具体实施方式
本发明人经过广泛而深入的研究,制备了一类具有式I所示结构的化合物,并发现其具有JAK1和/或JAK2抑制及降解活性。且所述的化合物在极低浓度下,即可对JAK1和/或JAK2产生抑制及降解作用,活性相当优异,因而可以用于治疗与JAK1和/或JAK2活性或表达量相关的疾病如肿瘤。在此基础上完成了本发明。
本发明提供了一类新化合物及其降解酪氨酸蛋白激酶JAK1和/或JAK2的用途。本发明的化合物能够抑制或降解JAK1和/或JAK2,并且可用作JAK1和/或JAK2抑制剂或降解剂而挥免疫调节作用,以及用于治疗哺乳动物的类风湿性关节炎、银屑病、银屑病性关节炎、强直性脊柱炎、特应性皮炎、干燥性角膜结膜炎以及克罗恩病等疾病。
术语
本发明中,术语“C 1-8烃基”是指只含有碳、氢两种原子的官能团,其中,碳原子的个数为1~8。烃基可以看作是相应的烃失去一个氢原子后剩下的自由基,其可为烷基、环烷基、烯基或炔基等;其结构可为直链、支链或环状;可以是脂肪族的,也可以是芳香族的。

下面更具体地描述本发明式I结构化合物的制备方法,但这些具体方法不对本发明构成任何限制。本发明化合物还可以任选将在本说明书中描述的或本领域已知的各种合成方法组合起来而方便的制得,这样的组合可由本发明所属领域的技术人员容易的进行。
下列反应流程阐述本发明化合物的制备。除非另有指示,反应流程和随后的讨论中的A、B、W、Y、Z、X、a、b、c、d、e、R 1、R 2、R 3、R 4、R 5、R 6、R 7、R 8、R 9、R 10是如上述定义的。
一般而言,可以如下述方案中所述的从式III获得式I的化合物:

在化合物II中,当W为醚时,可采用式III(W1=OH)在碱作用下与含有离去基团的中间体直接亲核取代制备,或采用光延反应与醇反应来制备;当W为酯、氨基甲酸酯(NHCO 2)时,可采用式III(W1=OH)在碱作用下与酰氯、活化酯(酰胺)、羧酸、异氰酸酯反应制备;当W为胺时,可采用式III(W1=NH 2)在碱作用下与含有离去基团的中间体直接亲核取代制备,也可采用式III(W1=NH 2)与醛/酮进行还原胺化反应来制备;当W为酰胺、烷氧羰基胺(OCONH)及脲时,可采用式III(W1=NH 2)在碱作用下与相应酰氯、活化酯(酰胺)、羧酸、异氰酸酯反应制备。
在化合物I中,当A与氮原子以C-N键连接时,可用直接取代或还原胺化制备;当A与氮原子以酰胺、脲、氨基甲酸酯、磺酰胺、硫酰胺方式连接时,可用相应的酰氯、活化酯(酰胺)、羧酸、异氰酸酯、磺酰氯、硫酰氯制备。
一般的,根据连接结构A与W的情况,化合物I也可以通过先把式IV与中间的链连接,再与式III反应得到。所用化学合成方法同上所述。
式III、IV的化合物可通过已知的合成方法获得或容易通过商业购买获得。
式I化合物的应用
所述的式I化合物可用于以下的一种或多种用途:
(a)制备治疗与JAK1和/或JAK2活性或表达量相关的疾病的药物;
(b)制备JAK1和/或JAK2靶向抑制剂或降解剂;
(c)体外非治疗性地抑制或降解JAK1和/或JAK2;
(d)体外非治疗性地抑制肿瘤细胞增殖;和/或
(e)治疗与JAK1和/或JAK2活性或表达量相关的疾病(如自身免疫性疾病)。
在另一优选例中,所述与JAK1和/或JAK2活性或表达量相关的疾病为器官移植排 斥、异种移植、红斑狼疮、多发性硬化、类风湿性关节炎、银屑病(牛皮癣)、癌症、哮喘、特应性皮炎、I型糖尿病与糖尿病并发症、自体免疫性甲状腺障碍、溃疡性结肠炎、克罗恩氏病、阿尔茨海默病、白血病、淋巴瘤、多发性骨髓瘤、斑秃、白癜风等。
本发明的式I化合物可用于制备一种药物组合物,所述的药物组合物包括:(i)有效量的式I化合物,或其药学上可接受的盐;和(ii)药学上可接受的载体。
在另一优选例中,所述的有效量是指治疗有效量或抑制有效量。
本发明的式I化合物还可以用于抑制或降解布JAK1和/或JAK2的方法,所述的抑制是体外非治疗性的抑制也可以是治疗性的抑制。
在另一优选例中,当对抑制对象施用抑制有效量的本发明的式I化合物或其药学上可接受的盐时,所述的抑制有效量为0.001-500nmol/L,较佳地为0.01-200nmol/L。
特别地,本发明还提供了一种治疗与JAK1和/或JAK2活性或表达量相关的疾病的方法,所述方法包括:对治疗对象施用治疗有效量的式I化合物,或所述含有式I化合物作为有效成分的药物组合物。
药物组合物和施用方法
由于本发明化合物可显著降解JAK1和/或JAK2从而发挥对JAK1和/或JAK2抑制活性,因此本发明化合物及其各种晶型,药学上可接受的无机或有机盐,水合物或溶剂合物,以及含有本发明化合物为主要活性成分的药物组合物可用于治疗、预防以及缓解由与JAK1和/或JAK2活性或表达量相关的疾病。根据现有技术,本发明化合物可用于治疗包括肿瘤等的疾病。
本发明的药物组合物包含安全有效量范围内的本发明化合物或其药理上可接受的盐及药理上可以接受的赋形剂或载体。其中“安全有效量”指的是:化合物的量足以明显改善病情,而不至于产生严重的副作用。通常,药物组合物含有1-2000mg本发明化合物/剂,更佳地,含有5-200mg本发明化合物/剂。较佳地,所述的“一剂”为一个胶囊或药片。
“药学上可以接受的载体”指的是:一种或多种相容性固体或液体填料或凝胶物质,它们适合于人使用,而且必须有足够的纯度和足够低的毒性。“相容性”在此指的是组合物中各组份能和本发明的化合物以及它们之间相互掺和,而不明显降低化合物的药效。药学上可以接受的载体部分例子有纤维素及其衍生物(如羧甲基纤维素钠、乙基纤维素钠、纤维素乙酸酯等)、明胶、滑石、固体润滑剂(如硬脂酸、硬脂酸镁)、硫酸钙、植物油(如豆油、芝麻油、花生油、橄榄油等)、多元醇(如丙二醇、甘油、甘露醇、山梨醇等)、乳化剂(如吐温

