胞内结构域。
术语“Notch core”是利用控制配体依赖性裂解和激活的最小支架,包括控制S2裂解位点对金属蛋白酶而言的可接近性的Lin12-Notch重复序列(LNR)、异源二聚化结构域(HD),以及包含Notch细胞内结构域(NICD)的释放所需的γ-分泌酶裂解位点的跨膜结构域(TMD)。
免疫细胞
在一些实施方案中,本发明提供了一种免疫细胞,该细胞表达质粒载体对。
在一些实施方案中,该免疫细胞能够局部分泌IL12或者调控表达IL12。
在一些实施方案中,该免疫细胞能够增强CAR-T细胞的肿瘤杀伤。
组合物
本发明的免疫细胞可以与CAR-T细胞组合使用。所述的药物组合物除了包括有效量的免疫细胞,还可包含有效量的CAR-T细胞。
与抗肿瘤药物联合
在一些实施方案中,本发明的免疫应答细胞可以与另一治疗剂联合给药。在一些实施方案中,所述另一治疗剂是化疗药。可以与本发明的免疫应答细胞联合应用的化疗药物包括但不限于有丝分裂抑制剂(长春花生物碱),包括长春新碱、长春花碱、长春地辛和诺维宾(TM)(长春瑞滨,5'-去氢硫化氢);拓扑异构酶I抑制剂,例如喜树碱化合物,包括CamptosarTM(伊立替康HCL)、HycamtinTM(托泊替康HCL)和衍生自喜树碱及其类似物的其它化合物;鬼臼毒素衍生物,例如依托泊苷、替尼泊苷和米多昔佐兹;烷基化剂顺铂、环磷酰胺、氮芥、三亚甲基硫代磷酰胺、卡莫司汀、白消安、苯丁酸氮芥、布列喹嗪、尿嘧啶芥末、氯洛芬和达卡巴嗪;抗代谢物,包括阿糖胞苷、氟尿嘧啶、甲氨蝶呤、巯嘌呤、硫唑嘌呤和丙卡巴肼;抗生素,包括但不限于多柔比星、博来霉素、更生霉素、柔红霉素、霉素霉素、丝裂霉素、肉瘤霉素C和道诺霉素;以及其它化疗药物,包括但不限于抗肿瘤抗体、达卡巴嗪、氮胞苷、阿姆沙康、美法仑、异环磷酰胺和米托蒽醌。
在一些实施方案中,可以与本发明的免疫应答细胞联合应用的化疗药物包括但不限于抗血管生成剂,包括抗VEGF抗体(包括人源化和嵌合抗体、抗VEGF适体和反义寡核苷酸)以及其他血管发生抑制剂,例如血管抑素、内皮抑制素、干扰素、视黄酸和金属蛋白酶-1和-2的组织抑制剂。
组合药盒

实施例1:核酸编码的嵌合抗原受体蛋白的慢病毒质粒的构建及病毒包装
下表1解释了本发明实例的嵌合抗原受体各部分的连接顺序。

1.antiGPC3-synNotch-GAL4VP64核酸片段扩增
1)GPC3scFv序列的扩增
通过常规的PCR的方法,获得antiGPC3的scFv的核酸序列(SEQ ID NO:2)
以PHR-PGK-antiCD19-synNotch-GAL4VP64(addgene购买)为模板通过PCR方式获得其他核酸序列。其中信号序列(SEQ ID NO:1)以引物对[上游引物:5’-tctcacgcgtcaagtggagc-3’(SEQ ID NO:14),下游引物:5’-tctgcaccagctgcacctcgaggtcctcttcagag-3’(SEQ ID NO:15)]进行PCR扩增;synNotch-GAL4VP64序列以引物对[上游引物:5’- atcctggactacagcttcacaggtggcgc-3’(SEQ ID NO:16),下游引物:5’-agagccggcagcaggccgcgggaag-3’(SEQ ID NO:17)]进行PCR扩增。
2.antiGPC3-synNotch-GAL4VP64核酸片段的拼接
将antiGPC3scfv核酸片段,与等摩尔的信号序列核酸片段以及等摩尔的synNotch-GAL4VP64核酸片段进行三片段拼接并PCR,拼接条件为:预变性:94℃,4min;变性:94℃,40s;退火:42℃,40s;延伸:68℃,3min 20s,进行7个循环,然后总延伸68℃,10min,补充DNA聚合酶及正向引物和反向引物后PCR扩增30个循环,扩增条件为预变性:94℃,4min;变性:94℃,40s;退火:60℃,40s;延伸:68℃,3min 20s,进行30个循环,然后总延伸68℃,10min。扩增获得的片段为antiGPC3-synNotch-GAL4VP64。
