さらに、本発明の代表的な態様である水性PVAゲル等の水性ゲル(本明細書中において、本発明の水性ゲルともいう)は、白血球や抗体等の透過を阻害できることに加えて、補体の透過も阻害できるため、免疫に関与する細胞や抗体の隔離に加えて、免疫の作用を補佐する補体の隔離も行うことができる。すなわち、本発明の水性PVAゲルは、透過すべき酸素、無機有機の栄養分、種々のホルモンの中で最大と想定される直径5nm程度の分子(例えばインスリンなどのホルモンを含む生理活性物質)を透過させ、透過を阻止すべき免疫関連細胞や免疫関連物質の中で最小と想定される直径50nm程度の分子(例えば抗体、補体など)を透過させない選択性を有しているため免疫隔離層として使用でき、優れた免疫抑制作用を有する。
本開示は、調製時には水溶液もしくはゾルとすることができ、-5℃以上の温度でゲル形成が可能な、生体内酵素により主鎖が切断されないゲル形成高分子材料、代表的には、シンジオタクティシティがトライアッド表示で32~40%であるポリビニルアルコール樹脂(A)を成分として含む水性ゲルを免疫隔離層として有する、細胞又は組織包埋デバイスを包含する。
本発明の水性ゲルの代表的態様は、調製時には水溶液もしくはゾルであり、-5℃以上の温度に下げることによってゲル化した、シンジオタクティシティをコントロールしたPVA樹脂(さらには、シンジオタクティシティがトライアッド表示で、32~40%のPVA樹脂)を成分として含み、細胞又は組織包埋デバイスの免疫隔離層を形成するための水性ゲルである。
本開示において、ゾルは、好ましくはヒドロゾルである。
本発明で使用される生体内酵素により主鎖が切断されないゲル形成高分子材料は、同目的に使用される水系ゲル形成材料であるゼラチンやアルギン酸等とは異なり、好ましくは、主鎖が生体内酵素によって切断されない高分子材料であり、例えば、主たる部分が切断されないものであれば、片末端あるいは両末端に切断される主鎖を追加的に含むことができる。このような材料の代表例としては、繰り返し単位の主鎖としてエチレン構造を持つ重合体、特にポリビニルアルコール系樹脂やポリアクリル酸系樹脂を挙げることができ、中でもその側鎖に水溶性およびゲル形成を行う機能性官能基を持つものであることが好ましい。このような材料を用いた場合には、生体内に長時間存在させても生体内酵素によって主鎖が切断されないことから、長時間デバイス形状を保持することが可能となる。
本発明で使用される生体内酵素により主鎖が切断されないゲル形成高分子材料の代表的態様であるPVA樹脂(A)は、シンジオタクティシティがトライアッド表示で、通常32~40%であり、33~39%であることが好ましく、34~38%であることがさらに好ましい。シンジオタクティシティが32%以上であれば水性ゲルになりやすく、40%以下であれば水性ゲルの作製が容易となる。
なおトライアッド表示のシンジオタクティシティは、PVA樹脂(A)を重DMSOに溶解し、プロトンNMR測定による水酸基のピークより求めることができる。
ビニルエステル重合体の製造方法としては、ビニルエステル系単量体を重合する方法であれば特に限定されず、従来公知の方法に従って良い。
重合の際には、重合容器の形状、重合攪拌機の種類、さらには重合温度や、重合容器内の圧力等いずれも公知の方法を使用してかまわない。重合方法としては、従来から公知のバルク重合、溶液重合、懸濁重合、乳化重合等の各種の重合方法が可能である。重合度の制御や重合後に行うケン化反応のこと等を考慮すると、アルコールを溶媒とした溶液重合、あるいは、水又は水及びアルコールを分散媒とする懸濁重合が好ましいが、これらに限定されるものではない。

また、重合の際、脂肪族ビニルエステルの加水分解を防止する目的で、酒石酸、クエン酸、酢酸等の有機酸を添加してもよい。
鹸化反応に用いられる溶媒としては、メタノール、エタノール等のアルコール類;酢酸メチル、酢酸エチル等のエステル類;アセトン、メチルエチルケトン等のケトン類;ベンゼン、トルエン等の芳香族炭化水素類;テトラヒドロフラン等が挙げられ、これらは単独で又は2種以上を組み合わせて用いることができる。また、鹸化温度、時間等に特に制限されない。
また、鹸化物の乾燥、粉砕、洗浄方法も特に制限はなく、公知の方法を使用してもかまわない。
本発明の水性PVAゲルにおける平均孔径は、例えば、5nm以上500nm未満であり、好ましくは5nm以上200nm未満であり、さらに好ましくは5nm以上50nm未満である。
前記平均孔径は、例えば、走査型電子顕微鏡(日立S-4000型、日立製作所製)でゲル表面の写真(SEM写真、倍率1,000倍~5,000倍)をとり、得られた写真を画像処理装置(本体名:日本アビオニクス株式会社製、TVイメージプロセッサTVIP-4100II、制御ソフト名:ラトックシステムエンジニアリング株式会社製、TVイメージプロセッサイメージコマンド4198)に取り込んで得られる像における孔径を所定数測定し、それを演算処理することにより、平均孔径を求めることができる。
