Abstract
The invention relates to an investigation or analysis of materials, specifically to production of specimens for investigation, in particular microparticles and micro-traces from a carrier object of plant or animal origin, and can be used in palynology, biology, ecology, medical science for spore and pollen analysis and for forensic, merchandising and ecological assessments. In a method for removing microparticles and micro-traces from an object of plant or animal origin from the surface of the carrier object, the material is washed by a physical and mechanical method at a temperature of 20-25°C in a monobasic carboxylic acid solution with the addition of a surface-active substance, followed by sedimentation. Alkaline hydrolysis of the sediment is then carried out using a mixture of ammonium hydroxide and alkali, followed by rinsing, drying and staining the microparticles and micro-traces. The technical result of the claimed invention consists in improving the quality of samples in the preparation stages and in producing as full a set of microparticles and micro-traces as possible by the choice of solvents and washing regimes, which makes it possible to accelerate routine operations for taking microparticles and micro-traces of plant or animal origin from a carrier object, and to increase the effectiveness of the results of an identification analysis as a whole.

Предлагаемое изобретение является доступным, эргономичным и экологически безопасным способом изъятия микрочастиц для получения «чистых» качественных проб с набором микрочастиц. Способ позволяет получить максимально полный набор микрочастиц и микроследов растительного и животного происхождения, в том числе пыльцевых зерен и спор высших растений, палиноморфов, мицелия, бактерий, простейших, фитолитов, обломков растительного и животного происхождения для характеристики свойств объекта- носителя. Способ также позволяет получить дифференцированный набор микрочастиц и микроследов растительного и животного происхождения с учетом времени роста объекта-носителя, т.к. дополнительным или горячим смывом снимают глубоко зафиксированные микрочастицы, осевшие в месте произрастания объекта-носителя; основным или холодным смывом - более поздние микроследы, связанные с упаковкой и перемещением объекта-носителя. Использование заявляемого способа ускоряет рутинные работы - время проведения работ в среднем 2,5 ч на 1 пробу, а также увеличивает эффективность результатов идентификационного анализа - количество трудно или неидентифицируемых микрообъектов колеблется в пределах 1-8%.

The method also allows to obtain a differentiated set of microparticles and micro-traces of plant and animal origin, taking into account the growth time of the carrier object, because deep or fixed microparticles settled in the place of growth of the carrier object are removed with an additional or hot wash; main or cold flush - later micro-traces associated with the packaging and movement of the carrier object. Using the proposed method speeds up routine work - the average time of work is 2.5 hours per 1 sample, and also increases the effectiveness of the results of identification analysis - the number of hard or unidentifiable micro-objects ranges from 1-8%.