3.GAL4UAS-IL12-PGK-mcherry核酸片段扩增
首先以pHR-GAL4UAS-tBFP-PGK-mcherry(购自addgene)为模板在tBFP与PGK-mcherry之间添加一个MluⅠ限制性酶切位点,再以此质粒为模板继续构建。
1)IL12序列的扩增
通过PCR扩增获得IL12序列(其中,IL12P40的核酸序列如SEQ ID NO:7所示,IL12P35的核酸序列如SEQ ID NO:8所示)。
2)其他部分的核酸序列
其他部分的核酸序列以pHR-GAL4UAS-tBFP-PGK-mcherry为模板通过PCR方式获得。其中GAL4UAS序列以引物对[上游引物:5’-agatcgc gatgggaaaaaattc-3’(SEQ ID NO:28),下游引物:5’-tgctgaacttcactctcatgatcc aacgaatgtcgag-3’(SEQ ID NO:29)]进行PCR扩增。
4.GAL4UAS-IL12核酸片段的拼接
分别将前述获得的IL12核酸片段,与等摩尔的GAL4UAS核酸片段进行两片段拼接并PCR,拼接条件为:预变性:94℃,4min;变性:94℃,40s;退火:42℃,40s;延伸:68℃,3min 20s,进行7个循环,然后总延伸68℃,10min,补充DNA聚合酶及正向引物和反向引物后PCR扩增30个循环,扩增条件为预变性:94℃,4min;变性:94℃,40s;退火:60℃,40s;延伸:68℃,3min 20s,进行30个循环,然后总延伸68℃,10min。扩增获得的片段为GAL4UAS-IL12。
5.慢病毒质粒载体的构建
慢病毒质粒载体使用的载体系统属于第三代自灭活慢病毒载体系统,该系统共有三个质粒即编码蛋白Gag/Pol、编码Rev蛋白的包装质粒psPAX2(购自addgene);编码VSV-G蛋白的包膜质粒PMD2.G(购自addgene)。
上述步骤1和2中获得的目的基因antiGPC3-synNotch-GAL4VP64通过MluⅠ和SacⅡ限制性内切酶双酶切,连入同样双酶切的pHRLSIN载体(购自addgene)中;以及目的基因GAL4UAS-IL12通过NruⅠ和MluⅠ限制性内切酶双酶切,连入同样双酶切的添加了MluⅠ的pHRLSIN载体中。构建成功的载体经序列测序正确后,可以准备用于慢病毒包装。
得到的含有目的片段的载体如下(功能元件及连接关系参见图1A和图1B):
pHRLSIN-antiGPC3-synNotch-GAL4VP64(即,pHR-antiGPC3-synNotch-GAL4VP64,其中antiGPC3-synNotch-GAL4VP64的序列如SEQ ID NO:21,所示);
pHRLSIN-GAL4UAS-IL12-PGK-mcherry(即,pHR-GAL4UAS-IL12-PGK-mcherry,其序列如SEQ ID NO:22所示)
6.质粒转染293T包装慢病毒
1)以5×10
6的密度接种培养至第6~10代的293T细胞于10cm培养皿中,37℃、5%CO
2培养过夜准备用于转染,培养基为含10%胎牛血清的DMEM;
2)分别将目的基因质粒pHRLSIN-antiGPC3-synNotch-GAL4VP64、pHRLSIN-GAL4UAS-IL12-PGK-mcherry 10μg分别与包膜质粒pMD2.G 3μg、包装质粒pAX2 7.5μg溶入800μl空白DMEM培养液,混匀;
3)将60μg聚醚酰亚胺(PEI)(1μg/μl)溶解于800μl的无血清DMEM培养液中,轻轻混匀(或1000rpm涡旋5秒)温孵育5min;
4)转染复合物的形成:将质粒混合液加入PEI混合液中,加入后立即涡旋混合或轻轻混匀,室温下孵育20min;
5)将转染复合物1.6ml滴加入含11ml DMEM培养基的10cm培养皿中(无需换液);