または、大気圧走査電子顕微鏡(例えば、日立ハイテクノロジーズ社製 AeroSurf 1500、日本電子株式会社製JASM-6200)、動的光散乱法(例えば株式会社堀場製作所製nano Partica SZ-100 Plus)又は走査型顕微光散乱法等により求めることができる。
本発明の水性ゲルに、生体組成物(B)を封入することにより、細胞又は組織包埋デバイスを形成することができる。
生体組成物(B)としては、特に限定されず、作製する細胞又は組織包埋デバイスの使用目的に応じて適宜選択することができる。
生体組成物(B)としては、本発明において水性ゲルを好ましく製造する温度(すなわち、-5~60℃)において安定に保存できる細胞(好ましくは生細胞)又は生体組織であれば、細胞又は生体組織の種類によらず、生理活性物質の供給能力が高い細胞又は組織包埋デバイスを得ることができるため、好ましい。
分化細胞としては、例えば、表皮細胞、平滑筋細胞、骨細胞、骨髄細胞、軟骨細胞、骨格筋芽細胞、膵実質細胞、膵島細胞、膵内分泌細胞、膵外分泌細胞、膵管細胞、肝臓細胞(例えば、肝細胞)、甲状腺細胞、副甲状腺細胞、副腎細胞、下垂体細胞、脾細胞、松果体細胞、腎臓細胞(腎細胞)、脾臓細胞、前葉細胞、成長ホルモン分泌細胞、ドパミン産生細胞、血液細胞、心筋細胞、骨格筋細胞、骨芽細胞、神経細胞、色素細胞、脂肪細胞等を用いることができる。上記細胞は、生体から単離された細胞のみならず、後述する幹細胞から分化誘導されたものであってもよい。
幹細胞(iPS細胞等)等の分化誘導されうる細胞については、分化誘導される細胞をデバイスに組み込んで、投与後に生体内で分化誘導させてもよいし、分化誘導した細胞をデバイスに組み込んで用いてもよい。
また、幹細胞としては、組織幹細胞(例えば、表皮幹細胞、毛嚢幹細胞、膵幹細胞・膵前駆細胞、肝幹細胞、神経幹細胞、網膜幹細胞、造血幹細胞、間葉系幹細胞等)、胚性幹細胞(ES細胞)、iPS細胞(induced pluripotent stem cell)等を用いることができるが、これらに限らない。
これらの細胞は、ヒト、ブタ、ラット又はマウス等の哺乳動物由来であって、患者等の生体にとって有用なホルモンやタンパク質等の生理活性物質を産生及び/又は分泌する細胞であることが好ましく、細胞の種類の選択は、移植する患者等の生体の疾患の種類によって決定することができる。また、これらの細胞がヒト細胞以外である場合、治療目的にヒト遺伝子を導入された細胞であってもよい。なお、生体にとって有用なホルモンとは、インスリン、甲状腺刺激ホルモン、甲状腺ホルモン、副甲状腺ホルモン、成長ホルモン、チロキシン、糖質コルチコイド、グルカゴン、エストラジオール、又はテストステロン等が例示される。生体にとって有用なタンパク質とは、具体的には、血液凝固因子、補体、アルブミン、グロブリン、各種酵素(代謝酵素又はアミラーゼ、プロテアーゼ、若しくはリパーゼ等の消化酵素)が例示される。その他の生理活性物質の例として、例えばドパミンなどの神経伝達物質等が挙げられる。
具体的に言えば、膵細胞(膵島細胞)、肝細胞、ドパミン産生細胞及びこれらの幹・前駆細胞が好ましく、膵細胞(膵島細胞)、肝細胞、及びこれらの幹・前駆細胞がより好ましく、膵細胞(膵島細胞)及び膵前駆(幹)細胞がより好ましい。
投与量は、医師が患者の年齢、性別、症状、副作用等を考慮して決定するので、一概には言えないが、通常成人1人当たり約1~10のデバイスを体内に移植することができる。例えば糖尿病患者に対する場合、患者の体重1kgあたり通常1000~100000IEQ(膵島量の国際単位:直径150μmの膵島の体積を1IEQと定義)、好ましくは5000~40000IEQ、より好ましくは10000~20000IEQの膵島を含有するデバイスを移植することができる。
デバイスの形状は特に限定されない。円盤状、球状、円柱状、楕円球状等を含むが、円盤状が好ましい。デバイスが円盤状の場合は、厚みと直径の積で表現すると、厚みは通常0.1mm~10cm、好ましくは0.1~5mm、より好ましくは0.5~2mmであり、直径は通常1mm~50cm、好ましくは1mm~10cm、より好ましくは2~4cm程度である。
材質は従来公知のものを用いてよい。
なお、本発明における生体組成物(B)としては、前記した細胞又は生体組織以外に、他の生体由来成分が含まれることを妨げるものではない。
また、本開示は、本発明の細胞又は組織包埋デバイスにおける細胞又は組織が、微生物生菌体由来である場合を除く場合を包含する。
本発明の水性ゲルに、生体組成物(B)とともに細胞培養成分(C)を封入することにより、細胞又は組織包埋デバイスを形成してもよい。
細胞培養成分(C)としては、特に限定されないが、例えば、Na、K、Cl、Ca、Glucose等を含有する酢酸あるいはリン酸緩衝液等が挙げられる。
