Abstract

본 발명은 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자군의 검출방법에 관한 것으로서, 더욱 상세하게는 쥐 흉선 림프종 세포에 저선량 저준위 방사선을 조사한 후 텔로미어 조절 유전자들에 대하여 발현량을 계측하여, 공통 또는 단독으로 발현되는 텔로미어 조절 유전자를 검출하는 방법에 관한 것이다. 상기와 같은 본 발명에 따르면, 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자군의 검출방법을 제공함으로써, 산업 및 의료종사자의 방사선 피폭과 발암과의 상관성을 평가할 수 있는 저준위 방사선 텔로미어 조절 유전자로 활용할 수 있고, 암 환자의 암 진행 및 치료정도를 평가할 수 있는 저준위 방사선 텔로미어 조절 유전자로 활용할 수 있을 뿐만 아니라 방사선과 암 발생 인과관계를 평가할 수 있는 저준위 텔로미어 조절 유전자 회복 지표로 활용할 수 있는 효과가 있다.

Description

저선량률 저준위 방사선 조사된 쥐 흉선 림프종 세포에서 발굴된 텔로미어 조절 유전자군과 그 검출방법
본 발명은 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자군의 검출방법에 관한 것으로서, 더욱 상세하게는 쥐 흉선 림프종 세포에 저선량 저준위 방사선을 조사한 후 텔로미어 조절 유전자들에 대하여 발현량을 계측하여, 공통 또는 단독으로 발현되는 텔로미어 조절 유전자를 검출하는 방법에 관한 것이다.
현재 사회적 논란이 되고 있는 방사선의 인체 영향에 대한 과학적 기전을 해석하고자, 개체뿐만 아니라 세포와 분자 수준에서의 과학적 근거를 마련하고자 노력하고 있지만, 100 mSv 이하의 저선량 방사선 영역에서는 현재까지 재현성 있는 사실 기전을 입증하지 못해 논쟁의 대상이 되고 있는 실정이다.
방사선은 조혈조직 및 장관조직에 손상을 주어 백혈구 감소증 (leukopenia)을 유발하고 장 점막으로부터 정상 세균총 (normal flora)의 침투성을 증가시키며, 따라서 특이 또는 비특이적인 면역방어 기전이 손상되어 감염성 질환에 대한 저항성이 감소되는 등 암과 같은 질병과 연관이 많은 인자로 알려져 있다. 이러한 방사선에 의한 영향은 혈액ㅇ조직 장벽 (blood tissue barrier)의 손상과 식세포 (phagocyte) 수의 감소, 그리고 섭식된 생물을 죽이는 능력과 혈청 보체수준의 감소 및 면역반응의 장해 등으로 나타난다.
2 내지 7 Gy의 방사선에 피폭되면 림프구의 수는 현저히 감소되어, 수 시간 내에 최소수로 되었다가 시간 경과에 따라 서서히 증가되지만, 방사선에 피폭된 후 3-4주가 지나야 정상수준으로 회복될 수 있다. 한편, 0.25 내지 1 Gy 정도의 저선량 방사선을 조사하면 방사선을 조사하지 않은 동물에 비하여 항체형성이 지연되며 일시적으로 매우 높은 항체역가 (antibody peak titer)가 관찰된다. 그러나 0.25 Gy 이하의 저선량 범위에서는 일어날 수 있는 기전이 명확하지 않아, 호메시스 이론과 대립되어 현재까지 논쟁의 여지로 남아 있다 [Stebbing, 1982].
실제 저선량 방사선은 개체성장을 촉진시키고, 생체의 면역기능 항진 및 수명을 연장시킨다는 보고가 있다[Luckey T.D. et al., 1982]. 사람에서도 이러한 방사선 호메시스에 대한 연구 보고가 있어 Bloom 등 (1987)은 0.5 Gy 이하에서는 인체의 세포성 면역이 항진된다고 보고하였고, Nambi와 Soman (1987)은 연간 0.03 uSv (0.3 mrem) 에서는 암의 발생률을 감소시킨다고 보고하였다. 그러나 방사선 노출에 대한 인체 영향 연구 시, 100 mSv 이하의 저선량 방사선의 영향이 인체 영향으로 표현되기까지 오랜 시간이 소요되고 이를 검출하는 데 많은 한계가 있다. 결국, 이와 관련한 연구는 현상학적 효과를 관찰하는 수준일 뿐, 구체적인 영향 기전이 밝혀지지 않아 여전히 논란의 여지를 남겨 놓고 있다.