6)4-5h后,用10%FBS的DMEM培基给转染的293T细胞换液;
7)在转染72h后,使用0.45μm滤器过滤收集病毒上清液;
8)5×PEG-8000NaCl浓缩病毒,每30ml初始液加入母液7.5ml。每隔20-30min混合 一次,共进行3-5次,4℃放置过夜;
9)次日4℃,4000rpm离心60min,弃离心上清,离心所得沉淀用1/10-1/50原液体积的溶解液进行重悬以100μl/管分装冻存于-80℃,得到慢病毒1。
7.慢病毒滴度测定
1)以1×10
5细胞数目接种293T细胞于12孔培养板,1ml/孔;按0.6μl/ml加入初始浓度10μg/μl的polybrene溶液,终浓度6μg/ml;37℃、5%CO
2培养1h,培养液为含10%胎牛血清的DMEM;
2)加10μl/孔的病毒浓缩液5倍稀释,3个梯度,37℃、5%CO
2培养;
3)感染72h后,胰酶消化(30s)293T细胞,加1ml DMEM(10%FBS)终止消化,将细胞悬液转移入2ml离心管中(二等分),5000rpm,离心5min,弃上清;PBS(2%N-溴代琥珀酰亚胺(NBS))洗两次;
4)pHRLSIN-antiGPC3-synNotch-GAL4VP64病毒对照组细胞直接PBS(2%NBS)洗两次,重悬后作为对照;检测组细胞如上所述,检测组细胞+50μl 1:50稀释的Alexa(R)-647标记的anti-myc抗体,冰上孵育45min,5000rpm/min,离心5min弃上清;重复两次;加入500μl PBS(2%NBS),转移至流式管中,流式细胞仪检测Alexa(R)-647通道。
5)pHRLSIN-GAL4UAS-IL12-PGK-mcherry病毒流式细胞仪直接检测PE-CF594通道;
6)以阳性率为5-20%的细胞数为宜,计算滴度(U/mL)=细胞数目×阳性率/病毒体积。经浓缩后所测定的病毒滴度约pHRLSIN-antiGPC3-synnotch-GAL4VP64:1×10
8U/ml;pHRLSIN-GAL4UAS-IL12-PGK-mcherry:3×10
8U/ml。
实施例2:重组慢病毒感染NK92细胞
将实施例1制备的慢病毒1感染NK92细胞,得到GPC3-SYN-IL12-NK92细胞,具体操作如下:
1)感染前一天以重组人纤连粘蛋白(Retronectin)包被24孔板,每孔加入380μl 5μg/ml的重组人纤连粘蛋白溶液(PBS),4℃孵育过夜;
2)感染时弃去24孔板中的重组人纤连粘蛋白溶液(PBS),1ml PBS洗两次。以MOI=30用上述重组慢病毒感染,同时补加终浓度10μg/ml的polybrene以提高感染效率,每孔细胞数目5×10
5,培基500μl,32℃,1800g,离心90min后,转移至细胞培养箱中;
3)感染后的细胞每隔一天采用5×10
5/ml的密度进行传代。
实施例3:GPC3-SYN-IL12-NK92细胞的鉴定
慢病毒感染的NK92细胞在培养第7天,通过流式细胞检测不同受体表达,由于antiGPC3的N端带有Myc标签,检测Myc表达即为pHRLSIN-antiGPC3-synNotch-GAL4VP64感染成功的阳性细胞,检测mcherry表达即为pHRLSIN-GAL4UAS-IL12-PGK-mcherry感染成功的阳性细胞,Myc和mcherry同时表达即为双感染成功的阳性细胞GPC3-SYN-IL12-NK92。
1)不同感染的NK92细胞分别取1×10
6细胞,二等分于2ml离心管内,4℃,5000rpm,离心5min,弃上清,PBS洗两次;
2)检测myc表达的对照组细胞加入直接PBS(2%NBS)洗两次,重悬后作为对照;检测组细胞如上所述,检测组细胞具体+50μl 1:50稀释的Alexa(R)-647标记的anti-myc抗体,冰上孵育45min;
3)加入2ml PBS(2%NBS)重悬细胞,4℃,5000rpm/min,离心5min弃上清;重复两次;