細胞培養成分(C)がNaを含有する場合、Na濃度は、好ましくは20~150mEq/L、より好ましくは80~140mEq/Lに調整する。
Kを含有する場合、K濃度は、好ましくは2.5~130mEq/L、より好ましくは3.5~40mEq/Lに調整する。
Clを含有する場合、Cl濃度は、好ましくは15~170mEq/L、より好ましくは100~150mEq/Lに調整する。
Caを含有する場合、Ca濃度は、好ましくは0.5~5mEq/L、より好ましくは1~3mEq/Lに調整する。
Glucoseを含有する場合、Glucose濃度は、好ましくは1~11mM、より好ましくは3~7mMに調整する。
また、細胞培養成分(C)は、他の成分(例えば、徐放性付与剤、等張化剤、pH調整剤など)と組み合わせて使用してもよい。
このような状態の細胞培養成分(C)の含有量は、特に限定されないが、細胞又は生体組織の増殖、生存及び/又は生理活性物質の分泌等を阻害しない範囲であって、本発明の目的を損なわない範囲であることが好ましい。
上記状態の細胞培養成分(C)の添加量は、PVA樹脂(A)100質量部に対して、例えば、100~2000質量部、好ましくは150~1000質量部(例えば、200~300質量部、175~300質量部など)程度であってもよい。
例えば、PVA樹脂(A)の水溶液9mLに対して、10倍濃度の細胞培養成分(C)1mLであってもよい。
他の成分としては、例えば、生細胞の増殖を促進又は制御する物質である細胞増殖因子、細胞から産生される活性物質であるサイトカイン、その他の生理活性物質、細胞又は組織包埋デバイスへの血流を促進する血流促進物質、神経栄養因子等が挙げられる。これらは、1種単独で又は2種以上を併用して使用することができる。
細胞増殖因子としては、例えば、血小板由来増殖因子(PDGF)、上皮増殖因子(EGF)、線維芽細胞増殖因子(FGF)、肝細胞増殖因子(HGF)、血管内皮増殖因子(VEGF)、インスリン等が挙げられる。
サイトカインとしては、例えば、造血因子(例えば、インターロイキン類、ケモカイン類、コロニー刺激因子等)、腫瘍壊死因子、インターフェロン類等が挙げられる。
その他の生理活性物質として、例えば、アミノ酸(例えば、グリシン、フェニルアラニン、リジン、アスパラギン酸、グルタミン酸等)、ビタミン類(例えば、ビオチン、パントテン酸、ビタミンD等)、血清アルブミン、抗生物質等が挙げられる。
血流促進物質としては、例えば、シトルリン又はその塩、カプサイシン、カプサイシノイド類が挙げられる。
神経栄養因子としては、例えば、NGF(nerve growth factor;神経成長因子)、BDNF(brain-derived neurotrophic factor;脳由来神経栄養因子)、NT-3(neurotrophin-3;ニューロトロフィン-3)、NT-4(neurotrophin-4;ニューロトロフィン-4)、GDNF(Glial-Cell Derived Neurotrophic Factor;グリア細胞株由来神経栄養因子)、ニュールツリン(neurturin)、アルテミン(artemin)、パーセフィン(persephin)等が挙げられる。
なお、前記他の成分の添加量は、特に限定されない。
本発明の細胞又は組織包埋デバイスに使用される水性ゲルは、PVA樹脂(A)を水性溶媒中で溶液化(溶解)しゲル化した水性ゲルを用いるのであれば特に限定されずに作製することができる。PVA樹脂(A)の溶解方法は、特に限定されず、例えば、加熱や撹拌等を行うことによって溶解することができる。また、溶解は、密閉下で行ってもよいし、必要に応じて加圧を行ってもよい。なお、水性溶媒としては、通常、水が使用される。
また、溶液化の時間は、温度や圧力、溶液濃度によって適宜変更できるが、例えば、1分~12時間、30分~10時間等である。
放置温度としては、生細胞の保存に適した温度であれば特に制限はないが、例えば、-5℃以上であり、好ましくは-5~60℃(例えば、0~60℃)であり、より好ましくは-3~50℃(例えば、0~50℃)であり、さらに好ましくは0~40℃である。このような範囲であれば、生存細胞数の減少が少ないため好ましい。また、放置温度は、PVA樹脂(A)水溶液(ゾル状態であってもよい)、又は水性ゲルを凍結させることなく、該水溶液、又はゲルで生細胞や生体組織を包埋できる温度が好ましい。本発明では、特定のシンジオタクティシティを有するPVA樹脂(A)を用いることにより、生細胞や生体組織の保存に適するよう比較的低い温度(例えば、上記範囲の温度)で、水性ゲルを作製することができる。
水性ゲルを作製する際の放置時間は、PVA樹脂(A)の濃度、放置温度等により適宜選択することができるが、通常は1時間~3、4日程度、好ましくは1時間~2日程度さらに好ましくは2~12時間程度である。1時間以上であれば、細胞又は組織包埋デバイスを体内に留置した際に容易に崩壊しない等の観点から、好ましい。
