好ましい具体的な実施例では、前記CAR−T細胞のCARは、SEQ ID NO:23、24または25に示される配列を有する。
いくつかの実施形態では、本発明は、プラスミドベクターペアを発現する免疫細胞を提供する。
本発明の免疫細胞は、CAR−T細胞と組み合わせて使用することができる。前記薬学的組成物は、有効量の免疫細胞に加えて、有効量のCAR−T細胞も含み得る。
いくつかの実施形態では、本発明の免疫応答細胞は、別の治療薬と組み合わせて投与することができる。いくつかの実施形態では、前記他の治療薬は化学療法薬である。本発明の免疫応答細胞と組み合わせて使用することができる化学療法薬は、ビンクリスチン、ビンブラスチン、ビンデシン、ノビビン(TM)(ビノレルビン、5’−硫化水素)などの有糸分裂阻害剤(ビンカアルカロイド);CamptosarTM(イリノテカンHCL)、HycamtinTM(トポテカンHCL)、およびカンプトテシンとその類似体に由来するその他の化合物を含む、カンプトテシン化合物などのトポイソメラーゼI阻害剤;エトポシド、テニポシド、ミドキシゾズなどのポドフィロトキシン誘導体;、アルキル化剤シスプラチン、シクロホスファミド、ナイトロジェンマスタード、トリメチレンチオホスホルアミド、カルムスチン、ブスルファン、クロラムブシル、ブリキアジン、ウラシルマスタード、クロプロフェンおよびダカルバジン;シタラビン、フルオロウラシル、メトトレキサート、メルカプトプリン、アザチオプリン、プロカルバジンなどの代謝拮抗剤;ドキソルビシン、ブレオマイシン、ダクチノマイシン、ダウノルビシン、マイシンマイシン、マイトマイシン、サルコマイシンCおよびダウノマイシンを含むがこれらに限定されない抗生物質;および抗腫瘍抗体、ダカルバジン、アザシチジン、アムサカム、メルファラン、イホスファミド、ミトキサントロンを含むがこれらに限定されないその他の化学療法薬と含むがこれらに限定されない。
本発明は、本発明の免疫細胞を含む組み合わせキットをさらに提供する。前記キットは、癌、病原体感染、免疫障害、または同種移植の治療または予防に使用できる。一実施形態では、キットは、1つまたは複数の単位剤形を含む有効量の免疫細胞を含有する治療用または予防用組成物を含み得る。
以下の表1は、本発明の例のキメラ抗原受容体の各部の接続順序を説明するものである。
1)GPC3 scFv配列の増幅
従来のPCR法により抗GPC3のscFvの核酸配列(SEQ ID NO:2)を取得する。
antiGPC3 scfv核酸断片と等モルのシグナル配列核酸断片および等モルのsynNotch−GAL4VP64核酸断片を3断片スプライシングし、PCRを実行し、スプライシング条件:事前変性:94 ℃、4分;変性:94 ℃,40秒;アニーリング:42 ℃、40秒;伸長:68 ℃、3分20秒、7サイクル行った後、合計伸長68 ℃、10分であり、DNAポリメラーゼおよびフォワードプライマーとリバースプライマーを補足した後、PCR増幅を30サイクル行い、増幅条件は、事前変性:94 ℃、4分;変性:94 ℃、40秒;アニーリング:60 ℃、40秒;伸長:68 ℃、3分20秒、30サイクル行った後、合計伸長68 ℃、10分である。増幅して得られた断片は、antiGPC3−synNotch−GAL4VP64である。
まず、pHR−GAL4UAS−tBFP−PGK−mcherry(addgeneから購入)をテンプレートとしてtBFPとPGK−mcherryとの間に1つのMluI制限酵素切断部位を添加し、次にこのプラスミドをテンプレートとして使用して構築を続行した。
PCR増幅によってIL12配列(ここで、IL12 P40の核酸配列は、SEQ ID NO:7に示され、IL12 P35の核酸配列は、SEQ ID NO:8に示される)を得た。