텔로미어(telomere)는 그리스어 '텔로스(끝)'와 '메로스(부분)'의 합성어로 6개의 뉴클레오티드(AATCCC, TTAGGG 등)가 수천번 반복 배열된 염색체의 끝단을 말한다. 즉, 염색체 말단의 염기서열 부위를 말한다. 이 부분에서는 세포분열이 진행될수록 길이가 점점 짧아져 나중에는 매듭만 남게 되고 세포복제가 멈추어 죽게 되는 것이 밝혀짐으로써 이것이 노화와 수명을 결정하는 원인으로 추정되고 있다. 그런데 정상세포에서 분열함에 따라 텔로미어가 짧아지는 것과 반대로 암세포에서는 텔로미어의 길이가 더 이상 짧아지지 않고 그대로 유지가 되는 것이 암의 유지와 발달의 중요한 역할을 한다는 것이 확인되었다. 그렇기 때문에 텔로미어의 길이를 조절하는 것이 암 치료의 중요한 타겟으로 생각되어짐에도 불구하고 저선량률 저준위 방사선에 의해 텔로미어의 길이가 어떻게 조절 되는지 알려지지 않았다.
본 발명에서는 텔로미어 길이 조절에 관여한다고 알려진 유전자들이 RNA와 단백질 수준에서 저선량률 저준위 방사선에 의해 어떻게 반응하는지 확인하였음. 본 발명에서 저선량률 저준위 방사선에 민감하게 반응하는 것으로 발굴된 유전자들은 이미 텔로미어 길이 조절에 중요한 역할을 한다고 알려져 있는 유전자군임이다. 또한 본 발명에서 발굴된 유전자들이 기존에 저선량률 저준위 방사선 조사에 민감한 유전자라 알려진 바 없으며, 특히나 RNA와 단백질 수준에서 저선량률 저준위 방사선에 의해 다르게 반응하는 유전자들을 발굴함으로써 저선량률 저준위 방사선 조사가 RNA 발현 조절을 통한 단백질 발현 조절이 아닌 RNA 발현과 독립적인 단백질 발현 조절이 가능하다는 것을 보여주었다. 결국 본 발명자들은 0.25 Gy이하의 영역의 저선량 저준위 방사선 자극이 세포의 텔로미어 조절 유전자군을 다르게 반응하도록 하는 것을, 측정 방법을 개발하여 확인할 수 있었고, RNA와 단백질 수준에서 확인함으로써 발명을 완성하였다.
본 발명자는 방사선에 의한 암 억제 연구시, 일반적으로 생쥐를 이용한 연구결과는 사람이나 동물에 바로 적용할 수 있으나 분자적 수준에서 해석하기 어렵다는 한계가 있으므로, 이러한 단점을 보완하기 위해 쥐 흉선 림프종 세포를 이용하여 분자적으로 어떠한 변화가 일어나는지 확인함으로써 본 발명을 완성하게 되었다.
1) 쥐 흉선 림프종 세포에 저선량률 저준위 방사선과 고선량률 저준위 방사선, 고준위 방사선을 조사한 후, 텔로미어 조절 유전자들의 변화를 관찰하였고, 2) 그 중에서 저선량률 저준위 방사선 조사에 민감하게 반응하는 텔로미어 조절 유전자를 중심으로 검증 후, 그 기능을 분석하였으며, 3) 텔로미어 길이 조절을 타겟으로 하는 암 치료에 저선량률 저준위 방사선의 효용성을 확인하고자 하였다.
결론적으로 본 발명의 목적은 (1) 쥐 흉선 림프종 세포 각각에 저선량률 저준위 방사선과 고선량률 저준위 방사선, 고준위 방사선을 조사하는 단계; (2) 조사된 세포 각각에서, 기존에 텔로미어 조절 유전자로 알려진 유전자들의 변화를 관찰하는 단계; (3) 텔로미어 조절 유전자들의 변화가 관찰된 것 중에서, 저선량률 저준위 방사선 조사에 의해 단독으로 발현되는 텔로미어 조절 유전자를 검출하는 단계; 및 (4) 검출된 유전자를 증폭하고 발현량을 측정하는 단계;를 포함하는 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자군의 검출방법을 제공하는데 있다.