水性ゲルに封入される直前の生体組成物(B)の生細胞数全体に対する、水性ゲル、又は細胞若しくは組織包埋デバイスにおける生存細胞数の割合は、例えば60~100%、好ましくは70~100%、より好ましくは80~100%である。生細胞数は、例えば、Fluorescein diacetate試薬による細胞質染色およびpropidium iodideによる細胞核染色により測定(FDA/PI測定と略すこともある)することができる。
得られる水性ゲルは、後述する免疫隔離層として機能し得るように、細胞を安定に保持するとともに、酸素やブドウ糖、インスリンなどの生体にとって有用なホルモン、その他の生理活性物質を透過させ、免疫系のタンパク質を透過させない構造を有する。
水性PVAゲルの応力は、例えば島津製作所製小型卓上試験機EZTest EZ-SXを用いその使用説明書に従って測定することができる。
また、該水性ゲルの成形方法としては、例えば、PVA樹脂(A)(さらに、所望により、好ましくは生体組成物(B)及び細胞培養成分(C))を含む水溶液(ゾル状態であってもよい)を、ゲル化する前に目的とする形状の型に流し込む方法、得られたゲルをナイフ等で目的の形状に加工する方法等が挙げられる。
なお、通常PVA樹脂(A)(さらに、所望により、好ましくは生体組成物(B)及び/又は細胞培養成分(C)等)を含む水溶液は、ゲル状態に至るまでにゾル状態を経由する。そのようなゾル状態も本発明の水性ゲル等価物として本発明の範囲内であると理解される。
PVA樹脂(A)を含む水溶液(ゾル状態であってもよい)の固形分濃度は、例えば、0.3~20%、好ましくは0.5~10%、より好ましくは1~8%である。このような範囲であれば、細胞又は組織包埋デバイスを動物に移植後、体内で長期にデバイス形態を維持し、免疫隔離能を保持できる等の観点から好ましい。
本発明の水性ゲルは、細胞又は組織包埋デバイスの免疫隔離層として使用することができる。
「免疫隔離層」とは、例えば、ブドウ糖;インスリン、甲状腺刺激ホルモン、甲状腺ホルモン、副甲状腺ホルモン、成長ホルモン、チロキシン、糖質コルチコイド、グルカゴン、エストラジオール、又はテストステロンなどのホルモン;血液凝固因子、アルブミン、グロブリン、各種酵素(代謝酵素又はアミラーゼ、プロテアーゼ、若しくはリパーゼ等の消化酵素)などのタンパク質;ドパミンなどの神経伝達物質等を透過しつつも、例えば抗体、補体、又は白血球などの免疫系のタンパク質を透過しない層を意味する。
細胞又は組織包埋デバイスは、生体組成物(B)を包埋又は含有していればよく、例えば、バイオ人工臓器などであってもよい。
細胞又は組織包埋デバイスの応力は、例えば島津製作所製小型卓上試験機EZTest EZ-SXを用いその使用説明書に従って測定することができる。
水性ゲルは、その補強及び/又は操作性の簡便化のために、補強材として有用な支持基材(D)を組み合わせて用いてもよい。
例えば、水性ゲルを薄膜シート状にする場合には、その補強及び操作性の簡便化のために、樹脂製メッシュシート等の基材(補強材)に固定してゲル化するのがよい。
支持基材(D)の素材は、限定されるものではないが、例えば、高分子[例えば、PET(ポリエチレンテレフタレート)、PE(ポリエチレン)、PP(ポリプロピレン)、テフロン(登録商標)等]、金属等が挙げられ、生体内で分解、変質しないことが好ましいが、一定期間経過後生体内で分解されるものであってもよい。
メッシュシートのメッシュ(網目)サイズは、透過すべき酸素、無機有機の栄養分、種々のホルモンの中で最大と想定される直径5nm程度の分子(例えばインスリンなどのホルモンを含む生理活性物質)を透過させ、透過を阻止すべき免疫関連細胞や免疫関連物質の中で最小と想定される直径50nm程度の分子(例えば抗体、補体など)を透過させないように、通常5~100nm、好ましくは10~50nm、より好ましくは20~30nmである。
図4に示されるように、膵臓から良質な膵島細胞が分離されて、移植用膵島が準備される(図4参照)。膵島細胞同士の凝集を防ぐために膵島細胞を上記したメッシュ(2枚)で固定する。このように準備された固定された膵島細胞とPVA樹脂(A)から本発明のデバイスが製造され、最内層は膵島細胞であり、インスリンを発散している。第2層はメッシュ層で細胞を支持している。最外層は、ゲル表面であって、免疫隔離膜を形成している。この免疫隔離膜は生体適合性が高く、インスリンを通過させるが免疫関連物質は通過させない(図5参照)。
本発明のデバイスはそのまま生体内に適用できるが、例えば、公知技術に従って容易に設けられる新生血管を用いて構築された網に収納されて医療効果を発揮する。デバイスは容易に取り出し、又は交換できる(図6参照)。
このデバイスは下記の特徴の少なくとも1つを奏することができる。
(1)包埋する細胞の質を高く維持する。
(2)移植膵島がホスト患者の免疫システムから適切に隔離されている。