他の部分の核酸配列は、pHR−GAL4UAS−tBFP−PGK−mcherryをテンプレートとしてPCR法によって得た。ここで、GAL4UAS配列は、プライマーペア[上流プライマー:5’−agatcgc gatgggaaaaaattc−3’(SEQ ID NO:28)、下流プライマー:5’−tgctgaacttcactctcatgatcc aacgaatgtcgag−3’(SEQ ID NO:29)]でPCR増幅を実行した。
上記で取得したIL12核酸断片と等モルのGAL4UAS核酸断片をそれぞれ2断片スプライシングし、PCRを実行した。スプライシング条件は、事前変性:94 ℃、4分;変性:94 ℃,40秒;アニーリング:42 ℃、40秒;伸長:68 ℃、3分20秒、7サイクル行った後、合計伸長68 ℃、10分であり、DNAポリメラーゼおよびフォワードプライマーとリバースプライマーを補足した後、PCR増幅を30サイクル行い、増幅条件は、事前変性:94 ℃、4分;変性:94 ℃,40秒;アニーリング:60 ℃、40秒;伸長:68 ℃、3分20秒、30サイクル行った後、合計伸長68 ℃、10分である。増幅して得られた断片は、GAL4UAS−IL12である。
レンチウイルスプラスミドベクターが使用するベクターシステムは、第3世代の自己不活性化レンチウイルスベクターシステムに属し、前記システムには、Gag/Polをコードするタンパク質と、Revタンパク質をコードするパッケージングプラスミドpsPAX2(addgeneから購入)、VSV−Gタンパク質をコードするエンベローププラスミドPMD2.G(addgeneから購入)の3つのプラスミドがある。
pHRLSIN−antiGPC3−synNotch−GAL4VP64(すなわち、pHR−antiGPC3−synNotch −GAL4VP64、ここで、antiGPC3−synNotch−GAL4VP64の配列は、SEQ ID NO:21に示さる);
pHRLSIN−GAL4UAS−IL12−PGK−mcherry(すなわち、pHR−GAL4UAS−IL12−PGK-mcherry、その配列は、SEQ ID NO:22に示さる)
6.プラスミドトランスフェクション293Tパッケージングレンチウイルス
1)トランスフェクションするために、6〜10継代まで培養した293T細胞を5×106の密度で10 cmペトリ皿に接種し、37 ℃、5% CO2で一晩培養し、培地は10%ウシ胎児血清を含むDMEMである;
2)10 μgの標的遺伝子プラスミドpHRLSIN−antiGPC3−synNotch−GAL4VP64、pHRLSIN−GAL4UAS−IL12−PGK−mcherryをそれぞれ3 μgのエンベローププラスミドpMD2.G、7.5 μgのパッケージングプラスミドpAX2と800 μlのブランクDMEM培養液に溶解し、均一に混合する;
3)60 μgのポリエーテルイミド(PEI)(1 μg/μl)を800 μlの無血清DMEM培養液に溶解し、穏やかに混合(または1000 rpmで5秒間ボルテックス)し、5分間インキュベートする;
4)トランスフェクション複合体の形成:プラスミド混合液をPEI混合液に加え、添加後すぐにボルテックスまたは穏やかに混合し、室温で20分間インキュベートする;
5)1.6 mlのトランスフェクション複合体を11 mlのDMEM培地を含む10 cmペトリ皿に滴下する(培地を交換する必要はない);
6)4−5時間後、トランスフェクトした293T細胞を10 %FBSのDMEM培地と交換する;
7)トランスフェクションの72時間後、ウイルス上清を0.45 μmのフィルターでろ過する;
8)5×PEG−8000 NaCl濃縮ウイルス、30 mlの初期溶液あたり7.5 mlの母液を加える。20−30分に1回、合計3−5回混合し、4 ℃で一晩放置する;