이러한 기술적 과제를 이루기 위한 본 발명은, (1) 림프종 세포 각각에 저선량률 저준위 방사선과 고선량률 저준위 방사선을 조사하는 단계; (2) 상기 (1)단계에서 조사된 세포 각각에서, 기존에 텔로미어 조절 유전자로 알려진 유전자들의 변화를 관찰하는 단계; (3) 상기 (2)단계에 의해 텔로미어 조절 유전자들의 변화가 관찰된 것 중에서, 저선량률 저준위 방사선 조사에 의해 단독으로 발현되는 텔로미어 조절 유전자를 검출하는 단계; 및 (4) 상기 (3)단계에 의해 검출된 유전자를 증폭하고 발현량을 측정하는 단계;를 포함하는 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자의 검출방법을 제공한다.
상기 (1)단계에서 저선량률 저준위 방사선량이 23.22 mGy/시간일 수 있다.
상기 (1)단계에서 23.22 mGy/시간의 저선량률 저준위 방사선 환경에서 총 누적선량이 557.28 mGy가 되도록 배양할 수 있다.
상기 4)단계에서 텔로미어 관련 유전자는 tert, wrap53, dkc1, ctc1, pot1, tpp1, tin2, trf1, trf2, rap1, rtel1 및 telo2 유전자 서열에서 선택되는 프라이머로 증폭될 수 있다.
상기 (4)단계에서 정량적 핵산증폭법과 특수단백질 검출검사를 통해서 발현량을 측정할 수 있다.
상기와 같은 본 발명에 따르면, 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자군의 검출방법을 제공함으로써,
산업 및 의료종사자의 방사선 피폭과 발암과의 상관성을 평가할 수 있는 저준위 방사선 텔로미어 조절 유전자로 활용할 수 있고, 암 환자의 암 진행 및 치료정도를 평가할 수 있는 저준위 방사선 텔로미어 조절 유전자로 활용할 수 있을 뿐만 아니라 방사선과 암 발생 인과관계를 평가할 수 있는 저준위 텔로미어 조절 유전자 회복 지표로 활용할 수 있는 효과가 있다.
도 1은 실험예 2.의 선량률과 선량에 따른 텔로미어 길이 조절 암억제 지표 유전자들의 발현 변화에 대해, 핵산증폭법을 이용하여 상대적인 발현양을 확인한 그래프.이다.
도 2는 실험예 3.의 선량률과 선량에 따른 암 억제 관련 텔로미어 조절 단백질의 변화 분석결과를 나타낸 그래프이다. 6개 유전자(TRF2, Tin2, Rap1, Pot1, Rtel1, CTC1)의 단백질 발현량이 변화하였음을 알 수 있다.
도 3은 쥐 흉선 림프종 세포에서 저선량률 저준위 방사선 조사에 의한 텔로미어 조절 유전자들의 변화를 도식화한 그림이다. TRF2, Tin2, Rap1, Pot1은 유전자들의 발현이 증가하고, Rtel1, CTC1은 감소하며, 이로 인해 텔로미어 길이가 줄어들고, 발암가능성도 줄어들게 된다.
이하, 본 발명을 상세히 설명한다.
일 관점에서 본 발명은, (1) 림프종 세포 각각에 저선량률 저준위 방사선과 고선량률 저준위 방사선을 조사하는 단계; (2) 상기 (1)단계에서 조사된 세포 각각에서, 기존에 텔로미어 조절 유전자로 알려진 유전자들의 변화를 관찰하는 단계; (3) 상기 (2)단계에 의해 텔로미어 조절 유전자들의 변화가 관찰된 것 중에서, 저선량률 저준위 방사선 조사에 의해 단독으로 발현되는 텔로미어 조절 유전자를 검출하는 단계; 및 (4) 상기 (3)단계에 의해 검출된 유전자를 증폭하고 발현량을 측정하는 단계;를 포함하는 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자의 검출방법을 제공한다. 여기서 림프종 세포는, 쥐의 흉선 림프종 세포(EL4)일 수 있다. 또한 (1)단계에서, 비교 대상으로 고준위 방사선을 조사하는 세포를 추가할 수 있다.