(3)酸素・グルコースが供給され、かつ適切なインスリン応答が可能である。
(4)低侵襲な移植が可能であり、必要に応じて体外への容易な取出しや取り換えが可能である。
(1)シンジオタクティシティ
d6-DMSO溶液中で1H-NMRを測定しトライアッド表示のシンジオタクティシティ(%)を求めた。
なお、トライアッド表示のシンジオタクティシティ(%)とは、4~5ppmの3本の水酸基プロトン(低磁場側からアイソタクティック[I]、ヘテロタクティック[H]、シンジオタクティック[S])比率から求めたもので、下記計算式に基づく。

d6-DMSO溶液中で1H-NMRを測定し鹸化度(モル%)を求めた。
(3)重合度
JISK6725記載のベンゼン溶液、30℃におけるポリ酢酸ビニル換算の重合度を測定した。
[合成例1]
攪拌機、温度計、滴下ロ-ト及び還流冷却器を取り付けたフラスコ中に、ピバリン酸ビニル800部、メタノール190部を仕込み、系内の窒素置換を行った後ジャケットを80℃に加熱し、還流が発生した時点で2,2’-アゾビス(2,4-ジメチルバレロニトリル)0.07部をメタノール10部に溶解した溶液を添加し重合を開始し、6時間後に重合停止剤としてm-ジニトロベンゼンを添加し、重合を停止した。重合収率は58%であった。
得られた反応物からメタノール、ピバリン酸ビニルを除去後、アセトンを加えてポリピバリン酸ビニルを溶解し40%アセトン溶液を得た。
この40%アセトン溶液250部に水酸化カリウムの25%メタノール溶液110部とを加えてよく混合し、50℃で2時間鹸化反応を行った。得られたゲル状物を粉砕し、アセトン150部、水酸化カリウムの25%メタノール溶液340部中でさらに50℃で4時間鹸化反応を行った。反応後酢酸で中和し、固液分離を行い、メタノール、水で洗浄、乾燥後、30gのPVA樹脂1を得た。
得られたPVA樹脂1の鹸化前の重合度は1450であり、鹸化度は99.91モル%であった。また、トライアッド表示によるシンジオタクティシティは37.1%であった。
ピバリン酸ビニル800部を、ピバリン酸ビニル685部に変更し、酢酸ビニル115部を追加した以外は合成例1と同様にしてPVA樹脂2を作製した。なお、モル比(ピバリン酸ビニル/酢酸ビニル)は80/20であった。得られたPVA樹脂2の鹸化前の重合度は1510であり、鹸化度は99.90モル%であった。また、トライアッド表示によるシンジオタクティシティは35.7%であった。
ピバリン酸ビニル800部を、ピバリン酸ビニル553部に変更し、酢酸ビニル247部を追加した以外は合成例1と同様にしてPVA樹脂3を作製した。なお、モル比(ピバリン酸ビニル/酢酸ビニル)は60/40であった。得られたPVA樹脂3の鹸化前の重合度は2030であり、鹸化度は99.89モル%であった。また、トライアッド表示によるシンジオタクティシティは34.0%であった。
攪拌機、温度計、滴下ロ-ト及び還流冷却器を取り付けたフラスコ中に、ピバリン酸ビニル2000部、メタノール38部を仕込み、系内の窒素置換を行った後ジャケットを80℃に加熱し、還流が発生した時点で2,2’-アゾビス(2,4-ジメチルバレロニトリル)0.03部をメタノール2部に溶解した溶液を添加し重合を開始し、3時間後に重合停止剤としてm-ジニトロベンゼンを添加し、重合を停止した。重合収率は17%であった。
得られた反応物からメタノール、ピバリン酸ビニルを除去後、アセトンを加えてポリピバリン酸ビニルを溶解し25%アセトン溶液を得た。
この25%アセトン溶液200部に水酸化カリウムの25%メタノール溶液54部とを加えてよく混合し、50℃で2時間鹸化反応を行った。得られたゲル状物を粉砕し、アセトン150部、水酸化カリウムの25%メタノール溶液168部中でさらに50℃で4時間鹸化反応を行った。反応後酢酸で中和し、固液分離を行い、メタノール、水で洗浄、乾燥後、17gのPVA樹脂4を得た。
得られたPVA樹脂4の鹸化前の重合度は4440であり、鹸化度は99.86モル%であった。また、トライアッド表示によるシンジオタクティシティは36.7%であった。
ピバリン酸ビニル2000部を、ピバリン酸ビニル1600部に変更し、酢酸ビニル268部を追加した以外は合成例4と同様にしてPVA樹脂5を作製した。なお、モル比(ピバリン酸ビニル/酢酸ビニル)は80/20であった。得られたPVA樹脂5の鹸化前の重合度は4530であり、鹸化度は99.93モル%であった。また、トライアッド表示によるシンジオタクティシティは35.5%であった。
ゾル状態のPVA樹脂(A)水溶液Aとして、細胞培養成分(C)を含むゾル状態のPVA樹脂(A)水溶液を2種作製した。作製して得られたゾル状態のPVA樹脂(A)水溶液を以下、ゾルA-1、及びゾルA-2とも表記する。
