在细胞培养实验中,必须对培养板,尤其是二次使用的培养板进行无菌处理。由于培养板不能进行高压灭菌处理,一般的实验室又没有辐射灭菌的条件。所以,人们通常在无菌条件的超净台内,对培养板用紫外灯照射的方法进行灭菌处理。为了保证紫外灯照射的效果,紫外灯与培养板之间的距离要控制在30厘米以内。因此,需要在超净台寻找一些物体垫高培养板,使其靠近紫外灯的光源。
但在超净台的台面上放置试管架、移液器架、注射器和收纳上述实验小器材的容器,或直接将上述的实验小器材散放在超净台的台面上,会较大的占用超净台的台面空间,导致在超净台台面上出现的实验样本、实验试剂和实验器材的拥挤现象,这些现象会妨碍科研实验快速和高效的实施过程,影响科研实验的精度和准确率。严重时可导致科研实验的失败。在超净台寻找一些物体垫高培养板进行紫外灯照射消毒时,由于超净台对放置的物品有一定的要求,寻找这些垫高物会比较麻烦,有时还出现垫高物不稳导致培养板的跌落。

在定位锁锁体内设置定位锁硅胶垫,定位锁硅胶垫与磁性底座立柱的抱合,使定位锁固定在磁性底座立柱所需的高度位置上。

使用方法:
预处理:用消毒液对超净台用试管、收纳盘、移液器、注射器架、灭菌托盘各构件部分进行擦拭灭菌,以确保安装在超净台内的具有磁性底座的试管、收纳盘、移液器、注射器架、灭菌托盘不会改变超净台内的无菌环境;如频繁使用,则将其长期放置在超净台中,免除用消毒液的擦拭灭菌;
磁性底座的安装:将恒磁铁旋转开关(5)旋至关的位置;此时,恒磁铁(8)的N和S极处于水平位置,磁力线在被非导磁材料(9)隔开的两块软磁材料(10)中形成闭路,磁性座吸附面(4)没有磁性,可自由移动;将磁性底座(1-7)放置所需位置,具体操作为:将磁性座吸附面(4)与超净台内铁质的垂直边板接触,磁性座底面(7)与超净台内的底面接触;旋转恒磁铁旋转开关至开的位置,此时恒磁铁的N和S极处于上下位置,恒磁铁的N和S极分别对准被非导磁材料隔开的两块软磁材料,从N极到软磁材料到超净台内的铁质垂直边板到另一块软磁材料到S极,形成磁力线闭合,可将磁性座吸附面牢牢的吸附在超净台内的铁质垂直边板上;
试管架水平构件的安装:选择所需的高度,安装试管架水平构件高度定位锁(11-14);具体步骤如下:松开定位锁紧固螺丝(12),将定位锁中间的柱形孔套入立柱(1),选择所需的高度,拧紧定位锁紧固螺丝,使定位锁的硅胶垫(11)与立柱紧紧抱合;高度定位锁安装完毕后,取试管架水平构件(15-18),将试管架连接套筒(18)套进立柱,连接套筒的下缘位于高度定位锁的上缘,完成试管架水平构件的安装和高度定位;
收纳盘水平构件的安装:选择所需的高度,安装收纳盘水平构件高度定位锁,其安装方法与安装试管架水平构件高度定位锁的方法相同;高度定位锁安装完毕后,取收纳盘水平构件(19-22),将该构件的连接套筒(22)套进立柱,连接套筒的下缘位于高度定位锁的上缘,完成高度定位;完成收纳盘水平构件的安装和高度定位;