本发明化合物或药物组合物的施用方式没有特别限制,代表性的施用方式包括(但并不限于):口服、瘤内、直肠、肠胃外(静脉内、肌肉内或皮下)、和局部给药。
用于口服给药的固体剂型包括胶囊剂、片剂、丸剂、散剂和颗粒剂。在这些固体剂型中,活性化合物与至少一种常规惰性赋形剂(或载体)混合,如柠檬酸钠或磷酸二钙,或与下述成分混合:(a)填料或增容剂,例如,淀粉、乳糖、蔗糖、葡萄糖、甘露醇和硅酸;(b)粘合剂,例如,羟甲基纤维素、藻酸盐、明胶、聚乙烯基吡咯烷酮、蔗糖和阿拉伯胶;(c)保湿剂,例如,甘油;(d)崩解剂,例如,琼脂、碳酸钙、马铃薯淀粉或木薯淀粉、藻酸、某些复合硅酸盐、和碳酸钠;(e)缓溶剂,例如石蜡;(f)吸收加速剂,例如,季胺化合物;(g)润湿剂,例如鲸蜡醇和单硬脂酸甘油酯;(h)吸附剂,例如,高岭土;和(i)润滑剂,例如,滑石、硬脂酸钙、硬脂酸镁、固体聚乙二醇、十二烷基硫酸钠,或其混合物。胶囊剂、片剂和丸剂中,剂型也可包含缓冲剂。
固体剂型如片剂、糖丸、胶囊剂、丸剂和颗粒剂可采用包衣和壳材制备,如肠衣和其它本领域公知的材料。它们可包含不透明剂,并且,这种组合物中活性化合物或化合物的释放可以延迟的方式在消化道内的某一部分中释放。可采用的包埋组分的实例是聚合物质和蜡类物质。必要时,活性化合物也可与上述赋形剂中的一种或多种形成微胶囊形式。
用于口服给药的液体剂型包括药学上可接受的乳液、溶液、悬浮液、糖浆或酊剂。除了活性化合物外,液体剂型可包含本领域中常规采用的惰性稀释剂,如水或其它溶剂,增溶剂和乳化剂,例知,乙醇、异丙醇、碳酸乙酯、乙酸乙酯、丙二醇、1,3-丁二醇、二甲基甲酰胺以及油,特别是棉籽油、花生油、玉米胚油、橄榄油、蓖麻油和芝麻油或这些物质的混合物等。