1.425gの上記PVA樹脂1を蒸留水23.575gと混合し、焦げなどが生じないように160℃、35rpmで3時間回転撹拌して溶解し、90℃まで冷却した後に得られたゾル状態のPVA樹脂(A)水溶液を、予め90℃に温めたガラス製バイアル瓶に回収し、90℃で保温した。その後、作製したゾル状態のPVA樹脂(A)水溶液を42度のウォーターバスへ入れ、ゾル状態のPVA樹脂(A)水溶液8.8mLに10倍濃度HBSS(ハンクス平衡塩溶液)を1.0mL、1M HEPESを0.2mL加え、チューブを上下に震盪させ攪拌した。その後、遠心機(久保田商事株式会社製、商品名:ハイブリッド高速冷却遠心機6200)でスピンダウンし、42℃で静置することでゾルA-1(PVA樹脂濃度:5w/v%)を得た。得られたゾルA-1を3.5cmディッシュに移した。
1.135gの上記PVA樹脂1を蒸留水23.865gと混合し、焦げなどが生じないように160℃、35rpmで3時間回転撹拌して溶解し、90℃まで冷却した後に得られたゾル状態のPVA樹脂(A)水溶液を、予め90度に温めたガラス製バイアル瓶に回収し、90℃で保温した。その後、作製したゾル状態のPVA樹脂(A)水溶液を42℃のウォーターバスへ入れ、ゾル状態のPVA樹脂(A)水溶液8.8mLに10倍濃度HBSS(ハンクス平衡塩溶液)を1.0mL、1M HEPESを0.2mL加え、チューブを上下に震盪させ攪拌した。その後、遠心機(久保田商事株式会社製、商品名:ハイブリッド高速冷却遠心機6200)でスピンダウンし、42℃で静置することでゾルA-2を得た。得られたゾルA-2を3.5cmディッシュに移した。
上記で作製したゾルA-1及びA-2について、下記にしたがって固形分濃度を測定した。
ゾルの固形分濃度は、加熱乾燥式水分計(エーアンド・デイ、MS-70)を用いて測定した。水分計の試料皿にガラス繊維シートを置き、各ゾル約1gをシートに均一に浸み込ませ、試料皿温度120℃、測定時間(加温時間)15分の条件で各ゾルの固形分を測定した。測定時の水分計の表示は固形分(%)を選択した。固形分を算出する計算式は、乾燥後質量/乾燥前質量×100(%)である。ゾルA-1の固形分濃度は6.5質量%、ゾルA-2の固形分濃度は5.5質量%と算出された。
11~14週齢の雄性Lewisラット(日本エスエルシー)を膵島分離に使用した。0.8mg/mL collagenase type V(Sigma-Aldrich社製)を溶解した冷ハンクス緩衝液(HBSS)をラット総胆管から注入した膵臓を、37℃で12分間消化し膵組織から膵島を分離した。Histopaque-1119(Sigma-Aldrich社製)とLymphoprep(AXIS-SHIELD、Norway)を用いて濃度勾配遠心を行い、膵島を回収した。膵島は5.5 mmol/l グルコースと10%胎児ウシ血清(Fetal Bovine Serum:FBS)を含むRPMI1640培地で37℃、5%CO2下で一晩培養した後、実施例および比較例の検討に使用した。
スライドガラスの上に、上記ディッシュ上にのせられた上記作製済みのゾルA-1又はA-2について、60μLをのせ、4℃にて10分間静置した。その上に直径15mmの円形型PETメッシュ(株式会社サンプラティック社製、商品名:PETメッシュシート(呼称TN120))をかぶせ、ゾルA-1又はA-2 60μLに、上記で調製した膵島細胞から培地成分を可及的に取り除いたものを懸濁させて得られた懸濁液を上記PETメッシュ上にのせた。ゲルローディングチップを用いて膵島細胞の懸濁液をPETメッシュ上に拡げ、膵島細胞の懸濁液を挟み込む様にPETメッシュをさらにその上にかぶせた。さらにPETメッシュの上にゾルA-1又はA-2を250μLのせ、その上からスライドガラスをかぶせた。このようにして構築したゾルを湿箱の中に留置し、4℃下で3時間静置し、膵島包埋デバイス(水性ゲル)を得た。なお、上記膵島包埋デバイスはいずれも、膵島量は800IEQsであった。
[実施例1]
上記にて構築したゾルA-1を用いた膵島包埋デバイス(水性ゲル)をスライドガラスから外し、6wellプレートに5mL/wellの割合で保存培地(グルコース濃度を5.5mMに調整し、10%FBSを含有したRPMI1640培地)に浸し、4℃下で2時間保存した。
ストレプトゾトシン誘発糖尿病C57BL/6Jマウスの皮下に、上記4℃下で2時間保存後のゾルA-1を用いた膵島包埋デバイス(水性ゲル)を留置することで移植を行った。
上記移植後、経時的に血糖値を測定して治癒効果を確認した(n数=4)。その平均値±標準偏差を図1及び表1に示す。なお、血糖値200mg/dl以下で○(糖尿病治癒)、200mg/dl超を×(糖尿病状態)と判定した。
(PVA溶液組成)
重合度5000、鹸化度99.3モル%、トライアッド表示によるシンジオタクティシティが29.5%のPVAを用い、PVAが3%、FBS(Fetal bovine serum: 牛胎児血清)が10%、Dimethyl sulfoxide(ジメチルスルホキシド)が5%、Nicotinamide(ニコチンアミド)が10mMとなるように、ETK(ET-Kyoto)溶液に溶解させて得られたPVA溶液を凍結融解式膵島デバイス作製に用いた。