注射器架水平构件的安装:选择所需的高度,安装注射器水平构件高度定位锁,其安装方法与安装试管架水平构件高度定位锁的方法相同;高度定位锁安装完毕后,取注射器架水平构件(27-30),将注射器架连接套筒(30)套进立柱,连接套筒的下缘位于高度定位锁的上缘,完成注射器架水平构件的安装和高度定位;
灭菌托盘水平构件的安装:选择所需的高度,安装灭菌托盘水平构件高度定位锁,其安装方法与安装试管架水平构件高度定位锁的方法相同;高度定位锁安装完毕后,取灭菌托盘水平构件(31-34),将灭菌托盘连接套筒(34)套进立柱,连接套筒的下缘位于高度定位锁的上缘,完成灭菌托盘水平构件的安装和高度定位;
上述5个水平构件安装的顺序,可根据实验需要,任意选择;连接套筒的内圈直径要略大于立柱的外直径,做到既能让连接套筒在立柱上自由旋转,又不能让其在立柱上产生明显的摆动;至此,已完成该具有磁性底座的试管、收纳盘、移液器、注射器架、灭菌托盘的安装;为保险起见,开紫外灯二次灭菌,二次灭菌完成后,关紫外灯;
带盖塑料微型试管(以下简称试管)的放置:根据实验需要,旋转调整试管架板(17)的水平位置;根据试管的规格,选择向不同规格的试管下挂孔(16)垂直放置或取出试管,由于试管的管口端的直径要大于试管管体的直径,所以直径小的管体可垂直插入试管下挂孔中,而直径大的管口端却卡在试管下挂孔的上方,保证了试管的垂直放置;
收纳物的放置:根据实验需要,水平旋转调整收纳盘的水平空间位置,收纳物放置板边框(20)围绕圆形收纳物放置板(21),并高于收纳物放置板的板面1cm,用于防止收纳物的脱落;根据实验需要在收纳物放置板上放入或取出包括滴管吸头、吸管吸头、酒精棉球、培养瓶盖、打火机、眼科镊的实验常用小器材;
移液器的放置:根据实验需要,旋转调整移液器架板(24)的水平位置;向移液器下挂槽(25)垂直放置或取出移液器,由于移液器根部呈向两侧伸展,所以其宽度要大于移液器的其他部分的直径;所以宽度小的移液器部分能插入移液器下挂槽中,而宽度大的移液器根部却卡在移液器下挂槽的上方,实现了移液器在移液器架上的垂直放置;
注射器的放置:根据实验需要,旋转调节注射器架板(28)的水平位置;向注射器水平放置槽(29)水平放置或取出注射器;
塑料材质的细胞培养板(以下简称培养板)的放置:根据实验需要,水平旋转培养板水平放置板(32)的位置,将培养板和培养板盖放入由防滑脱边框(33)包绕的空间中,培养孔和培养板盖的内面要面向紫外灯;打开超净台内的紫外灯对他们实施灭菌处理;灭菌处理结束后,关掉紫外灯,在无菌条件下,盖好培养板盖,从灭菌托盘上取下已灭菌的培养板,备用;
拆卸:需要从超净台内取出该具有磁性底座的试管、收纳盘、移液器、注射器架、灭菌托盘时,按从上向下的顺序,先垂直向上从立柱移出并取下灭菌托盘水平构件,松开定位锁紧固螺丝,垂直向上移动,从立柱上取出灭菌托盘高度定位锁;取出注射器、移液器、收纳盘和试管架水平构件的方法与取出灭菌托盘水平构件的方法相同,取出注射器、移液器、收纳盘和试管架高度定位锁的方法与取出灭菌托盘高度定位锁的方法相同;最后将恒磁铁旋转开关旋至关的位置,取出磁性底座即可。

    拆卸:需要从超净台内取出该具有磁性底座的试管、收纳盘、移液器、注射器架、灭菌托盘时,按从上向下的顺序,先垂直向上从立柱移出并取下灭菌托盘水平构件,松开定位锁紧固螺丝,垂直向上移动,从立柱上取出灭菌托盘高度定位锁;取出注射器、移液器、收纳盘和试管架水平构件的方法与取出灭菌托盘水平构件的方法相同,取出注射器、移液器、收纳盘和试管架高度定位锁的方法与取出灭菌托盘高度定位锁的方法相同;最后将恒磁铁旋转开关旋至关的位置,取出磁性底座即可。