4)加入500μl PBS(2%NBS),转移至流式管中。流式细胞仪检测Alexa(R)-647以及PE-CF594通道,确定阳性NK92细胞比例。
结果如图2所示,GPC3-SYN-IL12-NK92细胞中Myc和Mcherry标签双阳性率为40.6%,其中Myc阳性率为63.1%,Mcherry阳性率为54.2%。
GPC3-SYN-IL12-NK92细胞以细胞密度为5×10
5/ml隔天传代培养、计数,培养第11天约有20-40倍的扩增,表明双感染的NK92细胞在体外能够进行一定数量的扩增,为后续体外毒性试验及体内试验提供了保证。
实施例4:GPC3-SYN-IL12-NK92细胞的IL12调控表达
1)调控表达使用材料如下:肝癌细胞系(Huh7(高表达GPC3)、PLC/PFR/5(低表达GPC3)、SK-Hep-1(不表达GPC3)、SK-Hep-1-GPC3(过表达GPC3))作为靶细胞抗原刺激,各靶细胞GPC3的表达情况如图3所示。效应细胞为体外培养12天的嵌合抗原受体表达的NK92细胞;
2)效靶比(effector:target)按1:1铺12孔板,2×10
5/孔;400g离心1min增加相互接触;
3)其中各实验组和各对照组如下:
各实验组:各靶细胞+GPC3-SYN-IL12-NK92
对照组:各靶细胞+NK92
4)37℃、5%CO
2条件下共培养24h后收集上清,Elisa试剂盒检测IL12的表达。
参见图4A,由Elisa结果可以看出,分别与中高表达GPC3的细胞系Huh7、PLC、及过表达GPC3的SK-Hep-1共孵育,本发明的GPC3-SYN-IL12-NK92细胞的IL12分泌量显著性升高,而对不表达GPC3的SK-Hep-1细胞IL12只有一点基础表达,表明GPC3-SYN-IL12-NK92细胞分泌的IL12依赖靶抗原的刺激调控。
GPC3-Syn-IL12NK92细胞与Huh7细胞和SK-Hep-1-GPC3共孵育中,在不同时间点的IL12分泌情况如图4B所示,显示随着时间的延长,IL12分泌量大大提高。
实施例5:CAR-T细胞的构建
作为示例性的,本实施例构建了靶向GPC3的CAR-T细胞的CAR具有SEQ ID NO:23所示的氨基酸序列。应理解,其他代次的CAR也具有类似的效果,如SEQ ID NO:24、SEQ ID NO:25、SEQ ID NO:24和SEQ ID NO:26所示GPC-BBZ、GPC-28BBZ、GPC3-z。
以pRRLSIN-cPPT.EF-1α为载体,构建了表达人源化抗体huGPC3的二代嵌合抗原受体的慢病毒质粒,PRRLSIN-cPPT.EF-1α-huGPC3-28Z。HuGPC3-28Z序列由CD8α信号肽(SEQ ID NO:9)、huGPC3scFV(SEQ ID NO:2)、CD8hinge(SEQ ID NO:10)、CD28跨膜区(SEQ ID NO:11)和胞内信号传导结构域(SEQ ID NO:12)以及CD3的胞内段CD3ξ(SEQ ID NO:13)组成。
参照实施例1的操作转染293T包装慢病毒,得到慢病毒2。
采用慢病毒2感染T细胞,得到GPC3-28Z CAR-T细胞。
实施例6:体外实验
采用CytoTox 96非放射性细胞毒性检测试剂盒(Promega公司),具体参照CytoTox96非放射性细胞毒性检测试剂盒说明书。
1)靶细胞:分别接种50μL 2×10
5/mL的huh7细胞、PLC/PRF/5、SK-Hep-1-GPC3、SK-Hep-1细胞于96well板;
2)效应细胞:按效靶比3:1、1:1或1:3加UTD、UTD+GPC3-Syn-IL12、GPC3-28Z、 及GPC3-28Z+GPC3-Syn-IL12细胞。
效应细胞与靶细胞共孵育12小时,实验结果如图6所示。
实施例7:体内抑瘤实验
a.NSG小鼠接种Huh7移植瘤
通过皮下接种Huh7细胞,2×10
6/只。
b.准备UTD、GPC3-28Z CAR-T细胞和双感染GPC3-SYN-IL12-NK92