상기 (1)단계에서 저선량률 저준위 방사선량이 23.22 mGy/시간인 것을 특징으로 한다.
상기 (1)단계에서 23.22 mGy/시간의 저선량률 저준위 방사선 환경에서 총 누적선량이 557.28 mGy가 되는 것을 특징으로 한다.
상기 4)단계에서 텔로미어 관련 유전자는 tert, wrap53, dkc1, ctc1, pot1, tpp1, tin2, trf1, trf2, rap1, rtel1 및 telo2 유전자 서열에서 선택되는 프라이머로 증폭되는 것을 특징으로 한다.
상기 (4)단계에서 정량적 핵산증폭법과 특수단백질 검출검사를 통해서 발현량을 측정하는 것을 특징으로 한다.
쥐 흉선 림프종 세포에서 저선량률 저준위 방사선 조사에 의한 텔로미어 조절 유전자들의 변화를 보면, TRF2, Tin2, Rap1, Pot1은 유전자들의 발현이 증가하고, Rtel1, CTC1은 감소하여, 6개 유전자가 변화하였음을 확인하였다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지는 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
이 발명에서 발굴한 저선량률 저준위 방사선에 의해 민감하게 반응하는 6개의 유전자 (TRF2, Tin2, Rap1, Pot1, Rtel1, CTC1)는 이전에 발명되지는 않았지만, 텔로미어 길이 조절과 관련된 특성에 대한 발명 보고는 있었다. TRF2는 텔로머레이즈와는 독립적으로 텔로미어의 빠른 분해를 유도한다고 알려졌다(Munoa 등, 2006). Tin2는 텔로머레이즈에 의한 텔로미어 연장을 억제한다고 알려졌다(Kim 등, 1999). Rap1이 부족하게 되면 텔로미어 길이가 연장되는 것으로 보아 Rap1는 텔로미어 길이를 억제하는 기능을 가진다고 알려졌다(O'Connor 등, 2004). Pot1은 텔로미어의 길이 연장을 억제한다고 보고되었다 (Kendellen 등, 2009). Rtel1은 텔로미어의 길이를 유지하는데 중요한 역할을 한다고 알려졌다(Uringa 등, 2010). CTC1은 인간 흑색종 세포에서 텔로미어 길이가 짧아지는 것과 세포고사를 억제한다고 알려졌다(Luo 등, 2014).
암 억제를 위해 텔로미어 길이를 조절하는 것이 중요한 치료방법이 될 수 있다는 것은 알려져 있으나 실제로 어떻게 암세포에서 텔로미어의 길이 유지를 억제할 수 있는지에 대해선 아직도 많은 연구가 필요한 상태이다. 본 발명에서는, 저선량률 저준위 방사선 조사에 의해 암세포에서 텔로미어 길이 조절 유전자들의 발현이 어떻게 변하고 이를 통해 암 억제가 되는지 분석하고자 하였다.
실험예 1. 쥐 흉선 림프종 세포 준비와 방사선 조사
쥐 흉선 림프종 세포를 ATCC에서 구매하여 37도, 5% CO2세포배양실에서 안정화시켰다. 쥐 흉선 림프종 세포에 누적선량 557.28mGy의 저준위 방사선을 조사하였으며 이를 또한 2개의 그룹으로 나눠, 저선량률 저준위 방사선 조사와 고선량률 저준위 방사선 조사로 분리하였다. 저준위 방사선의 효과와 비교하기 위해 2Gy의 고준위 방사선을 조사한 세포도 준비하였다.
실험예 2. 텔로미어 조절 유전자들의 발현량 계측과 프라이머들
쥐 흉선 림프종 세포에서 저선량률 저준위 방사선에 민감하게 반응하는 텔로미어 조절 유전자를 발굴하고 기능을 해석하기 위해 필요한 발현량의 계측은, 정량적 핵산 증폭법과 특수단백질 검출검사를 이용하여 분석하였다.
한편, 저선량률 저준위 (23.22mGy/시간) 방사선이 조사된 쥐 흉선 림프종 세포에서 민감하게 반응하는 텔로미어 조절 유전자들에 대하여 발현량을 계측하기 위해 사용한 프라이머 염기서열은 [표 1]에 나타내었다.