9)翌日4 ℃、4000 rpmで60分間遠心し、上清を廃棄し、遠心分離によって得られた沈殿物を1/10−1/50 の元の容量の溶解溶液で再懸濁し、−80 ℃で100 μl/チューブに保存して、レンチウイルス1を得た。
1)293T細胞を1×105細胞の数で12ウェル培養プレートに接種し、1 ml/ウェル;0.6 μl/ mlで初期濃度10 μg/μlのpolybrene溶液に加え、最終濃度6 μg/ ml;37 ℃、5% CO2で1時間培養し、培養液は、10%ウシ胎児血清を含むDMEMである;
2)10 μl/ウェルのウイルス濃縮液を加えて3つの勾配で5倍に希釈し、37 ℃、5% CO2で培養する;
3)感染から72時間後、293T細胞をトリプシン消化(30秒)し、1 ml DMEM(10%FBS)を加えて消化を停止し、細胞懸濁液を2 mlの遠心チューブに(二等分)に移し、5000 rpmで5分間遠心し、上清を廃棄し;PBS(2% N−ブロモスクシンイミド(NBS))で2回洗浄する;
4)pHRLSIN−antiGPC3−synNotch−GAL4VP64ウイルス対照群細胞を直接にPBS(2% NBS)で2回洗浄し、対照として再懸濁し;実験群細胞は上記のとおり、実験群細胞+50 μl 1:50で希釈したAlexa(R)−647標識anti−myc抗体を氷上で45分間インキュベートし、5000 rpm/分、5分間遠心し、上清を廃棄し;2回繰り返し;500 μl PBS(2% NBS)を加え、フローチューブに移し、フローサイトメトリーでAlexa(R)−647チャネルを確認する;
5)pHRLSIN−GAL4UAS−IL12−PGK−mcherryウイルスフローサイトメトリーでPE−CF594チャネルを直接に検出する;
6)陽性率が5−20%の細胞数を適宜取り、力価(U/mL)=細胞数×陽性率/ウイルス体積を計算する。濃縮後に測定されたウイルス力価は、約pHRLSIN−antiGPC3−synnotch−GAL4VP64:1×108U/ml;pHRLSIN−GAL4UAS−IL12−PGK−mcherry:3×108U/mlである。
実施例1で調製されたレンチウイルス1でNK92細胞を感染させて、GPC3−SYN−IL12−NK92細胞を得、具体的な操作は以下の通りである:
1)感染の1日前に24ウェルプレートに組換えヒトフィブロネクチン(Retronectin)をコーティングし、5 μg/ mlの組換えヒトフィブロネクチン溶液(PBS)380 μlを各ウェルに加え、4℃で一晩インキュベートする;
2)感染時に24ウェルプレート内の組換えヒトフィブロネクチン溶液(PBS)を廃棄し、PBS 1 mlで2回洗浄する。MOI = 30で上記の組換えレンチウイルスを感染させ、ポリブレン(polybrene)を最終濃度10 μg/mlで添加して感染効率を高め、ウェルあたりの細胞数は5×105で、培地は500 μl、32 ℃、1800 g、90分間遠心分離してから細胞インキュベーターに移し;
3)感染後の細胞を5×105/mlの密度で隔日継代する。
レンチウイルスに感染したNK92細胞は、培養7日目にフローサイトメトリーによって異なる受容体発現をテストされ、抗GPC3のN末端にはMycタグがあるため、Mycの発現の検出は、pHRLSIN−antiGPC3−synNotch−GAL4VP64に感染した陽性細胞であり、mcherryの発現の検出は、pHRLSIN−GAL4UAS−IL12−PGK−mcherryに感染した陽性細胞であり、Mycとmcherryの同時発現は、二重感染が成功した陽性細胞GPC3−SYN−IL12−NK92である。
2)myc発現を検出するための対照群細胞を直接PBS(2%NBS)で2回洗浄し、対照として再懸濁し;実験群細胞は上記のとおり、実験群細胞+50 μl 1:50で希釈したAlexa(R)−647標識anti−myc抗体を氷上で45分間インキュベートする;