除了活性化合物外,悬浮液可包含悬浮剂,例如,乙氧基化异十八烷醇、聚氧乙烯山梨醇和脱水山梨醇酯、微晶纤维素、甲醇铝和琼脂或这些物质的混合物等。
用于肠胃外注射的组合物可包含生理上可接受的无菌含水或无水溶液、分散液、悬浮液或乳液,和用于重新溶解成无菌的可注射溶液或分散液的无菌粉末。适宜的含水和非水载体、稀释剂、溶剂或赋形剂包括水、乙醇、多元醇及其适宜的混合物。
用于局部给药的本发明化合物的剂型包括软膏剂、散剂、贴剂、喷射剂和吸入剂。活性成分在无菌条件下与生理上可接受的载体及任何防腐剂、缓冲剂,或必要时可能需要的推进剂一起混合。
本发明化合物可以单独给药,或者与其他药学上可接受的化合物联合给药。
使用药物组合物时,是将安全有效量的本发明化合物适用于需要治疗的哺乳动物(如人),其中施用时剂量为药学上认为的有效给药剂量,对于60kg体重的人而言,日给药剂量通常为1~2000mg,优选5~500mg。当然,具体剂量还应考虑给药途径、病人健康状况等因素,这些都是熟练医师技能范围之内的。
本发明的主要优点包括:
1.提供了一种如式I所示的化合物。

将2.14g 6-溴己酰氯和1.37g泊马度胺溶于50ml THF(四氢呋喃)中,搅拌回流8h,降至室温,浓缩后40℃真空干燥得化合物(1)2.19 g。MS(ESI):450[M+H] +

将4-羟基沙利度胺100mg,三甘醇单苄醚96mg,三苯基膦100mg溶解在10ml无水THF中,滴加DIAD(偶氮二甲酸二异丙酯)95mg,室温反应2h。减压去除THF,柱层析纯化后得化合物(4)110mg。MS(ESI):497[M+H] +
第二步:

实施例5用Caliper assay法测试化合物对JAK1/2的抑制活性
实验步骤如下:
配置1×激酶反应缓冲液(50 mM HEPES,PH 7.5;0.0015%Brij-35)和激酶反应终止液(100 mM HEPES,PH 7.5;0.0015%Brij-35;0.2%Coating Reagent;50mM EDTA);
配置实验样品:在96孔板中加入100加入品:在t的样品溶液(100%DMSO溶解),得到50×样品溶液。在同一块板上设置两个只含有100溶液。在同一块板上设置两的孔作为对照。一个作为不加样品对照,另一个作为不加酶对照。在96孔板中加入10中加样品和90和加入照×激酶反应缓冲液,作为中转板。将此中转板震荡10分钟。
准备试验板:取96孔中转板中配置好的样品各5中转板至384孔板中。
激酶反应:在5酶反应:×化合物溶液(用DMSO溶解,用水稀释10倍)中加入10中加的2.5×的JAK1/2激酶溶液(激酶用1×激酶反应缓冲液稀释),室温孵育10min后再加入10加入n的2.5×底物肽溶液(在1×激酶反应缓冲液稀释的FAM标记肽和ATP)。
终止激酶反应:在28℃下反应一段时间后加入25反应一激酶反应终止液。
在Caliper上测试荧光度(F),并收集数据。
计算激酶活性抑制率:对激酶活性的百分抑制率=(F DMSO对照-F 样品)/(F DMSO对照-F 阴性对照)×100,以DMSO为溶液对照,以不加激酶为阴性对照。
结果显示,化合物(浓度100 nM)对JAK1/2的抑制活性见下表:

实施例6用Western blot检测化合物降解JAK1和JAK2蛋白的活性
细胞株:Jurkat细胞株用含10%小牛血清的RPMI1640培养基于37℃、5%CO 2、饱和湿度孵育箱内培养。
设置DMSO对照组、化合物干预组(10组、,),处理24小时收集细胞后加入100μL预冷的细胞裂解液,冰上裂解30 min,提取细胞总蛋白,二喹啉甲酸(BCA)法测定蛋白浓度并进行定量。常规制胶、上样、电泳,然后转膜,封闭,分别加入兔抗人JAK1(1∶500)和兔抗人JAK2(1∶500),4℃孵育过夜,漂洗后加入辣根过氧化物酶标记的山羊抗兔 IgG(1∶5 000),漂洗后ECL显影液显色,Bio-Rad凝胶成像系统扫描成像,计算机软件处理分析。以磷酸甘油醛脱氢酶(GAPDH)内参对照。
使用Image J软件对各条带进行灰度分析,计算化合物降解JAK1或JAK2蛋白的降解率。
结果显示,化合物(10显示)对Jurkat细胞中JAK1/2蛋白的降解活性见下表:

备注:上述表格中,“-注代表无降解活性,“+”代表降解率为10%-30%,“++”代表降解率为30%-50%,“+++”代表降解率为50%-90%,“++++”代表降解率为大于90%。
实施例7 CCK8法检测式化合物对Jurkat细胞(人白血病T淋巴细胞)增殖的抑制作用
在体外用CCK8法实验测定化合物对Jurkat细胞的增殖抑制作用。具体步骤如下:
取Jurkat细胞培养于10%小牛血清的1640培养基中,接种于96孔板,2板,中 5细胞/孔,置于37℃,5%CO 2孵化箱中。将化合物溶解于二甲亚砜(DMSO)中,得到浓度为10 mM的溶液,再用磷酸缓冲液稀释至所需浓度,分别加入上述96孔板中,每个浓度2孔,每孔10每孔,每个浓度作两个平行测试。将DMSO相应作梯度稀释后加入板中,作为对照。
将上述96孔板在37℃、5%CO 2细胞培养箱中培养48小时后,每孔中加入10后,每孔中加入溶液,继续在培养箱中保温1~4小时。测定465nm光吸收值。
根据光吸收值计算化合物处理后细胞相对存活率。
通过软件计算化合物对Jurkat细胞的IC 50
结果显示,合成的化合物对Jurkat细胞的体外增殖抑制作用见下表:

实施例8测试体内抗类风湿性关节炎活性
以全弗氏佐剂诱导Wistar大鼠类风湿性关节炎为模型,测定化合物(3)的抗类风湿性关节炎作用。具体步骤如下:
1)试验样品
样品:化合物(3)。
2)配制方法
样品:化合物(3),配制时用生理盐水注射液溶解
3)动物和佐剂
Wistar大鼠18只,雄性,体重170-200g。
佐剂:全弗氏佐剂(CFA),Sigma。
4)试验方法
将Wistar大鼠随机均分为3组,每组6只。分别为空白对照组、模型组、(化合物(3)组(20mg/kg ip)。于大鼠左后足跖皮内注射CFA 0.1 mL诱导类风湿关节炎。大鼠造模后第11天开始给药治疗,每天一次,第22天治疗结束。治疗结束后测量左后足体积,计算足肿胀抑制率。
5)试验结果
化合物(3)对全弗氏佐剂诱导大鼠类风湿关节炎的足肿胀抑制率见下表:

  5. 如权利要求1所述的化合物,或其药学上可接受的盐,其特征在于,R 4选自H、氰基、带有或不带有取代基的C 1-6烷基。

  7. 一种药物组合物,其特征在于,所述的组合物含有权利要求1所述的化合物或其药学上可接受的盐、前药,以及药学上可接受的载体。
  8. 一种如权利要求1所述的化合物或其药学上可接受的盐的用途,用于:
    (a)制备治疗与JAK1和/或JAK2活性或表达量相关的疾病的药物;
    (b)制备JAK1和/或JAK2靶向抑制剂或降解剂;
    (c)体外非治疗性地抑制或降解JAK1和/或JAK2;
    (d)体外非治疗性地抑制肿瘤细胞增殖;和/或
    (e)治疗与JAK1和/或JAK2活性或表达量相关的疾病。
  9. 如权利要求8所述的用途,其特征在于,所述与JAK1和/或JAK2活性或表达量相关的疾病为肿瘤或自身免疫疾病。
  10. 一种抑制或降解JAK1和/或JAK2的方法,其特征在于,包括步骤:对作用对象施用有效量的如权利要求1所述的式I化合物或其药学上可接受的盐,或对抑制对象施用抑制有效量的如权利要求7所述的药物组合物。