저선량률 저준위 (23.22mGy/시간) 방사선이 조사된 쥐 흉선 림프종 세포에서 텔로미어 길이 조절에 관련되어 있다고 알려진 (표 1)의 12개 유전자들에 대하여 발현량을 계측하기 위하여 핵산증폭법을 이용하였다. 그 결과, (도 1)에 나타낸 바와 같이 저선량률 저준위 방사선에 조사된 쥐 흉선 림프종 세포에서 tert, wrap53, dkc1, ctc1 및 trf1까지 5개 유전자가 특이적으로 방사선에 민감하게 반응하여 발현량이 증가 또는 감소하였다.
실험예 3. 저선량률 저준위 방사선에 조사된 쥐 흉선 림프종 세포의 단백질 분석
저선량률 저준위 (23.22mGy/시간) 방사선 조사된 단백질과, 대조군으로 고선량률 저준위 (800mGy/분, 총 선량: 557.28mGy) 방사선 조사된 단백질을 분석하였다. 그 결과 (도 2)에 나타낸 바와 같이 저선량률 저준위 방사선 조사된 쥐 흉선 림프종 세포에서 TRF2, Tin2, Rap1, Pot1, Rtel1및 CTC1 단백질이 특이적으로 민감하게 반응하여 발현량이 증가 또는 감소하였다는 것을 확인하였다.
위와 같은 결과를 보면, 최종적으로 저선량률 저준위 방사선 조사에 의해 민감하게 반응하는 텔로미어 길이 조절 관련 유전자들로, 6개의 유전자 (TRF2, Tin2, Rap1, Pot1, Rtel1, CTC1)를 발굴하여 RNA와 단백질 수준에서 변화를 확인하였다. 이를 (도 3)과 같이 요약할 수 있는데, 쥐 흉선 림프종 세포에서 저선량률 저준위 방사선 조사에 의한 텔로미어 조절 유전자들의 변화를 도식화한 그림이다. TRF2, Tin2, Rap1, Pot1은 유전자들의 발현이 증가하고, Rtel1, CTC1은 감소하며, 이로 인해 텔로미어 길이가 줄어들고, 발암가능성도 줄어들게 된다.
이상, 본 발명내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적인 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의해 정의된다고 할 것이다.

Claims (5)

  1. (1) 림프종 세포 각각에 저선량률 저준위 방사선과 고선량률 저준위 방사선을 조사하는 단계;
    (2) 상기 (1)단계에서 조사된 세포 각각에서, 기존에 텔로미어 조절 유전자로 알려진 유전자들의 변화를 관찰하는 단계;
    (3) 상기 (2)단계에 의해 텔로미어 조절 유전자들의 변화가 관찰된 것 중에서, 저선량률 저준위 방사선 조사에 의해 단독으로 발현되는 텔로미어 조절 유전자를 검출하는 단계; 및
    (4) 상기 (3)단계에 의해 검출된 유전자를 증폭하고 발현량을 측정하는 단계;를 포함하는 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자의 검출방법.
  2. 제 1항에 있어서,
    상기 (1)단계에서 저선량률 저준위 방사선량이 23.22 mGy/시간인 것을 특징으로 하는 저선량률 저준위 방사선에 만응하는 텔로미어 조절 유전자의 검출방법.
  3. 제 1항에 있어서,
    상기 (1)단계에서 23.22 mGy/시간의 저선량률 저준위 방사선 환경에서 총 누적선량이 557.28 mGy가 되도록 배양하는 것을 특징으로 하는 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자의 검출방법.
  4. 제 1항에 있어서,
    상기 4)단계에서 텔로미어 관련 유전자는 tert, wrap53, dkc1, ctc1, pot1, tpp1, tin2, trf1, trf2, rap1, rtel1 및 telo2 유전자 서열에서 선택되는 프라이머로 증폭되는 것을 특징으로 하는 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자의 검출방법.
  5. 제 1항에 있어서,
    상기 (4)단계에서 정량적 핵산증폭법과 특수단백질 검출검사를 통해서 발현량을 측정하는 것을 특징으로 하는 저선량률 저준위 방사선에 반응하는 텔로미어 조절 유전자의 검출방법.