3)2 ml PBS(2% NBS)を加えて細胞を再懸濁し、4 ℃、5000 rpm/分で5分間遠心し、上清を廃棄し;2回繰り返す;
4)500 μl PBS(2% NBS)を加え、フローチューブに移す。フローサイトメトリーでAlexa(R)−647チャネルおよびPE−CF594チャネルを確認し、陽性のNK92細胞の割合を決定する。
1)調節発現は、次のとおりの材料を使用する:肝癌細胞株(Huh7(GPC3高発現)、PLC/PFR/5(GPC3低発現)、SK−Hep−1(GPC3発現しない)、SK−Hep−1−GPC3(GPC3過剰発現)を標的細胞抗原として刺激し、各標的細胞GPC3の発現状況は図3に示したとおりである。エフェクター細胞は、インビトロで12日間培養されたキメラ抗原受容体を発現するNK92細胞である;
2)エフェクターと標的の比率(effector:target)は、1:1で12ウェルプレートを2×105/ウェル;400gで1分間遠心して相互の接触を増加する;
3)実験群と対照群は以下のとおりである:
各実験群:各標的細胞 + GPC3−SYN−IL12−NK92
対照群:各標的細胞 + NK92
4)37 ℃、5% CO2で24時間共培養後、上清を回収し、ElisaキットでIL12の発現を検出する。
一例として、本実施例ではGPC3のCAR−T細胞を標的とCARは、SEQ ID NO:23に示されるアミノ酸配列を有する。SEQ ID NO:24、SEQ ID NO:25、SEQ ID NO:24およびSEQ ID NO:26に示されるGPC−BBZ、GPC−28BBZ、GPC3−zのように、他の世代のCARも同様の効果があることを理解すべきである。
CytoTox96非放射性細胞毒性検出キット(Promega社)を使用し、CytoTox96非放射性細胞毒性検出キットの指示を参照してください。
2)エフェクター細胞:エフェクターと標的の比率3:1、1:1または1:3でUTD、UTD+GPC3−Syn−IL12、GPC3−28Z、およびGPC3−28Z+GPC3−Syn−IL12細胞を加える。
a.NSGマウスにHuh7移植腫瘍を接種する
Huh7細胞を、マウスあたり2×106個に皮下に接種する。
2)1日目に、24ウェル細胞培養プレートを組換えヒトフィブロネクチンでコーティングし:組換えヒトフィブロネクチンの濃度は5 μg/ml、380 μl/ウェル、4 ℃で一晩インキュベートする;
3)2日目に、ウイルス感染のために活性化細胞を採取し、各ウイルスに感染したPBMCの数は3×106でMOI = 10であった。細胞とウイルス液を組換えヒトフィブロネクチンでコーティングした24ウェルプレートに移し、感染条件は32 ℃、1800 rpmで40分間遠心する;
4)3日目に、感染したPBMC細胞を低速(500 rpm、10分)で遠心分離して死んだ細胞を取り除き、培地を交換する;
5)5日目に、CD3−CD28抗体でコーティングされた磁気ビーズを磁極で取り除き、12日間インキュベートする。

請求項30に記載の組み合わせキット。 - 前記免疫細胞の第1のベクターの細胞外ドメインによって特異的に認識される腫瘍抗原およびCAR−T細胞によって認識される腫瘍抗原はすべてGPC3であることを特徴とする
請求項31に記載の組み合わせキット。 - 前記免疫細胞の第1のベクターの細胞外ドメインによって特異的に認識される腫瘍抗原およびCAR−T細胞によって認識される腫瘍抗原はすべて肝臓癌で発現されることを特徴とする
請求項32に記載の組み合わせキット。 - 前記免疫細胞の第1のベクターは、SEQ ID NO:21に示される配列を含み、前記第2のベクターは、SEQ ID NO:22に示される配列を含み、前記CAR−T細胞のCARは、SEQ ID NO:23、24または25に示される配列を有することを特徴とする