培地成分を可及的に取り除いたLewisラットの膵島細胞のみを1.5mLチューブに入れ、4℃のセルバンカー(十慈フィールド株式会社製)1.0mLを加えて膵島細胞を懸濁し、氷冷下に1分間置いた後、セルバンカーを取り除いた。1mmスペーサー付スライドガラスの上に、上記PVA溶液を塗布したPETメッシュ(株式会社サンプラティック社製、商品名:PETメッシュシート(呼称TN120)、10×15mm)をのせ、該PETメッシュ上に、160μLのPVA溶液に膵島細胞のみを懸濁させて得られた懸濁液を拡げてから、さらにPVA溶液を塗布したPETメッシュを被せた。この上にスライドガラスをのせて、厚さ1mmの凍結融解式膵島デバイスを作製した。
作製した凍結融解式膵島デバイスを-80℃で24時間静置した後、型から外して氷冷UW溶液(University of Wisconsin solution、臓器保存溶液)中で融解した。さらに、氷冷UW溶液に5分間ずつ3回浸漬し、ゲル内の溶液組成をUW溶液に置換してから、UW溶液中に4℃下で24時間保存した。
氷冷した膵島培養用培地(RPMI1640培地:5.5mMグルコース、10%FBS、抗生剤含)10mLでデバイス表面のUW溶液を洗浄してから培地(氷冷)に5分間ずつ3回浸漬し、ゲル内の溶液組成を培地に置換した後、3mLの培地で37℃24時間培養した。
皮下よりもグラフト生着が容易であることが知られているストレプトゾトシン誘発糖尿病C57BL/6Jマウスの腹腔内に、上記保存後の凍結融解式膵島デバイスを留置することで移植を行った。
移植後、実施例1と同様に経時的に血糖値を測定し治癒効果を評価した。その結果を表1及び図2に示す。
表2に示すように、移植膵島量および移植するデバイスの個数を変更した以外は比較例1と同様に凍結融解式膵島デバイスを作製、移植し糖尿病治癒評価を行った。その結果を表1、図2及び図3に示す。
ゾルA-1の製造方法において、PVA樹脂1に代えて、PVA樹脂2又は3を用いた以外は同様にして、膵島包埋デバイス(水性ゲル)を得た。得られた水性ゲルを用いて試験例1と同様に移植試験を行ったところ、いずれの水性ゲルを用いた場合においても、PVA樹脂1を用いた場合と実質的に同じ効果が得られた。
また、ゾルA-1の製造方法において、PVA樹脂1に代えて、トライアッド表示によるシンジオタクティシティが36.5%又は39.1%となるように、重合温度を変更して調製したPVA樹脂を用いた以外は同様にして、膵島包埋デバイス(水性ゲル)を得た。得られた水性ゲルを用いて試験例1と同様に移植試験を行ったところ、いずれの水性ゲルを用いた場合においても、PVA樹脂1を用いた場合と実質的に同じ効果が得られた。
(FDA/PI測定)
表3に示すPVA樹脂(A)、濃度に変更し、実施例1で使用した膵島デバイスと同様の方法で膵島デバイスを作製し、3mL/wellのPBS(室温)で3分間ずつ2回洗浄した。アセトン(和光純薬工業、東京、日本)で5mg/mLに溶解したFluorescein diacetate(FDA: Calbiochmem、San Diego、USA)15μLと、蒸留水で0.5mg/mLに溶解したPropidium iodide (PI: Sigma-Aldrich, St.Louis、MO、USA) 20μLを6wellプレートに入れた3mLのPBSに添加してFDA/PI染色溶液とし、その中にPBSで洗浄後の膵島デバイスを移し、暗所で5分間染色後、3mLのPBS溶液で3分間洗浄した。膵島デバイスをカバーガラス(松浪硝子工業株式会社、大阪、日本)の上にのせ、蛍光顕微鏡(BZ-900:キーエンス、東京、日本)を用いて、FDA(励起波長470/40nm、吸収波長525/50)とPI(励起波長540/25nm、吸収波長605/60)の膵島内における局在を観察した。
何れの実施例においても、FDA測定(染色)では生細胞の存在を確認(FDA(+))し、死細胞はほぼ認められなかった。一方PI測定では細胞核が染色されず(PI(-))、FDA染色の結果と同様に死細胞がほぼ存在しないことが明らかとなった。
比較例1で作製した膵島デバイスを実施例と同様の方法でFDA/PI測定を行った。FDA測定では染色像の存在が確認できず、PI測定では明瞭な細胞核の染色が見られ、両測定結果から比較例におけるデバイス内の膵島は広範に渡り壊死を引き起こしている事が確認された。
(水性ゲル、細胞又は組織包埋デバイスの固形分濃度、応力測定)
PVA樹脂1と水を使用し160℃にて溶解、90℃に冷却後、直径34mmの円柱容器内に充填後、42℃30分、4℃3時間放置することで、直径34mm×高さ17mmの水性ゲルを作製した。
得られた水性ゲルの固形分濃度(PVA樹脂1濃度)は5.0%で、20℃における応力は0.7kPaであった。