1)d0天,取PBMC细胞3×10
7,按CD3-CD28抗体包被磁珠:细胞=2:1添加磁珠,培养基为AIM-V(2%AB血清),总体积为5ml。并添加终浓度为500IU/ml的IL-2;
2)d1天,以重组人纤连粘蛋白包被24孔细胞培养板:重组人纤连粘蛋白浓度为5μg/ml,380μl/孔,4℃孵育过夜;
3)d2天,取活化后的细胞进行病毒感染,每种病毒感染PBMC数目为3×10
6、MOI=10。将细胞及病毒液转移至包被了重组人纤连粘蛋白的24孔板内,感染条件为32℃、1800rpm离心40min;
4)d3天,对感染后的PBMC细胞低速离心(500rpm,10min)去除死细胞后换液;
5)d5天,通过磁极去除CD3-CD28抗体包被磁珠,培养至12天。
c.对Huh7移植瘤进行GPC3-28Z CAR-T细胞和GPC3-SYN-IL12-NK92细胞联合过继免疫治疗。
d18天,测量NSG小鼠Huh7移植瘤体积,约为150mm
3。小鼠分为4组,包括:UTD对照组、GPC3-28Z CAR-T单独治疗组、GPC3-SYN-IL12-NK92单独治疗组以及GPC3-28Z CAR-T+GPC3-SYN-IL12-NK92联合治疗组,每组小鼠6只;并对分组后的小鼠进行过继免疫治疗。通过尾静脉注射200μl细胞。
(a)UTD组(没有转染载体的T细胞组):1×10
6/只UTD。
(b)GPC3-28Z CAR-T细胞组:1×10
6/只阳性细胞。
(c)GPC3-SYN-IL12-NK92组:1×10
6/只阳性细胞。
(d)GPC3-28Z CAR-T+GPC3-SYN-IL12-NK92联合治疗组:分别1×10
6/只阳性细胞,之后每隔3-4天,再次给与GPC3-SYN-IL12-NK92细胞,共给与四次,每次1×10
6/只阳性细胞。
d.每隔3-4天测量Huh7移植瘤体积的大小,记录每组小鼠瘤体积的变化,瘤体积计算公式为:(长×宽
2)/2。结果如图5A所示,可看出GPC3-28Z CAR-T+GPC3- SYN-IL12-NK92联合治疗组能显著抑制肿瘤生长。
在Day 45,将小鼠引颈处死,剥离小鼠皮下肿瘤并称量瘤重,结果如图5B所示,与对照组相比,GPC3-28Z CAR-T+GPC3-SYN-IL12-NK92联合治疗组小鼠肿瘤重量较轻。
在记录每组小鼠瘤体积的变化过程中,小鼠的体重变化如图5C所示,各治疗组小鼠体积变化没有统计学差异。数据的统计方式为±SEM,*P<0.05,**P<0.01,***P<0.001,****P<0.0001,2way ANOVA。
采用常规组织切片制备方法,将小鼠的肿瘤组织制成组织切片。将组织切片进行免疫组化染色,采用抗人CD3抗体(Thermo Scientific)来检测肿瘤组织切片中人的CAR-T细胞,结果如图7所示,显示GPC3-28Z CAR-T+GPC3-SYN-IL12-NK92联合治疗组有更多的CAR-T细胞浸润,以上结果说明该联合治疗组的两种细胞间有协同作用。
结论:体外实验中,GPC3-28Z+GPC3-Syn-IL12细胞与对照组相比,对肿瘤细胞的杀伤效果无明显差异;而在体内实验中,GPC3-28Z CAR-T+GPC3-SYN-IL12-NK92联合治疗组与对照组相比,小鼠肿瘤生长受到明显抑制。
以上实施例的说明只是用于帮助理解本发明的核心思想。应当指出,对于本领域的普通技术人员而言,在不脱离本发明原理的前提下,还可以对本发明的固体粉末组合物及其制备方法进行若干改进和修饰,但这些改进和修饰也落入本发明权利要求请求保护的范围内。
Claims (35)
- 质粒载体对,其包括第一载体和第二载体,其特征在于,所述第一载体包含Notch核心,还包括其他胞外段和胞内段的编码核酸分子,所述胞内段优选地含有转录激活因子;所述第二载体含有与所述第一载体中的胞内段特异性识别和结合的核酸分子以及编码IL12的核酸分子。