固形分濃度は、加熱乾燥式水分計(エーアンド・デイ、MS-70)を用いて測定した。水分計の試料皿にガラス繊維シートを置き、水性ゲル約1gをシートに均一に浸み込ませ、試料皿温度120℃、測定時間(加温時間)15分の条件で水性ゲルの固形分を測定した。測定時の水分計の表示は固形分(%)を選択した。固形分を算出する計算式は、乾燥後質量/乾燥前質量×100(%)である。
応力測定は、島津製作所製小型卓上試験機EZTest EZ-SXを使用し、その使用説明書に従って測定した。具体的には、直径34mm×高さ17mmの水性ゲルを直径20mmの円柱治具を用い、20%押し込み時の応力を求めた。
表4に示す通り、PVA樹脂(A)の種類、濃度を適宜変更し、4℃における放置時間を変更した以外は実施例6と同様に水性ゲルを作製し、固形分濃度、応力を求めた。
表5に示す通り、PVA樹脂(A)の種類、濃度を適宜変更し、HBSS(ハンクス平衡塩溶液)を使用し、4℃における放置時間を変更した以外は実施例6と同様に細胞又は組織包埋デバイスを作製し、固形分濃度、応力を求めた。
Claims (20)
- シンジオタクティシティがトライアッド表示で32~40%であるポリビニルアルコール樹脂(A)を成分として含む水性ゲルを免疫隔離層として有する、細胞又は組織包埋デバイス。
- 水性ゲルが、-5℃以上でのゲル化履歴を有することを特徴とする請求項1に記載の細胞又は組織包埋デバイス。
- 水性ゲルが、応力0.3~30kPaを有することを特徴とする請求項1又は2記載の細胞又は細胞包埋デバイス。
- ポリビニルアルコール樹脂(A)の鹸化度が90~99.99モル%、重合度が100~10000である請求項1~3のいずれか記載の細胞又は組織包埋デバイス。
- ポリビニルアルコール樹脂(A)が、ピバリン酸ビニルを重合成分とする重合体の鹸化物であって、ピバリン酸ビニル単位を含む請求項1~4のいずれかに記載の細胞又は組織包埋デバイス。
- 免疫隔離層に、生体組成物(B)及び細胞培養成分(C)が封入されている請求項1~5のいずれか1項に記載の細胞又は組織包埋デバイス。
- 生体組成物(B)が、膵島細胞、膵管細胞、肝臓細胞、神経細胞、甲状腺細胞、副甲状腺細胞、腎細胞、副腎細胞、下垂体細胞、脾細胞、脂肪細胞、骨髄細胞、間葉系幹細胞、ES細胞及びiPS細胞からなる群から選択される1以上である請求項6に記載の細胞又は組織包埋デバイス。
- 生体組成物(B)が、膵島細胞又は肝臓細胞である請求項6に記載の細胞又は組織包埋デバイス。
- 細胞培養成分(C)が、Na、K、Cl、Ca及びGlucoseからなる群から選択される1以上を含有する酢酸又はリン酸緩衝液である請求項6~8のいずれか1項に記載の細胞又は組織包埋デバイス。
- 支持基材(D)を含有する請求項1~9のいずれか1項に記載の細胞又は組織包埋デバイス。
- 支持基材(D)の素材が、PET、PE、PP、テフロン(登録商標)及び金属からなる群から選択される1以上である請求項10に記載の細胞又は組織包埋デバイス。
- 応力0.3~30kPaを有することを特徴とする請求項1~11のいずれかに記載の細胞又は細胞包埋デバイス。
- ポリビニルアルコール樹脂(A)を含む水溶液と、細胞培養成分(C)を混合した後に、生体組成物(B)を混合して得られる混合物をゲル化せしめる工程を含む請求項6~12のいずれか1項に記載の細胞又は組織包埋デバイスの製造方法。
- 水性ゲルを作製する際の温度を-5℃以上にする請求項13に記載の細胞又は組織包埋デバイスの製造方法。
- シンジオタクティシティがトライアッド表示で32~40%であるポリビニルアルコール樹脂(A)を含む水性ゲルを含有する細胞又は組織包埋デバイスの免疫隔離層形成剤。
- ポリビニルアルコール樹脂(A)の鹸化度が90~99.99モル%、重合度が100~10000である請求項15に記載の剤。
- ポリビニルアルコール樹脂(A)が、ピバリン酸ビニルを重合成分とする重合体の鹸化物であって、ピバリン酸ビニル単位を含む請求項16に記載の剤。
- (1)ポリビニルアルコール樹脂(A)を含み、(2)封入されている生体組成物(B)の分泌成分を透過させ、且つ免疫関連細胞又は免疫関連物質の透過を抑制することを特徴とする水性ゲルを免疫隔離層として有する、細胞又は生体組織包埋デバイス。
- -5℃以上でゲル化する性質を有することを特徴とする、シンジオタクティシティがトライアッド表示で32~40%であるポリビニルアルコール樹脂(A)、生体組成物(B)及び細胞培養成分(C)を含有する混合物。
- 生理活性物質産生細胞または組織を直接、あるいは支持材料で支持したものに対して、生体内酵素により主鎖が切断されないゲル形成高分子材料を含む保護ゲル層形成材料の溶液もしくはゾルを適用し、その後-5℃以上の温度でゲル形成を行う、生理活性物質産生細胞または組織に対する保護ゲル層の形成方法。