- 根据权利要求1所述的质粒载体对,其特征在于,所述第一载体的Notch核心具有SEQ ID NO:3所示的核酸序列;优选地,所述第一载体的胞内段为GAL4VP64,所述第二载体含有GAL4UAS。
- 权利要求1所述的质粒载体对,其中所述胞外段含有抗原或该抗原的抗体或其片段,或含有受体或该受体的配体。
- 权利要求1所述的质粒载体对,其特征在于,所述第一载体的胞外段能够特异性识别肿瘤抗原;优选地,所述肿瘤抗原为实体瘤抗原,所述肿瘤抗原选自前列腺特异性膜抗原(PSMA)、癌胚抗原(CEA)、IL13Ralpha、HER-2、CD19、NY-ESO-1、HIV-1Gag、Lewis Y、MART-1、gp100、酪氨酸酶、WT-I、hTERT、间皮素、EGFR、EGFRvIII、GPC3、EphA2、HER3、EpCAM、MUC1、MUC16、CLDN18.2、叶酸受体、CLDN6、CD30、CD138、ASGPR1、CDH16、GD2、5T4、8H9、αvβ6整合素、B细胞成熟抗原(BCMA)、B7-H3、B7-H6、CAIX、CA9、CD20、CD22、κ轻链(kappa light chain)、CD33、CD38、CD44、CD44v6、CD44v7/8、CD70、CD123、CD171、CSPG4、EGP2、EGP40、ERBB3、ERBB4、ErbB3/4、FAP、FAR、FBP、胚胎型AchR、GD2、GD3、HLA-AI MAGE A1、MAGE3、HLA-A2、IL11Ra、KDR、Lambda、MCSP、NCAM、NKG2D配体、PRAME、PSCA、PSC1、ROR1、Sp17、SURVIVIN、TAG72、TEM1、TEM8、VEGRR2、HMW-MAA、VEGF受体、和/或纤连蛋白、腱生蛋白或肿瘤坏死区的癌胚变体中的一种或多种,更优选地,所述肿瘤抗原为GPC3或CLDN18.2。
- 权利要求1的质粒载体对,其中所述胞外段含有scFv;优选anti-GPC3scFv。
- 权利要求1的质粒载体对,其中所述第一载体还包括标签,例如,C-myc,FLAG和/或HA。
- 权利要求1的质粒载体对,其中所述第二载体还包括报道子,例如,GFP,BFP和/或mcherry。
- 权利要求1的质粒载体对,其中所述第一载体的开放阅读框依序包括PGK启动子-信号肽-myc标签–anti-GPC3scFv-Notch核心-GAL4-接头-VP64,所述第 二载体依序包括GAL4UAS-CMV minimal启动子-IL12-PGK启动子–mcherry。
- 权利要求1的质粒载体对,其中所述第一载体的开放阅读框的核苷酸序列含有如SEQ ID NO:21所示的序列,所述第二载体的开放阅读框的核苷酸序列含有如SEQ ID NO:22所示的序列。
- 权利要求1-9任一项所述的质粒载体对,其中所述第一载体或第二载体还包括各自的功能性变体,该变体的核苷酸序列分别与所述第一载体或第二载体的核苷酸序列具有至少50%,优选70%、75%、80%、85%或90%,更优选95%、96%、97%、98%或99%的同一性;优选地,anti-GPC3scFv具有SEQ ID NO:2所示的核苷酸序列具有至少90%的同一性。
- 基因修饰的免疫细胞,其特征在于,所述细胞转染有权利要求1-10任一项的质粒载体对。
- 权利要求11的免疫细胞,其特征在于,所述免疫细胞为T细胞或NK细胞;优选地,所述免疫细胞为NK细胞。
- 根据权利要求12所述的免疫细胞,其特征在于,所述NK细胞为来自血液或者来自细胞系;优选地,来自细胞系;更优选地,所述来自细胞系的NK为NK92细胞系。
- 权利要求1-10任一项的质粒载体对或权利要求11-13任一项的免疫细胞在制备用于局部分泌IL12或者调控表达IL12的药物中的应用。
- 权利要求1-10任一项的质粒载体对或权利要求11-13任一项的免疫细胞在制备用于增强CAR-T细胞杀伤作用的药物中的应用。
- 根据权利要求15所述的用途,其特征在于,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与所述CAR-T细胞识别的肿瘤抗原相同。
- 根据权利要求15所述的用途,其特征在于,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与所述CAR-T细胞识别的肿瘤抗原均表达在同一种肿瘤上。
- 根据权利要求16所述的用途,其特征在于,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原均为GPC3。
- 根据权利要求17所述的用途,其特征在于,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原均在肝癌中有表达。
- 根据权利要求17所述的用途,其特征在于,所述的免疫细胞的第一载体含有如SEQ ID NO:21所示的序列,所述第二载体含有如SEQ ID NO:22所示的序列;所述CAR-T细胞的CAR具有SEQ ID NO:23、24或25所示的序列。
- 根据权利要求14-20任一项所述的用途,其特征在于,所述的免疫细胞与CAR- T细胞的细胞给予比例为1:1~4:1。
- 根据权利要求14-21任意所述的用途,其特征在于,所述的免疫细胞在一个给药周期内可以分为多次给予。
- 包含有权利要求11-13任一项所述的免疫细胞及CAR-T细胞的组合物。
- 根据权利要求23所述的组合物,其特征在于,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原相同。
- 根据权利要求23所述的组合物,其特征在于,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原均表达在同一种肿瘤上。
- 根据权利要求24所述的组合物,其特征在于,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原均为GPC3。
- 根据权利要求25所述的组合物,其特征在于,所述的免疫细胞的第一载体的胞外段特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原均在肝癌中有表达。
- 根据权利要求23所述的组合物,其特征在于,所述的免疫细胞的第一载体含有如SEQ ID NO:21所示的序列,所述第二载体含有如SEQ ID NO:22所示的序列,所述CAR-T细胞的CAR具有SEQ ID NO:23、24或25所示的序列。
- 根据权利要求23-28任一项所述的组合物,其特征在于,所述的免疫细胞与所述的CAR-T细胞的细胞给予比例为1:1~4:1。
- 组合药盒,包括含有权利要求11-13任一项所述的免疫细胞的药盒A及含有CAR-T细胞的药盒B。
- 根据权利要求30所述的组合药盒,其特征在于,所述的免疫细胞的第一载体的胞外域特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原相同。
- 根据权利要求30所述的组合药盒,其特征在于,所述的免疫细胞的第一载体的胞外域特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原均表达在同一种肿瘤上。
- 根据权利要求31所述的组合药盒,其特征在于,所述的免疫细胞的第一载体的胞外域特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原均为GPC3。
- 根据权利要求32所述的组合药盒,其特征在于,所述的免疫细胞的第一载体的胞外域特异性识别的肿瘤抗原与CAR-T细胞识别的肿瘤抗原均在肝癌中有表达。
- 根据权利要求30所述的组合药盒,其特征在于,所述的免疫细胞的第一载体含有SEQ ID NO:21所示的序列,所述第二载体含有如SEQ ID NO:22所示的序列,所述CAR-T细胞的CAR具有SEQ ID NO:23、24或25所示的序列。
